Abstract:Nanoparticle-based vaccine delivery systems have been extensively used to promote and induce immune responses to protein antigens. The properties of the nanoparticles, such as size, surface charge, and antigen loading mode, have been proved to significantly influence the adjuvant effect and immunoreactivity of nanoparticle-based vaccine delivery systems. The purpose of the study was to investigate how the surface charge and antigen loading mode of nanoparticles impact the immune responses. In this study, three ovalbumin (OVA)-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles with different surface charges and antigen loading modes were developed. The three nanoparticles were designed as antigen encapsulated with negatively charged (Angelica sinensis polysaccharide (ASP)-PLGA/OVA), antigen encapsulated with polyethylenimine (PEI)-coated (ASP-PLGA/OVA-PEI), and antigen adsorbed on PEI-coated (ASP-PLGA-PEI-OVA) nanoparticles. The Angelica sinensis polysaccharide (ASP) was used as the immunopotentiator and encapsulated into three nanoparticles. The results demonstrated that both PEI-coated (positively charged) nanoparticles promoted the antigen escape from the endosome, which led to the cytoplasmic antigen delivery to generate cross presentation, compared to negatively charged nanoparticles. In addition, PEI-coated nanoparticles activated the DCs in lymph nodes 5 days after the primary vaccination. In vivo experiments demonstrated that both antigen-encapsulated nanoparticles induced more potent and long-term antigen-specific antibody responses, compared to that of antigen-adsorbed nanoparticles. Thus, the PEI-coated and antigen-encapsulated nanoparticles (ASP-PLGA/OVA-PEI) as a vaccine adjuvant delivery system have the potential to induce strong and long-term humoral and cellular immune responses.

Abstract:Enhancing immune response activation through the synergy of effective antigen delivery and immune enhancement using natural, biodegradable materials with immune-adjuvant capabilities is challenging. Here, we present NAPS L.p that can activate the Toll-like receptor 4 (TLR4) pathway, an amphiphilic exopolysaccharide, as a potential self-assembly adjuvant delivery platform. Its molecular structure and unique properties exhibited remarkable self-assembly, forming a homogeneous nanovaccine with ovalbumin (OVA) as the model antigen. When used as an adjuvant, NAPS L.p significantly increased OVA uptake by dendritic cells. In vivo imaging revealed prolonged pharmacokinetics of NAPS L. p -delivered OVA compared to OVA alone. Notably, NAPS L.p induced elevated levels of specific serum IgG and isotype titers, enhancing rejection of B16-OVA melanoma xenografts in vaccinated mice. Additionally, NAPS L.p formulation improved therapeutic effects, inhibiting tumor growth, and increasing animal survival rates. The nanovaccine elicited CD4 + and CD8 + T cell-based immune responses, demonstrating the potential for melanoma prevention. Furthermore, NAPS L.p -based vaccination showed stronger protective effects against influenza compared to Al (OH) 3 adjuvant. Our findings suggest NAPS L.p as a promising, natural self-adjuvanting delivery platform to enhance vaccine design across applications.

Abstract:The plant polysaccharides are recognized as an effective biological response modifier with low toxicity. In this study, the water-soluble polysaccharide from the roots of Actinidia eriantha (AEPS) was evaluated for its toxicity and adjuvant potential on the specific cellular and humoral immune responses to ovalbumin (OVA) in mice. AEP did not cause any mortality and side effects when mice were administered subcutaneously twice at the dose up to 5.0mg at intervals of 7 days. The mice were immunized subcutaneously with OVA 100μg alone or with OVA 100μg dissolved in saline containing Quil A (10μg) or AEPS (25, 50, or 100μg) on days 1 and 15. Two weeks later, splenocyte proliferation, natural killer (NK) cell activity, production and mRNA expression of cytokines from splenocytes, and serum OVA-specific antibody titers were measured. The Con A-, LPS-, and OVA-induced splenocyte proliferation and the serum OVA-specific IgG, IgG1, IgG2a, and IgG2b antibody titers in the immunized mice were significantly enhanced by AEPS (P<0.05, P<0.01 or P<0.001). AEPS also significantly promoted the production of Th1 (IL-2 and IFN-γ) and Th2 (IL-10) cytokines and up-regulated the mRNA expression of IL-2, IFN-γ, IL-4 and IL-10 cytokines and T-bet and GATA-3 transcription factors in splenocytes from the immunized mice (P<0.05, P<0.01 or P<0.001). Besides, AEPS remarkably increased the killing activities of NK cells from splenocytes in the immunized mice (P<0.01 or P<0.001). The results indicated that AEPS had strong potential to increase both cellular and humoral immune responses and elicit a balanced Th1/Th2 response, and that AEPS may be a safe and efficacious adjuvant candidate suitable for a wide spectrum of prophylactic and therapeutic vaccines.

Abstract:Nanoparticle vaccine delivery systems have been emerging strategies for inducing potent immune responses to prevent and treat infectious diseases and cancers. The properties of nanoparticle vaccine delivery systems, such as nanoparticle size, surface charge, and antigen release kinetics, have been extensively studied and proven to effectively influence the efficacy of vaccine responses. However, a few types of research have focused on the influence of administration routes of nanoparticle vaccines on immune responses. Herein, to investigate how the administration routes affect the immune responses of nanoparticles vaccines, we developed a nanoparticles system (NPs), in which the ovalbumin (OVA) and Angelica sinensis polysaccharide (ASP) were incorporated into poly(lactic-co-glycolic acid) (PLGA) nanoparticles and the polyethylenimine (PEI) was coated on the surface of nanoparticles. The NPs vaccine was intramuscularly and subcutaneously injected (im and sc) into mice, and the immune responses induced by these two delivery routes were compared. The results showed that both im and sc administration of NPs vaccines elicited strong antigen-specific IgG, IgG1, and IgG2a antibody responses, with no significant difference. In contrast, NP vaccines with sc administration significantly enhanced immune responses, such as enhancing the recruitment and activation of dendritic cells (DCs) in lymph nodes (LNs), promoting the antigen transport into draining lymph nodes, increasing the secretion of cytokines, improving the ratio of CD4+T cells to CD8+ T cells, activating cytotoxic T lymphocyte response, and inducing a strong cellular immune response. These results may provide a new insight onto the development of vaccine delivery systems.
Thymopentin treatment of selective IgA deficiency.

Abstract:Thymic hormones have been shown to modulate immunoglobulin production in a number of experiments and it is generally agreed that this action is mediated by modulation of helper and/or suppressor T cell activities. The possibility of upregulating the immunoglobulins is of particular relevance in patients with hypogammaglobulinemias and this paper reports on the results of thymopentin treatment in 9 patients with selective IgA deficiency. Two out of 4 patients responded positively in an open-label trial; in one the serum IgA values remained stable up to 8 weeks after discontinuation of treatment whereas there was a rapid fall in the other. Both responders had consistently normal T4/T8 ratios during the treatment, whereas the nonresponders revealed high ratios with large fluctuations of the T4/T8 ratio. In a subsequent (still ongoing) double-blind trial in 5 patients (3 thymopentin, 2 placebo) no significant change of serum or secretory IgA levels has been observed. Taken together, the data suggest that the tested dose regimen of thymopentin (i.e. daily i.m. injections of 1 mg/kg for 2 weeks, then same dose 3 time weekly for 10 weeks) may only work in a subset of patients with selective IgA deficiency. In the present study we did not attempt to distinguish to which of the three known subgroups the 9 patients belonged, nor did we try alternative dose regimens of thymopentin.

Abstract:Recently, nanoparticles have been widely used in drug and vaccine adjuvant delivery. Dendrobium devonianum Polygonatum (DP), a main biologically active ingredient isolated from Dendrobium devonianum, has been widely used in the clinic as an immunostimulant to stimulate and improve immune responses, contributing to its excellent biological activity. To increase the immune efficacy of DP, macrophage cell membrane-coated drug nanocrystals featuring homologous immune escape, targeting ability and low toxicity are in high demand. In this study, a new drug and vaccine adjuvant delivery system, PEI-MM-PLGA-DP/OVA, was designed and developed. This study aimed to report the macrophage immunomodulatory activity of PEI-modified macrophage cell membrane-coated PLGA nanoparticles encapsulating Dendrobium devonianum polysaccharides. PEI-MM-PLGA-DP/OVA could promote antigen uptake by macrophage and lymphocyte proliferation, increase the expression levels of MHC II, CD80 and CD86, and upregulate the ratio of CD4+ to CD8+ T cells in immunized mice. PEI-MM-PLGA-DP/OVA induced the highest TNF-α, IFN-γ, IL-4, and IL-6 cytokine secretion levels and the levels of OVA-specific antibodies (IgG) compared with the other groups. The above results indicated that PEI-MM-PLGA-DP/OVA had better adjuvant activity than PLGA-DP/OVA and MM-PLGA-DP/OVA.

Abstract:Recent pharmacological studies have demonstrated that Viola philippicapolysaccharide (VPP) exhibits a modulating effect on immune activity. However, its utilization has been hampered by its large particle size and complex spatial structure. Poly‐lactic‐co‐glycolic acid copolymer (PLGA) is recognized as an effective drug delivery carrier, exhibiting excellent biochemical properties. In this experiment, VPP was encapsulated with PLGA to form Viola philippica polysaccharide PLGA nanoparticles (VPP‐PLGA NPs). The morphological structure and immunomodulatory effects of VPP‐PLGA NPs were evaluated. The particle size of VPP‐PLGA NPs was reduced compared to VPP, and the optimal preparation conditions were as follows: the ratio of the organic phase to the internal aqueous phase was 8:1, the ratio of the external aqueous phase to the primary emulsion was 7:1, and the concentration of PLGA was 20 mg/mL. Additionally, VPP‐PLGA NPs significantly increased the NO content, IL‐4, and IFN‐γ levels in RAW264.7 cells, as well as enhanced their phagocytic activity. Furthermore, VPP‐PLGA NPs were found to increase NO content and IFN‐γ secretion in bone marrow‐derived dendritic cells (BMDCs). These findings suggest that VPP‐PLGA NPs could enhance the immune activity and may be utilized as a VPP delivery system for inducing strong immune responses.

Abstract:Alhagi honey polysaccharide (AHP) exhibit an excellent immune adjuvant effect, but low bioavailability in the body limits its application. Cationic polymer-modified poly (lactic-co-glycolic acid) (PLGA) nanoparticles have been widely investigated as vaccine delivery systems owing to their excellent antigen-loading efficiency. In this study, three kinds of cationic polymer were used to coat AHP-encapsulated PLGA nanoparticles (AHPP) to build positively charged antigen carriers. Among them, H5N1-loaded PEI-AHPP formulation could induce highest hemagglutination inhibition (HI) titer, IgG-subtype, and cytokines, activated dendritic cells (DCs) in lymph nodes, and CD3e+CD4+ and CD3e+CD8a+ T cells in the spleen of immunized mice. PEI-AHPP could stimulate DCs to highly express MHCI and MHCII molecules and had good antigen slow-release effect at the injected site along with lymph node targeting. These findings demonstrate that PEI-AHPP has the potential to be an effective adjuvant to induce strong and long-lasting Th1 and Th2 mixed immune responses.

Abstract:In order to enhance the cellular immune response of vaccines, numerous vaccine delivery systems have been developed, especially cationic nanoparticle carriers. Cationic polymer dots (PDs) have been widely used for biomedical imaging and drug delivery due to their excellent photoluminescence, small size and abundant positive charge. In this study, polyethyleneimine (600 Da) (PEI600)-modified redox-responsive hyperbranched poly(amido amine) (PAA-PEI600) and partially carbonized PAA-PEI600 PDs were designed and prepared as vaccine carriers to deliver the model antigen protein ovalbumin (OVA). Then, OVA-specific immune responses induced by PAA-PEI600/OVA and PDs/OVA nanoparticles were evaluated in vivo. The results suggest that the PAA-PEI600/OVA and PDs/OVA nanoparticles enhanced OVA-specific immune responses when compared to OVA alone. Further, PDs/OVA nanoparticles induced more potent OVA-specific cellular immune responses, including higher levels of the OVA-specific IgG2a/IgG1 antibody ratio, splenocyte proliferation, IL-12 and IFN-γ cytokines, maturation of dendritic cells, effector memory CD4+ T cells and CD8+ T cells as well as cytotoxic T lymphocytes (CTLs) than PAA-PEI600/OVA nanoparticles did. Moreover, subcutaneously injected PDs/OVA nanoparticles significantly inhibited tumor growth of the mice bearing E.G7-OVA tumor and extended mice survival. All the results show that immunization with PDs/OVA nanoparticles elicited more effective OVA-specific cellular immune responses. PDs could serve as promising vaccine delivery systems for cancer immunotherapy.

Abstract:Polysaccharides derived from Eucommia ulmoides leaves have immune-regulating, hypoglycemic, anti-tumor, and other beneficial functions. As a drug delivery system, polylactic-glycolic acid copolymer (PLGA) offers advantages like immunogenicity enhancement, improved drug absorption, and increased drug bioavailability. Currently, macrophage membranes are successfully employed in biomimetic nanomaterials to enhance drug utilization and extend drug administration time in the body, exhibiting good immunogenicity and organ targeting. In this study, we developed biomimetic PsEUL PLGA nanoparticles (NPs) coated with macrophage membrane (MM-PPsEUL) and examined their immune activity in vitro and in vivo. In vitro , MM-PPsEUL increased the phagocytic efficiency of macrophages without affecting their activity. In vivo , MM-PPsEUL significantly elevated the immune organ index of mice and upregulated the level of ovalbumin-specific IgG antibodies in serum. Simultaneously, it increased the expressions of cytokines IFN-γ and IL-4, as well as the levels of TLR4, MyD88, TRAF6, and NF-κB p65 proteins in mice spleen. MM-PPsEUL also exhibited a targeting effect on inguinal lymph nodes and slowed down the release rate of antigens, thereby inducing a prolonged immune response. In summary, MM-PPsEUL provides sustained release capabilities and can induce both humoral and cellular immune responses. This makes MM-PPsEUL NPs a promising antigen-targeted delivery system.

Sensitivity of the nasal vessels of the cat to chemical agents after sympathetic and parasympathetic denervation.

Abstract:The nasal mucosa was denervated sympathetically and parasympathetically in 19 cats and the effects of phenylephrine, noradrenaline and adrenaline on the vessels were recorded as pressure changes in water-filled balloons located in the nasal cavities. One of the cavities was used as a control in the same cat. The responses to noradrenaline were significantly greater on the sides where the superior cervical ganglia had been extirpated 2 to 8 weeks earlier. These differences are not mainly due to supersensitivity, but probably to effects of dilated blood vessels in the denervated sides. When both sides were sympathetically denervated, one chronically and the other acutely, the responses differed significantly only when the durations were compared and these differences seem to be due to supersensitivity. No significant differences in threshold doses and in responses to supraliminal doses of the drugs could be demonstrated when the postganglionic parasympathetic nerves had been cut.
Enhanced immunization via dissolving microneedle array-based delivery system incorporating subunit vaccine and saponin adjuvant

Abstract:PURPOSE: To enhance the immunogenicity of the model subunit vaccine, ovalbumin (OVA) was combined with platycodin (PD), a saponin adjuvant. To reduce the toxicity of PD, OVA, and adjuvant were loaded together into liposomes before being incorporated into a dissolving microneedle array.METHODS: OVA- and PD-loaded liposomes (OVA-PD-Lipos) were prepared using the film dispersion method. Their uptake behavior, toxicity to mouse bone marrow dendritic cells (BMDCs), and hemolytic activity to rabbit red blood cells (RBCs) were evaluated. The OVA-PD-Lipos were incorporated into a dissolving microneedle array. The chemical stability of OVA and the physical stability of OVA-PD-Lipos in microneedle arrays were investigated. The immune response of Institute of Cancer Research mice and potential skin irritation reaction of rabbits to OVA-PD-Lipos-MNs were evaluated.RESULTS: The uptake of OVA by mouse BMDCs was greatly enhanced when OVA was prepared as OVA-PD-Lipos, and in this form, the toxicity of PD was dramatically reduced. OVA was chemically stable as OVA-PD-Lipos, when OVA-PD-Lipos was incorporated into a dissolving microneedle array. Institute of Cancer Research mice treated with OVA-PD-Lipos-MNs showed a significantly enhanced immune response. PD combined with OVA elicited a balanced Th1 and Th2 humoral immune response in mice, with minimal irritation in rabbit skin.CONCLUSION: The dissolving microneedle array-based system is a promising delivery vehicle for subunit vaccine and its adjuvant.

Abstract:A polymersome system for delivering protein antigen ovalbumin (OVA) based on amphiphilic polyphosphazene grafting with N,N-diisopropylethylenediamine (DPA) and poly(ethylene glycol) (PEG) groups (poly[(DPA)m (PEG)n phosphazene], PEDP) was designed and constructed. The 200-240 nm-size OVA-loaded polymersomes displayed high stability at physiological pH, slow internalization through clathrin-mediated endocytosis pathway, and then a pH-triggered sustained OVA release in acidic environment, leading to extensive antigen access to cytosol. Prime-boost vaccine kept high antibody titers for 8 weeks and the subcutaneous vaccine of OVA polymersomes biased the immune response towards a type 1 T helper (Th1) response. Animal experiment results showed that the antigen-specific prophylactic vaccination by PEDP polymersomes delivery was much more rapid and efficient in depressing tumor growth and progress when compared with the therapeutic vaccination. These results suggested that PEDP-based polymersomes are very promising in controlled cytosolic delivery of protein antigens, and enhanced Th1 specific immune response.

Abstract:Abstract Background Poly(D, L-lactic-co-glycolic acid) (PLGA) nanoparticles have potential applications as a vaccine adjuvant and delivery system due to its unique advantages as biodegradability and biocompatibility. Experimental We fabricated cationic solid lipid nanoparticles using PLGA and dimethyl-dioctadecyl-ammonium bromide (DDAB), followed by loading of model antigen OVA (antigen ovalbumin, OVA 257-264 ) to form an OVA@DDAB/PLGA nano-vaccine. And we investigated the intracellular signaling pathway in dendritic cells in vitro and antigen transport pathway and immune response in vivo mediated by an OVA@DDAB/PLGA nano-vaccine. Results In vitro experiments revealed that the antigen uptake of BMDCs after nanovaccine incubation was two times higher than pure OVA or OVA@Al at 12 h. The BMDCs were well activated by p38 MAPK signaling pathway. Furthermore, the nano-vaccine induced antigen escape from lysosome into cytoplasm with 10 times increased cross-presentation activity than those of OVA or OVA@Al. Regarding the transport of antigen into draining lymph nodes (LNs), the nano-vaccine could rapidly transfer antigen to LNs by passive lymphatic drainage and active DC transport. The antigen + cells in inguinal/popliteal LNs for the nano-vaccine were increased over two folds comparing to OVA@Al and OVA at 12 h. Moreover, the antigen of nano-vaccine stayed in LNs for over 7 days, germinal center formation over two folds higher than those of OVA@Al and OVA. After immunization, the nano-vaccine induced a much higher ratio of IgG2c/IgG1 than OVA@Al. It also effectively activated CD4 + T, CD8 + T and B cells for immune memory with a strong cellular response. Conclusion These results indicated that DDAB/PLGA NP was a potent platform to improve vaccine immunogenicity by p38 signaling pathway in BMDCs, enhancing transport of antigens to LNs, and higher immunity response. Abstract:Schematic representation of the process of preparing γ‐PGA/CS/OVA NPs. To improve the stability and immunization of proteins or peptides after vaccination, choosing biodegradable and biocompatible materials to encapsulate protein ovalbumin (OVA) is necessary. In this study, we aim to construct OVA‐loaded γ‐polyglutamic acid (γ‐PGA)/chitosan (CS) nanoparticles (NPs) using ionic cross‐linking and poly‐electrolyte composite methods and to explore its immune effect in mice. The as‐constructed γ‐PGA/CS/OVA NPs, with a positive charge on the surface, were uniformly round. Additionally, the particle size range of the as‐constructed γ‐PGA/CS/OVA NPs was 200–400 nm. Furthermore, the sodium dodecyl sulfate (SDS)‐PAGE analysis showed that OVA in γ‐PGA/CS/OVA NPs had high structural integrity. The conditions of preparing γ‐PGA/CS/OVA NPs were optimized in detail, and it was found that the resultant loading capacity of NPs achieved 207.7 ± 11.1 μg/mg. MTT assay showed that γ‐PGA/CS NPs have good biocompatibility. The results of immune experiments in mice showed that compared with OVA not wrapped with γ‐PGA/CS composite carrier, γ‐PGA/CS/OVA NPs can induce a stronger immune response through humoral immunity in vivo. Overall, these findings demonstrated that γ‐PGA/CS NPs can be regarded as a good protein delivery system to enhance the immune response in vivo.

Preparation of Modified Konjac Glucomannan Nanoparticles and their Application as Vaccine Adjuvants to Promote Ovalbumin-Induced Immune Response in Mice

Abstract:PurposeHerein, we reported a facile strategy for synthesis of two types of modified konjac glucomannan nanoparticles (NPs). The goal of this project was to explore the potential of the NPs as vaccine adjuvants.MethodsFirstly, anionic carboxymethylated konjac glucomannan (CKGM) and cationic quaternized konjac glucomannan (QKGM) were synthesized by chemical modification of konjac glucomannan (KGM). Subsequently, two types of NPs, CKGM/QKGM and sodium tripolyphosphate (TPP)/QKGM, were prepared through polyelectrolyte complex method and ionic cross-linking method, respectively. The thus-synthesized NPs were then loaded with ovalbumin (OVA) to further evaluate the effect of NPs on immune response in mice.ResultsThe encapsulation efficiency of OVA for CKGM/QKGM/OVA and TPP/QKGM/OVA NPs could be 49.2% and 67.7%, respectively, while the drug loading capacity could reach 10.9% and 60%. The NPs showed irregular spherical shape and exhibited good sustained-release properties. In vitro cytotoxicity assay revealed that both the blank and OVA-loaded NPs were not toxic to cells. The OVA-specific IgG, splenocytes proliferation and cytokine levels indicated that the OVA-induced humoral and cellular immune responses were up-regulated by OVA-loaded NPs. What’s more, CKGM/QKGM/OVA NPs elicited both higher IL-2 and IFN-γ production, while TPP/QKGM/OVA NPs elicited both higher IL-4 and IL-10 production.ConclusionsThese results suggest that TPP/QKGM and CKGM/QKGM NPs are promising to be used as vaccine adjuvants. The TPP/QKGM/OVA NPs could induce stronger humoral immune response, while CKGM/QKGM/OVA NPs could enhance the cellular immune response more effectively.

Abstract:Oil-in-water emulsion-based adjuvants have demonstrated acceptable safety in many disease indications, while their adjuvant activities for vaccines still need to be improved. Recently, the strategy of combining adjuvants with multiple types of immunostimulants has been shown to enhance immune responses. In this study, astragalus polysaccharides were combined with simvastatin as an immunostimulant to construct a compound O/W emulsion adjuvant. The formulations were optimized according to the OVA-specific antibody responses induced in mice. For this reason, high (5 mg/mL), medium (2.5 mg/mL), and low (1.25 mg/mL) concentrations of astragalus polysaccharides and high (10 mg/mL), medium (1 mg/mL), and low (0.1 mg/mL) concentrations of simvastatin were selected. The final optimal formulation of the immunostimulant was a high concentration of astragalus polysaccharides combined with a medium concentration of simvastatin. The optimal compound O/W emulsion adjuvant could induce effective humoral and cellular immune responses that were stronger and more stable than those induced by aluminum adjuvant and Freund's adjuvant. The OVA/HAPS-MSim-OE induced dramatically strong and persistent IgG expressions and Th1-polarized immune responses. What's more, the highest CD4 + /CD8 + lymphocyte ratios were observed in OVA/HAPS-MSim-OE group. In addition, compound O/W emulsion adjuvant groups significantly promoted the secretion of IFN-γ and IL-6, which also indicated that the compound O/W emulsion adjuvants could induce both enhanced Th1 and Th2-mediated immune responses but prefer the Th1-mediated ones. This study would contribute to an interesting and promising direction in the development of emulsion-based adjuvants.

Abstract:Cancer vaccines have recently garnered tremendous interest. However, the targeted delivery of antigens and adjuvants to dendritic cells (DCs) still remains challenging. In this study, we developed glycosylated poly(lactic-co-glycolic acid) nanoparticles (NPs) loaded with the SIINFEKL peptide (OVA) as a tumor-specific antigen and CpG oligodeoxynucleotide (CpG) as an adjuvant for an effective DC-targeted cancer vaccine. Surface modification of NPs with galactose (Gal) or mannose (Man) was carried out by a double-emulsion solvent evaporation method, and the products were respectively named OVA-CpG Gal-NPs and OVA-CpG Man-NPs. They exhibited a uniform particle size, high loading capacity, robust stability, and extended release. The OVA-CpG Gal-NPs were found to rapidly enhance antigen uptake and DC maturation. In the in vivo study, OVA-CpG Gal-NPs via intravenous (i.v.), intranasal (i.n.) and subcutaneous (s.c.) routes had rapidly accumulated in the spleen. Moreover, the non-glycosylated OVA-CpG NPs after s.c. immunization could rapidly be trafficked to distal lymph nodes and sustained higher levels. All of these formulations increased the level of cluster of differentiation 4-positive (CD4+) T cells and interferon (IFN)-γ in the spleen, then promoted the cytotoxic CD8+ tumor-infiltrating lymphocytes against E.G7-OVA lymphomas. In conclusion, galactosylated NPs provided an effective platform to enhance the DC targeting to induce cellular immunity and T-cell recruitment into tumor sites in vivo, thus showing great potential to be developed as a prophylactic vaccine for cancer immunotherapy.
