Abstract:Intact iodine-replete or iodinedeficient rats were injected with doses of 10–500 μCi 131I. One week later, they were injected with a tracer dose of 125I 4 or 24 hr before autopsy. One thyroid lobe was examined histologically. The other lobe was homogenized and its resistance to 4-hr digestion with 1% pancreatin or with 0.003% pancreatin after pretreatment of the iodoprotein with 0.05M mercaptoethanol was studied. Distribution of both 131I and 125I radioactivity was also determined by quantitative chromatography after 24-hr digestion with 1% pancreatin. In both the iodine-replete and iodine-deficient rats, a dose of 250 or 500 μCi 131I produced increased resistance to digestion in both the 131Iand 125I-labeled iodoprotein and a reduction in the percent of total chromatograph radioactivity of both labels in the iodothyronine component.
