Abstract:A therapeutic breakthrough in cancer treatment has recently been made by using magnetic nanoparticles (MNPs) for heating in hyperthermia therapy. Unapproachable tumors are being effectively destroyed by focused heat produced by MNPs. The major challenges regarding practical application of this therapy include the control and improvement of induction heating ability of MNPs and hyperthermia temperature range between 42–47 °C, for secure treatment at targeted area. Here, in this study, we have reported the synthesis and characterization of core–shell structured silica-coated cobalt ferrite nanoparticles (SiO2 coated CoFe2O4 MNPs), which are potential candidates for use as heat source in magnetic hyperthermia therapy. The SiO2 coated CoFe2O4 MNPs were synthesized using the reverse micelle method, with the SiO2 coating performed simultaneously during MNP synthesis. Various analytical tools were utilized for the characterization. The structural measurements were probed by X-ray diffraction (XRD). Monodisperse MNPs with nearly spherical core–shell structure was revealed by Transmission electron microscopy (TEM) results. The average diameter of MNPs obtained from TEM analysis was 15 nm. The surface coating of cobalt ferrite MNPs with silica was verified by Fourier transform infrared spectrometry (FTIR). The saturation magnetization values obtained using vibrating sample magnetometer (VSM) measurements were 45.74 emu/g, revealing the superparamagnetic nature of the nanoparticles. The heating efficiency of different concentrations of synthesized nanoparticles was evaluated in an aqueous solution under an alternating magnetic field of strength 5.5 kA/m at a frequency of 260 kHz. A saturation temperature of 42 °C was attained at an optimum concentration of 1.7 mg/mL, while a magnetic field strength of 3.9 kA/m achieved 42 °C at a fixed concentration of 3.5 mg/ml. The obtained specific absorption rate (SAR) values for all the samples were between 56.23 to 100.1 W/g. The correlation between SAR values, MNP concentration, and applied magnetic field strength was also examined. The high saturation magnetization, efficient heating rates, and high SAR values make our synthesized silica-coated cobalt ferrite nanoparticles promising candidates for magnetic hyperthermia treatments, potentially improving therapeutic outcomes for cancer patients.

Abstract:In this report, the heating efficiencies of γ-Fe2O3 and hybrid γ-Fe2O3-TiO2 nanoparticles NPs under an alternating magnetic field (AMF) have been investigated to evaluate their feasible use in magnetic hyperthermia. The NPs were synthesized by a modified sol-gel method and characterized by different techniques. X-ray diffraction (XRD), Mössbauer spectroscopy and electron microscopy analyses confirmed the maghemite (γ-Fe2O3) phase, crystallinity, good uniformity and 10 nm core sizes of the as-synthesized composites. SQUID hysteresis loops showed a non-negligible coercive field and remanence suggesting the ferromagnetic behavior of the particles. Heating efficiency measurements showed that both samples display high heating potentials and reached magnetic hyperthermia (42 °C) in relatively short times with shorter time (~3 min) observed for γ-Fe2O3 compared to γ-Fe2O3-TiO2. The specific absorption rate (SAR) values calculated for γ-Fe2O3 (up to 90 W/g) are higher than that for γ-Fe2O3-TiO2 (~40 W/g), confirming better heating efficiency for γ-Fe2O3 NPs. The intrinsic loss power (ILP) values of 1.57 nHm2/kg and 0.64 nHm2/kg obtained for both nanocomposites are in the range reported for commercial ferrofluids (0.2-3.1 nHm2/kg). Finally, the heating mechanism responsible for NP heat dissipation is explained concluding that both Neel and Brownian relaxations are contributing to heat production. Overall, the obtained high heating efficiencies suggest that the fabricated nanocomposites hold a great potential to be utilized in a wide spectrum of applications, particularly in magnetic photothermal hyperthermia treatments.

Abstract:Uniform magnetic nanoparticle-loaded polymer nanospheres with different loading contents of manganese ferrite nanoparticles were successfully synthesized using a flexible emulsion process. The MnFe2O4-loaded polymer nanospheres displayed an excellent dispersibility in both water and phosphate buffer saline. The effect of loading ratio and size of MnFe2O4 nanoparticles within the nanospheres on the specific absorption rate (SAR) under an alternating magnetic field was investigated. Our results indicate that a large size (here 18 nm) and a low loading ratio are preferable for a high SAR. For a smaller particle size (6 nm), the low loading ratio did not result in an enhancement of the SAR value, while a very low SAR value is expected for 6 nm. In addition, the SAR of low-content MnFe2O4 (18 nm)-loaded polymer nanospheres in the agarose gel which is simulated for in vivo environment is the highest among the samples and does not change substantially in physiological environments. This differs largely from the behaviour of singly dispersed nanoparticles. Our results have paved the way for the design of MnFe2O4-loaded polymer nanospheres as magnetic hyperthermia agents for in vivo bio-applications.

Abstract:Nanoparticles possess unique properties, which can be applied in medical applications; they address targets such as cellular therapy, tissue repair, Nano-biosensors, drug delivery, magnetic resonance imaging and magnetic fluid hyperthermia. In this work, different sized cobalt ferrite nanoparticles (CFNP) were synthesized with narrow size distribution, by using chemical precipitation methods, to aim for finding the optimum particle size which has high heating efficiency in the applied magnetic field. The obtained powder was calcined at different temperature (600°C, 800°C, 900°C, and 1000°C). The sample which characterized by Transmission Electron Microscopy (TEM) confirmed the formation of single-phase CFNP in the range 10–115 nm depending on the annealing temperature and Vibrating Sample Magnetometer (VSM) to get the magnetization and coercivity of the powder. Localized magnetic particle hyperthermia treatment using ferrimagnetic nanoparticles continue to be an active area of medical application. So, homemade induction heater was designed. The induction heater was designed to be capable of generating high frequency, strong alternative magnetic fields (8 kA·m–1, 135 kHz). In vitro heating efficiencies in test tube, at a MNPs concentration of 250 mg CFNP·ml-1, were measured in the applied field. The temperature increase (ΔT) of the tube content at 60 s was 29.9°C for MNPs of 18 nm, 26.7°C for 25 nm, 25°C for 60 nm and 22.9°C for MNPs of 95 nm. The smallest nanoparticles (18 nm) exhibiting a high heating efficiency. In conclusion, we found that the size of the CFNP increased with increasing the calcined temperature at which the synthesis of the nanoparticles was performed. The heating efficiency of the particles was improved with decreasing particle size from 95 nm to 18 nm in the alternating magnetic field.
Synthesis, Surface Modification and Magnetic Properties Analysis of Heat-Generating Cobalt-Substituted Magnetite Nanoparticles

Abstract:Here, we present the results of the synthesis, surface modification, and properties analysis of magnetite-based nanoparticles, specifically Co0.047Fe2.953O4 (S1) and Co0.086Fe2.914O4 (S2). These nanoparticles were synthesized using the co-precipitation method at 80 °C for 2 h. They exhibit a single-phase nature and crystallize in a spinel-type structure (space group Fd3¯m). Transmission electron microscopy analysis reveals that the particles are quasi-spherical in shape and approximately 11 nm in size. An observed increase in saturation magnetization, coercivity, remanence, and blocking temperature in S2 compared to S1 can be attributed to an increase in magnetocrystalline anisotropy due to the incorporation of Co ions in the crystal lattice of the parent compound (Fe3O4). The heating efficiency of the samples was determined by fitting the Box-Lucas equation to the acquired temperature curves. The calculated Specific Loss Power (SLP) values were 46 W/g and 23 W/g (under HAC = 200 Oe and f = 252 kHz) for S1 and S2, respectively. Additionally, sample S1 was coated with citric acid (Co0.047Fe2.953O4@CA) and poly(acrylic acid) (Co0.047Fe2.953O4@PAA) to obtain stable colloids for further tests for magnetic hyperthermia applications in cancer therapy. Fits of the Box-Lucas equation provided SLP values of 21 W/g and 34 W/g for CA- and PAA-coated samples, respectively. On the other hand, X-ray photoelectron spectroscopy analysis points to the catalytically active centers Fe2+/Fe3+ and Co2+/Co3+ on the particle surface, suggesting possible applications of the samples as heterogeneous self-heating catalysts in advanced oxidation processes under an AC magnetic field.

Abstract:Magnetic hyperthermia is a technique that describes the heating of material through an external magnetic field. Classic hyperthermia is a medical condition where the human body overheats, being usually triggered by a heat stroke, which can lead to severe damage to organs and tissue due to the denaturation of cells. In modern medicine, hyperthermia can be deliberately induced to specified parts of the body to destroy malignant cells. Magnetic hyperthermia describes the way that this overheating is induced and it has the inherent advantage of being a minimal invasive method when compared to traditional surgery methods. This work presents a particle system that offers huge potential for hyperthermia treatments, given its good loss value, i.e., the particles dissipate a lot of heat to their surroundings when treated with an ac magnetic field. The measurements were performed in a low-cost custom hyperthermia setup. Additional toxicity assessments on Jurkat cells show a very low short-term toxicity on the particles and a moderate low toxicity after two days due to the prevalent health concerns towards nanoparticles in organisms.

Abstract:Using magnetic nanoparticles for extracorporeal magnetic heating applications in bio-medical technology allows higher external field amplitudes and thereby the utilization of particles with higher coercivities (HC). In this study, we report the synthesis and characterization of high coercivity cobalt ferrite nanoparticles following a wet co-precipitation method. Particles are characterized with magnetometry, X-ray diffraction, Mössbauer spectroscopy, transmission electron microscopy (TEM) and calorimetric measurements for the determination of their specific absorption rate (SAR). In the first series, CoxFe3−xO4 particles were synthesized with x = 1 and a structured variation of synthesis conditions, including those of the used atmosphere (O2 or N2). In the second series, particles with x = 0 to 1 were synthesized to study the influence of the cobalt fraction on the resulting magnetic and structural properties. Crystallite sizes of the resulting particles ranged between 10 and 18 nm, while maximum coercivities at room temperatures of 60 kA/m for synthesis with O2 and 37 kA/m for N2 were reached. Magnetization values at room temperature and 2 T (MRT,2T) up to 60 Am2/kg under N2 for x = 1 can be achieved. Synthesis parameters that lead to the formation of an additional phase when they exceed specific thresholds have been identified. Based on XRD findings, the direct correlation between high-field magnetization, the fraction of this antiferromagnetic byphase and the estimated transition temperature of this byphase, extracted from the Mössbauer spectroscopy series, we were able to attribute this contribution to akageneite. When varying the cobalt fraction x, a non-monotonous correlation of HC and x was found, with a linear increase of HC up to x = 0.8 and a decrease for x > 0.8, while magnetometry and in-field Mössbauer experiments demonstrated a moderate degree of spin canting for all x, yielding high magnetization. SAR values up to 480 W/g (@290 kHz, 69 mT) were measured for immobilized particles with x = 0.3, whit the external field amplitude being the limiting factor due to the high coercivities of our particles.

Abstract:In this study, Mg 1 −x Co x Fe 2 O 4 (0 ≤ x ≤ 1 with ∆x = 0.1) or MCFO nanoparticles were synthesized using a chemical co-precipitation method and annealed at 200, 400, 600, and 800°C respectively to investigate the structural properties of the materials by X-ray diffractometer (XRD), transmission electron microscopy (TEM), and Fourier-transform infrared spectroscopy (FTIR). Controlled annealing increased particle size for each value of x. The aim was to investigate how specific loss power (SLP) and maximum temperature (T max ) during local magnetic hyperthermia were affected by structural alterations associated with particle size and composition. The lattice parameter, X-ray density, ionic radius, hopping length, bond length, cation-cation distance, and cation-anion distance increase with an increase in Co 2+ content. Raman and FTIR spectroscopy reveal changes in cation distribution with Co 2+ content and particle size. Magnetic properties measured by the physical property measurement system (PPMS) showed saturation magnetization (M s ), coercivity (H c ), remanent magnetization (M r /M s ), and anisotropy constant (K 1 ) of the Mg 1 −x Co x Fe 2 O 4 nanoparticles increase with Co 2+ content and particle size. When exposed to an rf magnetic field, the nanohybrids experienced an increase in both the SLP (specific loss power) and T max (maximum temperature) as the particle size initially increased. However, these values reached their peak at critical particle size and subsequently decreased. This occurs since a modest increase in anisotropy, resulting from the presence of Co 2+ and larger particle size, facilitates Néel and Brownian relaxation. However, for high anisotropy values and particle size, the Néel and Brownian relaxations are hindered, leading to the emergence of a critical size. The critical size increases as the Co 2+ content decreases, but it decreases as the Co 2+ content increases, a consequence of higher anisotropy with the increase in Co 2+ . Additionally, it is noteworthy that the maximum temperature (T max ) rises as the concentration of nanohybrids grows, but the specific loss power (SLP) decreases. An increased concentration of chitosan-MCFO nanohybrids inhibits both the Néel and Brownian relaxation processes, reducing specific loss power.

Abstract:Superparamagnetic iron oxide nanoparticles (SPIONs) have found applications in the magnetic fluid hyperthermia (MFH) due to their unique magnetic properties, chemical stability and biocompatibility. However, challenges exist in attaining high heating efficiencies of the SPIONs under the applied alternating magnetic fields below Hergt's biological safety limit. Here, we present synthesis of single surfactant (pyromellitic acid (PMA)/2-aminoterephthalic acid (ATA)) and dual surfactants (PMA-ATA) coated SPIONs via chemical co-precipitation method and characterization to determine their phase purity, surface coatings and particle sizes. The hydrodynamic sizes/zeta potentials values of the SPIONs were determined for studying their water-dispersibility. Finally, the impact of heating on specific absorption rate (SAR) and intrinsic loss of power (ILP) were determined. SPIONs were found to exhibit magnetite phase and particle sizes in the range of 9-10 nm, good water dispersibility with (i) hydrodynamic diameters ranging from 190-304 nm and (ii) zeta potentials ranging from -38 mV to -49 mV. The ATA and PMA-ATA coated SPIONs showed better time-dependent temperature rise that resulted in higher heating efficacies-i.e., SAR and ILP values ranging from 58.6-79.5 W/gFe and 1.7-2.3 nHm²/Kg, respectively as compared to the PMA coated SPIONs. Thus, ATA and PMA-ATA coated SPIONs were found to be very promising candidates for their usage in MFH applications.

Abstract:Magnetic hyperthermia is an effective modality for treating cancerous cells selectively at the intracellular level via proper administration, surface tagging and appropriate choice of magnetic nanoparticle. Also, achieving the therapeutic temperature with as much low concentration of nanoparticles, as possible, is desirable. Recently, ferromagnetic nanoparticles or ferrofluid are gaining attention as effective magnetic hyperthermia agents due to their enhanced heating potential under in vivo conditions. In the present work, we have fabricated citrate capped cobalt ferrite ferrofluid via a simple two-step process and investigated the physicochemical, magnetic and induction heating performance. The average size of the synthesized nanoparticle is 9 ± 4.3 nm which is suitable for intracellular hyperthermia and has the advantage that it can be easily eliminated from the body through renal clearance. The M ( T ) measurement of the ferrofluid confirmed the soft ferromagnetic behavior. The induction heating potential of the ferrofluid prepared was tested at different concentrations (1, 2, 5 & 10 mg/ml), suspension medium and alternating magnetic fields (15, 30 & 45 kA/m) with a fixed frequency of 316 kHz. Under a viscous environment, 1 mg/ml sample subjected to 45 kA/m reached the hyperthermia temperature within 160 s and the specific loss power was found to be 297 W/g (Co+Fe) . Hence, the synthesized ferrofluid is suitable for intratumoural hyperthermia.

Abstract:Cobalt ferrite magnetic nanoparticles were synthesized through a hydrothermal route at various reaction temperatures: 100 °C, 130 °C, 160 °C, and 190 °C in order to study their hyperthermia potential. The heating properties of these samples were investigated by measuring time-dependent temperature curves in an external magnetic field (200 kHz, 100 Oe). Magnetic properties validated by cation distribution through octahedral and tetrahedral sites of the spinel structure done by the MAUD program. The results showed that temperature rising leads to migration of cobalt ions from octahedral to the tetrahedral site leading to change the reverse spinel structure to mixed structure. It was found that the nanoparticles synthesized at 160 °C reaction temperature had the maximum specific absorption rate (SAR) and intrinsic loss power parameter (ILP), as well as the highest saturation magnetization (Ms) and lowest coercivity (Hc) value.

Abstract:In this study, the effect of the shell thickness on the structural and magnetic properties of the CoFe2O4/NiFe2O4 core/shell is studied. A single-phase core/shell nanocomposite was prepared by the hydrothermal method. The shell thickness was found to control the magnitudes of saturation magnetization and coercive field of the prepared samples. The thickness of the NiFe2O4, which covered cubic CoFe2O4 particles of 15 nm, was 1.8 nm, leading to an increase in the saturation magnetization by 26% and a decrease in the coercive field by 50% compared to bare CoFe2O4. However, a further increase in shell thickness caused interfacial dislocations due to the lattice mismatch between the core and the shell. Finally, the heating ability at high frequency was measured for all samples. Comparing the temperature rise under the influence of AC magnetic field, which indicates power loss, relative to bare CoFe2O4, it was enhanced by 100% for a shell thickness of 26 nm. The results of this study point to potential applications for these samples in the field of magnetic hyperthermia for cancer therapy and drug delivery.

Abstract:AC induction heating properties of colloidal nano-aggregates of ferrimagnetic cobalt ferrite magnetic nanoparticles (MNPs) are reported in this study. Bio-compatible chitosan polymer-coated CoFe2O4 MNPs are synthesized using a co-precipitation method. Powder X-ray diffraction indicates the formation of mixed spinel structures for the uncoated (CP) and chitosan-coated (CP–CHN) MNPs, which is also supported by the cation distributions obtained from the Mössbauer spectra. The presence of chitosan coating on the surface of the CP-CHN MNPs is confirmed using X-ray photoelectron and Fourier transform infrared spectroscopy studies. Transmission electron microscopy shows primary particle sizes of ∼13 nm, which is larger than the superparamagnetic size limit of the CoFe2O4 MNPs. Hence, the CP and CP-CHN MNPs exhibit ferrimagnetic behaviour at room temperature with estimated saturation magnetization values of ∼77.4 emu/g and ∼74.4 emu/g, respectively. The average hydrodynamic diameter is found to be ∼90 ± 8 nm for an aqueous dispersion of the CP-CHN MNPs, which indicate the formation of colloidal nano-aggregates due to the ferrimagnetic interaction of the primary MNPs. The CP-CHN sample exhibits a significantly high AC induction heating efficiency of ∼267.2 ± 4.0 W/gFe, where the higher heating efficiency is attributed to the combination of hysteresis and relaxation-mediated magneto-thermal energy conversion, as confirmed using Stoner-Wohlfarth model-based dynamic hysteresis loop calculations. Further, the heating efficiency decreases with increasing sample concentration due to an increase in dipolar interaction, which is confirmed using semi-empirical calculations, where a lowering of the initial susceptibility is observed at higher concentrations. The higher AC induction heating efficiency, coupled with the demonstrated significant bio-compatibility during in vitro cytotoxicity studies, make the cobalt ferrite nano-aggregates potential candidates for magnetic hyperthermia.

Abstract:The adoption of magnetic hyperthermia as either a stand-alone or adjunct therapy for cancer is still far from being optimised due to the variable performance found in many iron oxide nanoparticle systems, including commercially available formulations. Herein, we present a reproducible and potentially scalable microwave-based method to make stable citric acid coated multi-core iron oxide nanoparticles, with exceptional magnetic heating parameters, viz. intrinsic loss parameters (ILPs) of up to 4.1 nH m(2) kg(-1), 35% better than the best commercial equivalents. We also probe the core-to-core magnetic interactions in the particles via remanence-derived Henkel and ΔM plots. These reveal a monotonic dependence of the ILP on the magnetic interaction field Hint, and show that the interactions are demagnetising in nature, and act to hinder the magnetic heating mechanism.

Abstract:Monodispersed Mn0.5Zn0.5Fe2O4 (~ 14 nm) nanoparticles were prepared by using a chemical co-precipitation technique. The direct mixing method has been used to prepare ferrofluid (base fluid-water) and to investigate their properties for the application of magnetic hyperthermia. Phase identification, crystal structure, and average crystallite size of the prepared samples were revealed in the XRD analysis. It confirms the Fd3m space group cubic spinel structure and average crystallite size ~ 14 nm. From TEM evaluation, the average particle size was found at ~ 16 nm. The 41.12 emu/g magnetization value of the Manganese Zinc Ferrite (MZF) nanoparticles (NPs) was found at room temperature. A negligible coercivity and remanence values confirm the superparamagnetic behavior of MZF nanoparticles at room temperature were confirmed by vibrating sample magnetometer (VSM) analysis. The induction heating analysis on water dispersed MZF Magnetic NPs at 4 kA/m AC magnetic field amplitude and 280 kHz frequency was conducted to use them in the application of hyperthermia. The result specifies that the heating potential of prepared ferrofluids can be reached within 65 s of hyperthermia temperature (42 °C) at lower 4 mg/mL content, indicating that the prepared material can be used as a heating agent for the treatment of magnetic hyperthermia. The specific absorption rate (SAR) was found to be 110.90 W/g. Thus, the obtained results suggest that the prepared MZF nanoparticles are a promising candidate for hyperthermia therapy due to their high heat-generating ability with less time at a lower concentration.
Physical and in vitro evaluation of ultra-fine cohenite particles for the prospective magnetic hyperthermia application

Abstract:We report here the structural and biocompatibility studies of bare nanoparticles of θ-Fe3C and its ferrofluid, prepared using pluronic acid F127 as a stabilizer. For both the cases, this carbide was compatible (~ 80% cell viability after 48 h) with A549 human lung carcinoma cells up to a concentration of 2 mg/mL which was comparable to that of its magnetic iron oxide counterparts. The X-ray diffraction and transmission electron microscopy validated its orthorhombic phase having an average particle size of ~ 6 nm. The surface property of the θ-Fe3C sample was analyzed by X-ray photoelectron spectroscopy (XPS), which designates the existence of only Fe and C. The Mössbauer spectroscopy for the sample also verified this carbide phase. The room temperature saturation magnetization for the sample at 2 T was around 78.2 Am2/kg. This value was more than that of magnetic iron oxide nanoparticles of similar size. Its ferrofluid displayed substantial temperature rise with time during the magnetic hyperthermia experiment. Consequently, the obtained specific loss power, as well as intrinsic loss power values, were 46 W/g and 0.526 nHm2/kg, respectively, at a field of 23 mT and 261 kHz. Both the values indicated its suitability for magnetic hyperthermia application.

Abstract:Monodispersed Fe3O4 magnetic nanoparticles (MNPs) having size of 7 nm have been prepared from iron oleate and made water dispersible by functionalization for biomedical applications. Three different reactions employing thioglycolic acid, aspartic acid and aminophosphonate were performed on oleic acid coated Fe3O4. In order to achieve a control on particle size, the pristine nanoparticles were heated in presence of ferric oleate which led to increase in size from 7 to 11 nm. Reaction parameters such as rate of heating, reaction temperature and duration of heating have been studied. Shape of particles was found to change from spherical to cuboid. The cuboid shape in turn enhances magneto-crystalline anisotropy (Ku). Heating efficacy of these nanoparticles for hyperthermia was also evaluated for different shapes and sizes. We demonstrate heat generation from these MNPs for hyperthermia application under alternating current (AC) magnetic field and optimized heating efficiency by controlling morphology of particles. We have also studied intra-cellular uptake and localization of nanoparticles and cytotoxicity under AC magnetic field in human breast carcinoma cell line.

Abstract:Magnetic particle hyperthermia, in which colloidal nanostructures are exposed to an alternating magnetic field, is a promising approach to cancer therapy. Unfortunately, the clinical efficacy of hyperthermia has not yet been optimized. Consequently, routes to improve magnetic particle hyperthermia such as designing hybrid structures comprised from different phase materials are actively pursued. Here we demonstrate enhanced hyperthermia efficiency in relative large spherical Fe/Fe-oxide core/shell nanoparticles through the manipulation of interactions between the core and shell phases. Experimental results on exemplary samples with diameters in the range 30-80 nm indicated a direct correlation of hysteresis losses to the observed temperature elevation rate with a maximum efficiency of around 0.9 kW/g. The absolute particle size, the core/shell ratio, and the interposition of a thin w\"ustite interlayer, are shown to have powerful effects on the specific absorption rate. By comparing our measurements to micromagnetic calculations we have unveiled topologically non-trivial magnetisation reversal modes under which interparticle interactions become negligible, aggregates formation is minimized, and the energy that is converted into heat is increased. This information has been overlooked till date and is in stark contrast to the existing knowledge on homogeneous particles.

Abstract:Cells of the obligate halophile Halobacterium cutirubrum were grown to the stationary phase in the presence of 32P-orthophosphate, 35S-sulfate, or 14C-labelled precursors (acetate, malonate, mevalonate, glycerol). 32P was incorporated into the phosphatide components in proportion to their relative concentrations, thus: phosphatidyl glycerophosphate (diether analogue) > unidentified minor phosphatide > phosphatidyl glycerol (diether analogue). 35S-Sulfate was incorporated mostly into the glycolipid sulfate ester component, but small amounts of 35S also appeared in a more polar sulfolipid and in a minor unidentified sulfolipid.14C-Mevalonate was the most efficient precursor for the phytanyl chains, followed in decreasing order by acetate and glycerol. Only traces of 14C from the malonate precursor were incorporated, and these appeared entirely in the phytanyl groups.
