Abstract:Holographic techniques enable precise laser manipulation, but suffer from two considerable limitations: speckle and deterioration of axial distribution. Here, we propose a cylindrical quadratic phase (CQP) method with temporal focusing (TF) to generate speckle-free holographic illumination with high axial resolution. TF-CQP utilizes a superposed cylindrical phase as the initial guess to iteratively optimize phase hologram, realizing speckle-free holographic reconstruction on the target focal plane and eliminating secondary focus on the defocused planes. TF-CQP further disperses defocused beams symmetrically by a blazed grating, placed conjugate to the focal plane, which enhances axial confinement. Simulation and experimental results show that TF-CQP reconstructs speckle-free illumination with arbitrary shapes and <10 µm axial resolution. Compared to TF-GS (Gerchberg-Saxton algorithm), widely used in holographic optogenetics, TF-CQP shows increased uniformity of 200% and improved modulation efficiency of 32.33% for parallel holographic illumination, as well as a 10% increment in axial resolution.

Abstract:A fast and high-resolution reconstruction algorithm with variable resolution increasing ratio and down-sampling ratio is proposed. Since the digital holography requires that the space-bandwidth product of the recording system should be greater than that of the object so as to ensure no loss of information, usually much redundant information and pixels exist in the recorded digital hologram. According to that, we propose utilizing a flexible down-sampling process with variable ratio to eliminate the redundancy. The technical characteristics are analyzed. The validity of this method is shown by both calculations based on theory analysis and experimental results.
Single-exposure cepstrum phase-shifting in-line digital holography based on dual-CCD

Abstract:Limited by the property of charge-coupled device (CCD), the recording optical path of in-line digital holography is commonly used to take full advantage of the CCD sampling space, which is in order to improve the performance of digital holographic measurement. But the reconstructed image of in-line digital holography is overlapped by the direct wave and the conjugate image, which is hard to be separated. To achieve the real-time high-resolution measurement of in-line digital holography, the dual-CCD single-exposure cepstrum phase-shifting in-line digital holography is put forward, which is based on the single phase-shifting cepstrum technology. Then two key technical schemes of this technology, including the method of adjusting dual-CCD's mirroring overlapping position and the optical structure of dual-CCD single-exposure π/2 phase-shifting, are given. Finally, the experimental results show that, the dual-CCD single-exposure cepstrum phase-shifting in-line digital holography can solve the overlapping problem well. Moreover, the real-time recording and reconstruction of in-line digital holography are initially realized.
Research on Improving Reconstruction Quality from Holograms of Marine Algae in Digital Holographic Microscopy

Abstract:In this paper, we improve reconstruction quality from in-line holograms of marine algae by two phase-retrieval-based reconstruction methods. Digital holographic microscopy, with its high-resolution and volume recording capability, has been applied in the area of monitoring marine algae. We designed a digital in-line holographic microscopy laboratory system to observe and analyze marine algae. Due to the "twin-image" problem caused by the loss of phase information in hologram recording, the reconstructed quality from holograms of marine algae reduced significantly. To enhance reconstruction quality, the alternating projections methods and inverse methods are introduced and implemented on the marine algae holograms. Compared with the traditional backpropagation method, both of the above methods have good performance in improving reconstruction quality from in-line marine algae holograms.

Abstract:The research proposes a new method to solve the spectrum aliasing and improve the resolution of off-axis digital holograms based on Kronecker interpolation and backpropagation algorithms. The method suppresses the direct current (DC) image and increase the low-frequency information with Kronecker interpolation of the hologram. The size of the Fresnel reconstructed image can be controlled with the reconstruction distance, and this feature is used to select the appropriate reconstruction distance for the Fresnel amplitude reconstruction of interpolated holograms to separate the real and imaginary images. To obtain the complete first-order spectral information for the object, the amplitude of the desired image is obtained with spatial domain filtering and then inverted. Finally, the amplitude and phase of the object are reconstructed according to the angular spectrum reconstruction algorithm. The results show that the method can solve the spectrum aliasing problem of the holograms and thus improve the resolution of off-axis holograms. In addition, the background noise of the reconstructed phase is reduced because the influence of the zero-order spectrum can be completely avoided.

Abstract:The measurement of microstructured components is a challenging task in optical engineering. Digital holographic microscopy has attracted intensive attention due to its remarkable capability of measuring complex surfaces. However, speckles arise in the recorded interferometric holograms, and they will degrade the reconstructed wavefronts. Existing speckle removal methods suffer from the problems of frequency aliasing and phase distortions. A reconstruction method based on the antialiasing shift-invariant contourlet transform (ASCT) is developed. Salient edges and corners have sparse representations in the transform domain of ASCT, and speckles can be recognized and removed effectively. As subsampling in the scale and directional filtering schemes is avoided, the problems of frequency aliasing and phase distortions occurring in the conventional multiscale transforms can be effectively overcome, thereby improving the accuracy of wavefront reconstruction. As a result, the proposed method is promising for the digital holographic measurement of complex structures. (C) 2018 Society of Photo- Optical Instrumentation Engineers (SPIE).

Abstract:The imaging quality of in-line digital holography is challenged by the twin-image and aliasing effects because sensors only respond to intensity and pixels are of finite size. As a result, phase retrieval and pixel superresolution techniques serve as the two essential ingredients for high-fidelity holographic imaging. In this work, we combine the two within a unified algorithmic framework. Pixel super-resolution phase retrieval is recast as an optimization problem and is solved via gradient descent-based algorithms. Regularization techniques and Nesterov’s momentum are introduced to further speed up data acquisition and iterative reconstruction. The proposed algorithms are verified through a proof-of-concept lensless on-chip microscope. We demonstrate experimentally the capability of pixel super-resolution phase retrieval techniques in revealing the subpixel and quantitative phase information of complex biological samples.

Abstract:A method for improving the resolution of single-frame off-axis holograms based on extrapolation iteration and Kronecker interpolation is proposed to address the problem of low resolution of image sensors in digital holographic systems. Low-frequency information in the image is increased and the zero-order image is suppressed by the numerical computation in the frequency domain using the Kronecker interpolation method. High-frequency information outside the received plane is calculated using an extrapolation iterative algorithm. The obtained low-frequency and high-frequency information are then combined to improve the resolution of off-axis holography. Compared to conventional reconstruction methods, the proposed method eliminates the need to add complex optical elements to the optical path, greatly simplifying the optical system when increasing resolution. Simulation and optical experimental results demonstrate the feasibility of the proposed method.
Iterative Reconstruction of Diffraction Limited Digital Holography

Abstract:Digital holographic imaging has the characteristics of fast and flexible with simple set up, and it can quantitatively obtain the intensity and phase of objects. The resolution of the reconstructed image is greatly limited by the pixelated imaging detectors during the digitization processing of holograms. Due to the limited diffraction bandwidth, the diffracted beam of each object point can be viewed as a diffractive cone. The lack of diffraction components causes the reconstructed quality to decrease when the detector cannot collect a complete diffracted wave field in the inline holography. In this work, we studied the reconstructed quality of holograms under different sampling forms. The losing diffraction components can be reconstructed by using an iterative algorithm based on energy attenuation and support constraints. We propose three iterative methods. The first is the bicubic interpolation and extrapolation iterative method based on a single small detector. The second is multi-plane extrapolation iterative method. The third is a faithful iterative reconstruction based on a sparse sensor array. When the diffraction component recorded by the hologram is missing, the losing diffraction components can be reconstructed by using the principle of the holographic redundancy. The reduction of reconstruction quality caused by the loss of sampling in the small-sized detector and sparse sensor array in the digital holography can be compensated.
Holographic Imaging Using an Imperfect Plane Wave Illumination With a Background Phase

Abstract:Aberrations in the optical components and misalignments in the optical system cause a background phase in the coherent illumination. To reconstruct the object phase, the background phase illuminating the object must be measured and subtracted. For diffraction imaging and in-line holography, the traditional phase retrieval method reconstructs the phase diffracting from clear edges. However, it falls into stagnation when solving a background phase slowly varying in the spatial domain. In this study, we propose to solve the background phase using a modulation-based phase retrieval method. Alternative structured phase modulation (ASPM) can be the phase constraint to avoid stagnation when solving the background phase without clear edges. With ASPM, the background phase in the experiment can be efficiently retrieved when 16 phase patterns are employed. The ASPM acts as a phase grating to concentrate the intensities and provides robustness to noise. Compared to the conventional random phase modulations, the ASPM method had a smaller error value in the reconstruction iterations, which leads to a better reconstruction quality. After measuring and subtracting the background phase, the object phase was retrieved using a coherent diffraction imaging system. A phase plate can be accurately reconstructed under three different background phases.

Abstract:We investigate the influence of phase-shifting error on the quality of the reconstructed image in digital holography and propose a method of error elimination for a perfect image. In this method the summation of the intensity bit errors of the reconstructed image is taken as an evaluation function for an iterative algorithm to find the exact phase-shifting value. The feasibility of this method is demonstrated by computer simulation.

Abstract:Lensless holographic imaging is challenged by the twin-image artifact due to the missing phase and the aliasing effect due to the undersampled measurement. Therefore, phase retrieval and pixel super-resolution (PSR) techniques serve as the essential ingredients for high-fidelity holographic imaging. In this work, we combine the two in a unified framework by formulating the PSR phase retrieval as a non-convex feasibility problem. An adaptive smoothing strategy for escaping local minima is introduced. Numerical and experimental results are presented and discussed. The proposed framework can be generalized to various physical settings, and is compatible with the state-of-the-art iterative projection algorithms.

Abstract:Digital holographic microscopy is a promising method for measuring complex micro-structures with high slopes. A quasi-common path interferometric apparatus is adopted to overcome environmental disturbances, and an acousto-optic tunable filter is used to obtain multi-wavelength holograms. However, the phase shifting error caused by the acousto-optic tunable filter reduces the measurement accuracy and, in turn, the reconstructed topographies are erroneous. In this paper, an accurate reconstruction approach is proposed. It corrects the phase-shifting errors by minimizing the difference between the ideal interferograms and the recorded ones. The restriction on the step number and uniformity of the phase shifting is relaxed in the interferometry, and the measurement accuracy for complex surfaces can also be improved. The universality and superiority of the proposed method are demonstrated by practical experiments and comparison to other measurement methods.

Abstract:This study proposes a gradient descent method for enhancing the quality of reconstructed images encoded by double-phase holograms (DPHs). Holographic displays are often noisy because of optical hardware problems. However, owing to the conversion from complex-amplitude holograms to phase-only holograms, the reconstructed images still have noise and cannot be ignored. In this paper, the proposed method uses a spectrum loss function instead of image loss function in the gradient descent optimization process. The spectrum loss function ensures that DPHs can be optimized by gradient descent method, and this method requires only several iterations. The simulations and experimental results have validated the effectiveness of the proposed method.

Abstract:The morphology and dynamics of label-free tissues can be exploited by sample-induced changes in the optical field from quantitative phase imaging. Its sensitivity to subtle changes in the optical field makes the reconstructed phase susceptible to phase aberrations. We import variable sparse splitting framework on quantitative phase aberration extraction based on alternating direction aberration free method. The optimization and regularization in the reconstructed phase are decomposed into object terms and aberration terms. By formulating the aberration extraction as a convex quadratic problem, the background phase aberration can be fast and directly decomposed with the specific complete basis functions such as Zernike or standard polynomials. Faithful phase reconstruction can be obtained by eliminating global background phase aberration. The aberration-free two-dimensional and three-dimensional imaging experiments are demonstrated, showing the relaxation of the strict alignment requirements for the holographic microscopes.

Abstract:In the digital holographic microscopy, the phase aberration is a key problem limiting the quantitative phase measurement, especially for the microstructure whose phase distribution is dense with few and discontinuous background region. A numerical phase aberration compensation method with global curve fitting preprocessing and automatic extraction of the background region for microstructure testing is proposed. The global curve fitting is implemented to compensate most of the tilt, quadratic and partial high order aberrations and make the profile of the measured object significantly distinguishable. Then the background region is automatically extracted by directly segmenting the converted grayscale phase image. At last, the Zernike polynomial fitting is performed on the extracted background region, and the obtained coefficients are used to calculate the phase aberration of the whole phase image. Simulation and experimental results demonstrated that the proposed method is able to accurately extract background region and compensate the phase aberration for the complicated dense microstructure.
Comparison and Improvement of Image Quality Evaluation of Zero-order Elimination Methods in Holographic Reproduction

Abstract:During the reconstruction of holograms,the presence of the zero-order diffraction image which occupies the majority of energy reduces the contrast of the reproduced image,and affects the observation and recording of the original object.So to make the useful reconstructed image clear,it needs to be eliminated.In this paper,it is given an introduction and theoretical analysis of five measures commonly used for eliminating the zero-order diffraction image.We applied them in computer-generated hologram(CGH),and composed numerical reproduction simulations respectively.Then two common objective evaluation criteria were used to compare and judge the effectiveness of those measures.The results show that all the measures can effectively eliminate the bright spots caused by zero-order image;meanwhile the original images or conjugate images are preserved completely.The contrast of the reproduced images is improved.After general comparison,it is found that the measure of frequency filtering can get the best effect,but it does not jibe with human's intuitive perception.Based on the analysis of the advantages and disadvantages of these two methods and the frequency spectrum of zero-order image,it is proposed a new local method grounded on the attenuation rate of the central bright spot to evaluate the quality of reproduced image,and experiments were conducted to testify the method.The experimental results show that,according to this new method,a result is obtained to be consistent with the intuitive feel through human eyes looking at images directly,and thus it can be considered to be effective.

Abstract:An iterative method with bandwidth constraint strategy is proposed to design phase holograms for high-quality speckle-free optical reconstruction. The bandwidth properties of the reconstructed field are analyzed theoretically based on the sampling theory, which helps in properly allocating the sampling resources for efficiently describing the speckles and artifacts in the reconstructed field. Iterative calculation with bandwidth constraint strategy of the reconstructed field and quadratic initial phase can optimize the phase hologram without stagnation problem, which provides effective controls of the reconstructed intensity fluctuations and helps to suppress the speckles and artifacts. Numerical and optical experiments have been performed to validate the proposed method can achieve excellent image fidelity.

Abstract:The influence caused by different phase-shifting a ngle of reference wave in recording and the phase-shifting error on the quality of the reconstructed image is investigated in two-step phase-shifting in-line digital holography, and an effective method of eliminating the phase-shifting error is presented, in which the summation of the intensity bit errors of the reconstructed image is taken as an evaluation function for an iterative algorithm to find the exact phase-shifting value. Then the optimal choice of the phase-shifting value of reference wave is simulated by computers. The simulation results show that the reconstructed image noise is weak when the phase-shifting value is in a certain range. At last, the feasibility of the method of eliminating the phase-shifting error is demonstrated by computer simulation also.

As a result, phase retrieval and pixel super-resolution techniques serve as the two essential ingredients for high-fidelity holographic imaging. In this work, we combine the two as a unified optimization problem, and propose a generalized algorithmic framework for pixel-super-resolved phase retrieval. In particular, we introduce the iterative projection algorithms and gradient descent algorithms for solving this problem. The basic building blocks, namely the projection operator and the Wirtinger gradient, are derived and analyzed. The algorithms are verified with both simulated and experimental data. The proposed framework generalizes well to various physical settings, and is compatible with many state-of-the-art optimization algorithms.
