Abstract:This article presents a case report on a 17-year-old girl who sustained a dislocation fracture of the talus, and was treated with external fixation. The fracture healed with a good result. After a 1-year follow-up, the patient had completely normal, pain-free mobility in the ankle joint. There were no radiographic signs of osteonecrosis.

Abstract:Microbes, including bacteria and fungi, easily form stable biofilms on many surfaces. Such biofilms have high resistance to antibiotics, and cause nosocomial and postoperative infections. The antimicrobial and antiviral behaviors of Ag and Cu nanoparticles (NPs) are well known, and possible mechanisms for their actions, such as released ions, reactive oxygen species (ROS), contact killing, the immunostimulatory effect, and others have been proposed. Ag and Cu NPs, and their derivative NPs, have different antimicrobial capacities and cytotoxicities. Factors, such as size, shape and surface treatment, influence their antimicrobial activities. The biomedical application of antimicrobial Ag and Cu NPs involves coating onto substrates, including textiles, polymers, ceramics, and metals. Because Ag and Cu are immiscible, synthetic AgCu nanoalloys have different microstructures, which impact their antimicrobial effects. When mixed, the combination of Ag and Cu NPs act synergistically, offering substantially enhanced antimicrobial behavior. However, when alloyed in Ag-Cu NPs, the antimicrobial behavior is even more enhanced. The reason for this enhancement is unclear. Here, we discuss these results and the possible behavior mechanisms that underlie them.
Copper As an Antibacterial Agent for Human Pathogenic Multidrug Resistant Burkholderia Cepacia Complex Bacteria

Abstract:The Burkholderia cepacia complex (Bcc) consists of 17 closely related multidrug resistant bacterial species that are difficult to eradicate. Copper has recently gained attention as an antimicrobial agent because of its inhibitory effects on bacteria, yeast, and viruses. The objective of this study was to evaluate the antibacterial activity of copper surfaces and copper powder against members of the B. cepacia complex. The antibacterial activity of different copper surfaces was evaluated by incubating them with Bcc strain suspensions (5×10(7)cfu/ml). The bacterial survival counts were calculated and the data for various copper surfaces were compared to the data for stainless steel and polyvinylchloride, which were used as control surfaces. The antibacterial activity of copper powder was determined with the diffusimetrical technique and the zone of inhibition was evaluated with paper disks. A single cell gel electrophoresis assay, staining assays, and inductively coupled plasma mass spectroscopy were performed to determine the mechanism responsible for the bactericidal activity. The results showed a significant decrease in the viable bacterial count after exposure to copper surfaces. Moreover, the copper powder produced a large zone of inhibition and there was a significantly higher influx of copper ions into the bacterial cells that were exposed to copper surfaces compared to the controls. The present study demonstrates that metallic copper has an antibacterial effect against Bcc bacteria and that copper adversely affects the bacterial cellular structure, thus resulting in cell death. These findings suggest that copper could be utilized in health care facilities to reduce the bioburden of Bcc species, which may protect susceptible members of the community from bacterial infection.

Abstract:Norepinephrine (NE) and phenylephrine (Phe) were employed to study the ionic requirements for alpha adrenoceptor activation in the teleost Poecilia reticulata melanophores. As expected the beta adrenoceptor blocker, propranolol, increased the sensitivity of the preparation to NE (5.8 times), and was therefore employed in all the experimental procedures. Neither cocaine (a neuronal uptake blocker) nor dexamethasone (an extraneuronal uptake blocker) enhanced the sensitivity of the preparation to NE, suggesting that these inactivating mechanisms would not play a role in P. reticulata pigmentary system. However, in the absence of calcium, the dose-response curve (DRC) to NE was displaced to the left about 3.5 times, whereas the DRC to Phe was not affected. These results indicate that a neuronal uptake is active, but was not demonstrated by the classical pharmacological tools, probably due to an assymmetric display of the nervous endings. The DRC to NE was rightward displaced (14.1 times) in the presence of the calcium channel blocker Verapamil, whereas the DRC to Phe was not affected. These data suggest that P. reticulata melanophores possess a mixed population of alpha 1 and alpha 2 adrenoceptors, the activation of the latter eliciting an extracellular calcium influx. In sodium-free saline, the DRC to NE was rightward shifted (6.6 times) and the response to Phe was impaired in such a way that the maximal response was not achieved. Copper as an antimicrobial agent against opportunistic pathogenic and multidrug resistant Enterobacter bacteria

Abstract:Infections by Enterobacter species are common and are multidrug resistant. The use of bactericidal surface materials such as copper has lately gained attention as an effective antimicrobial agent due to its deadly effects on bacteria, yeast, and viruses. The aim of the current study was to assess the antibacterial activity of copper surfaces against Enterobacter species. The antibacterial activity of copper surfaces was tested by overlying 5×10 6 CFU/ml suspensions of representative Enterobacter strains and comparing bacterial survival counts on copper surfaces at room temperature. Iron, stainless steel, and polyvinylchloride (PVC) were used as controls. The mechanisms responsible for bacterial killing on copper surfaces were investigated by a mutagenicity assay of the D-cycloserin (cyclA gene), single cell gel electrophoresis, a staining technique, and inductively coupled plasma mass spectroscopy. Copper yielded a significant decrease in the viable bacterial counts at 2 h exposure and a highly significant decrease at 4 h. Loss of cell integrity and a significantly higher influx of copper into bacterial cells exposed to copper surfaces, as compared to those exposed to the controls, were documented. There was no increase in mutation rate and DNA damage indicating that copper contributes to bacterial killing by adversely affecting cellular structure without directly targeting the genomic DNA. These findings suggest that copper’s antibacterial activity against Enterobacter species could be utilized in health care facilities and in food processing plants to reduce the bioburden, which would increase protection for susceptible members of the community.
Metal Bionanohybrids against Microbiologically Influenced Corrosion (MIC) Consortia

Abstract:In search of new materials that would help to prevent microbiologically influenced corrosion (MIC), we have designed and synthetized six different copper and copper-silver nanoparticle-enzyme hybrids using a mild-conditions method carried out in water and r.t. Characterization analyses exhibited the presence of small crystalline nanoparticles with diameters from 2 to 20 nm. X-ray diffraction determined that the Cu hybrids were composed of different copper species, depending on the synthetic protocol used, while the Cu-Ag hybrids were mainly composed of copper and silver phosphate metallic species. Then, the bacterial viability of three MIC-relevant enrichments, sulfate-reducing bacteria (SRB), slime-forming bacteria (SFB), and acid-producing bacteria (APB), was studied in the presence of the bionanohybrids. The results demonstrated a notable effect of all bionanohybrids against SRB, one of the most prominent bacteria associated with MIC. In particular, Cu-2 and Cu-Ag-2 showed a reduction in bacterial cells of 94% and 98% after 48 h, respectively, at a concentration of 100 ppm. They also exhibited high efficiencies against SFB, with Cu-Ag-1 and Cu-Ag-2 hybrids being the best, with bacterial reduction percentages of 98% after 45 h of exposition at a concentration of 100 ppm. However, in the case of APB, the effect of the hybrids was lost due to the low pH level generated during the experiment. Finally, the capacity of Cu-2 and Cu-Ag-2 to inhibit the adhesion of SRB to the surface of carbon steel coupons was evaluated. Fluorescence imaging of the surface of the coupons at 24 h demonstrated that the presence of the hybrids inhibited the growth of SRB, obtaining a maximum reduction of 98% with Cu-2. Overall, the results of this study demonstrate that these novel nanomaterials have a wide-range antibacterial effect and may have a promising future in the prevention and treatment of MIC.

Abstract:In this work, we report the synthesis of copper nanoparticles (Cu NPs), employing the chemical reduction method in an aqueous medium. We used copper sulfate pentahydrate (CuSO4·5H2O) as a metallic precursor; polyethylenimine (PEI), allylamine (AAM), and 4-aminobutyric acid (AABT) as stabilizing agents; and hydrated hydrazine as a reducing agent. The characterization of the obtained nanoparticles consisted of X-ray, TEM, FTIR, and TGA analyses. Through these techniques, it was possible to detect the presence of the used stabilizing agents on the surface of the NPs. Finally, a zeta potential analysis was performed to differentiate the stability of the nanoparticles with a different type of stabilizing agent, from which it was determined that the most stable nanoparticles were the Cu NPs synthesized in the presence of the PEI/AAM mixture. The antimicrobial activity of Cu/PEI/AABT toward P. aeruginosa and S. aureus bacteria was high, inhibiting both bacteria with low contact times and copper concentrations of 50–200 ppm. The synthesis method allowed us to obtain Cu NPs free of oxides, stable to oxidation, and with high yields. The newly functionalized Cu NPs are potential candidates for antimicrobial applications.

Abstract:The constant advent of major health threats such as antibacterial resistance or highly communicable viruses, together with a declining antimicrobial discovery, urgently requires the exploration of innovative therapeutic approaches. Nowadays, strategies based on metal nanoparticle technology have demonstrated interesting outcomes due to their intrinsic features. In this scenario, there is an emerging and growing interest in copper-based nanoparticles (CuNPs). Indeed, in their pure metallic form, as oxides, or in combination with sulfur, CuNPs have peculiar behaviors that result in effective antimicrobial activity associated with the stimulation of essential body functions. Here, we present a critical review on the state of the art regarding the <i>in vitro</i> and <i>in vivo</i> evaluations of the antimicrobial activity of CuNPs together with absorption, distribution, metabolism, excretion, and toxicity (ADMET) assessments. Considering the potentiality of CuNPs in antimicrobial treatments, within this Review we encounter the need to summarize the behaviors of CuNPs and provide the expected perspectives on their contributions to infectious and communicable disease management.This article has not yet been cited by other publications.

Synergistic Polymer Coatings with Antibacterial and Antiviral Properties for Healthcare Applications

Abstract:The role of frequently touched surfaces in the transmission of infectious diseases is well-documented, and the urgent need for effective surface technologies with antipathogen activity has been highlighted by the recent global pandemic and rise in antimicrobial resistance. Here, we have explored combinations of up to 3 different classes of compounds within a polymeric matrix to enable the fabrication of coatings with broad-spectrum activity. Compounds were either based on metals or metal oxides, namely, copper, silver, and copper oxide, essential oils, namely, cinnamaldehyde, tea tree oil, and carvacrol oil, or cationic polymers, namely, poly(ε-lysine) and poly(hexamethylene biguanide). These compounds were mixed into a polymer matrix, coated, and dried to yield durable coatings. Coatings containing up to 7.5% (w/w) of the compounds were assessed in the zone of inhibition and biofilm assays using Staphylococcus aureus and Pseudomonas aeruginosa, as well as infectivity assays using human coronavirus OC43. Our data demonstrate that a selected combination of additives was able to provide a 5-log reduction in the colony-forming units of both bacteria and a 4-log reduction in viral infectivity. This simple but highly effective technology is expected to find applications in environments such as hospitals, aged care facilities, or public transport.
Biocompatible Copper Oxide Nanoparticle Composites from Cellulose and Chitosan: Facile Synthesis, Unique Structure, and Antimicrobial Activity

Abstract:Copper in various forms has been known to have bactericidal activity. Challenges to its application include preventing mobilization of the copper, to both extend activity and avoid toxicity, and bioincompatibility of many candidate substrates for copper immobilization. Using a simple ionic liquid, butylmethylimmidazolium chloride as the solvent, we developed a facile and green method to synthesize biocompatible composites containing copper oxide nanoparticles (CuONPs) from cellulose (CEL) and chitosan (CS) or CEL and keratin (KER). Spectroscopy and imaging results indicate that CEL, CS, and KER remained chemically intact and were homogeneously distributed in the composites with CuONPs with size of 22 ± 1 nm. Electron paramagnetic resonance (EPR) suggests that some 25% of the EPR-detectable Cu(II) is present as a monomeric species, chemically anchored to the substrate by two or more nitrogen atoms, and, further, adopts a unique spatially oriented conformation when incorporated into the [CEL + CS] composite but not in the [CEL + KER] composite. The remaining 75% of EPR-detectable Cu(II) exhibited extensive spin-spin interactions, consistent with Cu(II) aggregates and Cu(II) on the surface of CuONPs. At higher levels of added copper (>59 nmol/mg), the additional copper was EPR-silent, suggesting an additional phase in larger CuONPs, in which S > 0 spin states are either thermally inaccessible or very fast-relaxing. These data suggest that Cu(II) initially binds substrate via nitrogen atoms, from which CuONPs develop through aggregation of copper. The composites exhibited excellent antimicrobial activity against a wide range of bacteria and fungi, including methicillin-resistant Staphylococcus aureus; vancomycin-resistant Enterococcus; and highly resistant Escherichia coli, Streptococcus agalactiae, Pseudomonas aeruginosa, Stenotrophomonas maltophilia, and Candida albicans. Expectedly, the antibacterial activity was found to be correlated with the CuONPs content in the composites. More importantly, at CuONP concentration of 35 nmol/mg or lower, bactericidal activity of the composite was complemented by its biocompatibility with human fibroblasts.

Abstract:Elucidation of the role played by cells of the innate immune system, particularly the dendritic cell (DC) populations, has led to a precipitous growth in our understanding of immunity to pathogens and foreign antigens. Much of this information has been derived from studies using mouse model systems. However, mice and human DCs differ drastically in the relative distribution of the toll-like receptors (TLRs) critical for immune activation. This is particularly true for the plasmacytoid DCs (pDCs), which are activated almost exclusively through TLR signaling. Variation in this DC subpopulation has been implicated in a number of pathological syndromes; therefore, a thorough understanding of their steady state and activation profiles in human patients is essential. A number of factors, including the relatively low numbers of these cells in blood, have precluded careful analysis in clinical trials. To overcome these limitations, we have developed a technique for studying the steady state and activation profile of pDCs collected from small amounts of human blood. This technique can be performed with 10,000 cells to obtain the immune transcriptome of the pDCs analyzed by quantitative PCR using amplified RNA. In addition, we have used multiplex enzyme-linked immunosorbent assays to measure secreted proteins. We demonstrate the validity of this technique and document its potential for use with blood from human study populations.

Abstract:This work reports a nanocatalytic antimicrobial agent (Si@Cu NSs) with enhanced CDT via p‐d orbital hybridization strategy and synergistic photothermal effect. This strategy enhances the Fenton‐like reaction of copper atoms and addresses the limited application of nanoparticles in pathophysiological environments. Excellent therapeutic effects demonstrate the feasibility of constructing novel CDT agents by orbital hybridization for the catalytic treatment of IAIs. Recurrent bacterial infections, impenetrable microbial biofilms, and irremediable antibiotic resistance are the most perilous threats in orthopedic implant‐associated infections (IAIs). Recently, chemodynamic therapy (CDT) has been considered a promising therapy, while the clinical practices for existing CDT agents are limited by the low therapeutic efficiency in physiological circumstances. Herein, it is demonstrated that silica‐copper nanosheets (Si@Cu NSs) exhibit a combined therapeutic photothermal‐chemodynamic strategy for IAIs treatment with superior capacity and biocompatibility. This unique design endows the nanostructure with enhanced copper‐based Fenton‐like properties via p‐d orbital hybridization by incorporating copper nanoclusters (Cu NCs) on silicene nanosheets (Si NSs) matrix, integrating inherent photothermal performance of Si NSs with specific catalytic anti‐infection of Cu NCs against planktonic bacteria and biofilms both in vitro and in vivo. This study not only reveals the promising bio‐application prospects of 2D nanocatalytic biomaterials but also demonstrates the feasibility of constructing novel CDT agents by orbital hybridization for the catalytic treatment of IAIs.

Abstract:Antibacterial properties of copper have been known for ages. With the rise of antimicrobial resistance (AMR), hospital-acquired infections, and the current SARS-CoV-2 pandemic, copper and copper-derived materials are being widely researched for healthcare ranging from therapeutics to advanced wound dressing to medical devices. We cover current research that highlights the potential uses of metallic and ionic copper, copper alloys, copper nanostructures, and copper composites as antibacterial, antifungal, and antiviral agents, including those against the SARS-CoV-2 virus. The applications of copper-enabled engineered materials in medical devices, wound dressings, personal protective equipment, and self-cleaning surfaces are discussed. We emphasize the potential of copper and copper-derived materials in combating AMR and efficiently reducing infections in clinical settings.
Biocompatibility and antimicrobial activity of copper(II) oxide hybridized with nano silicate platelets

Abstract:Nano-sized copper(II) oxide (CuO) is an interesting material with proficient antibacterial activities but limited due to its toxicity to mammalian cells. In this study, CuO nano-sized needles were coated on the surface of nano-silicate platelets (NSP), which were exfoliated from natural montmorillonite clays and with the thickness of about 1 nm, to form nanohybrids. Our findings indicate that NSPs can effectively prevent the aggregation of CuO needles, and significantly enhance antibacterial efficiency without affecting biocompatibility. An optimal antibacterial activity against Staphylococcus aureus exceeding 99% could be obtained, while that for Escherichia coli was about 88%. Electron paramagnetic resonance results suggest that •OH radicals, one of the reactive oxygen species generated by CuO/NSP, might damage the cellular components and thus result in the death of bacterial.

Abstract:Copper is a cheaper alternative to various noble metals with a range of potential applications in the field of nanoscience and nanotechnology. However, copper nanoparticles have major limitations, which include rapid oxidation on exposure to air. Therefore, alternative pathways have been developed to synthesize metal nanoparticles in the presence of polymers and surfactants as stabilizers, and to form coatings on the surface of nanoparticles. These surfactants and polymeric ligands are made from petrochemicals which are non- renewable. As fossil resources are limited, finding renewable and biodegradable alternative is promising.The study aimed at preparing, characterizing and evaluating the antibacterial properties of copper nanoparticles. Copper nanoparticles were prepared using gelatin biopolymer, CuSO4.5H2O ions and hydrazine as stabilizer, precursor salt and reducing agent respectively. However, vitamin C and NaOH solution were also employed as an antioxidant and pH adjuster. The synthesized copper nanoparticles were characterized using UV-visible spectroscopy (UV-vis), thermogravimetric analysis (TGA), zeta potential measurements powder, X-ray diffraction (XRD), field emission scanning electron microscope and transmission electron microscope (TEM). The UV-visible absorption spectrum confirms the formation of the CuNPs, which showed maximum absorbance at 583 nm. Results obtained from TEM indicated a decrease in size of particle from a low concentration to high concentration of the supporting materials. The optimum concentration of gelatin was found to be 0.75 wt%. The supporting materials used for this synthesis are biocompatible and the obtained products are stable in air. The synthesized CuNPs display promising antibacterial activities against B. subtilis (B29), S. aureus (S276), S. choleraesuis (ATCC 10708) and E. coli (E266) as gram positive and negative bacteria respectively.

Abstract:Rampant pathogenesis induced by communicable microbes has necessitated development of technologies for rapid and sustained disinfection of surfaces. Copper nanoparticles (CuNPs) have been widely reported for their antimicrobial properties. However, nanostructured copper is prone to oxidative dissolution in the oil phase limiting its sustained use on surfaces and coatings. The current study reports a systematic investigation of a simple synthesis protocol using fatty acid stabilizers (particularly essential oils) for synthesis of copper nanoparticles in the oil phase. Of the various formulations synthesized, rosemary oil stabilized copper nanoparticles (RMO CuNPs) were noted to have the best inactivation kinetics and were also most stable. Upon morphological characterization by TEM and EELS, these were found to be monodispersed (ᴓ 5-8 nm) with copper coexisting in all three oxidation states on the surface of the nanoparticles. The nanoparticles were dropcast on woven fabric of around 500 threads per inch and exposed to gram positive bacteria (Staphylococcus aureus), gram negative bacteria (Escherichia coli and Pseudomonas aeruginosa), enveloped RNA virus (phi6), non-enveloped RNA virus (MS2) and non-enveloped DNA virus (T4) to encompass the commonly encountered groups of pathogens. It was possible to completely disinfect 107 copies of all microorganisms within 40 minutes of exposure. Further, this formulation was incorporated with polyurethane as thinners and used to coat non-woven fabrics. These also exhibited antimicrobial properties. Sustained disinfection with less than 9% cumulative copper loss for upto 14 washes with soap water was observed. Based on the studies conducted, RMO CuNP in oil phase was found to have excellent potential of integration on surface coatings, paints and polymers for rapid and sustained disinfection of microbes on surfaces.

Abstract:Antimicrobial resistance (AMR), caused by microbial infections, has become a major contributor to morbid rates of mortality worldwide and a serious threat to public health. The exponential increase in resistant pathogen strains including Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) poses significant hurdles in the health sector due to their greater resistance to traditional treatments and medicines. Efforts to tackle infectious diseases caused by resistant microbes have prompted the development of novel antibacterial agents. Herein, we present selenium and copper oxide monometallic nanoparticles (Se-MMNPs and CuO-MMNPs), characterized using various techniques and evaluated for their antibacterial potential via disc diffusion, determination of minimum inhibitory concentration (MIC), antibiofilm, and killing kinetic action. Dynamic light scattering (DLS), scanning electron microscopy (SEM/EDX), and X-ray diffraction (XRD) techniques confirmed the size-distribution, spherical-shape, stability, elemental composition, and structural aspects of the synthesized nanoparticles. The MIC values of Se-MMNPs and CuO-MMNPs against S. aureus and E. coli were determined to be 125 μg/mL and 100 μg/mL, respectively. Time–kill kinetics studies revealed that CuO-MMNPs efficiently mitigate the growth of S. aureus and E. coli within 3 and 3.5 h while Se-MMNPs took 4 and 5 h, respectively. Moreover, CuO-MMNPs demonstrated better inhibition compared to Se-MMNPs. Overall, the proposed materials exhibited promising antibacterial activity against S. aureus and E. coli pathogens.

Copper Oxide Electrochemical Deposition to Create Antiviral and Antibacterial Nanocoatings

Abstract:The impact of the reaction environment on the formation of the polycrystalline layer and its biomedical (antimicrobial) applications were analyzed in detail. Copper oxide layers were synthesized using an electrodeposition technique, with varying additives influencing the morphology, thickness, and chemical composition. Scanning electron microscopy (SEM) images confirmed the successful formation of polyhedral structures. Unmodified samples (CuL) crystallized as a mixture of copper oxide (I) and (II), with a thickness of approximately 1.74 μm. The inclusion of the nonconductive polymer polyvinylpyrrolidone (PVP) during synthesis led to a regular and compact CuO-rich structure (CuL-PVP). Conversely, adding glucose resulted in forming a Cu2O-rich nanostructured layer (CuL-D(+)G). Both additives significantly reduced the sample thickness to 617 nm for CuL-PVP and 560 nm for CuL-D(+)G. The effectiveness of the synthesized copper oxide layers was demonstrated in their ability to significantly reduce the T4 phage titer by approximately 2.5-3 log. Notably, CuL-PVP and CuL-D(+)G showed a more substantial reduction in the MS2 phage titer, achieving about a 5-log decrease. In terms of antibacterial activity, CuL and CuL-PVP exhibited moderate efficacy against Escherichia coli, whereas CuL-D(+)G reduced the E. coli titer to undetectable levels. All samples induced similar reductions in Staphylococcus aureus titer. The study revealed differential susceptibilities, with Gram-negative bacteria being more vulnerable to CuL-D(+)G due to its unique composition and morphology. The antimicrobial properties were attributed to the redox cycling of Cu ions, which generate ROS, and the mechanical damage caused by nanostructured surfaces. A crucial finding was the impact of surface composition rather than surface morphology on antimicrobial efficacy. Samples with a dominant Cu2O composition exhibited potent antibacterial and antiviral properties, whereas CuO-rich materials showed predominantly enhanced antiviral activity. This research highlights the significance of phase composition in determining the antimicrobial properties of copper oxide layers synthesized through electrodeposition.

Abstract:Metallic nanoparticles are a new class of materials with applications in medicine, pharmaceutical and agriculture. Using biological, chemical and physical approaches, nanoparticles with amazing properties are obtained. Copper is one of the most-found elements and plays an important part in the normal functioning of organisms. Coper nanoparticles have superior antibacterial properties when comparing them to present day antibiotics. Moreover, apart from their antibacterial role, antifungal, antiviral and anticancer properties have been described. Although the mechanism of actions is not completely understood, copper nanoparticles can become a viable alternative in fighting multi-resistant bacteria strains. We hereby review the already existing data on copper nanoparticle synthesis, effects and mechanisms of action as well as toxicity.
Synergistic antibacterial effect of copper and silver nanoparticles and their mechanism of action

Abstract:Abstract Bacterial infections are one of the leading causes of death worldwide. In the case of topical bacterial infections such as wound infections, silver (Ag) has historically been one of the most widely used antibacterials. However, scientific publications have demonstrated the adverse effects of silver on human cells, ecotoxicity and insufficient antibacterial effect for the complete elimination of bacterial infections. The use of Ag in the form of nanoparticles (NPs, 1–100 nm) allows to control the release of antibacterial Ag ions but is still not sufficient to eliminate infection and avoid cytotoxicity. In this study, we tested the potency of differently functionalized copper oxide (CuO) NPs to enhance the antibacterial properties of Ag NPs. The antibacterial effect of the mixture of CuO NPs (CuO, CuO–NH 2 and CuO–COOH NPs) with Ag NPs (uncoated and coated) was studied. CuO and Ag NP combinations were more efficient than Cu or Ag (NPs) alone against a wide range of bacteria, including antibiotic-resistant strains such as gram-negative Escherichia coli and Pseudomonas aeruginosa as well as gram-positive Staphylococcus aureus , Enterococcus faecalis and Streptococcus dysgalactiae . We showed that positively charged CuO NPs enhanced the antibacterial effect of Ag NPs up to 6 times. Notably, compared to the synergy of CuO and Ag NPs, the synergy of respective metal ions was low, suggesting that NP surface is required for the enhanced antibacterial effect. We also studied the mechanisms of synergy and showed that the production of Cu + ions, faster dissolution of Ag + from Ag NPs and lower binding of Ag + by proteins of the incubation media in the presence of Cu 2+ were the main mechanisms of the synergy. In summary, CuO and Ag NP combinations allowed increasing the antibacterial effect up to 6 times. Thus, using CuO and Ag NP combinations enables to retain excellent antibacterial effects due to Ag and synergy and enhances beneficial effects, since Cu is a vital microelement for human cells. Thus, we suggest using combinations of Ag and CuO NPs in antibacterial materials, such as wound care products, to increase the antibacterial effect of Ag, improve safety and prevent and cure topical bacterial infections.

Abstract:Recent clinical outbreaks of infectious diseases caused by pathogenic microorganisms, such as viruses, bacteria, and fungi, along with the emergence of unwanted microorganisms in industrial settings, have significantly reduced efficiency. Graphene has recently attracted significant attention as a potential antimicrobial agent because of its low toxicity, ease of production and functionalization, and high solubility in water. The presence of oxygen functional groups allows the interaction of the compound with bacteria and other biomolecules, making it an interesting candidate for antimicrobial therapy. Moreover, integrating graphene into copper coatings has been shown to enhance their antimicrobial properties. However, the implementation of copper–graphene composite coatings is currently limited by the difficulty of uniformly distributing graphene within the copper matrix. Copper (Cu)-doped graphitic nanoplatelets (CuGnPs), one option to overcome this challenge, are made via a mechanochemical reaction between solid graphite and Cu powder. The configuration of C–Cu bonds within CuGnPs can be identified using a range of analytical techniques, including transmission electron microscopy, X-ray photoelectron spectroscopy, energy-dispersive X-ray spectroscopy, scanning electron microscopy, and time-of-flight secondary ion mass spectrometry. To evaluate the antibacterial activity of the Cu-GnPs, we employed Escherichia coli or Staphylococcus aureus. Various amounts (250, 500, 750, and 1000 μg/mL) of prepared CuGnP samples were incubated in a bacterial suspension for 3 or 6 h at 150 rpm and 37 °C for a colony-forming unit assay. Three hours and six hours of treatment of the bacteria with CuGnPs led to a significant difference in bacterial survival compared with that of the control. It was observed that CuGnPs, with copper bound to graphene oxide, more effectively inhibited the proliferation of E. coli compared with nanoplatelets containing graphene oxide alone. These findings suggest that the unique properties of CuGnPs, such as C–Cu bonds, high surface area, and the coexistence of micropores and mesopores, are valuable for exerting strong antimicrobial effects making CuGnPs effective at preventing bacterial colonization on industrial surfaces.
