Abstract:Research indicates general willingness to drink (i.e., "How willing are you to drink tonight?") fluctuates day-to-day and is associated with daily-level drinking. However, it is unknown whether willingness to engage in specific alcohol-related behaviors are associated with actual engagement in those behaviors above and beyond general willingness to drink at the daily-level. The present study examined whether daily-level willingness to engage in specific behaviors (i.e., pre-gaming, blacking out, hooking up) predicted engaging in those respective behaviors on drinking days above and beyond one's general willingness to drink. Participants included adolescents and young adults who were part of a longitudinal ecological momentary assessment (EMA) study on cognitions and alcohol use. Participants (N = 675; 43 % male) who reported drinking on weekend days (i.e., Friday and Saturday [N = 3,727 days]), were included. The study involved a 3-week EMA burst design with bursts occurring quarterly over 12 months. Multilevel logistic regressions indicated on drinking days when participants reported being more willing than their own average to pre-game (OR = 1.77, p <.001), black out (OR = 1.46, p <.05), or drink before hooking up (OR = 1.68, p <.001), they were more likely to pregame, black out, and hook up, respectively, whereas general willingness to drink was not associated with any outcomes at the daily level. Results suggest willingness to engage in specific behaviors may be essential to target in prevention programming in comparison to general willingness to drink when aiming to reduce specific risk behaviors.

Correlates and motives of pre-drinking with intoxication and harm around licensed venues in two cities

Abstract:Introduction and aims: The study investigates the prevalence of pre-drinking culture in the night-time economy (NTE) and its impact upon intoxication and alcohol-related harm and violence experienced by patrons. Design and methods: Cross-sectional surveys were conducted in and around licensed venues in Newcastle (NSW) and Geelong (Victoria) during peak trading hours (typically 9pm-1am). Participants completed a five minute structured interview which targeted: demographics, past and planned movements on the survey night, safety/experience of harm, and patron intoxication. 3949 people agreed to be interviewed, a response rate of 90.7%. Around half (54.9%) of interviewees were male and mean age was 24.4 years (SD = 5.8). Results: 66.8% of participants reported pre-drinking prior to attending licensed venues. On a 1-10 scale measuring self-rated intoxication, pre-drinkers scored significantly higher compared to non pre-drinkers (P < 0.001). Compared to non-pre-drinkers, patrons who had consumed 6-10 standard pre-drinks were 1.5 times more likely to be involved in a violent incident in the past 12 months (OR = 1.50, 95%CI 1.03-2.19, P = 0.037) increasing to 1.8 times more likely for patrons who had 11-15 drinks (OR = 1.80, 95%CI 1.04-3.11 P = .036). Pre-drinking was also associated with both self-rated and observer-rated intoxication, as well as increased probability of illicit drug use. Amongst pre-drinkers, price was the most commonly reported motive for pre-drinking (51.8%). Discussion and conclusions: 'Pre-drinking' was normal behaviour in the current sample and contributes significantly to the burden of harm and intoxication in the NTE. Price disparity between packaged vs. venue liquor is a key motivator for pre-drinking.

Abstract:Aim: To describe the research, policy and prevention implications of pre-drinking or pre-gaming; that is, planned heavy drinking prior to going to a public drinking establishment. Methods: The authors describe the phenomenon of pre-drinking, motivations for pre-drinking and its associated risks using available research literature, media and popular internet vehicles. Results: Heavy drinking prior to going out has emerged as a common and celebrated practice among young adults around the world. Apparent motivations are: (i) to avoid paying for high priced drinks at commercial drinking establishments; (ii) to achieve drunkenness and enhance and extend the night out; and (iii) to socialize with friends, reduce social anxiety or enhance male group bonding before going out. Limited existing research on pre-drinking suggests that it is associated with heavy drinking and harmful consequences. We argue that policies focused upon reducing drinking in licensed premises may have the unintended consequence of displacing drinking to pre-drinking environments, possibly resulting in greater harms. Conclusions: Effective policy and prevention for drinking in licensed premises requires a comprehensive approach that takes into account the entire drinking occasion (not just drinking that occurs in the licensed environment), as well as the 'determined drunkenness' goal of some young people.

Abstract:Background Pregaming is a high‐risk drinking behavior that is associated with heavy drinking and negative alcohol‐related consequences. College students may engage in pregaming for several reasons, including to enhance social experiences or cope with negative affect. Research shows that associations between drinking, social anxiety, and depression are multifaceted. However, our understanding of the complex associations of mental health symptoms with pregaming motives and behaviors remains limited. Methods This study examined heterogeneity in how pregaming motives, social anxiety, and depression associate with past 30‐day pregaming variables (quantity, frequency, and negative consequences). We used latent profile analysis (LPA) in a sample of heavy‐drinking university students (N = 479). Results LPA results indicated four profiles: mild/moderate social anxiety and depression symptoms, moderate motives (n = 285), minimal social anxiety and depression symptoms, low motives (n = 61), subclinical/elevated social anxiety and depression symptoms, high motives (n = 75), and clinically elevated social anxiety and depression symptoms, moderate motives (n = 58). The subclinical/elevated symptoms, high motives profile reported the highest pregaming frequency and consequences, including blackouts. Individuals in the moderate motives clinically elevated symptoms, moderate motives profile reported more consequences than those in the minimal symptoms, low motives and mild/moderate symptoms, moderate motives profiles, subclinical/elevated symptoms, high motives profiles. Individuals in the minimal symptoms, low motives profile reported the fewest consequences. Conclusions Among these students, social anxiety and pregaming motives were associated with more frequent pregaming and social anxiety and depression were associated with greater negative consequences. Interventions that target pregaming‐specific motives among students with mental health symptoms may be warranted to reduce this risky behavior and associated drinking‐related harm.

Abstract:Emerging Adulthood, Ahead of Print. Social anxiety symptoms may increase risk for heavy drinking and alcohol-related negative consequences during pregaming (drinking before a social event); efforts to identify malleable psychosocial risk factors are needed. This study examined cross-sectional relationships between social anxiety symptoms, pregaming behaviors, negative alcohol-related consequences, and pregaming-specific motives, moderated by gender. Undergraduates who endorsed current drinking (N = 7528) completed a self-report survey on drinking behaviors as part of a large, multisite study. Social anxiety symptoms were not associated with lifetime history of pregaming. Among participants who engaged in past 30-day pregaming, social anxiety symptoms were related to more negative alcohol-related consequences, greater interpersonal enhancement motives, and lower situational control motives. Social anxiety symptoms were not associated with pregaming quantity. The associations between social anxiety symptoms and pregaming frequency, situational control, and intimate pursuit motives varied by gender. Findings highlight social anxiety symptoms and gender as important factors associated with pregaming-specific motives and behaviors.

Abstract:Objective: Playing drinking games (DGs) is a common behavior among university students, which is concerning given that DG participation is related to more alcohol use/consequences. People's motivations for playing DGs are associated with certain DG behavior patterns and negative consequences. However, few studies have accounted for general drinking motives and other key covariates when examining the unique associations between DG-specific motivations and DG outcomes. The present study used a large, multisite national sample of university students in the United States to determine cross-sectional associations between DG-specific motives (e.g. sexual pursuit, enhancement/thrills, conformity) and DG frequency, quantity, and negative consequences, above and beyond relevant demographics and general drinking motives. Method: University students ( N = 8922) completed a self-report survey that assessed DG-specific behaviors, consequences, and motives, as well as general drinking behaviors and motives. Results: Multivariate findings indicated that enhancement/thrills and sexual pursuit motives are the riskiest DG motives, given their association with DG frequency, quantity, and consequences. Conformity was positively associated with negative consequences while competition and boredom were positively associated with DG frequency. Social lubrication was unrelated to all outcomes. Conclusion: Enhancement/thrills, sexual pursuit, and conformity DG motives may be particularly important targets for DG-specific prevention and intervention efforts.

Abstract:Individuals consume alcohol for a variety of reasons (motives), and these reasons may be differentially associated with the types of drinking outcomes that result. The present study examined whether specific affect-relevant motivations for alcohol use (i.e., coping, enhancement) are associated with distinct types of consequences, and whether such associations occur directly, or only as a function of increased alcohol use. It was hypothesized that enhancement motives would be associated with distinct problem types only through alcohol use, whereas coping motives would be linked directly to hypothesized problem types. Regularly drinking undergraduates (N = 192, 93 female) completed self-report measures of drinking motives and alcohol involvement. Using structural equation modeling, we tested direct associations between coping motives and indirect associations between enhancement motives and eight unique alcohol problem domains: risky behaviors, blackout drinking, physiological dependence, academic/occupational problems, poor self-care, diminished self-perception, social/interpersonal problems, and impaired control. We observed direct effects of coping motives on three unique problem domains (academic/occupational problems, risky behaviors, and poor self-care). Both coping and enhancement motives were indirectly associated (through use) with several problem types. Unhypothesized associations between conformity motives and unique consequence types also were observed. Findings suggest specificity in the consequences experienced by individuals who drink to cope with negative affect versus to enhance positive affect, and may have intervention implications. Findings depict the coping motivated student as one who is struggling across multiple domains, regardless of levels of drinking. Such students may need to be prioritized for interventions.

Abstract:Introduction: Pregaming is a popular but high-risk drinking behavior common among college students. Although sexual and gender minority (SGM) college students are a vulnerable population with regards to hazardous alcohol use and alcohol consequences, there is currently limited research investigating the pregaming behavior of this group. The present study aimed to (1) examine mean level differences in pregaming behaviors and motives between SGM and non-SGM college students and (2) explore how SGM status was associated with pregaming behaviors and if SGM status moderated the association between motives and pregaming behaviors. Methods: The sample consisted of 485 college student drinkers in the US, with 19% ( n = 93) identifying as SGM. All participants completed measures of past 30-day pregaming frequency and quantity (yielding a total pregaming drinks outcome) and drinking consequences experienced on pregaming days. Results: SGM participants consumed significantly fewer pregaming drinks than non-SGM participants, but did not significantly differ on alcohol-related consequences or drinking motives. The pregaming motive of intimate pursuit moderated the association between SGM status and total pregaming drinks, such that non-SGM participants with high intimate pursuit motives drank the heaviest. Conclusions: Our findings suggest that SGM students consume significantly fewer pregaming drinks than their non-SGM counterparts. However, they may be at a similar risk of experiencing pregaming consequences as non-SGM students. SGM students were less susceptible to the effect of intimate pursuit motives on pregaming drink consumption. This study offers support for past research regarding the effects of certain pregaming motives on pregaming drink consumption and consequences.

Drinking locations and alcohol-related harm: Cross-sectional and longitudinal associations in a sample of young Swiss men

Abstract:Background: Alcohol consumption--in particular drinking volume (DV) and risky single occasion drinking (RSOD)--has been related to a wide range of negative consequences and health problems. Previous studies also suggested that drinking in certain locations may be more strongly associated with the occurrence of alcohol-related harm than drinking in others. However, they were conducted in countries culturally and legally different from European countries and were limited to cross-sectional designs. This study investigates the cross-sectional and longitudinal associations of alcohol-related harm with DVs in different locations in a sample of young Swiss men. Methods: A representative sample of 4536 young Swiss male drinkers completed baseline and 15-month follow-up questionnaires. These assessed DVs in 11 locations, alcohol-related harm (i.e. number of alcohol-related consequences and alcohol use disorder criteria) and frequency of RSOD. Cross-sectional and longitudinal associations of alcohol-related harm with DVs in each location were tested using regression models, with and without adjustment for frequency of RSOD. Results: Both cross-sectional and longitudinal analyses showed significant positive associations between alcohol-related harm and DVs at friends' homes, in discos/nightclubs and in outdoor public places, when controlling for frequency of RSOD. In contrast, the contribution of DVs at one's own home and in restaurants was consistently not significant when adjusted for frequency of RSOD. When controlling for RSOD, associations between alcohol-related harm and DVs in bars/pubs, when playing sports, during other leisure activities, at cinemas/theatres, during sporting events, and during special events were not consistent between cross-sectional and longitudinal analyses. Conclusion: Results suggest that prevention interventions should not only target reducing the overall volume of alcohol consumed and the frequency of RSOD in general, but they should additionally focus on limiting alcohol consumption in outdoor public places, discos/nightclubs, and in friends' homes in particular, or at least on preventing harm occurring in these occasions.
Indirect associations between impulsivity and alcohol outcomes through motives for drinking responsibly among U.S. college students: an integration of self-determination theory and the acquired preparedness model

Abstract:The aim of this study was to conduct a preliminary investigation of the associations between facets of impulsivity and alcohol outcomes through motives for drinking responsibly described by self-determination theory (SDT) among college students. Participants ( N = 2808) were part of a multisite investigation of college student drinking across 10 universities in 8 states in the USA who reported past-month drinking. Results of a structural equation model testing all possible indirect associations simultaneously indicated that one-third (20 out of 60) of the indirect associations were statistically significant ( p <.01). Facets with higher scores representing higher impulsivity (negative/positive urgency) were negatively associated with more internalized motives (autonomous motivation and introjected regulation) and positively associated with less internalized motives (external regulation and amotivation) for drinking responsibly. Facets with higher scores representing lower impulsivity (perseverance and premeditation) demonstrated opposite patterns of associations with motives for drinking responsibly. In turn, more internalized motives were related to higher frequency of protective behavioral strategies use, lower alcohol use severity, and fewer negative alcohol-related consequences; less internalized motives demonstrated an opposite pattern of associations with these alcohol outcomes. The present findings should be replicated using experimental and longitudinal studies for appropriately testing mediation but offer support for a novel hypothesis for motivational pathways from impulsivity to alcohol outcomes that may provide insight into intervention targets, if further supported by future research.

Abstract:This study advances the psychometric testing and evaluation of the factor structure of the Positive Drinking Consequences Questionnaire (PDCQ) in adolescents. Our findings identify four factors of positive alcohol‐related consequences: sociability, liquid courage, sexual enhancement, and tension reduction. The PDCQ has demonstrated good internal consistency, test–retest reliability, as well as concurrent and discriminant validity. Furthermore, the PDCQ demonstrates incremental validity beyond alcohol expectancies and drinking motives. These results confirm that the PDCQ is a reliable and valid measure in adolescents. Background The Positive Drinking Consequences Questionnaire (PDCQ) was developed to measure positive consequences of alcohol use endorsed by college drinkers. Efforts to assess positive drinking consequences experienced by adolescents have been much more limited. The aim of the present study was to advance the psychometric testing and evaluation of the factor structure of the PDCQ in adolescents. Methods The current sample consisted of 173 adolescents at T1 (mean age = 15 years, range = 13–17; 61% female) who reported alcohol use in the past 12 months. Data were collected at two time points over a 12‐month interval in the United States. Confirmatory factor analyses, internal consistency, test–retest reliability, and discriminant, concurrent, predictive, and incremental validity were tested. Results Our analyses supported four factors of positive alcohol‐related consequences: sociability, liquid courage, sexual enhancement, and tension reduction. Internal consistency was moderate to high (α = 0.78–0.94, ω = 0.86–0.91 at T1; α = 0.59–0.93, ω = 0.85–0.93 at T2). Test–retest reliability was fair to good (ICC = 0.46–0.55). The PDCQ total and subscale factor scores demonstrated discriminant validity from negative alcohol expectancy. PDCQ total and subscale factor scores were positively associated with current alcohol consumption (ρs = 0.19–0.50 at T1; ρs = 0.17–0.46 at T2), indicating concurrent validity. Predictive validity analyses showed that the overall PDCQ scale score and the sociability subscale positively predicted maximum drinks 1 year later (ρs = 0.18–0.22). However, the sexual enhancement subscale was negatively predictive of typical drinking frequency 1 year later. Finally, the PDCQ showed incremental validity for concurrent alcohol consumption beyond that for alcohol expectancies and drinking motives. Conclusion The present findings support for the reliability and validity of PDCQ for use in adolescents where it may have utility as an assessment tool for characterizing various aspects of positive drinking.

Prevalence and correlates of daily‐level reasons not to drink among young adults who use alcohol

Abstract:Background This study examined reasons not to drink in young adults in relation to demographics, alcohol use patterns, timing (weekend vs. weekday), and typical drinking motives. Methods Young adults who reported past 30‐day alcohol use and at least one nondrinking day (n = 614; mean age = 21.5 years ±0.53) completed a survey of alcohol‐related measures (e.g., typical drinking motives) and up to 14 daily surveys that included 12 reasons not to drink assessed on nondrinking days. Multilevel logistic regressions were estimated for each reason not to drink and related covariates. Results The most common reasons not to drink on a given day were "wasn't interested in drinking" (83.4% of nondrinking days) and "didn't want to get drunk" (81.8% of nondrinking days), with over 96% of participants endorsing each of these at least once. On days (11.6%; by 29.5% of participants) when another drug was used instead of alcohol, 81.8% used cannabis. Sex, race/ethnicity, weekend (vs. weekday), and drinking motives were differentially linked to reasons not to drink. Reporting high‐intensity drinking (i.e., ≥10 drinking on a day) versus binge (5–9 drinks on a day) in the past 2 weeks was linked to "had a hangover recently" (odds ratio = 2.85) as a reason not to drink. Conclusions Findings suggest that reasons not to drink reflect personal decisions and highlight ways to acknowledge situational barriers (e.g., saving money for food and essentials) that can be emphasized in brief interventions. Furthermore, reasons not to drink and alcohol motives may work in tandem within the motivational model to impact alcohol use behaviors.

Abstract:Objective: We examined alcohol use and consequences across five categories of same-day drinking intentions and willingness and tested whether same-day motives and protective strategies predicted differences in outcomes across categories of intentions and willingness. Method: In a 14-week ecological momentary assessment design, undergraduate student participants (N = 196) reported drinking intentions and behaviors over 13 surveys weekly (four morning surveys [Thursday through Sunday]; three midday, early, and late evening surveys [Thursday through Saturday]). On average, participants were 20.61 years old (SD = 1.50; range 17–25), 63% identified as female (n = 124), 29% as male (n = 57), and 8% identified as neither male nor female (n = 15; i.e., nonbinary; transgender; genderqueer; agender). Participants reported numbers of drinks consumed on the evening (past 2 hr) and morning (previous day) surveys. Multilevel generalized linear models tested effects of drinking intentions/willingness categories, motives, protective strategies, and interactions between key variables on alcohol use and consequences in several models. Results: Rates and quantities of drinking were highest on planned drinking days, and especially high when students planned to get drunk. When enhancement and social motives were elevated, students were more likely to drink and consumed more drinks even on unplanned drinking days, and especially when socializing with others. Effects of coping motives were weaker and sparse. Harm reduction protective strategies were associated with more positive and negative consequences with little variation across planned and unplanned drinking days. Conclusion: Jointly considering drinking intentions and willingness narrows the intention-behavior gap in student drinking and suggests potential areas of focus for messaging around responsible drinking. Abstract:OBJECTIVE: Pre-drinking, the practice of consuming alcohol prior to attending a subsequent event, increases the risk of alcohol-related harm, and is common in undergraduate student populations. The current study tested an integrated behaviour change model to identify the motivational, social-cognitive, and implicit predictors of pre-drinking.DESIGN: University students (N = 289) completed an online questionnaire comprising measures of motivational and social-cognitive constructs related to reducing pre-drinking alcohol consumption and past behaviour, and an implicit association test for drinking identity. Participants reported their pre-drinking alcohol consumption at follow-up, 4 weeks from baseline.MAIN OUTCOME MEASURES: Self-reported pre-drinking alcohol consumption.RESULTS: A variance-based structural equation model revealed that few model hypotheses were supported. Although the effects of past behaviour, perceived behavioural control, and implicit drinking identity, on follow-up pre-drinking alcohol consumption were statistically significant, the effect of intention was not.CONCLUSIONS: Current findings indicate pre-drinking alcohol consumption is associated with past behaviour, perceived behavioural control and implicit drinking identity, and no intentions to reduce pre-drinking alcohol consumption. The finding raise questions over the validity of applying the integrated model in this context. Interventions should consider these factors and attempt to facilitate the formation of intentions that lead to subsequent behaviour.

Abstract:Emerging Adulthood, Ahead of Print. Evidence demonstrates an association between symptoms of attention-deficit/hyperactivity disorder (ADHD) and alcohol outcomes, though mechanisms underlying relations are unclear. Given that drinking motives (coping-anxiety, coping-depression, enhancement, social, and conformity) appear to serve as a mediator through which other factors influence drinking behavior, the current multi-site study examined the relation between ADHD symptoms and alcohol outcomes through motives. Past-month drinkers completed online measures assessing ADHD symptoms, alcohol use and problems, and drinking motives. A multiple-mediator model found inattention symptoms were: 1) positively associated with coping-depression, which positively related to alcohol use, which positively related to alcohol problems; 2) positively associated with enhancement motives, which positively related to use, which positively related to problems; and 3) positively associated with coping-anxiety, which negatively related to use, which positively related to problems. No indirect relations via motives were found for hyperactive-impulsive symptoms. Findings highlight unique associations depending on ADHD symptoms and specific drinking motives.

Abstract:Introduction Drinking intensity among young adults is associated with greater negative alcohol-related consequences, but often studied using categorical drinking thresholds. This study examined how alcohol-related consequences varied as a continuous function of number of drinks consumed, without imposing thresholds, to identify drink ranges for which risk is greatest. Methods Analyses included daily surveys from the Young Adult Daily Life study (2019-22) in which individuals reported drinking 1 or more alcoholic drinks (n=5,219 days; 832 individuals). Time-varying effect models estimated total number of negative alcohol-related consequences and five individual consequences (hangover, drank more than planned, nausea/vomiting, did something embarrassing, blacked out) as a function of the number of drinks consumed among young adult males and females. Results Often, increases in the number and prevalence of negative consequences were a nonlinear function of the number of drinks consumed. Females reported few negative consequences on 1-to-3-drink occasions but steep increases in consequences on 4-to-9-drink occasions. Among females, the prevalence of blacking out increased six-fold from 4-drink (3.4%; 95% CI=2.6,4.7) to 9-drink occasions (20.6%; 95% CI=16.1,26.1). Among males, the prevalence of some consequences, while not linear, increased across the full drink range. Blacking out among males increased from 9.6% (95% CI=6.2,14.7) on 10-drink occasions to 23.9% (95% CI=15.0,35.8) on 15-drink occasions. Conclusions Substantial heterogeneity is missed when typical drinking categories are used. In particular, variability in consequences across levels of the traditional binge drinking category suggests that important differences in risk may be missed when assessed as a uniform category.

[Interaction between carbaryl and calf thymus DNA enhanced via Cu2+].

Abstract:The interaction between carbaryl and calf thymus DNA with Cu2+ was studied using fluorescence spectroscopy (FS) and UV spectrum. The quenching process was proved to be single static quenching and the quenching constant decreases with temperature increasing. The experimental results showed that the carbaryl can be intercalated into the twin-screw structure of calf thymus DNA, forming ctDNA-carbaryl adducts. The UV spectrum showed that ctDNA can also result in the increasing color and red shift. In the presence of Cu2+, it was discovered that the apparent association constant (Kalpha) increases and the binding sites number (n) of carbaryl molecules on ctDNA obviously increases. The interaction of the carbaryl and ctDNA was driven mainly by electrostatic force which was enhanced by Cu2+, thus the contribution of deltaH to deltaG increased in the presence of Cu2+ So the interaction between carbaryl and calf thymus DNA was enhanced via Cu+.

Abstract:Does evening circadian preference predict university students’ motives for drinking alcohol (i.e. social, enhancement, conformity, or coping motives)? Drinking to cope, which is associated with alcohol problems, may be more common in evening types because of their sleep problems and difficulties in dealing with stress. Two hundred and nineteen university students (M age = 21.80, SD = 6.80) completed online the Composite Scale of Morningness, the Drinking Motives Questionnaire Revised, the Sleep Quality Scale, the COPE measure of coping with stress, a measure of socially desirable responding, and gave information about gender and age. Evening preference was associated with greater use of all drinking motives, and with poorer sleep and poorer coping with stress. Multiple regression showed that drinking to cope was best predicted by poor sleep, social drinking, and avoidant coping with stress (R 2 = 0.45). Poor sleep and drinking to cope are a cause for concern because of possible reciprocal causality.

Abstract:This study investigated the physiological effects of inhaled corticosteroids, which are used widely to treat asthma. The application of fluticasone propionate (FP, 100 μM) induced sustained increases in the short-circuit current (I(SC)) in human airway Calu-3 epithelial cells. The FP-induced I(SC) was prevented by the presence of H89 (10 μM, a protein kinase A inhibitor) and SQ22536 (100 μM, an adenylate cyclase inhibitor). The FP-induced responses involved bumetanide (a Na(+)-K(+)-2Cl(-) cotransporter inhibitor)-sensitive and 4,4'-dinitrostilbene-2,2'-disulfonic acid (an inhibitor of HCO(3)(-)-dependent anion transporters)-sensitive components, both of which reflect basolateral anion transport. Further, FP augmented apical membrane Cl(-) current (I(Cl)), reflecting cystic fibrosis transmembrane conductance regulator (CFTR)-mediated conductance, in the nystatin-permeabilized monolayer. In I(SC) and I(Cl) responses, FP failed to enhance the responses to forskolin (10 μM, an adenylate cyclase activator). Nevertheless, we found that FP synergistically increased cytosolic cAMP concentrations in combination with forskolin. All these effects of FP were reproduced with the use of budesonide. Collectively, inhaled corticosteroids such as FP and budesonide stimulate CFTR-mediated anion transport through adenylate cyclase-mediated mechanisms in a nongenomic fashion, thus sharing elements of a common pathway with forskolin. However, the corticosteroids cooperate with forskolin for synergistic cAMP production, suggesting that the corticosteroids and forskolin do not compete with each other to exert their effects on adenylate cyclase. Considering that such synergism was also observed in the FP/salmeterol combination, these nongenomic aspects may play therapeutic roles in mucus congestive airway diseases, in addition to genomic aspects that are generally recognized.

Abstract:There are a growing number of commercial uses of nanoparticles which involve direct people contact with the potential for absorption through the skin. Nanoparticles are present in a range of consumer products including colloidal health drinks, carbon fibre sports equipment, sunscreens, cosmetics, electronic products and as antibacterial components of toys, cooking products and wound dressings. Environmental sources of ultra-fine nanoparticles have been present for millennia and anthropogenic sources of similar materials result from industrial processes. Recent technological advances have enabled improvements in both the manufacture of nanoparticles and in the study of their safety. With subcellular dimensions, the physical properties of a nanoparticle cannot be easily predicted from the properties of a microparticle with identical chemical composition. Recent studies in animals and humans have sought to document the safety of consumer nanomaterials. In this article, the safety of such materials is critically reviewed.
