Abstract:To reduce cadmium (Cd) pollution of food chains, screening and breeding of low-Cd-accumulating genotypes have received increasing attention. However, the mechanisms involving Cd tolerance and accumulation are not fully understood. Here, we investigated the physiological responses and metabolomics profiling on two wheat (Triticum aestivum L.) genotypes, a low-Cd-accumulating genotype in grains (Aikang58, AK58) and a high-Cd-accumulating genotype in grains (Zhenmai10, ZM10), in hydroponic culture treated without/with Cd for 7 days. The results showed that AK58 was a Cd tolerant genotype with higher capacity of antioxidant systems in root. In addition, the concentrations of Cd bound to root cell walls were higher in AK58 than ZM10, of which pectin and hemicellulose played important roles in Cd binding. Moreover, subcellular distribution manifested that Cd sequestrated in the vacuoles was another tolerance mechanism in AK58. Simultaneously, metabolomics profiling showed that, in AK58, phenylalanine metabolism, alanine, aspartate and glutamate metabolism, isoquinoline alkaloid biosynthesis, arginine and proline metabolism, arginine biosynthesis and glyoxylate and dicarboxylate metabolism are highly related to antioxidant defense system, cell wall biosynthesis and metabolisms of phytochelatins together with other organic ligands, playing crucial roles in Cd tolerance and Cd fixation mechanisms in roots. These novel findings should be useful for molecular assisted screening and breeding of low Cd-accumulating genotypes for wheat crop.

Abstract:Although several studies on the effects of cadmium (Cd) on wheat have been reported, the gene expression profiles of different wheat tissues in response to gradient concentrations of Cd, and whether soil microorganisms are involved in the damage to wheat remain to be discovered. To gain further insight into the molecular mechanisms of Cd-resistance in wheat, we sowed bread wheat (Triticum aestivum) in artificially Cd-contaminated soil and investigated the transcriptomic response of the wheat roots, stems, and leaves to gradient concentrations of Cd, as well as the alteration of the soil microbiome. Results indicated that the root bioaccumulation factors increased with Cd when concentrations were < 10 mg/kg, but at even higher concentrations, the bioaccumulation factors decreased, which is consistent with the overexpression of metal transporters and other genes related to Cd tolerance. In the Cd-contaminated soil, the abundance of fungal pathogens increased, and the antimicrobial response in wheat root was observed. Most of the differentially expressed genes (DEGs) of wheat changed significantly when the Cd concentration increased above 10 mg/kg, and the transcriptional response is much greater in roots than in stems and leaves. The DEGs are mainly involved in Cd transport and chelation, antioxidative stress, antimicrobial responses, and growth regulation. COPT3 and ZnT1 were identified for the first time as the major transporters responding to Cd in wheat. Overexpression of the nicotianamine synthase and pectinesterase genes suggested that nicotianamine and pectin are the key chelators in Cd detoxification. endochitinase, chitinase, and snakin2 were involved in the anti-fungal stress caused by Cd-induced cell damage. Several phytohormone-related DEGs are involved in the root’s growth and repair. Overall, this study presents the novel Cd tolerance mechanisms in wheat and the changes in soil fungal pathogens that increase plant damage.
Physio-Biochemical and Transcriptomic Features of Arbuscular Mycorrhizal Fungi Relieving Cadmium Stress in Wheat

Abstract:Arbuscular mycorrhizal fungi (AMF) can improve plant cadmium (Cd) tolerance, but the tolerance mechanism in wheat is not fully understood. This study aimed to examine the physiological properties and transcriptome changes in wheat inoculated with or without Glomus mosseae (GM) under Cd stress (0, 5, and 10 mg·kg-1 CdCl2) to understand its role in wheat Cd tolerance. The results showed that the Cd content in shoots decreased while the Cd accumulation in roots increased under AMF symbiosis compared to the non-inoculation group and that AMF significantly promoted the growth of wheat seedlings and reduced Cd-induced oxidative damage. This alleviative effect of AMF on wheat under Cd stress was mainly attributed to the fact that AMF accelerated the ascorbate-glutathione (AsA-GSH) cycle, promoted the production of GSH and metallothionein (MTs), improved the degradation of methylglyoxal (MG), and induced GRSP (glomalin-related soil protein) secretion. Furthermore, a comparative analysis of the transcriptomes of the symbiotic group and the non-symbiotic group revealed multiple differentially expressed genes (DEGs) in the 'metal ion transport', 'glutathione metabolism', 'cysteine and methionine metabolism', and 'plant hormone signal transduction' terms. The expression changes of these DEGs were basically consistent with the changes in physio-biochemical characteristics. Overall, AMF alleviated Cd stress in wheat mainly by promoting immobilization and sequestration of Cd, reducing ROS production and accelerating their scavenging, in which the rapid metabolism of GSH may play an important role.
Comparative physiological and transcriptomic analyses reveal ascorbate and glutathione coregulation of cadmium toxicity resistance in wheat genotypes

Abstract:Background: Cadmium (Cd) is a heavy metal with high toxicity that severely inhibits wheat growth and development. Cd easily accumulates in wheat kernels and enters the human food chain. Genetic variation in the resistance to Cd toxicity found in wheat genotypes emphasizes the complex response architecture. Understanding the Cd resistance mechanisms is crucial for combating Cd phytotoxicity and meeting the increasing daily food demand. Results: Using two wheat genotypes (Cd resistant and sensitive genotypes T207 and S276, respectively) with differing root growth responses to Cd, we conducted comparative physiological and transcriptomic analyses and exogenous application tests to evaluate Cd detoxification mechanisms. S276 accumulated more H2O2, O2-, and MDA than T207 under Cd toxicity. Catalase activity and levels of ascorbic acid (AsA) and glutathione (GSH) were greater, whereas superoxide dismutase (SOD) and peroxidase (POD) activities were lower in T207 than in S276. Transcriptomic analysis showed that the expression of RBOHA, RBOHC, and RBOHE was significantly increased under Cd toxicity, and two-thirds (22 genes) of the differentially expressed RBOH genes had higher expression levels in S276 than inT207. Cd toxicity reshaped the transcriptional profiling of the genes involving the AsA-GSH cycle, and a larger proportion (74.25%) of the corresponding differentially expressed genes showed higher expression in T207 than S276. The combined exogenous application of AsA and GSH alleviated Cd toxicity by scavenging excess ROS and coordinately promoting root length and branching, especially in S276. Conclusions: The results indicated that the ROS homeostasis plays a key role in differential Cd resistance in wheat genotypes, and the AsA-GSH cycle fundamentally and vigorously influences wheat defense against Cd toxicity, providing insight into the physiological and transcriptional mechanisms underlying Cd detoxification.

Abstract:Root radial transport is important for cadmium (Cd) absorption and root-shoot translocation. However, the relationship between root structural characteristics and radial transport of Cd in wheat is still unclear. Six wheat cultivars with different Cd tolerance and accumulation characteristics were used to investigate the roles of root phenotype, microstructure, and apoplastic and symplastic pathways in Cd uptake and root-shoot transport in pot culture. Longer root length, smaller root diameter, and more numerous root tips were more conducive to Cd absorption, while thicker roots were able to retain more Cd, thus reducing root-shoot transport and improving Cd tolerance of shoots. Cd stress can induce the deposition of apoplastic barriers in wheat roots, and the deposition of the apoplastic barrier increases under greater stress. The formation of apoplastic barriers can reduce Cd absorption and transfer to the shoot, and the presence of passage cells can weaken this effect. The cell wall thickening induced by Cd stress enhanced Cd adsorption capacity in wheat roots, but there was no significant correlation between Cd content and polysaccharide content in the cell wall. The up-regulated expression of TaHMA3 and TaVP1, which encode proteins related to Cd compartmentalization, was associated with increased Cd tolerance in wheat and decreased Cd translocation to aboveground parts. The morphology and anatomy of roots appear to play critical roles in Cd tolerance, uptake, and translocation in wheat. The present study provides useful information for the selection of wheat cultivars with low Cd accumulation.

Abstract:Cadmium is commonly recognized as toxic to plant growth, low-level Cd has promoting effects on growth performance, which is so-called hormesis. Although Cd toxicity in wheat has been widely investigated, knowledge of growth response to a broad range of Cd concentrations, especially extremely low concentrations, is still unknown. In this study, the morphological, physiological, and biochemical performance of wheat seedlings to a wide range of Cd concentrations (0–100 μΜ) were explored. Low Cd treatment (0.1–0.5 μM) improved wheat biomass and root development by enhancing the photosynthetic system and antioxidant system ability. Photosynthetic rate ( P n ) was improved by 5.72% under lower Cd treatment (1 μΜ), but inhibited by 6.05–49.85% from 5 to 100 μΜ. Excessive Cd accumulation induced oxidative injury manifesting higher MDA content, resulting in lower photosynthetic efficiency, stunted growth, and reduction of biomass. Further, the contents of ascorbate, glutathione, non-protein thiols, and phytochelatins were improved under 5–100 μΜ Cd treatment. The ascorbate peroxidase activity in the leaf showed a hormetic dose–response characteristic. Correlation analysis and partial least squares (PLS) results indicated that antioxidant enzymes and metabolites were closely correlated with Cd tolerance and accumulation. The results of the element network, correlation analysis, and PLS showed a crucial role for exogenous Cd levels in K, Fe, Cu, and Mn uptake and accumulation. These results provided a deeper understanding of the hormetic effect of Cd in wheat, which would be beneficial for improving the quality of hazard and risk assessments.

Multiomics reveals an essential role of long-distance translocation in regulating plant cadmium resistance and grain accumulation in allohexaploid wheat (Triticum aestivum)

Abstract:Cadmium (Cd) is a highly toxic heavy metal that readily enters cereals, such as wheat, via the roots and is translocated to the shoots and grains, thereby posing high risks to human health. However, the vast and complex genome of allohexaploid wheat makes it challenging to understand Cd resistance and accumulation. In this study, a Cd-resistant cultivar of wheat, 'ZM1860', and a Cd-sensitive cultivar, 'ZM32', selected from a panel of 442 accessions, exhibited significantly different plant resistance and grain accumulation. We performed an integrated comparative analysis of the morpho-physiological traits, ionomic and phytohormone profiles, genomic variations, transcriptomic landscapes, and gene functionality in order to identify the mechanisms underlying these differences. Under Cd toxicity, 'ZM1860' outperformed 'ZM32', which showed more severe leaf chlorosis, poorer root architecture, higher accumulation of reactive oxygen species, and disordered phytohormone homeostasis. Ionomics showed that 'ZM32' had a higher root-to-shoot translocation coefficient of Cd and accumulated more Cd in the grains than 'ZM1860'. Whole-genome re-sequencing (WGS) and transcriptome sequencing identified numerous DNA variants and differentially expressed genes involved in abiotic stress responses and ion transport between the two genotypes. Combined ionomics, transcriptomics, and functional gene analysis identified the plasma membrane-localized heavy metal ATPase TaHMA2b-7A as a crucial Cd exporter regulating long-distance Cd translocation in wheat. WGS- and PCR-based analysis of sequence polymorphisms revealed a 25-bp InDel site in the promoter region of TaHMA2b-7A, and this was probably responsible for the differential expression. Our multiomics approach thus enabled the identification of a core transporter involved in long-distance Cd translocation in wheat, and it may provide an elite genetic resource for improving plant Cd resistance and reducing grain Cd accumulation in wheat and other cereal crops.
Effects of Zinc Application on Cadmium (cd) Accumulation and Plant Growth Through Modulation of the Antioxidant System and Translocation of Cd in Low- and High-Cd Wheat Cultivars.

Abstract:Cadmium (Cd) contamination is a big challenge for managing food supply and safety around the world. Reduction of the bioaccumulation of cadmium (Cd) in wheat is an important way to minimize Cd hazards to human health. This study compared and highlighted the effects of soil and foliar applications of Zn on Cd accumulation and toxicity in cultivars with high Cd accumulation (high-Cd wheat) and low Cd accumulation (low-Cd wheat). Both foliar and soil Zn applications provided effective strategies for reducing wheat grain Cd concentrations in the high-Cd wheat by 26-49% and 25-52%, respectively, and these also significantly reduced the concentrations in wheat stems and leaves. Foliar and soil Zn applications significantly reduced Cd in leaves and stems of the low-Cd wheat but had no effects on grain Cd. Both soil and foliar Zn applications significantly alleviated Cd toxicity by regulation of Cd transport genes, as reflected by the increased grain yield and antioxidant enzyme activity in the wheat tissues. Gene expression in response to zinc application differed in the two wheat cultivars. Down-regulation of the influx transporter (TaNramp5) and upregulation of the efflux transporters (TaTM20 and TaHMA3) in the high-Cd wheat may have contributed to the Zn-dependent Cd alleviation and enhanced its tolerance to Cd toxicity. Additionally, foliar Zn applications down-regulated the leaf TaHMA2 expression that reduced root Cd translocation to shoots, while soil Zn applications down-regulated the root TaLCT1 expression, which contributed to the reduction of root Cd concentrations. Soil (99 kg ZnSO4 center dot 7H(2)O ha(-1)) and foliar (0.36 kg ZnSO4 center dot 7H(2)O ha(-1)) Zn applications can effectively decrease the Cd in grains and guarantee food safety and yield, simultaneously. Abstract:Cadmium (Cd) contamination is a serious challenge in agricultural soils worldwide, resulting in Cd entering the food chain mainly through plant-based food and threatening human health. Minimizing Cd bioaccumulation in wheat is an important way to prevent Cd hazards to humans. Hydroponic and pot experiments were conducted to comprehensively evaluate the effects of zinc sulfate (ZnSO 4 ) and zinc oxide nanoparticles (nZnO) on Cd uptake, translocation, subcellular distribution, cellular ultrastructure, and gene expression in two wheat genotypes that differ in grain Zn accumulation. Results showed that high-dose nZnO significantly reduced root Cd concentration (52.44%∼56.85%) in two wheats, in contrast to ZnSO 4 . The S216 exhibited higher tolerance to Cd compared to Z797. Importantly, Zn supplementation enhanced Cd sequestration into vacuoles and binding to cell walls, which conferred stability to ultracellular structures and photosynthetic apparatus. Down-regulation of influx transporter ( TaHMA2 and TaLCT1 ) and up-regulation of efflux transporters ( TaTM20 and TaHMA3 ) in Z797 might contribute to Zn-dependent alleviation of Cd toxicity and enhance its Cd tolerance. Down-regulation of ZIP transporters ( TaZIP3 , -5 , and -7 ) might contribute to an increase in root Zn concentration and inhibit Cd absorption. Additionally, soil Zn provided an effective strategy for the reduction of grain Cd concentrations in both wheats, with a reduction of 26%∼32% (high ZnSO 4 ) and 11%∼67% (high nZnO), respectively. Collectively, these findings provide new insights and perspectives on the mechanisms of Cd mitigation in wheats with different Zn fertilizers and demonstrate that the effect of nZnO in mitigating Cd stress is greater than that of ZnSO 4 fertilizers.

Abstract:Soil cadmium (Cd) pollution poses a serious threat to both the productivity and quality of wheat. This study aimed to investigate the genotypic variation in grain Cd concentration in wheat through field and pot experiments. Among 273 wheat genotypes, a significant genotypic difference was found in grain Cd concentration, ranging from 0.01 to 0.14 mg kg-1. Two contrasting genotypes, X321 (a low grain Cd accumulator) and X128 (a high grain Cd accumulator), were selected for pot experiments. X321 exhibited a 17.9% greater reduction in yield and a 10.2% lower shoot-to-grain Cd translocation rate than X128 under Cd treatment. Grain Cd content showed a positive correlation with soil available Cd content and a negative correlation with Cu content. Soil catalase activity significantly decreased in X128 under Cd stress, whereas no difference was found in X321. The grains of X321 exhibited a more compact spatial distribution of starch grains and protein matrix than those of X128. Moreover, the size of A-type starch in X128 was larger than in X321. Meanwhile, X128 contained much B-type starch, with some surface pits observed on A-type granules under Cd stress. Cd treatment increased the abundance of rhizosphere microorganism communities, with Ellin6067 and Ramlibacter being enriched in X128 under Cd treatment, which might facilitate Cd uptake. The accumulation of Cd in grains demonstrated a strong positive correlation with the rhizosphere bacterial diversity (correlation coefficient = 0.78). These findings provide new insights into the basis of grain Cd accumulation in wheat and have potential implications for developing new verities with low Cd accumulation to ensure food safety and minimize human exposure.

Abstract:Microorganisms can modulate the contents of cadmium (Cd) and zinc (Zn) in wheat grains. Increasing the essential nutrient element Zn and decreasing the toxic element Cd in wheat grains can significantly improve human health. To characterize the specific bacterial communities associated with Cd and Zn accumulation in wheat, we conducted a field experiment by planting wheat cultivars differing in their capacity for Cd and Zn accumulation. The grain Cd contents in wheat cultivars YN23 (0.078 mg kg−1), JN17 (0.080 mg kg−1), YN836 (0.081 mg kg−1) and LM2 (0.091 mg kg−1) were significantly lower than those in ZM32 (0.16 mg kg−1). The Zn contents were significantly higher in the grains of JN17 (44.36 mg kg−1), LM2 (42.22 mg kg−1) and ZM32 (43.19 mg kg−1) than YN23 (27.05 mg kg−1) and YN836 (29.70 mg kg−1). On the basis of contents and bio-concentration factors of Cd and Zn in wheat grain, JN17 and LM2 were identified as low-Cd- and high-Zn-accumulating cultivars, YN23 and YN836 were low-Cd- and low-Zn-accumulating cultivars, and ZM23 was a high-Cd- and high-Zn-accumulating cultivar. The relative abundance values of Gemmatimonadaceae, Sphingomonadaceae and Beijerinckiaceae in the rhizospheres of low-Cd cultivars were significantly higher than those of high-Cd cultivars. High-Zn cultivars had higher abundance of Rhodanobacteraceae in the rhizosphere than did low-Zn cultivars. The low-Cd- and high-Zn-accumulating cultivars were enriched in Alphaproteobacteria and Gemmatimonadaceae, and strengthened nitrification function including aerobic_ammonia_oxidation and aerobic_nitrite_oxidation in the rhizosphere soil, thus contributing to the decreased Cd and increased Zn contents in wheat grains. Microbial technology is a promising method to control the contents of Cd and Zn in wheat grains.

Abstract:Abstract Background Heavy metal cadmium (Cd) is a common environmental pollutant in soils, which has an negative impacts on crop growth and development. At present, cadmium has become a major soil and water heavy metal pollutant, which not only causes permanent and irreversible health problems for humans, but also causes a significant reduction in crop yields. Results This study examined the chemical forms of Cd in the roots of two wheat varieties (M1019 and Xinong20) by continuous extraction and analyzed differences in distribution characteristics of Cd in the root cell wall, cytoplasm, and organelles by elemental content determination and subcellular separation. Furthermore, we conducted proteomics analysis of the roots of the two varieties under Cd pollution using mass spectrometry quantitative proteomics techniques. A total of 11,651 proteins were identified, of which 10,532 proteins contained quantitative information. In addition, the differentially expressed proteins in the two varieties were related to DNA replication and repair, protein metabolism, and the glutathione metabolism pathway. Conclusion The results of this study improve our understanding of the mechanism of plant responses to Cd stress.

Abstract:Many soils face dual challenges of cadmium (Cd) contamination and salinization. However, the response of crops, especially wheat, to combined Cd and salinity stress is not understood. Here, wheat was grown in a hydroponic model for 14 days under single and combined Cd and NaCl stresses. Growth parameters, tissue Cd2+ and Na+ contents, and leaf chlorophyll (Chl), O2•-, and MDA levels were determined. Comparative transcriptomic and metabolomic analyses of the leaves were performed. The results showed that combined stress had a greater inhibitory effect on Chl contents and generated more O2•- and MDA, resulting in more severe wheat growth retardation than those under Cd or NaCl stress. Stress-induced decrease in Chl levels may be attributed to the inhibition of Chl biosynthesis, activation of Chl degradation, or a decline in glutamate content. Cd addition weakened the promotional effect of NaCl on SOS1 gene expression, thereby increasing the Na+ content. Contrastingly, NaCl supplementation downregulated the Nramp and ZIP gene expressions related to Cd uptake and transport, thereby impeding Cd2+ accumulation. All stresses enhanced tryptophan content via promoting tryptophan biosynthesis. Meanwhile, Cd and NaCl stresses activated phenylpropanoid biosynthesis and purine metabolism, respectively, thereby increasing the levels of caffeic acid, fumaric acid, and uric acid. Activating the TCA cycle was important in the wheat's response to combined stress. Additionally, NaCl and combined stresses affected starch and sucrose metabolism, resulting in sucrose and trehalose accumulation. Our findings provide a comprehensive understanding of the response of wheat to the combined Cd and salinity stress.
RNA-sequencing analysis reveals transcriptional changes in the roots of low-cadmium-accumulating winter wheat under cadmium stress

Abstract:Wheat is the second most important food crop worldwide, and thus, it is important to protect both its yield and quality. Cadmium (Cd) is a heavy metal with strong soil–plant mobility and biological toxicity. Thus, screening low-Cd-accumulating genotypes to elucidate the underlying molecular regulatory mechanisms and gene networks related to Cd stress may be useful to reduce dietary exposure to this heavy metal. Here, the root transcriptomes of the low-Cd-accumulating genotype YM16 treated (YT) or not treated (YC) with Cd were analyzed using the Illumina Hiseq4000 data. The gene expression analysis of YT and YC revealed 1427 differentially expressed genes (DEGs; 429 upregulated and 998 downregulated). Of these, 298 upregulated DEGs were assigned to 41 gene ontology (GO) terms, whereas 537 downregulated DEGs were assigned to 61 GO terms. The pathway enrichment analysis showed that phenylpropanoid biosynthesis, glutathione metabolism, sulfur metabolism, and nitrogen metabolism were significantly enriched in upregulated DEGs, which are involved in defense and detoxification in response to Cd stress. The expression pattern of eight selected DEGs was also analyzed by quantitative real-time polymerase chain reaction, and the results were consistent with those obtained by RNA-sequencing, confirming data reliability. Overall, our study might increase the understanding of complex molecular mechanisms that regulate the responses of plants to Cd stress, and help develop new improved wheat varieties with low Cd content in the grain.

Abstract:Corn (Zea Mays L.) and wheat (Triticum aestivum L. ) seedlings were grown in four cadmium (Cd) concentration levels (0 - 1 mg/l) in a hydroponic system to analyze the antioxidant enzyme system, Cd concentration in the shoots and roots of plants, proline contents, growth responses and chlorophyll contents in the leaves of corn and wheat. The results showed that there was a significant increase in malondialdehyde (MDA) concentration from 0 to 1 mg/l, and peroxidase (POD) andcatalase (CAT) activities in the corn and wheat subjected to 0 – 1 mg/l of Cd. However, there was a significant decrease in the superoxide dismutase (SOD) activities in the corn and wheat subjected to 0 - 1 mg/l of Cd. This indicated that Cd stress induced an oxidative stress response in corn and wheat seedlings, characterized by an accumulation of MDA, decrease in the activities of SOD and increase in the activities of POD and CAT. Root and leaf Cd concentrations of corn and wheat increased with their exposure to the Cd level,s and the highest Cd concentration occurred in roots, followed by the leaves. An increase in proline in the leaves of corn and wheat seedlings exposed to Cd occurred as well as a decrease in chlorophyll (CHL) contents and shoots and roots biomass. Thus, Cd levels negatively affected the corn and wheat seedlings growth. Key words: Cadmium, corn (Zea mays L.), wheat (Triticum aestivum L.).

Abstract:The accumulation and elimination kinetics for Cd described by one-compartment model were determined in five subcellular fractions of wheat after exposure to 5.0μM Cd with or without pre-exposure to 0.1μM Cd or 5.0μM Zn. The results show that the acclimation of wheat to Cd or Zn generates its resistance to Cd and decreased Cd accumulation in wheat. The acclimation of wheat to Cd or Zn enhanced the accumulation of Cd in the biological detoxified fractions especially the heat-stable fraction and decreased the accumulation of Cd in the metal sensitive fractions especially the heat-denatured proteins over time. Subsequently, the acclimation to Cd or Zn increased the activities of CAT and SOD in root and shoot to different extents compared to the unacclimated wheat. This study for the first time suggests that the acclimation of plant to low-level metal affects the accumulation and the toxicity of metal in plant.
Trade-offs of reproductive growth and Cd remobilization regulated Cd accumulation in wheat grains (Triticum aestivum L.)

Abstract:Minimization of cadmium (Cd) accumulation in wheat grain (Triticum aestivum L.) is an important way to prevent Cd hazards to humans. However, little is known about the mechanisms of varietal variation of Cd accumulation in wheat grain. This study explores the physiological mechanisms of Cd bioaccumulation through field and hydroponic experiments on two wheat varieties of low-Cd-accumulating variety (L-6331) and high-Cd-accumulating variety (H-6049). Field study showed that average Cd accumulative rates in spikes of H-6049 were 1.57-fold of L-6331 after flowering, ultimately grain-Cd of H-6049 was 1.70-fold of L-6331 in Cd-contaminated farmland. The hydroponic experiment further confirmed that more vegetative tissues of L-6331 were involved in the remobilization of Cd, which jointly mitigated the process of Cd loaded to grains when leaf-cutting conducted after Cd stress. Additionally, the L1 and N1 of L-6331 play an especially important role in regulating Cd remobilization, and the larger EVB areas in N1 have the morphological feature that facilitates the transfer of Cd to L1. Overall results implied that low-Cd-accumulating variety initiated more trade-offs of reproductive growth and Cd remobilizatoin under Cd-stress after flowering compared with high-Cd-accumulating variety, and provided new insights into the processes of Cd loaded into wheat grains among different varieties.

Abstract:BACKGROUND:Cadmium (Cd) is a severe detrimental environmental pollutant. To adapt to Cd-induced deleterious effects, plants have evolved sophisticated defence mechanisms. In this study, a genome-wide transcriptome analysis was performed to identify the mechanisms of Cd tolerance using two barley genotypes with distinct Cd tolerance.RESULTS:Microarray expression profiling revealed that 91 genes were up-regulated by Cd in Cd-tolerant genotype Weisuobuzhi and simultaneously down-regulated or non-changed in Cd-sensitive Dong17, and 692 genes showed no change in Weisuobuzhi but down-regulated in Dong17. Novel genes that may play significant roles in Cd tolerance were mainly via generating protectants such as catalase against reactive oxygen species, Cd compartmentalization (e.g. phytochelatin-synthase and vacuolar ATPase), and defence response and DNA replication (e.g. chitinase and histones). Other 156 up-regulated genes in both genotypes also included those encoding proteins related to stress and defence responses, and metabolism-related genes involved in detoxification pathways. Meanwhile, biochemical and physiological analysis of enzyme (ATPase and chitinase), phytohormone (ethylene), ion distribution and transport (Cd, Na(+), K(+), Ca(2+), ABC transporter) demonstrated that significantly larger Cd-induced increases of those components in Weisuobuzhi than those in Dong17. In addition, Cd-induced DNA damage was more pronounced in Dong17 than that in Weisuobuzhi.CONCLUSIONS:Our findings suggest that combining microarray, physiological and biochemical analysis has provided valuable insights towards a novel integrated molecular mechanism of Cd tolerance in barley. The higher expression genes in Cd tolerant genotype could be used for transgenic overexpression in sensitive genotypes of barley or other cereal crops for elevating tolerance to Cd stress.

Abstract:Cadmium (Cd) pollution in cultivated soils has posed a great risk to human health through the soil-plant-human pathway. Therefore, it is important to derive soil thresholds for the low-Cd accumulating genotype of wheat (Triticum aestivum L) to promote its application in agricultural production on Cd-contaminated sites. Here, a pot experiment was performed to explore the transfer characteristics of Cd in two contrasting wheat genotypes at three different soils and the effect of soil parameters together with soil safety Cd thresholds derivation. Generally, grain Cd highly accumulating wheat genotype (Zhenmai10, HT) showed higher Cd accumulation in grains than grain Cd weakly accumulating wheat genotype (Aikang58, LT). Stepwise multiple linear regression (SMLR) analysis (log-transformed Freundlich-type) indicated that Cd accumulation in wheat grains was strongly related to soil total Cd concentration and pH for both genotypes (R2 = 0.907*** for HT; R2 = 0.910*** for LT). Combining the simple regression model of soil-plant transfer system with the risk assessment method based on human health, soil total Cd thresholds for three soils were calculated with the values of 0.62, 0.82, and 0.62 mg kg-1 in LT genotype and 0.31, 0.77, and 0.49 mg kg-1 in HT genotype. Therefore, we suggested that when deducing soil thresholds, the ability of wheat genotypes to accumulate Cd and soil properties should be considered because of the large differences in soil thresholds between different genotypes and types of soils. We believe our results will promote the application of low-Cd wheat genotypes to agricultural production, thereby ensuring the safety of their products.

Abstract:Cadmium (Cd) is detrimental to both plants and humans. Maize (Zea mays L.) genotypes exhibit variations in Cd accumulations. This study examined variations in Cd accumulation and tolerance among four maize genotypes with contrasting root morphology. The four maize genotypes were cultivated in a semi-hydroponic system with three Cd concentrations (0, 10, 20 μmol L-1). The effects of Cd on plant growth and physiology were assessed 39 days after transplanting. Results showed that root characteristics were positively correlated with root Cd accumulation and the bioconcentration factor under Cd20 treatment. Genotypes Shengrui999 and Zhengdan958 exhibited higher total Cd content than Xundan29 and Zhongke11 under Cd20 conditions. Cd toxicity led to membrane degradation of chloroplast mesophyll cells, loosening and swelling of grana lamella, and reduced starch reserves. The greater tolerance of Shengrui999 and Zhengdan958 was contributed to factors such as root biomass, shallower root depth, higher Cd content, accumulation of osmolyte such as soluble protein, antioxidant activities such as catalase (CAT), and the presence of phytohormone gibberellic acid. The study establishes a link between root morphology, Cd accumulation, and tolerance in maize plants, as demonstrated by the higher Cd accumulation and shallower root system in Cd-tolerant genotypes. This research provides a foundation for breeding maize cultivars better suited for adaptation to moderate Cd-contaminated environments.
