Abstract:Capscaicin was separated from capsaicinoide by reversed-phase preparative liquid chromatography. Using a ODS column (200 mm×10.0 mm), the effects of mobile phase and flow rate, feed concentration and volume on the purity, productivity, recovery and mobile phase consumption were studied respectively, and the suitable operation conditions were obtained as follows: the mobile phase is methanol/water (75/25 v/v) , and the feed concentration and volume are 180 mg·mL-1 and 2.08 mL, respectively; while the flow rate is 4 mL·min-1. Under above conditions, the capscaicin with a purity of 95.23% could be achieved, and the productivity, recovery and mobile phase consumption are 4.38 g·L-1·h-1, 93.27% and 0.087 L·g-1, respectively. This technology proposed provides a simple and effective method for the preparation of capscaicin.

Abstract:A high performance liquid chromatographic method was developed for the separation and determination of centellasaponin A and asiaticoside isomers in Centella asiatica products. The method comprises separation by reversed phase chromatography on a Synergi 4μ Hydro-RP 80A reversed-phase column using acetonitrile-water (21:79, v/v) as mobile phase. Centellasaponin A and asiaticoside were separated with high resolution by the developed method. The method's linearity, precision, and recovery were validated. The effects of mobile phase composition, temperature, and flow rate on the separation of the two isomers were also investigated. Experimental results indicated that acetonitrile-water as the mobile phase offered enhancing resolution to the polar isomers. Furthermore, it was observed that the sequence of centellasaponin A and asiaticoside was reversed in peak consequence by using methanol-water as the mobile phase compared to acetonitrile-water, which could be used for the separation of other isomeric triterpenoid saponins. Construction of On-Line Supercritical Fluid Extraction with Reverse Phase Liquid Chromatography–tandem Mass Spectrometry for the Determination of Capsaicin

Abstract:A novel on-line system composed of supercritical fluid extraction（SFE）, dilution line and reverse phase liquid chromatography/mass spectrometry（RPLC/MS） was constructed for on-line extraction and reverse phase separation of some fat-soluble components in foods. Three columns including a trap column,concentration column and analytical column were used for trapping the fat-soluble components, on-line enrichment and reverse phase separation, respectively. Capsaicinoids were on-line extracted by a CO 2 supercritical fluid, then concentrated and separated by using the C 18 columns, finally detected by mass spectrometry（MS）. Capsaicin eluted at 10.1 min and limit of detection（LOD, S/N=3） for the standard solution is 0.55pg. The linearity was calculated with a value of coefficient of determination（R~2）≥0.998 in the range of 1.1–8.5 ng. Concentrations of capsaicin in the green, yellow, and red bell peppers were determined to be 60.33 ng/g, 31.79 ng/g, 35.38ng/g, respectively.

Abstract:A speedy and selective ultra-HPLC-MS/MS method for simultaneous determination of deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON), 15-ADON. nivalenol and fusarenon X in traditional Chinese medicines (TCMs) was developed. The method was based on one-step sample cleanup using reliable homemade cleanup cartridges. A linear gradient mobile-phase system, consisting of water containing 0.2% aqueous ammonia and acetonitrile/methanol (90:10, v/v) at a flow rate of 0.4 mL/min, and an Acquity UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m) were employed to obtain the best resolution of the target analytes. [C-13(15)]-DON was used as the internal standard to accomplish as accurate as possible quantitation. The established method was further validated by determining the linearity (R-2 >= 0.9990), sensitivity (LOQ, 0.29-0.99 mu g/kg), recovery (88.5-119.5%) and precision (RSD <= 15.8%). It was shown to be a suitable method for simultaneous determination of DON, 3-ADON, 15-ADON, nivalenol and fusarenon X in various TCM matrices. The utility and practical impact of the method was demonstrated using different TCM samples.

Abstract:A multi-channel counter-current chromatography (CCC) method has been designed and fabricated for the high-throughput fractionation of natural products without complications sometimes encountered with other conventional chromatographic systems, such as irreversible adsorptive constituent losses and deactivation, tailing of solute peaks and contamination. It has multiple independent CCC channels and each channel connects independent separation column(s) by parallel flow tubes, and thus the multi-channel CCC apparatus can achieve simultaneously two or more independent chromatographic processes. Furthermore, a high-throughput CCC fractionation method for natural products has been developed by a combination of a new three-channel CCC apparatus and conventional parallel chromatographic devices including pumps, sample injectors, effluent detectors and collectors, and its performance has been displayed on the fractionation of ethyl acetate extracts of three natural materials Solidago canadensis, Suillus placidus, and Trichosanthes kirilowii, which are found to be potent cytotoxic to tumor cell lines in the course of screening the antitumor candidates. By combination of biological screening programs and preparative high-performance liquid chromatography (HPLC) purification, 22.8mg 6β-angeloyloxykolavenic acid and 29.4mg 6β-tigloyloxykolavenic acid for S. canadensis, 25.3mg suillin for S. placidus, and 6.8mg 23,24-dihydrocucurbitacin B for T. Kirilowii as their major cytotoxic principles were isolated from each 1000mg crude ethyl acetate exctract. Their chemical structures were characterized by electrospray ionization mass spectrometry, one- and two-dimensional nuclear magnetic resonance. The overall results indicate the multi-channel CCC is very useful for high-throughput fractionation of natural products for drug discovery in spite of the solvent balancing requirement and the lower resolution of the shorter CCC columns.

Abstract:From nutritional points of view, carotenoids, capsaicinoids, and tocopherols are valuable constituents in pungent peppers. A rapid and reliable high performance liquid chromatography (HPLC) method for the simultaneous determination of phytonutrients in spice red peppers and chili products was developed and validated. The method included simultaneous detection by fluorescence and diode-array detectors. The major capsaicinoids, two tocopherols and 43 carotenoid components, were simultaneously separated, detected, and identified in the appointed pepper powder (containing Capsicum annuum and Capsicum frutescens) for method validation. The separation was performed on a Nucleosil C18 reverse phase column and optimized gradient elution. Resolution ranged between 0.96 and 1.46 with the highest values corresponding to γ-tocopherol and α-tocopherol. The limits of detection and quantification of target compounds ranged between 18.77 and 148.08 ng mL−1. Recoveries were between 89.83–100.26 and 79.72–88.86% when standard materials were spiked at low and high amounts, respectively. The most sufficient extraction of the different phytonutrients was achieved by mixture of methanol and acetone, although it was only slightly better than the mixture of methanol and acetonitrile. These results suggest that the developed method could be used for rapid, one-step determination of a wide range of phytonutrients in chili and pepper powders.

Abstract:INTRODUCTION:Charged aerosol detection (CAD) has the merits of high sensitivity, high universality and response uniformity. The strategy that combines the quantitative analysis of multi-components by single marker (QAMS) with CAD has certain advantages for the quantification of multi-components. However, relevant research was limited.OBJECTIVES:To comprehensively investigate the crucial factors that affect the performance of the HPLC-CAD-QAMS approach and validate the credibility and feasibility of the method.METHODOLOGY:Multiple components of Qishen Yiqi dripping pills (QSYQ) were assayed using the high-performance liquid chromatography (HPLC)-CAD-QAMS approach. Some factors that affect the sensitivity and accuracy of the approach were sufficiently studied. After the method verification, principal component analysis (PCA) was applied to evaluate the quality consistency of three types of samples: normal samples, expired samples and negative samples.RESULTS:A HPLC-CAD-QAMS method was successfully developed for the multi-component determination of QSYQ. First, chromatographic conditions were optimised by a definitive screening design, and the optimised ranges of operating parameters were obtained with a Monte Carlo simulation method. Next, a new method to select the internal reference standards was successfully introduced based on the heatmap of Pearson correlation coefficients of the response factors. Then, the multi-point method was selected to calculate the relative correction factors, and a robustness test was conducted with Plackett-Burman design. Finally, the PCA was proved to be effective for the quality consistency evaluation of different samples.CONCLUSION:The developed HPLC-CAD-QAMS method can be a reliable and superior means for the multi-component quantitative analysis of QSYQ.
Determination of Capsaicin and Dihydrocapsaicin in Some Chilli Varieties using Accelerated Solvent Extraction Associated with Solid-Phase Extraction Methods and RP-HPLC-Fluorescence

Abstract:Reversed phase-HPLC with fluorescence detection of two major capsaicinoids was described. Isocratic elution using a ratio of methanol and deionized water (66:34, v/v) as mobile phase was used at a flow rate of 0.9 mL/min with well achieved separation within 6 min. Under optimum conditions, their analytical figures of merit for the HPLC method were validated. The linearity was in the range of 1.0-25.0 μg/mL with multiple determination coefficients of higher than 0.995. The limit of detection was ranged of 0.008-0.01 μg/mL. The repeatability and reproducibility of the retention time and peak area for these compounds were in good precision with their relative standard deviations (RSDs) lower than 1% and 5%, respectively. Both capsaicin and dihydrocapsaicin were extracted using an accelerated solvent extraction (ASE) of methanol as an extraction solvent for 5 min static time with 3 cycles. The methanolic extracts were subjected to clean up with C 18 solid-phase extraction (SPE) with its recoveries ranking of 90.2-98.0%. The method recoveries of real samples were found to be 60.7-98.6%. The optimized extraction method were applied for the determination of the two capsaicinoids in ten vareities of hot chilli pepper samples. Total contents of capsaicinoids were found in the range of 2,307.0-9047.3 μg/g DW with their corresponding Scoville heat unit (SHU) of 34,600-135,700. Additionally, the contents of capsaicinoids using external calibration method comparing with those of standard addition were not significantly different, indicating accuracy of the method. Mostly, the contents of capsaicin found in these real samples were rather higher than those of dihydrocapsaicin.
Analysis Of Polycyclic Aromatic Hydrocarbons By Capillary Electrochromatography By Using Capillary Columns Packed With Polycyclic-Aromatic-Hydrocarbon-Specific Particles

Abstract:As a result of their carcinogenic, teratogenic and mutagenic properties, polycyclic aromatic hydrocarbons are considered as priority pollutants by the US Environmental Protection Agency and the analysis of polycyclic aromatic hydrocarbons in environment draws extensive attention in the analytical community. The diversity of polycyclic aromatic hydrocarbons in terms of chemical structures and properties leads to the requirement for highly efficient separation technologies. This study describes the usage of polycyclic-aromatic-hydrocarbon-specific particles as a stationary phase for the separation of polycyclic aromatic hydrocarbons by capillary electrochromatography. A comparison of the separations of 16 polycyclic aromatic hydrocarbons performed on capillary columns packed with polycyclic-aromatic-hydrocarbon-specific particles and C-18 silica microspheres indicated that polycyclic-aromatic-hydrocarbonspecific particles showed a wider distribution and larger gap in chromatographic retention factor (0.6-41.8) toward polycyclic aromatic hydrocarbons than C-18 (2.0-14.2). As a result, resolutions of polycyclic aromatic hydrocarbons on columns packed with polycyclic-aromatic-hydrocarbon-specific particles (1.2-9.9) were significantly superior over those on C-18 -packed capillary columns (0.5-6.3). A mobile phase of 20 mM Tris (pH 8.5)/acetonitrile (15:85 v/v) was optimized for the rapid separation of polycyclic aromatic hydrocarbons at a pressurized flow rate of 0.2 mL min(-1) under an applied voltage of -16 kV. Limits of detection of 16 polycyclic aromatic hydrocarbons were obtained in the range 0.020-0.89 mg L-1 with good precisions (intracolumn reproducibilities of 0.2-1.9% for retention time, 1.4-4.2% for peak height and 1.4-4.4% for peak area, and intracolumn reproducibilities of 2.6-4.2% for retention time, 3.4-7.2% for peak height and 6.9-9.7% for peak area). The proposed method was employed for polycyclic aromatic hydrocarbons analysis in fresh water with good recovery (87.6-112.4%), where polycyclic aromatic hydrocarbons in all water samples were present at levels below the detection limits.
Development of an RP-HPLC Method for Quantifying Diclofenac Diethylamine, Methyl Salicylate, and Capsaicin in Pharmaceutical Formulation and Skin Samples

Abstract:An RP-HPLC method with a UV detector was developed for the simultaneous quantification of diclofenac diethylamine, methyl salicylate, and capsaicin in a pharmaceutical formulation and rabbit skin samples. The separation was achieved using a Thermo Scientific ACCLAIMTM 120 C18 column (Waltham, MA, USA, 4.6 mm × 150 mm, 5 μm). The optimized elution phase consisted of deionized water adjusted to pH = 3 using phosphoric acid mixed with acetonitrile in a 35:65% (v/v) ratio with isocratic elution. The flow rate was set at 0.7 mL/min, and the detection was performed at 205 nm and 25 °C. The method exhibits good linearity for capsaicin (0.05–70.0 μg/mL), methyl salicylate (0.05–100.0 μg/mL), and diclofenac diethylamine (0.05–100.0 μg/mL), with low LOD values (0.0249, 0.0271, and 0.0038 for capsaicin, methyl salicylate, and diclofenac diethylamine, respectively). The RSD% values were below 3.0%, indicating good precision. The overall greenness score of the method was 0.61, reflecting its environmentally friendly nature. The developed RP-HPLC method was successfully applied to analyze Omni Hot Gel® pharmaceutical formulation and rabbit skin permeation samples.

Abstract:A newly developed portable capillary liquid chromatograph was investigated for the separation of various pharmaceutical and illicit drug compounds. The system consists of two high-pressure syringe pumps capable of delivering capillary-scale flow rates at pressures up to 10,000 psi. Capillary LC columns packed with sub-2 μm particles are housed in cartridges that can be inserted into the system and easily connected through high-pressure fluidic contact points by simply applying a specific, pre-determined torque rather than using standard fittings and less precise sealing protocols. Several over-the-counter analgesic drug separations are demonstrated, along with a simple on-line measurement of tablet dissolution. Twenty illicit drug compounds were also separated across six targeted drug panels. The results described in this study demonstrate the capability of this compact LC instrument to address several important drug-related applications while simplifying system operation, and greatly reducing solvent usage and waste generation essential for on-site analysis. Abstract:Comprehensive two-dimensional separation by capillary liquid chromatography coupled with capillary micellar electrokinetic chromatography was investigated. Fast micellar electrokinetic chromatography was achieved by speeding up electroosmotic flow and improving solute mass transfer in micellar pseudo-stationary phase. High efficiency separation was achieved by selecting operating conditions based on theoretical relationships and experimental data. A new gating interface from the first dimension to the second dimension based on effluent stacking-injection was developed. The reproducibility of the interface for sample transfer efficiency, migration time, and peak height was studied. The overall system performance was verified in the separation of complex neutral components. Resolution of hundreds of compounds present in traditional Chinese medicines was demonstrated.
UFLC simultaneous determination of two narcotics compositions in a novel spice

Abstract:Objective: To establish an UFLC method for simultaneous determination of two compositions(JWH-073,JWH-018) in a new type spice. Methods: A UFLC method was applied with a Shimpack XRODS Ⅱ chromatographic column(2. 0 mm × 75 mm,2. 2 μm),the mobile phase consisted of acetonitrile and water,eluted in gradient mode(0-10 min,65%→85% acetonitrile; 10-11 min,85%→65% acetonitrile; 11-15 min, 65% acetonitrile). The mobile phase was pumped at a ow rate of 0. 25 mL·min-1. The UV detection wavelength was set at 280 nm. The column temperature was controlled at 35 ℃. The injection volume was 5 μL. The contents of JWH-073 and JWH-018 in the new type spice were calculated by external standardization method. Results: There were good linear relationships for JWH-073 and JWH-018. The average recoveries(n = 9) were 98. 58%, 98. 72%,respectively. Conclusion: The established UFLC method is simple,sensitive,reproducible and feasible for simultaneous determination of the two compositions,JWH-073 and JWH-018 in the new type spice.
A Novel Nonionic Micellar Liquid Chromatographic Method for Simultaneous Determination of Pseudoephedrine, Paracetamol, and Chlorpheniramine in Cold Compound Preparations

Abstract:A novel analytical method of nonionic micellar high-performance liquid chromatography has been developed to simultaneously separate and determine three active ingredients in cold compound preparations, in which the contents of paracetamol were significantly higher than that of pseudoephedrine and chlorpheniramine. A C18 column was used as the stationary phase and ultraviolet detection wavelengths were selected at 210 nm for chlorpheniramine and pseudoephedrine and 300 nm for paracetamol. The micellar mobile phase was composed of an aqueous solution (containing 3.0 × 10−3 mol/L Brij-35 and 2.0 × 10−2 mol/L potassium dihydrogen phosphate)-methanol (96:4, v/v) at pH 3. Under the conditions, the analytes were separated in 10 min. The method was validated by limit of quantitation, limit of detection, linearity, stability, precision, and recovery. The possible separation mechanism of the proposed method was also briefly discussed. Compared with sodium dodecyl sulfate and cetyltrimethylammonium bromide, the ultraviolet absorption produced by Brij-35 can be neglected. The novel method used much lower amount of organic solvent (4%, v/v). Programmed ultraviolet detection improved the accuracy of simultaneous determination of the analytes whose contents were in great disparity. The method was successfully applied to determine pseudoephedrine, paracetamol, and chlorpheniramine in cold compound preparations.

Deep Eutectic Solvent-Based Dispersive Liquid–Liquid Microextraction Coupled with LC-MS/MS for the Analysis of Two Ochratoxins in Capsicum

Abstract:Ochratoxins, a common class of mycotoxin in capsicum, and techniques and methods for the determination of mycotoxins in spices have been increasingly developed in recent years. An innovative and eco-friendly method of dispersive liquid–liquid microextraction (DLLME) was demonstrated in this study, based on a synthesized deep eutectic solvent (DES) combined with LC-MS/MS, for the quantification and analysis of two ochratoxins in capsicum. The DES-DLLME method parameters entail selecting the DES type (thymol:decanoic acid, molar ratio 1:1) and DES volume (100 μL). The volume of water (3 mL) and salt concentration (0 g) undergo optimization following a step-by-step approach to achieve optimal target substance extraction efficiency. The matrix effect associated with the direct detection of the target substance in capsicum was significantly reduced in this study by the addition of isotopic internal standards corresponding to the target substance. This facilitated optimal conditions wherein quantitative analysis using LC-MS/MS revealed a linear range of 0.50–250.00 μg/mL, with all two curves calibrated with internal standards showing correlation coefficients (r2) greater than 0.9995. The method's limits of detection (LODs) and limits of quantification (LOQs) fell in the ranges of 0.14–0.45 μg/kg and 0.45–1.45 μg/kg, respectively. The method's spiked recoveries ranged from 81.97 to 105.17%, indicating its sensitivity and accuracy. The environmental friendliness of the technique was assessed using two green assessment tools, AGREE and complexGAPI, and the results showed that the technique was more in line with the concept of sustainable development compared to other techniques for detecting ochratoxins in capsicum. Overall, this study provides a new approach for the determination of mycotoxins in a complex food matrix such as capsicum and other spices using DES and also contributes to the application of green analytical chemistry methods in the food industry.

Non-psychoactive cannabinoids identification by linear retention index approach applied to a hand-portable capillary liquid chromatography platform

Abstract:The aim of the present research was the application of the linear retention index (LRI) system for the identification of non-psychoactive cannabinoids using a portable LC instrument. The miniaturization, viz. the use of very low quantities of mobile phase, enabled the development of a compact mobile system to be used for in situ analysis, also according to a green and cost-saving approach. In particular, new capillary LC (cap-LC) methods coupled with UV detection were developed for the analysis of extracts of Cannabis sativa L. Two setups were explored to achieve the efficient separation of twenty-four cannabinoids: a single column setup which exploited a sub-2 µm packing to increase the chromatographic resolution, and a dual-column setup based on the serial connection of two different stationary phases, each coupled to an UV detector. The latter allowed the determination of two LRI values for each analyte, thus increasing the identification power. Moreover, since two different wavelengths were used on the LED-based UV detectors, the ratio of the absorbances measured on each chromatographic trace represented a third identification criterion, thus fulfilling the recommendations of the Scientific Working Group for The Analysis of Seized Drugs (SWDRUG) about the categories of analytical techniques to be used and the minimum number of parameters required for the unambiguous identification of drugs. The obtained results could be used for the development of a novel analytical method for fast and automatic in situ forensic investigations and hemp breeding programs, also minimizing the consumption of both sample and solvent.
Quantification of Captopril using Ultra High Performance Liquid Chromatography

Abstract:We develop an efficient, sensitive, accurate, cost effective, and greener liquid chromatography method for the determination of a quantification method concerning Captopril in pharmaceutical dosage forms by using ultra high performance liquid chromatography (UHPLC) equipment. Satisfactory resolution was able to be achieved using a Water Aquity UHPLC BEH C18 1.7 $\mu$m $\times$ 2.1 mm $\times$ 50 mm column and an isocratic mobile phase composition of Methanol, Milli-Q water and Trifluoroacetic acid in the ratio of 55:45:0.05 v/v/v respectively at a flow rate of 0.1mL/min and a wavelength detection of 220nm. The retention time for Captopril was found to be 1.744 minutes while for Captopril disulfide it was found to be 2.657 minutes. The method was validated based on the ICH and USP guidelines.

Abstract:Phenolic acids are important active components of certain Traditional Chinese Medicines (TCM) and have a wide range of biological effects. Separation and purification of phenolic acids remains challenging due to difficulties with quality control using existing chromatographic methods The purpose of this study was to compare the effects of different chromatographic columns and conditions for the separation of phenolic acids. The BEH column was determined to be optimal, providing efficient separation in the shortest time (17.00 min) using gradient elution with carbon dioxide as the mobile phase, methanol/acetonitrile (70:30, v/v) with 1% TFA as the modifier, and a flow rate of 0.8 mL/min. Good peak shapes were obtained, and the peak asymmetry values were close to 1.00 for all phenolic acids. The resolution was more than 2.83 for all separated peaks. The developed method was subsequently applied to the determination of phenolic acids in Xanthii Fructus. These results are beneficial for quality control and standardization of herbal drugs using UPC2, providing an efficient, rapid and environmentally friendly scientific basis for future analysis of phenolic acids. Abstract:A fast analytical method by direct analysis in real time ionization coupled to mass spectrometer (DART-MS/MS) has been developed for the estimate the capsaicin (CAP) and dihydrocapsaicin (DHCP) concentration in fermented pepper paste. Firstly, these mass fragmentation schemes of CAP and DHCP by DART-MS/MS are proposed, and compared with those of electrospray ionization source (ESI). After conducting a series of optimizations, the samples were extracted by a simple procedure using acetonitrile, the samples extracts were then directly evaluated by DART-MS/MS without chromatographic separation. A limit of detection (LOD) of 0.0234 and 0.0510 μg/mL was achieved for CAP and DHCP, respectively. The results obtained with the newly developed DART-MS/MS method are in good agreement with those by the conventional ultra-high-performance liquid chromatography-electrospray ionization-mass spectrometry method. The results showed that DART-MS/MS is a high-throughput tool for determination of CAP and DHCP concentration in food products.

Abstract:Enantiomeric separation of chiral pharmaceuticals was carried out by means of in capillary liquid chromatography (CLC) and pressurized capillary electrochromatography (pCEC) using a vancomycin chiral stationary phase (CSP). A 100 microm I.D. fused-silica capillary was packed with 5 microm diameter silica particles modified with vancomycin. Enantiomeric resolution of fifteen beta3-receptor blockers and analogs was studied by polar organic CLC mode and reversed-phase pCEC mode using mobile phases containing methanol-isopropanol-acetic acid-triethylamine and TEAA buffer-methanol, respectively. Several factors affecting chiral separation were investigated in both CLC and pCEC mode. Good enantiomeric resolution was achieved by CLC mode for propranolol, celiprolol, esmolol, bisoprolol, atenolol, metoprolol and carteolol using methanol-isopropanol-acetic acid-triethylamine (70:30:0.05:0.05, v/v/v/v) as mobile phase and for clenbuterol, bambuterol, terbutaline, and salbutamol using methanol-isopropanol-acetic acid-triethylamine (50:50:0.05:005 or 50:50: 0.025:0.05, v/v/v/v) as mobile phase. The baseline was achieved by pCEC mode for the separation of esmolol, bisoprolol, atenolol, metoprolol, carteolol in the mobile phase containing MeOH-0.05%TEAA (pH 7.0) (90:10, v/v) (-10 kV), and that of propranolol and celiprolol in the mobile phase containing MeOH-0.025%TEAA (pH 7.0) (90:10, v/v)(-10 kV). Comparative enantioseparations performed in polar organic CLC and reversed phase pCEC mode revealed significant difference.
