Abstract:Lighting is typically static for indoor production of leafy greens. However, temporal spectrum differentiation for distinct growth phases can potentially control age-specific desirable traits. Spectral effects can be persistent yet dynamic as plants mature, necessitating characterization of time-dependent responses. We grew red-leaf lettuce (Lactuca sativa L.) "Rouxai" in a growth room at 23°C and under a 20-h photoperiod created by warm-white (WW), blue (B; peak = 449 nm), green (G; peak = 526 nm), red (R; peak = 664 nm), and/or far-red (FR; peak = 733 nm) light-emitting diodes. From day 0 to 11, plants received six static lighting treatments with the same total photon flux density (400–800 nm): WW180, R180, B20R160, B20G60R100, B20R100FR60, or B180 (subscripts denote photon flux densities in μmol⋅m–2⋅s–1). On day 11, plants grown under each of the six treatments were transferred to all treatments, which created 36 temporal spectrum alternations. Plant growth, morphology, and coloration were measured on days 11 and 25. Increasing B radiation from 0 to 100% in static treatments decreased shoot fresh and dry weights and increased foliage redness of seedlings and mature plants. Compared to B20R160, B20R100FR60 increased shoot fresh weight, but not dry weight, on both days. However, other phenotypic responses under static treatments changed over time. For example, leaf length under B180 was 35% lower on day 11 but similar on day 25 compared to that under R180. In the B20 background, substituting G60 for R radiation did not influence shoot weight on day 11 but decreased it by 19% on day 25. When plants were switched from one treatment to another on day 11, the treatments applied before day 11 influenced final shoot weight and, to a lesser extent, leaf length and foliage coloration on day 25. In comparison, effects of the treatments applied after day 11 were more pronounced. We conclude some phenotypic responses to light quality depend on time and sequential light quality treatments had cumulative effects on lettuce growth. The temporal complexity of spectral responses is critical in photobiological research and creates opportunities for time-specific spectrum delivery to optimize crop characteristics.

Abstract:Chlorophyll content reflects plants' photosynthetic capacity, growth stage, and nitrogen status and is, therefore, of significant importance in precision agriculture. This study aims to develop a spectral and color vegetation indices-based model to estimate the chlorophyll content in aquaponically grown lettuce. A completely open-source automated machine learning (AutoML) framework (EvalML) was employed to develop the prediction models. The performance of AutoML along with four other standard machine learning models (back-propagation neural network (BPNN), partial least squares regression (PLSR), random forest (RF), and support vector machine (SVM) was compared. The most sensitive spectral (SVIs) and color vegetation indices (CVIs) for chlorophyll content were extracted and evaluated as reliable estimators of chlorophyll content. Using an ASD FieldSpec 4 Hi-Res spectroradiometer and a portable red, green, and blue (RGB) camera, 3600 hyperspectral reflectance measurements and 800 RGB images were acquired from lettuce grown across a gradient of nutrient levels. Ground measurements of leaf chlorophyll were acquired using an SPAD-502 m calibrated via laboratory chemical analyses. The results revealed a strong relationship between chlorophyll content and SPAD-502 readings, with an R2 of 0.95 and a correlation coefficient (r) of 0.975. The developed AutoML models outperformed all traditional models, yielding the highest values of the coefficient of determination in prediction (Rp2) for all vegetation indices (VIs). The combination of SVIs and CVIs achieved the best prediction accuracy with the highest Rp2 values ranging from 0.89 to 0.98, respectively. This study demonstrated the feasibility of spectral and color vegetation indices as estimators of chlorophyll content. Furthermore, the developed AutoML models can be integrated into embedded devices to control nutrient cycles in aquaponics systems.

Abstract:Leaf photosynthetic pigments play a crucial role in evaluating nutritional elements and physiological states. In facility agriculture, it is vital to rapidly and accurately obtain the pigment content and distribution of leaves to ensure precise water and fertilizer management. In our research, we utilized chlorophyll a (Chla), chlorophyll b (Chlb), total chlorophylls (Chls) and total carotenoids (Cars) as indicators to study the variations in the leaf positions of Lycopersicon esculentum Mill. Under 10 nitrogen concentration applications, a total of 2610 leaves (435 samples) were collected using visible-near infrared hyperspectral imaging (VNIR–HSI). In this study, a "coarse–fine" screening strategy was proposed using competitive adaptive reweighted sampling (CARS) and the iteratively retained informative variable (IRIV) algorithm to extract the characteristic wavelengths. Finally, simultaneous and quantitative models were established using partial least squares regression (PLSR). The CARS–IRIV–PLSR was used to create models to achieve a better prediction effect. The coefficient determination (R2), root mean square error (RMSE) and ratio performance deviation (RPD) were predicted to be 0.8240, 1.43 and 2.38 for Chla; 0.8391, 0.53 and 2.49 for Chlb; 0.7899, 2.24 and 2.18 for Chls; and 0.7577, 0.27 and 2.03 for Cars, respectively. The combination of these models with the pseudo-color image allowed for a visual inversion of the content and distribution of the pigment. These findings have important implications for guiding pigment distribution, nutrient diagnosis and fertilization decisions in plant growth management.

Abstract:Plant production in urban areas is receiving much attention due to its potential role in feeding the rapidly growing population of city dwellers. However, higher energy demands in urban plant factories are among the key challenges that need to be addressed. Artificial lighting is responsible for the most significant levels of energy consumption in plant factories; therefore, lighting systems must be modulated in consideration of the sustainable food–energy nexus. In this context, low light irradiation using blue (B) and red (R) LED was applied in a plant factory for the growth of red leaf lettuce (Lactuca sativa L. var Lollo rosso) to evaluate the growth performance and functional quality. The tested B (450 nm) and R (660 nm) light ratios were B/R = 5:1; 3:1; 1:1; 1:3, and 1:5, with a photosynthetic photon flux density (PPFD) of 90 ± 3 μmol m−2 s−1. In the plant factory, the photoperiod, temperature, RH, and CO2 conditions were 16 h d−1, 20 ± 0.5 °C, 65% ± 5%, and 360 ± 10 μL L−1, respectively. The lettuce was harvested 10 and 20 days after the commencement of LED light treatment (DAT). In this study, normal photosynthetic activity and good visual quality of the lettuce were observed. The results show that a higher fraction of R (B/R = 1:5) significantly increased plant growth parameters such as plant height, leaf area, specific leaf area, plant fresh and dry weight, and carbohydrate content. By contrast, a higher fraction of B (B/R = 5:1) significantly increased the photosynthetic parameters and contents of pigment and phenolic compounds. The rate of photosynthetic performance, carbohydrates (except starch), and content of phenolic compounds were highest after 10 DAT, whereas the pigment contents did not significantly differ at the different growth stages. It is concluded that high R fractions favor plant growth and carbohydrate content, while high B fractions favor photosynthetic performance and the accumulation of pigments and phenolic compounds in red leaf lettuce under limited lighting conditions. This study will help in designing artificial lighting conditions for plant factory production to reduce energy demands.
Regulation of the Photon Spectrum on Growth and Nutritional Attributes of Baby-Leaf Lettuce at Harvest and during Postharvest Storage

Abstract:The photon flux density (PFD) and spectrum regulate the growth, quality attributes, and postharvest physiology of leafy vegetables grown indoors. However, limited information is available on how a photon spectrum enriched with a broad range of different wavebands regulates these factors. To determine this, we grew baby-leaf lettuce ‘Rouxai’ under a PFD of 200 μmol m−2 s−1 provided by warm-white (WW; control) light-emitting diodes (LEDs) supplemented with either 30 μmol m−2 s−1 of ultraviolet-A (+UV30) or 50 μmol m−2 s−1 of blue (+B50), green (+G50), red (+R50), or WW (+WW50) light. We then quantified growth attributes and accumulated secondary metabolites at harvest and during storage in darkness at 5 °C. Additional +G50 light increased shoot fresh and dry weight by 53% and 59% compared to the control. Relative chlorophyll concentration increased under +UV30, +G50, and especially +B50. At harvest, +B50 increased total phenolic content (TPC) by 25% and anthocyanin content (TAC) by 2.0-fold. Additionally, +G50 increased antiradical activity (DPPH) by 29%. After each day of storage, TPC decreased by 2.9 to 7.1% and DPPH by 3.0 to 6.2%, while TAC degradation was less pronounced. Principal component analysis indicated a distinct effect of +G50 on the lettuce at harvest. However, concentrations of metabolites before and during storage were usually greatest under the +B50 and +R50 treatments.

Abstract:The growth and development of green lettuce plants can be modulated by the prevailing light conditions around them. The aim of this study was to evaluate the effect of ambient light enrichment with different LED light spectra on agronomic characteristics, polyphenol concentration and relative gene expression of enzymes associated with polyphenol formation in 'Levistro' lettuce grown hydroponically in a Nutrient Film Technique (NFT) system for 28 days in a greenhouse. The spectra (blue:green:red:far-red) and red:blue (R:B) ratios obtained by enriching ambient light with Blue (B), White (W), Blue-Red (BR) and Red (R) LED light were B: 47:22:21:10, 0.5:1; W: 30:38:23:9, 0.8:1; BR: 33:15:44:8, 1.3:1 and R: 16:16:60:8, 3.8:1, respectively, and photosynthetically active radiation (PAR) under the different treatments, measured at midday, ranged from 328 to 336 μmoles m−2 s−1. The resulting daily light integral (DLI) was between 9.1 and 9.6 mol m−2 day−1. The photoperiod for all enrichment treatments was 12 h of light. The control was ambient greenhouse light (25:30:30:15; R:B = 1.2:1; PAR = 702 μmoles m−2 s−1; DLI = 16.9 mol m−2 day−1; photoperiod = 14.2 h of light). Fresh weight (FW) and dried weight percentage (DWP) were similar among the enrichment treatments and the control. The leaf number increased significantly under BR and R compared to B lights. The relative index of chlorophyll concentration (RIC) increased as plants grew and was similar among the enrichment treatments and the control. On the other hand, the concentration of chlorogenic acid and chicoric acid increased under BR and B lights, which was consistent with the higher relative expression of the coumarate 3-hydroxylase enzyme gene. In view of the results, it is inferred that half of the PAR or DLI is sufficient to achieve normal growth and development of 'Levistro' lettuce plants, suggesting a more efficient use of light energy under the light enrichment treatments. On the other hand, the blue and combined blue-red lights promoted the accumulation of phenolic compounds in the leaves of 'Levistro' lettuce plants.

Abstract:Lettuce is a widely consumed leafy vegetable; it became popular due to its enhanced nutritional content. Recently, lettuce is also regarded as one of the model plants for vegetable production in plant factories. Light and nutrients are essential environmental factors that affect lettuce growth and morphology. To evaluate the impact of light spectra on lettuce, butter lettuce was grown under the light wavelengths of 460, 525, and 660 nm, along with white light as the control. Plant morphology, physiology, nutritional content, and transcriptomic analyses were performed to study the light response mechanisms. The results showed that the leaf fresh weight and length/width were higher when grown at 460 nm and lower when grown at 525 nm compared to the control treatment. When exposed to 460 nm light, the sugar, crude fiber, mineral, and vitamin concentrations were favorably altered; however, these levels decreased when exposed to light with a wavelength of 525 nm. The transcriptomic analysis showed that co-factor and vitamin metabolism- and secondary metabolism-related genes were specifically induced by 460 nm light exposure. Furthermore, the pathway enrichment analysis found that flavonoid biosynthesis- and vitamin B6 metabolism-related genes were significantly upregulated in response to 460 nm light exposure. Additional experiments demonstrated that the vitamin B6 and B2 content was significantly higher in leaves exposed to 460 nm light than those grown under the other conditions. Our findings suggested that the addition of 460 nm light could improve lettuce's biomass and nutritional value and help us to further understand how the light spectrum can be tuned as needed for lettuce production.

Abstract:Green light, as part of the photosynthetically active radiation, has been proven to have high photosynthetic efficiency once absorbed by plant leaves and can regulate plant physiological activities. However, few studies have investigated the appropriate and efficient way of using the green light for plant production. Thus, the objective of this study was to investigate a moderate amount of green light, partially replacing red and blue light, for plant growth and development. In this experiment, four treatments were set up by adjusting the relative amount of green light as 0 (RB), 30 (G30), 60 (G60), and 90 (G90) μmol m −2 s −1 , respectively, with a total photosynthetic photon flux density of 200 μmol m −2 s −1 and a fixed red-to-blue ratio of 4:1. Lettuce ( Lactuca sativa cv. ‘Tiberius’) plant growth and morphology, stomatal characteristics, light absorptance and transmittance, photosynthetic characteristics, and nutritional quality were investigated. The results showed that: (1) shoot dry weight increased by 16.3 and 24.5% and leaf area increased by 11.9 and 16.2% under G30 and G60, respectively, compared with those under RB. Plant stem length increased linearly with increasing green-to-blue light ratio; (2) light transmittance of lettuce leaf under treatments employing green light was higher than that under RB, especially in the green region; (3) stomatal density increased, whereas stomatal aperture area decreased with the increase in the relative amount of green light; and (4) carbohydrate accumulation increased under G60 and G90. Soluble sugar contents under G60 and G90 increased by 39.4 and 19.4%, respectively. Nitrate contents under G30, G60, and G90 decreased by 26.2, 40.3, and 43.4%, respectively. The above results indicated that 15–30% green light replacing red and blue light effectively increased the yield and nutritional quality of lettuce plants.

Abstract:Abstract LED lighting in indoor farming systems allows to modulate the spectrum to fit plant needs. Red (R) and blue (B) lights are often used, being highly active for photosynthesis. The effect of R and B spectral components on lettuce plant physiology and biochemistry and resource use efficiency were studied. Five red:blue (RB) ratios (0.5-1-2-3-4) supplied by LED and a fluorescent control (RB = 1) were tested in six experiments in controlled conditions (PPFD = 215 μmol m −2 s −1 , daylength 16 h). LED lighting increased yield (1.6 folds) and energy use efficiency (2.8 folds) as compared with fluorescent lamps. Adoption of RB = 3 maximised yield (by 2 folds as compared with RB = 0.5), also increasing leaf chlorophyll and flavonoids concentrations and the uptake of nitrogen, phosphorus, potassium and magnesium. As the red portion of the spectrum increased, photosystem II quantum efficiency decreased but transpiration decreased more rapidly, resulting in increased water use efficiency up to RB = 3 (75 g FW L −1 H 2 O). The transpiration decrease was accompanied by lower stomatal conductance, which was associated to lower stomatal density, despite an increased stomatal size. Both energy and land surface use efficiency were highest at RB ≥ 3. We hereby suggest a RB ratio of 3 for sustainable indoor lettuce cultivation.

Abstract:LED illumination can have a narrow spectral band; its intensity and time regime are regulated within a wide range. These characteristics are the potential basis for the use of a combination of LEDs for plant cultivation because light is the energy source that is used by plants as well as the regulator of photosynthesis, and the regulator of other physiological processes (e.g., plant development), and can cause plant damage under certain stress conditions. As a result, analyzing the influence of light spectra on physiological and growth characteristics during cultivation of different plant species is an important problem. In the present work, we investigated the influence of two variants of LED illumination (red light at an increased intensity, the “red” variant, and blue light at an increased intensity, the “blue” variant) on the parameters of photosynthetic dark and light reactions, respiration rate, leaf reflectance indices, and biomass, among other factors in lettuce (Lactuca sativa L.). The same light intensity (about 180 μmol m−2s−1) was used in both variants. It was shown that the blue illumination variant increased the dark respiration rate (35–130%) and cyclic electron flow around photosystem I (18–26% at the maximal intensity of the actinic light) in comparison to the red variant; the effects were dependent on the duration of cultivation. In contrast, the blue variant decreased the rate of the photosynthetic linear electron flow (13–26%) and various plant growth parameters, such as final biomass (about 40%). Some reflectance indices (e.g., the Zarco-Tejada and Miller Index, an index that is related to the core sizes and light-harvesting complex of photosystem I), were also strongly dependent on the illumination variant. Thus, our results show that the red illumination variant contributes a great deal to lettuce growth; in contrast, the blue variant contributes to stress changes, including the activation of cyclic electron flow around photosystem I.

Abstract:Understanding the responses of plant growth and secondary metabolite synthesis to different light wavelengths is important for optimizing lighting conditions for vegetable production in indoor vertical farms. Basil ( Ocimum basilicum ) ‘Improved Genovese Compact’ (green leaf) and ‘Red Rubin’ (purple leaf), green mustard ‘Amara’ ( Brassica carinata ), red mustard ‘Red Giant’ ( Brassica juncea ), green kale ‘Siberian’ ( Brassica napus var. pabularia ), and red kale ‘Scarlet’ ( Brassica oleracea ), which are high-value and multifunctional culinary herbs and leafy greens, were used to characterize the effects of red (R), blue (B), and green (G) wavelengths on plant photosynthesis, morphology, biomass production, and secondary metabolites accumulation. Light quality treatments consisted of three R and B light combinations, R 88 B 12 (the proportions of R and B wavelengths were 88% and 12%, respectively), R 76 B 24 , and R 51 B 49 , and two white light combinations, R 44 B 12 G 44 (the proportions of R, B, and G wavelengths were 44%, 12%, and 44%, respectively) and R 35 B 24 G 41 . Experiments were conducted in a walk-in growth room with a photosynthetic photon flux density set at 224 μmol·m −2 ·s −1 and a 16-hour photoperiod. Results indicated that the net photosynthesis in purple basil and green kale were positively correlated with B proportions (BP), and that higher BP increased the relative chlorophyll concentration in purple basil and red kale. In contrast, higher BP suppressed stem elongation and leaf expansion and reduced shoot biomass in all tested species except red mustard. Higher BP increased phytochemical concentrations but decreased the total amounts of phytochemicals per plant. For all basil and brassica ( Brassica sp.) cultivars, the inclusion of G wavelengths decreased shoot biomass compared with that of plants grown under R and B light combinations with similar BP. Inclusion of G wavelengths stimulated stem elongation in green basil and green mustard under 12% BP; whereas it suppressed stem elongation in purple basil, green kale, red kale, and green mustard under 24% BP. The effects on phytochemical accumulation were species-specific for the inclusion of G wavelengths. Considering biomass production, nutritional values, and working environment for growers, a white light with lower BP and G proportions is recommended for culinary herbs and Brassica leafy greens production at vertical farms.

Abstract:Light-emitting diodes (LEDs) are an artificial light source used in closed-type plant factories and provide a promising solution for a year-round supply of green leafy vegetables, such as lettuce (Lactuca sativa L.). Obtaining high-quality seedlings using controlled irradiation from LEDs is critical, as the seedling health affects the growth and yield of leaf lettuce after transplantation. Because key molecular pathways underlying plant responses to a specific light quality and intensity remain poorly characterised, we used a multi-omics–based approach to evaluate the metabolic and transcriptional reprogramming of leaf lettuce seedlings grown under narrow-band LED lighting. Four types of monochromatic LEDs (one blue, two green and one red) and white fluorescent light (control) were used at low and high intensities (100 and 300 μmol·m−2·s−1, respectively). Multi-platform mass spectrometry-based metabolomics and RNA-Seq were used to determine changes in the metabolome and transcriptome of lettuce plants in response to different light qualities and intensities. Metabolic pathway analysis revealed distinct regulatory mechanisms involved in flavonoid and phenylpropanoid biosynthetic pathways under blue and green wavelengths. Taken together, these data suggest that the energy transmitted by green light is effective in creating a balance between biomass production and the production of secondary metabolites involved in plant defence.

Abstract:There is a growing interest in the impact of light intensity and temperature on vegetable growth and dietary quality characteristics. Lettuce (Lactuca sativa L.), as one of the model vegetables in plant factories, was grown under two light levels (G1: 160 µmol m–2 s–1, G2: 200 µmol m–2 s–1) with different day/night temperatures (T1: 20 ± 1°C/15 ± 1°C, T2: 24 ± 1°C/19 ± 1°C) to investigate mutual effects of light intensity and temperature on its growth and quality properties. The results revealed that the T2G2 treatment showed higher plant height, biomass and net photosynthetic rate, while T1G2 resulted in decreased leaf area and lower maximum photochemical efficiency of photosystem II (Fv/Fm) compared to T1G1. Simultaneous increases in light intensity and temperature based on T1G1 significantly increased the contents of anthocyanin, vitamin C (Vc) and soluble protein by 58%, 21% and 172%, respectively. The higher light intensity promoted the nutritional values of lettuce, characterised by lower nitrate level and soluble protein contents. Principal component analysis showed a negative collinearity between the nitrate content and soluble protein and Vc contents. In conclusion, light intensity and temperature co-regulated the lettuce's growth and development, and their simultaneous increases better for growth and nutritional values of lettuce. These results provide a better understanding of the cultivation of plants in an artificial plant factory while maintaining yield and quality.
Blue and Far-Red Light Affect Area and Number of Individual Leaves to Influence Vegetative Growth and Pigment Synthesis in Lettuce

Abstract:Published work indicates that high percentage of blue light can enhance pigment levels but decreases growth, while addition of far-red light to growth light can increase quantum efficiency and photosynthesis in leafy greens. Combining high-energy blue light with low-energy far-red light may increase both vegetative growth and pigment levels. However, the effect of high-energy blue and low-energy far-red light on the vegetative growth and pigments synthesis is unclear. This information can be potentially useful for enhancing the levels of pigments with nutritional value (e.g., beta-carotene and anthocyanins) in the produce grown in vertical farms. We grew romaine lettuce (cv. Amadeus) under similar light intensity (approximately 130 μmol⋅m –2 ⋅s –1 ) but different proportions of red: blue: far-red including 90:10: 0 (“High-R”), 50: 50: 0 (“High-B”), and 42: 42: 16 (“High-B+FR”) for 31 days. Results indicated that canopy area and leaf photosynthetic rate of lettuce plants was reduced in the High-B, thereby reducing plant growth. We did not observe photosynthesis enhancement in the High-B+FR. Instead, plants clearly showed photomorphogenic effects. The phytochrome photostationary state (PSS) decreased with far-red addition, resulting in reduced leaf number per plant. This was likely to shift the allocation of resources toward elongation growth for shade avoidance. Further, we observed an increase in the area of individual leaves, canopy area, and shoot dry weight in the High-B+FR. However, these appear to be an indirect consequence of decreased leaf number per plant. Our results also indicate that changes in expansion growth at individual leaf scale largely regulated pigment concentration in plants. As individual leaf area became smaller (e.g., High-B) or larger (e.g., High-B+FR), the levels of pigments including chlorophylls and beta-carotene increased or decreased, respectively. Area of individual leaves also positively influenced canopy area (and likely light interception) and shoots dry weight (or vegetative growth). Our study provides additional insights into the effects of high-energy blue and low-energy far-red light on individual leaf number and leaf growth, which appear to control plant growth and pigment levels in lettuce.

Yield and Quality of Romaine Lettuce at Different Daily Light Integral in an Indoor Controlled Environment

Abstract:In this study, the effect of different photosynthetic photon flux density (PPFD) provided by LEDs (Light Emitting Diodes) and photoperiod on biomass production, morphological traits, photosynthetic performance, sensory attributes, and image texture parameters of indoor cultivated romaine lettuce was evaluated. Two cultivars of lettuce Lactuca sativa var. longifolium namely ‘Casual’ (Syngenta)—midi romaine lettuce with medium-compact heads—and ‘Elizium’ (Enza Zaden)—a mini type (Little Gem) with compact heavy heads—were used. PPFD of 160 and 240 μmol m−2 s−1 and photoperiod of 16 and 20 h were applied, and Daily Light Integral (DLI) values were 9.2, 11.5, 13.8, and 17.3 mol m−2 day−1. The experiment lasted 30 days in the Indoor Controlled Environment Agriculture facility. DLI equal to 17.3 mol m−2 per day for cv. ‘Casual’ and 11.5–17.3 mol m−2 per day for cv. ‘Elizium’ allowed to obtain a very high fresh weight, 350 and 240 g, respectively, within 30 days of cultivation in an indoor plant production facility. The application of the lowest PPFD 160 μmol m−2 s−1 and 16 h photoperiod (9.2 mol m−2 per day DLI) resulted in the lowest fresh weight, the number of leaves and head circumference. The level of nitrate, even at the lowest DLI, was below the limit imposed by European Community Regulation. The cv. ‘Elizium’ lettuce grown at PPFD 240 μmol m−2 s−1 and 16 h photoperiod had the highest overall sensory quality. The cv. ‘Casual’ lettuce grown at PPFD 160 μmol m−2 s−1 and 20 h photoperiod had the lowest sensory quality. The samples subjected to different photoperiod and PPFD were also successively distinguished in an objective and non-destructive way using image features and machine learning algorithms. The average accuracy for the leaf samples of cv. ‘Casual’ lettuce reached 98.75% and for cv. ‘Elizium’ cultivar—86.25%. The obtained relationship between DLI and yield, as well as the quality of romaine lettuce, can be used in practice to improve romaine lettuce production in an Indoor Controlled Environment.
Effects of UV-B and UV-C Spectrum Supplementation on the Antioxidant Properties and Photosynthetic Activity of Lettuce Cultivars

Abstract:Indoor farming systems enable plant production in precisely controlled environments. However, implementing stable growth conditions and the absence of stress stimulants can weaken plants' defense responses and limit the accumulation of bioactive, health-beneficial phytochemicals. A potential solution is the controlled application of stressors, such as supplemental ultraviolet (UV) light. To this end, we analyzed the efficiency of short-term pre-harvest supplementation of the red–green–blue (RGB, LED) spectrum with ultraviolet B (UV-B) or C (UV-C) light to boost phytochemical synthesis. Additionally, given the biological harm of UV radiation due to high-energy photons, we monitored plants' photosynthetic activity during treatment and their morphology as well as sensory attributes after the treatment. Our analyses showed that UV-B radiation did not negatively impact photosynthetic activity while significantly increasing the overall antioxidant potential of lettuce through enhanced levels of secondary metabolites (total phenolics, flavonoids, anthocyanins), carotenoids, and ascorbic acid. On the contrary, UV-C radiation-induced anthocyanin accumulation in the green leaf cultivar significantly harmed the photosynthetic apparatus and limited plant growth. Taken together, we showed that short-term UV-B light supplementation is an efficient method for lettuce biofortification with healthy phytochemicals, while UV-C treatment is not recommended due to the negative impact on the quality (morphology, sensory properties) of the obtained leafy products. These results are crucial for understanding the potential of UV light supplementation for producing functional plants.

Abstract:The spectrum and intensity of light play a significant role in the primary and secondary metabolism of plants. Low intensity can make the photosynthetic process less efficient, while inadequate spectrum can impair plant growth and quality. This study investigates the effect of different LED light spectra at low intensity on germination and growth of lettuce (Lactuca sativa L.) seedlings under a temperature-controlled chamber and the subsequent impact on mature plants grown in a greenhouse under natural light. The purpose was to reach a commercial plant seedling using a low amount of energy to achieve the yield potential in a shorter period. The experiment was carried out in three trials. In trial 1, the effect of different LED light wavelengths [100% blue (B); 100% red (R); mixed light 1 (52% blue, 27% green and 21% red) (BGR1), and mixed light 2 (29% blue, 53% green and 17% red and 1% far red) (BGR2)] at low intensity (55 μmol m–2 s–1 and 12 h light photoperiod) and darkness (control) on germination of two lettuce cultivars [‘Levistro’ (green) and ‘Carmolí’ (red)] was evaluated in a controlled temperature chamber (20±1.2°C). In trial 2, the effect of the same light conditions of the first experiment on agronomic characteristics and pigment contents of lettuce seedlings compared to the natural light (control: 451±66 μmol m–2 s–1) were evaluated. In trial 3, the seedlings developed under different LED light wavelengths were transplanted to evaluate the subsequent effect on the growth of baby lettuce cultivated hydroponically in the greenhouse under natural light. The results of this study show that red wavelength reduced germination percentage, while lights with a higher blue component (B and BGR1) accelerated germination and increased the number of germinated seeds in ‘Levistro’. Red also delayed germination and decreased the number of germinated seeds in ‘Carmolí’ compared to darkness. Seedlings of ‘Levistro’ had a higher fresh weight (FW) than ‘Carmolí’. In addition, FW increased under BGR2 and R, which coincided with the highest number of leaves and leaf length. Nevertheless, fresh weight was higher under BGR2 and B after transplanting, coinciding with the highest number of leaves. A higher blue component of the light (B and BGR1) increased the dry matter percentage (DMP) of seedlings, but there was no significant difference after transplanting. Chlorophyll (CHL) a and b content increased under BGR2; however, the highest CHL a/b ratio was observed under BGR1 in ‘Levistro’ and B in ‘Carmolí’, but it was higher after transplanting when seedlings were grown under B. The anthocyanin (ANT) content of ‘Carmolí’ seedlings was promoted by a higher blue component of the light (B and BGR1) but significantly increased under natural light (control) at the highest intensity. This work shows that varying the spectrum at low intensity can positively modify the growth and biochemical characteristics of lettuce seedlings, although the effect depends on the cultivar. This modification improves the performance of plants during greenhouse growth after transplanting, especially seedlings grown under B and BGR2. Highlights - Blue light enhanced germination and increased the number of germinated seeds of green lettuce. - High blue component lights improved the morphology, dry matter percentage, and chlorophyll a/b ratio of lettuce seedlings. - Blue and full-spectrum lights applied to lettuce seedlings affect fresh weight after transplanting. - The anthocyanin content of seedlings was stimulated by blue light at 55 μmol m–2 s–1, but even more so by PAR of natural light at 451 μmol m–2 s–1.

Space-qualified, abuttable packaging for LBNL p-channel CCDs, part II

Abstract:Fully depleted, back-illuminated, p-channel CCDs developed at Lawrence Berkeley National Laboratory exhibit high quantum efficiency in the near-infrared (700-1050nm), low fringing effects, low lateral charge diffusion (and hence small, well-controlled point spread function), and high radiation tolerance. Building on previous efforts, we have developed techniques and hardware that have produced space-qualified 4-side abuttable, high-precision detector packages for 10.5μm pixel, 3.5k x 3.5k p-channel LBNL CCDs. These packages are built around a silicon carbide mounting pedestal, providing excellent rigidity, thermal stability, and heat transfer. Precision fixturing produces packages with detector surface flatness better than 10μm P-V. These packages with active areas of 36.8mm square may be packed on a detector pitch as small as 44mm. LBNL-developed Front End Electronics (FEE) packages can mount directly to the detector packages within the same footprint and detector pitch. This combination, along with identically interfaced NIR detector/FEE packages offers excellent opportunities for high density, high pixel count focal planes for space-based, ground-based, and airborne astronomy.
The Effect of LED Light Spectra on the Growth, Yield and Nutritional Value of Red and Green Lettuce (Lactuca sativa)

Abstract:Controlled Environment Agriculture (CEA) is a method of increasing crop productivity per unit area of cultivated land by extending crop production into the vertical dimension and enabling year-round production. Light emitting diodes (LED) are frequently used as the source of light energy in CEA systems and light is commonly the limiting factor for production under CEA conditions. In the current study, the impact of different spectra was compared with the use of white LED light. The various spectra were white; white supplemented with ultraviolet b for a week before harvest; three combinations of red/blue lights (red 660 nm with blue 450 nm at 1:1 ratio; red 660 nm with blue 435 nm 1:1 ratio; red 660 nm with blue at mix of 450 nm and 435 nm 1:1 ratio); and red/blue supplemented with green and far red (B/R/G/FR, ratio: 1:1:0.07:0.64). The growth, yield, physiological and chemical profiles of two varieties of lettuce, Carmoli (red) and Locarno (green), responded differently to the various light treatments. However, white (control) appeared to perform the best overall. The B/R/G/FR promoted the growth and yield parameters in both varieties of lettuce but also increased the level of stem elongation (bolting), which impacted the quality of grown plants. There was no clear relationship between the various physiological parameters measured and final marketable yield in either variety. Various chemical traits, including vitamin C content, total phenol content, soluble sugar and total soluble solid contents responded differently to the light treatments, where each targeted chemical was promoted by a specific light spectrum. This highlights the importance of designing the light spectra in accordance with the intended outcomes. The current study has value in the field of commercial vertical farming of lettuce under CEA conditions.
