Abstract:Truffles, belonging to the genus Tuber , are ectomycorrhizal (ECM) fungi that form underground ascocarps and primarily establish symbiosis with oaks and hazels. The cultivation of Tuber spp. involves transplanting inoculated seedlings that have formed ectomycorrhiza with Tuber species, with mulching being effective for truffle cultivation. In this study, we investigated the effects of mulching on the mycelial growth of four Tuber species (T. himalayense, T. koreanum, T. melanosporum, and T. borchii ) in the Korean natural environment, highlighting the potential for Korea as a truffle cultivation site. We developed and tested species-specific primers for quantifying the soil mycelial biomass of Tuber spp. by qRT-PCR, determined the superior mulch color for mycelial growth, and identified the Tuber species exhibiting the highest growth rate in the Korean field environment. Our results demonstrated that white mulch significantly enhanced mycelial growth in Tuber species than black mulch, likely owing to its ability to maintain low soil temperatures, control weeds, and improve host plant growth. Among the Tuber species, T. himalayense showed the greatest growth potential in the Korean natural environment. Additionally, a significant and positive correlation was observed between the mycelial biomass of Tuber species and the growth of inoculated seedlings, as measured by the total stem length and the number of leaves, thereby indicating the importance of symbiosis between ECM fungi and host plants. This study provides valuable insights into truffle cultivation in Korea and highlights the potential of using white mulch to promote mycelial growth, thereby contributing essential data for understanding the appropriate environmental conditions for Tuber spp. cultivation in Korea. Further study is needed to assess the long-term impact of mulching and to explore the effectiveness of other mulching materials.

Abstract:Penicillium marneffei is an important thermal dimorphic fungus that is endemic to Southeast Asia and China and causes penicilliosis, an AIDS-defining disease. Dimorphic switching is considered an important growth characteristic associated with its pathogenicity. In recent years, the molecular mechanisms underlying both dimorphic switching and monomorphic growth have been studied. However, little is known about the physical and chemical factors that impact either dimorphic switching or monomorphic growth of this organism. Further, the natural history of the disease is unclear. Our experiments focus upon the effects of temperature, pH and salinity on both growth phases of P. marneffei. We compared 11 isolates of P. marneffei and found that all could grow at a wide temperature range (8.0-39.8 degrees C), but growth was dramatically inhibited at 40 degrees C. The morphological switch from hyphae to yeast growth was initiated at 32 degrees C. However, the sensitivity to elevated temperatures during this transition varied among isolates. Both hyphae and yeast growth forms grew much better at acidic (pH 5, 6) and neutral pH than at alkaline conditions. While similar sensitivities were observed at high concentrations of NaCl and CaCl(2), in general, yeast cells displayed a greater sensitivity to both compounds. Our data demonstrate that isolate differences occur in growth patterns. Importantly, the growth requirements defined in our study may shed light on the environmental conditions that favor its survival, a subject that is not completely resolved in the current literature.

Abstract:A white-colored truffle Tuber japonicum, indigenous to Japan, is an ascomycetous ectomycorrhizal fungus. To clarify the physiological characteristics of this fungus, we investigated the influence of culture medium, temperature, and sources of nitrogen (N) and carbon (C) on the growth of five strains. Tuber japonicum strains grew better on malt extract and modified Melin-Norkrans medium, and showed peak growth at 20 °C or 25 °C. This fungus utilized inorganic (NH4 + and NO3 -) and organic N sources (casamino acids, glutamine, peptone, urea, and yeast extract). Additionally, this fungus utilized various C sources, such as monosaccharide (arabinose, fructose, galactose, glucose, and mannose), disaccharide (maltose, sucrose, and trehalose), polysaccharide (dextrin and soluble starch), and sugar alcohol (mannitol). However, nutrient sources that promote growth and their effects on growth promotion widely varied among strains. This can result from the strain difference in enzyme activities involved in the assimilation and metabolism of these sources.

Abstract:The vegetative propagation of ectomycorrhizal truffle fungi is limited by their slow mycelial growth. Many factors including media, isolate genotypes and environmental conditions can alter fungal mycelial growth rates. This study aimed to improve the in vitro growth rate of Tuber brumale by determining the optimal carbohydrate and nitrogen sources, temperature and pH. After 8 weeks, the highest level of growth and densest hyphal branching were recorded in the medium containing glucose as the main carbohydrate. For nitrogen, glutamine (200 mg N l-1) provided the greatest hyphal growth and density compared to the other amino acid treatments. Regarding temperature, 16?C proved to be optimal for T. brumale growth and branching. Media of pH 6 and pH 7 were most favourable for the growth of T. brumale. The results from this research provide baseline data on the vegetative nutrition of T. brumale and have implications for the in vitro culture of winter truffle hyphae.

Abstract:The conidia germination and appressoria formation of Colletotrichum gloeosporioides, the pathogen of persimmon anthracnose, was tested on concave glass slides in different environment. As the concentration of glucose went up, rates of conidia germinating increased but the percentage of appressoria formation decreased, and the length of germ tubes augmented and the diameter of appressoria was steady; with time prolonging, the percentage of conidia germinating and appressoria formation all climbed. The conidia could germinate and form appressoria over wide pH range from 2. 0 to 9. 0; the optimal pH for conidia germination and appressoria formation was between pH 5. 0 and pH 6. 0. The pathogenicity test under different pH condition illuminated that the disease occurred on twigs at 23℃ within pH 4. 0 - 8. 0, also appeared at 17℃ and pH 6. 0 without spore masses on lesions of twigs surface, however, was absent at 17℃ and pH 5. 0 as well as at 15℃ and pH 5. 0 to pH 6. 0. The optimal temperature for the mycelium growth was about 25℃ , and higher temperature inhibited its growth. The scanning electron microscopy examination showed that the length of germ tube, growing longitudinally along the ridge and transversely cross the ridges and grooves, varied greatly, and the appressoria formation occurred at the bottom of groove or near.

Abstract:A new kind of endogenetic fungus,found in the corms of Crocus sativus L.,was identified to be Alternaria alternata(Fr.: Fr.) Keissler based on its morphological and biological characteristics.The fungus would cause the corm rotten under certain conditions,and the symptom would be more serious when it incorporates with penicillium.The effect of environmental and nutritional factors on its growth and reproduction was remarkable.ⅰ) The temperature from 10 to 35 ℃ were is suitable for mycelial growth,sporogenesis and spore germination.However,the optimum temperature is 28 ℃.For spores,the lethal temperature is 65 ℃ lasting 10 minutes.ⅱ) When pH ranges 4.0-7.0 in the media,it is favourable for mycelial growth,sporogenesis and spore germination.However,the optimum pH was 6.0.ⅲ) The relative humidity must be higher than 80% for mycelial growth and sporogenesis,and the lowest RH for spore germination is 85%.ⅳ) The best as to carbon sources for mycelial growth and sporogenesis are glucose or saccharose,while starch or glycerin can not be diretly used,unless they were decomposed previously.ⅴ) As to nitrogen sources,the fungus could utilize NO_3~——N(nitrate) very well,while NH_4~+-N(ammonium) inhibit its growth and sporogenesis.

Abstract:Background: Certain microorganisms during the preparation of substrate for growing Agaricus bisporus are of great interest for the conversion of organic material and for the excretion of a wide range of metabolites with growth regulation activities that may affect the mycelial growth processes and mushroom yield. Adding beneficial microorganisms in the substrate may be a biotechnological alternative to optimize A. bisporus cultivation. Objective: Isolate and evaluate thermophilic microorganisms from self-heating pasteurized substrate with growth-promoting effects on A. bisporus cultivation. Methods: Different microorganisms were isolated and selected at 45 and 55 °C. They were tested for siderophore production, 1-octen-3-ol consumption, and phosphate solubilization in coculture with A. bisporus to determine their growth effects on potato dextrose agar (PDA) and on sterile pangola grass (Digitaria eriantha). Results: Of the 106 microorganisms isolated, 88 % were able to grow in the presence of 1-octen-3-ol, while 1 % had the capacity to produce siderophores, and 55 % had the ability to solubilize phosphate. The strains Bacillus hisashii ECS-B-65, B. licheniformis ECS-B-78, Rhizomucor pusillus ECS-710 and ECS-712, Aspergillus fumigatus ECS-709, and Thermomyces sp. ECS-711 were found to have a positive effect on A. bisporus mycelial growth.

Abstract:In recent decades, an enormous potential of fungal-based products with characteristics equal to, or even outperforming, classic petroleum-derived products has been acknowledged. The production of these new materials uses mycelium, a root-like structure of fungi consisting of a mass of branching, thread-like hyphae. Optimizing the production of mycelium-based materials and fungal growth under technical conditions needs to be further investigated. The main objective of this study was to select fast-growing fungi and identify optimized incubation conditions to obtain a dense mycelium mat in a short time. Further, the influence of the initial substrate characteristics on hyphae expansion was determined. Fungal isolates of Ganoderma lucidum , Pleurotus ostreatus , and Trametes versicolor were cultivated for seven days on substrate mixtures consisting of various proportions of pine bark and cotton fibers. Furthermore, the substrates were mixed with 0, 2, and 5 wt.% calcium carbonate (CaCO 3 ), and the incubator was flushed with 0, 5, and 10 vol.% carbon dioxide (CO 2 ). All samples grew in the dark at 26 °C and a relative humidity of 80%. Evaluation of growth rate shows that cotton fiber-rich substrates performed best for all investigated fungi. Although Pleurotus ostreatus and Trametes versicolor showed comparatively high growth rates of up to 5.4 and 5.3 mm d −1 , respectively, mycelium density was thin and transparent. Ganoderma lucidum showed a significantly denser mycelium at a maximum growth rate of 3.3 mm d −1 on a cotton fiber-rich substrate (75 wt.%) without CaCO 3 but flushed with 5 vol.% CO 2 during incubation.

Abstract:High temperature stress caused by global warming presents a challenge to the healthy development of forestry. Cenococcum geophilum is a common ectomycorrhizal fungus (ECMF) in the forest system and has become an important fungus resource with application potential in forest vegetation restoration. In this study, three sensitive isolates of C. geophilum (ChCg01, JaCg144 and JaCg202) and three tolerant isolates of C. geophilum (ACg07, ChCg28 and ChCg100) were used to analyze the physiological and molecular responses to high temperature. The results showed that high temperature had a significant negative effect on the growth of sensitive isolates while promoting the growth of tolerant isolates. The antioxidative enzymes activity of C. geophilum isolates increased under high temperature stress, and the SOD activity of tolerant isolates (A07Cg and ChCg100) was higher than that of sensitive isolates (ChCg01 and JaCg202) significantly. The tolerant isolates secreted more succinate, while the sensitive isolates secreted more oxalic acid under high temperature stress. Comparative transcriptomic analysis showed that differentially expressed genes (DEGs) of six C. geophilum isolates were significantly enriched in “antioxidant” GO entry in the molecular. In addition, the “ABC transporters” pathway and the “glyoxylate and dicarboxylic acid metabolic” were shared in the three tolerant isolates and the three sensitive isolates, respectively. These results were further verified by RT-qPCR analysis. In conclusion, our findings suggest that C. geophilum can affect the organic acid secretion and increase antioxidant enzyme activity in response to high temperature by upregulating related genes.

Abstract:To study the cultural characteristics of mycelia of Ganoderma gibbosum, a medicinal fungus used in China. The growth rate and biomass of G. gibbosum mycelia were measured under different temperature, lightning carbon and nitrogen sources conditions. It showed that the optimal growth temperature for mycelia was 25 degrees C. Darkness was beneficial for mycelium growth. The initial pH 5.5 was suitable. The sucrose was the best carbon source and yeast extract the best nitrogen source, the optimal carbon-nitrogen ratio 60:2. These conclusions will offer references for further artificial cultivation and liquid fermentation.
Optimization of Biomass Cultivation from Tuber borchii and Effect of Additives on Triterpenoid Production

Abstract:Edible fungi are renowned for producing biologically active secondary metabolites that possess anti-tumor activity, protect the liver and have other benefits. The cultivation of truffle mycelia through submerged fermentation has gained interest in the production of metabolites for bio-medicinal purposes. In the present study, Tuber borchii was cultivated by submerged fermentation to produce both biomass and triterpenoids. Various additives, including palmitic acid, stearic acid, linoleic acid, chitosan, CaCl2 and limonene, were investigated to enhance triterpenoid production. It was observed that increasing the medium’s linoleic acid concentration to 1 g/L increased the production of triterpenoids to 129.29 ± 6.5 mg/L, which was 2.94 times higher than the control. A number of variables, including potassium and magnesium ion concentrations and carbon and nitrogen sources and concentrations, were considered to ascertain the ideal conditions for T. borchii growth in submerged fermentation. The best concentrations for glucose, yeast extract, peptone, malt extract, KH2PO4 and MgSO4·7H2O in submerged fermentation were 19.45, 4.58, 7.91, 5.3, 0.58 and 0.82 g/L, respectively, according to response surface methodology. Validation analysis revealed that the experimental values and the predicted values were in good agreement. Under ideal circumstances, the maximum dry cell weight (2.980.18 g/L), which was 1.39 times greater than the control, was attained. Finally, the addition of 1.5 g/L linoleic acid on day 14 to the optimal medium elevated the triterpenoid production to 212.63 ± 16.58 mg/L, which was a 4.84-fold increase compared to the control.

Black Truffles Affect Quercus aliena Physiology and Root-Associated nirK- and nirS-Type Denitrifying Bacterial Communities in the Initial Stage of Inoculation

Abstract:Truffles ( Tuber spp.) are edible ectomycorrhizal fungi with high economic value. Bacteria in ectomycorrhizosphere soils are considered to be associated with the nutrient uptake of truffles and hosts. Whether Tuber spp. inoculation can affect the growth of Quercus aliena , the ectomycorrhizosphere soil, and the rhizosphere nirK and nirS -denitrifier communities at the ectomycorrhizae formation stage is still unclear. Therefore, we inoculated Q. aliena with the black truffles Tuber melanosporum and Tuber indicum , determined the physiological activity and morphological indices of Q. aliena seedlings, analyzed the physicochemical properties of ectomycorrhizosphere soils, and applied DNA sequencing to assess the nirK and nirS - denitrifier community structure in ectomycorrhizosphere soils. Peroxidase activity was higher in the seedlings inoculated with T. melanosporum than in the T. indicum inoculation and uninoculated control treatments. The available phosphorus contents were lower and nitrate contents were higher in those with truffle inoculation, and T. melanosporum treatment differed more from the control than the T. indicum treatment. The richness of the nirK -community was highest in the T. indicum treatment and lowest in the uninoculated treatment. The differences in nirK -community composition across treatments were not statistically significant, but the nirS communities were different. The nirS -type bacteria correlated with three environmental factors (pH, available phosphorus, and nitrate contents), whereas the nirK -type bacteria were only associated with the nitrate contents. Generally, this work revealed that inoculation with Tuber spp. would change a few nutrient contents and richness of nirK -type bacteria and had little effects on growth of Q. aliena seedlings in the initial stage of inoculation. The results of this study may provide in-depth insights into the relationships between Tuber spp. and hosts, which should be taken into account when developing truffle production methods.

Comparison of Plasma Osteopontin Levels between Patients with Borderline Ovarian Tumours and Serous Ovarian Carcinoma.

Abstract:Osteopontin (OPN) is a novel biomarker of various cancers including ovarian carcinoma. OPN is a promising adjunct to a major biomarker of ovarian cancer, CA125, in diagnosis, differential diagnosis and prognosis. The aim of our study was to measure the plasma level of OPN and CA125 in patients with borderline ovarian tumours (BOTs), serous ovarian carcinoma, and controls to determine its potential role in the differential diagnosis between serous ovarian carcinoma and BOT. The plasma samples of 66 women were analysed using Luminex technology, designed to simultaneously measure multiple specific protein targets. The mean OPN plasma level for the control group was 23.3 ng/ml; for BOT 26.3 ng/ml; and for patients with serous ovarian carcinoma 59.5 ng/ml. Specifically, there was a significant difference between the OPN levels in patients with ovarian carcinoma and BOT (P < 0.001) as well as controls (P < 0.001). There was no difference between the mean levels of OPN in patients with BOT and the control group (P = 0.286). Using the receiver operating characteristic (ROC), we determined the utility of OPN and CA125 to differentiate between BOT and serous ovarian carcinoma. The area under the ROC curve (AUC) for OPN was 0.793 (95% confidence interval (CI) 0.669-0.917, P < 0.001) and for CA125 0.766 (95% CI 0.626-0.907, P = 0.002). Based on our data, we suggest that OPN can be used as a possible differential diagnostic biomarker to distinguish between malignant serous ovarian carcinoma and BOT.
Description, identification, and growth of Tuber borchii Vittad. mycorrhized Pinus sylvestris L. seedlings on different lime contents

Abstract:Tuber borchii forms ectomycorrhiza with oaks, hazel, and pines, including Pinus sylvestris . However, its ectomycorrhiza morphotype with P. sylvestris was not comprehensively described so far, and molecular analyses are missing despite a high danger of misidentification of T. borchii ectomycorrhiza with other closely related and less valuable truffle species. We described for the first time the morphology and anatomy of T. borchii-P. sylvestris ectomycorrhiza using differential interference contrast technique and semi-thin sections in combination with molecular confirmation of identity. Color of ectomycorrhiza is reddish to dark brown, and morphotypes are unevenly but densely covered by warts-bearing pin-like cystidia. All layers of the hyphal mantle are pseudoparenchymatous with outer mantle layer formed of epidermoid cells. T. borchii ectomycorrhiza was identified by a molecular comparison with fruitbodies used for inoculation and its respective ectomycorrhizae. T. borchii has a wide ecological amplitude. To get a better insight in mycorrhization requirements, we investigated growth of P. sylvestris and its ectomycorrhiza infection rate with T. borchii in substrate with different lime content. The mycorrhization of P. sylvestris with T. borchii in the mycorrhization substrate and cultivation in greenhouse conditions was successful, with colonization of P. sylvestris varying between 36.5 and 48.1%. There was no significant correlation of mycorrhization to applied lime contents, and consequently to pH in substrate, while the increased levels of lime improved growth of the P. sylvestris seedlings.

Comparative Transcriptome Profiles of the Response of Mycelia of the Genus Morchella to Temperature Stress: An Examination of Potential Resistance Mechanisms

Abstract:Temperature and moisture belong to the most important environmental factors affecting the growth and development of fungi. However, the effect of temperature on the mycelia of the edible Morchella mushrooms has not been determined. Here, a comprehensive analysis was performed to determine the influence of culture temperature on 13 strains of mycelia of three Morchella species (Morchella sextelata, Morchella septimelata, and Morchella importuna) at 5 °C, 10 °C, 15 °C, 20 °C, 25 °C, and 30 °C. The mycelial branching and growth rate data showed that 15–20 °C was a suitable temperature range for the mycelial growth of the 13 Morchella strains. RNA sequences revealed that a total of 2843, 2404, 1973, 1572, and 1866 differentially expressed genes (DEGs) were identified at 5 °C, 10 °C, 15 °C, 25 °C, and 30 °C compared with 20 °C. A Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis further indicated that the purine nucleotide and tyrosine metabolism pathways were crucial for mycelium development. Moreover, the enrichment of autophagy of mitochondria, regulation of cell morphogenesis, and piecemeal microautophagy of the nuclei at 25 °C (vs. 20 °C) indicated the damage caused by heat stress in Morchella mycelia. Notably, a total of four unique module eigengenes (MEs) were identified through a weighted gene coexpression network analysis (WGCNA). Among them, 2293 genes in the turquoise module were significantly positively correlated with temperature (r = 0.946, p < 0.001), whereas 739 genes in the blue module were significantly negatively correlated with temperature (r = −0.896, p < 0.001), suggesting that the effect of high temperatures on mycelial genes was significantly greater than that of low temperatures. Moreover, the coexpression network indicated that high culture temperatures accelerated the oxidative stress response and energy metabolism in mycelia, while upregulation of purine nucleotide catabolism and ribosomal protein-related genes were improved by low-temperature tolerance. In addition, the upregulated expression of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), and heat shock protein (HSP) genes in mycelia was associated with reactive oxygen species (ROS)-mediated damage at high temperatures. Overall, this study provides an important theoretical basis and application value for optimizing Morchella cultivation techniques.

Effect of Environmental Factors on Spore Germination, Spore Survival, and Growth of Gloeosporium Musarum

Abstract:Optimum temperature for growth, sporulation and conidial germination in G. musarum was in the range of 27–30°. The rate of germination was somewhat slower in light than in the dark, but under optimum conditions, maximum percentage of germination was obtained in both instances within 8 hours. Spores germinated in 6–12 hours at 98–100% relative humidity, but germination was delayed for 24–48 hours at 96%; germination failed to occur at 92%. Lowered oxygen tension slowed spore germination. Completely anaerobic conditions prevented spore germination, but did not prevent growth of the mycelium on agar medium. Mycelial growth was limited in an atmosphere of CO2, and there was evidence for CO2 toxicity. Optimum pH for spore germination was about pH 6, but germination occurred over the pH range of 4--10. Conidia survived on glass beads for 60 or more days, except over Drierite, where they survived less than 16 days. Germinated spores with appressoria were found to be more resistant to solutions of 0.25% sodium hypochlorite than were nongerminated spores.

Abstract:The synthesis of truffle ecomycorrhizae and the ecology of truffle-colonized seedlings in the early symbiotic stage are important for the successful truffle cultivation. In this study, two black truffle species, Tuber melanosporum and Tuber indicum, were selected to colonize Pinus armandii seedlings. 2, 4, 6 and 8 months after inoculation, the growth performance of the host and the rhizosphere soil properties were detected. The dynamic changes of two mating type genes in substrate were also monitored to assess the sexual distribution of truffles. Additionally, the variation of soil bacterial communities encoded by phoD alkaline phosphatase genes was investigated through next-generation sequencing. The results indicated that both T. melanosporum and T. indicum colonization promoted the growth of P. armandii seedlings to some extent, including improving their biomass, total root surface area, root superoxide dismutases and peroxidase activity. The organic matter and available phosphorus in rhizosphere soil were also significantly enhanced by two truffles' colonization. The phoD-harboring bacterial community structure was altered by both truffles, and T. melanosporum decreased their diversity or richness on the 6th and 8th month after inoculation. Pseudomonas, Xanthomonas, and Sinorhizobium, a N2-fixer with phoD genes, were found more abundant in truffle-colonized treatments. The mating type distribution of the two truffles was uneven, with MAT1-1-1 genes occupying the majority. Overall, T. melanosporum and T. indicum colonizationaffected the micro-ecology of truffle symbionts during the early symbiotic stage, and these results could give us a better understanding on the truffle-plant-soil-microbe interactions, which would be beneficial to the subsequent truffle cultivation.
