Abstract:This paper introduces Cellulose Paper Sorptive Extraction (CPSE) as a method to simplify the sample preparation process for the analysis of chlorpyrifos (CPF) in postmortem biological matrices (blood, urine and gastric lavage). The method involves adsorption of CPF onto unmodified cellulose paper from diluted samples followed by desorption into acetonitrile (ACN) for subsequent gas chromatography – mass spectrometry (GC-MS) analysis. The method's optimization explored various parameters affecting extraction efficiency, resulting in 1" × 1′′ filter papers and five pieces as optimal sizes and quantities, respectively. ACN emerged as the most effective elution solvent, and 10 min of elution time produced optimal results. Additionally, factors such as extraction time (60 min), agitation speed (200 rpm), and sample volume (10 mL) were optimized. The proposed method was validated according to the guidelines of SWGTOX and exhibited robust performance, with limit of detection (LOD) ranging from 0.027 to 0.030 μg mL −1 , and limit of quantification (LOQ) from 0.089 to 0.099 μg mL −1 . The method exhibited wide linearity ranging from 0.1 to 10 μg mL −1 , with correlation coefficients falling within the range of 0.9989–0.9995. The accuracy fell between 97 and 110%. Intra and inter-day precision were always less than 6 and 10%, respectively. The method exhibited very good enrichment factors (EF%) for CPF to be in the range of 65–70 for all tested matrices. Adhering to the concept of White Analytical Chemistry (WAC), CPSE-GC-MS method demonstrated a whiteness score of 98.0. The practicality of the method was evaluated using recently introduced Blue Applicability Grade Index (BAGI) and was deemed practical for routine forensic toxicology analysis of organophosphorus poisoning cases. The method was successfully applied for CPF analysis in two cases of death investigation due to its fatal intoxication.

Abstract:RATIONALE Maternal diet during pregnancy has the potential to affect airway development and to promote T-helper-2-cell responses during fetal life. This might increase the risk of developing childhood asthma or allergy. OBJECTIVES We investigated the influence of maternal food consumption during pregnancy on childhood asthma outcomes from 1 to 8 years of age. METHODS A birth cohort study consisting of a baseline of 4,146 pregnant women (1,327 atopic and 2,819 nonatopic). These women were asked about their frequency of consumption of fruit, vegetables, fish, egg, milk, milk products, nuts, and nut products during the last month. Their children were followed until 8 years of age. Longitudinal analyses were conducted to assess associations between maternal diet during pregnancy and childhood asthma outcomes over 8 years. MEASUREMENTS AND MAIN RESULTS Complete data were obtained for 2,832 children. There were no associations between maternal vegetable, fish, egg, milk or milk products, and nut consumption and longitudinal childhood outcomes. Daily consumption of nut products increased the risk of childhood wheeze (odds ratio [OR] daily versus rare consumption, 1.42; 95% confidence interval [95% CI], 1.06-1.89), dyspnea (OR, 1.58; 95% CI, 1.16-2.15), steroid use (OR, 1.62; 95% CI, 1.06-2.46), and asthma symptoms (OR, 1.47; 95% CI, 1.08-1.99). CONCLUSIONS Results of this study indicate an increased risk of daily versus rare consumption of nut products during pregnancy on childhood asthma outcomes. These findings need to be replicated by other studies before dietary advice can be given to pregnant women.

Abstract:In the present work, a disposable microextraction device with a polyamide 6 nano-fibrous supported liquid membrane (SLM) is employed for the pretreatment of minute volumes of biological fluids. The device is placed in a sample vial for an at-line coupling to a commercial capillary electrophoresis instrument with UV-Vis detection (CE-UV) and injections are performed fully automatically from the free acceptor solution above the SLM with no contact between the capillary and the membrane. Up to 4-fold enrichment of model basic (nortriptyline, haloperidol, loperamide, and papaverine) and acidic (ibuprofen, naproxen, ketoprofen, and diclofenac) drugs is achieved by optimizing the ratio of the donor to the acceptor solution volumes (16 to 4 μL, respectively). The actual setup enables SLM extractions from less than a drop of sample and is suitable for pretreatment of scarce human body fluids. Two unique methods are reported for efficient clean-up and enrichment of the basic and acidic drugs from capillary blood (formed as dried blood spot), serum, and urine samples, which enable their determination at therapeutic and/or toxic levels. The hyphenation of the SLM extraction with CE-UV analysis provides good repeatability (RSD, 2.4–14.9%), linearity (r2, 0.988–1.000), sensitivity (LOD, 0.017–0.22 mg L−1), and extraction recovery (ER, 20–106%) at short extraction times (10 min) and with minimum consumption of samples and reagents.

Abstract:The quantification and interpretation of drug concentrations in biological matrices to optimize pharmacotherapy and to perform the therapeutic drug monitoring (TDM) is particularly important for compounds with narrow therapeutic ranges, known to cause adverse effects. In these cases, the biomonitoring is essential to avoid the toxicity and side effects. In this study, an innovative Fabric Phase Sorptive Extraction (FPSE) followed by high performance liquid chromatography-photodiode array detection (FPSE–HPLC–PDA) method was optimized and validated for the extraction and quantitative evaluation of seven antidepressant drugs (ADs, venlafaxine, citalopram, paroxetine, fluoxetine, sertraline, amitriptyline, and clomipramine) in human whole blood, urine, and saliva samples.The best chromatographic separation was obtained using a reverse phase column and ammonium acetate (50 mM, pH 5.5) and acetonitrile (AcN) as mobile phases, with 0.3% of triethylamine (TEA) for the best peak shape. The used sample preparation technique, FPSE, developed in 2014, has offered numerous advantages such as low consumption of organic solvents, no sample pretreatment, and reduced overall sample preparation time. Among all tested membranes, sol-gel carbowax (CW 20 M) sorbent, coated on cellulose FPSE media, was the most efficient. The developed method provides satisfactory limit of detection of 0.06 μg/mL for all analytes except for venlafaxine that was 0.04 μg/mL. Both RSD% and BIAS% gave values below ±15%, according to current guidelines. Finally, real samples analyzes were carried out, comparing the obtained data with the anamnestic data of the subjects, confirmed the validity of the method.
Sustainable beeswax modified cellulose paper for the determination of tricyclic antidepressants in biofluids

Abstract:This article describes the synthesis of sorptive phases for bioanalysis based on the modification of cellulose paper with natural beeswax as sorbent, resulting in a substrate completely renewable and sustainable. The preparation of the sorptive phases consisted of the dissolution of beeswax in hexane, followed by its drop-casting on cellulose paper and subsequent evaporation of the solvent. The beeswax modification of paper renders it hydrophobic, enabling the extraction of the target analytes, i.e., imipramine, desipramine, amitriptyline and trimipramine, via hydrophobic interactions. The main variables affecting the extraction performance were investigated (e.g., pH, ionic strength, extraction time, eluent composition, agitation speed). The analytical workflow combines a straightforward sampling, simultaneous extraction of 30 samples in 1 h, and the rapid (<2 min) determination of the analytes via direct infusion mass spectrometry. The method provided limits of detection in the range 2.0 and 3.2 μg L −1 , and the precision, expressed as relative standard deviation, was better than 5.4 % and 8.5 % for intra and inter-day analyses, respectively. The accuracy, in terms of relative recovery, ranged from 90 % to 121 % using saliva as model biofluid.

A dual paper-based nucleic acid extraction method from blood in under ten minutes for point-of-care diagnostics

Abstract:Nucleic acid extraction (NAE) plays a crucial role for diagnostic testing procedures. For decades, dried blood spots (DBS) have been used for serology, drug monitoring, and molecular studies. However, extracting nucleic acids from DBS remains a significant challenge, especially when attempting to implement these applications to the point-of-care (POC). To address this issue, we have developed a paper-based NAE method using cellulose filter papers (DBSFP) that operates without the need for electricity (at room temperature). Our method allows for NAE in less than 7 min, and it involves grade 3 filter paper pre-treated with 8% (v/v) igepal surfactant, 1 min washing step with 1× PBS, and 5 min incubation at room temperature in 1× TE buffer. The performance of the methodology was assessed with loop-mediated isothermal amplification (LAMP), targeting the human reference gene beta-actin and the kelch 13 gene from P. falciparum . The developed method was evaluated against FTA cards and magnetic bead-based purification, using time-to-positive (min) for comparative analysis. Furthermore, we optimised our approach to take advantage of the dual functionality of the paper-based extraction, allowing for elution ( eluted disk ) as well as direct placement of the disk in the LAMP reaction ( in situ disk ). This flexibility extends to eukaryotic cells, bacterial cells, and viral particles. We successfully validated the method for RNA/DNA detection and demonstrated its compatibility with whole blood stored in anticoagulants. Additionally, we studied the compatibility of DBSFP with colorimetric and lateral flow detection, showcasing its potential for POC applications. Across various tested matrices, targets, and experimental conditions, our results were comparable to those obtained using gold standard methods, highlighting the versatility of our methodology. In summary, this manuscript presents a cost-effective solution for NAE from DBS, enabling molecular testing in virtually any POC setting. When combined with LAMP, our approach provides sample-to-result detection in under 35 minutes.

Abstract:Diabetes ketoacidosis (DKA) is a life-threatening complication and requires immediate medical attention in the case of diabetes subjects, especially in the case of type 1 diabetes mellitus. In the condition of DKA, the body produces an excess amount of ketone bodies after unregulated fat degradation, causing blood to become acidic and hampering the regular metabolic activities of the body. The current diagnostic technique for DKA condition is based on monitoring ketone bodies, especially β-hydroxybutyric acid, from human urine and blood samples. The detection of serum ketone bodies in pathology is sometimes limited due to false positive results and the lack of standardization for precise quantification of analytes. In this study, a paper-based patch operating on the thin film solid-phase microextraction (TF-SPME) principle was developed and it was coupled with gas chromatography-mass spectrometry for simple quantification of β-hydroxybutyric acid (BHB) ketone body from a phosphate-buffered saline matrix. To fabricate the paper-based TF-SPME patches, a regular A4 sheet paper sheet was utilized as the substrate and uniform coating by multiwalled carbon nanotubes (MWCNT), polydimethylsiloxane (PDMS) and divinyl benzene (DVB) compounds was performed with an automatic film applicator. The 70 μm paper-based coated sheet was trimmed into 4 cm × 1 cm dimension pieces to obtain multiple patches from a single sheet. Extraction of the BHB ketone body into the closed vials was performed by exploiting the individual DVB/PDMS and DVB/CNT/PDMS paper patches followed by desorption with acetonitrile before quantification by gas chromatography-mass spectrometry analysis. Our study showed that the BHB extraction efficiency of DVB/PDMS-coated patches was higher than that of DVB/CNT/PDMS. The outcome showed a good linearity ( R 2 = 0.99) within the 500–20 000 ng mL −1 concentration range of BHB by paper-based DVB/PDMS patches. This study demonstrated the feasibility of utilizing simple, cost-effective paper-based disposable TF-SPME patches as a sampling kit for future screening of diabetes ketoacidosis without the need for prolonged traditional sample preparation in pathology.

Redundancy of stomatal control for the circadian photosynthetic rhythm in Kalanchoë daigremontiana Hamet et Perrier.

Abstract:In continuous light, the Crassulacean acid metabolism plant Kalanchoe daigremontiana Hamet et Perrier has a circadian rhythm of gas exchange with peaks occurring during the subjective night. The rhythm of gas exchange is coupled to a weak, reverse phased rhythm of quantum yield of photosystem II (Phi (PSII)). To test if the rhythm of Phi (PSII) persists in the absence of stomatal control, leaves were coated with a thin layer of translucent silicone grease which prevented CO2 and H2O exchange. In spite of this treatment, the rhythm of Phi (PSII) occurred with close to normal phase timing and with a much larger amplitude than in uncoated leaves. The mechanism underlying the Phi (PSII) rhythm in coated leaves can be explained by a circadian activity of phosphoenolpyruvate carboxylase (PEPC). At peaks of PEPC activity, the small amount of CO2 contained in the coated leaf could have become depleted, preventing the carboxylase activity of Rubisco and causing decreases in electron transport rates (observed as deep troughs of Phi (PSII) at 23-h in LL and at ca. 24-h intervals afterwards). Peaks of Phi (PSII) would be caused by a downregulation of PEPC leading to improved supply of CO2 to Rubisco. Substrate limitation of photochemistry at 23 h (trough of Phi (PSII)) was also suggested by the weak response of ETR in coated leaves to stepwise light enhancement. These results show that photosynthetic rhythmicity in K. daigremontiana is independent of stomatal regulation and may originate in the mesophyll.
Humic acid–coated paper: An affordable mixed-mode weak-cation exchanger for isolating cocaine, codeine, and methadone from saliva samples for their direct infusion mass spectrometric analysis

Abstract:The use of natural materials as precursors for synthesizing sorptive phases can reduce the environmental impact of the sample preparation step. In this article, two natural materials, i.e., cellulose and humic acids, are combined, resulting in a porous and flat sorptive phase that combines hydrophobic and cation exchange interactions. The simple preparation, with low reagent consumption, and the high production capacity make this process affordable. The performance of the humic acid–coated paper has been studied towards three basic drugs (cocaine, codeine, and methadone), and it has allowed the determination of the targets in saliva by direct infusion tandem mass spectrometry. The simultaneous extraction of many samples and the rapid analysis (2 min) provide a high sample throughput. The resulting method has been validated following the International Conference on Harmonization (ICH) M10 validation guide. Working at the optimum conditions, the linear range was established from 5 to 500 μg·L −1 and the detection limits were established at 1.5 μg·L −1 for all the analytes. At the LOQ, the inter-day precision and intra-day precision, expressed as relative standard deviation, were better than 13.6% and 17.9%, respectively. For low/mid/high quality controls, the inter-day precision and intra-day precision were better than 11.8% and 9.1%, respectively. The accuracy, calculated as relative recovery, was in the range of 83.8–106.1%. Several samples from patients under treatment with codeine-containing prescriptions were finally analyzed.

A Simplified and Completely Automated Workflow for Regulated Lc-Ms/Ms Bioanalysis Using Cap-Piercing Direct Sampling and Evaporation-Free Solid Phase Extraction

Abstract:Automated sample extraction for regulated bioanalysis by liquid chromatography/tandem mass spectrometry (LC-MS/MS) still presents significant challenges. A new sample preparation methodology with a simplified and completely automated workflow was developed to overcome these challenges using cap piercing for direct biofluid transfer and evaporation-free solid phase extraction (SPE). Using pierceable cap sample tubes, a robotic liquid handler was able to sample withouf uncapping or recapping during sample preparation. Evaporation for SPE was eliminated by using a mobile phase-compatible elution solvent followed by sample dilution prior to LC-MS/MS analysis. Presented here are three LC-MS/MS assays validated using this methodology to support three CNS drug development programs: (1) BMS-763534 and its metabolite, BMS-790318, in dog plasma; (2) BMS-694153 in monkey plasma; and (3) Pexacerfont (BMS-562086) and two metabolites, BMS-749241 and DPH-123554, in human plasma. These assays were linear from 1.00 to 1000 or 2.00 to 2000 ng/mL for each analyte with excellent assay accuracy, precision and reproducibility. These assays met acceptance criteria for regulated bioanalysis and have been successfully applied to drug development study samples. The methodology described here successfully eliminated all manual intervention steps achieving fully automated sample preparation without compromising assay performance. Importantly, this methodology eliminates the potential exposure of the bioanalyst to any infectious biofluids during sample preparation. (C) 2013 Elsevier B.V. All rights reserved.
Novel capsule phase microextraction in combination with liquid chromatography-tandem mass spectrometry for determining personal care products in environmental water

Abstract:A novel sample preparation technique named capsule phase microextraction (CPME) is presented here. The technique utilizes a miniaturized microextraction capsule (MEC) as the extraction medium. The MEC consists of two conjoined porous tubular polypropylene membranes, one of which encapsulates the sorbent through sol-gel technology, while the other encapsulates a magnetic metal rod. As such, MEC integrates both the extraction and stirring mechanisms into a single device. The aim of this article is to demonstrate the application potential of CPME as sample preparation technique for the extraction of a group of personal care products (PCPs) from water matrices. Among the different sol-gel sorbent materials (UCON®, poly(caprolactone-dimethylsiloxane-caprolactone) (PCAP-DMS-CAP) and Carbowax 20M (CW-20M)) evaluated, CW-20M MEC demonstrated the best extraction performance for the selected PCPs. The extraction conditions for sol-gel CW-20M MEC were optimized, including sample pH, stirring speed, addition of salt, extraction time, sample volume, liquid desorption solvent, and time. Under the optimal conditions, sol-gel CW-20M MEC provided recoveries, ranging between 47 and 90% for all analytes, except for ethylparaben, which showed a recovery of 26%. The method based on CPME with sol-gel CW-20M followed by liquid chromatography-tandem mass spectrometry was developed and validated for the extraction of PCPs from river water and effluent wastewater samples. When analyzing different environmental samples, some analytes such as 2,4-dihydroxybenzophenone, 2,2-dihydroxy-4-4 methoxybenzophenone and 3-benzophenone were found at low ng L−1.
Inexpensive, effective novel activated carbon fibers for sample cleanup: application to multipesticide residue analysis in food commodities using a QuEChERS method

Abstract:Phenolic resin based activated carbon fibers (ACFs) were applied for the first time as a reversed-dispersive solid-phase extraction (r-DSPE) sorbent. A modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) method was applied to determine 26 pesticides (organophosphates, organochlorines, synthetic pyrethroids, and herbicides) in different complex matrices, including cauliflower, cucumber, banana, apple, wheat, and black gram. Different physicochemical characterization techniques were used to investigate the engineering and structural properties of the r-DSPE sorbent. All the chromatographic analyses were performed with a gas chromatograph equipped with an electron capture detector. The recoveries of all 26 pesticides were acceptable (70–120%), with relative standard deviations of less than 15%. The limit of detection and the limit of quantification were 1.13–5.48 ng/g and 3.42–16.60 ng/g, respectively. In the original QuEChERS method, primary secondary amine is extensively used as the r-DSPE sorbent in the cleanup process, but it is eightfold more expensive than the ACFs used in this study. Therefore, the modified QuEChERS method using ACFs during the cleanup process is more efficient, cheaper, and more robust to determine pesticides from different types of matrices, including vegetables, grains, and fruits, and ACFs could be used as a cost-effective alternative to primary secondary amine.Graphical AbstractSample clean-up using PSA and ACF as r-DSPE sorbent in QuEChERS method

Abstract:In recent years, using natural materials to prepare sorbents has been a clear trend in the sample preparation context. This research line is supported by sustainable considerations, as it responds to the tenth principle of Green Analytical Chemistry. Also, the affordability and availability of natural materials open the door to developing global analytical solutions. Among all the available natural materials, cellulose paper presents unique characteristics (porosity, mechanical resistance, and easy chemical/physical modification of the substrate) that can be exploited in analytical extractions. This article reviews the role of paper-based sorptive phases in the current sample preparation realm, putting more emphasis on their potential than on the description of specific applications. The article covers the main synthetic routes described in the literature for preparing these phases and the reported coatings. In contrast with the classical analytical workflow (based on the separated extraction, elution, and analysis steps), the direct combination of sample preparation with instrumental techniques permits further simplification of the analytical procedures. Cellulose paper can operate under both workflows, and the main approaches based on spectroscopic and spectrometric analysis will be discussed. Finally, the combination of both instrumental techniques (using the so-called dual substrates) will be outlined.

Abstract:In this study, a green and rapid method called deep eutectic solvent dispersed liquid–liquid microextraction with magnetic particles-assisted retrieval (DES-DLLME-MPAR) was proposed, which was applied for extracting antidepressants from biofluids as a proof-of-concept. Vortex-assisted DES extraction enabled rapid extraction and reusable magnetic particles was utilized to assist in the rapid retrieval of the DES. Ethanol, also low in toxicity, was used as the desorption solvent. Optimal conditions for the extraction were achieved as follows: 10 μL of DES composed of methyltrioctylammonium bromide and capryl alcohol in a molar ratio of 1:3 as the extraction solvent, a sampling solution pH of 10.0, a vortex time of 1 min for both extraction and desorption, 10 mg of magnetic particles for assisting DES retrieval, and a desorption solution of 100 μL ethanol. Under these conditions, the extraction recoveries were above 83.4 % and the whole extraction procedure could be completed in approximately 3 min. Linear calibration curves in the concentration range of 0.01–10.00 μg/mL were obtained for four antidepressants, with correlation coefficients exceeding 0.999. The limits of quantification ranged from 0.068 to 0.092 μg/mL, meeting the demand for therapeutic drug monitoring. Satisfied accuracies (relative recoveries: 93.8–111.6 %) and precisions (RSD ≤ 6.0 %) were achieved. The detection selectivity was enhanced by the efficient removal of the interferences. Three greenness evaluation metrics collectively demonstrate the excellent greenness of the developed method. The proposed DES-DLLME-MPAR method was rapid, green, simple, inexpensive and accurate, offering a promising alternative approach for the analysis of hydrophobic drugs in clinical biofluids.

Abstract:In this study, a combination of single-interface hollow fiber liquid-phase microextraction and electromembrane extraction was applied for the extraction of some basic drugs from biological samples. The extraction process was followed by the HPLC-UV instrument. In this dynamic system, 1-octanol was impregnated into the pores of hollow fiber wall as an acceptor phase, the sample solution (pH = 12) was pumped into the lumen of hollow fiber by a syringe pump, the extraction efficiency was improved by filling and emptying (25 times) of sample solution (step 1). The extracted analytes in the organic acceptor phase were back-extracted by a fast electromembrane extraction procedure (step 2). Effective parameters on the extraction efficiency of both methods were investigated and optimized. Under optimized conditions (organic acceptor phase: 1-octanol, sample solution volume: 1500 µL, flow rate: 2.0 mL min-1, the number of filling and emptying cycles: 25, voltage: 100 V, back-extraction time: 2 min, and aqueous acceptor phase: 100 mM HCl), the proposed method provided good linearity with determination coefficients ranging from 0.992 to 0.996 over a concentration range of 2.5-1000 ng mL-1. Extraction recoveries were obtained in the range of 65.1 and 88.8%, which resulted in preconcentration factors in the range of 32.6-44.4. The limits of detection were found to be within the range of 0.12-0.36 ng mL-1, while the corresponding repeatability ranged from 3.7 to 9.3% (n = 3). Finally, the optimized method was applied for the quantification of propranolol, diltiazem and, lidocaine in urine and, plasma samples with relative recoveries ranged between 94.1 and 105.4%, indicating the reliability of the method.

Abstract:Capsule phase microextraction (CPME) is an efficient bioanalytical technique that streamlines the sample preparation by integrating the filtration and stirring mechanism directly into the device. A novel composite sorbent designed to be selective towards the target analytes consisting of mixed-mode sorbent chemistry synthesized by sol–gel technology is found promising and superior to the conventional C 18 sorbents. Herein we describe the encapsulation of an ionic liquid (IL)/Carbowax 20M-functionalized sol–gel sorbent (sol–gel IL/Carbowax 20 M) in the lumen of porous polypropylene tubes for the capsule phase microextraction of three phosphodiesterase-5 inhibitors namely avanafil, sildenafil, and tadalafil in human serum and urine samples. The CPME device was characterized by Scanning Electron Microscopy (SEM) and Fourier-Transform Infrared Spectroscopy (FT-IR). The experimental parameters of CPME procedure (e.g. sample pH and ionic strength, extraction time, stirring rate, elution solvent and volume) were carefully optimized to achieve the highest possible extraction efficiency for the analytes. Method validation was conducted in terms of precision, linearity, accuracy, matrix effect, lower limits of quantification, and limits of detection (LOD). The method linearity was investigated in the range of 50–1000 ng mL −1 for all analytes while the precision was less than 11.8 % in all cases. For all analytes, the LOD values were 17 ng mL −1 . The IL/CW 20M-functionalized microextraction capsules could be reused at least 25 times both for urine and serum samples. The green character and the applicability of the proposed method were evaluated using the ComplexGAPI and BAGI indexes. The optimized CPME protocol exhibited reduced consumption of organic solvent and generation of waste, cost-effectiveness, and simplicity. Finally, the proposed method was successfully applied to the analysis of sildenafil in human urine after administration of drug-containing formulation.

Abstract:In this work, a novel layered sorbent for microextraction by packed sorbent (MEPS) was introduced, which has been prepared by coating graphene oxide/polyamide (GO/PA) nanocomposite (NC) onto cellulose paper through solvent exchange method. Scanning electron microscopy (SEM) was applied to investigate the surface characteristic and morphology of PA and GO/PA NC coated on cellulose paper. The prepared MEPS device was used for extraction of organophosphorous pesticides (OPPs) including chlorpyrifos, fenthion, fenithrothion, ethion, edifenphos and phosalone in environmental aqueous samples followed by detection using gas chromatography-flame ionisation detector (GC-FID). Important parameters affecting the MEPS method including pH of sample solution, extraction draw-discard cycles, sorbent layers, desorption solvent volume and desorption draw-eject number were studied and optimised using central composite design (CCD). Based on the method validation, limits of detection (LODs) were in the range of 0.2–1 µg L−1. The calibration graphs for chlorpyrifos, fenthion and edifenphos are linear in the concentration range of 1 to 500 µg L−1; for ethion and phosalone are linear in the range of 1–1000 µg L−1 and for fenithrothion is linear in the range of 3–1000 µg L−1. The method precision (RSD %) with six replicates determinations was in the range of 3 to 9.4 % and 3.9 to 11.9% for distilled water and spiked river water sample, respectively, at the concentration level of 300 µg L−1 . The developed method was applied successfully to determine OPP compounds in river, dam and tap water samples; accordingly, the relative recoveries (RR%) were obtained in the range of 77.8 to 113.3%.

Abstract:Common methodologies such as liquid-liquid extraction and solid-phase extraction are applied for the extraction of opioids from biological specimens i.e., blood and urine. Techniques including LC-MS/LC-MSMS, GC-MS, etc. are used for qualitative or quantitative determination of opioids. The goal of the present work is to design a green, economic, rugged, and simple extraction technique for famous opioids in human blood and urine and their simultaneous quantification by GC-MS equipped with an inert plus electron impact (EI) ionization source at SIM mode to produce reproducible and efficient results. Morphine, codeine, 6-acetylmorphine, nalbuphine, tramadol and dextromethorphan were selected as target opioids. Anhydrous Epsom salt was applied for dSPE of opioids from blood and urine into acetonitrile extraction solvent with the addition of sodium phosphate buffer (pH 6) and n-hexane was added to remove non-polar interfering species from samples. BSTFA was used as a derivatizing agent for GC-MS. Following method validation, the LOD/LLOQ and ULOQ were determined for morphine, codeine, nal-buphine, tramadol, and dextromethorphan at 10 ng/mL and 1500 ng/mL, respectively, while the LOD/LLOQ and ULOQ were determined for 6-acetylmorphine at 5 ng/mL and 150 ng/mL, respectively. This method was applied to real blood and urine samples of opioid abusers and the results were found to be reproducible with true quantification.
