Abstract:To study the effect of exogenous fecal microbiota intervention on intestinal barrier function in recipient pigs,twelue litters of "Duroc×Landrace×Yorkshire" suckling piglets were randomly allocated into two groups on the basis of similar body weight,birth time and parity.Each group had six litters (10～12 piglets/litter).All piglets were infused by gavage with exogenous fecal suspension or same amount of sterile phosphate buffered solution (PBS) every other day from the first day after birth to 10 days of age.All piglets in each litter had free access to sow and water during the experiment.Piglets were killed and samples were collected on 12 days of age and 27 days of age.The results indicated that:Exogenous fecal microbiota intervention significantly decreased the incidence of diarrhea and increased the average daily gain and survival rate in suckling piglets (P＜0.05).Exogenous fecal microbiota intervention significantly decreased crypt depth in the intestine of suckling piglets at 27 days of age (P＜0.05) and increased villus height/crypt depth in jejunum and ileum at 12 and 27 days of age (P＜0.05).Exogenous fecal microbiota intervention significantly increased the number of goblet cells and SIgA+ cells (12 days of age) and improved the relative expression of Muc2 and tight junction protein ZO-1 and Occludin in the intestine (P＜0.05).While the number of SIgA+ cells had no significant difference between the treatment group and the control group at 27 days of age (P＞0.05).It is suggested that exogenous fecal microbiota intervention can increase the number of goblet cells and SIgA+ cells,improve the expression of Muc2 and tight junction protein and regulate intestinal barrier function in recipient pigs.Exogenous fecal microbiota intervention has the prospect of maintaining intestinal physiology homeostasis and promoting intestinal health in pigs.
Clostridium Butyricum Relieves Diarrhea by Enhancing Digestive Function, Maintaining Intestinal Barrier Integrity, and Relieving Intestinal Inflammation in Weaned Piglets

Abstract:The current study aimed to determine how Clostridium butyricum (C. butyricum) relieved diarrhea by evaluating its effects on intestinal digestive enzyme activity, barrier integrity, inflammatory responses, and oxidative stress in weaned piglets. A total of 360 weaned piglets that were 25 days old (180 males and 180 females) were randomly assigned to 5 groups in 6 pens, with 12 pigs per pen, and offered the same antibiotic-free cornsoybean-type basal diet for 30 d. The groups were as follows: no additives (control), basal diet + 250 mg/kg C. butyricum (CB1), basal diet + 500 mg/kg C. butyricum (CB2), basal diet + 1,000 mg/kg C. butyricum (CB3), and basal diet + 2,000 mg/kg C. butyricum (CB4). The incidence of diarrhea decreased quadratically as the amount of C. butyricum supplementation was increased, and this decrease was minimized in the CB2 group. Compared with those in the control group, the duodenal amylase and lipase activities were increased quadratically and maximized in the CB2 group, and the jejunal lipase activity was increased cubically in the CB1 group, while the pancreatic trypsin activity was increased linearly and maximized in the CB4 group. The serum concentration of D-lactic acid responded to the increases in the levels of C. butyricum in a quadratic manner; the minimum level was observed in the CB2 group. Jejunal zonula occludens-1 expression displayed linear and quadratic trends following C. butyricum supplementation, with the maximum response observed in the CB3 group. Quadratic and cubic increases were observed in jejunal alkaline phosphatase activity in the CB3 group. The jejunal IL-10 concentration increased, while the jejunal vascular endothelial growth factor concentration decreased quadratically in response to dietary C. butyricum supplementation in the CB2 group. No significant differences among the groups were observed in terms of the jejunal mucosal concentrations of oxidation-associated enzymes and products. In conclusion, dietary supplementation with appropriate doses of C. butyricum could relieve diarrhea in weaned piglets by enhancing digestive enzyme activity, maintaining intestinal barrier integrity, and relieving intestinal inflammation.

Abstract:Objectives Butyrate, a volatile fatty acid (VFA) produced by gut bacteria, regulates gut barrier function and immune response. However, the incorporation of butyrate into foods is challenging. Tributyrin (TB), a triglyceride with 3 butyrate molecules, represents an alternative to butyrate. Herein, the effects of encapsulated TB with gamma-cyclodextrin (ɣ-CD/TB) on gut microbiota composition and VFA concentrations were assessed in dextran sulfate sodium (DSS)-induced colitis piglets. Methods Two-day-old piglets (n = 59) were randomized to three diets: control (CON) [Advance Baby Pig Liqui-Wean], CD [CON + 9.0 mM ɣ-CD], or CDTB [CON + 9.0 mM ɣ-CD/TB]. Half of the piglets in each group received DSS (1.25 g/kg body weight) from postnatal day (PND) 14 to 18. Ileal (IL), ascending colon (AC) and rectal (RC) contents were collected at PND 27. Microbiota were assessed by 16S rRNA gene sequencing and VFA were measured by gas chromatography. Results Permutational multivariate analysis of variance showed diet affected the beta diversity in IL (P = 0.045) with CON differing from CD or CDTB. Relative abundances of ileal Weissellaand Actinobacillus were higher and Turicibacter were lower in CON than CD or CDTB piglets (P < 0.05). While diet did not affect overall bacterial composition in AC and RC, it impacted proportions of some bacterial genera. Bacteroides was lower, Holdemanella and Ruminococcus were higher in AC of CON than CDTB. Rectal Bacteroides were lower and UCG-010 were higher in CON compared to CDTB (P < 0.05). Colitis influenced beta diversity in AC and RC (P = 0.002). Compared to piglets without colitis, proportions of 10 bacterial genera were lower and 6 were higher in AC, while 9 were lower and 11 were higher in RC of colitis piglets (P < 0.05). Diet impacted alpha diversity in IL and AC with observed features in IL, Shannon and evenness indices and phylogenetic diversity (Faith PD) in AC being higher in CON than CDTB (P < 0.05). Colitis increased Faith PD in RC (P = 0.004). For VFA, colitis decreased isobutyrate and isovalerate in IL and increased acetate, propionate, butyrate and valerate in RC, regardless of diet. Conclusions Encapsulated ɣ-CD/TB modulates the bacterial community along the piglet intestinal tract, while colitis influences both the microbiota composition and VFA concentrations. Abstract:Oral tolerance has previously been shown effective in preventing several immune-mediated disorders in animal models. The aims of this study were to investigate the effect of oral colitis-extracted proteins (CEP) on dextran sulfate sodium (DSS)-induced colitis in BALB/c mice and to explore the relative role of the intestinal mucosal gamma delta T cells.The effect of five low oral doses of CEP on colitis was evaluated by clinical manifestation and histological lesions. Serum cytokines were measured by enzyme-linked immunosorbent assay. The percentages of the intestinal mucosal gamma delta T cells were evaluated by flow cytometry.CEP-fed colitis mice showed less severe symptoms and histological injury than bovine serum albumin (BSA)-fed control mice. Tolerized mice developed an increase in TGF-beta 1 and no change in IFN-gamma serum levels. Increases in TCR gamma delta(+) T cells and CD8 alpha+TCR gamma delta(+) T cells in small intestinal mucosal lymphocytes and no quantitative change in large intestinal mucosal lymphocytes were demonstrated in colitis mice compared to untreated mice. The proportions of TCR gamma delta(+) T cells and CD8 alpha+TCR gamma delta(+) T cells in large intestinal mucosal lymphocytes from CEP-fed colitis mice were significantly higher compared to BSA-fed controls. The disease activity index negatively correlated with the percentages of large intestinal mucosal gamma delta T cells. Furthermore, mucosal repair in repair-period mice was also accompanied by increases in TCR gamma delta(+) T cells and CD8 alpha+TCR gamma delta(+) T cells in large intestinal mucosal lymphocytes.Improvement of DSS-induced colitis that resulted from oral administration of colitis-extracted proteins is associated with an increase in gamma delta T cells in large intestinal mucosa.

Abstract:Despite the marked changes that crustacean muscle undergoes during the molt cycle, pyruvate kinase is present as the same form throughout the molt cycle. This pyruvate kinase was subject to feed-forward activation by fructose-1, 6 bisphosphate (FBP) as well as feed-back inhibition by MgATP. The enzyme showed a high affinity for phosphoenolpyruvate (Km = 0.1 mM) but showed no cooperativity in substrate binding. The addition of 0.05 mM FBP reduced the PEP Km to 0.05 mM. MgATP inhibition showed a Ki of 1.8 mM versus PEP. The inhibition due to MgATP could be reversed by FBP. Various other compounds inhibited the enzyme, including citrate, alpha-ketoglutarate, tryptophan, and malate, although at rather high levels. Measurements of the reversal of this pyruvate kinase, taken together with the low levels of phosphoenolpyruvate carboxykinase and pyruvate carboxylase, predict only minimal levels of gluconeogenic flux in crustacean muscle.
Effects of Tributyrin on Growth Performance, Intestinal Microflora and Barrier Function of Weaned Pigs

Abstract:This study evaluated the effects of tributyrin supplementation on the growth performance, intestinal microflora and barrier function of weaned pigs. A total of 144 piglets (Duroc x (Landrace x Yorkshire), average initial weight of 6.82 kg weaned at 24 +/- 1 day) were allotted to 4 treatments for 2 weeks, each of which had 6 pens with 6 pigs per pen (3 barrows and 3 gilts). The four treatments were the control (basal diet), and the basal diet supplemented with 250, 500 and 750 mg/kg tributyrin, respectively. After the feeding experiment, 6 pigs per treatment were slaughtered to investigate the effects of tributyrin on intestinal microflora and barrier function. The results showed that tributyrin supplementation in increasing concentrations increased (P < 0.05) average daily gain (ADG), reduced (P < 0.05) the fecal scores, improved (P < 0.05) intestinal morphology, as indicated by greater villus height and villus height: crypt depth, and intestinal barrier function reflected by increased transepithelial electrical resistance (TER) and decreased paracellular flux of dextran (FD4). Moreover, the 16s rRNA results showed that incremental levels of tributyrin increased (P < 0.05) the ratio of Lactobacilli to Escherichia coli (E. coli) in the jejunum and colon of weaned piglet. The results indicated that incremental levels of tributyrin increased average daily gain and decreased fecal scores of weaned pigs. The improvements in growth performance may result from the improved the intestinal microflora, morphology and barrier function. Thus, dietary tributyrin offers a promising approach to alleviative post-weaning intestinal tract disorders in pigs.
Changes of CD8⁺ T Cells in Dextran Sulfate Sodium-Induced Colitis Mice Pretreated with Oral Immune Regulation.

Abstract:Background 11 has been reported that CD8(+) regulatory cells could be induced upon oral tolerance. The purpose of this study was to investigate the changes of CD8 alpha(+) T cells in dextran sulfate sodium (DSS)-induced colitis mice pretreated by oral immune regulation.Methods The effects of five low oral doses of colitis-extracted proteins (CEP) on colitis were evaluated by clinical manifestation and histological lesions. The percentages of CD8 alpha(+) T cells gating on CD3(+) T cells were evaluated in the gut-associated lymphoid tissues (GALT) and the spleens by flow cytometry. Differences between the two groups were compared by Student's t test or Mann-Whitney U test.Results Compared to bovine serum albumin (BSA)-fed control mice, administration of CEP resulted in marked alleviation of colitis. The proportion of CD8 alpha(+) T cells, not only in intraepithelial lymphocytes (IELs) and lamina propria lymphocytes (LPLs) of the large intestine (LI) but also in spleen from CEP-fed colitis mice, was significantly higher than that from BSA-fed colitis mice (LI-IELs: (71.5 +/- 5.4)% vs. (60.1 +/- 4.3)%, P<0.01; LI-LPLs: (60.7 +/- 5.2)% vs. (51.9 +/- 4.7)%, P <0.01; spleen: (24.1 +/- 3.6)% vs. (20.3 +/- 4.1)%, P <0.05; n=8). Mucosal repair in repair-period mice five days after termination of DSS treatment was also accompanied by an increase of CD8 alpha(+) T cells in large intestinal mucosal lymphocytes (LI-IELs: (72.1 3.7)% vs. (61.5 4.5)%, P<0.01; LI-LPLs: (62.1 +/- 5.7)% vs. (52.7 +/- 3.6)%, P<0.01; n=8). The proportion of CD3(+) T cells increased in Peyer's patches (PPs) and decreased in mesenteric lymph nodes (MLNs) from colitis mice compared to untreated mice, whereas the change pattern of CD3(+) T cells in PPs and MLNs from CEP-fed colitis mice was just on the contrary.Conclusion Improvement of DSS-induced colitis resulted from oral immune regulation is associated with an increase in CD8 alpha(+) T cells in spleen and large intestinal mucosa. Long-term Outcomes of Infliximab Use for Pediatric Crohn Disease: A Canadian Multicenter Clinical Practice Experience

Abstract:Background: Data on long-term real-world outcomes of infliximab in pediatric Crohn disease are limited. Aim: The aim of the study was to evaluate infliximab optimization and durability in children with Crohn disease. Methods: We performed a retrospective review of children with Crohn disease who started infliximab from January 2008 to December 2012 in 4 Canadian tertiary care centers. A priori factors associated with optimization and discontinuation from loss of response were evaluated using logistic regression and Cox proportional hazards model, respectively. Results: One hundred eighty children (54.4% boys) started infliximab; all completed induction. Median age at infliximab start was 14.3 years (Q1, Q3: 12.8, 15.9 years) and median time from diagnosis to infliximab start was 1.5 years (Q1, Q3: 0.6, 3.5 years). At last follow-up, 87.1% were maintained on infliximab (median duration follow-up 85.9 weeks [Q1, Q3: 43.8, 138.8 weeks]). Infliximab optimization occurred in 57.3% (dose escalation 15.2%, interval shortening 3.9%, both 38.2%), primarily due to loss of response. Younger age at diagnosis (<10 years old) and nonstricturing, nonpenetrating behavior were associated with optimization (odds ratio 6.5, 95% confidence interval [CI] 2.0–21.1 and odds ratio 2.1, 95% CI 1.0–4.2, respectively). The 1- and 2-year durability of infliximab (percentage in follow-up who were continuing on infliximab) were 95.5% (95% CI 90.4–98.3) and 91.0% (95% CI 82.4–96.3), respectively. Annual discontinuation due to loss of response occurred at 3.2% per year (95% CI 1.1–5.2). Conclusions: Children with Crohn disease maintain a durable response to infliximab. Optimization occurs frequently and allows for continued use. Younger age at diagnosis and nonstricturing, nonpenetrating behavior are associated with increased need for infliximab optimization.

Abstract:Backgrounds and aims There is increasing evidence that gut-derived intraepithelial lymphocytes have potent cytolytic and immunoregulatory functions, which they use to sustain epithelial integrity. The aims of this study were to investigate the roles of small intestinal intraepithelial lymphocytes (SI-IELs) in oral tolerance and dextran sodium sulfate (DSS)-induced colitis. Methods SI-IELs or sorted γδ T cells from untreated, colitis, and colitis-extracted protein (CEP)-fed colitis mice were adoptively transferred to BALB/c mice; colitis was then induced with DSS. Cytokines were analyzed in sera from mice and culture supernatants. Results Transfer of SI-IELs or sorted γδ T cells from untreated and colitis mice all alleviated experimental colitis. Mice orally administered with five low doses of CEP showed less severe symptoms and histological injury. SI-IELs from CEP-fed colitis mice more significantly ameliorated colitis than those from control mice (weight, 94.1 ± 2.5% vs. 89.8 ± 2.6%, p < 0.05; disease activity index, 7.2 ± 1.2 vs. 8.7 ± 1.9, p < 0.05; histological scores, 22.1 ± 2.8 vs. 25.7 ± 2.1, p < 0.05, n = 8 per group); however, not did SI-γδ IELs from CEP-fed colitis mice. Alleviation of colitis was accompanied by an increase of TGF-β1 secretion and no change of IFN-γ in sera and culture supernatants. The level of serum TGF-β1 was negatively related to the severity of colitis. Conclusions The protective effects of SI-IELs in DSS-induced colitis were partly accomplished by γδ T cells and could be mediated by TGF-β but were not associated with IFN-γ. Oral tolerance strengthens the suppressive effects of regulatory subsets in SI-IELs.

Abstract:ObjectiveTo evaluate the state of oral tolerance and its therapeutic role in mice with dextran sulfate sodium (DSS)‐induced colitis.MethodsDelayed‐type hypersensitivity (DTH) was determined 7 and 14 days after DSS‐induced colitis and control mice. Disease activity index (DAI) score and colonic histopathological score were measured 7 days after colonic extracted protein (CEP) or bovine serum albumin (BSA) (control) was administrated, with the evaluation of Th1–Th2 balance in the spleen, Peyer's patch and γδ T cells in intraepithelial lymphocytes and lamina proper lymphocytes in the intestine.ResultsAfter fed with 250 μg ovalbumin oral tolerance was induced in 7 days in both DSS‐induced colitis and control mice, while oral tolerance persisted in the control mice but vanished in DSS‐induced colitis 14 days after ovalbumin challenge. DAI and colonic histopathological scores were decreased significantly after the ingestion of CEP (controlled by BSA) in DSS‐induced colitis with significant reduction of Th1 and the ratio of Th1 to Th2 in Peyer's patch as well as the γδ T cells in lamina proper lymphocytes in the intestine. No significant difference in Th1–Th2 balance in the spleen and γδ T cells in intraepithelial lymphocytes in the intestine were observed.ConclusionsThere is a defect in oral tolerance at day 7 in DSS‐induced colitis. If taken orally, CEP may have a protective role in DSS‐induced colitis, which may be related to the deflection from Th1 to Th2 in Peyer's patch and the reduction of γδ T cells in lamina proper lymphocytes in the intestine.
Supplementation of tributyrin improves the growth and intestinal digestive and barrier functions in intrauterine growth-restricted piglets

Abstract:Background & aims: Intrauterine growth restriction (IUGR) neonates suffer from growth restriction. Tributyrin (TB), a pro-drug of butyrate, can facilitate the growth of animals. This study was to investigate the effects of TB supplementation on the growth of IUGR neonatal piglets. Methods: Sixteen IUGR and 8 NBW (normal body weight) neonatal piglets were chosen, weaned at 7th day and fed basic milk diets (NBW and IUGR group) or the basic diets supplemented with 0.1% tributyrin (IT group, IUGR piglets fed with tributyrin) until day 21 (n = 8). The body weights of the piglets on days 0, 7, 10, 14, 17, and 20 were measured. The digestive enzyme activity, intestinal morphology, immunoglobulin levels and gene expression of IgG, FcRn and GPR41 in the small intestines were analyzed. Results: The body weights of the piglets in the IUGR and IT group were similar, and both were lower than the NBW group on days 10 and 14. However, after day 17, the IT group exhibited improved (P < 0.05) body weights compared to that of the IUGR group. The piglets were sacrificed on day 21. Compared with the NBW piglets, IUGR impaired the development of immune organs and small intestines, impaired the intestinal villus morphology, decreased (P < 0.05) most of the tested intestinal digestive enzyme activities, decreased (P < 0.05) the ileal sIgA and IgG levels, and down-regulated (P < 0.05) the intestinal IgG and GPR41 expression. Piglets in the IT group exhibited a better-developed (P < 0.05) spleen and small intestines, improved intestinal villus morphology, increased (P < 0.05) intestinal villus surface areas, enhanced (P < 0.05) digestive enzyme activities, and up-regulated (P < 0.05) expression of IgG and GPR41 mRNA compared to those of the IUGR group. Conclusions: TB supplementation improves the growth and the intestinal digestive and barrier functions in IUGR piglets during the suckling period.
Banxia Xiexin Decoction Protects Against Dextran Sulfate Sodium-Induced Chronic Ulcerative Colitis in Mice.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE:Banxia Xiexin decoction (BXD), one of a traditional Chinese medicine chronicled in Shang Han Lun, is commonly used to treat gastroenteritis, ulcerative colitis and diarrhea. In our study, we used current biomedical approaches to investigate the therapeutic efficacy of BXD and possible protective mechanism involved in inhibiting dextran sulfate sodium (DSS)-induced chronic ulcerative colitis model.MATERIALS AND METHODS:Chronic DSS colitis was induced in C57BL/6 male mice by three cycles of 5 days of 2% DSS in drinking water, alternating with 5 days of normal water, totaling 30 days. In BXD group, the mice were administered at a dose of 8.7g/kg BXD for 5 days before and during DSS treatment via oral gavage per day. Mice in vehicle group and DSS group were given orally the same volume of drinking water, instead. Body weight, stool characters and hematochezia were observed everyday. The colorectal tissues were used to detect levels of TNF-α, IL-4, IL-10, IL-1β, IL-17, IL-23 and MPO by ELISA or qRT-PCR. The expression of COX-2, 8-Oxoguanine and Nrf2 were examined by IHC, and p-p65 was examined by western blotting. ThOD and the content of MDA were measured according to kits respectively.RESULTS:BXD significantly protected against DSS-induced chronic ulcerative colitis by amelioration of body weight loss, DAI and histology score. The level of TNF-α, IL-1β, IL-17, IL-23, COX-2 and p-p65 were decreased significantly, while the level of IL-10 improved with the treatment of BXD. MDA, MPO and 8-Oxoguanine were decreased, meanwhile SOD activity and Nrf2 expression were elevated significantly by BXD.CONCLUSIONS:BXD possesses the potential of anti-inflammation and anti-oxidation to treat colitis. The protective mechanism of BXD may involve in inhibition of NF-κBp65 activation and increasement of Nrf2 expression in colorectums of mice.

Abstract:Background: Tauroursodeoxycholic acid (TUDCA), a hydrophilic bile acid, is the main medicinal component of bear bile and is commonly used to treat a variety of hepatobiliary diseases. Meanwhile, TUDCA has been shown to modulate the intestinal barrier function and alleviate DSS-induced colitis in mice. However, the effect of TUDCA on the intestinal barrier of weaned piglets remains largely unclear. Methods: The weaned piglets and porcine IPEC-J2 intestinal epithelial cells were used to investigate the effects of TUDCA on intestinal barrier function in weaned piglets and explore the possible underlying mechanisms. In vivo, 72 healthy weaned piglets were randomly allocated into 2 groups according to their gender and body weight, and piglets were fed the basal diet with 0 (control, CON) and 200 mg/kg TUDCA for 30 d, respectively. Three female and three male piglets reflecting the average bodyweight were slaughtered in each group and samples were collected. In vitro, IPEC-J2 cells were subjected to 100 μmol/L TUDCA to explore the possible underlying mechanisms. Results: Our results demonstrated that dietary TUDCA supplementation significantly reduced the diarrhea incidence of weaned piglets, possibly attributing to the TUDCA-enhanced intestinal barrier function and immunity. In addition, TUDCA supplementation altered serum metabolites and the relative abundance of certain gut bacteria, which might contribute to the improved intestinal barrier function. Furthermore, the in-vitro results showed that TUDCA improved the E. coli-induced epithelial barrier impairment of IPEC-J2 cells and increased Takeda G-coupled protein receptor 5 (TGR5) protein expression. However, knockdown of TGR5 and inhibition of myosin light chain kinase (MLCK) pathway abolished the TUDCA-improved epithelial barrier impairment in E. coli-treated IPEC-J2 cells, indicating the involvement of TGR5-MLCK in this process. Conclusions: These findings showed that TUDCA improved intestinal barrier function associated with TGR5-MLCK pathway and the alteration of serum metabolites and gut bacteria in weaned piglets, suggesting the potential application of TUDCA in improving gut health in piglet production.

Abstract:Background: Whether the probiotic Clostridium butyricum (CB) alleviates enterotoxigenic Escherichia coli (ETEC) K88-induced inflammation by regulating the activation of the toll-like receptor (TLR) signaling pathway is not clear, thus, we carried out this study. A total of 72 piglets (average body weight 7.09 ± 0.2 kg) were randomly divided into three groups of 24 piglets per group. Pigs were either fed a daily diet (NC, negative control), a diet tested every day by 1 × 109 CFU/mL ETEC K88 (PC, positive control), or a basal diet supplemented with 5 × 105 CFU/g CB and challenged with ETEC K88 (PC + CB group). Results: Our results showed that CB pretreatment attenuated the effect of ETEC K88 by decreasing C-reactive protein (CRP), which resulted in tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) production. Histological examination revealed that CB pretreatment alleviated intestinal villi injury caused by ETEC K88 challenge. Furthermore, CB pretreatment promoted mRNA expression of the negative regulators of TLR signaling, including myeloid differentiation factor (MyD88), toll-interacting protein (Tollip), and B cell CLL/lymphoma 3 (Bcl-3), in the intestines of ETEC K88-challenged piglets. ETEC K88-induced activation of nuclear factor kappa B (NF-κB) and nuclear factor of kappa light polypeptide gene enhancer in B cells inhibitor alpha (IκBα) was attenuated by CB pretreatment. Conclusion: These findings indicate that CB helps to maintain and strengthen the shape of intestinal villi and limits detrimental inflammatory responses, partly by inhibiting toll-like receptor 2 (TLR-2), toll-like receptor 4 (TLR-4), and toll-like receptor 5 (TLR-5) expression and inhibiting NF-κB p65, and promoting IκBα activation and synergism among its negative regulators.

Abstract:INTRODUCTION AND OBJECTIVE Acute disseminated encephalomyelitis (ADEM) is an inflammatory demyelinating disease of the central nervous system. This study is based on clinical symptoms and diagnostic tests employed. PATIENTS AND METHODS We describe a seven children series indicating the initial neurologic abnormalities, diagnostic tests, treatments used and clinical-neuroradiological evolution. RESULTS The mean presentation age was 4.1 years. Initial neurologic symptoms were mainly spastic hemi/paraparesis, cerebellous and pyramidal syndrome, consciousness changes, meningeal signs, seizures and cranial nerve palsies. The cerebrospinal fluid was abnormal in four patients with positive serologic tests in two of them (Coxsackie B). Electrophysiological studies were affected in 50%. MRI findings consisted of multifocal supratentorial white matter lesions. Clinical evolution revealed a progressive improvement with resolution after two months. Follow-up was made between six months and five years. The treatment was based on aciclovir and corticosteroids. CONCLUSIONS ADEM runs a monophasic course of progressive neurologic abnormalities. Diagnosis is based on suggestive clinical and neuroimaging findings. Generally speaking, MRI showed resolution of multifocal lesions in conjunction with clinical improvement.
Effects of dietary tributyrin on intestinal mucosa development, mitochondrial function and AMPK-mTOR pathway in weaned pigs

Abstract:Background: The objective of this experiment was to investigate the influence of dietary tributyrin on intestinal mucosa development, oxidative stress, mitochondrial function and AMPK-mTOR signaling pathway. Methods: Seventy-two pigs were divided into two treatments and received either a basal diet or the same diet supplemented with 750 mg/kg tributyrin. Each treatment has six replicates of six pigs. After 14 days, 6 pigs from each treatment were selected and the jejunal samples were collected. Results: Results showed that supplemental tributyrin increased (P < 0.05) villus height and villus height: crypt depth of weaned pigs. Pigs fed tributyrin had greater (P < 0.05) RNA/DNA and protein/DNA ratios than pigs on the control group. The mRNA levels of sodium glucose transport protein-1 and glucose transporter-2 in the jejunum were upregulated (P < 0.05) in pigs fed the tributyrin diet. Dietary tributyrin supplementation lowered (P < 0.05) the malondialdehyde and hydrogen peroxide (H2O2) content in jejunum, enhanced (P < 0.05) the mitochondrial function, as demonstrated by decreased (P < 0.05) reactive oxygen species level and increased (P < 0.05) mitochondrial membrane potential. Furthermore, tributyrin increased (P < 0.05) mitochondrial DNA content and the mRNA abundance of genes related to mitochondrial functions, including peroxisomal proliferator-activated receptor-γ coactivator-1α, mitochondrial transcription factor A, nuclear respiratory factor-1 in the jejunum. Supplementation with tributyrin elevated (P < 0.05) the phosphorylation level of AMPK and inhibited (P < 0.05) the phosphorylation level of mTOR in jejunum compared with the control group. Conclusions: These findings suggest that dietary supplementation with tributyrin promotes intestinal mucosa growth, extenuates oxidative stress, improves mitochondrial function and modulates the AMPK-mTOR signal pathway of weaned pigs.
Alleviation Effects of Bifidobacterium animalis subsp. lactis XLTG11 on Dextran Sulfate Sodium-Induced Colitis in Mice

Abstract:Inflammatory bowel disease (IBD) is a chronic immune-related disease, which can occur through the dysfunction of the immune system caused by the imbalance of gut microbiota. Previous studies have reported the beneficial effects of Bifidobacterium on colitis, while the related mechanisms behind these effects have not been fully elucidated. The aim of our study is to investigate the alleviation effect of Bifidobacterium animalis subsp. lactis XLTG11 (B. lactis) on dextran sulfate sodium (DSS)-induced colitis and its potential mechanism. The results showed that B. lactis XLTG11 significantly decreased weight loss, disease activity index score, colon shortening, myeloperoxide activity, spleen weight, and colon tissue damage. Additionally, B. lactis XLTG11 significantly decreased the levels of pro-inflammatory cytokines and increased the level of anti-inflammatory cytokine. Meanwhile, high doses of B. lactis XLTG11 significantly up-regulated the expression of tight junction proteins and inhibited activation of Toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MYD88)/nuclear factor-κB (NF-κB) signaling pathway. Furthermore, B. lactis XLTG11 increased the gut microbiota diversity and modulated gut microbiota composition caused by DSS. Moreover, Spearman’s correlation analysis also found that several specific gut microbiota were significantly correlated with colitis-related indicators. These results demonstrated that B. lactis XLTG11 can alleviate DSS-induced colitis by inhibiting the activation of the TLR4/MYD88/NF-κB signaling pathway, regulating inflammatory cytokines, improving intestinal barrier function, and modulating the gut microbiota.

Abstract:Background: Inflammatory bowel disease (IBD) is typically managed using medications such as 5-aminosalicylic acid (5-ASA), glucocorticoids, anti-TNFα Ab, or anti-IL-12/23 Ab. However, some patients do not respond well to these treatments or frequently experience relapses. Therefore, alternative therapeutic options are needed. Since the activation of the inflammasome is crucial to the pathogenesis of IBD, inhibiting the inflammasome may be beneficial for patients. Materials and methods: We tested the efficacy of taurodeoxycholate (TDCA), which is a known G-protein coupled receptor 19 (GPCR19) agonist, in a mouse colitis model induced by dextran sodium sulfate (DSS). Results: In the mouse colitis model, TDCA prevented loss of body weight, shortening of the colon, production of pro-inflammatory cytokines, infiltration of pro-inflammatory cells, and mucosal ulceration in the colon. In vitro, TDCA inhibited the activation of NF-κB in bone marrow-derived macrophages (BMDMs) by activating the cAMP-PKA axis. TDCA downregulated the expression of purinergic receptor P2X7 (P2X7R) and enhanced the colocalization of P2X7R with GPCR19, and inhibited the Ca2+ mobilization of BMDMs when stimulated with ATP or BzATP, which plays a pivotal role in activating the NLRP3 inflammasome (N3I) via P2X7R. TDCA inhibited the oligomerization of NLRP3-ASC and downregulated the expression of NLRP3 and ASC, as well as suppressed the maturation of pro-caspase-1 and pro-IL-1β. TDCA also increased the percentage of M2 macrophages while decreasing the number of M1 macrophages, Th1, Th2, and Th17 cells in the colon. Conclusion: TDCA ameliorated DSS-induced colitis in mice, possibly by inhibiting both the priming phase (via the GPCR19-cAMP-PKA-NF-κB axis) and the activation phase (via the GPCR19-P2X7R-NLRP3-Caspase 1-IL-1β axis) of N3I signaling.
