Abstract:The aqueous two-phase extraction process (APTE) for polysaccharides from the fruits of Ilex cornuta (Lindl. et Paxt.) (PFIC) was studied, and its monosaccharide composition and antioxidant activities were analyzed. After screening and selection of isopropanol/ammonium sulfate as the aqueous two-phase system, various parameters of crude extract amount, ultrasonic time, extraction temperature and extraction time on the extraction process was investigated in detail and optimized by response surface methodology (RSM). The obtained PFIC was characterized by FT-IR and its monosaccharide composition was analyzed by GC-MS. Furthermore, the antioxidant activities of PFIC were evaluated by measuring the scavenging rates of DPPH radical, hydroxyl radical and superoxide anion radical. Under the optimal conditions in which crude extract amount was 2.23 g, ultrasonic time was 17.5 min, extraction temperature was 69 â„ƒ and the extraction time was 1.55 h, the yield of polysaccharides reached 50.87% which was close to the predicted value of 50.78%. FT-IR analysis confirmed that the extract was polysaccharide, and GC-MS analysis indicated that the monosaccharide components of the polysaccharides were rhamnose, arabinose, xylose, mannose, glucose and galactose. Antioxidant results showed that PFIC showed strong scavenging ability to DPPH radical, fine scavenging ability to hydroxyl radical and certain scavenging ability to superoxide anion radical. The ultrasonic-assisted APTE process for PFIC optimized by RSM is reasonable and reliable, and the obtained PFIC has the potential to be utilized as natural antioxidants.
Enzyme-Assisted Extraction of Polysaccharides from Dendrobium Chrysotoxum and Its Functional Properties and Immunomodulatory Activity

Abstract:Dendrobium chrysotoxum has long been used as a health-promoting and therapeutic tonic in China. The polysaccharides from D. chrysotoxum (DCP) are considered to play an important role in its medical effects. The present study aimed at optimizing the technology for enzyme-assisted extraction (EAE) of DCP and at investigating the physicochemical characteristics and functional properties of DCP-E obtained by EAE and DCP-H obtained by hot water extraction (HVVE). Based on single factor tests and orthogonal experiments of extraction pH, extraction temperature, cellulase amount and extraction time, the optimum conditions for CP-E were extraction pH value of 5.5, extraction temperature of 40 degrees C, cellulase amount of 10 g/L, extraction time of 3.0 h and solid/liquid ratio of 1:25. Under these conditions, the yield of DCP-E was 8.41 g/100 g dry weight, which was 1.25 times of that of DCP-H. As compared to DCP-H, DCP-E displayed an increased purity, a decreased molecular weight and relative viscosity as well as a changed monosaccharide compositions. Foam stabilization and splenic cell proliferation experiments showed that DCP-E had higher cell proliferation rate while DCP-H had better foam stabilization activity. These results indicate that different extraction methods would influence the physicochemical characteristics and subsequently functional properties of polysaccharides, suggesting that the suitable extraction method should be selected according to the application intention of polysaccharides. Structural Characterization and Immunomodulatory Activity of Polysaccharides from Polygonatum sibiricum Prepared with Deep Eutectic Solvents

Abstract:Deep eutectic solvents (DES), as a new, efficient, and green solvents, have attracted more attention in recent years. This study was performed to extract Polygonatum sibiricum polysaccharides using DES (DPSP). Subsequently, DPSP was subjected to DEAE-Sepharose Fast Flow and Sephadex G-100 column for further purification (DPSP-3). Structural characterization and immunomodulatory activity in RAW264.7 cells were performed. Results showed that the yield of DPSP was (15.62 ± 0.71)%, and its yield was 1.53 times that of the polysaccharides extracted using the pharmacopoeia method (PSP). DPSP-3 had a smaller molecular weight (3.2 × 106 Da) and a higher ratio of galactose (65.75%) and mannose (19.76%) as compared to PSP. The scanning electron microscope (SEM) showed that DPSP-3 had curly shape, lamellar surface, and a few spherical edges, which were significantly different from PSP and DPSP. Both PSP and DPSP-3 could significantly improve phagotrophic capacity and increase the release of ROS, NO, IL-6, and TNF-α in RAW 264.7 cells, and DPSP-3 had better immunomodulatory activity. In summary, compared to the pharmacopoeia method, polysaccharides prepared using the DES extraction method showed higher yield and improved immunomodulatory activity.

Abstract:The ultrasonic-assisted deep eutectic solvent method was used to extract the polysaccharides of Pericarpium Citri Reticulatae (PCRP), and the ultrasound-assisted DES extraction process was optimized by Box-Behnken response surface test using the extraction rate of the PCRP as an index; the in vitro activities of purified the PCRP(PCRPs-1) were investigated by determining the scavenging rate of DPPH• and ABTS• + as well as by enzyme inhibition assay. The monosaccharide composition was analyzed by HPLC. The best process conditions for response surface optimization were a material-liquid ratio of 1:37 g/mL, water content of 44%, time of 89 min, and power of 320 W. The polysaccharide extraction rate was measured to be 5.41%, which was well optimized when compared with that of the ordinary aqueous extraction method of 3.92%. By α-glucosidase and α-amylase inhibition activity test, it showed that the PCRPs-1 had hypoglycemic activity. The DPPH radical scavenging activity test and ABTS + scavenging activity test indicated that the PCRPs-1 had good biological activity. Analysis of the monosaccharide fractions showed that the PCRPs-1 consisted of mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, xylose, and arabinose, with molar ratios of 1:39.24:4.41:8.91:7.83:86.00:1.02:9.17. The activity studies showed that PCRPs-1 possessed certain hypoglycaemic and antioxidant activities.

Abstract:A green extraction method was developed using deep eutectic solvent extraction for the polysaccharide from Acanthopanax senticosus (A. senticosus). Among the eight types of DES prepared, the DES with a ratio of 1:4 L-malic acid to L-proline was found to be a suitable extraction solvent based on the extraction efficiency. The extraction parameters were optimized by Plackett–Burman and response surface methodology (RSM). The best extraction conditions were found for L-malic acid. Under the conditions of an L-malic acid/L-proline ratio of 1:4, ultrasonic power of 240 W, material–liquid ratio of 31.068 g/mL, water content of 32.364%, extraction time of 129.119 min, and extraction temperature of 60 °C, the extraction rate of A. senticosus polysaccharides was 35.452 ± 0.388 mg-g−1. This rate was higher than that of polysaccharides obtained by hot water extraction (13.652 ± 0.09 mg-g−1). The experimental results were best fitted by the quasi-secondary kinetic model when compared to two other kinetic models. Electron microscopic observations showed that DESs were more destructive to plant cells. The polysaccharide extracted from DESs had more monosaccharide components, a lower molecular weight, a higher antioxidant capacity, and superior anti-glycation activity compared to polysaccharides extracted from water (ASPS-PW). This study demonstrates the effectiveness of DESs in obtaining polysaccharides from A. senticosus.

Abstract:Deep eutectic solvent (DES) and hot-water extraction (HWE) methods were utilized to extract polysaccharides from Polygonatum sibiricum, referred to as DPsP and WPsP, respectively. The extracted polysaccharides were purified using the Superdex-200 dextran gel purification system, resulting in three components for each type of polysaccharide. The structures of these components were characterized. The molecular weight analysis revealed that DPsP components had slightly larger molecular weights compared with WPsP, with DPsP-A showing a slightly higher dispersity index and broader molecular weight distribution. The main monosaccharide components of both DPsP and WPsP were mannose and glucose, while DPsP exhibited a slightly greater variety of sugar components compared with WPsP. FTIR analysis demonstrated characteristic polysaccharide absorption peaks in all six PSP components, with a predominance of acidic pyranose sugars. NMR analysis revealed the presence of pyranose sugars, including rhamnose and sugar aldehyde acids, in both DPsP-B and WPsP-A. DPsP-B primarily exhibited β-type glycosidic linkages, while WPsP-A predominantly displayed α-type glycosidic linkages, with a smaller fraction being β-type. These findings indicated differences in monosaccharide composition and structure between PSPs extracted using different methods. Overall, this study provided experimental evidence for future research on the structure–function relationship of PSPs.

Abstract:The effects of extraction conditions on the yield of polysaccharides from the fruit of Chaenomeles sinensis (FCS) using a hot compressed water method were investigated. The results showed that an appropriately high temperature (150 °C) and a moderate extraction time (45 min) at a material to water ratio of 1 to 10 g/mL led to a high yield of alcohol precipitation polysaccharide (PA). The purified polysaccharides (CSP-1, CSP-2, and CSP-3) were successfully obtained using a DEAE-52 chromatographic column. Chemical analysis showed that CSP-2 and CSP-3 were homogenous and exhibited characteristics of esterified pectins, whereas CSP-2 mainly consisted of galacturonic acid (GalA), galactose (Gal), arabinose (Ara), rhamnose (Rha), and mannose (Man) with an average molecular weight of 59.1 kDa. Furthermore, CSP-1 possessed stronger antioxidant ability according to DPPH scavenging and reducing power compared with CSP-2 and CSP-3. However, it was weaker with respect to OH scavenging. The technical data presented in this study could help the industry make use of polysaccharides from FCS as a source of pectin for a range of pharmaceutical, culinary, and cosmetic products.
Optimization of ultrasonic-assisted deep eutectic solvent for the extraction of polysaccharides from Indocalamus tessellatus leaves and their biological studies

Abstract:To the valorization of Indocalamus tessellatus(I.tessellatus) leaves for the functional food industry, the process of ultrasonic-assisted deep eutectic solvent (DES) extraction of I.tessellatus leaves polysaccharides (U-DES-ICP) was optimized by response surface methodology. In comparison to ultrasonic extracted polysaccharides (UICP) and water extracted polysaccharides (WICP), their physicochemical properties were analyzed by HPLC, HPGPC-MALLS, and FTIR, and the structure morphology was observed by SEM. Their antioxidant activities were evaluated based on DPPH and ABTS + assay as well. The results showed that the best DES system for U-DES-ICP was a mixture of choline chloride and malonic acid in the molar ratio of 1:4 (10% water content). The highest yield of U-DES-ICP (0.6812%) was obtained by following optimized conditions: 60 min of sonication time, 17 mL/g of liquid-solid ratio, 500W of sonication power, and 63.5 °C of sonication temperature, which was significantly higher than that of the conventional methods. The structural analysis indicated that U-DES-ICP had more uronic acid with higher molecular weight compared with UICP and WICP, the relative content of uronic acid of U-DES-ICP reached 8.364%, which was twice as much as the other two polysaccharides; its molecular weight reached up to 6.201 × 103 KD, and the highest values of molecular weight of UICP and WICP were 1.684 × 103 KD and 995.4 KD, U-DES-ICP which were confirmed by SEM. The antioxidant assays reflected that U-DES-ICP possessed stronger scavenging capacities both on DPPH and ABTS + radicals with IC50 (0.48 mg/mL and 0.61 mg/mL) than the other two biomacromolecules. IC50s for DPPH and ABTS radical scavenging by UICP were 0.51 and 0.73 mg/mL, while 0.82 and 0.94 mg/mL were IC50s for both radical scavenging by WICP, respectively. Therefore, this study evidenced that a novel DES system could boost the extraction of I.tessellatus leaves polysaccharides, which produced a higher yield of product with stronger biological activities compared with traditional techniques.

Abstract:Plantaginis Semen (PS) are the dried mature seeds of Plantago asiatica L. or Plantagodepressa Willd. in the Plantago family. Its polysaccharides are important components of PS . Response surface methodology was used to optimize the ultrasonic-assisted deep eutectic solvent (DES) extraction process of PS polysaccharides (PSP). The results showed that the optimal extraction parameters were a solid-liquid ratio of 1:35 g/mL, an extraction time of 73 min, and a molar ratio of 2:1. The yield of PSP was 0.64% and 1.20% by water immersion and ultrasonic water extraction, respectively, indicating that the DES extraction method (2.21 ± 0.06%) is superior to these two methods, and the optimization effect is good. Through the α-glucosidase and α-amylase inhibition activities experiment, it was found that the IC 50 values of PSPs-1 were 1122 and 220.5 μg/mL. DPPH·and ABTS + scavenging activity experiments showed that the IC 50 values of PSPs-1 were 19.2 and 4.3 μg/mL, respectively. Its molar ratio of monosaccharide composition is rhamnose: galactose: galacturonic acid: glucose: glucuronic acid: arabinose: mannose: xylose = 33.6:13.3:6.5:3:2.6:2:1.4:1. Therefore, this study can provide an experimental basis for the establishment of an industrialized production process of polysaccharides and the study of their biological activities.

Abstract:Ultrasound extraction (UE) enhanced with deep eutectic solvent (DES) was used to extract Lentinus edodes polysaccharides. Box‐Behnken design (BBD) was applied to investigate the influences of water content (10‐90%), solid‐liquid solvent (1:10‐1:50 g/mL), time (4‐12 min), temperature (40‐80 °C) and ultrasonic power (100‐500 W) on the yield of Lentinus edodes polysaccharides. The optimal extraction conditions were ultrasonic power of 300 W, extraction time of 8 min, water content of 80%, a solid‐liquid ratio of 1:30 g/mL and a temperature of 60 °C, respectively. The highest extraction yield of Lentinus edodes polysaccharide was 10.17% under optimal conditions. The results of FT‐IR, SEM, and monosaccharide composition confirmed that the extracts possessed the characteristics of polysaccharides. In addition, the polysaccharides obtained with the UE enhanced with DES method exhibited higher antioxidant activities than the polysaccharides extracted with the UE method and HWE method. This extraction method can further expand the production efficiency and structural diversity of Lentinus edodes polysaccharides and meet the supply and demand relationship. It can be foreseen that this method can be applied to the extraction of more active substances.

Abstract:In this study, a high-efficiency and non-pollution extraction procedure, ultrasound-assisted technique with deep eutectic solvents (DESs), was applied for extraction of polysaccharides from Morchella importuna (MIP-D). The results exhibited that the system of DES was: mole ratio between choline chloride and oxalic acid of 2:1, water content of 90% (v/v), and the optimal extraction parameters were as follows: extraction time of 31.2 min, extraction temperature of 62.1 ° C, and the liquid–solid ratio of 32.5:1 (v/w). Under these extraction parameters, the extraction yield of MIP-D was 4.5 times higher than hot water extraction (HWE) method and had higher carbohydrate (85.27%) and sulfate contents (34.16%). Moreover, high-performance liquid chromatography (HPLC) and Fourier-transform IR (FTIR) spectrum analysis indicated that MIP-D was comprised of glucosamine, galactose, glucose, and mannose, with molar ratios of 0.39:1.88:3.82:3.91, which contained the pyranose ring skeleton. High-performance gel permeation chromatography (HPGPC) analysis revealed that MIP-D showed three fractions with molecular weights of 2.6 × 10 6 , 7.3 × 10 4 , and 3.7 × 10 3 Da, which were lower than those of polysaccharides extracted by HWE. In-vitro tests proved that MIP-D possessed excellent antioxidant and inhibited α-amylase and α-glucosidase inhibitory activities. Therefore, DESs (choline chloride-oxalic acid) as a high-efficiency and non-pollution solvent alternative can be applied to the separation of bioactive polysaccharides from Morchella importuna ( M. importuna ).

Abstract:This study aimed to isolate polysaccharides from Penthorum chinense Pursh and evaluate their rheological characteristics, physicochemical properties, and antioxidant activity. The optimal conditions for the maximal extraction yield of Penthorum chinense Pursh polysaccharides (4.05 ± 0.12%) were determined by employing a single-factor test and response surface methodology which included an extraction time of 3 h, a liquid–solid ratio of 20 mL/g, and three separate extraction times. The rheological experiments showcased that the P. chinense polysaccharides exhibited typical shear-thinning behavior, with their apparent viscosity being influenced by various parameters such as concentration, pH, temperature, salt content, and freeze–thaw. The purified polysaccharides (PCP-100), having an average molecular weight of 1.46 × 106 Da, mainly consisted of glucose (18.99%), arabinose (22.87%), galactose (26.72%), and galacturonic acid (21.89%). Furthermore, the PCP-100 exhibited high thermal stability and displayed an irregular sheet-like morphology. Its superior reducing power and free radical scavenging ability implied its significant antioxidant activity in vitro. Collectively, these findings provide important insights for the future application of P. chinense polysaccharides in the food industry.

Attachment , Entrepreneurial Role Stress and Quality of Life in Metal Shop Owners

Abstract:Attachment is a deep and enduring bond that is emotional and it connects one person to another (Anisworth, 1973). Entrepreneurs are rational decision makers who assume risk and provide management for the firm (Kilby, 1971). Role stress can be defined as a discrepancy between expectations and behaviour; when such discrepancies exist, the entrepreneur will be exposed to role stress (Wincent & Örtqvist, 2009). Quality of Life is defined by as appraisal of and satisfaction with their current level of functioning as compared to what they perceive to be possible or ideal (Lehto, Ojanen & Kellokumpu-Lehtinen, 2005).The objective of this study was to find the relationship between attachment, entrepreneurial role stress, and quality of life among father and son metal shop owners and to observe whether fathers and sons differ with respect to these variables. A non-probability purposive sampling method was employed to collect data from 120 small scale entrepreneurs of metal supplying stockiest (60 fathers and 60 sons).The Inventory of Parent and Peer Attachment (Armsden, 1987), Entrepreneurial Role Stress Scale (Pareek, 1997), and WHO Quality of life-BREF questionnaire (WHOQOL-BREF,WHO, 1997) were administered to the participants. Results showed that sons experience a higher financial and health satisfaction than the fathers. Significant correlations were found between attachment, entrepreneurial role stress and quality of life in fathers and sons. These findings illuminate the role of family business succession and communication among fathers and sons in enhancing the level of satisfaction in the coming generations.

Abstract:Dendrobium huoshanense is a famous edible and medicinal herb, and polysaccharides are the main bioactive component in it. In this study, response surface methodology (RSM) combined with a Box–Behnken design (BBD) was used to optimize the enzyme-assisted extraction (EAE), ultrasound–microwave–assisted extraction (UMAE), and hot water extraction (HWE) conditions and obtain the polysaccharides named DHP-E, DHP-UM, and DHP-H. The effects of different extraction methods on the physicochemical properties, structure characteristics, and bioactivity of polysaccharides were compared. The differential thermogravimetric curves indicated that DHP-E showed a broader temperature range during thermal degradation compared with DHP-UM and DHP-H. The SEM results showed that DHP-E displayed an irregular granular structure, but DHP-UM and DHP-H were sponge-like. The results of absolute molecular weight indicated that polysaccharides with higher molecular weight detected in DHP-H and DHP-UM did not appear in DHP-E due to enzymatic degradation. The monosaccharide composition showed that DHPs were all composed of Man, Glc, and Gal but with different proportions. Finally, the glycosidic bond types, which have a significant effect on bioactivity, were decoded with methylation analysis. The results showed that DHPs contained four glycosidic bond types, including Glcp-(1→, →4)-Manp-(1→, →4)-Glcp-(1→, and →4,6)-Manp-(1→ with different ratios. Furthermore, DHP-E exhibited better DPPH and ABTS radical scavenging activities. These findings could provide scientific foundations for selecting appropriate extraction methods to obtain desired bioactivities for applications in the pharmaceutical and functional food industries.

Polysaccharides extraction from Ganoderma lucidum using a ternary deep eutectic solvents of choline chloride/guaiacol/lactic acid

Abstract:In this study, a purposefully formulated ternary deep eutectic solvents (DESs), consisting of choline chloride, guaiacol, and lactic acid in a molar ratio of 1:1:1, was synthesized for the extraction of polysaccharides from Ganoderma lucidum. The physicochemical properties of the synthesized DESs, including viscosity, density, pH, and hydrogen bonds, were comprehensively examined. Verification of the formation of the ternary DESs was accomplished through Fourier transform infrared and Nuclear magnetic resonance spectroscopies. Subsequently, response surface methodology was applied to optimize crucial parameters for polysaccharide extraction using DESs, resulting in a maximal extraction yield of 94.72 mg/g under the optimized conditions. Cyclic experiments demonstrated the commendable cyclic stability of the DESs, with a recovery rate exceeding 88 %. Furthermore, experiments on monosaccharide composition, molecular weight, and antioxidant activity of the isolated polysaccharides were conducted. Density functional theory was employed to gain insights into the molecular mechanism of polysaccharide extraction by DESs. The findings revealed a triple hydrogen bond interaction and a high binding energy (65.29 kcal/mol) between the DESs and glucose, highlighting their significant contribution to the high extraction effectiveness. This molecular-level understanding underscores the inherent superiority of DESs in the polysaccharide extraction processes, providing valuable insights for future applications in this field.

Abstract:Abstract Penthorum chinense Pursh is a well-known traditional Chinese medicine, however, little attention has been paid to the polysaccharides of P. chinense (PCP). Therefore, it is great significance to investigate the characteristics and activities of PCP. In this study, response surface methodology was applied to optimize the extraction parameters of PCP. Then, two polysaccharides fractions (PCP-1 and PCP-2) were purified from PCP by using DEAE-52 chromatography, and there preliminary chemical properties and in vitro antioxidant activities were investigated. Results revealed that the optimum extraction conditions of PCP were identified as follow: ratio of water to raw material 20.6 mL/g; extraction time 3.5 h and extraction temperature 85°C. Based on these conditions, the maximum yield of PCP was 3.12% ± 0.19%. Purified fractions PCP-1 and PCP-2 were all acidic heteropolysaccharides, and PCP-1 mainly consisted of galactose and arabinose while PCP-2 mainly consisted of rhamnose, galacturonic acid, galactose and arabinose. Moreover, PCP-2 exhibited stronger scavenging activities against DPPH radical, hydroxyl radical and superoxide anion radical and chelating activity on Fe 2+ in vitro .

Abstract:In the present study, subcritical water extraction (SWE) assisted with deep eutectic solvent (DES) is used to extract Lentinus edodes polysaccharides (LEP). In addition, the antioxidant activity of the polysaccharide samples was also investigated. Based on a single factor test and response surface test, the optimal extraction factors were a liquid–solid solvent of 40:1 mL/g, extraction temperature of 147.23 °C, water content of 39.76% and extraction time of 17.58 min. Under these extraction conditions, the yield of LEP was 6.26 ± 0.08%. Compared with the SWE and hot water extraction (HWE), it improved by 19.24% and 17.01%, respectively. In addition, the results of monosaccharide composition, molecular weight, FT-IR, UV and SEM confirmed that the extracts had the features of polysaccharides. Interestingly, the polysaccharides obtained with the SWE assisted with the DES procedure showed a higher DPPH scavenging activity, hydroxyl radical scavenging activity and hydrogen peroxide scavenging activity, which indicated that the polysaccharides with this method had a stronger antioxidant activity. These findings demonstrated that the SWE-assisted DES is a strong method to obtain polysaccharides from Lentinus edodes for food, biopharmaceutical and other industrial production.

Polysaccharides from Radix Peucedani: Extraction, Structural Characterization and Antioxidant Activity

Abstract:In this study, an ultrasound-assisted green extraction method was applied for the extraction of polysaccharides from Radix Peucedani based on deep eutectic solvents (DESs), and the result showed that a DES system composed of betaine and 1,2-propylene glycol with a molar ratio of 1:2 possessed the optimal extraction efficiency for polysaccharides. Single-factor and Box–Behnken designs were used to determine the optimum extraction conditions for the maximum yields of polysaccharides from Radix Peucedani by using DESs. The maximum yields of polysaccharides attained 11.372% within a DES water content of 19%, an extraction time of 36 min, an extraction temperature of 54 °C, a solid–liquid ratio of 1:30 and an ultrasonic irradiation power of 420 W. The physicochemical properties of polysaccharides were analyzed using ICS and FT-IR, and the structure morphology was observed by SEM. The polysaccharides extracted from Radix Peucedani exhibited general antioxidant activities in vitro including DPPH, Hydroxyl and ABTS+ radical-scavenging activity. The antioxidant mechanism of Radix Peucedani polysaccharides was investigated using network pharmacology and molecular docking methods. The result showed that the high binding activity of glucose and IL1B, galactose and CASP3 was recognized as a potential mechanism for the antioxidant effects of Radix Peucedani polysaccharides.

Abstract:Polygonatum sibiricum polysaccharides (PSPs) have important biological functions, such as antioxidation, immunomodulatory, and hypolipidemic functions. Different extraction methods have effects on their structures and activities. In this study, six extraction methods, including hot water extraction (HWE), alkali extraction (AAE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), microwave-assisted extraction (MAE), and freeze–thaw-assisted extraction (FAE) were used to extract PSPs, and their structure–activity relationships were analyzed. The results showed that all six PSPs had similar functional group compositions, thermal stability, and glycosidic bond compositions. PSP-As (PSPs extracted by AAE) exhibited better rheological properties due to their higher molecular weight (Mw). PSP-Es (PSPs extracted by EAE) and PSP-Fs (PSPs extracted by FAE) had better lipid-lowering activity due to their lower Mw. PSP-Es and PSP-Ms (PSPs extracted by MAE), which do not contain uronic acid and have a moderate Mw, had better 1,1-diphenyl-2-picrylhydrazyl (DPPH)-radical-scavenging activity. On the contrary, PSP-Hs (PSPs extracted by HWE) and PSP-Fs, with the Mw of uronic acid, had the best OH-radical-scavenging activity. The high-Mw PSP-As had the best Fe2+-chelating ability. In addition, mannose (Man) may play an important role in the immunomodulatory activity. These results indicate that different extraction methods affect the structure and biological activity of polysaccharides to varying degrees, and these results are helpful for understanding the structure–activity relationship of PSPs.

Abstract:Deep eutectic solvent as a new green and safe solvent system has attracted more and more attention in recent years. In this study, three deep eutectic solvents (DES) were combined with ultrasound irradiation to extract tea polysaccharides (TPs) from Anji white tea, which was compared with conventional hot water extraction (HW). The physicochemical, structural, and biological properties of TPs extracted by ultrasound-assisted DES and hot water (HWP) were further investigated. Results showed that the DES system composed of choline chloride and 1,6-hexanediol (CH) with the molar ratio of 1:2 exhibited the optimal extraction yield (19.18%) and in vitro antioxidant activities for TPs (CHP). Furthermore, compared to the HWP, the CHP had a higher extraction yield and total carbohydrate content and a lower molecular weight. Monosaccharide composition analysis displayed that the molecular structure of CHP exhibited more arabinose but less glucose, mannose, galacturonic acid, and glucuronic acid than HWP. Little difference was observed in the preliminary structural characteristics between HWP and CHP from Fourier transform infrared analysis. Besides, CHP possessed better α-glucosidase inhibitory and hypoglycemic activity in L6 cells than HWP. Therefore, the ultrasound-assisted DES extraction method can be a promising strategy for extracting TPs with excellent bioactivities for future applications in functional foods.
