Abstract:Efficient nitrogen (N) removal from mariculture wastewater is one of the important measures to protect the marine ecological environment. A moving bed biofilm reactor (MBBR) inoculating heterotrophic nitrification and aerobic denitrification (HNAD) bacteria was set and the effects of chemical oxygen demand (COD) and total nitrogen (TN) (COD/TN) ratios on N removal performance, enzymatic activity, biofilm characteristics and microbial community were discussed. Results showed that a more stable N removal performance was obtained in the MBBR process inoculating HNAD bacteria under low COD/TN ratio compared with the conventional MBBR. The removal efficiency of nitrate nitrogen (NO 3 − -N) declined with COD/TN ratio decrease, and protein (PN) and polysaccharide (PS) concentration decreased from 284.3 and 37.1 mg (g VSS) −1 to 42.9 and 14.4 mg (g VSS) −1 with low ammonia monooxygenase (AMO), nitrite reductase (NIR) and nitrate reductase (NR) activities. Sequencing analysis revealed that strain Zobellella B307 became the dominant microorganism (the relatively abundance is 2.33–5.60 %), and its relatively abundance and the functional genes related to N metabolism reduced from 15.38 % to 9.66 % and 0.61 % to 0.55 % with COD/TN ratio decrease to explain the reason for the decrease of N removal efficiency. This study can provide a deep insight into N removal mechanism of the reactor inoculating HNAD bacteria under different COD/TN ratios.

Abstract:This research investigated two proposed modified biofilm carriers' performances in treating recirculating aquaculture systems (RAS) wastewater under different salinities (12%o, 26%o, and 35%o) for about 92 days. Three moving bed biofilm reactors (MBBRs; R1, R2, and R3) were filled with unmodified novel sponge biocarriers (SB) served as a control, modified novel SB with ferrous oxalate (C2FeO4@SB), and modified novel SB with combined ferrous oxalate and activated carbon (C2FeO4-AC@SB), respectively. Under the highest saline condition, a significantly higher ammonia removal efficiency of 98.86 +/- 0.7% (p > 0.05) was obtained in R3, whereas R2 and R1 yielded 95.18 +/- 2.8% and 91.66 +/- 1.5%, respectively. Microbial analysis showed that Vibrio, Ruegeria, Formosa, Thalassospira, and Denitromonas were predominant genera, strictly halophilic heterotrophic nitrifying bacteria involved in nitrogen removal. In conclusion, the synergistic effects of novel sponge, C2FeO4 and AC accelerated biofilm formations and stability, subsequently enhanced the removal of ammonia from the mariculture RAS wastewater by the C2FeO4-AC@SB carriers in R3.
Denitrification performance and bacterial ecological network of a reactor using biodegradable poly(3-hydroxybutyrate-co-3-hydroxyvalerate) as an electron donor for nitrate removal from aquaculture wastewater

Abstract:Nitrate accumulation is a common phenomenon in aquaculture that can lead to eutrophication of surrounding water bodies. This study used poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) as a carbon source and substrate and performed a microbial co-occurrence network ecological analysis to elucidate the denitrification processes in two packed-bed reactors with different salinities. The denitrification rate reached maximum values of 0.438 and 0.446 kg m−3 d−1 in reactor I (salinity 0 ‰) and reactor II (salinity 20 ‰), respectively. Although ammonia was formed in both systems based on dissimilation nitrate reduction to ammonia (DNRA), the concentration was very low (2.47 ± 1.99 and 2.84 ± 1.79 mg L−1); moreover, the nitrite content was average (1.01 ± 0.87 and 0.96 ± 0.86 mg L−1). These results suggested that denitrification dominated in both reactors. PHBV generally presented a stable release of DOC, although a sharp increase was observed in the start-up period of reactor II. 16S rRNA results showed that reactor I had richer microbial diversity than reactor II. Among the top ten taxa, Betaproteobacteria was the dominant class in reactor I while Gammaproteobacteria was the dominant class in reactor II. In the stable period, Thauera and Denitromonas was the most abundant genera in reactor I and reactor II, respectively. In addition, the bacterial co-occurrence network showed that reactor I had a more complex node and edge network and faster start-up time compared to reactor II; however, reactor II had a more stable nitrogen removal capacity. Higher expression of NorB and NosZ genes in reactor II indicated higher efficient denitrification in seawater system. The SEM and FTIR showed bacterial development and materials surface erosion. These findings verified the denitrification performance and niche differences between freshwater and seawater environments.
Performance and microbial communities of different biofilm membrane bioreactors with pre-anoxic tanks treating mariculture wastewater

Abstract:The performance of pollutant removals, activated sludge characteristics, and microbial communities of two biofilm membrane bioreactors coupled with pre-anoxic tanks (BF-AO-MBRs) (one using fiber bundle bio-carriers (FB-MBR) and the other using suspended bio-carriers (MB-MBR)) were compared at the salinity between zero and 60 g/L. At all salinities, three bioreactors showed good COD average removal efficiencies (> 94.1%), and FB-MBR showed the best TN removal efficiency (90.4% at 30 g/L salinity). Moreover, FB-MBR had the faster process start-up time and better salt shock resistance. At high salinities (30-60 g/L), more extracellular polymeric substances were produced by the BF-AO-MBRs to avoid the penetration of salt and protect the bacterial community. Because of the different attachment patterns of biofilms, the microbial community structure in the FB-MBR exposed to 30 g/L salinity had higher nitrite-oxidizing/ammonia-oxidizing bacteria ratio (6.44) with more abundance of denitrifiers, which contribute to higher TN removal efficiency and lower nitrite accumulation.

Abstract:As a novel trend, solid carbon sources are applied to act as electron donors and biofilm carrier in biological denitrification process. In this study, simultaneous nitrate and ammonium removal process in an airlift sequencing batch reactor using 3-hydroxybutyrate-co-3-hydroxyvalerate (PHBV) as carbon source and biofilm carrier under intermittent aeration conditions was established to treat effluent of synthetic marine recirculating aquaculture system. The results showed that maximum nitrate and ammonia nitrogen removal rates of 0.45 and 0.09 kg m- 3 d-1 were achieved. No significant nitrite accumulation was found during 200-day operation, while effluent dissolved organic carbon accumulation and particle size reduction significantly increased. Microbial community analysis and batch tests illuminate that the generated sludge and attached biofilm played important roles in nitrogen removal. This study demonstrates the potential mechanism for the nitrogen removal process mediated by 3-hydroxybutyrateco-3-hydroxyvalerate and provide a new idea for the alternative solutions of solid carbon sources.

Abstract:This study investigated the relation between 17 beta-estradiol (E2) degradation and nitrification in synthetic mariculture wastewater by ammonia oxidizing bacterium Nitrosomonas europaea and moving bed biofilm reactor (MBBR). Batch experiments showed that E2 degradation by N. europaea in wastewater followed zero-order reaction kinetics (r(2) = 0.944, 4.07 mu g/ L h(-1)) when ammonia presented. Nitrite yield in N. europaea inoculation decreased by 77.8% exposed to 1 mg/L E2. The inhibitory impact on ammonia oxidation was enhanced with increasing E2 dosage from 50 ng/L to 1 mg/L. Notably, E2 as low as 50 ng/L still had significant interference with nitrite production, bacterial density and ammonia monooxygenase (AMO) activity of N. europaea. Still, the following continuous 68-day degradation test revealed that 84.5%-98.7% E2 could be removed by a benchscale MBBR. Whereas, ammonia removal remarkably decreased from 94.7% +/- 2.1% to 85.6% +/- 2.1% (p < .05) along with the enhanced E2 removal (from 84.5% +/- 2.0% to 98.7% +/- 0.4%, p < .05) when inlet E2 increased from 10 mu g/L to 1 mg/L, indicating the great role of heterotrophs in E2 degradation. In contrast, nitrite oxidation was not affected upon E2 exposure irrespective of E2 concentrations. In summary, nitrification was effective in removing E2, while E2 interfered with ammoxidation process, but this interference was negligible at the reactor level given the low level of E2 in practical field conditions. Abstract:Traditional biotechnology including activated sludge and biofilm process have the problems of poor tolerance and low removal efficiency in treating high ammonia and salt wastewater. In this study, a moving bed biofilm reactor (MBBR) started up with heterotrophic nitrification-aerobic denitrification (HN-AD) bacteria inoculation was proposed, with an initial nitrogen concentration of 600 mg·L−1. The nitrogen removal and microbial characteristics of the reactor were analyzed in comparison to the MBBR startup with activated sludge inoculation (S-MBBR) under different salinities. The results showed that the nitrogen removal efficiency of H-MBBR was 40 % higher than that of S-MBBR under low salinity (<1 %), and it obtained the maximum ammonia removal rate of 10.6 mg·L−1·h−1 at the salinity of 2 %. At 5 % salinity, the H-MBBR system still showed a 22.5 % and 14.5 % increase in ammonia and total nitrogen removal. High-throughout sequencing results showed that the halophilic bacteria Pseudomonas, Halomonas and Nitrincola played a key role at 5 % salinity, with the first two synthesizing tetrahydropyrimidine in H-MBBR to resist osmotic stress. Co-occurrence network analysis showed that a close-knit community structure leaded to an increased nitrogen removal effect in H-MBBR. This study proposes a novel approach for treating high ammonia nitrogen and high salt wastewater.

Abstract:To overcome long start-up time, poor ammonia tolerance and removal performance of traditional moving bed biofilm reactor (MBBR) inoculated with activated sludge for high-ammonia wastewater treatment, a novel MBBR based on heterotrophic nitrification-aerobic denitrification (HN-AD) was proposed. Start-up of MBBR was firstly performed via inoculated with HN-AD bacteria. Start-up time was shortened from 39 d to 15 d, NH4+ tolerance was enhanced from 200 mg/L to 1000 mg/L, and TN removal was increased from 30.4% to 80.7%. The carrier types and NH4+ concentration had significant effects on nitrogen removal and microbial ecology. When the NH4+ concentration was increased to 900 mg/L in MBBR using polyvinyl alcohol gel as carrier, the TN removal, the abundance of HN-AD bacteria Acinetobacter, Pseudomonas and Paracoccus, which played a key role in TN removal and ammonia tolerance, and the abundance of genes related to nitrogen removal were much higher than those of MBBR using kaldness.

Abstract:This paper used a moving bed biofilm reactor( MBBR) to test and analyze aquaculture wastewater treatment. Results indicated that reactor started- up quickly and operated stably in the process of simulated wastewater treatment using MBBR. The removal efficiency for both chemical oxygen demand( COD) and ammonia nitrogen was over 80%,and it reached 50% for total phosphorous( TP),when hydraulic retention time( HRT) was 8 h and DO remained 2. 0 ～ 3. 0 mg·L- 1. Under the condition of organic loading at( 0. 76 ± 0. 03) kg·m- 3·d- 1,the average removal efficiency of total nitrogen( TN) and ammonia was 71. 73% and 98. 42%,respectively,representing the best removal results among all operation periods. To achieve better TN removal rate,organic loading should keep no lower than 0. 5 kg·m- 3·d- 1. TN removal efficiency reached the maximum when dissolved oxygen( DO) was at( 3.00 ±0. 25) mg·L- 1,which was the most suitable to both nitrification and denitrification reactions. To maintain the high removal efficiency of ammonia nitrogen and reduce the nitrite accumulation,DO should keep no less than 2. 0 mg·L- 1in the reactor. Shorter HRT decreased the ammonia removal efficiency and could cause nitrite accumulation. When MBBR was operated in sequencing mode,the removal efficiency of TP was better than that in continuous operation mode. However,nitrite content increased and accumulated at the end of the operation cycle and would be harmful to fish culture.

Abstract:The nitrogen removal performance and mechanisms of Bacillus subtilis H1 isolated from a mariculture environment were investigated. Strain H1 efficiently removed NH 4 + -N, NO 2 − -N, and NO 3 − -N in simulated wastewater with removal efficiencies of 85.61%, 90.58%, and 57.82%, respectively. Strain H1 also efficiently degraded mixed nitrogen (NH 4 + -N mixed with NO 2 − -N and/or NO 3 − -N) and had removal efficiencies ranging from 82.39 to 89.54%. Nitrogen balance analysis revealed that inorganic nitrogen was degraded by heterotrophic nitrification–aerobic denitrification (HN-AD) and assimilation. 15 N isotope tracing indicated that N 2 O was the product of the HN-AD process, while N 2 as the final product was only detected during the reduction of 15 NO 2 − -N. The nitrogen assimilation and dissimilation pathways by strain H1 were further clarified using complete genome sequencing, nitrification inhibitor addition, and enzymatic activity measurement, and the ammonium oxidation process was speculated as NH 4 + → NH 2 OH → NO → N 2 O. These results showed the application prospect of B. subtilis H1 in treating mariculture wastewater.

Abstract:The increasing concern for environmental remediation has led to a search for effective methods to remove eutrophic nutrients. In this study, Methylobacterium gregans DC-1 was utilized to improve nitrogen removal in a sequencing batch biofilm reactor (SBBR) via aerobic denitrification. This bacterium has the extraordinary characteristics of strong auto-aggregation and a high ability to remove nitrogen efficiently, making it an ideal candidate for enhanced treatment of nitrogen-rich wastewater. This strain was used for the bioassessment of a test reactor (SBBRbio), which showed a shorter biofilm formation time compared to a control reactor (SBBRcon) without this strain inoculation. Moreover, the enhanced biofilm was enriched in TB-EPS and had a wider variety of protein secondary structures than SBBRcon. During the stabilization phase of SBBRbio, the EPS molecules showed the highest proportion of intermolecular hydrogen bonding. It is possible that bioaugmentation with this strain positively affects the structural stability of biofilm. At influent ammonia loadings of 100 and 150 mg. L-1, the average reduction of ammonia and nitrate-nitrogen was higher in the experimental system compared to the control system. Additionally, nitrite-N accumulation was lower and N2O production decreased compared to the control. Analysis of the microbial community structure demonstrated successful colonization in the bioreactor by a highly nitrogen-tolerant strain that efficiently removed inorganic nitrogen. These results illustrate the great potential of this type of denitrifying bacteria in the application of bioaugmentation systems.

Abstract:Transurethral ureterolithotripsy (TUL) was performed on 200 patients in the outpatient clinic without fluoroscopic control. Continuous, or one-shot caudal epidural anesthesia was generally adopted to secure the patient's walk after the treatment. The overall rate of stone removal in one session was 51%, however, if confined to the lower ureteral stone, it was 67%. The stone-free rate for all outpatients was 80% in the followuped 183 cases. As acute complications, fever (>37•Ž), bleeding and ureteral perforation occurred in 3 (2%), 5 (3%) and 7 (4%) cases, respectively. As late complications, ureteral stricture developed in 4 cases. Two of them were operated on, and the other 2 were controlled by placement of a double -J stent. The results indicate that TUL can be performed in the outpatient clinic on selected cases.

Abstract:Recently, the functional microorganisms capable of eliminating nitrogenous waste have been applied in mariculture systems. As a potential candidate for treating mariculture wastewater, strain DN3 eliminated 100% of ammonia and nitrate and 96.61%–100% of nitrite within 72 h, when single nitrogen sources at concentrations of 0–50 mg/L. Strain DN3 also exhibited the efficient removal performance of mixed-form nitrogen (ammonia, nitrate, and nitrite) at salinity 30 ‰, C/N ratio 20, and 180 rpm. The nitrogen assimilation pathway dominated inorganic nitrogen metabolism, with less nitrogen (14.23%-25.02% of TN) lost into the air via nitrification and denitrification, based on nitrogen balance analysis. Moreover, the bacterial nitrification pathway was explored by enzymatic assays and inhibition assays. These complex nitrogen assimilation and dissimilation processes were further revealed by bacterial genome analysis. These results provide important insight into nitrogen metabolism of Halomonas sp. and theoretical support for treating mariculture wastewater with strain DN3.

Identification, interactions, nitrogen removal pathways and performances of culturable heterotrophic nitrification-aerobic denitrification bacteria from mariculture water by using cell culture and metagenomics

Abstract:The rapid expansion of aquaculture industry brings about significant environmental concerns, especially nitrogen pollution. Compared to nitrogen bioconversion implemented by the conventional autotrophic nitrifiers and anaerobic denitrifiers, bacteria capable of heterotrophic nitrification-aerobic denitrification (HNAD) in mariculture environments have yet to be well understood. In this study, twenty-five species of new halophilic HNAD bacteria were isolated and identified from mariculture water. By these strains co-cultured in the synthetic mariculture water (ammonia: 5 mg/L, C/N: 5, salinity: 30‰), microbial dynamic analysis showed that ammonia were mainly removed by dominant genera of Marinomonas, Marinobacterium, Halomonas, and Cobetia which simultaneously had positive correlations to total nitrogen removal. Metagenomic annotations revealed that inorganic-N was converted into gaseous-N and organic-N by these HNAD bacteria through nitrogen metabolism pathways of assimilation, partial nitrification, nitroalkane oxidation, nitrate/nitrite dissimilation reduction, and denitrification. Among them, due to the interspecific coexistence and cooperation, Marinomonas communis & Halomonas titanicae, Marinomonas communis & Cobetia marina, Marinomonas aquimarina & Halomonas titanicae, and Marinomonas aquimarina & Cobetia marina exhibited significantly better inorganic-N removal efficiency and stability. The four novel bacterial consortia could transform approximately 60% of initial ammonia into intracellular organic-N (18–20%) and gaseous-N (36–38%), which were significantly higher than those of their single strains. These findings will contribute to understanding and developing the culturable HNAD bacteria as promising candidates for nitrogen pollution control and water bioremediation in mariculture or other saline environments.

Abstract:A heterotrophic nitrification-aerobic denitrification (HN-AD) strain isolated from membrane aerated biofilm reactor (MABR) was identified as Pseudomonas sp. B-1, which could effectively utilize multiple nitrogen sources and preferentially consume NH4-N. The maximum degradation efficiencies of NO3-N, NO2-N and NH4-N were 98.04%, 94.84% and 95.74%, respectively. The optimal incubation time, shaking speed, carbon source, pH, temperature and C/N ratio were 60 h, 180 rpm, sodium succinate, 8, 30 °C and 25, respectively. The strain preferred salinity of 1.5% and resisted heavy metals in the order of Mn2+ > Co2+ > Zn2+ > Cu2+. It can be preliminarily speculated from the results of enzyme assay that the strain removed nitrogen via full nitrification-denitrification pathway. The addition of strain into the conventional MABR significantly intensified the HN-AD performance of the reactor. The relative abundance of the functional bacteria including Flavobacterium, Pseudomonas, Paracoccus, Azoarcus and Thauera was obviously increased after the bioaugmentation. Besides, the expression of the HN-AD related genes in the biofilm was also strengthened. Thus, strain B-1 had great application potential in nitrogen removal process.
In-situ enrichment and application of heterotrophic nitrification-aerobic denitrification bacteria in membrane aerated biofilm reactor

Abstract:In order to reduce the resource and energy consumption of traditional biological nitrogen removal (BNR) process, heterotrophic nitrification-aerobic denitrification (HN-AD) bacteria was in situ enriched in membrane aerated biofilm reactor (MABR) by inoculating conventional activated sludge. Contaminants removal performance, EPS composition and microbial community were explored. The results indicated that the average removal efficiency of COD and TN under optimal condition reached 84.13% and 91.54%, respectively, which demonstrated that the reactor possessed excellent contaminants removal capacity. EPS analysis suggested that abundant protein especially tryptophan protein-like substance played a vital role in maintaining the microbial stability of biofilms. Multiple HN-AD genera, mainly Paracoccus, were detected with the highest relative abundance of 54.70%, which confirmed the successful enrichment of the HN-AD bacteria. Conventional nitrifiers and denitrifiers also thrived in biofilm, which demonstrated the synergistic nitrogen removal of multiple microorganisms. This study provided important insights into application of HN-AD bacteria and synergistic nitrogen removal in BNR system.

Abstract:To address the challenges of nitrogen removal and stability problems in biological treatment of high-salinity wastewater, a new technology based on salt-tolerant aerobic denitrifying (SAD) bacteria to strengthen the two-stage bio-contact oxidation reactor (TBOR) and moving bed biofilm reactor (MBBR) dual-stage process was proposed. The results showed that the NH4 +-N removal rate of TBOR and TN removal rate of MBBR increased by 26.05 % and 15.90 %, respectively, after bioaugmentation with SAD bacteria at salinity of 5 %. The COD, NH4 +-N and TN removal rates in real wastewater (at salinity of 4 ~ 5 %) treatment were 93.45 %, 97.57 % and 90.83 %, respectively. Microbial analysis showed that at salinity of 5 %, SAD bacteria Halomonas and Truepera were the main functional bacteria responsible for nitrogen removal. Microbial functional analysis and metabolic predictions further revealed that bioaugmentation enhanced the expression of the nir gene, thereby further promoting the process of dissimilatory nitrite reduction to ammonium (DNRA). Furthermore, bioaugmentation can also resist the external high-salinity environment by exporting Na+ extracellularly, promoting extracellular polymeric substances (EPS) secretion and amino acid metabolism. This study demonstrates that bioaugmentation with SAD bacteria is a suitable approach for nitrogen and carbon removal of high-salinity wastewater.

Deciphering performance and microbial characterization of marine anammox bacteria-based consortia treating nitrogen-laden hypersaline wastewater: Inhibiting threshold of salinity

Abstract:Hypersaline wastewater posed a challenge to microbial nitrogen removal processes. Herein, halophilic marine anammox bacteria (MAB) were applied to treat nitrogen-rich wastewater with 35-90 g/L salts for the first time. It was found that MAB, with low relative abundance (2.3-6.9 %), still exhibited good nitrogen removal efficiency (>90 %) under 35-70 g/L salts. The specific anammox activity peaked at 180.16 mg N/(g·VSS·d) at 65 g/L salts. MAB secreted more extracellular polymeric substances to resist the adverse effects of hypersaline stress. Nevertheless, the nitrogen removal deteriorated at 75 g/L salts, and further collapsed as the salinity increased. At 90 g/L salts, total nitrogen removal rate decreased by 74 % compared with that of 35 g/L salts. Besides, SBR1031 increased from 12.0 % (35 g/L salts) to 17.4 % (90 g/L salts) and became the dominant bacterial genus in the reactor. This work shed light on the treatment of hypersaline wastewater through MAB.

Abstract:To overcome poor ammonia tolerance and removal performance of bio-contact oxidation (BCO) reactor inoculated with activated sludge for high-ammonia nitrogen ( NH4+-N ) chemical wastewater treatment, this study compared inoculating heterotrophic nitrification-aerobic denitrification (HN-AD) bacteria in moving bed biofilm reactor (MBBR) with activated sludge inoculation in BCO reactor under simulated high NH4+-N conditions. Results revealed that MBBR achieved faster biofilm formation (20 days vs. 100 days for BCO) with notable advantages: 27.6 % higher total nitrogen (TN) and 29.9 % higher NH4+-N removal efficiency than BCO. Microbial analysis indicated optimal enrichment of the key nitrogen removal (NR) bacterium Alcaligenes , leading to increased expression of NR enzymes hydroxylamine reductase, ensuring the superior NR efficiency of the MBBR. Additionally, functional enzymes and genes analysis speculated that the NR pathway in MBBR was: NH4+-N → NH 2 OH → NO3--N → NO2--N → NO → N 2 O → N 2 . This research offers a practical and theoretical foundation for extending HN-AD bacteria-inoculated MBBR processes.

Abstract:Microorganisms in heterotrophic nitrification-aerobic denitrification (HN-AD) mixed bacteria-Chlorella symbiotic system are difficult to be fixed and easily lost in continuous flow process. Meanwhile, excessive aeration in traditional aeration method might result in CO 2 stripping. In this study, the membrane-aerated biofilm reactor (MABR) was employed to achieve efficient nitrogen and phosphorus removal with different COD/TN ratios based on HN-AD mixed bacteria-Chlorella symbiotic system. The results showed that compared with the HN-AD bacteria system, the symbiotic system reduced 33.3 % carbon demand and 43.6 % CO 2 emission. The removal efficiency of TN and TP increased by 14.9 % and 51.2 %, respectively. The SEM, EPS and EEM analysis indicated that, the biofilm mass and intensity reached its best conditions at the optimal COD/TN ratio of 8. 16S rRNA high-throughput sequencing and functional gene prediction indicated that Chlorella increased the abundance of HN-AD functional bacteria ( Acinetobacter , Pseudomonas and Paracoccus played a key role) and the expression of functional gene ( Nas ). This paper provides new insights for the application of the novel HN-AD mixed bacteria-Chlorella symbiotic system in MABR, which will be beneficial for future research.
