Abstract:Objective To construct a prokaryotic expression system of gingipain R(Rgp) genes to identify the correlation between Porphyromonas gingivalis(Pg) gingipain R and chronic periodontitis.Methods The routing Phenol-chloroform method was applied to prepare genomic DNA of Pg ATCC33277.Oligonucleotide primers were designed to amplify the RgpAcat and RgpB genes according to the published corresponding nucleotide sequences.Prokaryotic expression system was established,and Ni-NTA affinity chroma-tography was applied to extract and purify the recombinant proteins.rRgpAcat and rRgpB were applied to human monocyte THP-1 cell strain.Flow-cytometric analyses were performed to detect the expression of CD-14 on the cell surface.Enzyme-linked immunosorbent assay( ELISA ) was used to detect the inhibition of lipopolysaccharides-induced IL-1β production from human monocytes by rRgpAcat or rRgpB.Results In comparison with the corresponding sequences from GeneBank,homologies of the nucleotide sequences of the cloned RgpAcat and RgpB genes were ＞ 97％.rRgpAcat and rRgpB output of the constructed prokaryotie expression system was approximate 50％ of the total bacterial proteins.The intensity of fluorescence of expression of CD-14 by THP-1 cells was 68.97.After rRgpAcat or rRgpB treatment with THP-1 cells for 0.5 h,it reduced to 45.30,46.47 (P ＜ 0.05 ),and the level of IL-1 β was also decreased (P ＜0.01 ).Conclusions Prokaryotic expression systems of RgpAcat and RgpB genes were successfully established in this study.rRgpAcat and rRgpB could cleave monocyte CD-14,block the THP-1 cells secreting cytokines,and as a consequence sustain chronic inflammation.
Correlation between levels of porphyromonas gingivalis outer membrane protein A in subgingival plaques of chronic periodontitis and periodontal lesions

Abstract:Objective To detect porphyromonas gingivalis outer membrane protein A(PgmA) in subgingival plaques from chronic periodontitis patients and to elucidate the correlation between PgmA levels and periodontal lesions.Methods SDS-PAGE was used to monitor the recombined PgmA(rPgmA) expression of prokaryotic expression system and the purity of rPgmA extracted by Ni-NTA affinity chromatography.Rabbits were immunized with rPgmA to obtain antiserum.ELISA method was established to examine PgmA in 209 subgingival plaques from 56 chronic periodontitis patients and to analyse the correlation between PgmA levels and periodontal lesions.Results rPgmA output was approximately 50% of the total bacterial proteins.The extrated rPgmA after SDS-PAGE only showed a single fragment.90.0% of subgingival plaques(188/209)were ELISA PgmA positive.The PgmA levels in the subgingival plaque samples from advanced periodontitis were significantly higher than those in the samples from mild periodontitis(P<0.05).No difference of the PgmA level was found between the samples from mild/moderate periodontitis and the samples from moderate/advanced periodontitis(P>0.05).Conclusion The lesions of chronic periodontitis were associated with the subgingival PgmA level.PgmA might play a role in the pathogenicity of porphyromonas gingivalis.

Abstract:To understand the correlation of the subgngival infection and co-inection with humancytomegalovirus(HCMV) and Epstein-Barr virus(EBV) in chronic periodontitis with the severity of the periodontal lesions,the glucoprotein gene(gB) of HCMV and the EBV nuclear antigen-2 gene(ebnA2) from 920 subgingival specimens taken from 130 cases of chronic periodontitis,65 cases of gingivitis and 35 healthy individuals without periodontal lesions were detected by means of nested PCR assay.The gingival index(GI) attachment loss(AL) and pocket depth(PD) were used as the observation indicators.It was found that the positive rates of HCMV,(63.5%),EBV-1(27.9%) and EBV-2(9.8%) infections in chronic periodontitis were obviously higher than those in gingivitis(49,2%,16.9% and 3.1%),as well as those of the healthy individuals without peroodontal lesions(40.0%,13.6% and 0.07%) respectively.However,no significant difference existed in the detection rate between the gingivitis samples and samples taken from healthy individuals.Also,no correlation could be found among the EBV-1 or EBV-2 infection and GI,AL and PD.Although the HCMV infection,co-infection with HCMV and EBV-1 or HCMV EBV-2 showed no relation to GI,but definite correlations could be demonstrated among infection or co-infection with AL and PD.All these results and statistical analysis lead to the conclusion that the infections with HCMC,EBV-1 or EBV-2 are involved in the development of chronic periodontitis in humans,and the co-infection with HCMV and EBV-1 can exacerbate the severity of disease.
Correlation Between Infection of Different Glycoprotein B Genotypes of Human Cytomegalovirus and Human Chronic Periodontitis

Abstract:OBJECTIVE:To investigate the correlation between infection of different human cytomegalovirus (HCMV) glycoprotein B (gB) genotypes and human chronic periodontitis. METHODS:A nested-polymerase chain reaction (nPCR) was employed to detect HCMV gB gene in the subgingival plaque samples from 65 chronic periodontitis patients and in the gingival crevicular fluid samples from 24 periodontally healthy control. The amplification fragments of gB gene were further genotyped by restriction fragment length polymorphism (RFLP). The correlation among infection with the different HCMV genotypes and the severity of periodontal lesion were evaluated. RESULTS:In terms of teeth examined, the prevalence of HCMV (59.23%, 154/260) in the chronic periodontitis lesions was significantly higher than that of HCMV (32.29%, 31/96) in the periodontally healthy control (P < 0.01). Of the HCMV DNA positive samples from the chronic periodontitis lesions, 11.7% (18/154) was genotyped as gB I, 80.5% (124/154) as gB II, and 7.8% (12/154) as gB I and gB II co-infection, and of the HCMV DNA positive samples from the periodontal healthy control, 45.2% (14/31) was genotyped as gB I, 38.7% (12/31) as gB II, and 16.1% (5/31) as gB I and gB II co-infection. The gB II genotype was more dominant among the chronic periodontitis lesions compared with that among the periodontally healthy control (P < 0.01). In chronic periodontitis, no statistical significance could be found between infection of different HCMV gB genotypes and the different clinical parameters of CAL, PD and GI (P > 0.05). CONCLUSIONS:Subgingival infection with HCMV is closely associated with chronic periodontitis. Infection of HCMV may not correlate directly with severity of periodontitis. However, gB II may be the dominant genotype of HCMV, which is associated with the chronic periodontitis.

Abstract:OBJECTIVE:To establish a 16S rDNA multiplex polymerase chain reaction (PCR) for simultaneously detecting P. gingivalis, A. actinomycetemcomitans and T. denticola in clinical specimens of chronic periodontitis and to study the correlation between different modes of infection and severity of the disease.METHODS:Periodontal pocket specimens from 152 patients with mild, moderate or advanced chronic periodontitis and gingival sulcus specimens from 30 periodontally healthy individuals were collected and placed in 200 microl lysis buffer. The specimens were incubated in 100 degrees C for 10 min and 10 microl of the supernatant was directly used as PCR template. DNAs from P. gingivalis strain ATCC33277, A. actinomycetemcomitans strain Y4, T. denticola strain FM and E. coli strain DH5alpha were used as positive and negative controls in PCR with all of which were prepared by routing phenol-chloroform method. A multiplex PCR assay, using three sets of primers specific to 16S rDNA genes of the three anaerobes, was developed to detect the specimens. The target amplification fragments from 3 cases of PCR products positive for all the three anaerobes were sequenced after T-A cloning. Chi-square test was applied to analyze the correlation between different coinfection of the three anaerobes and severity of the disease.RESULTS:The established 16S rDNA multiplex PCR assay was able to detect P. gingivalis, A. actinomycetemcomitans and T. denticola at a minimum of 10, 20 and 20 cells, respectively. In comparison with the reported corresponding sequences, similarities of the nucleotide sequences from the three anaerobes 16S rDNA amplification fragments were as high as 99.45%, 97.08% and 96.59%, respectively. Of the 30 periodontally healthy gingival sulcus specimens, only one (3.3%) positive for P. gingivalis and two (6.7%) for A. actinomycetemcomitans could be identified but all of the rest were negative. In the 152 CP periodontal pocket specimens, 147 cases (96.7%) were positive for P. gingivalis, A. actinomycetemcomitans and/or T. denticola, respectively, and 5 cases (3.3%) were negative for all the three anaerobes. The positive rate of P. gingivalis detection (91.5%, 139/152) was significantly higher than those of A. actinomycetemcomitans (72.4%, 110/152) and T. denticola (80.9%, 123/152) (chi(2) = 7.07, 18.67; P < 0.01). 89.8% of the specimens from patients showed coinfections with two (26.5%) or three anaerobes (63.3%), and the coinfection rates in the specimens from moderate and advanced CP were remarkably higher than that from mild CP (chi(2) = 10.43, P < 0.01).CONCLUSION:The 16S rDNA multiplex PCR established in this study showed high sensitivity and specificity, which could be used to detect P. gingivalis, A. actinomycetemcomitans and T. denticola in clinical specimens. CP was an disease caused by multiple pathogenic microbes while the synergistic pathopoiesis of the three microbes was closely related to the severity of the disease.
Correlation between the level of Porphyromonas gingivalis collagenase in subgingival plaques of chronic periodontitis and the periodontal lesions

Abstract:Objective To detect prtC gene and its expression product (PrtC) of P.gingivalis in subgingival plaques from chronic periodontitis patients and to elucidate the correlation between PrtC levels and periodontal lesions, and to identify the effect of PrtC on inducing host cell to secrete inflammatory cytokines. Methods SDS-PAGE was used to monitor the recombinant PrtC (rPrtC) expression of prokaryotic expression system and the purity of rPrtC extracted by Ni-NTA affinity chromatography. Rabbits were immunized with rPrtC to obtain antiserum. A multiple PCR was established to detect 16S rDNA and prtC genes of P.gingivalis in the subgingival plaque samples from 209 teeth sites of 56 chronic periodontitis patients. ELISA method was also established to examine PrtC in the samples and the correlation between the PrtC levels and periodontal lesions. The effects of rPrtC on inducing human umbilical vein endothelial cell line EVC-304 to secrete IL-1α, IL-8 and TNF-α. Results rPrtC output was approximate 50% of the total bacterial proteins. The extracted rPrtC after SDS-PAGE only showed a single fragment, 96.1% (201/209) and 92.3% (193/209) of the subgingival plaque samples were P.gingivalis 16S rDNA and prtC genes positive confirmed by PCR. 91.4% of the subgingival plaque samples (191/209) were PrtC ELISA positive. The PrtC levels in the subgingival plaque samples from advanced periodontitis were significantly higher than those in the samples from mild and moderate peridontitis (P<0.05). No difference of the PrtC level was found between the samples from mild/moderate peridontitis and the samples from moderate/advanced peridontitis (P>0.05). After co-incubation with 1 μg rPrtC for 24 h and with 5 or 10 μg rPrtC for 12 h, [CM(43]the levels of IL-1α, IL-8 and TNF-α secreted by the EVC-304 cells significantly increased (P<0.05). Conclusion[CM)] P.gingivalis has high carrying and expression frequencies of prtC gene. The lesions of chronic periodontitis are closely relative to the subgingival PrtC levels. rPrtC has the activity to directly induce host cells to synthesize and secrete IL-1α, IL-8 and TNF-α.

Abstract:Strains of two novel Corynebacterium species were cultured from samples of human nostrils and skin collected in the United States and Botswana. These strains demonstrated growth on Columbia Colistin-Nalidixic Acid agar with 5% sheep blood and in liquid media (brain heart infusion and tryptic soy broth) supplemented with Tween 80, a source of the fatty acid oleic acid. Cells were Gram-positive, non-spore-forming, non-motile bacilli that showed catalase but not oxidase activity. Major fatty acids in both of these species were 18:1 ω9c (oleic acid), 16:0 (palmitic acid), and 18:0 (stearic acid). Analysis of the 16S ribosomal RNA gene sequences identified these strains as belonging to the genus Corynebacterium (family Corynebacteriaceae). Whole-genome sequencing revealed that these strains formed distinct branches on a phylogenomic tree, with C. tuberculostearicum being the closest relative but with average nucleotide identities of < 95% relative to all previously described species. These results indicate that these strains represent novel species of Corynebacterium, for which we propose the names Corynebacterium hallux sp. nov., with the type strain CTNIH22T (=ATCC TSD-435T=DSM 117774T), and Corynebacterium nasorum sp. nov., with the type strain KPL3804T (=ATCC TSD-439T=DSM 117767T). We also describe the characteristics of two strains isolated from human nasal passages that are members of the recently named species Corynebacterium yonathiae.

Abstract:The phenomenon of enhanced flow of phosphosilicate glass (PSG) by ion implantation is reported here. By implanting As into PSG, excellent reflow has been obtained in inert ambients at temperatures as low as 750C. Sensitivity of reflow to percentage of oxygen (in nitrogen ambient) as well as to the energy of the As ions has been investigated. For the case of B, BF2, F, Ar, Se, and Sb implants, no similar reflow has been observed. CV and conduction experiments have shown the dielectric integrity to be unaffected by the implant. RBS depth profiling and resistivity measurements show As redistribution with no penetration to the substrate. This technology has been implemented into a VLSI process.
Relationship of serum and gingival crevicular fluid antibody levels with the amount of subgingival homologous bacterium in patients with periodontal disease]

Abstract:39 patients were selected in this study, including 12 patients with rapidly progressive periodontitis (RPP), 17 with adult periodontitis (AP) and 10 with marginal gingivitis (MG). The levels of antibody to Porphyromonas gingivalis (Pg) in serum (Sr) and in gingival crevicular fluid (GCF) from their 156 teeth were measured by ELISA. The Pg in subgingival plaque was detected by indirect immunofluorescence technique. Data showed different GCF/Sr ratio of anti--Pg antibody level between periodontitis patients (lower than 1) and MG patients (greater than 1). The amount of subgingival Pg was negatively correlated with clinical periodontal parameters, i.e, probing depth, attachment loss, plaque index and bleeding index. It was suggested that low GCF/Sr ratio of antibody level plus high counts of homologous bacterium might be used as an indicator of periodontal disease activity.

Abstract:Abstract The oral microbiome plays an important role in the human microbial community and in maintaining the health of an individual. Imbalances in the oral microbiome may contribute to oral and systemic diseases. The progression of periodontal disease is closely related to the growth of bacteria, such as Porphyromonas gingivalis , in the oral cavity. However, the pathogen growth mechanism specific to periodontal disease remains unknown. This study aimed to identify bacteria associated with periodontal health by focusing on hemolytic bacteria. Unstimulated saliva samples were collected from ten periodontitis patients and five healthy subjects to detect and identify the presence of hemolytic bacteria. The saliva of healthy subjects contained a higher proportion of G. haemolysans than saliva samples from patients with periodontitis. Growth inhibition assays indicated that the protein components contained in the culture supernatant of G. haemolysans directly suppressed the growth of P. gingivalis . This study shows that the presence of G. haemolysans in saliva is associated with periodontal health and that it inhibits the growth of P. gingivalis in vitro.

Abstract:In previous studies, it was demonstrated that Corynebacterium pseudodiphtheriticum 090104, isolated from the human nasopharynx, modulates respiratory immunity, improving protection against infections. Here, the antagonistic effect of the 090104 strain on respiratory pathogens, including Streptococcus pneumoniae, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Acinetobacter baumannii, was explored. In a series of in vitro studies, the capacity of C. pseudodiphtheriticum 090104, its bacterium-like particles, and its culture supernatants to coaggregate, inhibit the growth, and change the virulent phenotype of pathogenic bacteria was evaluated. The results showed that the 090104 strain was able to exert a bacteriostatic effect on K. pneumoniae and S. pneumoniae growth. In addition, C. pseudodiphtheriticum 090104 coaggregated, inhibited biofilm formation, and induced phenotypic changes in all the respiratory pathogens evaluated. In conclusion, this work demonstrated that, in addition to its beneficial effects exerted by host–microbe interactions, C. pseudodiphtheriticum 090104 can enhance protection against respiratory pathogens through its microbe–microbe interactions. The mechanisms involved in such interactions should be evaluated in future research.

Abstract:Aim: To investigate the antibacterial effects of Corydalis Saxicola bunting total alkaloid (CSBTA) on Porphyromonas gingivalis . Methods: SEM, chemical staining, RT-qPCR and ELISA were used to detect effects of CSBTA on P. gingivalis . Results: CSBTA treatment caused shrinkage and rupture of P. gingivalis morphology, decreased biofilm density and live bacteria in biofilm, as well as reduced mRNA expression of virulence genes hagA , hagB , kgp , rgpA and rgpB of P. gingivalis . Furthermore, NOK cells induced by CSBTA-treated P. gingivalis exhibited lower IL-6 and TNF-α expression levels. Conclusion: CSBTA is able to kill free P. gingivalis , disrupt the biofilm and weaken the pathogenicity of P. gingivalis . It has the potential to be developed as a drug against P. gingivalis infection.

Titer and Avidity of Serum Antibody to Porphyromonas Gingivalis Before and after Periodontal Therapy

Abstract:OBJECTIVE:To observe the changes of titer and avidity of serum antibody to Porphyromonas gingivalis (Pg) before and one month after periodontal therapy, and to observe the correlation of titer and avidity of anti-Pg IgG with clinical indices. METHODS:ELISA and DEA-ELISA were used to determine the titer and avidity of serum IgG antibody to Pg381 before and one month after periodontal treatment in 17 periodontitis patients and 6 healthy control subjects. RESULTS:The titers of anti-Pg IgG antibody were higher in periodontitis patients than in healthy control subjects (P < 0.001). All patients were seropositive (titers to Pg were higher than 2-fold above the median for control subjects). IgG antibody avidity to Pg in periodontitis patients were not found to be significantly lower than that in control group. The titers to Pg decreased significantly at one month after periodontal therapy, but antibody avidity didn't change. Serum anti-Pg antibody titers had a tendency of negative correlation with probing depth. The antibody titers were significant higher in periodontitis patients with < or = 5 mm mean probing depth than in patients with > 5 mm mean probing depth. CONCLUSIONS:It is suggested that the humoral immune response to Pg381 is protective and this protective effect mainly depends on the titer of antibody.
Prevalence of Actinomyces spp. in patients with chronic periodontitis

Abstract:This study investigated the prevalence of Actinomyces spp. in shallow, deep and very deep pockets of patients with chronic periodontitis compared to healthy controls and correlated the results with clinical status. Twenty patients with chronic periodontitis and 15 healthy subjects were enrolled in this study. Clinical indices were recorded in a six-point measurement per tooth. From each patient samples of supra and subgingival plaque were taken separately from teeth with shallow, deep and very deep pockets. Samples of supragingival plaque and sulcular microflora were collected from the healthy subjects. All the samples were cultivated on different media at 37̊C in an anaerobic atmosphere for 7 days. All the suspect colonies were identified using a rapid ID 32 A system (bioMèrieux) and MALDI-TOF-MS analysis using an Autoflex II Instrument (Bruker Daltonics) together with in house developed identification software and a reference spectra database. A total of 977 strains were identified as Actinomyces. Actinomyces naeslundii/oris/johnsonii (430 isolates) was the most prevalent species and was found in all patients and in almost all of the healthy subjects. Significant differences (p=0.003) between the groups were found for Actinomyces odontolyticus/meyeri and Actinomyces israelii which were associated with periodontitis patients. Actinomyces dentalis was found in higher percentage (p=0.015) in the periodontitis group. Actinomyces gerencseriae and Actinomyces massiliensis were significantly more often found supragingivally than subgingivally (p=0.004, p=0.022, respectively) in the periodontitis group. Whether some Actinomyces species, definitely important plaque formers, are actively involved in the pathogenicity of chronic periodontitis needs further investigation.
Characterization of a hyaluronic acid-based hydrogel containing an extracellular oxygen carrier (M101) for periodontitis treatment: An in vitro study

Abstract:Periodontitis is an inflammatory disease associated with anaerobic bacteria leading to the destruction of tooth-supporting tissues. Porphyromonas gingivalis is a keystone anaerobic pathogen involved in the development of severe lesions. Periodontal treatment aims to suppress subgingival biofilms and to restore tissue homeostasis. However, hypoxia impairs wound healing and promotes bacterial growth within periodontal pocket. This study aimed to evaluate the potential of local oxygen delivery through the local application of a hydrogel containing Arenicola marina's hemoglobin (M101). To this end, a hydrogel (xanthan (2%), hyaluronic acid (1%)) containing M101 (1-2 g/mL) (Xn(2%)-HA(1%)-M101) was prepared and characterized. Rheological tests revealed the occurrence of high deformation without the loss of elastic properties. Dialysis experiment revealed that incorporation of M101 within the gel did not modify its oxygen transportation properties. Samples of release media of the gels (1g/L (10%) and 2g/L (10%) M101) decreased significantly the growth of P.gingivalis after 24h validating its antibacterial effect. Metabolic activity measurement confirmed the cytocompatibility of Xn(2%)-HA(1%)-M101. This study suggests the therapeutic interest of Xn(2%)-HA(1%)-M101 gel to optimize treatment of periodontitis with a non-invasive approach.

Periodontol Pathogens in Coronary Atherosclerotic Plaques

Abstract:Objective To compare the detection rate of periodontol bacteria in coronary atherosclerotic plaques between severe periodontitis and mild periodontitis.Methods The coronary atherosclerotic plaques and subgingival plaques from 29 patients for coronary artery bypass surgery were collected,and the DNA from the bacteria of the plaques was extracted using the chelex-100 method.Three kinds of periodontal bacteria Porphyromonas gingivalis(P.g),Actinobacillus actinomycetemcomtans(A.a) and Tannerella forsythensis(T.f) were determined by PCR.There was 16 patients with severe periodontitis and 13 with mild periodontitis.Results In coronary atherosclerotic plaques,the detection rates of P.g,Aa and T.f were higher in severe periodontitis than in mild periodontitis(P0.05).The concordant presence of the same periodontal bacterium in subgingival plaques and coronary atherosclerotic plaques in the same patient was P.g: 5 cases in severe periodontitis,2 in mild periodontitis;A.a: 2 in severe periodontitis,O in mild periodontitis;T.f: 3 in severe periodontitis,1 in mild periodontitis.Conclusion The detection rate of periodontal bacteria in coronary atherosclerotic plaques in severe periodontitis is higher than that in mild periodontitis.

Abstract:OBJECTIVE:To detect periodontopathic bacterial DNA in atherosclerotic vascular lesions in a group of Chinese patients by using polymerase chain reaction (PCR) techniques.METHODS:Ten human specimens of atherosclerotic plaque were obtained sterilely. The sclerotic lesions were blade into fine pieces and DNA was isolated from the samples. To detect Porphyromonas gingivalis(Pg), Tannerella forsythia(Tf), Aggregatibacter actinomycetemcomitans(Aa), Prevotella intermedia(Pi), Prevotella nigrescens(Pn), Treponema denticola(Td), Campylobacter rectus(Cr), PCR amplification of bacterial 16S ribosomal DNA (rDNA) was carried out.RESULTS:PCR assays for bacterial 16S rDNA indicated the presence of periodontal pathogens in 3 out of 10 surgical specimens. DNA of Pg were found in 3 samples, Tf was found only in one among the 3 samples and Pn was found in another sample among the 3 samples.CONCLUSION:The data confirm that DNA of periodontal pathogens present in atherosclerotic plaques. Pg, Tf, Pn may play a role in the development and progression of atherosclerosis in these Chinese patients. Further studies with large size samples are needed.
