Abstract:This paper describes a mechanism for a mastication robot using a human skull model. Since 1986 the authors have been developing the WJ (Waseda Jaw) series of mastication robots by using a dental study model, a duralumin mandibular, DC motors, wires, etc. From 1986 to 1991 the authors developed mastication robots with a mechanical mandibular model, and experimented with these robots. However, in order to analyze the human masticatory system, the authors needed a model closer to the human. The characteristics of the new mastication robot mechanism developed in 1992 are, that this robot has a structure similar to the human by using a skull model and, that the argument of the temporalis muscle force vector is variable by using two actuators. On the basis of this robot, the authors will be able to clarify and construct a quantitative and dynamic masticatory model for human mastication because it has human-like structures.
A comparative study on the growth and nitrogen and phosphorus uptake characteristics of 15 wetland species

Abstract:The biomass, root morphology and nutrient uptake capacities of 15 species of local wetland plants were investigated in hydroponic culture. The wetland plants were exposed to 38.5mg center dot L-1 of NH[image omitted]-N, 132.8mg center dot L-1 of NO[image omitted]-N and 10mg center dot L-1 of dissolved inorganic P for 28days. Mean total biomass of the 15 species ranged from 1.2 to 21.6gplant-1, with above/below ground ratios (AG:BG) in the range 1.7-5.5. Mean NH[image omitted]-N, NO[image omitted]-N uptake rates ranged from 3.7 to 14.3mgN center dot day-1 (accounting for 8.0-49.4% of the NH[image omitted]-N supply) and 17.8 to 59.4mgN center dot day-1 (17.8-59.6% of the NO[image omitted]-N supply). Mean P uptake rate ranged from 1.71 to 4.61mgP center dot day-1 (24.1-61.5% of the P supply). The N and P concentrations in plant tissues ranged from 28.2 to 606.1mgN center dot plant-1 and 4.1 to 53.1mgP center dot plant-1, with AG:BG ratio in the range 1.7-7.0 and 1.6-4.6, respectively. The accumulation of N and P in plant tissues was both significantly correlated with plant biomass and root surface area. Among the different species, Canna generalis, Typha latifolia, Thalia dealbata and Lythrum salicaria had greater above- and below-ground biomass, larger root surface area, and greater nutrient uptake and storage rates than the other plants. Our results suggest (or indicate) that the selection of plant species suitable for constructed wetlands can be based on plant biomass and root surface area.
Effects of Plant Diversity on Greenhouse Gas Emissions in Microcosms Simulating Vertical Constructed Wetlands with High Ammonium Loading.

Abstract:Wastewater with relatively high nitrogen concentrations is a major source of nitrous oxide (N2O) and methane (CH4) emissions and exerts multiple stresses on the environment. Studies have shown that plant diversity plays an important role in ecosystem functioning. However, the effects of plant species diversity on CH4 and N2O emissions under high ammonium (NH4+-N) loading rates remain unclear. In this study, a microcosm experiment simulating vertical constructed wetlands supplied with high NH4+-N water levels was established. The treatments included four species richness levels (1, 2, 3, 4) and 15 species compositions. There was no significant relationship between species richness and N2O emissions. However, N2O emissions were significantly reduced by specific plant species composition. Notably, the communities with the presence of Rumex japonicus L. reduced N2O emissions by 62% compared to communities without this species. This reduction in N2O emissions may have been a result of decreased N concentrations and increased plant biomass. CH4 emissions did not respond to plant species richness or species identity. Overall, plant species identity surpassed species richness in lowering N2O emissions from constructed wetlands with high NH4+-N water. The results also suggest that communities with R. japonicus could achieve higher N removal and lower greenhouse gas emissions than other wetland species.
Responses of Dissimilatory Nitrate Reduction to Ammonium and Denitrification to Plant Presence, Plant Species and Species Richness in Simulated Vertical Flow Constructed Wetlands

Abstract:This study investigated the effects of plant presence, plant species and their species richness on plant biomass production, pH, dissolved oxygen (DO), oxidation-reduction potential (ORP), denitrification (DNF), dissimilatory NO3 − reduction to ammonium (DNRA) and two associated bacterial community compositions in thirty vertical flow microcosm wetlands fed with the Hoagland solution, where three plant species richness levels (i.e. unvegetated, monocultured and 4-species polycultured treatment, respectively) were established using four macrophytes. Plant presence increased DO and ORP values, as well as the terminal restriction fragment (TRF) richness and Shannon-Weaver index of the DNRA community and also improved both potential DNF and DNRA rates. The microcosms monocultured with Cyperus alternifolius exhibited the greatest DO, ORP, smallest plant biomass parameters and DNF rates among all of the monocultured microcosms, whereas the microcosms monocultured with Canna glauca and Iris pseudacorus harbored the smallest pH, DO, ORP, the greatest plant biomass parameters and DNRA rates. Compared to both unvegetated and monocultured treatments, the 4-species polycultured treatment was effective in increasing both potential DNF and DNRA rates due to the greatest plant biomass parameters as confirmed by the correlation analysis, but was ineffective in terms of changing both DNF and DNRA community compositions.
Linking the Rhizosphere Effects of 12 Woody Species on Soil Microbial Activities with Soil and Root Nitrogen Status

Abstract:The rhizosphere effect depicts the changes in soil physical, chemical, and biological properties due to altered microbial activity in response to root activity, which has important implications for soil carbon and nutrient cycling. However, how the plant-induced changes in rhizosphere soil microbial activity correlate with root and soil nutrient status remains uncertain. Here we targeted 12 woody species of distinct growth forms, i.e., tree vs. shrub, in an urban park on a university campus in Beijing, China. We measured root carbon and nitrogen concentrations and soil properties of paired rhizosphere and bulk soils for each plant. We investigated how the rhizosphere effects on microbial properties, as indicated by microbial biomass and potential activity of soil extracellular hydrolases, varied among the 12 woody species and between trees and shrubs, as well as how they were regulated by bulk soil and root nitrogen status. In general, across the 12 species, root nitrogen content, which is a potential indicator of plant life span and nutrient acquisition strategy, was positively correlated with the rhizosphere effect on potential enzyme activity, while the inverse was true for root carbon: nitrogen ratio, which further verified the role of root functional traits in controlling soil nutrient cycling through the rhizosphere effect. Moreover, soil organic carbon, total nitrogen, and available nitrogen concentrations were all found to be negatively correlated with the rhizosphere effect on microbial properties, which highlighted the stimulation of rhizosphere microbial activity for nitrogen acquisition (the nitrogen-mining hypothesis). Trees and shrubs exhibited no significant differences in their rhizosphere effects, and there was a potential difference in the role of rhizosphere effect in regulating soil nitrogen cycling between legumes and non-legumes. Overall, our study highlights the role of plant nutrient acquisition strategy and soil nutrient status in regulating the rhizosphere effect of woody species on microbial-driven soil processes, which is essential for a better understanding of mechanisms by which plants sustain their nutrient supply.
Denitrifying Bacterial Community Dominantly Drove Nitrogen Removals in Vertical Flow Constructed Wetlands As Impacted by Macrophyte Planting Patterns

Abstract:The study aims to identify relations of denitrifying bacterial and fungal communities to nitrogen removals in vertical flow wetland microcosms (VFWMs) using four macrophyte species (Iris pseudacorus, Canna glauca, Scirpus validus and Cyperus alternifolius) and three species richness levels (unplanted, monocultured and 4-species mixture) as fixed factors. Results showed that among four macrophyte species, only Canna glauca planting significantly decreased nitrate removal by 87.7% in the VFWMs. The 4-species mixture improved TN and nitrate removals by 84.0% and 91.3%, but decreased ammonium removal by 94.5%. Heatmap and nonmetric multidimensional scaling analyses identified a significant difference in denitrifying bacterial community structure across macrophyte richness levels, but did not identify the difference in denitrifying fungal communities. The redundancy analysis revealed that denitrifying bacterial community individually explained 99.4% and 93.0% variance of nitrogen removals among four macrophyte species and across macrophyte richness levels, while the fungal community only explained 30.7% and 21.8% variance of nitrogen removals. Overall, the macrophyte richness and bacterial denitrifiers are the critical factors of nitrogen removals in the VFWMs, thus providing useful data to design a vertical flow constructed wetland at a full scale.
Effects of Urban Green Infrastructure Designs on Soil Bacterial Community Composition and Function

Abstract:Green infrastructures (GIs) are environmental designs mainly used to treat urban stormwater runoff pollution. The influences of green infrastructure designs are associated with the bacteria composition and diversity in their soils. This study evaluated the effects of GI design on the composition and function of urban soil bacterial communities in Wuhan, China. GI designs were divided into five different types: engineered (road bioswales, rain gardens, and sunken green spaces) and non-engineered (conventional road greenbelts and urban lawns) designs. The engineered soil combined with the connectivity to urban runoff of the GI design can have strong influences on bacterial community composition and function. Across the five GI design types, we found that the composition of bacterial communities was dominated by Proteobacteria , Acidobacteria , Bacteroidetes and Actinobacteria at the phylum level. The relative abundance of Proteobacteria and Bacteroidetes in the engineered designs was significantly higher than the non-engineered designs and was highest in road bioswales. We detected nine predominant ecological function groups related to the carbon (3 groups) and nitrogen (6 groups) cyclings, with their relative abundance in engineered soils significantly higher than those in non-engineered soils, especially in road bioswales and sunken green spaces. Road bioswales (RBS), with direct connection to urban runoff from street, had the highest denitrification enzyme activity (DEA). DEA of green infrastructures was highly correlated with soil organic carbon, moisture and total nitrogen. Road bioswales appear to function as denitrification hotspots and have higher potential denitrification than other GI designs.

Abstract:In micro-polluted constructed wetland (CW), the low pollutant concentrations and the low COD/N ratios (chemical oxygen demand: total nitrogen in influent), make the biological treatment more difficult. It is expected that root exudates drive microbial-based transformations within plant rhizosphere. In this research, the roles of root exudates of three aquatic plants (Phragmites australis, Typha angustifolia and Cyperus alternifolius) in improving the growth of heterotrophic denitrifying bacteria were determined in a micro-polluted CW. In studied root rhizospheres, the total organic carbon (TOC) released from the plant roots varied significantly among plant species and seasons; the average TOC ranged from 0.1715 to 0.9221mgg-1rootDMd-1, which could fuel a denitrification rate of approximately 156-841kgNO3--Nha-1year-1 if all were used by the denitrifying bacteria; the abundances of nirK- and nirS-encoding bacteria were significantly influenced by the concentration of sucrose and glucose (0.869≤r≤0.933, p<0.05), and microbial community richness and diversity had response to root exudates. The results revealed that root exudates can act as endogenous carbon sources for heterotrophic denitrifying bacteria and ultimately determine the microbe distribution patterns in micro-polluted CW.

Abstract:<h3 class="Heading">Aims</h3>Wetland plants affect ammonia-oxidation rate (AOR) and nitrogen-fixation rate (NFR), resulting in changes in plant-available nitrogen and wetland nitrogen-cycling. This study aims to investigate the underlying mechanisms and influencing factors associated with ammonium oxidation and nitrogen fixation in response to wetland-emergent aquatic plants.<h3 class="Heading">Methods</h3>A field experiment was conducted to study the response of activity, abundance, and community structure of ammonia-oxidizers and diazotrophs to three wetland-emergent aquatic plants in summer and autumn. The root exudates and soil physicochemical properties were determined due to their important role in rhizosphere interactions.<h3 class="Heading">Results</h3>AOR and NFR were significantly higher in rhizosphere than in bulk soils. The increases of AOR and NFR in rhizosphere were significantly positively affected by oxidation-reduction potential, additionally, NFR was also significantly affected by sugar-containing root exudates. Furthermore, NFR strongly correlated with diazotroph abundance, whereas AOR was linearly correlated with ammonia-oxidizing archaea (AOA) activity and mostly determined by a specific AOA taxon.<h3 class="Heading">Conclusions</h3>Emergent plant rhizosphere exhibited strong positive effects on AOR and NFR by altering community structure and abundance of related microorganisms. Release of available oxygen and carbon in plant rhizospheres is vital for promoting ammonium oxidation and nitrogen fixation and might further mediate wetlands nitrogen-cycling.

Abstract:Vegetated drainage ditches (eco-ditches) have drawn much attention in recent years for the ability to remediate diffuse contaminants in rural wastewater through sediment retention, plant uptake and interception, and microbial metabolic activities. However, the effect of plant species on microbial community structure and nitrogen (N) retention in ditch sediment remains poorly understood. In this study, mesocosm plastic drums were planted with eight plant species commonly found in ditches and nurtured with wastewater for 150 days. Sediment total nitrogen (TN) was greatly increased after 150-day nurturing with rural wastewater, from 296.03 mg∙kg−1 (Iris japonica Thunb) to 607.88 mg∙kg−1 (Acorus gramineusO). This study also presents the effect of different plant species on sediment microbial communities, thus providing insight into N removal mechanisms in eco-ditch. Fifty-eight differentially abundant taxa were identified, and sediment microbial community structure for no plant (CK), Acg, Canna indica (Cai), and Typha latifolia L. (Tyl) was primarily linked to sediment NH4+-N and TN. Extremely small proportions of ammonia oxidizing bacteria (AOB) and nitrifying bacteria were detected for all treatments, but large proportions of Crenarchaeota, which comprises the widely existent ammonium oxidized archaea (AOA), were found in CK, Acg and Cai. The abundance of Nitrosotalea from Crenarchaeota presented positive correlations with sediment NH4+-N contents and ammonia oxidation function predicted by Faprotax, indicating Nitrosotalea might be the dominant ammonium-oxidizing microbes in sediment samples. The probable NH4+-N removal pathway in wastewater sediment was through a combined effect of AOA, nitrifying bacteria, and anammox.

Abstract:Plant rhizosphere is the most active hotspot for microbial nitrogen (N) removal in constructed wetlands (CWs), but the effects of plant root activities (i.e., radial oxygen loss, ROL and root exudates, REs) on different microbial N transformation processes, including nitrification, denitrification, anaerobic ammonia oxidation (anammox) and dissimilatory nitrate reduction to ammonia (DNRA) have not been comprehensively studied. In this study, N removal performance and transformation processes in the rhizosphere of CWs were investigated using rhizobox, planar optode system, and metagenomic approaches. Results suggested nitrification was promoted in the rhizosphere, due to the high dissolved oxygen (DO) concentration caused by ROL and the humic acid-like substance in REs, making the rhizosphere a higher removal efficiency of NH 4 + -N than in the nonrhizosphere. Out of expectation, the denitrification process was not promoted by the REs, the potential "carbon source", but inhibited by the high DO concentration in the rhizosphere and the phenol-like substance (a kind of biological denitrification inhibitor) in REs. In addition, both anammox and DNRA processes were, for the first time, found to be inhibited in the rhizosphere. The humic acid-like substance in REs and ammonia-oxidation production hydroxylamine inhibited the activity of anammox microorganisms and genes, and there was a symbiotic relationship between anammox microorganisms and DNRA microorganisms. This study provides better and more complete insights into the biogeochemical cycle of N occurred in the rhizosphere of CWs.

Abstract:The root exudates released from macrophytes are critical for pollutant removal and associated microorganisms, but their role in nitrogen removal remains poorly understood in constructed wetlands (CWs). The release of root exudates by Myriophyllum aquaticum was quantified using a hydroponic experiment, and then four treatments with and without environmentally relevant concentrations of root exudates were conducted to investigate the effect of root exudates on nitrogen removal in CWs. The dissolved organic carbon (DOC) release rate of M. aquaticum was 1.1-2.2 mg g-1 h-1, significantly higher than that of Typha orientalis (0.58-0.78 mg g-1 h-1) and Iris pseudacorus (0.13-0.14 mg g-1 h-1). The DOC release rate of M. aquaticum was affected by the ammonium concentration and harvesting frequency. The removal efficiencies of ammonium and total nitrogen were higher in the CWs with plants than in those without plants, confirming the crucial role of plants in plant -microbe interactions and nitrogen removal in CWs. The CWs showed significantly higher ammonium removal efficiencies under the root exudate treatments (mean 90.4% and 69.3%) than the no-root exudate treatments (87.3% and 62.0%), while total nitrogen presented no significant difference. The phylum-level taxonomic classification of microbial sequences detected in the sediments was slightly different between the root exudate and no-root exudate treatments. The results suggested that the root exudate mass naturally released by plants may be insufficient to change the carbon to nitrogen ratios of wastewater. More gene copies were observed in the rhizosphere sediments than the bulk sediments (p < 0.05), indicating that root exudates had a positive effect on microbes distribution within the rhizosphere. Therefore, high levels of root exudates as external carbon sources should be added to enhance nitrogen removal in CWs for carbon-limited wastewater treatment. The present study provided new insights into the role of macrophytes in pollutant removal and management strategy of CWs for wastewater treatment.
Microbial regulation of soil carbon properties under nitrogen addition and plant inputs removal

Abstract:Background Soil microbial communities and their associated enzyme activities play key roles in carbon cycling in terrestrial ecosystems. Soil microbial communities are sensitive to resource availability, but the mechanisms of microbial regulation have not been thoroughly investigated. Here, we tested the mechanistic relationships between microbial responses and multiple interacting resources. Methods We examined soil carbon properties, soil microbial community structure and carbon-related functions under nitrogen addition and plant inputs removal (litter removal (NL), root trench and litter removal (NRL)) in a pure Larix principis-rupprechtii plantation in northern China. Results We found that nitrogen addition affected the soil microbial community structure, and that microbial biomass increased significantly once 100 kg ha −1 a −1 of nitrogen was added. The interactions between nitrogen addition and plant inputs removal significantly affected soil bacteria and their enzymatic activities (oxidases). The NL treatment enhanced soil microbial biomass under nitrogen addition. We also found that the biomass of gram-negative bacteria and saprotrophic fungi directly affected the soil microbial functions related to carbon turnover. The biomass of gram-negative bacteria and peroxidase activity were key factors controlling soil carbon dynamics. The interactions between nitrogen addition and plant inputs removal strengthened the correlation between the hydrolases and soil carbon. Conclusions This study showed that nitrogen addition and plant inputs removal could alter soil enzyme activities and further affect soil carbon turnover via microbial regulation. The increase in soil microbial biomass and the microbial regulation of soil carbon both need to be considered when developing effective sustainable forest management practices for northern China. Moreover, further studies are also needed to exactly understand how the complex interaction between the plant and below-ground processes affects the soil microbial community structure.

Vegetation Restoration Alters the Diversity and Community Composition of Soil Nitrogen-Fixing Microorganisms in the Loess Hilly Region of China

Abstract:In this study, we determined the variations in the diversity and structure of nitrogen-fixing microorganisms during afforestation and how this variability correlates with the soil physicochemical factors. Soils were sampled from replicated plots representing three revegetation types (Robinia pseudoacacia L. [Rp], Caragana korshinskii Kom [Ck], and abandoned land) with adjacent farmland as the control at a research site in Ansai District, Yan'an City, Shaanxi Province, China. Using high-throughput sequencing of the nifH rRNA gene, we found that revegetation significantly increased the Shannon diversity index, and for the richness index Chao1 was highest in Ck and have no significant difference in other three lands. From farmland to vegetation types abandoned land, Ck, and Rp, the dominant phylum of nifH microorganisms changed from Actinobacteria to Proteobacteria. Furthormore, variation in the Chao1 richness index was significantly influenced by pH (p = 0.002), soil organic carbon (SOC; p = 0.025) and total nitrogen (TN; p = 0.015). The Shannon diversity index was significantly influenced by SOC, nitrate nitrogen (NN), C to P ratio, TN, AN, N to P ratio, TP, and sand. The changes of nifH microorganism composition at the phylum level were significantly correlated with the soil N to P ratio, which explained 53.1% of the variation among environmental factors (F = 11.8, p = 0.002). The ratio of N to P (interpretation rate 65.2%, F = 18.8, p = 0.002) was also a key factor that influenced the composition at the genus level. Based on the diversity and composition, Ck was a more advantageous recovery method to increase the capacity for N fixation. This study provides a basis for the study of N-fixation characteristics in the Loess Hilly Region.

Abstract:AimIn agricultural basins, riparian buffers maintained along stream channels reduce nitrogen (N) concentrations in agricultural runoff, thereby improving water quality. Investigating the role of riparian vegetation in the related processes will provide insights into the mechanisms by which riparian zones retain soil N.MethodsIn our study, the proportion of plant N uptake, fine root (diameter < 1mm) biomass and fine root morphology at five soil depths (0-15, 15-30, 30-45, 45-60, and 60-75 cm) for Acorus calamus, Canna indica and Phragmites communis were measured in Taihu Lake Basin.ResultsThe soil layer from which the majority of N was absorbed was 0-15 cm (50.8±1.0, 56.0±1.4 and 37.5±3.2% for A. calamus, C. indica, and P. communis, respectively). The N uptake from 45 to 75 cm for P. communis (21.2±2.2%) was significantly higher than A. calamus (14.9±0.7%) and C. indica (9.2±1.5%) (P < 0.05). Our results showed that N uptake was directly proportional to root morphological characteristics such as specific root surface area (SRA, P < 0.05) and specific root length (SRL, P < 0.01), but the relationships varied among species. Changes in environmental factors caused by soil depth strongly influenced some of the root morphological indicators (e.g., fine root biomass, mean diameter (D)).ConclusionsSoil environmental factors and plant root morphology jointly influenced plant N uptake. During vegetation selections of the riparian restoration projects, plants with high SRL or SRA should be given priority due to their expected high capacity in N uptake. But cautions need to be taken as the positive relationships between SRL and SRA and plant N uptake may vary between plant species. Optimal selection of diverse species with complementary nutrient uptake strategies could maximize N uptake at various soil depths and overall N removal from agricultural runoff.

Abstract:Abstract Forests are composed of various plant species, and rhizosphere soil microbes are driven by root exudates. However, the interplay between root exudates, microbial communities in the rhizosphere soil of canopy trees, understory shrubs, grasses, and their responses to nitrogen (N) deposition remains unclear. Pinus tabulaeformis , Rosa xanthina , and Carex lancifolia were used to investigate root exudates, rhizosphere soil microbial communities, and their responses to N application in forest ecosystem. Root exudate abundances of P. tabulaeformis were significantly higher than that of R. xanthina and C. lancifolia , with carbohydrates dominating P. tabulaeformis and R. xanthina root exudates, fatty acids prevailing in C. lancifolia root exudates. Following N application, root exudate abundances of P. tabulaeformis and R. xanthina initially increased before decreasing, whereas those of C. lancifolia decreased. Microbial number of rhizosphere soil of C. lancifolia was higher than that of P. tabulaeformis and R. xanthina , but there was insignificant variation of rhizosphere soil microbial diversity among plant species. N application exerted promotional and inhibitory impacts on bacterial and fungal numbers, respectively, while bacterial and fungal diversities were increased by N application. Overall, N application had negative effects on root exudates of P. tabulaeformis , inhibiting rhizosphere soil microbial populations. N application suppressed rhizosphere soil microbial populations by increasing root exudates of R. xanthina . Conversely, N application elevated nutrient content in the rhizosphere soil of C. lancifolia , reducing root exudates and minimally promoting microbial populations. This study highlights the importance of understory vegetation in shaping soil microbial communities within forests under N deposition.

Abstract:Numerous reforestation projects have been conducted to improve soil fertility in degraded forests, often causing alterations to the soil microbial communities. However, it remains unclear whether microbial functional groups are affected and how these groups correlate with an increase in the nutrient contents during reforestation. We investigated the abundance and composition of free-living nitrogen-fixing microorganisms (diazotrophs) by quantifying and sequencing the marker gene nifH in bulk soils from five reforestation approaches, including legumes and non-legumes, in subtropical China. The relationships between diazotrophic community attributes and soil nitrogen (N) content [NO3 -, NH4 +, and microbial biomass N (MBN)] were examined under various approaches. Abundance of diazotrophs was highest in the native tree plantation (Schima spp. and Michelia macclurei) and Acacia mangium monoculture (AM), and lowest in the Pinus massoniana monoculture. The diazotrophic abundance correlated positively with soil organic matter and water content while there was a negative correlation to pH. The composition of diazotrophic community differed significantly among the five reforestation approaches examined and was closely correlated with variations in soil pH, NH4 + and water content. Diazotrophic community composition was closely related to soil NH4 + content, whereas abundance was not. The AM contained higher NH4 +, NO3 - and MBN contents than the other reforestation approaches, which may be associated with the indicator species of diazotrophs (Actinobacteria, Proteobacteria, and Firmicutes). However, there were more indicator species of Proteobacteria in the mixed Acacia plantation (Acacia mangium and Acacia crassicarpa) than in AM, which might have contributed to the remarkedly lower N content compared to AM. Overall, the soil N content under reforestation appeared to be more related to the composition of diazotroph community than to the abundance of diazotrophs.

Abstract:Nitrogen removal in eutrophic water was studied by monoculture and mixture plants in ecological floating bed system in the greenhouse. At the same time, the numbers of nitrifying and denitrifying bacteria population and the nitrification and denitrification rate in the floating bed plant rhizospere were quantified. The results showed that nitrogen removal in mixture system was outperformed the monoculture and no plant system, and the plant system was outperformed no plant system. There was significant difference between the monoculture and mixture system by the numbers of nitrifying bacteria population and denitrifying bacteria population and denitrification rate. In contrast to watercress system and mixture system, the numbers of nitrifying bacteria population and nitrification rate decreased obviously (P <0.05) in the water dropwort system, the order of the numbers of denitrifying bacteria population and denitrification rate after 15d treatment is: water dropwort system＞ watercress system ＞mixture system ＞no plant system.

Abstract:The root exudates released from macrophytes are critical for pollutant removal and associated microorganisms, but their role in nitrogen removal remains poorly understood in constructed wetlands (CWs). The release of root exudates by Myriophyllum aquaticum was quantified using a hydroponic experiment, and then four treatments with and without environmentally relevant concentrations of root exudates were conducted to investigate the effect of root exudates on nitrogen removal in CWs. The dissolved organic carbon (DOC) release rate of M. aquaticum was 1.1–2.2 mg g−1 h−1, significantly higher than that of Typha orientalis (0.58–0.78 mg g−1 h−1) and Iris pseudacorus (0.13–0.14 mg g−1 h−1). The DOC release rate of M. aquaticum was affected by the ammonium concentration and harvesting frequency. The removal efficiencies of ammonium and total nitrogen were higher in the CWs with plants than in those without plants, confirming the crucial role of plants in plant-microbe interactions and nitrogen removal in CWs. The CWs showed significantly higher ammonium removal efficiencies under the root exudate treatments (mean 90.4% and 69.3%) than the no-root exudate treatments (87.3% and 62.0%), while total nitrogen presented no significant difference. The phylum-level taxonomic classification of microbial sequences detected in the sediments was slightly different between the root exudate and no-root exudate treatments. The results suggested that the root exudate mass naturally released by plants may be insufficient to change the carbon to nitrogen ratios of wastewater. More gene copies were observed in the rhizosphere sediments than the bulk sediments (p < 0.05), indicating that root exudates had a positive effect on microbes distribution within the rhizosphere. Therefore, high levels of root exudates as external carbon sources should be added to enhance nitrogen removal in CWs for carbon-limited wastewater treatment. The present study provided new insights into the role of macrophytes in pollutant removal and management strategy of CWs for wastewater treatment.

Abstract:Wetland plants that cover the wetlands play an important role in reducing pollutants. The aim of this study was to investigate the effect of two plant species on microbial communities and nitrogen-removal genes and to evaluate the contributions of absorbing pollutants by Canna indica (CI) and Cyperus alternifolius (CA) to the removal performance in both a vertical subsurface flow constructed wetland and a horizontal subsurface flow constructed wetland, which were part of a full-scale hybrid constructed wetland system. The microbial assemblages were determined using 16S rRNA high-throughput sequencing. Results showed that the presence of CI and CA positively affected microbial abundance and community in general and which was positive for the total bacteria and ammonia nitrogen removal in the CWs. The higher abundance of Nitrospirae appeared in the non-rhizosphere sediment (NRS) than that in the rhizosphere sediment (RS). More denitrification genes were found in NRS than in RS. The copy numbers of narG, nirS and nosZ genes for CA were higher than those for CI. Wetland plant species can significantly (P < 0.05) affect the distribution of microbial communities in RS. Plant selection is important to promote the development of microbial communities with a more active and diverse catabolic capability and the contribution of plant absorption to the overall removal rate of wetland system can be neglected.
