Abstract:LZ-B-1, a water-soluble peptidoglycan, was prepared from the fruiting bodies of Ling Zhi or Reishi medicinal mushroom, Ganoderma lucidum, by ultrafiltration, DEAE-Sepharose Fast-Flow column, and Sepharose CL-6B column. To develop functional food or pharmaceuticals using LZ-B-1 and to investigate the relationship between the structure and bioactivity of LZ-B-1, the composition and immunomodulatory capacity of LZ-B-1 were studied herein. The peptidoglycan had a relative molecular weight of 1.12 x 10(4) Da, and total carbohydrate and peptide percentages were determined to be similar to 94.2% and similar to 5.4%, respectively. The carbohydrate moiety was mainly composed of L-fucose, D-galatose, and D-glucose in a molar ratio of approximately 1:5:2, together with small amounts of L-arabinose and D-mannose. The peptide moiety mainly contained arginine, threonine, glycine, serine, and aspartic acid in a molar ratio of approximately 1:1:2:2:1. Methylation reaction and GC-MS analysis indicated that the carbohydrate moiety mainly consisted of 1,6-disubstituted-galactopyranosyl, 1,2,6-trisubstituted-galactopyranosyl, 1-substituted-fucopyranosyl, 1,3-disubstituted- glucopyranosyl, 1,4,6-trisubstituted-glucopyranosyl, and 1-substituted-glucopyranosyl. Mouse spleen cell proliferation assay in vitro showed that a concentration of 200 mu g/mL of LZ-B-1 was the optimum concentration to promote the proliferation of mouse spleen lymphocytes (MSLs). In general, the higher the mouse spleen cell proliferation rate, the stronger the immunomodulatory activity. How the peptidoglycan stimulates the proliferation of MSLs and the bioactivity-structure relationship requires more research.

Abstract:Herein we describe the antioxidant, antimicrobial, antibiofilm, anti-inflammatory and wound-healing potential of aqueous and polysaccharide extracts from three widely appreciated mushrooms: Agrocybe aegerita, Laetiporus sulphureus and Agaricus bisporus. Moreover, we present their detailed phenolic, polysaccharide and protein profiles and ATR-FTIR spectra. The study found that polysaccharide extracts (PEs) from mushrooms had higher total and β-glucan levels than aqueous extracts (AEs), with A. aegerita showing the highest content. L. sulphureus had a higher total protein content, and A. aegerita AE had the highest phenolic content. Our results indicate that all the tested extracts have high potential regarding their bioactive properties, with A. aegerita being the most promising one. Namely, the antibacterial activity assay showed that the development of the skin-infection-causing agent, Staphylococcus aureus, was inhibited with a minimal inhibitory concentration of 4.00 mg/mL and minimal bactericidal concentration of 8.00 mg/mL, while the results regarding wound healing showed that, over the course of 24 h, the A. aegerita extract actively promoted wound closure in the HaCaT keratinocyte cell line model. The anti-inflammatory activity results clearly showed that when we used S. aureus as an inflammation-inducing agent and the A. aegerita aqueous extract in treatment, IL-6 levels reduced to the level of 4.56 pg/mL. The obtained data suggest that the tested mushroom extracts may serve as a source of bioactive compounds, with potential applications in the cosmeceutical, pharmaceutical and food industries. Furthermore, potential skin preparations carefully crafted with mushroom extract may help restore the skin's barrier function, decrease the probability of staph infections and minimize skin irritation.

Abstract:Ganoderma lucidum, a specie belonging to the class basidiomycetes, family polyporaceae of the order aphyllophorales has been widely utilized as a source of potential nutracuetical products. The cell wall of Ganoderma lucidum contains ergosterol which gets increased over different stage of the cultivation process. Ergosterol serves as the precursor for vitamin D, serves as the immunodulating agent in cancer suppression and has been the molecule of choice for antimycotic drug development. Thus, drug formulations of ergosterols or its conjugating complexes may enhance immunity and Vitamin D levels in the body. Ganoderma lucidum is also a rich source of pharmaceutical proteins and protein bound polysaccharides that can also serve as anti-cancer agents thereby improving the immune status of the individual. Other proteins found in Ganoderma lucidum are the morphogenesis related proteins such as hydophobins and an elaborate antioxidant enzyme machinery synthesized during the developmental stages. The intracellular and extracellular enzymatic profile of Ganderma exhibits enhanced activity during spawn run and pinning of the cultures. A positive role of hydrophobic amino acids and hydrophobin proteins has been identified in mushroom fructification process. Thus, medicinal mushroom such as Ganoderma can serve as a potential reservoir of bioactive compounds of pharmaceutic importance that can be utilized for prevention and cure of several disorders.

Abstract:The present study focused on the mushroom Ganoderma, which has been used in Eastern countries for centuries as a food and medicinal source. Specifically, the fruiting bodies of Ganoderma applanatum from the Kerala Forest Research Institute in Thirussur, Kerala, India, were analyzed for their nutritional and medicinal properties. The methanolic extracts of G. applanatum were used to examine secondary metabolites and proximate profiles, revealing the presence of various phytochemicals such as terpenoids, phenolics, glycosides, alkaloids, flavonoids, and saponins. Further analysis revealed the presence of significant amounts of calcium, sodium, phosphorus, and manganese. The compounds were characterized using chromatographic analysis, FTIR, and GC-MS, which revealed potential therapeutic compounds with C-H and C-O bonds in the amide group, β-glycosides, and C-C/C-O vibrations of phenolic substances. Mushroom extract at a concentration of 100 μg mL−1 exhibited potent antimicrobial activity against various pathogens. This study suggests that G. applanatum has a rich biochemical composition and pharmacological potential, making it a promising candidate for drug development and traditional medicine, and contributes valuable insights into its diverse therapeutic applications.

Evaluating the Application Potential of a Recombinant Ganoderma Protein as Bioactive Ingredients in Cosmetics

Abstract:The aim of this study was to evaluate the application potential of a recombinant fungal immunomodulatory protein from Ganoderma lucidum (rFIP-glu). First, a recombinant plasmid pPIC9K::FIP-glu-His was transferred into Pichia pastoris for the production of protein. The protein was then to assess its free radical scavenging abilities and the effect on the viability of both human immortalized keratinocytes (HaCaT cells) and mouse B16-F10 melanoma cells (B16 cells) in vitro, followed by the effect on the melanin synthesis of B16 cells. The results of SDS-PAGE and western blot showed that rFIP-glu was successfully expressed. Furtherly, a bioactivity assay in vitro indicated that the scavenging rate of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals reached 84.5% at 6.0 mg/mL (p ≤ 0.0001) of rFIP-glu, showing strong antioxidant activity. Subsequently, a safety evaluation demonstrated that rFIP-glu promoted the proliferation of HaCaT cells, with the cell viability reaching 124.3% at 48 μg/mL (p ≤ 0.01), regarding the cell viability of B16 cells after exposure to rFIP-glu (48 μg/mL) significantly inhibited, to 80.7% (p ≤ 0.01). Besides, rFIP-glu inhibited the melanin synthesis of B16 cells in a dose-dependent manner from 100–1000 μg/mL, and rFIP-glu at 500 μg/mL (p ≤ 0.01) exhibited the highest intracellular melanin amount reduction of 16.8%. Furthermore, a mechanism analysis showed that rFIP-glu inhibited tyrosinase (TYR) activity by up-regulating the expression of the microphthalmia-associated transcription factor (MITF) and down-regulating the gene expression of TYR and tyrosinase-related protein-1 (TYRP-1), thus inhibiting melanin synthesis. The data implied that rFIP-glu had significant antioxidant activity and whitening potency. It should be used as raw materials for cosmeceutical applications.

Abstract:Ganoderma lucidum, a species of the Basidiomycetes class, has been attracting international attention owing to its wide variety of biological activities and great potential as an ingredient in skin care cosmetics including "skin-whitening" products. However, there is little information available on its inhibitory effect against tyrosinase activity. Therefore, the objectives of this study were to investigate the chemical composition of G. lucidum and its inhibitory effects on melanogenesis. We isolated the active compound from G. lucidum using ethanol extraction and ethyl acetate fractionation. In addition, we assayed its inhibitory effects on tyrosinase activity and melanin biosynthesis in B16F10 melanoma cells. In this study, we identified a bioactive compound, ganodermanondiol, which inhibits the activity and expression of cellular tyrosinase and the expression of tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-associated transcription factor (MITF), thereby decreasing melanin production. Furthermore, ganodermanondiol also affected the mitogen-activated protein kinase (MAPK) cascade and cyclic adenosine monophosphate (cAMP)-dependent signaling pathway, which are involved in the melanogenesis of B16F10 melanoma cells. The finding that ganodermanondiol from G. lucidum exerts an inhibitory effect on tyrosinase will contribute to the use of this mushroom in the preparation of skin care products in the future.

Abstract:Ganoderma lucidum is an important medicinal mushroom with outstanding pharmaceutical properties, including anticancer, antioxidant, anti-inflammatory, and immunomodulatory activities due to its bioactive compounds. Such compounds could be suitably enhanced in mushrooms via an appropriate selection of growing substrates. This study aimed to evaluate the effect of phenolic-rich forest and agricultural wastes on the yield, antioxidant, and antimicrobial activities of G. lucidum . Oak sawdust (OS) (control) was used as a basal substrate to which phenolic-rich wastes, i.e., grape pomace (GP) and green walnut hulls (GWH), were mixed in percentages of 12.5, 25, and 50% each. Spawn running period (SRP), time to primordia initiation (DPI), time to first harvest (DFH), yield, and biological efficiency (BE) of G. lucidum were determined, and total phenolic content and antioxidant activity of the fruitbody were measured. The antimicrobial activity was assessed by examining extracts' effect on the growth of three Gram (+) and four Gram (−) bacteria and one yeast. Substrates supplemented with 12.5% GP significantly increased the yield and BE of G. lucidum (112.16 g/kg and 33%, respectively). Moreover, 12.5 and 25% GWH-supplemented substrates and 25% GP-supplemented ones resulted in a significant increase in the antioxidant capacity of the fruit bodies. The mushrooms cultivated on GP- and GWH-supplemented substrates exhibited higher antimicrobial activity when compared to those of OS. Our findings revealed that the incorporation of GP in the growing substrate of G. lucidum has the potential to improve its yield, antioxidant, and antimicrobial activities. Therefore, this agricultural waste can be successfully used as a good supplement in the production of G. lucidum .

Abstract:The cosmetic sector has experienced some of the strongest growth during the past ten years. In order to develop effective formulations, the cosmetic industry continually seeks high-quality organic components with pertinent bio-active properties such as anti-aging, anti-wrinkle, anti-pigmentation, anti-collagenase, antioxidant, and anti-inflammatory capabilities. There has been a noteworthy surge in the demand for cosmeceuticals utilizing fungal materials, driven by their unique biological and chemical characteristics in the past few decades. The sporophores of wild and cultivated mushrooms, are the sources of several nutraceuticals that can be used as nutricosmetics when taken orally. Not only the terrestrial members, fungi from the oceans generated more than 1000 metabolites, several of which are under investigation as potential cosmeceuticals and have therapeutic possibilities due to their diverse biological characteristics. Fungi-derived molecules such as kojic acid, chrysophanol, scytonemin, mycosporine-like amino acids, β-glucans, triterpenoids (ganoderic acids, ergosterols), ergothioneine, ceramide and chitosans are some key active components influencing the efficacy of cosmetic products. This review concentrates on several fungi-based cosmeceuticals, providing insights into their synthesis, mechanism of action, effectiveness, safety, as well as potential impact on the cosmetic market.

Physicochemical characterization of a high molecular weight bioactive β-D-glucan from the fruiting bodies of Ganoderma lucidum

Abstract:A purified polysaccharide coded as GLP20 was obtained by precipitating a hot-water extract from Ganoderma lucidum fruiting bodies with 20% (V/V) ethanol. Its total carbohydrate content was 95.9%. Structural analysis showed that GLP20 was a β-(1→3)-linked d-glucan with a (1→6)-β-d-glucopyranosyl side-branching unit on every third residue. Cell culture study revealed that GLP20 can significantly increase NO production of RAW264.7 macrophages. The analysis of light scattering and high performance size exclusion chromatography (HPSEC) showed that the molecular weight and polydispersity of GLP20 was 3.75 × 10(6)Da and 1.36, respectively. GLP20 had a rigid chain conformation in aqueous solution. A conformation transition occurred in the alkaline solution with NaOH concentration larger than 0.15M. The transition from ordered structure to single chain happened when GLP20 was heated above 135°C in water solution and was irreversible as demonstrated by differential scanning calorimetry (DSC). GLP20 existed as random coils in DMSO.

Abstract:The eROSITA instrument on the satellite Spektrum-Roentgen-Gamma will perform an all-sky survey as well as pointed observations in the low and medium X-ray energy regime. The 7 PNCCD detectors for this instrument are developed and manufactured at the MPI Semiconductor Laboratory. We summarize the characterization of this type of PNCCD regarding the spectral redistribution function in the photon energy range from 100 eV to 11 keV. We present the results of Geant4 simulations compared to our measurements. We explain the observed spectral features and their origin inside the detector.

Abstract:Ganoderma lucidum ( G. lucidum ) is a medicinal mushroom that is known for its ability to produce compounds with physiological effects on human health. This research was undertaken to amplify the production of bioactive components of G. lucidum under optimal cultivation conditions, obtained in a submerged state and utilized in solid state fermentation, with the purpose of enhancing antimicrobial and anticancer activities. The results indicated that titanium dioxide (TiO 2 NPs), magnesium oxide nanoparticles (MgO 2 NPs), and B6, along with glucose syrup and CLS syrups, were the most effective for producing GA, while wheat starch and whey protein, along with MgO 2 NPs and B6 vitamin, stimulated polysaccharide production using the One Factor at a Time (OFAT) method. After screening, the response surface method (RSM) statistically indicated that the media containing 42.11 g/L wheat starch with 22 g/L whey protein and 50 g/L glucose syrup with 30 g/L CSL were found to be the best conditions for polysaccharide (21.47% of dry weight biomass) and GA (20.35 mg/g dry weight biomass) production, respectively. The moss of the fruit body of G. lucidum produced under optimal GA conditions had the highest diversity in flavonoids and phenolic acids and significant antimicrobial activity against Esherichia coli ( E. coli ) and Bacillus subtilis ( B. subtilis ). In addition, the IC50 levels of shell and stem of G. lucidum were 465.3 and 485.7 μg/mL, respectively, while the moss did not reach 50% inhibition. In the end, the statistical approaches utilized in this research to elevate the levels of bioactive components in the fruiting body of G. lucidum produced a promising natural source of antimicrobial and anticancer agents.

Primary ovarian fibrosarcoma with long‐term survival: A report of two cases

Abstract:We report on two cases of primary ovarian fibrosarcoma detected in two women aged 44 and 34 years old. In the first case, an 18 × 17 × 10 cm multilobular tumor was detected in the left ovary. In the second case, a 13 × 9 × 3 cm and a 7 × 6 × 2 cm tumor were found in the left and right ovaries, respectively. In addition, a large quantity of ascitic fluid was detected. In both cases, a total hysterectomy with bilateral adnexectomy and an omentectomy were performed. No evidence of lymph‐node enlargement was detected in either case. Microscopic findings demonstrated densely, cellular spindle‐shaped tumor cells with increased mitotic activity (17 and 8/10 high‐power fields, respectively). Immunohistochemical analyses showed that tumor cells were positive for vimentin and negative for smooth muscle actin, S‐100, and estrogen receptor. Progesterone receptor was positive only in the second case. After surgery, both patients received several courses of combination chemotherapy. Neither patient demonstrated any evidence of disease recurrence during follow‐up for 10 years and 5 years, respectively.

Abstract:Ganoderma lucidum is a wood-degrading basidiomycete with numerous pharmacological effects. Since the mushroom is very rare in nature, artificial cultivation of fruiting bodies has been known on wood logs and on sawdust in plastic bags or bottles. Biotechnological cultivation of G. lucidum mycelia in bioreactors has also been established, both on solid substrates and in liquid media by submerged cultivation of fungal biomass. The most important pharmacologically active constituents of G. lucidum are triterpenoids and polysaccharides. Triterpenoids have been reported to possess hepatoprotective, anti-hypertensive, hypocholesterolemic and anti-histaminic effects, anti-tumor and anti-engiogenic activity, effects on platelet aggregation and complement inhibition. Polysaccharides, especially beta-d-glucans, have been known to possess anti-tumor effects through immunomodulation and anti-angiogenesis. In addition, polysaccharides have a protective effect against free radicals and reduce cell damage caused by mutagens.

Abstract:Mushrooms are edible fungi containing valuable nutrients. They provide attractive bio-active properties, which have confirmed anti-oxidants, anti-aging, and anti-inflammatory properties. Mushrooms possess abundant natural polymers affecting skin hydration and acting as moisturizers supporting skin barrier function. In this study, cloud ear mushroom (Auricularia polytricha) water extract (CW) was produced as a natural polymer to evaluate a new film-forming spray (FFS) containing CW to increase skin hydration and protect transepidermal water loss. CW contained polysaccharides as 748.2 ± 0.02 mg glucose/g extract. CW significantly inhibited the secretion of IL-6 and TNF-α and enhanced skin hydration by increasing aquaporin-3 (AQP3) and filaggrin (FLG) in HaCaT cells. The FFS was formulated using CW, sodium polystyrene sulfonate, and glycerin. The selected formulation contained brown Agaricus bisporus (BE-FFS) evaluated physical appearance, spray angle, spray pattern, and in vitro skin permeation. The BE-FFS has a transparent thin film with suitable occlusive properties, drying time, and physical appearance. Afterward, in vitro skin permeation and human hydration property studies presented the long-lasting effects and provided safety and hydration potential after 4 weeks of use. Overall, all results indicate that the BE-FFS is a natural film-forming spray for skin hydration improvement.

Abstract:Ganoderma a white rot fungus has been used as a folk remedy for promoting health and longevity for centuries. The vast amount of study has been performed on the medicinal properties of Ganoderma in general and Ganoderma lucidum in particular. The bioactivities of the metabolites reported from G. lucidum are immense. The main bioactive metabolites of G. lucidum consist of mainly polysaccharides and triterpenoids. The major bioactive polysaccharides isolated from Ganoderma species are β (1→3), β (1→4), and β (1→6)-D glucans. With respect to the pure chemical and structural points of view, G. lucidum polysaccharides are mostly composed of β-glucans, heteropolysaccharides, and glycoptoteins. The major component of this sugar molecule is glucose together with xylose, mannose, galactose, and fructose in different conformations. Many of these bioactive polysaccharides have shown activities against the major diseases of our time and the list of effects shown is huge. Various important bioactivities, namely, antitumor, antioxidant, cytotoxic, immunomodulatory, antibacterial, anti-inflammation, neuroprotective, hepatoprotective, anti-HIV, and so on have been shown by these bioactive polysaccharides. The main purpose of this review is to report the most bioactive polysaccharides from G. lucidum and other species of Ganoderma and to report their potential health benefits.

Abstract:Carbohydrates, including polysaccharide macromolecules, are the main constituents of the fungal cell wall. Among these, the homo- or heteropolymeric glucan molecules are decisive, as they not only protect fungal cells but also have broad, positive biological effects on the animal and human bodies. In addition to the beneficial nutritional properties of mushrooms (mineral elements, favorable proteins, low fat and energy content, pleasant aroma, and flavor), they have a high glucan content. Folk medicine (especially in the Far East) used medicinal mushrooms based on previous experience. At the end of the 19th century, but mainly since the middle of the 20th century, progressively more scientific information has been published. Glucans from mushrooms are polysaccharides that contain sugar chains, sometimes of only one kind (glucose), sometimes having several monosaccharide units, and they have two (α and β) anomeric forms (isomers). Their molecular weights range from 104 to 105 Da, and rarely 106 Da. X-ray diffraction studies were the first to determine the triple helix configuration of some glucans. It seems that the existence and integrity of the triple helix structure are criteria for their biological effects. Different glucans can be isolated from different mushroom species, and several glucan fractions can be obtained. The biosynthesis of glucans takes place in the cytoplasm, the processes of initiation and then chain extension take place with the help of the glucan synthase enzyme complex (EC 2.4.1.34), and the sugar units are provided by sugar donor UDPG molecules. The two methods used today for glucan determination are the enzymatic and Congo red methods. True comparisons can only be made using the same method. Congo red dye reacts with the tertiary triple helix structure, and the resulting glucan content better reflects the biological value of glucan molecules. The biological effect of β-glucan molecules is proportional to the integrity of the tertiary structure. The glucan contents of the stipe exceed the values of the caps. The glucan levels of individual fungal taxa (including varieties) differ quantitatively and qualitatively. This review presents in more detail the glucans of lentinan (from Lentinula edodes), pleuran (from Pleurotus ostreatus), grifolan (from Grifola frondose), schizophyllan (from Schizophyllum commune), and krestin (from Trametes versicolor), along with their main biological effects.

Abstract:Ganoderma has served as a valuable food supplement and medicinal ingredient with outstanding active compounds that are essential for human protection against chronic diseases. Modern pharmacology studies have proven that Ganoderma β-d-glucan exhibits versatile biological activities, such as immunomodulatory, antitumor, antioxidant, and antiviral properties, as well as gut microbiota regulation. As a promising polysaccharide, β-d-glucan is widely used in the prevention and treatment of various diseases. In recent years, the extraction, purification, structural characterization, and pharmacological activities of polysaccharides from the fruiting bodies, mycelia, spores, and fermentation broth of Ganoderma species have received wide attention from scholars globally. Unfortunately, comprehensive studies on the preparation, structure and bioactivity, toxicology, and utilization of β-d-glucans from Ganoderma species still need to be further explored, which may result in limitations in future sustainable industrial applications of β-d-glucans. Thus, this review summarizes the research progress in recent years on the physicochemical properties, structural characteristics, and bioactivity mechanisms of Ganoderma β-d-glucan, as well as its toxicological assessment and applications. This review is intended to provide a theoretical basis and reference for the development and application of β-d-glucan in the fields of pharmaceuticals, functional foods, and cosmetics.

Abstract:Among the most important sources of β-glucans are edible and medicinal mushrooms. These molecules are components of the cellular wall of basidiomycete fungi (mushrooms) and can be extracted even from the basidiocarp as the mycelium and its cultivation extracts or biomasses. Mushroom β-glucans are recognized by their potential effects as immunostimulants and immunosuppressants. They are highlighted as anticholesterolemic, anti-inflammatory, adjuvant in diabetes mellitus, mycotherapy for cancer treatment, as well as adjuvants for COVID-19 vaccines. Due to their relevance, several techniques of β-glucans extraction, purification, and analysis have already been described. Despite the previous knowledge of β-glucans' benefits for human nutrition and health, the main information about this topic refers to the molecular identification, properties, and benefits, as well as their synthesis and action on cells. Studies on biotechnology industry applications (product development) and the registered products of β-glucans from mushrooms are still limited and more common for feed and healthcare. In this context, this paper reviews the biotechnological production of food products containing β-glucans from basidiomycete fungi, focusing on food enrichment, and presents a new perspective on fungi β-glucans' use as potential immunotherapy agents.

Abstract:Ganoderma lucidum is a versatile mushroom. Polysaccharides and triterpenoids are the major bioactive compounds and have been used as traditional medicinal mushrooms since ancient times. They are currently used as nutraceuticals and functional foods. G. lucidum extracts and their bioactive compounds have been used as an alternative to antioxidants and antimicrobial agents. Secondary metabolites with many medicinal properties make it a possible substitute that could be applied as immunomodulatory, anticancer, antimicrobial, anti-oxidant, anti-inflammatory, and anti-diabetic. The miraculous properties of secondary metabolites fascinate researchers for their development and production. Recent studies have paid close attention to the different physical, genetic, biochemical, and nutritional parameters that potentiate the production of secondary metabolites. This review is an effort to collect biologically active constituents from G. lucidum that reveal potential actions against diseases with the latest improvement in a novel technique to get maximum production of secondary metabolites. Studies are going ahead to determine the efficacy of numerous compounds and assess the valuable properties achieved by G. lucidum in favor of antimicrobial and antioxidant outcomes.
Exploring the Biological Functions and Anti-Melanogenesis of Phallus indusiatus for Mushroom-Based Cosmetic Applications

Abstract:Phallus indusiatus, or bamboo mushroom, has been reported for its nutraceutical properties, while its cosmeceutical properties remain unclear. In this study, we conducted extractions of whole, fresh P. indusiatus using both aqueous and ethanolic methods. Among the extracts, ultrasonic-assisted extraction method with DI showed the highest antioxidant activity compared to the others. For cosmeceutical assessment, we evaluated the extracts’ inhibitory effects against ECM-degrading enzymes and found that they exhibited a modest inhibitory effect of approximately 50%. Remarkably, ultrasonic-assisted extraction with DI demonstrated promising cosmeceutical properties. Additionally, pressure-assisted extraction with DI showed a potentially protective effect against H2O2-induced DNA damage. To investigate the anti-melanogenic effect on MNT-1 cells, we treated them with the extracts and observed a significant decrease in cellular tyrosinase activity under α-MSH stimulation. This resulted in a relative reduction in melanin content. Notably, autoclaving extraction exhibited a significantly greater anti-melanogenic effect than the other extracts at the lowest concentration tested. Furthermore, the extracts demonstrated a reduction in NO production under LPS-induced inflammation. Hot water extraction with DI and ethanol exhibited a stronger anti-inflammatory effect compared to diclofenac, without any cytotoxicity. These findings highlight the hidden cosmeceutical properties of P. indusiatus and suggest its potential use as a bioactive ingredient in cosmetic formulations.
