Abstract:A method for remote radical C-H alkynylation and amination of diverse aliphatic alcohols has been developed. The reaction features a copper nucleophile complex formed in situ as a photocatalyst, which reduces the silicon-tethered aliphatic iodide to an alkyl radical to initiate 1,n-hydrogen atom transfer. Unactivated secondary and tertiary C-H bonds at β, γ, and δ positions can be functionalized in a predictable manner.

Abstract:Radical translocation processes triggered by nitrogen-centered radicals (NCRs), such as 1,5-hydrogen atom transfers (1,5-HAT), demonstrated by the well-established Hofmann-Löffler-Freytag (HLF) reaction, provide an attractive approach for the controllable and selective functionalization of remote inert C(sp 3 )–H bonds. Here we report an amidyl radical-triggered site-selective remote C(sp 3 )–H heteroarylation of amides under organic photoredox conditions. This approach provides a mild and highly regioselective reaction affording remote C(sp 3 )–H heteroarylated amides at room temperature under transition-metal free, weakly basic, and redox-neutral conditions. Non-prefunctionalized heteroarenes, such as purines, thiazolopyridines, benzoxazole, benzothiazoles, benzothiophene, benzofuran, thiazoles and quinoxalines, can be alkylated directly. Sequential and orthogonal C–H functionalization of different heteroarenes by taking advantage pH value or polarity of radicals has also been achieved. DFT calculations explain and can predict the site-selectivity and reactivity of this reaction. This strategy expands the scope of the Minisci reaction and serves as its alternative and potential complement.

Abstract:Herein, we developed a photoinduced remote selective C(sp3)−H alkylation of protected amines in the presence of cesium formate. The protected amines were synthesized from commercially available inexpensive 2‐iodo benzoyl chloride. Under mild reaction conditions, the in situ generated aryl radical is converted to alpha‐amidoalkyl radical via [1,5]‐HAT process, which combines with different Michael acceptors and affords corresponding alkylation products in good yields. Preliminary mechanistic studies revealed that formate anion acts as a source of carbon dioxide radical anion (CO2•−) and hydrogen atom donor was directly confirmed by isotope‐labeling studies.

Abstract:A deoxygenative radical alkylation of aromatic alkynes with alkyl aldehydes for the preparation of allylarenes has been successfully achieved. This transformation is accomplished through the reaction of alkyl aldehydes with alkynes in the presence of dipropylamine and Hantzsch ester catalyzed by a P/N heteroleptic Cu(I)-based photosensitizer under photoredox catalysis conditions. Preliminary mechanistic studies reveal that this aldehyde-alkyne coupling process comprises a regioselective radical addition of in situ-generated alkyl-substituted alpha-amino radicals to alkynes with subsequent 1,5-proton transfer, C-N bond cleavage, and concomitant isomerization of the resulting allyl radical species. Thus, in net result, dipropylamine serves as a traceless linker agent for the deoxygenative radical cross-coupling of alkyl aldehydes with alkynes under photoredox catalysis reaction conditions.

Abstract:Herein, a highly regioselective alkylation of propargylic carbonates for trisubstituted allenes with alkyl N-hydroxyphthalimide (NHP) esters was developed via a nickel-mediated radical cross-electrophile coupling process. Electron donor-acceptor (EDA) complex photoactivation strategies have emerged as versatile way for alkyl radical. Abstract:Direct acylation of amides at C(sp3)–H sites from aldehydes is one of the most convenient methods to access highly valuable keto-amides, yet it still remains challenging and less development. Herein, we reported a photoredox cooperative NHC/Pd-catalyzed method that enables C(sp3)–H acylation for quick construction of keto-amide derivatives from the readily available amides and aldehydes. This work demonstrates the generation of ketyl radicals from aldehydes under NHC/Pd catalysis and alkyl radicals via 1,5-hydrogen atom transfer from aryl radicals under photoredox/Pd catalysis, thus providing two- and three-component reactions for the synthesis of α- and γ-keto-amides through radical−radical coupling reactions. This work features with good compatibility with a large variety of functional groups as demonstrated by more than 75 examples, including complex bioactive compounds. Mechanistic studies including several control reactions shed light on the mechanism of this photoredox cooperative NHC/Pd-catalyzed reaction.

Abstract:An efficient, catalyst- and additive-free, visible-light-driven radical C3-H alkylation of quinoxalin-2(1H)-one derivatives has been developed. This reaction utilizes alkyl-NHP-esters as an alkyl radical donor and quinoxalin-2(1H)-one derivatives as an alkyl radical acceptor. The operationally simple protocol works under mild reaction conditions and tolerates a variety of functional groups. Furthermore, the synthetic utility of the methodology was successfully implemented for synthesizing biologically relevant C3-alkyl substituted quinoxalin-2(1H)-one derivatives.

Abstract:The site-selective functionalization of unactivated aliphatic amines is an attractive and challenging synthetic approach. We herein report a general strategy for the remote site-selective functionalization of unactivated C(sp3)–H bonds in amides by photogenerated amidyl radicals to form gem-difluoroalkenes with trifluoromethyl-substituted alkenes. The site selectivity is controlled by a 1,5-hydrogen atom transfer (HAT) process of the amide. Abstract:We reported the visible-light-mediated photoredox-catalyzed oxidative radical-polar crossover and 1,5-hydrogen atom transfer combined site-selective remote C(sp3)-N cross-coupling alkylamination of alkenes. Abstract:Alcohols play a pervasive role in the realm of chemistry, constituting inherent functional groups in numerous natural products and bioactive molecules. In this study, we introduce an efficient radical approach for the activation and transformation of unstrained aliphatic alcohols through β-scission of alkoxyl radicals under mild conditions, avoiding the need for metals, photocatalysts, and other additives. Additionally, we showcase the utility of this method in three-component cascade radical coupling reactions involving 1,3-butadiene or 1,3-enyne, respectively. Mechanistic studies reveal the excited state of the pyrazolone enolate as a key intermediate with dual functions as a radical precursor and an efficient reductant, leading to the formation of a radical pair in the reaction.

Abstract:The catalytic three‐component radical carboamination of alkenes has recently emerged as an alternative and robust platform for the rapid construction of diverse and valuable amines. Despite great advances in this field, new methods that enable highly selective access to new chemical space surrounding the amine functional groups are still in high demand. Herein, we report a generally applicable visible light‐induced copper‐catalyzed three‐component radical cyanoalkylamination reaction of alkenes, oxime carbonates, and benzoyloxycarbamates. This protocol demonstrates high chemo‐selectivity, broad substrate scope, and good functional group tolerance, providing access to a variety of cyanoalkylated aliphatic amines with good yields.

Abstract:We report a general photocatalytic protocol for the remote C(sp3)-H bond amination of free aliphatic alcohols. The electron transfer between the abundant and inexpensive catalyst FeCl3 and simple alkanols under blue LED irradiation enables the alkoxy radical formation under mild redox-neutral conditions, with no need for additional oxidant and prefunctionalization. The subsequent selective 1,5-hydrogen atom transfer (HAT) and amination provide a simple and efficient way to access molecular complexity from readily available and bulk alcohols.

Abstract:The photoredox electron donor–acceptor (EDA) complex-mediated radical coupling reaction has gained prominence in the field of organic synthesis, finding widespread application in two-component coupling reactions. However, EDA complex-promoted multi-component reactions are not well developed with only a limited number of examples have been reported. Herein, we report a photoinduced and EDA complex-promoted highly chemoselective three-component radical arylalkylation of [1.1.1]propellane, which allows the direct functionalization of C(sp3)–H with bicyclo[1.1.1]pentanes (BCP)-aryl groups under mild conditions. A variety of unnatural α-amino acids, featuring structurally diversified 1,3-disubstituted BCP moieties, were synthesized in a single-step process. Notably, leveraging the high tension release of [1.1.1]propellane, the highly unstable transient aryl radical undergoes rapid conversion into a relatively stable tertiary alkyl transient radical, and consequently, the competing side-reaction of two-component coupling was entirely suppressed. The strategic use of this transient radical conversion approach would be useful for the design of diverse EDA complex-mediated multi-component reactions. It is noteworthy that the highly chemoselective late-stage incorporation of the 1,3-disubstituted BCP pharmacophores into peptides was achieved both in liquid-phase and solid-phase reactions. This advancement is anticipated to have significant application potential in the future development of peptide drugs.
