Abstract:In this study, BaxA (GenBank: KM624029), which encodes the Bacillus amyloliquefaciens xylanase A (BaxA), was highly expressed in Pichia pastoris GS115 under the control of the AOX1 promoter. The recombinant xylanase, namely rePBaxA, was purified to homogeneity by using Ni-affinity resin and its molecular weight was 35.0kDa. The optimum temperature and pH of rePBaxA were 50°C and 5.0, respectively. The kinetic parameters Michaelis–Menten constant (Km) and maximum reaction rate (Vmax) of rePBaxA were 5.41mg/mL and 22.42μmol/min/mL, respectively. High–performance liquid chromatography results showed that after 6h of hydrolysis, rePBaxA released xylose–xylohexaose (X1–X6) mixture from beechwood and birchwood xylan, with xylobiose (X2) and xylotriose (X3) as the major products, respectively. The hydrolyates from oat spelt, wheat bran insoluble xylan and pretreated wheat bran by rePBaxA included X2–X6, with X6 having the highest concentration. The mode of action analysis revealed that rePBaxA was an endo-acting xylanase with transglycosylation activity. X2 might be the minimum oligomer hydrolyzed by rePBaxA. The pretreated wheat bran and wheat bran insoluble xylan could be directly hydrolyzed by rePBaxA. This study provided a basis for using agricultural waste by-products as substrates for manufacting value-added probiotics, namely, xylooligosaccharides.

Abstract:A novel β-xylosidase gene of glycosyl hydrolase (GH) family 3, xyl3A, was identified from the thermophilic fungus Humicola insolens Y1, which is an innocuous and non-toxic fungus that produces a wide variety of GHs. The cDNA of xyl3A, 2334 bp in length, encodes a 777-residue polypeptide containing a putative signal peptide of 19 residues. The gene fragment without the signal peptide-coding sequence was cloned and overexpressed in Pichia pastoris GS115 at a high level of 100 mg/L in 1-L Erlenmeyer flasks without fermentation optimization. Recombinant Xyl3A showed both β-xylosidase and α-arabinfuranosidase activities, but had no hydrolysis capacity towards polysaccharides. It was optimally active at pH 6.0 and 60°C with a specific activity of 11.6 U/mg. It exhibited good stability over pH 4.0-9.0 (incubated at 37°C for 1 h) and at temperatures of 60°C and below, retaining over 80% maximum activity. The enzyme had stronger tolerance to xylose than most fungal GH3 β-xylosidases with a high Ki value of 29 mM, which makes Xyl3A more efficient to produce xylose in fermentation process. Sequential combination of Xyl3A following endoxylanase Xyn11A of the same microbial source showed significant synergistic effects on the degradation of various xylans and deconstructed xylo-oligosaccharides to xylose with high efficiency. Moreover, using pNPX as both the donor and acceptor, Xyl3A exhibited a transxylosylation activity to synthesize pNPX2. All these favorable properties suggest that Xyl3A has good potential applications in the bioconversion of hemicelluloses to biofuels.
Improving the Quality of Wheat Flour Bread by a Thermophilic Xylanase with Ultra Activity and Stability Reconstructed by Ancestral Sequence and Computational-Aided Analysis

Abstract:Xylanase is an essential component used to hydrolyze the xylan in wheat flour to enhance the quality of bread. Presently, cold-activated xylanase is popularly utilized to aid in the development of dough. In this study, ancestral sequence reconstruction and molecular docking of xylanase and wheat xylan were used to enhance the activity and stability of a thermophilic xylanase. The results indicated that the ancestral enzyme TmxN3 exhibited significantly improved activity and thermal stability. The Vmax increased by 2.7 times, and the catalytic efficiency (Kcat/Km) increased by 1.7 times in comparison to TmxB. After being incubated at 100 °C for 120 min, it still retained 87.3% of its activity, and the half-life in 100 °C was 330 min, while the wild type xylanase was only 55 min. This resulted in an improved shelf life of bread, while adding TmxN3 considerably enhanced its quality with excellent volume and reduced hardness, chewiness, and gumminess. The results showed that the hardness was reduced by 55.2%, the chewiness was reduced by 40.11%, and the gumminess was reduced by 53.52%. To facilitate its industrial application, we further optimized the production conditions in a 5L bioreactor, and the xylanase activity reached 1.52 × 106 U/mL culture.

Optimization of fermentation conditions for xylanase production by engineered Pichia pastoris

Abstract:The optimal fermentation conditions for production of xylanase by engineered Pichia pastoris GS115-ATx in shake-flask cultivation was investigated. The results showed that the best carbon and nitrogen sources for xylanase production were bran and beef extract, respectively. Judging from their effect on xylanase production efficiency, organic nitrogen was better than inorganic nitrogen. The best methanol concentration was 0.5% for inducing xylanase expression. The xylanase activity was increased when 0.5% glycerin was added into the culture every 24 h. In addition, Toween 20, Toween 80, and betaine were found to increase the yield of xylanase, and among them betaine was the optimal stimulator. SDS-PAGE analysis revealed that the molecular weight of xylanase expressed by GS115-ATx was about 19.0 kDa. The xylanase activity of the culture supernatant reached its peak after 96-h methanol induction.
Combined strategies for improving the heterologous expression of a novel xylanase from Fusarium oxysporum Fo47 in Pichia pastoris

Abstract:Xylanase, an enzyme capable of hydrolyzing non-starch polysaccharides found in grain structures like wheat, has been found to improve the organizational structure of dough and thus increase its volume. In our past work, one promising xylanase FXYL derived from Fusarium oxysporum Fo47 and first expressed 779.64 U/mL activity in P. pastoris . It has shown significant potential in improving the quality of whole wheat bread, making it become a candidate for development as a new flour improver. After optimization of expression elements and gene dose, the xylanase activity of FXYL strain carrying three-copies reached 4240.92 U/mL in P. pastoris . In addition, 12 factors associated with the three stages of protein expression pathway were co-expressed individually in order in three-copies strain, and the translation factor Pab1 co-expression increased FXYL activity to 8893.53 U/mL. Nevertheless, combining the most effective or synergistic factors from three stages did not exhibit better results than co-expressing them alone. To further evaluate the industrial potential, the xylanase activity and protein concentration reached 81184.51 U/mL and 11.8 g/L in a 5 L fed-batch fermenter. These engineering strategies improved the expression of xylanase FXYL by more than 104-fold, providing valuable insights for the cost-effective industrial application of FXYL in the baking field. Abstract:A novel thermostable xylanase gene from Chaetomium sp. CQ31 was cloned and codon-optimized (CsXynBop). The deduced protein sequence of the gene shared the highest similarity of 75% with the glycoside hydrolase (GH) family 10 xylanase from Achaetomium sp. Xz-8. CsXynBop was over-expressed in Pichia pastoris GS115 by high-cell density fermentation, with the highest xylanase yield of 10,017 U/mL. The recombinant xylanase (CsXynBop) was purified to homogeneity and biochemically characterized. CsXynBop was optimally active at pH 6.5 and 85 °C, respectively, and stable over a broad pH range of 5.0-9.5 and up to 60 °C. The enzyme exhibited strict substrate specificity towards oat-spelt xylan (2, 489 U/mg), beechwood xylan (1522 U/mg), birchwood xylan (1067 U/mg), and showed relatively high activity towards arabinoxylan (1208 U/mg), but exhibited no activity on other tested polysaccharides. CsXynBop hydrolyzed different xylans to yield mainly xylooligosaccharides (XOSs) with degree of polymerization (DP) 2-5. The application of CsXynBop (200 U/g malt) in malt mashing substantially decreased the filtration time and viscosity of malt by 42.3% and 8.6%, respectively. These excellent characteristics of CsXynBop may make it a good candidate in beer industry.

Abstract:A novel endo-1,4-β-xylanase-encoding gene was identified in Alicyclobacillus mali FL18 and the recombinant protein, named Am Xyn, was purified and biochemically characterized. The monomeric enzyme worked optimally at pH 6.6 and 80 °C on beechwood xylan with a specific activity of 440.00 ± 0.02 U/mg and a good catalytic efficiency ( k cat /K M = 91.89 s −1 mLmg −1 ). In addition, the enzyme did not display any activity on cellulose, suggesting a possible application in paper biobleaching processes. To develop an enzymatic mixture for xylan degradation, the association between Am Xyn and the previously characterized β-xylosidase Am βXyl, deriving from the same microorganism, was assessed. The two enzymes had similar temperature and pH optima and showed the highest degree of synergy when Am Xyn and Am βXyl were added sequentially to beechwood xylan, making this mixture cost-competitive and suitable for industrial use. Therefore, this enzymatic cocktail was also employed for the hydrolysis of wheat bran residue. TLC and HPAEC-PAD analyses revealed a high conversion rate to xylose (91.56 %), placing Am Xyn and Am βXyl among the most promising biocatalysts for the saccharification of agricultural waste.

Bacterial Xylanase in Pseudomonas Boreopolis LUQ1 is Highly Induced by Xylose

Abstract:A xylanase producing bacterium was isolated from paper mill sludge in Thunder Bay, Canada. The newly isolated bacterium was identified as Pseudomonas boreopolis according to its 16S rRNA gene sequence and designated as LUQ1. The zymogram analysis indicated that there was one band of protein with xylanase activity, and the molecular weight of the enzyme was about 20 kDa. This xylanase works best at pH 6.0 and 65°C. The xylanase can be induced to express by xylose. The expression was enhanced on increasing the concentration of xylose, which reached its highest activity at 12 mM of xylose in fermentation. Wheat bran was the best carbon source in submerged fermentation. The highest xylanase activity of 25.61 U/ml was obtained at 96 h using wheat bran feedstock. When barley straw and wheat bran were used as feedstocks, the addition of 10 mM of xylose increased xylanase activity by ~50% and ~15%, respectively. The results showed that the strain LUQ1 has a great potential to produce xylanase for industrial applications.

Abstract:A novel alkaline xylanase gene ( xyn AI) from Bacillus amyloliquefaciens was highly expressed in Pichia pastoris , and the enzyme was purified and characterized. Sequence analysis indicated that XynAI belongs to xylanases family GH11 with 5 potential N-glycosylation sites and 3 salt bridges. The purified XynAI had molecular weight of 22 kDa, the optima temperature and pH of 55°C and pH 8.6, respectively. Furthermore, XynAI has high thermal (30–70°C) and pH (pH 5.0–10.0) stability.Mg 2+ , Ni 2+ , Ca 2+ and Fe 2+ at concentration of 1 mM enhanced the xylanase activity by 6.2, 6.5, 7.9 and 202.6%, respectively. Under optimal conditions, the K M and V max values of XynAI for beechwood xylan were 8 mg/mL and 83.3 µmol min –1 mg of protein –1 , respectively. In the culture supernatant, the maximal protein concentration and xylanase activity of XynAI reached 3.98 mg/mL and 292 U/mg, respectively. The biochemical properties of XynAI showed that it may be a promising candidate for industrial applications.

Abstract:Microbial xylanase has wide application in bioenergy, animal feed, environmental protection, the pulp and paper industry, and agricultural development. In this study, three xylanases from the microbiota of T. fuciformis with its companion strains were identified by metagenomics sequencing. The three enzymes were subjected to cloning and expression in E. coli or P. pastoris, purification, and characterization for their properties. The results showed that AsXyn1, from Annulohypoxylon stygium, among the three enzymes possessed high thermostability at 40 °C and broad pH tolerance in the range of 2.0–10.0, exhibiting its application potential. Furthermore, it was found that post-translational modification (such as glycosylation) of AsXyn1 enzyme modulated its activity, kinetic parameters, and thermostability. These results and findings provided a hint for enzyme modification and design in future.

Abstract:Xylanolytic enzymes cleave the β-1,4-glycosidic bonds within xylan, the primary polymer found in the hemicellulosic fraction of lignocellulosic biomass, converting it into xylose. This enzymatic class holds significant applications in various biotechnological processes, particularly within the pharmaceutical, food, and bioenergy industries. This study focuses on a cost-effective method for producing a xylanolytic blend (XB) through the solid-state fermentation of the low-cost coffee husk (CH) by-product, using Penicillium roqueforti ATCC 10110. Optimal bioprocess conditions were identified at 59% humidity and 16 °C, resulting in xylanolytic activity of 13.20 U/g. The XB exhibited favorable thermostability at 40 °C, with maximum activity at 50 °C and pH 5. The effect of solvents revealed significantly enhanced activity with dichloromethane and hexane. The presence of metallic salts, including Pb(C 2 H 3 O 2 ), Na 2 CO 3 , KCl, FeSO 4 , CuSO 4 , MgSO 4 , and ZnSO 4 , led to more than a 100% increase in enzyme activity, with Na 2 CO 3 demonstrating an outstanding 229.9% enhancement. Similarly, other organic compounds such as EDTA, SDS, Triton X-100, and Trolox significantly increased enzymatic activity (+ 286.69% for Triton X-100), while other salts such as CaCO 3 , MgCl 2 , and Al(NO 3 ) 3 led to inhibition. These results differ from previous reports of xylanases from this microorganism and position the developed XB as a promising sustainable catalyst for the saccharification of CH. The bio-based recycling approach elevates the value of CH and proposes an alternative to conventional fertilizer use. The basis developed here serves as guidelines for further investigations exploring the XB application in high-grade pharmaceuticals, food, and bioenergy in large-scale scenarios.

Abstract:Xyloglucanase had been extracted from the culture medium fermented by Paenibacillus xylanilyticus.Isolated and purified by methods of ammonium sulfate precipitation,HiPrep26/10 Desalting gel layer chromatography,HiPrepDEAE FF16/10 anion exchange column,HiPrep 16/60 Sephacryl S-100 gel layer chromatography and HiPrep 16/10 Source 30S cation exchange column.The molecular weight of xyloglucanase was about 39 kD,identified by SDS-PAGE.The optimal reaction temperature of xyloglucanase was 50℃,and there was good stabi-lity under 60℃.The optimal reaction pH of xyloglucanase was 7.0,and there was good stability between pH5.0 and pH10.0.The dynamic study demonstrated that the Km value of xyloglucanase was 65 g/L,the maximum reaction velocity(Vmax) was 6.49 μmol/min,and the kcat was 10.86 s-1.The substrate specificity of the xyloglucanase indicated that the xyloglucan had much higher specific activity than other substrates.The purified enzyme was analyzed by mass spectrometry,and the result showed that the purified enzyme had high homology with xyloglucanase from Paenibacillus pabuli.It was the first report about xyloglucanase product from Paenibacillus xylanilyticus.
A novel cold-active GH8 xylanase from cellulolytic myxobacterium and its application in food industry

Abstract:Although xylanase have a wide range of applications, cold-active xylanases have received less attention. In this study, a novel glycoside hydrolase family 8 (GH8) xylanase from Sorangium cellulosum with high activity at low temperatures was identified. The recombinant xylanase (XynSc8) was most active at 50 degrees C, demonstrating 20% of its maximum activity and strict substrate specificity towards beechwood and corncob xylan at 4 degrees C with V(max )values of 968.65 and 1521.13 mu mol/mg/min, respectively. Mesophilic XynSc8 was active at a broad range of pH and hydrolyzed beechwood and corncob xylan into xylooligosaccharides (XOS) with degree of polymerization greater than 3. Moreover, incorporation of XynSc8 (0.05-0.2 mg/kg flour) provided remarkable improvement (28-30%) in bread specific volume and textural characteristics of bread compared to commercial xylanase. This is the first report on a novel cold-adapted GH8 xylanase from myxobacteria, suggesting that XynSc8 may be a promising candidate suitable for bread making.
Production, Purification and Characterization of an Alkaliphilic Endo-Β-1,4-xylanase from a Microbial Community EMSD5

Abstract:In this study, a novel alkaliphilic extracellular endo-β-1,4-xylanase was purified from the culture filtrate of microbial community EMSD5, which could grow in liquid culture fermentation under static condition with corn stover as inducement. Single step purification was achieved using anion exchange chromatography. The purified enzyme showed a single band on SDS polyacrylamide gel electrophoresis (SDS-PAGE) with an apparent molecular weight of 22kDa and pI of 7.1. Stimulation of the activity by β-mercaptoethanol, and dithiotheritol (DTT) was observed. Moderately thermostable xylanase showed optimum activity at 50°C in pH 8.0. 16S rDNA clone library analysis indicated this community was mainly composed of bacteria related to genera Clostridium, Acinetobacter, Bacteroides, Lysinibacillus and Dysgonomonas. The present work reports for the first time the purification and characterization of an alkaliphilic xylanase from a microbial community, and the alkaliphilicity and moderate thermostability of this xylanase may have potential implications in pulp and paper industries.
Characterization of a GH10 extremely thermophilic xylanase from the metagenome of hot spring for prebiotic production

Abstract:A xylanase gene (named xyngmqa ) was identified from the metagenomic data of the Gumingquan hot spring (92.5 °C, pH 9.2) in Tengchong City, Yunnan Province, southwest China. It showed the highest amino acid sequence identity (82.70%) to endo-1,4-beta-xylanase from Thermotoga caldifontis . A constitutive expression plasmid (denominated pSHY211) and double-layer plate (DLP) method were constructed for cloning, expression, and identification of the XynGMQA gene. The XynGMQA gene was synthesized and successfully expressed in Escherichia coli DH5α. XynGMQA exhibited optimal activity at 90 °C and pH 4.6, being thermostable by maintaining 100% of its activity after 2 h incubated at 80 °C. Interestingly, its enzyme activity was enhanced by high temperatures (70 and 80 °C) and low pH (3.0–6.0). About 150% enzyme activity was detected after incubation at 70 °C for 20 to 60 min or 80 °C for 10 to 40 min, and more than 140% enzyme activity after incubation at pH 3.0 to 6.0 for 12 h. Hydrolytic products of beechwood xylan with XynGMQA were xylooligosaccharides, including xylobiose (X2), xylotriose (X3), and xylotetraose (X4). These properties suggest that XynGMQA as an extremely thermophilic xylanase, may be exploited for biofuel and prebiotic production from lignocellulosic biomass.

However, little is known about the genetic basic of utilizing xylan in this isolate. To further investigate the molecular mechanism of xylan metabolism, genome sequencing was carried out. The draft genome obtained has been deposited with GenBank. Endo-1,4-β-xylanase (Xyn) was acquired, followed by characterization of its cellular localization and expression profiles. Xyn proved to be a novel enzyme, differing from xylanases from similar species according to combined analyses from multiple sequence alignments and putative three-dimensional structure modeling. Moreover, recombinant expression was optimized in heterogeneous hosts to further determine optimum pH and temperature conditions as well as residual activity in the presence of metal ions and inhibitors. Finally, substrate specificity and kinetic parameters were also studied. In conclusion, our findings illustrate the genetic background related to xylan metabolism in this isolate. In addition, a novel Xyn that is expected to have potential in the bio-degradation of hemicellulosic materials was expressed, purified and characterized.

Abstract:Abstract Malignant ascites negatively impacts patient's quality of life and has significant impact on the health care resources. Majority of management guidelines are based on systemic reviews that have predominately relied on retrospective data. Therefore, there is lack of high-level evidence-based studies. In this review, the etiologies, pathophysiology, and various treatment methods including diuretic therapy, large volume paracentesis, indwelling catheter placement, peritoneovenous shunt, transjugular intrahepatic portosystemic shunt, and other available novel and/or experimental options are reviewed.
Contributions and characteristics of two bifunctional GH43 β-xylosidase /α-L-arabinofuranosidases with different structures on the xylan degradation of Paenibacillus physcomitrellae strain XB

Abstract:Xylan is one of the major polymeric hemicellulosic constituents of lignocellulosic biomass, and its effective utilization by microorganisms is crucial for the economical production of biofuels. In this study, Paenibacillus physcomitrellae XB exhibited different xylan degradation ability on different substrates of corncob xylan (CCX), oat spelt xylan (OSX), wheat flour arabinoxylan (AX) and beech wood xylan (BWX). The RT-QPCR result showed that two genes (Pph_0602 and Pph_2344) belonging to the glycoside hydrolase family 43 were up-regulated more than 5-fold on CCX and xylose. Substrate-specific assays with purified proteins Ppxyl43A (Pph_0602) and Ppxyl43B (Pph_2344) revealed that both exhibited β-xylosidase activity toward the chromogenic substrate p-nitrophenyl-β-D-xylopyranoside, and α-L-arabinofuranosidase activity toward p-nitrophenyl-α-L-arabinofuranoside, indicating their bifunctionality. By testing their degradation characteristics on different natural substrates, it was found that both Ppxyl43A and Ppxyl43B showed similar degradation ability on CCX and OSX. Both enzymes could hydrolyze xylohexaose and xylobiose completely to xylose, but could not hydrolyze BWX and AX, suggesting they mainly hydrolyze xylo-oligosaccharides by β-xylosidase activity. Further analysis showed that both of them displayed very high pH stability and thermostability on the β-xylosidase activity, but Ppxy143B exhibited wider pH and temperature ranges, higher pH and temperature stability, was less influenced by metal ions, and had a slower start-up response than Ppxyl43A. Given their predicted structure, it is likely that the enzymatic differences between Ppxyl43A and Ppxyl43B might be related to the extra C-terminus domain (GH43_C2) in Ppxyl43B, which could enhance the enzymatic stability while restricting the substrates' or metal ions' access to the active sites of Ppxyl43B. In conclusion, both Ppxyl43A and Ppxyl43B were β-xylosidase/α-L-arabinofuranosidase bifunctional enzymes and might be useful in xylan biomass conversion, especially in the hydrolysis of xylo-oligosaccharides into xylose.
