Abstract:In this narrative review, the authors summarise the relationship between stress and behaviour and how dairy cattle cope with stressors. Based on the available literature, the most common stressors in intensive dairy cattle farming, such as pain, disease, heat stress, poor comfort caused by technology, and social stress, are surveyed. The authors describe how these stressors modify the behaviour of dairy cattle, influencing their feeding patterns, social interactions, and overall well-being. Additionally, the review explores the effectiveness of various coping mechanisms employed by dairy cattle to mitigate stress, including physiological adaptations and behavioural responses. This review is a valuable resource for understanding and grading stress in dairy cattle through behavioural reactions. Elucidating the intricate interplay between stressors and behaviour offers insights into potential interventions to improve animal welfare and productivity in dairy farming. Furthermore, this review highlights areas for future research, suggesting avenues for more comprehensive behavioural studies to enhance our understanding of stress management strategies in dairy cattle.

Abstract:Increasing stress resilience of livestock is important for ethical and profitable meat and dairy production. Susceptibility to stress can entail damaging behaviours, a common problem in pig production. Breeding animals with increased stress resilience is difficult for various reasons. First, studies on neuroendocrine and behavioural stress responses in farm animals are scarce, as it is difficult to record adequate phenotypes under field conditions. Second, damaging behaviours and stress susceptibility are complex traits, and their biology is not yet well understood. Dissecting complex traits into biologically better defined, heritable and easily measurable proxy traits and developing biomarkers will facilitate recording these traits in large numbers. High-throughput molecular technologies (“omics”) study the entirety of molecules and their interactions in a single analysis step. They can help to decipher the contributions of different physiological systems and identify candidate molecules that are representative of different physiological pathways. Here, we provide a general overview of different omics approaches and we give examples of how these techniques could be applied to discover biomarkers. We discuss the genetic dissection of the stress response by different omics techniques and we provide examples and outline potential applications of omics tools to understand and prevent outbreaks of damaging behaviours.

Abstract:Animal husbandry serves as a source of provision of high-quality animal protein to the entire world population. Farm animals are exposed to different types of stressors at various stages of their life cycle. The climate change phenomenon also imposes additional stress on the animals due to extreme temperature (heat and cold) and unavailability of feed, etc. Such stress stimuli induce the release of stress hormones such as epinephrine, norepinephrine and cortisol in animals. These stress hormones reduce the host's production, reproduction and also influence the immune mechanisms. Recent researches revealed the stress hormone responsiveness of micro-organisms. Stress hormones were shown to increase the bacterial growth and virulence mechanisms, and the infectious disease process. This concept gains significance in the stress-related disease outbreaks in animals. Abstract:ABSTRACT Living animals are constantly faced with various environmental stresses that challenge normal life, including: oxygen limitation, very low or high temperature, as well as restriction of water and food. It has been well established that in response to these stresses, tolerant organisms regularly respond with a distinct suite of cellular modifications that involve transcriptional, translational and post-translational modification. In recent years, a new mechanism of rapid and reversible transcriptome regulation, via the action of non-coding RNA molecules, has emerged into post-transcriptional regulation and has since been shown to be part of the survival response. However, these RNA-based mechanisms by which tolerant organisms respond to stressed conditions are not well understood. Recent studies have begun to show that non-coding RNAs control gene expression and translation of mRNA to protein, and can also have regulatory influence over major cellular processes. For example, select microRNAs have been shown to have regulatory influence over the cell cycle, apoptosis, signal transduction, muscle atrophy and fatty acid metabolism during periods of environmental stress. As we are on the verge of dissecting the roles of non-coding RNA in environmental stress adaptation, this Commentary summarizes the hallmark alterations in microRNA expression that facilitate stress survival.

Abstract:Stress in livestock reduces productivity and is a welfare concern. At a physiological level, stress is associated with the activation of inflammatory responses and increased levels of harmful reactive oxygen species. Biomarkers that are indicative of stress could facilitate the identification of more stress-resilient animals. We examined twenty-one metabolic, immune response, and liver function biomarkers that have been associated with stress in 416 Italian Simmental and 436 Italian Holstein cows which were genotyped for 150K SNPs. Single-SNP and haplotype-based genome-wide association studies were carried out to assess whether the variation in the levels in these biomarkers is under genetic control and to identify the genomic loci involved. Significant associations were found for the plasma levels of ceruloplasmin (Bos taurus chromosome 1-BTA1), paraoxonase (BTA4) and γ-glutamyl transferase (BTA17) in the individual breed analysis that coincided with the position of the genes coding for these proteins, suggesting that their expression is under cis-regulation. A meta-analysis of both breeds identified additional significant associations with paraoxonase on BTA 16 and 26. Finding genetic associations with variations in the levels of these biomarkers suggests that the selection for high or low levels of expression could be achieved rapidly. Whether the level of expression of the biomarkers correlates with the response to stressful situations has yet to be determined.

Abstract:In swine production, stress is a common encounter that leads to serious bacterial infection and adverse effects on growth performance. Though antibiotics have been frequently used to control pathogen spread, sustained negative impacts from antibiotics have been found to affect intestinal integrity and the immune system. Multiple nutritional strategies have shown potential to counteract stress and replace antibiotics, including functional amino acids, low protein diet, plant extracts, organic acids, prebiotics, probiotics, minerals and vitamins. These additives relieve the stress response in swine via different mechanisms and signal transduction pathways. Based on the overview of signaling pathways and stress models, this review highlights the potential of nutritional strategies in swine for preventing or treating stress-related health problems. For wider application in the pig industry, the dose ranges measured require for further validation in different physiological contexts and formulations. In the future, microfluid devices and novel stress models are expected to enhance the efficiency of screening for new anti-stress candidates.

Unfortunately, there is a lack of understanding about the neuronal mechanisms that underlie well-being on an individual level. Neuroplasticity in the hippocampus, the subventricular zone (SVZ), the olfactory bulb (OB) and the hypothalamus may be essential regulatory components in the context of farm animal behaviour and welfare that may be altered by providing environmental enrichment (EE). The importance of pre-and probiotics as a form of EE and the microbiota-gut-brain-axis (MGBA) has come under the spotlight in the last 20 years, particularly in the contexts of research into stress and of stress resilience. However, it could also be an important regulatory system for animal welfare in livestock farming. This review aims to present a brief overview of the effects of EE on physiology and behaviour in farm animals and briefly discusses literature on behavioural flexibility, as well as inter-individual stress-coping styles and their relationship to animal welfare. Most importantly, we will summarise the literature on different forms of neural plasticity in farm animals, focusing on neurogenesis in various relevant brain regions. Abstract:Dairy ruminants provide a major part of the livestock and agriculture sectors. Due to the increase in world population and the subsequent increase in dairy product demands, the dairy sector has been intensified. Dairy farming intensification and the subsequent increase in animal nutritional demands and the increase in the average global temperature as well have subjected animals to various stress conditions that impact their health and welfare. Various management practices and nutritional strategies have been proposed and studied to alleviate these impacts, especially under heat stress, as well as during critical periods, like the transition period. Some of the nutritional interventions to cope with stress factors and ensure optimal health and production are the inclusion of functional fatty acids and amino acids and feed additives (minerals, prebiotics, probiotics, essential oils and herbs, phytobiotics, enzymes, etc.) that have been proven to regulate animals' metabolism and improve their antioxidant status and immune function. Thus, these nutritional strategies could be the key to ensuring optimum growth, milk production, and reproduction efficiency. Abstract:Abstract High ambient temperatures are a critical challenge in the poultry industry which is a key producer of the animal-based food. To evaluate heat stress levels, various parameters have been used, including growth rates, blood metabolites, and hormones. The most recent advances have explored expression profiling of genes that may play vital roles under stress. A high ambient temperature adversely affects nutrient uptake and is known to modulate the expression of genes encoding for sodium-dependent glucose transporters, glucose transporters, excitatory amino acid transporters, and fatty acid-binding proteins which are responsible for the absorption of macronutrients in the intestine. Various defensive activities are stimulated to protect the cell of different tissues from the heat-generated stress, including expression of early stress response genes coding for heat shock protein (HSP), c-FOS like protein, brain-derived neurotrophic factor (BDNF), and neuronal nitric oxide synthase (nNOS); antioxidant enzyme genes such as superoxide dismutase (SOD), catalase (CAT), and nicotinamide adenine dinucleotide phosphate oxidase (NOX4); and immune-related genes such as cytokines and toll-like receptors (TLRs). The potential role of HSPs in protecting the cell from stress and their presence in several tissues make them suitable markers to be evaluated under heat stress. BDNF and c-FOS genes expressed in the hypothalamus help cells to adapt to an adverse environment. Heat causes damage to the cell by generating reactive oxygen species (ROS). The NOX4 gene is the inducer of ROS under heat stress, which is in turns controlled by antioxidant enzymes such as SOD and CAT. TLRs are responsible for protecting against pathogenic attacks arising from enhanced membrane permeability, and cytokines help in controlling the pathogen and maintaining homeostasis. Thus, the evaluation of nutrient transporters and defense mechanisms using the latest molecular biology tools has made it possible to shed light on the complex cellular mechanism of heat-stressed chickens. As the impacts of heat stress on the above-mentioned aspects are beyond the extent to which the reduced growth performance could be explained, heat stress has more specific effects on the regulation of these genes than previously thought. Abstract:Heat stress is one of the greatest challenges for the global livestock industries as increased environmental temperature and humidity compromises animal production during summer leading to devastating economic consequences. Over the last 30 years, significant developments have been achieved in cooling and provision of shade and shelter to mitigate heat stress reducing some of the losses associated with heat stress in farm animals. However, the recent increase in the incidence of heat waves which are also becoming more severe and lasting longer, due to climate change, further accentuates the problem of heat stress. Economic losses associated with heat stress are both direct due to loss in production and animal life, and indirect due to poorer quality products as a result of poor animal health and welfare. Animal health is affected due to impaired immune responses and increased reactive oxygen species production and/or deficiency of antioxidants during heat stress leading to an imbalance between oxidant and antioxidants and resultant oxidative stress. Research over the last 20 years has achieved partial success in understanding the intricacies of heat stress impacts on oxidative stress and immune responses and developing interventions to ameliorate impacts of heat stress, improving immune responses and farm animal health. This paper reviews the body of knowledge on heat stress impacts on immune response in farm animals. The impacts of heat stress on both cell-mediated and humoral immune responses have been discussed identifying the shift in immune response from cell-mediated towards humoral response, thereby weakening the immune status of the animal. Both species and breed differences have been identified as influencing how heat stress impacts the immune status of farm animals. In addition, crosstalk signaling between the immune system and oxidative stress has been considered and the role of antioxidants as potential nutritional strategies to mitigate heat stress has been discussed.

[The stress response in the yeast Saccharomyces cerevisiae]

Abstract:All living organisms are subject to changing environmental conditions, to which they must adapt in order to survive. Recently, there have been significant advances leading to the comprehension of the different mechanisms implicated in the responses to stressful situations in the yeast S. cerevisiae. In nature, as well as in laboratory conditions or industrial processes, this yeast is subjected to different adverse environmental situations, such as osmotic, thermal and oxidative stresses. A general stress response pathway, mediated by protein kinase A, allows S. cerevisiae to cope with these three stressful conditions. However, there are also specific response pathways that include the HOG kinase for osmotic stress, the Heat Shock Factor for thermal stress and Yap1p and Yap2p transcription factors that regulate the oxidative stress response, among other enzymatic and non-enzymatic mechanisms. In this review, we describe the perception and signal transduction pathways that regulate gene expression leading to the adaptation to most common types of stress in S. cerevisiae. We also include information regarding the interaction between the signal transduction pathways involved in the different responses that allow this organism to coordinate its various physiological processes for optimal adaptation to the changing environment.

Abstract:High ambient temperatures affect animal production and welfare in tropical and sub-tropical regions of the world. Feed intake, growth rate, mortality, egg production, hatchability and other production traits related to the economic success of the poultry industry are adversely affected by severe heat stress. In general, heat stress induces the activity of the neuroendocrine system, resulting in activation of the hypothalamic-pituitary-adrenal (HPA) axis, and elevated corticosterone (CORT) concentrations, which affects metabolism and immune responses. These include negative regulation of metabolic hormones, antibody production and heterophil to lymphocyte (H/L) ratio. Heat stress increases mitochondrial activity, causing reactive species overproduction which disrupts the antioxidant balance, leading to oxidative stress damage of membranes, protein and DNA. Heat stress stimulates the central nervous system (CNS), which significantly reduces daily gain, feed intake and FCR in poultry. Consequently, from an animal husbandry perspective, intervention strategies to relieve heat stress conditions have been the focus of many published studies. This review describes the effect of high temperature on production, behavioural, biochemical and immune responses, including oxidative damage that occur during heat stress in poultry, in broilers and laying hens. Moreover, nutritional interventions to alleviate the negative consequence of heat stress is discussed.

Abstract:The gastrointestinal tract provides the biological environment for nutrient digestion and absorption. Its physical and chemical barriers are crucial to protect from invading pathogens and toxic substances. On this basis, the intactness of the gastrointestinal tract, with its multiple functions and impacts, is one of the key prerequisites for human and animal health. Undoubtedly, the functions of a healthy gut system also largely benefit the welfare and performance of animals in farming systems such as poultry industries. Broiler chickens grow rapidly, as a result of rigorous genetic programs, due to the high absorption capacity of intestinal epithelia for nutrients, the quick transport of nutrients to the muscle, and their efficient conversion into energy and biomass. Due to oxygen metabolism or enteric commensal bacteria, intestinal epithelial cells create reactive oxygen and nitrogen species physiologically. However, increased generation of these oxidants goes along with the formation of free radicals resulting in oxidative stress causing lipid peroxidation and dramatic molecular changes in the structure and function of the cell and mitochondrial membranes. These effects contribute to chronic oxidative stress and inflammation of the gastrointestinal tract and generally affect all chicken organs, tissues, and cells. Hence, all forms of chronic stress, regardless of the origin, negatively impact the chicken's overall performance, health, and welfare. This review article highlights some enteric inflammation models and biomarkers to evaluate gut integrity in chickens and discusses the repercussions that chronic stress and intestinal inflammation have on the health and performance of commercial poultry
Frequency of Dividing Cells as an Estimator of Bacterial Productivity

Abstract:It has recently been proposed that the frequency of dividing bacterial cells (FDC) can be used to predict growth rates of natural aquatic bacterial assemblages. We have examined the relationship between FDC and growth rate in bacteria from southern-temperate, coastal marine waters by using incubation under conditions of manipulated nutrient availability and exclusion of bacterivores. The regression of the natural logarithm of bacterial instantaneous growth rate (μ) on FDC resulted in a better fit than regression of untransformed μ on FDC. The regression equation was ln μ = 0.299FDC − 4.961. The coefficient of variation for predicted ln μ at mean FDC was 7%. The range of FDC-estimated bacterial instantaneous generation times for coastal Georgia waters was 12 to 68 h, and range of calculated bacterial production rates was 0.6 to 17.6 mg of C·m−3· h−1. Unresolved problems of and suggested improvements on the FDC method of predicting growth rate are discussed.

Abstract:Key PointsThis review summarizes the evidence that chronic or repeated stressors can cause neuronal disturbances that resemble the changes that are observed in the brain during depression. We focus on the stress hormones that are secreted by the adrenal gland (that is, primarily, cortisol in humans and corticosterone in rodents), which are secreted after stimulation by peptides that are released from the brain (corticotropin-releasing hormone or CRH) and pituitary gland (corticotropin or ACTH). The corticosteroids act together with the neuropeptides to facilitate adaptation to stress, but if dysregulated, they fail to do so during depression. The goal of the review is to identify mechanisms that are driven by the hormones that might explain why maladaptive changes produce stress-related disorders, such as depression, in some individuals, whereas others remain in excellent health under similar adverse conditions.In the first part of the review, we highlight the adaptive stress-related processes in the limbic brain, which is an area that is involved in the appraisal of new experiences, learning and memory processes. The corticosteroid actions are mediated by nuclear receptors, which are abundantly expressed in these limbic neurons. Two receptor types have been identified: one receptor system (mineralocorticoid receptors or MRs) has a role in the appraisal process and, therefore, in the onset of the stress response; the other system (glucocorticoid receptors or GRs) facilitates recovery from stress. We postulate that MRs and GRs function in a binary fashion.In the second part of the review, we discuss how an inappropriate stress response might lead to a vulnerable phenotype. One model of such an inappropriate response is the chronically stressed animal that shows structural and functional changes in the limbic brain. These changes include aspects of neurogenesis, structural remodelling and synaptic plasticity that might be related to behavioural impairments. Other animal models focus on the role of genetic background and early experience in shaping stress reactivity. Conditions are discussed that programme adaptation to stress depending on mother–infant interactions.In the third and final part of the review, we address how, in genetically predisposed humans, stressful situations might precipitate hypercortisolaemia, as seen in depression, or hypocortisolaemia, as found in post-traumatic stress disorder (PTSD). In the most extreme cases of hypercortisolaemia, psychotic symptoms occur (steroid psychosis). This research has proceeded to the point that genetic factors can be identified in, for example, MR- and GR-related genes, which render individuals either resilient or vulnerable to developing affective disorders.The review concludes with the statement that, together with other stress-system mediators, a binary response system that is operated by corticosteroids preserves health and homeostasis. Their proper function determines the capacity of the organism to contain stress reactions in the acute phase and in the management of the late recovery phase. Imbalance might cause a vulnerable phenotype for mental illness to evolve.

Abstract:When confronted with severe environmental stress, some animals are able to undergo a substantial reorganization of their cellular environment that enables long-term survival. One molecular mechanism of adaptation that has received considerable attention in recent years has been the action of reversible transcriptome regulation by microRNA. The implementation of new computational and high-throughput experimental approaches has started to uncover the vital contributions of microRNA towards stress adaptation. Indeed, recent studies have suggested that microRNA may have a major regulatory influence over a number of cellular processes that are essential to prolonged environmental stress survival. To date, a number of studies have highlighted the role of microRNA in the regulation of a metabolically depressed state, documenting stress-responsive microRNA expression during mammalian hibernation, frog and insect freeze tolerance, and turtle and marine snail anoxia tolerance. These studies collectively indicate a conserved principle of microRNA stress response across phylogeny. As we are on the verge of dissecting the role of microRNA in environmental stress adaptation, this review summarizes recent research advances and the hallmark expression patterns that facilitate stress survival.

Abstract:Oxidative stress is caused by an imbalance between the production of harmful oxygen molecules and the body's ability to repair their detrimental implications, leading to reduced growth rates, increased disease susceptibility, and decreased reproductive performance in animals. Vitamin A, comprising retinol, retinal, and retinoic acid, is crucial for normal growth, reproduction, and vision. Vitamin A possesses antioxidant properties by directly scavenging reactive oxygen species, boosting antioxidant enzyme activity, and promoting antioxidant defence mechanisms. Numerous studies have shown that livestock with adequate levels of vitamin A in their diet experience reduced oxidative stress compared to those with vitamin A deficiency. Moreover, vitamin A supplementation can mitigate oxidative stress in animals exposed to stressful conditions like heat stress. Adequate vitamin A status in livestock through dietary interventions and improved animal management practices can significantly benefit animal health and well-being. However, further research is still needed to optimize dosing strategies and fully understand the relationship between vitamin A and oxidative stress in different animal species and production systems. Therefore, continued research efforts are essential to fully harness the potential of vitamin A as an effective tool for mitigating oxidative stress and improving animal welfare.
