Abstract:Background Waterlogging is one of the main abiotic stresses that limit wheat production. Quantitative proteomics analysis has been applied in the study of crop abiotic stress as an effective way in recent years (e.g. salt stress, drought stress, heat stress and waterlogging stress). However, only a few proteins related to primary metabolism and signal transduction, such as UDP - glucose dehydrogenase, UGP, beta glucosidases, were reported to response to waterlogging stress in wheat. The differentially expressed proteins between genotypes of wheat in response to waterlogging are less-defined. In this study, two wheat genotypes, one is sensitive to waterlogging stress (Seri M82, named as S) and the other is tolerant to waterlogging (CIGM90.863, named as T), were compared in seedling roots under hypoxia conditions to evaluate the different responses at proteomic level. Results A total of 4560 proteins were identified and the number of differentially expressed proteins (DEPs) were 361, 640, 788 in S and 33, 207, 279 in T in 1, 2, 3 days, respectively. These DEPs included 270 common proteins, 681 S-specific and 50 T-specific proteins, most of which were misc., protein processing, DNA and RNA processing, amino acid metabolism and stress related proteins induced by hypoxia. Some specific proteins related to waterlogging stress, including acid phosphatase, oxidant protective enzyme, S-adenosylmethionine synthetase 1, were significantly different between S and T. A total of 20 representative genes encoding DEPs, including 7 shared DEPs and 13 cultivar-specific DEPs, were selected for further RT-qPCR analysis. Fourteen genes showed consistent dynamic expression patterns at mRNA and protein levels. Conclusions Proteins involved in primary metabolisms and protein processing were inclined to be affected under hypoxia stress. The negative effects were more severe in the sensitive genotype. The expression patterns of some specific proteins, such as alcohol dehydrogenases and S-adenosylmethionine synthetase 1, could be applied as indexes for improving the waterlogging tolerance in wheat. Some specific proteins identified in this study will facilitate the subsequent protein function validation and biomarker development.
Comparative study of standard heterosis for yield and its attributes in bread wheat under two different water regimes

Abstract:Water stress conditions have occurred in the past in various places of the world, affecting the yield and production of numerous crops, including wheat. The aim of this research was to estimate standard heterosis under two different water regimes for grain yield and its attributes in 33 crosses, which were obtained by crossing 11 lines and 3 testers in a Line × Tester mating design. The best cross combinations for yield and contributing traits under irrigated condition were C4, C8, C33, C24, and C23, compared to both checks HD2967 and PBW660. Whereas, in rainfed condition, C18, C14, C26, C21, and C20 crosses were superior to the checks. For both irrigated and rainfed conditions, the best cross combinations identified were C29, C15, C32, C2, and C31. As a result, these cross combinations could be used in wheat breeding programmes to improve bread wheat genotypes for increased grain yield, agro-morphological features, and water stress tolerance. The presence of high heterosis for yield-contributing traits not only aids in the development of hybrids by exploiting heterosis but also in the production of transgressive segregants to develop elite lines.
Comparative Analysis of Root Transcriptome Profiles Between Drought-Tolerant and Susceptible Wheat Genotypes in Response to Water Stress

Abstract:Water deficit is one of the major factors limiting crop productivity worldwide. Plant roots play a key role in uptaking water, perceiving and transducing of water deficit signals to shoot. Although the mechanisms of drought-tolerance have been reported recently, the transcriptional regulatory network of wheat root response to water stress has not been fully understood. In this study, drought-tolerant cultivar JM-262 and susceptible cultivar LM-2 are planted to characterize the root transcriptional changes and physiological responses to water deficit. A total of 8197 drought tolerance-associated differentially expressed genes (DEGs) are identified, these genes are mainly mapped to carbon metabolism, flavonoid biosynthesis, and phytohormone signal transduction. The number and expression level of DEGs involved in antioxidative and antiosmotic stresses are more enhanced in JM-262 under water stress. Furthermore, we find the DEGs related to root development are much more induced in JM-262 in phytohormone signal transduction and carbon metabolism pathway. In conclusion, JM-262 may alleviate the damage of drought by producing more osmoprotectants, ROS scavengers, biomass and energy. Interestingly, hormone signaling and cross-talk probably play an important role in promoting JM-262 greater root systems to take up more water, higher capabilities to induce more drought-related DEGs and higher resisitance to oxidative stresse.

Abstract:Wheat, staple food crop of the world, is sensitive to drought, especially during the grain-filling period. Water soluble carbohydrates (WSCs), stem reserve mobilization and higher invertase activity in the developing grains are important biochemical traits for breeding wheat to enhance tolerance to terminal drought. These traits were studied for three accessions of Triticum dicoccoides(a tetraploid wheat progenitor species) - acc 7054 (EC 171812), acc 7079 (EC 171837) and acc 14004 (G-194-3 M-6 M) selected previously on the basis of grain filling characteristics. Check wheat cultivars- PBW-343 (a popular bread wheat cultivar for irrigated environments) and C-306 (widely adapted variety for rain-fed agriculture) were also included in this set. Analysis of variance revealed significant genotypic differences for the content of water soluble carbohydrates, activity of acid invertase and alkaline invertase. Acc 7079 was found to be a very efficient mobilizer of water soluble carbohydrates (236.43 mg g-1 peduncle DW) when averaged over irrigated and rain-fed conditions. Acid invertase activity revealed marked genotypic differences between wild and cultivated wheats. Alkaline invertase activity was highest in Acc 7079 when pooled across both the environments. On the whole, acc 7079 qualifies as a suitable donor for enhancing tolerance of bread wheat to terminal drought. The association of physio-biochemical differences observed with grain filling attributes on one hand and molecular markers on the other could be of use in improving wheat for water stress conditions.

Performance of wheat crops with different chromosome ploidy: root-sourced signals, drought tolerance, and yield performance

Abstract:Pot-culture experiments were carried out to estimate the role of non-hydraulic root signals (nHRS) and the relation of these signals to drought tolerance and grain yield formation under drought stress in six wheat varieties. These were two modern hexaploid wheat ( Triticum aestivum L . , AABBDD) Plateau602 and Longchun8139-2 , two diploid wheat ( Triticum monococcum L., AB) MO1 and MO4 , and two tetraploid wheat ( Triticum dicoccum Schuebl L., AABB) DM22 and DM31 . In the two diploid relatives, the nHRS was switched on and off at a soil water content (SWC) of approximately 53–45% field water capacity (FWC). In contrast, in the modern hexaploid varieties, Longchun8139-2 and Plateau602 the nHRS occurred between a SWC of about 71 and 35% FWC, a much wider soil moisture range. The two tetraploid relatives, DM22 and DM31 , were generally intermediate. The nHRS threshold range in SWC also narrowed as all six varieties went through successive developmental stages from shooting to grain filling. The two hexaploid wheat varieties had the longest duration of survival after the water supply ceased, and the best yield stability under drought stress, similar to with tetraploid wheat varieties; the diploid wheat varieties were least robust. These two parameters were both significantly correlated with the nHRS soil moisture threshold range ( r =0.9456** and 0.8608*, respectively). Based on these patterns, we propose a ‘triple Z’ model to describe the features of non-hydraulic stomatal sensitivity versus soil drought in wheat growth.
Effect of Water Deficit on Self-Thinning Line in Spring Wheat (triticum Aestivum L.) Populations

Abstract:Monocultures of spring wheat (Triticum aestivum L.) were grown at overcrowded densities (10 000 and 3 000 plants per m(2)) under well-watered and water-stressed conditions to investigate the effects of water deficits on self-thinning. The results showed that density reduction in water-stressed populations was delayed compared with that in well-watered populations. Populations grown in well-watered conditions conformed to the -3/2-power law. Compared with the well-watered condition, there was no significant decrease of the self-thinning line underwater-stressed conditions in this experiment, although the rate of average shoot biomass accumulation decreased. This result implied that the exponent of the -3/2-power equation is not as sensitive as the rate of average shoot biomass accumulation to water stress. Further analysis indicated that, in each density treatment, the lines of the height versus shoot biomass relationships did not differ significantly between the two water conditions. However, the intercepts of the height versus shoot biomass relationships were greater in the higher-density populations (10 000/m(2)) than those in the lower-density populations (3 000/m(2)). These results showed that water deficit did not change plant geometry in this experiment. That is to say, shoot competition for light remains constant at a given biomass, although root competition for water becomes more serious in water deficit conditions. Based on these results and previous reports we propose that, to affect the thinning line slope, changes in symmetric competition are not as efficient as changes in asymmetric competition.

Abstract:Stomatal density (SD) and stomatal index (SI) on the abaxial flag leaf epidermis, gas exchange of flag leaves and yield of six wheat ( Triticum aestivum L.) cultivars differing in drought resistance were studied in plant growth chamber. There were significant differences among cultivars in SD, SI, epidermal cell density (ED), photosynthetic rate (P n ), transpiration rate (E), instantaneous transpiration efficiency (ITE), shoot dry weight yield (Ys), grain yield (Yd) and whole growth season water use efficiency (WUE), but not in stomatal conductance (g s ). Wheat cultivars with moderate drought resistance had higher P n and Yd than cultivars with strong or weak drought resistance. SD was negatively correlated with ITE (r=−0.85) and WUE (r=−0.84), but not with P n , E and Yd. There were significant relationships between ED and Yd (r=−0.92), between SI and P n (r=0.84). These results revealed that breeding wheat cultivar only to raise drought resistance might cause yield decrease when water stress was not too severe. Wheat cultivars with lower SD might improve ITE and WUE, but P n and yield (Ys and Yd) were not influenced significantly. Greater ED was not beneficial to Yd. SI might be a criterion to use in selecting parents for producing high-yielding wheat cultivars.
Effects of Elevated CO2 Concentration and Drought on the Photosynthesis and Stomatal Resistance and Transpiration Efficiency of Different Strains of Spring Wheat

Abstract:Two strains of spring wheat (Triticum aestivum L-high productive Gaoyuan 602 and drought-tolerating Dingxi 24) were grown at two different CO2 concentrations (350 and 700 μmol·mol-1) and three soil water levels (80%, 60%, 40%FWC) in open-top-chambers from sowing until final harvest. We examined the response of photosynthesis and stomatal resistance to increasing CO2 concentration and drought and found that the photosynthesis rate and stomatal resistance and transpiration efficiency in the two spring wheats had increased. However, the positive effect of high CO2 on Dingxi 24 was bigger than that on Gaoyuan 602. Dingxi 24 showed higher water saving effect. Based on the analysis of the results, we predict that drought-tolerating spring wheat cultivars will have selective advantages in future high CO2 environment.
Water and Nutrient Use Efficiency in Diploid, Tetraploid and Hexaploid Wheats

Abstract:Three diploid (Triticum boeoticum, AA; Aegilops speltoides, BB and Ae. tauschii, DD), two tetraploid (T. dicoccoides, AABB and T. dicoccon, AABB) and one hexaploid (T. vulgare, AABBDD) varieties of wheat, which are very important in the evolution of wheat were chosen in this study. A pot experiment was carried out on the wheat under different water and nutrient conditions (i) to understand the differences in biomass, yield, water use efficiency (WUE), and nutrient (N, P and K) use efficiency (uptake and utilization efficiency) among ploidies in the evolution of wheat; (ii) to clarify the effect of water and nutrient conditions on water and nutrient use efficiency; and (iii) to assess the relationship of water and nutrient use efficiency in the evolution of wheat. Our results showed that from diploid to tetraploid then to hexaploid during the evolution of wheat, both root biomass and above-ground biomass increased initially and then decreased. Water consumption for transpiration decreased remarkably, correlating with the decline of the growth period, while grain yield, harvest index, WUE, N, P and K uptake efficiency, and N, P and K utilization efficiency increased significantly. Grain yield, harvest index and WUE decreased in the same order: T. vulgare > T. dicoccon > T. dicoccoides > Ae. tauschii > Ae. speltoides > T. boeoticum. Water stress significantly decreased root biomass, above-ground biomass, yield, and water consumption for transpiration by 47-52%, but remarkably increased WUE. Increasing the nutrient supply increased wheat above-ground biomass, grain yield, harvest index, water consumption for transpiration and WUE under different water levels, but reduced root biomass under drought conditions. Generally, water stress and low nutrient supply resulted in the lower nutrient uptake efficiency of wheat. However, water and nutrient application had no significant effects on nutrient utilization efficiency, suggesting that wheat nutrient utilization efficiency is mainly controlled by genotypes. Compared to the other two diploid wheats, Ae. squarrosa(DD) had significant higher WUE and nutrient utilization efficiency, indicating that the D genome may carry genes controlling high efficient utilization of water and nutrient. Significant relationships were found between WUE and N, P and K utilization efficiency.
Genotypic Differences in Morphological, Physiological and Agronomic Traits in Wheat (Triticum aestivum L.) in Response to Drought

Abstract:Drought is one of the main environmental factors affecting crop growth, and breeding drought-tolerant cultivars is one of the most economic and effective ways of increasing yields and ensuring sustainable agricultural production under drought stress. To facilitate the breeding of drought-tolerant wheat, this study was conducted to evaluate genotypic differences in the drought tolerance of 334 wheat genotypes collected from China and Australia with the aim of screening for drought-tolerant and -sensitive genotypes and to elucidate the corresponding physiological mechanisms. A hydroponic-air experiment (roots exposed to air for 7 h/d and continued for 6 d) showed significant genotypic differences in shoot and root dry weights among the genotypes. The relative shoot and root dry weights, expressed as the percentage of the control, showed a normal distribution, with variation ranges of 20.2–79.7% and 32.8–135.2%, respectively. The coefficients of variation were in the range of 18.2–22.7%, and the diversity index was between 5.71 and 5.73, indicating a rich genetic diversity among the wheat genotypes for drought tolerance. Using phenotypic differences in relative dry weights in responses to drought stress, 20 of each of the most drought-tolerant and drought-sensitive genotypes were selected; these were further evaluated in pot experiments (watering withheld until the soil moisture content reached four percent). The results showed that the trends in drought tolerance were consistent with the hydroponic-air experiment, with genotypes W147 and W235 being the most drought-tolerant and W201 and W282 the most sensitive. Significant genotypic differences in water use efficiency in response to drought were observed in the pot experiment, with the drought-tolerant genotypes being markedly higher and the two sensitive genotypes being no different from the control. A marked increase in bound water content in the drought stress plants was observed in the two drought-tolerant genotypes, while a decrease occurred in the free water. The reductions in photochemical efficiencies of PSII, transpiration rates, net photosynthesis rates, chlorophyll contents and stomatal conduction in the drought-sensitive genotypes W201 and W282 under drought stress were higher than the two tolerant genotypes. This study provides a theoretical guide and germplasm for the further genetic improvement of drought tolerance in wheat.

Abstract:Climate change is one of the major threats to wheat cultivation globally. Among abiotic stress, imposed by climate change, drought stress causes a drastic effect on yield and productivity of wheat. Based on this context, research was carried out on eight genotypes including WS-I (Water stress), WS-II, WS-III, WS-IV, WS-V, WS-VI, WS-VII and Khirman (check variety) to check the effect of water stress at the experimental field of Nuclear Institute of Agriculture, Tandojam. The experiment was laid-out in a split-plot design (SPD) with factorial arrangement having four treatments i.e., T1 (normal six irrigations), T2 (one irrigation), T3 (two irrigations) and T4 (three irrigations) and three replications during Rabi season, 2019-2020 in order to assess the response of wheat genotypes under different water regimes conditions for vegetative, yield and yield-related traits. The results of analysis of variance results showed that genotypes were significantly different for all traits except spike length (cm). Similarly, a significant difference was observed among the treatments for all the traits except harvest index (%). While genotype x treatments interaction showed a significance level for most of the yield associated traits except few characters such as days to 75% heading, grain filling period, plant height (cm), peduncle length (cm), spikelets spike-1 and grains spike-1 indicating that genotypes perform similarly over the treatment. Maximum mean performance for all the traits was recorded under T3 treatment compared to the T2 and T1. Among the genotypes, WS-1V perform best for grains spike-1, grain weight spike-1 (g), 1000-grain weight (g), grain yield plot-1 and harvest index (%) under all treatments. However, WS-III also performed consistently under T2 and T3 treatment. Thus, genotypes like WS-III and WS-IV could be preferred for growing in those areas where growers face the problem of water shortage. Also, WS-III and WS-IV can be used as donor genotypes for developing drought tolerant varieties.
Genome-wide association mapping of genomic regions associated with drought stress tolerance at seedling and reproductive stages in bread wheat

Abstract:Understanding the genetic architecture of drought stress tolerance in bread wheat at seedling and reproductive stages is crucial for developing drought-tolerant varieties. In the present study, 192 diverse wheat genotypes, a subset from the Wheat Associated Mapping Initiative (WAMI) panel, were evaluated at the seedling stage in a hydroponics system for chlorophyll content (CL), shoot length (SLT), shoot weight (SWT), root length (RLT), and root weight (RWT) under both drought and optimum conditions. Following that, a genome-wide association study (GWAS) was carried out using the phenotypic data recorded during the hydroponics experiment as well as data available from previously conducted multi-location field trials under optimal and drought stress conditions. The panel had previously been genotyped using the Infinium iSelect 90K SNP array with 26,814 polymorphic markers. Using single as well as multi-locus models, GWAS identified 94 significant marker-trait associations (MTAs) or SNPs associated with traits recorded at the seedling stage and 451 for traits recorded at the reproductive stage. The significant SNPs included several novel, significant, and promising MTAs for different traits. The average LD decay distance for the whole genome was approximately 0.48 Mbp, ranging from 0.07 Mbp (chromosome 6D) to 4.14 Mbp (chromosome 2A). Furthermore, several promising SNPs revealed significant differences among haplotypes for traits such as RLT, RWT, SLT, SWT, and GY under drought stress. Functional annotation and in silico expression analysis revealed important putative candidate genes underlying the identified stable genomic regions such as protein kinases, O-methyltransferases, GroES-like superfamily proteins, NAD-dependent dehydratases, etc. The findings of the present study may be useful for improving yield potential, and stability under drought stress conditions.

Effect of drought stress on residual transpiration and its relationship with water use of wheat

Abstract:Increasing the water use efficiency (WUE) of wheat (Triticum spp.) has long been a goal in semiarid areas. Low rates of residual (cuticular) transpiration are thought to improve yield potential of wheat under dry conditions, although the linkage is tenuous. The objective of this work was to investigate the association of residual transpiration with water use, WUE, and leaf water status in hexaploid (T. aestivum L.) and tetraploid (T. turgidum L. var. durum) genotypes grown under two watering regimes in two glasshouse experiments. Single plants were grown in 0.1-m × 1-m (0.1-m × 0.5-m in exp. 2 low-stress treatment) PVC tubes filled with soil. The watering regimes consisted of weekly replenishment of water used (low stress), or addition of sufficient water to ensure plant survival (high stress). At anthesis, flag leaf residual transpiration (rate of water loss from excised leaves), stomatal conductance, relative water content (RWC), and osmotic potential (exp. 1 only) were measured. Water use was not correlated with residual transpiration rate in either experiment. Residual transpiration rate did not differ for the two stress treatments in exp. 1, but there were significant (P < 0.01) genotype by stress treatment interactions. Residual transpiration rate was not related to plant water status (leaf RWC or osmotic potential) as had been reported in other studies. Induction of water deficit tolerance in wheat due to exogenous application of plant growth regulators: membrane stability, water relations and photosynthesis

Abstract:Our experiment was carried out in order to explore effects of plant growth regulators (PGR; thidiazuron, paclobutrazol, and ascorbic acid) on physiological traits of wheat genotypes under water surplus and deficit conditions. Study revealed that relative water content, membrane stability index, chlorophyll content, photosynthetic rate (PN), and maximal quantum yield of PSII improved with PGRs application across the genotypes both under irrigation and water stress. The response of HD 2733 genotype was more positive toward PGRs treatment as compared to other genotypes under water stress. Higher PN and chlorophyll contents were observed in HD 2987 followed by C 306 genotype under water-stress conditions. Moreover, Rubisco small subunit (SSU) expression was lower in wheat genotypes under water stress as compared to irrigated conditions. Application of PGRs led to upregulation of SSU under water stress, while no significant change was found in Rubisco level and activity under irrigated condition in dependence on PGRs treatments. Yield-related traits showed also significant reduction under water-stress conditions, while application of PGRs enhanced the yield and its components. Results indicated that the PGRs exhibited a positive interaction and synergetic effect on water stressed wheat plants in terms of photosynthetic machinery and yield.

Morphological, Physiological, and Biochemical Characterization of Drought-Tolerant Wheat (Triticum spp.) Varieties

Abstract:Wheat is one of the most important cereal crops and extensively cultivated in wide ranges of altitudes in Ethiopia. With an alarming population growth in the era of climatic change, there is a need for further crop improvement for sustainable production. In this regard, the study was carried out at the Kulumsa Agricultural Research Center (KARC) in a rainout shelter to investigate the responses of durum and bread wheat varieties to soil water stress in terms of selected morphological, physiological, and biochemical parameters. The 2 factors were combined factorially and arranged in a randomized complete block design with 3 replications. The 12 wheat varieties, 6 bread wheat and other 6 durum wheat, were sown in pots under well-watered (100% field capacity) and water-stressed (30% field capacity) conditions. Results revealed that water stress resulted in 26%, 9%, 23%, 16%, and 11% reductions in plant height, spike length, number of spikelets spike−1, relative water, and chlorophyll contents, respectively. The tested wheat varieties under water stress produced 28% and 6% more proline content and total soluble sugar, respectively, as mitigation strategies against drought. Results further exhibited that wheat varieties significantly differed in all of the measured traits except for the plant height and relative water content. The present study verified that the biochemical parameters needs to be considered as better traits to select wheat (Triticum spp.) varieties for drought tolerance under water stress conditions.
Production and scavenging of reactive oxygen species confer to differential sensitivity of rice and wheat to drought stress

Abstract:Drought poses a serious threat to crop production worldwide, and is expected particularly to affect rice production and hence food security. Given that wheat is known to tolerate drought better than rice, we compare rice and wheat (cv. Weebill) to understand the species level differences in drought adaptive mechanisms. We also compare two contrasting rice genotypes (IR64, drought susceptible, and Apo, drought tolerant) for such mechanisms under well-watered (100% field capacity, 100%FC) and water-limited (60%FC) conditions. The reduction in biomass of wheat under water limitation was smaller due to a higher rate of photosynthesis associated with maintenance of tissue turgor compared to rice genotypes. Drought caused greater inhibition of Photosystem II quantum efficiency, carboxylation efficiency, and photosynthetic capacity parameters in IR64 than in Apo. Transcript levels of photosynthesis-related genes were also significantly more repressed by water limitation in IR64, whilst the wheat genotype showed smaller reduction than Apo. Despite higher non-photochemical quenching (NPQ), a smaller increase in scavenging enzymes in IR64 resulted in more accumulation of reactive oxygen species (ROS) in 60%FC than in 100%FC compared to Apo. As a photoprotection mechanism, increased levels of NPQ resulted in lower ROS accumulation in wheat despite the similar increase in scavenging enzyme transcript levels as in Apo, signifying the importance of preventing oxidative burst for enhanced drought tolerance. In Apo, upregulation of the 9-cis-epoxycarotenoid dioxygenase 2 gene implies the use of xanthophyll pool for the ABA biosynthesis. Our data suggest that regulating photosynthesis and oxidative protection in the wheat genotype enhanced drought tolerance. Improving these traits for rice is crucial to develop drought-tolerant rice genotypes.
Physiological and antioxidant responses of synthetic hexaploid wheat germplasm under drought

Abstract:Background: As a result of the world population and climate change impact increases (especially in arid environments), there is a critical need for high-yield, drought-tolerant wheat. Synthetic hexaploid wheat derived lines (SHW-DL), were created artificially by crossing different durum wheat cultivars (AABB) with accessions of Aegilops tauschii (DD), a beneficial source of new genes for common bread wheat (Triticum aestivum L). Here, we studied the response of a panel of 91 SHW-DL for drought tolerance based on physiological, antioxidant enzyme activities, and drought tolerance indices. Results: A wide range of variation and high values of heritability observed for grain yield, physiological and antioxidant traits indicating that the SHW-DL panel constitutes a valuable gene source for drought tolerance improvement of wheat. Despite decreases in grain yield (YLD), leaf area index (LAI), and relative water content (RWC) an increase in the content of malondialdehyde (MDA) was observed. Moreover, drought streass increased the antioxidant enzyme activities of ascorbate peroxidase (APX), catalase (CAT) and peroxidase (POD), and also photosynthetic pigments, proline (Pro), and MDA content. With higher values of grain yield, physiological and biochemical traits such as photosynthetic pigments, and RWC, and lower content of MDA, and peroxidase (H2O2) activity, SHW-DL performed better compared to common wheat lines under water stress conditions. Conclusions: Different responses to water stress within the germplasm and between synthetic and common wheat suggest that selection for adaptive and suitable genotypes is possible for drought tolerance in synthetic wheat germplasm. Genotypes 54, 98, 102, 105, 122, 124, 143, 159, 196, and 198 were identified to be directly used in breeding programs or indirectly by crossing them with other wheat germplasm collections.
The Comparation of Drought Resistance Between Caragana Species (caragana Arborescens, C.korshinskii, C.microphylla) and Two Chickpea (cicer Arietinum L.) Cultivars

Abstract:Water stress is the main environmental factor limiting plantproductivity,especially in arid and semi-arid areas.Within limits,plants adapt to water stress through physiological and morphological adaptations,and leaf water potential(LWP) and leaf relative water content(RWC) are widely used to determine plant water status.Several studies have demonstrated the relationship between RWC and LWP to quantify the dehydration tolerance of tissues: tissues which maintain a high RWC as LWP decreases are more tolerant to dehydration.Caragana arborescens,C.korshinskii,C.microphylla,deciduous shrubs,commonly found in desert and semi-desert zones,northwestern China,and have important ecological and economic values: including playing a key role in vegetation succession from shifting sand dune to sandy grassland,helping to restore degraded land and serving as supplemental forage for livestock.Chickpea(Cicer arietinum L.),annual legume crop,is also considered one of the most drought-tolerant cool season food legumes in these areas.In the present study,predawn LWP and RWC were measured in C.arborescens,C.korshinskii,C.microphylla and two chickpea cultivars,a desi type,cv.Rupali,and a kabuli type,cv.Almaz,when subjected to water deficit in the glasshouse.For each species,20 plants were randomly designated to one of two treatments: well-watered control(WW) and water-stressed(WS).The WS treatment was imposed by cessation of watering and WW plants were watered every 2-3 days to maintain the soil water content(SWC) above 80% field capacity.RWC and LWP were measured at 2-3 day interval.The results showed that RWC in chickpea decreased with declining LWP and had significant linear relationship with LWP.However,in C.arborescens,C.korshinskii and C.microphyll,RWC did not decreased(maintaining a steady high water content about 100%) until LWP dropped below-2.4MPa,-2.5MPa and-1.5 MPa,respectively.After that,RWC decreased with declining LWP and had significant linear relationship with LWP.The results showed that RWC was a good parameter to determine the water status in chickpea,but as far as C.arborescens,C.korshinskii and C.microphyll were concerned,it was not.The maintenance of steady high water content in C.arborescens,C.korshinskii and C.microphyll at the beginning of water-deficit period maybe result from rigid and inelastic leaf cell wall in these species,which would provide the basis for sustaining high pressure potential and enhanced plant drought resistance.LWP(ψ)-RWC(R) relationships of these species were analyzed and the regressive slope of Caragana species were slightly lower compared to that of chickpea species,but minimum value of LWP in chickpea species was-4.5 MPa,and the corresponding relative soil water content was 14%,while in Caragana species,equivalent values were-6.7MPa and 6%,respectively.The results indicated that Caragana species had stronger abilty to resist drought stress than chickpea.A better adaptation to drought in Caragana species maybe attribute to accumulation of osmotically-active solutes in plant tissues under water stress.

Abstract:Climate change is emerging phenomena and causing frequent drought which lead to scaricity of water, which ultimately nagetively affecting wheat (Triticumaestivum L.) yield all around the world. The aim of this study was to explore the potential deought tolerant wheat genotypes for candidate genes exploration. This study was conducted during the year 2014-2015 at Plant Physiology Division, Nuclear Institute of Agriculture (NIA) Tandojam. The six wheat genotypes (cv. MT-1/13, MT-2/13, MT-3/13, MT-4/13 Chakwal-86 and Khirman) were investigated for their response at germination and seedling stage under different water stress treatments (0, -0.5, -0.75 and -1.0 MPa) in controlled conditions. The results of experiments with reference to genotypes revealed that genotype Chakwal-86 shows maximum seed germination (82.58 %), while the genotype Khirman shows maximum shoot length (7.23 cm), root length (15.1 cm), shoot fresh wt. (5.85 g 10-1shoots), root fresh wt. (3.45 g 10-1roots), shoot dry wt. (1.33 g 10-1shoots), root dry wt. (0.69 g 10-1roots). Among the genotypes tested Khirman and MT-4/13 are the tolerant genotypes had the potential to perform better under drought conditions, whereas MT-4/13 and Chakwal-86 were moderate tolerant under water stress conditions. Moreover, the genotypes i.e. MT-1/13 and MT-2/13 are the sensitive genotypes under drought environment. It is concluded from present in-vitro studies that osmotic stress significantly reduced the seed germination shoot/root length fresh and dry weight in all six wheat genotypes. The maximum reduction was found at higher osmotic stress induced by PEG-6000 (-1.0 MPa) significantly.

Abstract:Drought and heat stress are potential problems that can reduce wheat yield, particularly during the terminal growth stages in arid and semiarid regions of the world. The current study intended to examine the impact of individual and combined drought and heat stress on the biochemical contents (antioxidant enzymes, proline, soluble proteins, and soluble sugars), physiological parameters (chlorophyll content, cell membrane stability, photosynthesis, stomatal conductance, and transpiration), plant–water relations (relative water content, water potential, osmotic potential, and pressure potential), agronomic traits (flag leaf area, plant height, number of tillers per plant, spike length, grains per spike, and thousand-grain weight), and gene expression ( TaHSF1a, TaWRKY-33, TaNAC2L , and TaGASR1 ) in four different thermostable and drought-tolerant wheat genotypes (i.e., Gold-16, HS-240, Suntop, and Hemai-13) collected from different countries. The tri-replicate experiment was conducted using two factorial arrangements in a randomized complete block design (RCBD). All measured traits, except total soluble sugars, proline, and cell membrane stability index, showed significant reduction under both combined and individual treatments. Furthermore, correlation analysis revealed a significant association between biochemical and physiological characteristics and crop agronomic productivity. Furthermore, principal component analysis (PCA) and heatmap analysis demonstrated significant levels of variation in traits according to the type of stress and nature of wheat genotype. The spectrographs and micrographs generated by scanning electron microscopy for the selected high- and low- tolerance samples revealed clear differences in mineral distribution and starch granulation. All studied genes showed comparatively high levels of relative expression under combined treatments of drought and heat stress in all wheat genotypes, but this expression was the highest in 'Gold-16' followed by 'HS-240', 'Suntop', and 'Hemai-13'. Overall, this study concluded that plants are proactive entities and they respond to stresses at all levels; however, the tolerant plants tend to retain the integrity of their biochemical, physiological, and molecular equilibrium.
