Abstract:The effects of biological factors on the vegetative growth process of mushrooms remain largely unexplored. We investigated the bacterial community in different growth stages of Pleurotus eryngii by high-throughput sequencing technology to explore the relationship between interacting bacteria and the growth and development of P. eryngii. We found significant variances in mushroom interacting association bacteria (MIAB) compositions among the samples from different growth stages, and 410 genera were identified. The bacteria in the full-bag and post-ripe stages were shifted to the biocontrol and growth-promotion ones. The mushroom growth-promoting bacteria (MGPB) were also isolated successfully and identified as B. cereus Bac1. The growth speed and density of mycelial pellets of P. eryngii, and activities of two exoenzymes (laccase and amylase), were analyzed by adding the different volumes of cell-free fermentation broth of B. cereus Bac1 to fungal culture media. The results showed that when a 5 mL cell-free fermentation broth was used, the growth speed of P. eryngii hyphae was enhanced by 1.15-fold over the control and reached 0.46 mm/h. The relative activity of laccase and amylase was increased by 26.9 and 43.83%. Our study revealed that the abundant interacting bacteria coexist with P. eryngii hyphae. Moreover, the abundance of some bacteria exhibiting a positive correlation with the growth periods of their host fungi can effectively promote the growth of the host, which will provide technical supports on the high-efficiency production of P. eryngii in factory cultivation.

Abstract:Volatile organic compounds (VOCs) produced by yeasts can positively affect crops, acting as antifungals or biostimulants. In this study, Aureobasidium pullulans and Metschnikowia pulcherrima were evaluated as potential antagonists of Trichoderma spp., common fungal pathogen in mushroom cultivation. To assess the biocontrol ability and biostimulant properties of the selected yeast species, in vitro co-culture and VOCs exposure assays were conducted. In both assays, VOCs produced by Aureobasidium spp. showed the stronger antifungal activity with a growth inhibition up to 30%. This result was further confirmed by the higher volatilome alcohol content revealed by solid phase microextraction-gas chromatography mass spectrometry (SPME/GC-MS). Overall, Aureobasidium strains can be potentially used as biocontrol agent in Pleorotus ostreatus and Cyclocybe cylindracea mycelial growth, without affecting their development as demonstrated by VOCs exposure assay and Fourier-transform infrared spectroscopy (FT-IR). Conversely, M. pulcherrima was characterized by a lower or absent antifungal properties and by a volatilome composition rich in isobutyl acetate, an ester often recognized as plant growth promoter. As confirmed by FT-IR, Lentinula mycelia exposed to M. pulcherrima VOCs showed a higher content of proteins and lipids, suggesting an improvement of some biochemical properties. Our study emphasizes that VOCs produced by specific yeast strains are potentially powerful alternative to synthetic fungicide in the vegetative growth of mushroom-forming fungi and also able to modify their biochemical composition.

Abstract:Microbial volatiles have beneficial roles in the agricultural ecological system, enhancing plant growth and inducing systemic resistance against plant pathogens without being hazardous to the environment. The interactions of plant and fungal volatiles have been extensively studied, but there is limited research specifically elucidating the effects of distinct volatile organic compounds (VOCs) on plant growth promotion. The current study was conducted to investigate the impact of VOCs from Cladosporium halotolerans NGPF1 on plant growth, and to elucidate the mechanisms for the plant growth-promoting (PGP) activity of these VOCs. The VOCs from C. halotolerans NGPF1 significantly promoted plant growth compared with the control, and this PGP activity of the VOCs was culture medium-dependent. Headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography–mass spectrometry (GC–MS) identified two VOC structures with profiles that differed depending on the culture medium. The two compounds that were only produced in potato dextrose (PD) medium were identified as 2-methyl-butanal and 3-methyl-butanal, and both modulated plant growth promotion and root system development. The PGP effects of the identified synthetic compounds were analyzed individually and in blends using N. benthamiana plants. A blend of the two VOCs enhanced growth promotion and root system development compared with the individual compounds. Furthermore, real-time PCR revealed markedly increased expression of genes involved in auxin, expansin, and gibberellin biosynthesis and metabolism in plant leaves exposed to the two volatile blends, while cytokinin and ethylene expression levels were decreased or similar in comparison with the control. These findings demonstrate that naturally occurring fungal VOCs can induce plant growth promotion and provide new insights into the mechanism of PGP activity. The application of stimulatory volatiles for growth enhancement could be used in the agricultural industry to increase crop yield.

Abstract:Microbial volatile organic compounds (VOCs) emitted from fungi are known as their secondary metabolites from environmental sources. However, their physiological roles remain to be unclear. Even though the roles are still unknown, VOCs are deliberately released to convey information to both homologous and non-homologous organisms. We investigated the effects of single VOCs (hexanal, benzaldehyde, heptanal, 2-ethyl-1-hexanol, 3-octanone, 2-undecanone, 3-octanol, 2-Phenylethanol, 2-phenyl-2-propanol, phenylbenzaldehyde, 2-pentadecanone, β-trans-bergamotene, β-bisabolene, 2-methyl-5 -(1-methylethyl)pyrazine) on the fungal growth. In parallel, application of the co-culturing system in a growth chamber allowed free gas and VOCs exchange between emitter colonies of Fusarium solani and Aspergillus fumigatus , or between colonies of different growth stages of the same species. Distinct self-inhibition occurred by the emitters of fungal growing colonies against receiver ones on the stage of conidial germination or against the younger colonies at an earlier stage in both fungi. Similarly, the phenomenon of allelopathy appeared to work between growing colonies of F . solani and the germinating conidia or young colonies of A . fumigatus or vice versa. Solid phase microextraction—gas chromatography/mass spectrometry revealed VOCs compounds of each fungi. In F . solani , hexanal and benzaldehyde appeared to be significant inhibitors for colony growth. Benzaldehyde inhibited filamentous growth but not conidial germination. In A . fumigatus , heptanal seemed to be an equivalent effector. The inhibitory effect of benzaldehyde was more distinct on the A . fumigatus conidial germination than its filamentous growth.

Abstract:Many volatile compounds secreted by bacteria play an important role in the interactions of microorganisms, can inhibit the growth of phytopathogenic bacteria and fungi, can suppress or stimulate plant growth and serve as infochemicals presenting a new type of interspecies communication. In this work, we investigated the effect of total pools of volatile substances and individual volatile organic compounds (VOCs) synthesized by the rhizosphere bacteria Pseudomonas chlororaphis 449 and Serratia plymuthica IC1270, the soil-borne strain P. fluorescens B-4117 and the spoiled meat isolate S. proteamaculans 94 on Arabidopsis thaliana plants. We showed that total gas mixtures secreted by these strains during their growth on Luria-Bertani agar inhibited A. thaliana growth. Hydrogen cyanide synthesis was unnecessary for the growth suppression. A decrease in the inhibition level was observed for the strain P. chlororaphis 449 with a mutation in the gacS gene, while inactivation of the rpoS gene had no effect. Individual VOCs synthesized by these bacteria (1-indecene, ketones 2-nonanone, 2-heptanone, 2-undecanone, and dimethyl disulfide) inhibited the growth of plants or killed them. Older A. thaliana seedlings were more resistant to VOCs than younger seedlings. The results indicated that the ability of some volatiles emitted by the rhizosphere and soil bacteria to inhibit plant growth should be considered when assessing the potential of such bacteria for the biocontrol of plant diseases.

Abstract:The dynamics of volatilomes emitted during the interaction between plant-growth-promoting bacteria (PGPB) and the phytopathogen Fusarium solani were evaluated for 5 days. The first screening was done to evaluate the antagonist activity of volatile compounds emitted by PGPB against F. solani. Volatilomes from 11 PGPB were determined individually and together with F. solani by using solid-phase microextraction coupled to gas-chromatography–mass spectrometry. Isolates of PGPB belonged to the Bacillus genus and inhibited from 18 to 24% the fungal mycelium growth. The isolates also induced morphological alterations of fungal hyphae, like small globular vesicles and the formation of chlamydospores, suggesting a stress mechanism response by the fungus. Volatilome profile showed 49 different compounds that appeared in the bacterial–fungal interaction, such as ketones, sesquiterpenes, monoterpenoids, alkanes, alkenes, carboxylic acids, and fatty acids. Some ketones and alcohols were detected in high abundance only in the interaction PGPB-fungus at 3 and 5 days. Bacillus circulans A19, Bacillus amyloliquefaciens A21, and Bacillus wiedmannii S18 shared a group of emitted alcohols and ketones when they were exposed to F. solani. F. solani produced its own volatilome profile, with the presence of sesquiterpenes, such as α-cubebene and caryophyllene, which increased significantly in co-incubation with the tested bacteria, suggesting chemical communication between them.

Abstract:Abstract Volatile organic compounds (VOCs) produced by Aureobasidium pullulans were investigated for antagonistic actions against Alternaria alternata and Botrytis cinerea . Conidia germination and colony growth of these two phytopathogens were suppressed by A. pullulans VOCs. A novel experimental setup was devised to directly extract VOCs using solid-phase microextraction-gas chromatography-mass spectrometry (SPME-GC-MS) from antagonist-pathogen culture headspace. The proposed system is a robust method to quantify microbial VOCs using an internal standard. Multivariate curve resolution-alternating least squares deconvolution of SPME-GC-MS spectra identified fourteen A. pullulans VOCs. 3-Methyl-1-hexanol, acetone, 2-heptanone, ethyl butyrate, 3-methylbutyl acetate and 2-methylpropyl acetate were newly identified in A. pullulans headspace. Partial least squares discriminant analysis models with variable importance in projection and selectivity ratio identified four VOCs (ethanol, 2-methyl-1-propanol, 3-methyl-1-butanol and 2-phenylethanol), with high explanatory power for discrimination between A. pullulans and pathogen. The antifungal activity and synergistic interactions of the four VOCs were evaluated using a Box-Behnken design with response surface modelling. Ethanol and 2-phenylethanol are the key inhibitory A. pullulans VOCs against both B. cinerea and A. alternata . Our findings introduce a novel, robust, quantitative approach for microbial VOCs analyses and give insights into the potential use of A. pullulans VOCs to control B. cinerea and A. alternata .

Abstract:The aims of this work were to screen isolated bacteria with a dual capacity: to inhibit Fusarium solani and to promote plant growth. Also, volatile compounds that would be responsible for that effect were identified. Seventy bacterial strains from the air, agricultural soils, hydrocarbons-contaminated soils, and extremophile soils were tested. The former were identified by Matrix-Assisted Laser Desorption/Ionization-time of flight mass spectrometry and 16S rDNA sequencing. The plant growth-promoting bacteria (PGPB) and their capability for phosphate solubilization, siderophores production, and indole production were determined. Twenty isolates from Bacillus and Pseudomonas genera inhibited the mycelial growth up to 40% in direct assays. Eleven isolates significantly inhibited mycelial growth in 18–24% via volatile emissions. Volatile compounds related to antifungal activity or stress response include ketones, sesquiterpenes, monoterpenoids, alkanes, and fatty acids. Our results support the potential of these PGPB to act as biocontrol agents against fungal pathogens via volatile emissions.

Volatile Organic Compound-Mediated Antifungal Activity of Pichia spp. and Its Effect on the Metabolic Profiles of Fermentation Communities

Abstract:Volatile organic compounds (VOCs) are chemicals responsible for antagonistic activity between microorganisms. The impact of VOCs on microbial community succession of fermentation is not well understood. In this study, Pichia spp. were evaluated for VOC production as a part of antifungal activity during baijiu fermentation. The results showed that the abundance of Pichia in the defect group (agglomerated fermented grains) was lower than that in control group, and a negative interaction between Pichia and Monascus was determined ( P < 0.05). In addition, the disruption of fungi was significantly related to the differences of metabolic profiles in fermented grains. To determine production of VOCs from Pichia and its effect on Monascus purpureus , a double-dish system was assessed, and the incidence of M. purpureus reduction was 39.22% after 7 days. As to antifungal volatile compounds, 2-phenylethanol was identified to have an antifungal effect on M. purpureus through contact and noncontact. To further confirm the antifungal activity of 2-phenylethanol, scanning electron microscopy showed that 2-phenylethanol widely and significantly inhibited conidium germination and mycelial growth of filamentous fungi. Metatranscriptomic analysis revealed that the Ehrlich pathway is the metabolic path of 2-phenylethanol in Pichia and identified potential antifungal mechanisms, including protein synthesis and DNA damage. This study demonstrated the role of volatile compound-mediated microbial interaction in microbiome assembly and discovered a plausible scenario in which Pichia antagonized fungal blooms. The results may improve the niche establishment and growth of the functional yeast that enhances the flavor of baijiu. IMPORTANCE Fermentation of food occurs within communities of interacting species. The importance of microbial interactions in shaping microbial structure and metabolic performance to optimize the traditional fermentation process has long been emphasized, but the interaction mechanisms remain unclear. This study applied metabolome analysis and amplicon sequencing along with metatranscriptomic analysis to examine the volatile organic compound-mediated antifungal activity of Pichia and its effect on the metabolism of ethanol during baijiu fermentation, potentially enhancing the establishment of the fermentation niche and improving ethanol metabolism.

Influence of the casing layer on the specific volatile compounds and microorganisms by Agaricus bisporus

Abstract:One of the major variables affecting yield of the mushroom Agaricus bisporus is the casing layer, which directly affects the productivity and mass. Here, volatile organic compounds were extracted by headspace solid-phase microextraction and high-throughput sequencing was used to analyze the microbial community diversity. The relationship between mushroom yield at different cropping stages and the contents of volatile organic compounds and microorganisms in three different casing layers: peat, peat + soil and soil were systematically evaluated. The result shows that Benzaldehyde and (E)-2-octenal which stimulate yield, obviously increased as mushrooms grew, while 3-octanone, which inhibits yield, decreased over time in all three casing layers. However, there was not a strong correlation between the concentration of volatile compounds and yield. In addition, more than 3,000 bacterial operational taxonomic units (OTUs) by performing high throughput sequencing of the microbes were obtained in the three casing layers. Interestingly, the microbial community compositions were very similar between the three casing layers at a later cropping stage, but the community richness varied significantly in different casing layers and at different cropping stages. At the phylum level, the communities had similar structures but were quantitively very different, and this was even more obvious at the genus level. Principal component analysis revealed significant alterations in microbial community structure in different casing layers. Sphingomonas , Dongia and Achromobacter were the dominant genera at cropping stage 1, and the stage 3 were abundant in Saccharibacteria_norank, Pseudomonas , Flavobacterium and Brevundimonas , which was positively correlated with yield, while the abundance of Pseudomonas at stage 1 and Lactococcus and Bacillus at stage 3 was negatively correlated with yield. These results provide a guide for the development and agricultural application of microbial agents for yield improvement in the production of A. bisporus .

Abstract:Oidium sp. has been recovered as an endophyte in Terminalia catappa (tropical chestnut) in Costa Rica. The volatile organic compounds (VOCs) of this organism uniquely and primarily consist of esters of propanoic acid, 2-methyl-, butanoic acid, 2-methyl-, and butanoic acid, 3-methyl-. The VOCs of Oidium sp. are slightly inhibitory to many plant pathogenic fungi. Previous work on the VOCs of Muscodor albus demonstrated that besides esters of small organic acids, a small organic acid and a naphthalene derivative were needed to obtain maximum antibiotic activity. Thus, the addition of exogenous volatile compounds such as isobutyric acid and naphthalene, 1,1'-oxybis caused a dramatic synergistic increase in the antibiotic activity of the VOCs of Oidium sp. against Pythium ultimum. In fact, at elevated concentrations, there was not only 100% inhibition of P. ultimum but killing as well. In addition, a coculture of Muscodor vitigenus (making only naphthalene) and Oidium sp. plus isobutyric acid produced an additive antibiosis effect against P. ultimum. The biological implications of multiple volatile compounds acting to bring about antibiosis in nature are discussed.

Abstract:Various branches of the food and medical industries widely use mushrooms, that is why their aromatic characteristics are quite important. This paper presents a comparative analysis of the aromatic and fatty acid composition of the mycelium of the edible oyster mushroom (Pleurotus ostreatus (Jacq.) P. Kumm., strain K-17) cultured on a glucose-peptone-yeast growth medium with wheat bran added. The quantitative and qualitative composition of the aromas has shown that wheat bran components affect the aroma-forming reactions during the growth of the mycelium of P. ostreatus. It has been established that the introduction of wheat bran increases the content of 1-octen-3-ol, the main fungal aromatic component. In the culture liquid, 1-octen-3-ol increases by 1.4 times, as compared with classic samples cultured on a growth medium without wheat bran. Also, in the mycelium cultured on a wheat bran-containing growth medium, the total quantity of identified aromatic components increases by 1.7 times. In this mycelium, the two main components of the mushroom’s aroma are formed: 1-octen-3-ol and hexanal. In other samples, these important components are absent. The results of this study confirm that the formation of the two main fungal aromatic components, 1‐octen-3-ol and hexanal, involves polyunsaturated fatty acids, namely linoleic acid. Its content decreased in parallel with the accumulation of aromatic components throughout culturing in the wheat bran-containing medium. The data obtained allow establishing how initiation of enzymatic oxidative reactions changes the aroma of P. ostreatus during surface culturing on a liquid medium. The research results reveal the regular patterns in the formation of aromatic components of macromycetes from lipid precursors. Addition of wheat bran to a growth medium unbalances the total content of aromatic components towards their accumulation in the mycelium. Since fruiting bodies and vegetative mycelium are similar in their biochemical composition, the results of studying the mycelium cultured on a liquid medium allow predicting how aromatic components will be formed in the fruiting bodies of oyster mushrooms.

Abstract:Many species or strains of the genus Pseudomonas have been characterized as plant growth promoting rhizobacteria (PGPR). We used a combination of phenotypic and genotypic techniques to analyze the community of fluorescent Pseudomonas strains in the rhizosphere of commercially grown Mentha piperita (peppermint). Biochemical techniques, Amplified rDNA Restriction Analysis (ARDRA), and 16S rRNA gene sequence analysis revealed that the majority of the isolated native fluorescent strains were P. putida. Use of two Repetitive Sequence-based PCR (rep-PCR) techniques, BOX-PCR and ERIC-PCR, allowed us to evaluate diversity among the native strains and to more effectively distinguish among them. PGPR activity was tested for the native strains and reference strain P. fluorescens WCS417r. Micropropagated M. piperita plantlets were exposed to microbial volatile organic compounds (mVOCs) emitted by the bacterial strains, and plant biomass parameters and production of essential oils (EOs) were measured. mVOCs from 11 of the native strains caused an increase in shoot fresh weight. mVOCs from three native strains (SJ04, SJ25, SJ48) induced changes in M. pierita EO composition. The mVOCs caused a reduction of metabolites in the monoterpene pathway, for example menthofuran, and an increase in menthol production. Menthol production is the primary indicator of EO quality. The mVOCs produced by native strains SJ04, SJ25, SJ48, and strain WCS417r were analyzed. The obtained mVOC chromatographic profiles were unique for each of the three native strains analyzed, containing varying hydrocarbon, aromatic, and alogenic compounds. The differential effects of the strains were most likely due to the specific mixtures of mVOCs emitted by each strain, suggesting a synergistic effect occurs among the compounds present.

Simultaneous Impact of Rhizobacteria Inoculation and Leaf-Chewing Insect Herbivory on Essential Oil Production and VOC Emissions in Ocimum basilicum

Abstract:Inoculation with rhizobacteria and feeding by herbivores, two types of abiotic stress, have been shown to increase the production of secondary metabolites in plants as part of the defense response. This study explored the simultaneous effects of inoculation with Bacillus amyloliquefaciens GB03 (a PGPR species) and herbivory by third-instar Spodoptera frugiperda larvae on essential oil (EO) yield and volatile organic compound (VOC) emissions in Ocimum basilicum plants. The density of glandular trichomes was also examined, given that they are linked to EO production and VOC emission. Herbivory increased EO content, but inoculation on its own did not. When combined, however, the two treatments led to a 10-fold rise in EO content with respect to non-inoculated plants. VOC emissions did not significantly differ between inoculated and non-inoculated plants, but they doubled in plants chewed by the larvae with respect to their undamaged counterparts. Interestingly, no changes were observed in VOC emissions when the treatments were tested together. In short, the two biotic stressors elicited differing plant defense responses, mainly when EO was concerned. PGPR did not stimulate EO production, while herbivory significantly enhanced it and increased VOC emissions. The combined treatment acted synergistically, and in this case, PGPR inoculation may have had a priming effect that amplified plant response to herbivory. Peltate trichome density was higher in inoculated plants, those damaged by larvae, and those subjected to the combination of both treatments. The findings highlight the intricate nature of plant defense mechanisms against various stressors and hint at a potential strategy to produce essential oil through the combined application of the two stressors tested here.

Volatile Organic Compounds Produced by Cacao Endophytic Bacteria and Their Inhibitory Activity on Moniliophthora roreri

Abstract:Several studies have reported that bacteria produce anti-fungal volatiles. We identified the organic volatile compounds produced by six cacao endophytic bacteria (CEB) strains (Bacillus amyloliquefaciens CFFSUR-B35, Bacillus megaterium CFFSUR-B32, Bacillus muralis CFFSUR-B39, Bacillus pumilus CFFSUR-B34, Bacillus subtilis CFFSUR-B31, and Novosphingobium lindaniclasticum CFFSUR-B36). We evaluated their inhibitory effect on mycelium growth and spore germination of the phytopathogenic fungus Moniliophtora roreri. The volatiles produced by these six CEB, were collected and identified by SPME and GC–MS. Moreover, the inhibitory effect of five synthetic volatile organic compounds, individually and in mixtures (dimethyl disulfide, 2–5 dimethyl pyrazine, α-pinene, 2-heptanone and 2-ethyl hexanol) on M. roreri mycelium growth and spore germination was evaluated. All strains examined produced volatiles in different amounts; 13 to 10 compounds were identified, including sulfide, alcohol benzene derivate, pyrazine, ketone, nitrogen and terpene compounds. The B. subtilis CFFSUR-B31 strain produced the largest number of volatiles, while B. pumilus CFFSUR-B34 produced the fewest and the lowest amounts. The volatile organic compounds produced by B. pumilus CFFSUR-B34, B. muralis CFFSUR-B39 and N. lindaniclasticum CFFSUR-B36 inhibited M. roreri mycelium growth by more than 35%, sporulation by more than 81% and spore germination by more than 74%. However, when synthetic compounds were evaluated individually and in mixtures, 2-ethyl hexanol at 100,000 ppm (20 mg/filter paper disc) inhibited M. roreri mycelium growth by 100%, followed by organic volatile compound mixtures C (dimethyl disulfide, 2,5-dimethyl pyrazine, α-pinene, 2-ethyl-hexanol, 2-Heptanone) and D (only the top four) at 100,000 ppm (4 and 5 mg/filter paper disc) which inhibited spore germination by 97 and 89%, respectively.Graphical Abstract

Abstract:Introducing Pseudomonas putida optimizes the microbial community within the casing layer, thereby enhancing Agaricus bisporus growth. Nevertheless, the underlying mechanisms governing microbial community succession within the casing layer following microbial agent introduction remained elusive. This study delineated the interplay among microbial populations in the casing layer across the growth cycle of A. bisporus, comparing cohorts treated with P. putida (AP) against untreated controls (CK). The inoculation with Pseudomonas promoted the growth of A. bisporus, shortened its growth cycle, and linearly enhanced the relative abundance of key bacterial genera integral to its growth, including Flavobacterium, Rhodoferax, Pseudarthrobacter, and Vogesella (adjusted R-2 = 0.238-0.866). Importantly, a positive correlation was identified between the proliferation of these beneficial bacteria and positive community cohesion (adjusted R-2 = 0.353-0.838). Additionally, analysis of the symbiotic network topology within the AP group indicated a more stable community following inoculation. The niche results (AP = 11.7672 +/- 1.8938, CK = 11.0481 +/- 3.1273, P < 0.05) underscored the positive impact of Pseudomonas inoculation on ecosystem dynamics. Concentrations of essential nutrients for A. bisporus growth, notably readily available phosphorus (On day 17, the AP group was 78.8333 +/- 1.26623 mg/kg, while the CK group was 30.0333 +/- 0.98658 mg/kg) and readily available potassium (On day 17, the AP group was 2693.9 +/- 14.79561 mg/kg, while the CK group was 1646.9333 +/- 10.71276 mg/kg), were significantly elevated in the AP group. These enhancements strongly correlated with the increased abundance of pivotal microbial populations. Our research provided insights into the application of microbial inoculants to enhance the production of edible fungi.

Abstract:This study investigated the impact of Lactiplantibacillus plantarum PC-004 (PL) and Kazachstania bulderi PY-1 (PY), selected from home-made Paocai, on the physicochemical properties and volatile flavor compounds of fermented Pleurotus eryngii. P. eryngii, fermented by PY, was maintained at about pH 5 throughout fermentation, while co-fermentation exhibited a higher acid production rate and lower post-acidification value compared to mono-fermentation by PL. The viable cell counts of PY exhibited no difference between the mono- and co-fermentation, while PL significantly decreased in the co-fermentation after 168 h. Compared to mono-fermented P. eryngii, co-fermentation had lower levels of certain biogenic amines. Co-fermented P. eryngii had a higher value of nitrite in the early fermentation stage, but this decreased during mono-fermentations. Additionally, among the three different groups, PY fermentation had more esters, PL fermentation had more ketones and aldehydes, and co-fermentation had more abundance and diverse volatile flavor compounds than mono-fermentations. Our results suggest that co-fermentation with K. bulderi and L. plantarum is suitable for producing fermented P. eryngii, highlighting their potential for the fermentation of edible mushrooms.

Abstract:Trichoderma aggressivum, a mycopathogen causing green mould disease, is a major problem in Agaricus bisporus cultivation due to crop loss, and resistance to chemical fungicides. There is an urgent need for novel biological ways to control mycopathogens without affecting the growth of A. bisporus. Bacteria from the mushroom-casing environment were identified and tested for antagonistic effect on T. aggressivum. Bacillus velezensis produced a large zone of inhibition and its supernatant inhibited the growth of T. aggressivum [−37%], and slightly stimulated A. bisporus growth [+2%]. Label free quantitative-proteomic (LFQ) analysis of changes in the abundance of T. aggressivum proteins following exposure to B. velezensis supernatant indicated increased abundance of proteins associated with catabolic processing of amino acids (40-fold), amino oxidase proteins (14-fold), oxidoreductase proteins (13-fold, 4-fold) and hydrolases (3-fold). Proteins that decreased in relative abundance were antioxidants (29-fold), NTF2 domain containing protein (17-fold), 60S ribosomal protein L-13 (14-fold), glucoamylase proteins (13-fold), proteasome subunit proteins (11-fold) and other ribosomal proteins (9-fold). LFQ analysis revealed that exposing A. bisporus to B. velezensis supernatant led to a decrease in: prohibitin (13-fold, 6-fold), proteasomal proteins (11-fold), cytosolic adaptor domain containing protein (5-fold), aldehyde dehydrogenase (4-fold), ribosomal proteins (4-fold), DLH domain-containing protein (4-fold) and PKS_ER domain containing protein (3-fold). The results indicate that A. bisporus was not under stress upon contact with B. velezensis. Whereas a detrimental effect of B. velezensis on T. aggressivum is shown by inhibition of growth and damage-preventing proteins and increased abundance of proteins associated with stress.

Abstract:Background: Certain microorganisms during the preparation of substrate for growing Agaricus bisporus are of great interest for the conversion of organic material and for the excretion of a wide range of metabolites with growth regulation activities that may affect the mycelial growth processes and mushroom yield. Adding beneficial microorganisms in the substrate may be a biotechnological alternative to optimize A. bisporus cultivation. Objective: Isolate and evaluate thermophilic microorganisms from self-heating pasteurized substrate with growth-promoting effects on A. bisporus cultivation. Methods: Different microorganisms were isolated and selected at 45 and 55 °C. They were tested for siderophore production, 1-octen-3-ol consumption, and phosphate solubilization in coculture with A. bisporus to determine their growth effects on potato dextrose agar (PDA) and on sterile pangola grass (Digitaria eriantha). Results: Of the 106 microorganisms isolated, 88 % were able to grow in the presence of 1-octen-3-ol, while 1 % had the capacity to produce siderophores, and 55 % had the ability to solubilize phosphate. The strains Bacillus hisashii ECS-B-65, B. licheniformis ECS-B-78, Rhizomucor pusillus ECS-710 and ECS-712, Aspergillus fumigatus ECS-709, and Thermomyces sp. ECS-711 were found to have a positive effect on A. bisporus mycelial growth.
