Abstract:Sphingolipids are major structural components of endomembranes and have also been described as an intracellular second messenger involved in various biological functions in all eukaryotes and a few prokaryotes. Ceramides (Cer), the central molecules of sphingolipids, have been depicted in cell growth arrest, cell differentiation, and apoptosis. With the development of lipidomics, the identification of ceramides has been analyzed in many species, mostly in model insects. However, there is still a lack of research in non-model organisms. Here we describe a relatively simple and sensitive method for the extraction, identification, and quantification of ceramides in Hemiptera Insects (brown planthooper), followed by Ultra-Performance Liquid Chromatography coupled to tandem mass spectrometry (UPLC-MS/MS). C18 is used as the separation column for quantitative detection and analysis on the triple quadruple liquid mass spectrometer. In this protocol, the standard curve method is adopted to confirm the more accurate quantification of ceramides based on the optional detection conditions.
[A clinicopathological study of eighteen autopsy cases with acquired toxoplasmosis].

Abstract:Eighteen autopsy cases of acquired toxoplasmosis in New York City were studied. Seventeen cases were with acquired immunodeficiency syndrome (AIDS) and one patient with Hodgkin's disease. All 18 cases involved the brain and nine of them disseminated to the heart (8 cases), lung (4 cases), pancreas (3 cases), alimentary tract (2 cases) and urogenital organs (3 cases). The authors divided the acquired toxoplasmosis into (1) immunocompetent, (2) immunocompromised and (3) immunodeficient types. The autopsy findings showed that the brain, heart and lung were the most susceptible organs. Pseudocysts were also found in lungs and alimentary tract, suggesting an autoinfection by swallowing sputum containing Toxoplasma. Toxoplasma in the urogenital organs might become a source for sexual transmitted toxoplasmosis.

Abstract:The in-depth isomeric and isobaric description of ultra-complex organic mixtures remains one of the most challenging analytical tasks. In the last two decades, ion mobility coupled to high-performance mass spectrometry added an additional structural dimension. Despite tremendous instrumental improvements, commercial devices are still limited in ion mobility and mass spectrometric resolving power and struggle to resolve isobaric species and complex isomeric patterns. To overcome these limitations, we explored the capabilities of cyclic ion mobility high-resolution mass spectrometry with special emphasis on petrochemical applications. We could show that quadrupole-selected ion mobility mass spectrometry gives closer insights into the isomeric distribution. In combination with slicing the specific parts of the ion mobility dimension, isobaric interferences could be drastically removed. Collision-induced dissociation (CID) allowed separating structural groups of polycyclic aromatic hydrocarbons and heterocycles (PAH/PASH), deploying up to 10 passes in the cyclic ion mobility device. Finally, we introduce a data processing workflow to resolve the 3.4 mDa SH4/C3 mass split by combining ion mobility and mass spectrometric resolving power. Cyclic ion mobility with the intelligent design of experiments and processing routines will be a powerful approach addressing the isobaric and isomeric complexity of ultra-complex mixtures.

Abstract:Sphingomyelins (SMs) are an essential class of lipids widely existing in different organisms. The sphingoid base and N-acyl chain are two building blocks which can undergo different types of modifications during lipogenesis, including desaturation, hydroxylation, and methyl branching. Current lipidomic analysis methods cannot provide detailed information on these structural motifs. Herein, we developed a tandem mass spectrometric method by harnessing radical-directed dissociation (RDD) from collision-induced dissociation (CID) of the bicarbonate anion adduct of SM ([M + HCO3](-)). A major RDD channel produced a high-abundance fragment carrying the intact N-acyl chain, termed as "N-acyl fragment", allowing the assignment of the sphingoid base/N-acyl composition and relative quantitation of compositional isomers of SM at high sensitivity. RDD also produced intrachain fragments in lower abundances, which helped localization of methyl branching and hydroxylation in SM. The acetone Paterno-Buchi (PB) reaction was found to be capable of derivatizing the Delta 4 carbon-carbon double bond (C=C) in sphingosine (SPH) base and producing C=C diagnostic ions upon CID, albeit at much lower efficiencies than those of the isolated C=C in alkyl chains. A liquid chromatography-mass spectrometry workflow was developed by incorporating MS2 CID of SM via [M + HCO3]- and PB-MS2 CID. The capability of profiling SM with detailed structural information was demonstrated by analyzing complex lipid extracts from porcine brain and Caenorhabditis elegans. These results provided visualization of the sphingoid base/N-acyl compositional isomers of SM lipids and revealed large structural diversity from each sample. These included identification of the sphingadiene base [d18:1(Delta 4,14)], C=C location isomers in N-acyls, C-2 hydroxylation of N-acyls, and iso-methyl branched SPH base.

Abstract:To coordinate cell signaling and other membrane‐mediated cellular functions, the cellular plasma membrane is organized into domains of differing protein and lipid composition. Though the distributions of specific membrane proteins can be imaged with many techniques, the precise spatial organization of sphingolipids within the plasma membrane is not well‐established. Here we have combined metabolic isotope incorporation with high‐resolution secondary ion mass spectrometry (SIMS) to chemically image the distributions of sphingolipids within the plasma membranes of intact fibroblast cells with at least 100 nm lateral resolution. Using this approach, we characterize how the sphingolipid distribution changes as a function of metabolic pulse‐labeling time. We show that changes in the short range (<1 micron) sphingolipid organization are not detectable, but the non‐random sphingolipid organization occurring on longer length scales (>5 microns) varies with the duration of selective isotope incorporation.

Abstract:When a packet of ions in a buffer gas is exposed to a weak electric field, the ions will separate according to differences in their mobilities through the gas. This separation forms the basis of the analytical method known as ion mobility spectroscopy and is highly efficient, in that it can be carried out in a very short time frame (micro- to milliseconds). Recently, efforts have been made to couple the approach with liquid-phase separations and mass spectrometry in order to create a high-throughput and high-coverage approach for analyzing complex mixtures. This article reviews recent work to develop this approach for proteomics analyses. The instrumentation is described briefly. Several multidimensional data sets obtained upon analyzing complex mixtures are shown in order to illustrate the approach as well as provide a view of the limitations and required future work.

Abstract:Understanding the composition, structure and stability of synthetic molecules is crucial for their design, yet currently the analytical tools commonly used do not readily provide this information, particularly for larger complex analytes above M = 500 Da. In this perspective, we show how ion mobility mass spectrometry (IM-MS), in combination with tandem mass spectrometry, computational methods and other complementary techniques, can be used to structurally characterise new synthetic molecules, make and predict new complexes, monitor disassembly processes and determine stability. Using IM-MS, we present an experimental and computational framework for the analysis and design of complex molecular architectures such as (metallo)supramolecular cages, interlocked molecules, rotaxanes, dendrimers, polymers and host-guest complexes.

Abstract:We developed a liquid chromatography/electrospray ionization tandem mass spectrometry method for the simultaneous quantitative determination of C18 sphingosine (Sph), C18 dihydrosphingosine (dhSph), C18 phytosphingosine (pSph), C18 sphingosine-1-phosphate (S1P), C18 dihydrosphingosine-1-phosphate (dhS1P), and C18 phytosphingosine-1-phosphate (pS1P). Samples were prepared by simple methanol deproteinization and analyzed in selected reaction monitoring modes. No peak tailing was observed on the chromatograms using a Capcell Pak ACR column (1.5 mm i.d. × 250 mm, 3 μm, Shiseido). The calibration curves of the sphingoids showed good linearity (r > 0.996) over the range of 0.050–5.00 pmol per injection. The accuracy and precision of this method were demonstrated using four representative biological samples (serum, brain, liver, and spleen) from mice that contained known amounts of the sphingoids. Samples of mice tissue such as plasma, brain, eye, testis, liver, kidney, lung, spleen, lymph node, and thymus were examined for their Sph, dhSph, pSph, S1P, dhS1P, and pS1P composition. Abstract:Shotgun lipid analysis based on electrospray ionization-tandem mass spectrometry (ESI-MS/MS) is increasingly used in lipidomic studies. One challenge for the shotgun approach is the discrimination of lipid isomers and isobars. Gas-phase charge inversion via ion/ion reactions has been used as an effective method to identify multiple isomeric/isobaric components in a single MS peak by exploiting the distinctive functionality of different lipid classes. In doing so, fatty acyl chain information can be obtained without recourse to condensed-phase separations or derivatization. This method alone, however, cannot provide carbon-carbon double bond (C=C) location information from fatty acyl chains. Herein, we provide an enhanced method pairing photochemical derivatization of C=C via the Paternò-Büchi reaction with charge inversion ion/ion tandem mass spectrometry. This method was able to provide gas-phase separation of phosphatidylcholines and phosphatidylethanolamines, the fatty acyl compositions, and the C=C location within each fatty acyl chain. We have successfully applied this method to bovine liver lipid extracts and identified 40 molecular species of glycerophospholipids with detailed structural information including head group, fatty acyl composition, and C=C location. Abstract:Domains enriched with cholesterol and sphingolipids, often referred to as lipid rafts, are believed to be present in the plasma membranes of eukaryotic cells and mediate important cellular functions, including cell signaling and virus budding. Changes in the abundances of cholesterol and sphingolipids influence many cellular processes. Yet, the distributions of cholesterol and sphingolipids within the plasma membrane have not been established. The inability to directly visualize lipids and cholesterol without the use of labels that may induce clustering renders characterizing the precise distribution of cholesterol and sphingolipids within the plasma membrane especially challenging. To address this problem, we have previously combined a chemically specific and spatially well-resolved imaging technique, high-resolution secondary ion mass spectrometry (SIMS), with metabolic labeling in order to image the distributions of sphingolipids within the plasma membranes of intact cells with at least 100 nm lateral resolution. We now report the use of this approach to image the sphingolipid and cholesterol distributions in parallel on the surface of intact fibroblast cells. We also quantitatively assess whether the cholesterol and sphingolipids are heterogeneously distributed and co-localized within the membrane. The results of these studies and their implications on models of membrane organization will be discussed.

Abstract:It is increasingly evident that a more detailed molecular structure analysis of isomeric lipids is critical to better understand their roles in biological processes. The occurrence of isomeric interference complicates conventional tandem mass spectrometry (MS/MS)-based determination, necessitating the development of more specialised methodologies to separate lipid isomers. The present review examines and discusses recent lipidomic studies based on ion mobility spectrometry combined with mass spectrometry (IMS-MS). Selected examples of the separation and elucidation of structural and stereoisomers of lipids are described based on their ion mobility behaviour. These include fatty acyls, glycerolipids, glycerophospholipids, sphingolipids, and sterol lipids. Recent approaches for specific applications to improve isomeric lipid structural information using direct infusion, coupling imaging, or liquid chromatographic separation workflows prior to IMS-MS are also discussed, including: 1) strategies to improve ion mobility shifts; 2) advanced tandem MS methods based on activation of lipid ions with electrons or photons, or gas-phase ion-molecule reactions; and 3) the use of chemical derivatisation techniques for lipid characterisation.
