Abstract:Numerous studies have shown that prenatal stress (PNS) can have profound effects on postnatal well-being. Here, the domestic pig (Sus scrofa) was used to investigate PNS effects owing to the direct relevance for farm animal welfare and the developing status of the pig as a large animal model in translational research. Pregnant primiparous sows were exposed, in mid-gestation, to either a social stressor (mixing with unfamiliar conspecifics) or were kept in stable social groups. The ratio of levels of mRNAs for corticotropin releasing hormone (CRH) receptors 1 and 2 in the amygdala, measured for the first time in the pig, was substantially increased in 10-week-old female, but not male, PNS progeny indicating a neurobiological propensity for anxiety-related behaviour. Mature female offspring were observed at parturition in either a behaviourally restrictive crate or open pen. Such PNS sows showed abnormal maternal behaviour in either environment, following the birth of their first piglet. They spent more time lying ventrally, more time standing and showed a higher frequency of posture changes. They were also more reactive towards their piglets, and spent longer visually attending to their piglets compared to controls. Associated with this abnormal maternal care, piglet mortality was increased in the open pen environment, where protection for piglets is reduced. Overall, these data indicate that PNS females have their brain development shifted towards a pro-anxiety phenotype and that PNS can be causally related to subsequent impaired maternal behaviour in adult female offspring.

[Human recombinant erythropoietin in the treatment of anemia in patients on long-term hemodialysis].

Abstract:Recombinant human erythropoietin (EPO) was administered i.v. to anaemic patients (pts) on hemodialysis in doses from 40 to 120/IU/kg 3 times a week. 20 out of 21 pts showed an increase in hemoglobin (Hb) level above 11 g/dl after 8-12 weeks. Maintenance doses to keep Hb value about 10 g/dl varied from 2 X 40 IU/kg to 3 X 40 IU/kg per week (subcutaneous). EPO improved the well-being and physical condition in all of pts. Six pts developed rise in blood pressure and most an increase in predialysis serum potassium and urea levels during first 16 weeks of treatment.

Abstract:Abstract Maternal stress during critical periods of fetal development is linked to the modified expression of several physiological processes. Despite attempts to minimize chronic stress and enhance sow well-being, some animals still experience stress, which can have short and long-term effects on future progeny. Therefore, we evaluated the impact of maternal stress on the immune phenotype and performance of pregnant gilts who had experienced pre- and post-natal stress and whether their offspring were affected. A total of 11 primiparous females (201.32 ± 17.18 kg at farrowing) clustered into treatment groups based on the treatment of their dams were used in this study. Their dams had been randomly allotted to receive either a placebo (CON) or Hydrocortisone Acetate capsules twice per day for 21 d during either mid (MID; d 51 to 72) or late (LATE; d 81 to 102) gestation. Prenatally stressed dams were sampled on gestational d 45, 60, 75, and 105 to determine descriptive and functional immune analysis. Productivity measures were assessed, including farrowing duration and the interval between piglets. Blood samples were taken from a subset of piglets born to these dams on d 7, 14, and 21 of lactation and then 24 h, 7, and 14 d post-wean. Data were analyzed using the Mixed procedure of SAS with repeated measures. Numbers of lymphocytes were elevated in gilts from LATE-treated dams compared with those from MID or CON dams for the entire study period (P < 0.05). All other descriptive immune measures, including neutrophil-to-lymphocyte ratio, were not different between groups during the gestational period. Mitogen-induced lymphocyte proliferation did not differ across treatments for prenatally stressed dams. No maternal treatment effects for any litter traits, farrowing duration, or interval between each piglet, but there was a tendency (P < 0.10) for piglets born to MID-stressed dams to be heavier than those born to CON dams. Total lymphocyte numbers were least in the progeny of MID-treated dams compared with piglets from LATE and CON dams for the entire lactation period. Over time, progeny from LATE-treated gilts had a more stimulated B-cell proliferation index during lactation (P < 0.05). Conversely, Concanavalin-A-induced lymphocyte proliferation tended to be least in offspring from MID-treated dams compared with CON. They also tended to have less percentage and numbers of lymphocytes than offspring from LATE and CON dams for the entire post-wean period. The offspring from MID-treated dams also had the greatest (P < 0.05) neutrophil-to-lymphocyte ratio for the entire period. While prenatal stress had minimal impact on the immune expression of the dam, its influence was evident in their progeny, particularly with MID. Results indicate that the programming effects may only be apparent in later generations. The study also suggests that mid-gestation is a more sensitive period for maternal stress in fetal development.

Abstract:Introduction Early life interactions with the sow or humans can have lifelong consequences on welfare and performance in pigs. It was hypothesised that piglets that received increased maternal contact and positive human contact would display improved responses to stressful events and improved performance. Methods Ninety-eight litters were allocated to a 2 x 2 factorial design for maternal contact (MC+)/restricted maternal contact (MC-); and positive human contact (HC+)/no additional human contact (HC-). Modified farrowing crates were used to restrict maternal contact (MC-) and litters in the HC+ treatment received five minutes of daily positive human interaction (stroking). Behavioural and physiological responses were assessed during processing at three days of age and during a behaviour test in which piglets were introduced to an empty arena, novel object, human hand and human standing in the arena at 18 days of age. Observations of behaviour in the home pen and growth and survival of piglets to weaning were also assessed. Results At processing, MC- piglets had more squeals (P = 0.015), tended to have more grunts (P = 0.063) and struggle attempts (P = 0.079), and had increased plasma cortisol concentrations (P = 0.009) after processing than MC+ piglets. During the behaviour test, HC+ piglets were more active (P = 0.014) and had more vocalisations (P < 0.05) in the empty arena than HC piglets. Furthermore, HC+ piglets were more likely to approach a human standing in the arena (P = 0.006) than HC- piglets. MC- piglets were less active (P = 0.008) and were less likely to approach the novel object (P = 0.049) than MC+ piglets. MC+ piglets tended to have higher weaning weights (P = 0.055) and more MC+ piglets were successfully weaned (P = 0.022) than MC- piglets. There was no significant effect of HC on piglet performance. Discussion While positive handling did not affect behavioural and physiological stress response to processing, HC+ reduced fear of novelty and humans. These findings highlight the importance of early life experiences has on stress resilience early in life.

Abstract:This study investigated the impact of early life interactions on stress resilience of weaner pigs. The hypotheses tested were that reduced maternal contact increases the stress response of pigs to weaning, and that positive human contact during lactation reduces, the stress response of pigs to weaning. Ninety-eight litters were allocated to a 2×2 factorial randomised block design for treatments maternal contact (MC+) / reduced maternal contact (MC-); and positive human contact (HC+) / control (HC-). Modified farrowing crates were used to reduce maternal contact (MC-). Litters in the HC+ treatment received five minutes of daily positive human interaction (stroking). Treatments ceased at weaning (22 days old). Forty-three pigs were mixed into pens of the same treatment at weaning. Behavioral observations were conducted for 2 hours post-weaning and for 4 hours at 2 weeks post-weaning, along with behavioural responses to humans and isolation at 3 and 4 weeks post-weaning, respectively. Cortisol, haptoglobin and IgA concentrations were collected the day prior to weaning at rest, two hours post-weaning, one hour following isolation and at 4 weeks post-weaning and at 5 weeks post-weaning at rest (basal). End weight and survival were also recorded. Results showed that in the first 90 minutes after weaning, HC+ pigs exhibited more inactive behaviours (P0.05), MC+ pigs exhibited more eating and active behaviours (P<0.05) from 30 to 60 minutes post-weaning. There were no significant treatment effects (P<0.05) on the physiological measures except at 5 weeks post-weaning where the IgA concentrations of the MC-/HC+ pigs remained steady while those of the other pigs increased five fold (P=0.002). While there were no treatment effects (P<0.05) on the behaviour of pigs at rest at 2-weeks post-weaning, in response to isolation at 4 weeks post-weaning or on performance post-weaning, there was limited evidence that the MC- pigs were less fearful of humans in the test at 3 weeks post-weaning. Based on the behavioural response of pigs immediately following weaning, early positive handling assisted pigs to cope with the stress of weaning. While previous research provided evidence of maternal contact on stress resilience during lactation, the present study provides limited evidence of maternal contact on stress resilience of the weaned pig.

Abstract:This experiment was conducted to investigate the effects of early weaning stress on intestinal morphology,mucosal barrier and p38 mitogen-activated protein kinase(p38 MAPK) signaling pathway of piglets.Piglets(Duroc×Landrace×Yorkshire) of 12 litters were chosen and 2 to 3 piglets(adjusted for body weight)from each litter were weaned and served as a weaned group.The remaining unweaned piglets were continued to nurse and served as an unweaned group.Six piglets of each group were chosen to slaughter at 22,24,28 and 35 days of age,respectively.The results showed as follows: compared with the unweaned group,jejunal villus height and the ratio of villus height to crypt depth at 22,24 and 28 days of age in the weaned group were decreased(P0.05),while there was no significant difference at 35 days of age(P0.05).The ratio of villus height to crypt depth of the weaner piglets at 24 and 28 days of age was lower than that at 22 days of age(P0.05).The weaner piglets had higher jejunal villus height and the ratio of villus height to crypt depth at 35 days of age than those at 22,24 and 28 days of age(P0.05).Compared with the unweaned group,plasma D-lactate content and diamine oxidase activity were increased with the increasing duration of weaning and still higher than those in the weaned group at 35 days of age(P0.05).Weaning stress activated p38 MAPK signaling pathway and significantly increased the ratio of colonic mucosa phospho-p38 MAPK and total p38 MAPK expression at 24 days of age compared with 22 days of age(P0.05).The results indicate that early weaning induces the damage of intestinal morphology and mucosal barrier.Although damaged intestinal morphology is near to reinstitution entirely at 35 days of ages,intestinal mucosal barrier is not recovered,indicating the recovery of mucosal barrier is slower than that of morphology.Early weaning stress activates p38 MAPK signaling pathway.

Abstract:【Objective】 The experiment studied the effect of early-weaned stress on serum cortisol,intestinal morphology and permeability in piglets.【Method】 A total of 32 piglets were weaned at 21 d.Intestinal morphology,serum cortisol,DAO activity,plasma D-lactate level,and urine lactulose mannitol ratio were measured at 21,22,25,28,35 d of age.【Result】 The results showed that the serum cortisol levels increased immediately at 22 d(P0.01),and remained high level in the 1 week postweaning(P0.05),even at 35 d(P0.05);Intestinal morphogenesis obviously changed during the early stage of weaning.Villous height declined,and crypt depth increased,especially in the 1 week postweaning;the intestinal tissue repaired at 28 d and recovered at 35 d.Plasma D-lactate level,DAO activity and urine lactulose mannitol ratio increased immediately at 22 d and significantly remained high level to day 35 compared with that before weaning(P0.05).【Conclusion】 It is concluded that intestinal mucosal barrier is subject to serious damage for early-weaned stress,and permeability increase is obviously prior to morphological changes,but the permeability lags behind the restoration of the morphological.So,intestinal barrier function better reflects the state of intestine.

Abstract:Introduction Over the last 40 years, the domestic pig has emerged as a prominent preclinical model as this species shares similarities with humans with regard to immunity, gastrointestinal physiology, and neurodevelopment. Artificial rearing of pigs provides a number of advantages over conventional rearing (i.e., true maternal care), including careful control of nutrient intake and environment conditions. Yet there remains a gap in knowledge when comparing brain development between sow-reared and artificially reared domestic pigs. Thus, our research sought to model brain development and assess recognition memory in a longitudinal manner by directly comparing rearing environments. Methods Forty-four intact (i.e., not castrated) male pigs were artificially reared or sow-reared from postnatal day 2 until postnatal week 4. After postnatal week 4, all pigs were housed in a group setting within the same environment until postnatal week 24. Magnetic resonance imaging was conducted on pigs at 8 longitudinal time-points to model developmental trajectories of brain macrostructural and microstructural outcomes. Additionally, pigs behavior were tested using the novel object recognition task at postnatal weeks 4 and 8. Results Throughout the 24-week study, no differences between rearing groups were noted in weekly body weights, average growth and feed intake patterns, or feed efficiency. Whole brain, gray matter, white matter, and cerebrospinal fluid growth patterns also did not differ between pigs assigned to different early-life rearing environments. Moreover, minimal differences in regional absolute volumes and fractional anisotropy developmental trajectories were identified, though artificially reared pigs exhibited higher initial rates of myelination in multiple brain regions compared with sow-reared pigs. Furthermore, behavioral assessment at both PNW 4 and 8 suggested little influence of rearing environment on recognition memory, however, an age-dependent increase in object recognition memory was observed in the sow-reared group. Conclusion Our findings suggest that early-life rearing environment influences the rate of development in some brain regions but has little influence on overall brain growth and object recognition memory and exploratory behaviors in the domestic pig. Artificial rearing may promote maturation in certain brain areas but does not appear to elicit long-term effects in outcomes including brain structure or object recognition memory.

Abstract:A suboptimal early nutritional environment (i.e., excess of energy, sugar, and fat intake) can increase susceptibility to diseases and neurocognitive disorders. The purpose of this study was to investigate in nonobese Yucatan minipigs (Sus scrofa) the impact of maternal diet [standard diet (SD) vs. Western diet (WD)] during gestation and 25 d of lactation on milk composition, blood metabolism, and microbiota activity of sows (n = 17) and their piglets (n = 65), and on spatial cognition (n = 51), hippocampal plasticity (n = 17), and food preferences/motivation (n = 51) in the progeny. Milk dry matter and lipid content, as well as plasma total cholesterol and free fatty acid (FFA) concentrations (P < 0.05) were higher in WD than in SD sows. Microbiota activity decreased in both WD sows and 100-d-old piglets (P < 0.05 or P < 0.10, depending on short-chain FAs [SCFAs]). At weaning [postnatal day (PND) 25], WD piglets had increased blood triglyceride and FFA levels (P < 0.01). Both SD and WD piglets consumed more of a known SD than an unknown high-fat and -sucrose (HFS) diet (P < 0.0001), but were quicker to obtain HFS rewards compared with SD rewards (P < 0.01). WD piglets had higher working memory (P = 0.015) and reference memory (P < 0.001) scores, which may reflect better cognitive abilities in the task context and a higher motivation for the food rewards. WD piglets had a smaller hippocampal granular cell layer (P = 0.03) and decreased neurogenesis (P < 0.005), but increased cell proliferation (P < 0.001). Influence of the housing environment during sow gestation on maternal health, and offspring immunity and survival

Abstract:In pig husbandry, pregnant females are often exposed to stressful conditions that may affect their physiology, behaviour or health and consequently the ability of their offspring to survive and develop. This study compared two different housing systems from insemination to 106 days of gestation, a conventional (C) and an enriched one (E), for maternal cortisol and immune parameters, as well as for the immune function and survival of their offspring. The C and E systems differed essentially for the housing design (slatted floor, 2.4 m2 per sow vs a deep straw litter and 3.4 m2 per sow). Results from Expt 1 (n = 108) demonstrated that C sows had higher salivary cortisol than E sows at gestational Day 102 (DG 102). Results from Expt 2 (n = 48) showed that, at DG 102, C sows had higher granulocyte counts and total plasma immunoglobulin G concentration than E sows, whereas lymphocyte number, haematocrit and haptoglobin plasma concentration were similar. Results from Expt 3 (n = 47) showed that plasma immunoglobulin G level at 5 and 27 days of age, hematocrit, white blood cell numbers and in vitro lymphocyte proliferation of piglets at 27 days of age were not influenced by maternal housing conditions. However, reproductive data collected in Expt 2 and 3 indicated that pre-weaning mortality was higher in C than E litters. In conclusion, the C system was associated with markers of increased stress and microbial aggression in gestating sows, and with greater piglet mortality during the suckling period without identified alterations in piglet immune function.

The site of action of epidural fentanyl: what can be learned by studying the difference between infusion and bolus administration? The importance of history, one hopes.

Abstract:I t is now approaching a quarter of a century since “selective spinal analgesia” was suggested as a goal for the spinal administration of opioids (1). In an ensuing study of epidural meperidine, each patient received the same bolus dose of meperidine (100 mg) IV and epidurally on separate occasions. This study permitted within-subject comparisons of cerebrospinal fluid (CSF) and blood meperidine concentrations after the two routes of administration, in parallel with analgetic response (2). The rigorous study design permitted a clear conclusion that, at early time points, part of the analgetic effect of meperidine was due to CSF-borne drug, presumably acting at the spinal cord. However, at later time points, blood-borne meperidine reached analgetic blood concentrations in some patients, presumably resulting in multimodal analgesia (3). The same group subsequently measured blood, as well as lumbar and cervical CSF concentrations of fentanyl after an epidural fentanyl bolus dose (100 g); fentanyl concentrations were found to be large in lumbar CSF, negligible in cervical CSF, and small in blood (4). This study, in patients with severe back pain, suggested that single bolus doses of fentanyl exerted a localized spinal analgetic effect. It is extraordinary that at this late stage there is no clear evidence as to whether epidural fentanyl bolus or infusion exert predominantly spinal or supraspinal analgesia. The studies by Ginosar et al. (5,6) in this issue of Anesthesia & Analgesia provide an inventive attempt to solve an often asked and still controversial question: Does epidurally administered fentanyl act locally, on spinal opioid receptors, or systemically, on supraspinal opioid receptors? While the authors have framed their paper, in many places, in terms of an either/or dichotomy, they have set out to test a milder hypothesis that the analgetic effect observed with two pain stimulus models in healthy volunteers after the epidural administration of fentanyl is predominantly mediated by a spinal mechanism if the drug is injected as a bolus, and by a supraspinal mechanism if infused. Indeed, the experimental results obtained support this hypothesis QED! Or do they? Some pharmacokinetic-pharmacodynamic reasoning is appropriately used by the authors in interpreting the results, hence some further background is relevant. It is sensible not to think of supraspinal and spinal mechanisms in terms of a dichotomy. Despite commentary about lipophilic drugs like fentanyl distributing into epidural fat, countless studies with epidurally injected opioids and local anesthetics (remember that fentanyl and bupivacaine have similar physicochemical properties) have demonstrated that systemically absorbed drug has a similar blood concentration profile to that after IM injection. The biphasic absorption patterns found can be interpreted as a “portion” of the dose being absorbed reasonably rapidly with a half-life of around 5 to 10 min, generating the “peak” arterial blood concentration at around 10 min after injection. The remaining “portion,” presumably that distributed into fatty tissues, is absorbed more slowly with a half-life of several hours, thereby sustaining the blood drug concentrations compared with IV drug administration. Blood-borne drug will thus be delivered to both supraspinal and spinal receptor sites in proportion to the distribution of cardiac output, in addition to drug delivered by local mechanisms of bulk flow and diffusion (3,4,7). Although the amount of blood-borne drug delivered supraspinally may be small after epidural compared with IV injection, one has to remember, as pointed out in the early days of spinal opioid pharmacology, that the dual Accepted for publication August 13, 2003. Abstract:Early social experience has profound effects on a wide spectrum of behaviors and neurochemical correlates in the rat. Repeated separation of rat pups from their dam during the early neonatal period causes acute perturbation of neuroendocrine and physiological status. The chronic sequelae of repeated separations have not been studied as extensively as the acute responses. Altered social experience at a later developmental stage, postweaning isolation rearing, is known to induce enduring changes in the behavioral responses to reward and reward-related stimuli in maturity. We have evaluated the influence of repeated early maternal separation on the responses to both primary and conditioned incentives in mature rats. Separated animals showed enhanced weight gain, a blunted locomotor response to a novel environment and a blunting of the response to both negative and positive contrast effects. Female separated animals, but not males, exhibited a profound attenuation of the acquisition of a conditioned anticipatory locomotor response to the presentation of food. These data are discussed with respect to the putative involvement of ventral striatal dopamine systems in reward mechanisms and the potential utility of early maternal separation as an animal model of depression.
Effects of chronic stress: A comparison between tethered and loose sows

Abstract:The present study aimed to investigate whether long-lasting, recurrent tethering of sows leads to enduring effects on measures that may be indicative of chronic stress. Sows that had experienced tethering for about 1.5 or 4.5years and age-matched sows kept in a social housing system (loose sows) were compared. Immediately after slaughter, blood samples were taken to measure plasma cortisol levels, and the brain, spleen, and adrenals were dissected and weighed. Gene expression in the frontal cortex and hippocampus was analyzed. Plasma cortisol levels were higher in the tethered sows than in the loose sows. The older, but not the younger, tethered sows had heavier adrenal glands than their loose counterparts. The weight of the spleen was not affected by the housing conditions, but the pituitary gland was lighter in tethered sows than in loose sows. Microarray analyses revealed an increased expression of beta-globin mRNA in the hippocampus and to a lesser extent in the frontal cortex of the older tethered sows, compared with the older loose sows. Taken together, the findings indicate that chronically stressed pigs develop depression-like symptoms. However, it can be questioned whether the pig subjected to repeated, long-term stress can be regarded an animal model of major depression.

Abstract:AbstractRationaleEarly-life stress (ELS) can increase anxiety, reduce prosocial behaviors, and impair brain regions that facilitate emotional and social development. This knowledge greatly stems from assessing disrupted mother–child relationships, while studies investigating the long-term effects of father-child relationships on behavioral development in children are scarce. However, available evidence suggests that fathers may uniquely influence a child’s behavioral development in a sex-specific manner. Rodent models examining mother–offspring interaction demonstrate relationships among ELS, neuroinflammatory mediators, and behavioral development; yet, the role paternal care may play in neuroimmune functioning remains unreported.ObjectivesUsing the biparental California mouse (Peromyscus californicus), we examined to what extent paternal deprivation impairs social and anxiety-like behaviors, augments peripheral corticosterone (CORT) response, and alters central proinflammatory cytokine production following an acute stressor in adulthood.MethodsBiparentally reared and paternally deprived (permanent removal of the sire 24 h post-birth) adult mice were assessed for sociability, preference for social novelty, social vigilance, and social avoidance behaviors, followed by novelty-suppressed feeding (NSF) testing for general anxiety-like behavior. Following an acute stressor, circulating CORT concentrations and region-specific proinflammatory cytokine concentrations were determined via radioimmunoassay and Luminex multianalyte analysis, respectively.ResultsIn response to a novel same-sex conspecific, social vigilance behavior was associated with reduced sociability and increased avoidance in paternally deprived mice—an effect not observed in biparentally reared counterparts. Yet, in response to a familiar same-sex conspecific, social vigilance persisted but only in paternally deprived females. The latency to consume during NSF testing was not significantly altered by paternal deprivation. In response to an acute physical stressor, lower circulating CORT concentrations were observed in paternally deprived females. Compared to control-reared males, paternal deprivation increased hypothalamic interleukin-1β, but decreased hippocampal IL-6 protein concentration.ConclusionGreater social vigilance behavior was demonstrated in paternally deprived mice while they avoided social interaction with a novel same-sex conspecific; however, in response to a familiar same-sex conspecific, paternal deprivation increased social vigilance behavior but only in females. It is possible that different neurobiological mechanisms underlie these observed behavioral outcomes as sex-specific central proinflammatory cytokine and stress responsivity were observed in paternally deprived offspring.

Abstract:In a randomised double‐blind vehicle controlled trial of 0.05% tretinoin cream in the treatment of photodamaged skin, the histological results of paired biopsies from 28 individuals who applied tretinoin for 26 weeks are compared with 28 paired biopsies from a control group applying vehicle alone. There was a significant increase in epidermal thickness in the tretinoin‐treated group (P.001). Epidermal atrophy was reversed in ten patients applying tretinoin cream.

Abstract:Maternal separation in early life has been observed to have lasting, detrimental effects that impair personal and social development and can persist into adulthood. Maternal separation during infancy can be most detrimental during adolescence, leading to long-term adverse effects on development and social behavior. This research study compared the effects of sibling and maternal separation in infancy on anxiety, sociability, or memory later in adolescence (postnatal day, PND, 50–58) in male and female Long-Evans Rats ( Rattus norvegicus ). Rat pups were semi-randomly assigned into eight conditions for daily isolation (PND 1–14). The groups were separated by the duration of isolation between 15 minutes (control group) or 180 minutes (experimental group) and the sex of the rat. They were also separated by comfort conditions with the dam present in an adjoining cage versus not present and siblings present or not present during isolation. The result was a 2 (15-min vs. 180-min) x 2 (dam vs. no dam) x 2 (single vs. grouped) x 2 (male vs. female) design. Once pups had reached adolescence (PND 50), researchers tested for differences in anxiety, activity, and social behavior using elevated plus-maze, open field habituation, a three-chamber social interaction, and a social discrimination task. Results indicate that longer isolation was more stressful and caused lower body weight. The female rats showed more anxious behavior in the open field but only if they were in the shorter isolation group. Social interaction showed that the rats isolated with the dam had different effects of isolation. In males, shorter isolation with the dam increased sociability but decreased sociability in females. These complicated findings may be due to the effects of inoculation, which describes how moderate stress combined with comfort may produce adaptation or immunity to stress and affect males and females differently.

Abstract:Postpartum depression is a complex illness that often occurs in genetically predisposed individuals. Closely related inbred rat strains are a great resource to identify novel causative genes and mechanisms underlying complex traits such as postpartum behavior. We report differences in these behaviors between the inbred depression model, Wistar Kyoto (WKY) More Immobile (WMI), and the isogenic control Wistar Kyoto Less Immobile (WLI) dams. WMI dams showed significantly lower litter survival rate and frequency of arched back and blanket nursing, but increased pup-directed licking, grooming, and retrieval during postpartum days (PPD) 1–10, compared to control WLIs. This increased pup-directed behavior and the frequency of self-directed behaviors segregated during selective breeding of the progenitor strain of WKY, which is also a depression model. These behaviors are manifested in the WMIs in contrast to those of WLIs. Furthermore, habitual differences in the self-directed behavior between light and dark cycles present in WLIs were missing in WMI dams. Hypothalamic transcript levels of the circadian rhythm-related gene Lysine Demethylase 5A (Kdm5a), period 2 (Per2), and the maternal behavior-related oxytocin receptor (Oxtr), vasopressin (Avp), and vasopressin receptor 1a (Avpr1a) were significantly greater in the post-weaning WMI dams at PPD 24 compared to those of WLIs, and also to those of WMI dams whose litter died before PPD 5. Expression correlation amongst genes differed in WLI and WMI dams and between the two time-points postpartum, suggesting genetic and litter-survival differences between these strains affect transcript levels. These data demonstrate that the genetically close, but behaviorally disparate WMI and WLI strains would be suitable for investigating the underlying genetic basis of postpartum behavior.

Abstract:This study was designed to determine whether exposure to low-dose endotoxin (lipopolysaccharide; LPS) during gestation can enhance immunity to a subsequent LPS challenge in piglets after weaning. Pregnant sows (parity: 2.6 ± 1.4) were assigned to prenatal immune stimulation (PIS; n = 7; administered 2.5 μg/kg BW LPS, i.m.) or saline treatment groups (CON; n = 7) administered at day 78 ± 1.8 of gestation. From the two prenatal treatment groups, barrows ( n = 17 PIS, 17 CON) were identified at weaning (21 ± 1.3 day of age) to subsequently receive a post-weaning LPS challenge. On day −1, the pigs were fitted with indwelling jugular catheters and subcutaneous temperature loggers. On day 0, the pigs were challenged i.v. with LPS (10 μg/kg BW), and blood samples were collected at −2, 0, 1, 2, 4, 6, 8, 12, and 24 h relative to LPS challenge. There was a treatment × time interaction for subcutaneous temperature ( P < 0.01), where the temperature increased more quickly at 1 and 2 h post-challenge in PIS compared to CON pigs. There was a tendency ( P = 0.08) for less change in white blood cells, relative to baseline values, in PIS compared to CON pigs. There was a treatment × time interaction ( P = 0.01) for lymphocyte concentrations where the concentrations were reduced in PIS compared to CON pigs at 8 h post-challenge. There was also a treatment × time interaction ( P = 0.01) for the change in eosinophil concentrations, where there was less change in eosinophil concentrations from 1 to 12 h in PIS compared to CON pigs. There was a tendency ( P ≤ 0.06) for a treatment × time interaction for serum interleukin-6 (IL-6) and IL-8. Granulocyte-macrophage colony-stimulating factor tended to be greater, and tumor necrosis factor-α tended to be reduced in PIS compared to CON pigs ( P ≤ 0.08). These data suggest that exposure to endotoxin in utero may influence the postnatal innate immune response to endotoxin. More research is necessary to further understand the mechanism behind the differences observed and the potential long-term influence of prenatal immune stimulation on pig offspring.

Abstract:Rodent pups use a variety of ultrasonic vocalizations (USVs) to facilitate maternal care. Importantly, infant USV repertoires are dependent on both the age and early life experiences of the pups. We have shown that an adverse rearing environment modeled with the maternal separation (MS) paradigm alters caregiving behavior, but little is known about how pup USVs differentially elicit maternal attention. In the present study, maternal approach towards a vocalizing pup over a non-vocalizing pup was tested in a Y-maze apparatus at two developmental time points over the course of MS. At the postnatal day (P) 10, dams engaged in longer interaction times with the vocalizing pup compared to the non-vocalizing pup. This effect was modulated by rearing environment and the sex of the pup, with only MS dams spending more time with vocalizing male pups. As expected at P20, dams did not show a preference for either the vocalizing or non-vocalizing pups regardless of rearing environment, however, MS dams spent a greater amount of time in the center of the apparatus as compared to control dams, which can be interpreted as a measure of uncertainty and indecision. These effects are important considering the sex specific effects of MS exposure across all developmental stages. Our novel findings support the hypothesis that sex-specific pup-dam interactions may drive later life outcomes following adversity.
