Abstract:Introduction: Given the limited success to date in using non-specific membrane makers, in identifying cardiac stem cells, the significance of finding a much more potential cardiac mesenchymal stem cell (C-MSC) maker from transcription factors in nuclear is very important. Hypothesis: We want to test the feasibility of using GATA4, an early cardiac-specific transcription factor, as biomarker to identify a subpopulation of C-MSC that could be highly efficacious for cardiovascular repair. Methods: A lentiviral vector in which the expression of eGFP reporter is driven by a 1kb fragment of the mouse GATA4 promoter was constructed. Murine C-MSC with high GFP intensity were sorted by FACS. Results: Western blot shows that the C-MSC population with high GFP intensity exhibits higher GATA4 protein expression as compared with unsorted C-MSC. Interestingly, we observed that sorted GATA4high C-MSC also exhibit higher HIF-1α protein expression as compared with unsorted C-MSC. We compared hypoxia- and serum deprivation induced-apoptosis between sorted GATA4high and unsorted C-MSC by TUNEL staining. We found that the GATA4high exhibited lower rate of TUNEL positivity as compared with unsorted C-MSC, suggesting GATA4high C-MSC are more resistant to hypoxia- and serum deprivation-induced apoptosis in comparison with unsorted C-MSC. In mouse model of myocardial infarction by permanent left coronary ligation, GATA4high C-MSC or unsorted C-MSC was injected into the border zone area. Left ventricular ejection fraction (EF) were serially (days 1 and 14) assessed using echocardiography. At day 1, no differences in EF were observed between GATA4high C-MSC and unsorted C-MSC group (n=9, ns). At day 14, left ventricular ejection fraction in GATA4high C-MSC cell-treated group was improved about 11.2% compared with the unsorted C-MSC group (n=9, P Conclusions: Our results suggest that GATA4high, as compared with unsorted C-MSC, exhibit enhanced survival capacity which will consequently lead to improvement of heart contractile function in the infarcted heart, therefore, GATA4high C-MSC can be considered as a much more potential progenitor for heart repair.
SDF-1 expression by mesenchymal stem cells results in trophic support of cardiac myocytes after myocardial infarction.

Abstract:Stem cell transplantation at the time of acute myocardial infarction (AMI) improves cardiac function. Whether the improved cardiac function results from regeneration of cardiac myocytes, modulation of remodeling, or preservation of injured tissue through paracrine mechanisms is actively debated. Because no specific stem cell population has been shown to be optimal, we investigated whether the benefit of stem cell transplantation could be attributed to a trophic effect on injured myocardium. Mesenchymal stem cells secrete SDF-1 and the interaction of SDF-1 with its receptor, CXCR4, increases survival of progenitor cells. Therefore, we compared the effects of MSC and MSC engineered to overexpress SDF-1 on cardiac function after AMI. Tail vein infusion of syngeneic MSC and MSC: SDF-1 1 day after AMI in the Lewis rat led to improved cardiac function by echocardiography by 70.7% and 238.8%, respectively, compared with saline controls 5 wk later. The beneficial effects of MSC and MSC: SDF-1 transplantation were mediated primarily through preservation, not regeneration of cardiac myocytes within the infarct zone. The direct effect of SDF-1 on cardiac myocytes was due to the observation that, between 24 and 48 h after AMI, SDF-1-expressing MSC increased cardiac myocyte survival, vascular density (18.2 +/- 4.0 vs. 7.6 +/- 2.3 vessels/mm(2), P<0.01; SDF-1: MSC vs. MSC), and cardiac myosin-positive area (MSC: 49.5%; mSC: SDF-1: 162.1%) within the infarct zone. There was no evidence of cardiac regeneration by the infused MSC or endogenous cardiac stem cells based on lack of evidence for cardiac myocytes being derived from replicating cells. These results indicate that stem cell transplantation may have significant beneficial effects on injured organ function independent of tissue regeneration and identify SDF-1: CXCR4 binding as a novel target for myocardial preservation.

Abstract:Bone marrow mesenchymal stromal cells (BMSCs) have potential applications in cell and gene therapies for cardiac disease. The cardiac-specific transcription factors GATA-binding protein 4 (GATA4) and T-Box protein 5 (TBX5) are considered to be pivotal in cardiogenesis. The aim of this study was to investigate the effects of GATA4 and TBX5 on cardiomyogenic differentiation of BMSCs. The BMSCs were initially isolated and identified. Vectors harboring cardiac transcription factor genes GATA4 and TBX5 or empty vectors were transferred into BMSCs. Cardiomyogenic cells differentiated from BMSCs were identified by expression of cardiac-specific markers including cardiac troponin T, connexin 43, β-myosin heavy chain, and myosin light chain-2 using immunocytochemical staining, western blotting, and quantitative real-time PCR. The ultrastructures of the differentiated cells were examined by transmission electron microscopy, which were similar to those of fetal cardiomyocytes. The differentiated cells exhibited L-type calcium current activities reflective of the electrophysiological characteristics of cardiomyocytes. These findings indicate that exogenous expression of cardiac-specific transcription factors GATA4 and TBX5 enhance cardiomyogenic differentiation of BMSCs.
Overexpression of Csx/Nkx2.5 and GATA-4 Enhances the Efficacy of Mesenchymal Stem Cell Transplantation after Myocardial Infarction

Abstract:Background: The high death rate of the transplanted stem cells in the infarcted heart and low efficiency of differentiation toward cardiomyocytes show that mesenchymal stem cell (MSC) transplantation after myocardial infarction (MI) is not effective. Csx/Nkx2.5 and GATA-4 are considered to be key regulators of cardiogenesis. The aim of the present study was to investigate the effect of transplanting MSC overexpressing Csx/Nkx2.5 and GATA-4 (MSCs-CG) after MI.Methods and Results: According to acridine orange/ethidium bromide staining, MSCs-CG were more resistant to anoxia as compared with MSCs in vitro. In a mouse MI model, ejection fraction and fractional shortening were higher in the MSC-CG group than in the MSC or phosphate-buffered saline group. Wall thickness of the infarct area was increased and collagen deposition was clearly reduced in the MSC-CG group as compared with the other groups. There were more surviving MSCs in the MSC-CG group than in the MSC group. Most of the Y chromosome-positive cells expressed cardiac troponin T and connexin43 (Cx-43). Cx-43 was localized between Y chromosome-positive cells and recipient cardiomyocytes. Microvessel density in the pen-infarct regions and infarct regions increased significantly in the MSC-CG group.Conclusions: Transplantation of MSCs overexpressing Csx/Nkx2.5 and GATA-4 represents a new treatment strategy with the potential to improve cardiac function after MI. Optimizing Cardiomyocyte Differentiation: Comparative Analysis of Bone Marrow and Adipose-Derived Mesenchymal Stem Cells in Rats Using 5-Azacytidine and Low-Dose FGF and IGF Treatment

Abstract:Mesenchymal stem cells (MSCs) exhibit multipotency, self-renewal, and immune-modulatory properties, making them promising in regenerative medicine, particularly in cardiovascular treatments. However, optimizing the MSC source and induction method of cardiac differentiation is challenging. This study compares the cardiomyogenic potential of bone marrow (BM)-MSCs and adipose-derived (AD)-MSCs using 5-Azacytidine (5-Aza) alone or combined with low doses of Fibroblast Growth Factor (FGF) and Insulin-like Growth Factor (IGF). BM-MSCs and AD-MSCs were differentiated using two protocols: 10 μmol 5-Aza alone and 10 μmol 5-Aza with 1 ng/mL FGF and 10 ng/mL IGF. Morphological, transcriptional, and translational analyses, along with cell viability assessments, were performed. Both the MSC types exhibited similar morphological changes; however, AD-MSCs achieved 70–80% confluence faster than BM-MSCs. Surface marker profiling confirmed CD29 and CD90 positivity and CD45 negativity. The differentiation protocols led to cell flattening and myotube formation, with earlier differentiation in AD-MSCs. The combined protocol reduced cell mortality in BM-MSCs and enhanced the expression of cardiac markers (MEF2c, Troponin I, GSK-3β), particularly in BM-MSCs. Immunofluorescence confirmed cardiac-specific protein expression in all the treated groups. Both MSC types exhibited the expression of cardiac-specific markers indicative of cardiomyogenic differentiation, with the combined treatment showing superior efficiency for BM-MSCs.

Abstract:Objective ischaemic heart disease is the leading cause of morbidity and mortality all over the world. Cardiomyocytes from bone marrow-derived mesenchymal stem cells (MSCs) offer great potential for repairment of the infracted heart. However, this approach has been limited by inefficient differentiation of MSCs into cardiomyocytes. To overcome such a problem, the underlying regulation mechanisms for cardiac differentiation should be elucidated. MicroRNAs (miRNAs) are small noncoding RNAs of ∼23 nucleotides that control post-transcriptional gene expression. Recently, miRNA has been widely shown to regulate key cellular events such as cell proliferation, cell differentiation. The purpose of this study is to determine the role of miRNAs during cardiac differentiation from MSCs. Method Firstly, we established a model of cardiac differentiation from rat bone marrow-derived MSCs using 10 μM 5-Aza, and performed a global miRNA analysis using EXIQON miRNA array to identify characteristic miRNA at different stage of differentiation. After being validated by real-time qRT-PCR and target gene prediction, several miRNAs such as miRNA-145 were further chosen to reveal its function during cardiac differentiation. Results miRNA profiling revealed that miR-145 expression increased during cardiac differentiation, especially at 12 days of treatment (2.25-fold change vs untreated MSCs). Compared to other tissues such as liver, brain, kidney, miRNA-145 expression is highest in rat heart tissue. Gain-of-function methods using pre-miR-145 showed the enchancement of cardiac differentiation, confirmed by immunocytochemical staining with cardiac-specific myosin heavy chain antibody. Conclusion miR-145 may be a critical regulator for cardiomyocyte lineage in our 5-Aza-induced differentiation system.
Cardiac-derived stem cell-based therapy for heart failure: progress and clinical applications.

Abstract:Stem cell-based therapy is emerging as a promising strategy to treat end-stage heart failure, a leading cause of morbidity and mortality. Stem cells can be isolated from a variety of sources and exhibit unique characteristics that impact their potential therapeutic utility. The adult heart contains small populations of committed, multipotent cardiac stem cells (CSC), which are adapted to the cardiac microenvironment and participate in postnatal physiological and pathological cardiac renewal or repair. These cells can be isolated, expanded in culture, and administered therapeutically to improve cardiac function in the setting of heart failure. CSC can be differentiated into three distinct cardiovascular lineages and exhibit enhanced paracrine factor production and engraftment as compared with other types of mesenchymal stem cells, which in turn may translate into improved therapeutic efficacy. The cell surface marker expression and phenotype of these CSC, however, depends on the method of isolation, selection and propagation, which likely explains the variable experimental results obtained to date. Moreover, invasive procedures are required to obtain CSC from humans. Early trials using autologous CSC in patients with ischemic cardiomyopathy have demonstrated feasibility and safety, along with variable degrees of therapeutic efficacy in terms of enhancing myocardial viability and cardiac function. Further studies are needed to optimize methods of CSC isolation, manipulation and delivery. If fully realized, the potential of CSC therapy could fundamentally change the approach to the treatment of end-stage heart failure.
Transcription factor-induced activation of cardiac gene expression in human c-kit+ cardiac progenitor cells

Abstract:Although transplantation of c-kit+ cardiac progenitor cells (CPCs) significantly alleviates post-myocardial infarction left ventricular dysfunction, generation of cardiomyocytes by exogenous CPCs in the recipient heart has often been limited. Inducing robust differentiation would be necessary for improving the efficacy of the regenerative cardiac cell therapy. We assessed the hypothesis that differentiation of human c-kit+ CPCs can be enhanced by priming them with cardiac transcription factors (TFs). We introduced five different TFs (Gata4, MEF2C, NKX2.5, TBX5, and BAF60C) into CPCs, either alone or in combination, and then examined the expression of marker genes associated with the major cardiac cell types using quantitative RT-PCR. When introduced individually, Gata4 and TBX5 induced a subset of myocyte markers. Moreover, Gata4 alone significantly induced smooth muscle cell and fibroblast markers. Interestingly, these gene expression changes brought by Gata4 were also accompanied by morphological changes. In contrast, MEF2C and NKX2.5 were largely ineffective in initiating cardiac gene expression in CPCs. Surprisingly, introduction of multiple TFs in different combinations mostly failed to act synergistically. Likewise, addition of BAF60C to Gata4 and/or TBX5 did not further potentiate their effects on cardiac gene expression. Based on our results, it appears that GATA4 is able to potentiate gene expression programs associated with multiple cardiovascular lineages in CPCs, suggesting that GATA4 may be effective in priming CPCs for enhanced differentiation in the setting of stem cell therapy.

Human Bone Marrow-Derived Mesenchymal Stem Cells Transplanted into Damaged Rabbit Heart to Improve Heart Function.

Abstract:Objective: The present study was designed to test whether transplantation of human bone marrow-derived mesenchymal stem cells (hMSCs) in New Zealand rabbits with myocardial infarction can improve heart function; and whether engrafted donor cells can survive and transdifferentiated into cardiomyocytes. Methods: Twenty milliliters bone marrow was obtained from healthy men by bone biopsy. A gradient centrifugation method was used to separate bone marrow cells (BMCs) and red blood cells. BMCs were incubated for 48 h and then washed with phosphate-buffered saline (PBS). The culture medium was changed twice a week for 28 d. Finally, hematopoietic cells were washed away to leave only MSCs. Human MSCs (hMSCs) were premarked by BrdU 72 h before the transplantation. Thirty-four New Zealand rabbits were randomly divided into myocardial infarction (MI) control group and cell treated group, which received hMSCs (MI+MSCs) through intramyocardial injection, while the control group received the same volume of PBS. Myocardial infarction was induced by ligation of the left coronary artery. Cell treated rabbits were treated with 5×10 6 MSCs transplanted into the infarcted region after ligation of the coronary artery for 1 h, and the control group received the same volume of PBS. Cyclosporin A (oral solution; 10 mg/kg) was provided alone, 24 h before surgery and once a day after MI for 4 weeks. Echocardiography was measured in each group before the surgery and 4 weeks after the surgery to test heart function change. The hearts were harvested for HE staining and immunohistochemical studies after MI and cell transplantation for 4 weeks. Results: Our data showed that cardiac function was significantly improved by hMSC transplantation in rabbit infarcted hearts 4 weeks after MI (ejection fraction: 0.695±0.038 in the cell treated group ( n =12) versus 0.554±0.065 in the control group ( n =13) ( P <0.05). Surviving hMSCs were identified by BrdU positive spots in infarcted region and transdifferentiated into cardiomyocytes characterized with a positive cardiac phenotype: troponin. I. Conclusion: Transplantation of hMSCs could transdifferentiate into cardiomyocytes and regenerate vascular structures, contributing to functional improvement.
The Effect of Angiotensin II, Retinoic Acid, EGCG, and Vitamin C on the Cardiomyogenic Differentiation Induction of Human Amniotic Fluid-Derived Mesenchymal Stem Cells

Abstract:Human amniotic fluid-derived mesenchymal stem cells (AF-MSCs) may be potentially applied in cell therapy or regenerative medicine as a new alternative source of stem cells. They could be particularly valuable in restoring cardiac tissue after myocardial infarction or other cardiovascular diseases. We investigated the potential of biologically active compounds, namely, angiotensin II, retinoic acid (RA), epigallocatechin-3-gallate (EGCG), vitamin C alone, and the combinations of RA, EGCG, and vitamin C with angiotensin II to induce cardiomyogenic differentiation of AF-MSCs. We observed that the upregulated expression of cardiac gene markers (NKX2-5, MYH6, TNNT2, and DES) and cardiac ion channel genes (sodium, calcium, the potassium) as well as the increased levels of Connexin 43 and Nkx2.5 proteins. Extracellular flux analysis, applied for the first time on AF-MSCs induced with biologically active compounds, revealed the switch in AF-MSCs energetic phenotype and enhanced utilization of oxidative phosphorylation for energy production. Moreover, we demonstrated changes in epigenetic marks associated with transcriptionally active (H3K4me3, H3K9ac, and H4hyperAc) or repressed (H3K27me3) chromatin. All in all, we demonstrated that explored biomolecules were able to induce alterations in AF-MSCs at the phenotypic, genetic, protein, metabolic, and epigenetic levels, leading to the formation of cardiomyocyte progenitors that may become functional heart cells in vitro or in vivo.

Abstract:Myocardial infarction (MI) is a leading cause of death worldwide, resulting in extensive loss of cardiomyocytes and subsequent heart failure. Inducing cardiac differentiation of stem cells is a potential approach for myocardial regeneration therapy to improve post-MI prognosis. Mesenchymal stem cells (MSCs) have several advantages, including immune privilege and multipotent differentiation potential. This study aimed to explore the feasibility of chemically inducing human amniotic membrane MSCs (hAMSCs) to differentiate into cardiomyocytes in vitro. Human amniotic membrane (AM) samples were obtained from routine cesarean sections at Far Eastern Memorial Hospital. The isolated cells exhibited spindle-shaped morphology and expressed surface antigens CD73, CD90, CD105, and CD44, while lacking expression of CD19, CD11b, CD19, CD45, and HLA-DR. The SSEA-1, SSEA-3, and SSEA-4 markers were also positive, and the cells displayed the ability for tri-lineage differentiation into adipocytes, chondrocytes, and osteoblasts. The expression levels of MLC2v, Nkx2.5, and MyoD were analyzed using qPCR after applying various protocols for chemical induction, including BMP4, ActivinA, 5-azacytidine, CHIR99021, and IWP2 on hAMSCs. The group treated with 5 ng/ml BMP4, 10 ng/ml Activin A, 10 μM 5-azacytidine, 7.5 μM CHIR99021, and 5 μM IWP 2 expressed the highest levels of these genes. Furthermore, immunofluorescence staining demonstrated the expression of α-actinin and Troponin T in this group. In conclusion, this study demonstrated that hAMSCs can be chemically induced to differentiate into cardiomyocyte-like cells in vitro. However, to improve the functionality of the differentiated cells, further investigation of inductive protocols and regimens is needed.
Colorectal carcinoma: laparoscopic versus traditional open surgery. A clinical trial.

Abstract:BACKGROUND/AIMS The purpose of this perspective study was to define the role of laparoscopic surgery in the treatment of colorectal carcinoma. METHODOLOGY One hundred colorectal cancer patients were submitted to surgical treatment between 1993 and 1996. Fifty patients were operated on by videolaparoscopy, the other 50 were operated on according to the standard "open" technique. The two groups had similar demographic (age, gender), pathological (site, stage), and surgical (type and extent of resection) data. Early and late results, benefits and drawbacks of the minimally invasive technique are compared to those of standard open surgery. RESULTS No intra-operative complications and no operative mortality occurred in the two groups. Early results (complications within 30 days from surgery) were: 1 pneumonia, 3 wound sepsis, and 3 fistulas (one required a reoperation) in the laparoscopic group; 2 wound sepsis and 5 fistulas (spontaneously recovered) in the open group. Late complications occurred in the laparoscopic group only: 1 bowel bridle occlusion 2 months after surgery (that required a reoperation), and 2 stenoses of the colorectal Knight-Griffen anastomosis, successfully treated by dilatation. Concerning the oncologic results, data were calculated on 40 laparoscopic and 43 open curative resections (stage I, II and III): 20% (8/40) of the laparoscopic and 23% (10/43) of the open group patients resulted in neoplastic progression. The neoplastic recurrences were single site (liver or regional) in 3 laparoscopic and in 5 open patients; multiple sites of relapse were observed in 5 laparoscopic (liver, peritoneum and 1 trocar site) and in 5 open (liver, peritoneum and 1 scar) cases. Five-year disease-free survival rates (Kaplan-Meier method) were similar in the two groups: 73.2% in the laparoscopic and 70.1% in the open. CONCLUSIONS Laparoscopic surgery seems to be a feasible and effective treatment of colorectal cancer and, with the improvement of technology and surgeon skill, it will represent an excellent alternative to the more diffuse and consolidated open surgery technique.
Abstract P1103: A Combinatorial Stem Cell Therapy Using HiPSC-derived Cardiomyocytes And HiPSC-derived Mesenchymal Stem Cells For Cardiac Regeneration

Abstract:Introduction: Myocardial infarction (MI) results in the death of nearly a billion cardiomyocytes in the heart. While conventional therapies alleviate the injury to some extent, they do not induce myocardial regeneration. On the other hand, transplantation of stem cells and their derived differentiated cells like hiPSC-derived cardiomyocytes (hiPSC-CMs) and hiPSC-derived mesenchymal stem cells (hiPSC-MSCs) have the potential to promote myocardial regeneration by direct (replacing dead cardiomyocytes) or indirect (paracrine) mechanisms, respectively. Hypothesis: We hypothesized that following co-transplantation of hiPSC-CMs and hiPSC-MSCs in ischemic hearts, hiPSC-MSCs will release paracrine factors to promote angiogenesis, reduce fibrosis, and improve cardioprotection and engraftment of transplanted hiPSC-CMs. Results: We cultured hiPSC-CMs and hiPSC-MSCs at different ratios, 1:1, 1:2 and 1:3, respectively. We observed improved contractility (MEA, and calcium transients) in the hiPSC-CM + hiPSC-MSC co-cultures, as compared to single cell-type cultures. Additionally, following exposure to hypoxia (1%O 2 ), we observed increased survival of hiPSC-CMs when co-cultured with hiPSC-MSCs (TUNEL assay). Furthermore, intramyocardial transplantation of (a) 1.5 х 10 6 hiPSC-CMs, (b) 1.5 х 10 6 hiPSC-MSCs or (c) 1.5 х 10 6 hiPSC-CMs + hiPSC-MSCs (3:1) in a mouse MI model, resulted in an improved cardiac function, LVEF (p < 0.01) and FS (p<0.01) in all animals receiving cell transplantation as compared to MI only group. Additionally, we observed efficient engraftment (immunostaining of human-specific markers, cTnT and HNA) of transplanted hiPSC-CMs in both hiPSC-CM-only and hiPSC-CM + hiPSC-MSC groups up to 4 weeks post-MI. We also observed increased neoangiogenesis (α-SMA, vWF) near the graft in the hiPSC-MSC-only and hiPSC-CM + hiPSC-MSC groups, as compared to control and hiPSC-CM-only groups. Lastly, a significant reduction in fibrosis (Masson-trichrome staining) was seen following transplantation of hiPSC-CMs + hiPSC-MSCs, compared to the control animals. Conclusion: Overall, our results established that a combinatorial stem therapy using hiPSC-CMs and hiPSC-MSCs for cardiac regeneration post-MI.

Abstract:BACKGROUND Embryonic stem cells (ESCs) serve as a crucial ex vivo model, representing epiblast cells derived from the inner cell mass of blastocyst-stage embryos. ESCs exhibit a unique combination of self-renewal potency, unlimited proliferation, and pluripotency. The latter is evident by the ability of the isolated cells to differentiate spontaneously into multiple cell lineages, representing the three primary embryonic germ layers. Multiple regulatory networks guide ESCs, directing their self-renewal and lineage-specific differentiation. Apoptosis, or programmed cell death, emerges as a key event involved in sculpting and forming various organs and structures ensuring proper embryonic development. However, the molecular mechanisms underlying the dynamic interplay between differentiation and apoptosis remain poorly understood. AIM To investigate the regulatory impact of apoptosis on the early differentiation of ESCs into cardiac cells, using mouse ESC (mESC) models - mESC-B-cell lymphoma 2 (BCL-2), mESC-PIM-2, and mESC-metallothionein-1 (MET-1) - which overexpress the anti-apoptotic genes Bcl-2 , Pim-2 , and Met-1 , respectively. METHODS mESC-T2 (wild-type), mESC-BCL-2, mESC-PIM-2, and mESC-MET-1 have been used to assess the effect of potentiated apoptotic signals on cardiac differentiation. The hanging drop method was adopted to generate embryoid bodies (EBs) and induce terminal differentiation of mESCs. The size of the generated EBs was measured in each condition compared to the wild type. At the functional level, the percentage of cardiac differentiation was measured by calculating the number of beating cardiomyocytes in the manipulated mESCs compared to the control. At the molecular level, quantitative reverse transcription-polymerase chain reaction was used to assess the mRNA expression of three cardiac markers: Troponin T, GATA4, and NKX2.5. Additionally, troponin T protein expression was evaluated through immunofluorescence and western blot assays. RESULTS Our findings showed that the upregulation of Bcl-2 , Pim-2 , and Met-1 genes led to a reduction in the size of the EBs derived from the manipulated mESCs, in comparison with their wild-type counterpart. Additionally, a decrease in the count of beating cardiomyocytes among differentiated cells was observed. Furthermore, the mRNA expression of three cardiac markers - troponin T, GATA4, and NKX2.5 - was diminished in mESCs overexpressing the three anti-apoptotic genes compared to the control cell line. Moreover, the overexpression of the anti-apoptotic genes resulted in a reduction in troponin T protein expression. CONCLUSION Our findings revealed that the upregulation of Bcl-2 , Pim-2 , and Met-1 genes altered cardiac differentiation, providing insight into the intricate interplay between apoptosis and ESC fate determination.

Abstract:Mesenchymal stem cells (MSCs) specifically differentiate into cardiomyocytes as a potential way to reverse myocardial injury diseases, and uncovering this differentiation mechanism is immensely important. We have previously shown that histone acetylation/methylation and DNA methylation are involved in MSC differentiation into cardiomyocytes induced by islet-1. These modifications regulate cardiac-specific genes by interacting with each other in the promoter regions of these genes, but the molecular mechanism of these interactions remains unknown. In this study, we found that the key enzymes that regulate GATA4/Nkx2.5 expression are Gcn5/HDAC1, G9A, and DNMT-1. When α-methylene-γ-butyrolactone 3 (MB-3) was used to inhibit Gcn5 expression, we observed that the interactions among these key enzymes in the GATA4/Nkx2.5 promoters were blocked, and MSCs could not be induced into cardiomyocytes. Our results indicated that islet-1 could induce Gcn5 binding to GATA4/Nkx2.5 promoter regions and induce the interactions among Gcn5, HDAC1, G9A and DNMT-1, which upregulated GATA4/Nkx2.5 expression and promoted MSC differentiation into cardiomyocytes.
An Experimental Study on Inducing Bone Marrow Mesenchymal Stem Cell to Differentiate into Cardiomyogenic Cells in Cardiomyocyte Cultered Microenviroment in Vitro

Abstract:Objective:To study the effect of cardiomyocyte microenviroment in vitro on bone marrow mesenchymal stem cell(MSCs) in diffierentiating into cardiomyogenic cell. Methods:Bone marrow cells were isolated form SD rat’s leg bone by their adherence to plastic in vitro,the MSCs were purified by culture-expanded and passaged. After treaded with DNA demethylating agent 5-azacytidine(5-aza). In experiment group the MCSs were grown,which were cultured in dishes seeded with cardiomyocytes from neonatal rats. In control group the MSCs were cultured only. The MSCs differentiation was observed by microscope,immunohistochemistry staining for Demin and cardiac troponin I(cTnI),and the positive time were observed. Results:In control group,after treated with 5-aza for 10 days,the clones of adhenrent fibroblast-like cells were formed,and they connected with adjoining cells 15 days and formed myotube-like structures about 20 days later. But the experiment group they formed myotube-like structures after about 17 days. The results of immunohistochemistry staining indiated that the experiment group’s positive time was earlier than the control group’s. Conclusion:The cardiomyocyte microenviroment are thought to be a key factor in inducing bone marrow mesenchymal stem cells to differentiate into cardiomyogenic cells.
Comparison in Expression of Specific Myocardiac Proteins in Developing Myocardium and Differentiated Cardiac Stem Cells

Abstract:To explore the possibility of transplantation of marrow-derived cardiac stem cells(MCSCs) for repairing the ischemic myocardium,expression features of cardiac troponin T(cTnT) and connexin-43(Cx-43) in the developing rat myocardium and the cardiomyocytes differentiated from marrow-derived cardiac stem cells (MCSCs) induced by bone morphogenesis protein-2(BMP-2) were investigated.Expression of cTnT mRNA in the myocardium increased gradually from embryonic rats to adult rats.After induction with BMP-2 for one week, cTnT mRNA of the cells was detected.In week 3 and week 4 after induction,expression of cTnT mRNA of the cells increased significantly.In immunostaining,the cells induced for two weeks expressed cTnT.In week 3 and week 4 after induction,the cells appeared transverse striation-like structures.Cx-43 mRNA was detected in E11 rats.On day 7 after birth,expression of Cx-43 mRNA reached the peak.Then,Cx-43 mRNA expression decreased gradually.In day 17,it trended stable.Cx-43 protein expressed mostly beneath the cell membrane in embryonic period and at cell junctions from day 10 after birth.Changes in expression Cx-43 mRNA were similar with that of cTnT mRNA expression in the cells induced with BMP-2.Expression of Cx-43 protein was observed in week 2 after induction,Cx-43 protein expressed at cell junctions from week 3 to week 4 after induction.These results demonstrate that MCSCs may differentiate into cardiomyocytes under induction with BMP-2.The structural characteristics of differentiated cells are similar with that of mature cardiomyocytes.Therefore,MCSCs are desirable cells for cell transplantation therapy of ischemic heart diseases.

Abstract:A growing body of evidence supports the argument that bone marrow-derived mesenchymal stem cells (MSCs) can differentiate into cardiomyocyte-like cells in an appropriate cellular environment, but the differentiation rate is low. A cocktail method was designed: we investigated the role of 5-azacytidine (5-aza), salvianolic acid B (SalB), and cardiomyocyte lysis medium (CLM) in inducing MSCs to acquire the phenotypical characteristics of cardiomyocytes. The fourth-passage MSCs were treated with 5-aza, SalB, CLM, 5-aza+salB, 5-aza+CLM, SalB+CLM, and 5-aza+SalB+CLM for 2 weeks. Immunofluorescence results showed that cTnT expression in the 5-aza+salB+CLM group was stronger than other groups. Real-time qPCR and Western blotting analyses showed that cTnT, alpha-cardiac actin, mef-2c, Cx43, and GSK-3beta expression increased while beta-catenin expression decreased. The salB+5-aza+CLM group had the most evident effects. SalB combined with 5-aza and CLM improved cardiomyocyte differentiation from MSCs. In the MSCs differentiation process, the Wnt/beta-catenin signaling pathway had been inhibited.
