Abstract:We aimed to investigate the effects of Clostridium butyricum-, Bacillus subtilis-, and Bacillus licheniformis-based potential probiotics on the growth performance, intestinal morphology, immune responses, and caecal short chain fatty acids (SCFAs) and microbial structure in broiler chickens. Three treatment groups containing a total of 1200 one-day-old AA broilers were included: birds fed with a basal diet only (Con), birds fed with added 10(10) probiotics cfu/kg (ProL), and birds fed with added 10(11) probiotics cfu/kg (ProH). The dietary probiotics significantly improved the final and average body weights and serum immunoglobulins A, M, and Y. The probiotics also enhanced the ileal morphology and improved the caecal acetate, butyrate, and propionate contents. Furthermore, 16S rRNA sequencing revealed that dietary compound probiotics modulated the caecal microflora composition as follows: (1) all birds shared 2794 observed taxonomic units; (2) treatment groups were well separated in the PCA and PCoA analysis; (3) the relative abundance of Parabacteroides, Ruminococcaceae_UCG-014, Barnesiella, Odoribacter, [Eubacterium_coprostanoligenes_group], [Ruminococcus]_torques_group, and Butyricimonas significantly varied between treatments. The compound probiotics improved the growth performance, serum immune responses, the ratio of ileal villus height to crypt depth, and major caecal SCFAs in broiler chickens. The dietary C. butyricum-, B. subtilis-, and B. licheniformis-based probiotics improved overall broiler health and would benefit the poultry industry.

Abstract:A total of 600, 1 day old male Lingnan Yellow broiler chickens were used to investigate the effects of Clostridium butyricum (C. butyricum) on growth performance, nitrogen metabolism, intestinal morphology and cecal microflora in broiler chickens. The birds were randomly assigned into 5 treatments (6 replicate pens per treatment with 20 birds per pen) and fed the same antibiotic-free basal diets during a 42 days feeding experiment. The treatments were as follows: no addition (Control), 2.5 x 10(7) cfu C. butyricum kg(-1) of diet (CB1), 5x10(7) cfu C. butyricum kg(-1) of diet (CB2), 1x10(8) cfu. C. butyricum kg(-1) of diet (CB3) and 10 mg colistine sulfate kg(-1) of diet (Antibiotic). Compared with the control birds, birds fed either CB1 or CB2 or antibiotic diet had greater (p<0.05) Body Weight (BW) on day 21 and 42 and higher (p<0.05) Average Daily Gain (ADG) from day 1-42. Birds fed C. butyricum or antibiotic diet had lower (p<0.05) Feed-to-Gain ratio (F:G) than the control birds from day 1-42. Dietary C. butyricum decreased (p<0.05) the concentration of serum Uric Acid (UA) compared with the control diet on day 21. Supplementation with CB2 or CB3 decreased (p<0.05) serum ammonia concentration compared with the control diet on day 21 and 42. Birds fed C. butyricum diet had higher (p<0.05) ileal villus height than the control birds on day 21 and 42. Birds fed CB2 or CB3 diet had lower (p<0.05) ileal crypth depth than the control birds on day 21 and 42. Supplementation with CB1 or CB3 decreased (p<0.05) the population of cecal Escherichia coli (E. coli) compared with the control on day 21. Birds fed C. butyricum diet had higher (p<0.05) population of cecal Bifidobacterium on day 21 and birds fed CB2 diet had higher (p<0.05) cecal Bifidobacterium on day 42 compared with the control birds. Supplementation with CB1 or CB2 increased (p<0.05) the number of cecal Lactobacillus on day 21 and supplementation with CB2 increased (p<0.05) Lactobacillus on day 42 compared with the control or antibiotic groups. The results indicate that supplementation with C. butyricum promotes growth performance, modulates nitrogen metabolism improves intestinal morphology and balances cecal microflora in broiler chickens.

Abstract:This experiment was conducted to investigate the effects of Clostridium butyricum on growth per-formance, antioxidant capacity, immune function and serum biochemical parameters of broilers.A total of 540 one-day-old Ross 308 broilers were randomly assigned to 3 groups with 6 replicates per group and 30 birds per replicate for a 42-day trial.Dietary treatments were:an antibiotic-free basal diet ( control group, CON) , basal diet supplemented with 10 mg/kg colistin sulfate and 50 mg/kg bacitracin zinc ( antibiotic group, ANT) , and basal diet supplemented with 3×108 CFU/kg Clostridium butyricum (Clostridium butyricum group, CB).The results showed as follows:1) the body weight ( BW) , average daily gain ( ADG) and average daily feed in-take ( ADFI) of broilers in CB and ANT were significantly increased ( P<0.05) compared with those in CON. 2 ) The activities of serum glutathione peroxidase ( GSH-Px) and total superoxide dismutase ( T-SOD) in broil-ers at 42 days of age were significantly enhanced ( P<0.05) compared with those in CON, and GSH-Px activi-ty was 60.00%higher ( P<0.05) than that in ANT.At 7 and 21 days of age, serum T-SOD activity of broilers in ANT was significantly increased ( P<0.05) compared with that in CON.3) Compared with CON, serum immunoglobulin A ( IgA) content in broilers at 21 days of age in CB and ANT was increased by 28.70%( P<0.05) and 26.46%( P<0.05) , and serum immunoglobulin G ( IgG) content in broilers at 7, 21 and 42 days of age was increased by 36.60%( P<0.05) , 37.77%( P<0.05) and 27.03%( P<0.05) , respectively.Serum IgG content of broilers at 7 and 42 days of age in ANT was significantly higher ( P<0.05) than that in CON. Serum immunoglobulin M ( IgM) content of broilers at 21 and 42 days of age in CB was significantly increased compared with CON, while 7.92%( P>0.05) and 47.62%( P<0.05) higher than that in ANT.4) Serum to-tal protein ( TP) content of broilers in CB was significantly higher ( P<0.05) than that in CON, especially in-creased by 31.33%( P<0.05) and 52.27%( P<0.05) compared with that of broilers at 21 and 42 days of age in ANT.Meanwhile, serum ammonia content of broilers in CB was significantly decreased ( P<0.05) com-pared with that of broilers at 21 and 42 days of age in CON.These results indicated that supplementation with Clostridium butyricum in diet can enhance serum antioxidant capacity and immune function, promote protein metabolism, and then improve the growth performance of broilers;moreover, it can aslo reduce ammonia emis-sions from broilers.

Abstract:The effects of feeding dehydrated Enterococcus faecium on growth performance, immune response, and cecal microflora in broiler chickens challenged with Escherichia coli K88 were investigated. Two hundred eighty-eight 1-d-old birds were randomly assigned to 4 treatments: negative control birds (N-con) fed a basal diet and not challenged with E. coli K88; positive control birds (P-con) fed a basal diet and challenged with E. coli K88; birds fed a basal diet including dehydrated E. faecium (Ef) at 1 × 10(9) cfu/kg of feed and challenged with E. coli K88; and birds fed a basal diet including the antibiotic colistine sulfate (Anti) at 10 mg/kg of feed and challenged with E. coli K88. Birds fed E. faecium had greater (P < 0.05) BW on d 14, 21, and 28 and greater (P < 0.05) jejunal villus height on d 21 and 28 compared with birds on the other treatments. Jejunal crypt depth was decreased (P < 0.05) in birds fed either E. faecium or antibiotic compared with P-con treatment birds on d 10, 21, and 28. Birds fed E. faecium had a greater (P < 0.05) concentration of IL-4 in their jejunal mucosa than did those in the N-con treatment group on d 10, 14, and 21. Infected birds, with or without E. faecium feeding, had a higher (P < 0.05) tumor necrosis factor-α and secreted IgA in their jejunal mucosa than did those in the N-con treatment group on d 10 and 14. Birds fed E. faecium had lower (P < 0.05) concentrations of E. coli on d 14 and 28, less (P < 0.05) Clostridium perfringens on d 28, greater Lactobacillus counts on d 14 and 21, and greater (P < 0.05) Bifidobacterium in their cecal contents on d 21 than did the P-con birds. These results suggest that E. faecium can promote growth performance, improve intestinal morphology, and beneficially manipulate the cecal microflora in broilers challenged with E. coli K88.
Effects of Composite Bacillus sp. on Growth Performance, Cecal Microflora and Ammonia Emission from Excreta in Broilers

Abstract:A Feeding trial was conducted to study the effects of different concentrations of CompositeBacillus sp.(Clostridium butyricum and Bacillus licheniformis,CBG) on growth performance,cecal microflora,and ammonia emission from excreta in broilers,to explore its action mechanism.Six hundred one-day-old Lingnan yellow-feathered broilers were randomly assigned to 4 groups with three replicate pens of 50 broilers each.There were 4 treatments: Control group,fed basal diet supplemented with 20 mg/kg Virginiamycin and 50 mg/kg Rroxarsone;and 3 experimental groups receiving basal diet supplemented with different concentrations of Composite Bacillus sp.(100,150,200 mg/kg,respectively).The experiment lasted for 50 days.The results showed that compared to the control,no significant differences were observed for growth performance after the supplementation of 100 mg/kg and 150 mg/kg Composit e Bacillus sp.(P >0.05).Composite Bacillus sp.had a significantly increasing effect on the populations of Lactobacillus of caecum at d 50(P <0.05).At 50 d of age,the pH value of caecum was significantly decreased after supplementation of 150 mg/kg and 200 mg/kg Composite Bacillus sp.preparation(P<0.05),and the group with 150 mg/ kg CBG got the optimal effect.With the supplementation of 150 mg/kg Composite Bacillus sp.,and uric acid content of its feces and caecum in day 50 was significant decreased(P<0.05).Composite Bacillus sp.significantly decreased the pH value of manure at day 43(P <0.05),and decreased the ammonia emission from excreta effectively,especially a better result happening on 150 mg/kg Composite Bacillus sp.group.Those indicated that Composite Bacillus sp.could regulate the intestinal microflora,decrease pH and the nitrogen content of feces and caecum,thus reduce the ammonia emission from manure.Results demonstrated that 150 mg/kg Composite Bacillus sp.is the best dosage in diets of broilers.
Modulation of broilers' caecal microflora and metabolites in response to a potential probiotic Bacillus amyloliquefaciens.

Abstract:Studies have found that a dietary supplement of Bacillus amyloliquefaciens improved the growth performance, increased the nutrient digestibility of hosts and modulated the intestinal microflora. A total of 360 1-day-old Ross broilers were randomly divided into three treatments: a control group with a basal diet, an antibiotic group with a basal diet and added colistin sulphate, and a probiotics group with a basal diet and added Bacillus amyloliquefaciens. The HiSeq high-throughput sequencing analysis of 16S rRNA was used to investigate the differences in birds' caecal microflora, and metabolomics was used to analyse changes in caecal metabolites. Results showed that the supplementation of Bacillus amyloliquefaciens significantly improved the BW and ADG compared with the control birds. Results of sequencing indicated that (i) 645, 670, 596 unique operational taxonomic units (OTUs) were found in birds supplemented with Bacillus amyloliquefaciens on day 7, 21 and 42, separately, (ii) due to the diversity and relative abundance of the birds' caecal microflora, the OTUs of the caecal microflora clustered according to age and treatment, except on day 42, (iii) among the six predominate families (Ruminococcaceae, Lachnospiraceae, Enterobacteriaceae, Erysipelotrichaceae, Lactobacillaceae and Rikenellaceae), the supplementation of Bacillus amyloliquefaciens significantly increased Enterobacteriaceae on day 42, (iv) Bacillus amyloliquefaciens increased the relative abundance of Faecalibacterium and Ruminococcus on day 21, increased the Faecalibacterium and Blautia and decreased the Ruminococcus on day 42. The metabolomics of caecal metabolites showed that the dietary Bacillus amyloliquefaciens changed the caecal metabolites involved of amino acid metabolism and glyceride metabolism, and the antibiotics changed the caecal metabolites that were related to carbohydrates and amino acid metabolism on day 21.

Abstract:This trial was conducted to investigate the adhesion of Bacillus amyloliquefaciens to Caco-2 cells, the effects of Bacillus amyloliquefaciens on the mucosal immunity, cecal volatile fatty acids and microbial diversity in broiler chickens. Three hundred and sixty 1-d-old Ross 308 chicks were randomly divided into 3 dietary treatments groups, which birds fed with basal diet, basal diet supplemented with colistine sulfate, and basal diet supplemented with Bacillus amyloliquefaciens. Polymerase chain reaction denaturing gradient gel electrophoresis was used to analyze the change of cecal microflora, and Gas Chromatography was used to analyze the cecal volatile fatty acids. Data showed that: 1) Bacillus amyloliquefaciens had a good adhesion ability to epithelial cells; 2) the supplementation of Bacillus amyloliquefaciens significantly increased the concentration of ileal mucosal secretory IgA and interleukin 6, decreased (P Lass Than 0.05) the concentration of tumor necrosis factor-a; 3) resulted in the change of cecal microbial community, higher levels of acetic acid, methylacetic and isovaleric acid in the birds. Thus, we considered that Bacillus amyloliquefaciens enhanced the mucosal immunity, increased the cecal concentration of major volatile fatty acids and the diversity microflora community in broilers.
Intestinal Morphologic and Microbiota Responses to Dietary Bacillus spp. in a Broiler Chicken Model

Abstract:Dietary inclusion of probiotic Bacillus spp. beneficially affect the broiler chickens by balancing the properties of the indigenous microbiota causing better growth performance. The effects of three Bacillus spp. on the growth performance, intestinal morphology and the compositions of jejunal microflora were investigated in broiler chickens. A total of 480 1-day-old male Arbor Acres broilers were randomly divided into four groups. All groups had six replicates and 20 birds were included in each replicate. The control birds were fed with a corn-soybean basal diet, while three treatment diets were supplemented with Bacillus coagulans TBC169, B. subtilis PB6, and B. subtilis DSM32315 with a dosage of 1 × 109 cfu/kg, respectively. The experiment lasted for 42 days. The compositions and diversity of jejunal microflora were analyzed by MiSeq high-throughput sequencing. The B. coagulans TBC169 group showed marked improvements of growth performance, nutrient digestibility and intestinal morphology compared with the other B. subtilis treatments. B. coagulans TBC169 supplementation improved the average body weight (BW), average daily weight gain (ADG), total tract apparent digestibility of crude protein and gross energy (GE), and reduced feed conversion rate (FCR) compared with the control group (P < 0.05). The villus height to crypt depth ratio (VH/CD) of jejunum and duodenum was increased in the birds fed with B. coagulans TBC169 compared with the control group (P < 0.05). However, two B. subtilis treatments presented more positive variation of the jejunum microflora of chickens than that in the B. coagulans TBC169 group. B. subtilis PB6 and B. subtilis DSM32315 treatments improved the diversity of jejunal microbiota on day 21 compared with the control (P < 0.05), while which were decreased on day 42 (P < 0.05). The supplementation with B. coagulans TBC169 significantly improved the proportion of Firmicutes, otherwise two B. subtilis significantly improved the proportion of Proteobacteria, Bacteroidetes, Actinobacteria, and Acidobacteria at the phylum level during starter phase and decreased the proportion of Bacteroidetes during growing phase compared with the control. The supplementation with B.subtilis DSM32315 significantly improved the proportion of Clostridiales during starter phase, whereas two B. subtilis significantly improved the proportion of Pseudomonas, Burkholderia, Prevotella, DA101 during growing phase at the genus level compared with the control. In conclusion, the dietary supplementation with probiotic Bacillus spp. strains improved body weight and intestinal morphology in broiler chickens, which might be associated with the gut microbiota.

Abstract:The recent ban on the use of antibiotics as a feed additive has led to the search for alternative sources of antibiotics in the feed industry. Presently, probiotics are considered as a potential substitute for antibiotic as a live biotherapeutic agent to improve animal health and performance. Accordingly, study was focused on evaluating the effect of Saccharomyces boulardii (Sb) and Bacillus subtilis B10 (Bs) on ultrastructure modulation and mucosal immunity development in broiler chickens. A total of three hundred 1-d-old Sanhuang broilers (a Chinese cross breed) were randomized into 3 groups, each group with 5 replications (n = 20). The control group (Ctr) was fed a basal diet containing an antibiotic (virginiamycin, 20 mg/kg). Meanwhile, broilers in experimental groups received Sb and Bs (1 × 108 cfu/kg of feed) in addition to the basal diet for 72 d. The results of the experimental groups revealed a significant improvement in live BW and relative weight of bursa of Fabricius and thymus. Also, intestinal villus height, width, and number of goblet cells increased in the Sb and Bs groups. Meanwhile, modulation in the intestinal ultrastructure and increased mRNA expression levels of occluding, cloudin2, and cloudin3 (P < 0.05) were observed in the Sb and Bs groups. Moreover, IgA-positive cells significantly increased in the jejunum of Sb- and Bs-supplemented groups (P < 0.05). Intestinal cytokines interleukin-6, tumor necrosis factor-α, interleukin-10, transforming growth factor-β, and secretory IgA concentrations were (P < 0.05) improved in the probiotic groups; however, Sb induced inflammatory and antiinflammatory cytokines (P < 0.05) in comparison with the Ctr group. The present findings conclusively revealed that Sb and Bs increased IgA-positive cells in the lumen of the intestinal villus and revealed that Sb and Bs could modulate intestinal ultrastructure through increasing occluding, cloudin2, and cloudin3 mRNA expression levels in broiler intestine.

Abstract:Knowledge about the modulation of gut microbiota improves our understanding of the underlying mechanism by which probiotic treatment benefits the chickens. This study examined the effects of Bacillus subtilis DSM 32315 on intestinal structure and microbial composition in broilers. Broiler chicks were fed basal diets without or with B. subtilis supplementation (1.0 × 109 spores/kg of diet). Supplemental B. subtilis increased average body weight and average daily gain, as well as elevated villus height and villus height to crypt depth ratio of ileum in broilers. Multi-dimension analysis showed a certain degree of separation between the cecal microbiota from treatment and control groups. Increased Firmicutes abundance and reduced Bacteroidetes abundance in cecum were observed responded to B. subtilis addition, which also increased the abundances of Christensenellaceae and Caulobacteraceae, and simultaneously decreased the abundances of potentially harmful bacteria such as Vampirovibrio, Escherichia/Shigella and Parabacteroides. Network analysis signified that B. subtilis addition improved the interaction pattern within cecal microbiota of broilers, however, it exerted little influence on the metabolic pathways of cecal microbiota by comparison of the functional prediction of metagenomes. In conclusion, supplemental B. subtilis DSM 32315 improved growth performance and intestinal structure of broilers, which could be at least partially responsible by the manipulation of cecal microbial composition.

Abstract:In this study, we investigated the effects of dietary supplementation with Bacillus subtilis (QST713) on the performance and intestinal health of yellow feather broilers under Coccidia and Clostridium perfringens (CP) challenge or CP alone. One-day-old yellow-feathered broiler roosters (n = 600) were randomly assigned to 5 groups (6 replicates with 20 roosters per replicate): the Con blank group, the CIC.p group (d24 Coccidia+d28-30 of CP challenge), the CIC.p + BS group (CIC.p +100 mg/kg B. subtilis), the C.p group (d 28-34 of CP challenge), and the C.p +BS group (C.p +100 mg/kg B. subtilis). The experiment lasted 80 d. The birds were evaluated for parameters such as average daily gain (ADG), average daily feed intake (ADFI), feed efficiency (F/G), intestinal lesion score, villus histomorphometry, intestinal tight junctions, inflammatory factors, and cecal microorganisms. The results revealed that 1) C.p. increased the F/G of broilers from 22 to 42 d (P < 0.05), whereas CIC.p. significantly decreased the 42 d and 80 d body weights (BW) and 22-42 d and 1-80 d ADG (P < 0.05) and significantly increased the 22 to 42 d and 1 to 80 d F/G (P < 0.05). The number of intestinal lesions significantly increased at 35 d and 42 d (P < 0.05). CIC.p significantly decreased the jejunum and ileum villus height (VH) and the ileum villus height/crypt depth (P < 0.05) at 35 d. The challenge significantly upregulated the expression of Claudin-1 and IL-4 mRNAs in the jejunum at 35 d and significantly downregulated the expression of IL-10 mRNA in the ileum at 35 d (P < 0.05); the number of unique OTUs in the challenge group decreased significantly after challenge treatment, and the relative abundances of Romboutsia at 35 d and Cladomyces and Lactobacillus at 42 d decreased significantly (P < 0.05). 2) Compared with the challenge groups, the addition of BS decreased the F/G of broilers from 22 to 42 d. Compared with the CIC group, the addition of BS significantly increased the F/G of broilers from 22 to 42 d. Compared with that in the CIC.p group, the addition of BS significantly increased the VH in the jejunum and ileum at 35 d (P < 0.05). Compared with the challenge groups, the BS groups presented significantly lower mRNA expression levels of Claudin-1 (P < 0.05) in the jejunum at 35 d. The Shannon and Chao indices suggested that BS increased the alpha diversity of cecum microorganisms in broilers. Dietary supplementation with B. subtilis can alleviate the damage to intestinal morphology and intestinal barrier function, as well as the altered cecal flora structure in broilers caused by Coccidia and C. perfringens infections.

Abstract:This study aimed to investigate the impact of Bacillus subtilis HC6 on the growth performance, immunity, antioxidant capacity, and intestinal health of broilers. A total of 180 one-day-old white feather broilers were randomly divided into two experimental groups, each comprising six replicates of fifteen chicks from 1 to 50 d of age. The groups were either fed a basal diet (CON) or the same diet supplemented with 5 × 108 cfu/kg of Bacillus subtilis HC6 (BS). Our results indicated that compared with the CON, dietary supplementation with BS increased feed efficiency during d 21–50 and d 1–50 (p < 0.05). Moreover, BS supplementation enhanced antioxidant capacity in the serum and liver, and also decreased the activity of diamine oxidase and the level of endotoxins (p < 0.05). Additionally, BS treatment increased the villi height in the jejunum and ileum, increased the ratio of villus height/crypt depth in the ileum, upregulated the expression of tight junction proteins in the jejunal mucosa, and downregulated the levels of IL-22 and IFN-γ on day 50 (p < 0.05). Principal coordinates analysis yielded clear clustering of two groups; dietary BS increased the relative abundance of Bacteroidales_unclassified (genus) and Olsenella (genus), and decreased the abundance of genera Alistipes on day 50, which identified a strong correlation with FCR, serum differential metabolites, or differential gene expression in the jejunal mucosa by spearman correlation analysis. The PICRUSt2 analysis revealed that supplementation with BS enriched the pathways related to xenobiotics biodegradation and metabolism, carbohydrate metabolism, energy metabolism, signaling molecules and interaction, the digestive system, and transport and catabolism. These results demonstrated that dietary BS increased feed efficiency, antioxidant capacity, and the mRNA expression of pro-inflammatory cytokines in the jejunal mucosa; and decreased the activity of diamine oxidase in serum, which might be attributed to the modulation of community composition and the functions of cecal microbiota in white-feathered broilers.

Abstract:We aimed to investigate the effects of Clostridium butyricum-, Bacillus subtilis-, and Bacillus licheniformis-based potential probiotics on the growth performance, intestinal morphology, immune responses, and caecal short chain fatty acids (SCFAs) and microbial structure in broiler chickens. Three treatment groups containing a total of 1200 one-day-old AA broilers were included: birds fed with a basal diet only (Con), birds fed with added 1010 probiotics cfu/kg (ProL), and birds fed with added 1011 probiotics cfu/kg (ProH). The dietary probiotics significantly improved the final and average body weights and serum immunoglobulins A, M, and Y. The probiotics also enhanced the ileal morphology and improved the caecal acetate, butyrate, and propionate contents. Furthermore, 16S rRNA sequencing revealed that dietary compound probiotics modulated the caecal microflora composition as follows: (1) all birds shared 2794 observed taxonomic units; (2) treatment groups were well separated in the PCA and PCoA analysis; (3) the relative abundance of Parabacteroides, Ruminococcaceae_UCG-014, Barnesiella, Odoribacter, [Eubacterium_coprostanoligenes_group], [Ruminococcus]_torques_group, and Butyricimonas significantly varied between treatments. The compound probiotics improved the growth performance, serum immune responses, the ratio of ileal villus height to crypt depth, and major caecal SCFAs in broiler chickens. The dietary C. butyricum-, B. subtilis-, and B. licheniformis-based probiotics improved overall broiler health and would benefit the poultry industry.

Abstract:Four hundred and fifty 1-d-old male Lingnan Yellow broiler chickens were used to investigate the effects of Clostridium butyricum on growth performance, immune function, and cecal microflora. The birds were randomly assigned to 5 treatments and offered the same antibiotic-free basal diets for 42 d. The treatments were as follows: no addition (control), 1 × 10(7) cfu C. butyricum/kg of diet (CB1), 2 × 10(7) cfu C. butyricum/kg of diet (CB2), 3 × 10(7) cfu C. butyricum/kg of diet (CB3), and 10 mg of colistine sulfate/kg of diet (antibiotic). Birds fed either CB2 or antibiotic had greater overall BW than those in the control group. During d 1 to 7, d 21 to 42, and d 1 to 42, birds fed either CB2 or CB3 or the antibiotic diet had greater ADG compared with those in the control group. No significant differences were observed in BW or ADG among the CB2, CB3, and antibiotic groups. Birds fed the CB2 or CB3 diet had greater concentrations of IgA and IgG in the serum from d 14 to 42 and greater IgM in the serum from d 21 to 42 than those in the control group. Birds fed the CB3 diet had a greater concentration of complement component 3 in the serum than those in the control group from d 7 to 42. Dietary C. butyricum decreased (P < 0.05) Escherichia coli in cecal contents on d 14 and 42, and both CB2 and CB3 decreased (P < 0.05) cecal Salmonella and Clostridium perfringen from d 14 to 42 compared with the control. Broilers fed either CB2 or CB3 had greater cecal Lactobacillus and Bifidobacterium counts from d 21 to 42, and birds fed C. butyricum had greater cecal C. butyricum counts during the whole period compared with those in the control group. The results indicate that C. butyricum promotes growth performance and immune function and benefits the balance of the intestinal microflora in broiler chickens.

Abstract:This study investigated the impact of Bacillus coagulans (BC) and tributyrin (TB) supplementation on the growth performance, serum antioxidant capacity, intestinal morphology, and cecal microbiota of yellow-feathered broilers. Using a 2 × 2 factorial design, 480 broilers were randomly assigned to four experimental diets, comprising two levels of BC (0 and 1 g/kg) and two levels of TB (0 and 1 g/kg), over a 36-day period. A significant interaction was observed between BC and TB, impacting the average daily feed intake (ADFI) of broilers aged between 26 and 40 days (p < 0.01). BC and TB also displayed a significant interaction in relation to serum malondialdehyde levels and total antioxidant capacity (p < 0.05). Additionally, there was a significant interaction between BC and TB concerning the duodenal villus-to-crypt ratio, crypt depth, and jejunal villus-to-crypt ratio (p < 0.05). The addition of BC and TB significantly enhanced the richness and diversity of cecal microbiota, with a notable interactive effect observed for the abundance of Faecalibacterium, Ruminococcus_torques_group, and Phascolarctobacterium. In conclusion, supplementation with BC and TB can effectively improve the growth performance, serum antioxidant capacity, intestinal morphology, and cecal microbiota composition of yellow-feathered broilers, indicating the presence of an interactive effect.
Supplemental Bacillus subtilis PB6 Improves Growth Performance and Gut Health in Broilers Challenged with Clostridium perfringens

Abstract:Clostridium perfringens (CP) is the principal pathogenic bacterium of chicken necrotic enteritis (NE), which causes substantial economic losses in poultry worldwide. Although probiotics are known to provide multiple benefits, little is known about the potential effects of Bacillus subtilis (B. subtilis) application in preventing CP-induced necrotic enteritis. In this study, 450 male Arbor Acres broilers were divided into 5 experimental treatments: A: basal diet (control group); B: basal diet and CP challenge (model group); C: CP challenge+10 mg/kg enramycin (positive control group); D: CP challenge+ 4 × 10 7 CFU / kg of feed B. subtilis PB6 (PB6 low-dosage group); and E: CP challenge+ 6 × 10 7 CFU / kg of feed B. subtilis PB6 (PB6 high-dosage group). There were 6 replicate pens per treatment with 15 broilers per pen. The present research examined the effect of Bacillus subtilis PB6 (B. subtilis PB6) on growth performance, mRNA expression of intestinal cytokines and tight junctions, and gut flora composition in broilers challenged with CP. The entire experiment was divided into two phases: the non-CP challenge phase (d0–18) and the CP challenge phase (d18–26). PB6 did not increase the growth performance during the first stage, but the PB6 high-dosage group was found to have larger body weight gain and ADFI during the CP challenge stage. Feed supplementation with PB6 reduced the lesion score of challenged chicks, with increased tight junction-related gene expression (occludin and ZO-1) and decreased TNF-α expression compared with CP-infected birds. A decrease in the abundance of Clostridium XI, Streptococcus, and Staphylococcus was observed after CP infection ( P < 0.05 ), while supplementation with PB6 restored the ileal microbial composition. In conclusion, administration of B. subtilis PB6 improved growth performance, enhanced intestinal barrier function, and mitigated intestinal inflammation/lesions, which might be due to its restoring effects on the ileal microbial composition in CP-challenged broilers.

Abstract:Introduction: This study evaluated the effects of dietary supplementation of Bacillus subtilis and Pichia farinose mixture (BPM) on growth performance, apparent ileal digestibility, cecal bacteria counts, small intestinal morphology and digestive enzymes activities, and jejunal nutrient transporters gene expression in broiler chicks. Methods: A total of 768 one-day-old Ross 308 broiler chicks were randomly Q18 assigned into 3 groups based on the initial body weight (42.00 ± 0.08 g). The experimental periods were 35 days. There were 16 replicates per group and 16 birds per cage. Dietary treatments included a basal diet supplemented with 0, 0.1, or 0.2% BPM to form CON, BPM0.1 (consisting Bacillus subtilis with 1.0 × 107 viable spore and Pichia farinose with 1.0 × 107 viable spore per kg diet), and BPM0.2 (consisting Bacillus subtilis with 2.0 × 107 viable spore and Pichia farinose with 2.0 × 107 viable spore per kg diet) groups. Results and discussion: Dietary supplementation of graded levels of BPM has positive effects on growth performance of broiler chicks, manifesting in the increase of body weight gain during days 1–35 as well as the decrease of feed conversion ratio during days 1–7, 21–35, and 1–35. Moreover, BPM supplementation positively improved ileal energy and crude protein digestibility, increased Lactobacillus counts, optimized intestinal morphology, enhanced intestinal digestive enzymes activities, and upregulated jejunal SGLT-1, GLUT-2, and PEPT-1 expression. Therefore, BPM supplementation improved growth performance of broiler chicks, which was partially related to the improvement in intestinal nutrient absorption capacity.

Abstract:A total of 240 1-day-old Arbor Acres male broilers were randomly divided into five dietary treatments (control feed (CON), supplemented with 75 mg/kg aureomycin (ANT), supplemented with 7.5 × 108 CFU/kg (Ba1) and 2.5 × 109 CFU/kg (Ba1), and 7.5 × 109 CFU/kg (Ba3) Bacillus amyloliquefaciens TL106, respectively) to investigate the probiotic effect of TL106 instead of antibiotics in broilers. On days 1–21, the average daily gain of broilers in the Ba groups was increased compared with the CON group (p < 0.05). In addition, the feed/gain ratio of broilers in the Ba groups was lower than that of broilers in the CON and ANT groups on days 22–42 and days 1–42 (p < 0.05). Compared with the CON group, dietary TL106 increased the digestibility of crude fiber and crude protein (p < 0.05), and the effect was similar to that of the ANT group. The levels of IL-1β, IFN-γ, and IL-6 in serum, jejunum, and ileum of broilers fed TL106 were decreased compared with the control group (p < 0.05). The mRNA expression of tight junction proteins in broilers of ANT and Ba groups was higher than the control group (p < 0.05). After 21 days, villus height and the ratio of villus height to crypt depth of duodenum and jejunum of broilers fed TL106 were higher than the control group (p < 0.05). The concentrations of short-chain fatty acids such as lactate, acetate, propionate, and butyrate in cecal digesta of broilers dietary TL106 were higher than the control group (p < 0.05). The supplementation with TL106 altered the compositions and diversity of the cecal microbiota of broilers. Moreover, supplementation with TL106 improved the ratio of Firmicutes to Bacteroidetes and decreased the relative abundance of Proteobacteria on days 21 and 28, while the abundance of Peptostreptococcaceae, Ruminococcaceae and Lactobacillaceae was increased. On days 35 and 42, broilers fed TL106 had an increased total abundance of Firmicutes and Bacteroidetes and decreased abundances of Lactobacillaceae, while the abundance of Barnesiellaceae was increased. In conclusion, dietary supplementation with TL106 improved the broiler's growth performance, immune response capacity, gut health, modulated development, and composition of the gut microbiota in broilers. It is suggested that Bacillus amyloliquefaciens TL106 may be a suitable alternative to in-feed antibiotics to improve broiler health and performance.

Abstract:The aim of this study was to investigate the effects of the combination of probiotics replacing antibiotics on growth performance, serum biochemical parameters, intestinal morphology, and expression of tight junction proteins in intestinal mucosa of broilers. A total of 168 Arbor Acres broilers (45.04 ± 0.92 g) were randomly divided into three treatments, with seven replicates per treatment, and eight broilers per replicate. The experiment included phases 1 (d 0 to 21) and 2 (d 21 to 42). The dietary treatments contained a corn soybean meal-based diet (control group; CON); an antibiotic group (basal diet + 75 mg/kg chlortetracycline; CTC), and a probiotics group (basal diet + probiotics (500 mg/kg in phase 1 and 300 mg/kg in phase 2; Bacillus subtilis 5 × 109 CFU/g, Bacillus licheniformis 2.5 × 1010 CFU/g and Saccharomyces cerevisiae 1 × 109 CFU/g; PB). The results showed broilers fed PB had improved (p < 0.05) feed conversion ratio (FCR) in phase 1 and increased (p < 0.05) average daily gain (ADG) in phase 2, as well as improved (p < 0.05) ADG and FCR overall (d 0 to 42). The apparent total tract digestibility (ATTD) of dry matter, organic matter, gross energy, and crude protein was increased (p < 0.05) in broilers fed PB, while the ATTD of dry matter and organic matter was enhanced in broilers fed CTC compared with CON. Broilers fed PB showed increased (p < 0.05) serum total antioxidant capacity concentrations and tended to have higher (p = 0.06) level of serum immunoglobulin M in phase 1 compared with CON. These broilers also had increased (p < 0.05) level of serum immunoglobulin A in phase 2 in comparison with CON and CTC. Moreover, broilers fed CTC and PB showed increased (p = 0.05) villus height to crypt depth ratio in duodenum, as well as higher (p < 0.05) mRNA expression of zonula occludens-1 in jejunum compared with CON. In conclusion, dietary supplementation with PB as chlortetracycline substitute could improve the growth performance, nutrient digestibility, serum antioxidant capacity, jejunal mucosal barrier function, and intestinal morphology of broilers.

Abstract:This study investigated the effects of dietary supplementation with Bacillus subtilis (B. subtilis) or Bacillus licheniformis (B. licheniformis) on growth performance, immunity, antioxidant capacity, short chain fatty acid (SCFA) production, and the cecal microflora in broiler chickens. In total, 360 male, 1-day-old Cobb 500 birds were randomly divided into 3 groups: the control group was fed a basal diet; the B. subtilis group was fed a basal diet supplemented with 1.5 × 109 CFU/kg B. subtilis; the B. licheniformis group was fed a basal diet supplemented with 1.5 × 109 CFU/kg B. licheniformis. Results showed that chickens supplemented with either B. subtilis or B. licheniformis had comparatively higher (P < 0.05) body weight and average daily gain, whereas no difference (P > 0.05) was observed in feed efficiency. Concentrations of serum IgA, IgY, and IgM, as well as anti-inflammatory IL-10 were significantly increased (P < 0.05), and proinflammatory IL-1β and IL-6 were significantly decreased (P < 0.05) by B. subtilis or B. licheniformis supplementation. Moreover, chickens fed with diets supplemented by either B. subtilis or B. licheniformis had greater antioxidant capacity, indicated by the notable increases (P < 0.05) in glutathione peroxidase, superoxide dismutase, and catalase, along with decrease (P < 0.05) in malondialdehyde. Compared to the control group, levels of SCFA, excluding acetic and propionic acid, in cecal content had improved (P < 0.05) by adding B. licheniformis, and significant increase (P < 0.05) in acetic and butyric acid was observed with B. subtilis supplementation. Microbial analysis showed that both B. subtilis or B. licheniformis supplementation could increase butyrate-producing bacteria such as Alistipes and Butyricicoccus, and decrease pathogenic bacteria such as the Synergistetes and Gammaproteobacteria. In summary, dietary supplemented with B. subtilis or B. licheniformis improved growth performance, immune status, and antioxidant capacity, increased SCFA production, and modulated cecal microbiota in chickens. Moreover, B. licheniformis was more effective than B. subtilis with the same supplemental amount.
