Abstract:Lotaustralin from the root of Rhodiola rosea L was extracted using supercritical carbon dioxide with methanol as modifier. Response surface methodology using Box-Behnken experimental design was utilized to explore parameters for supercritical carbon dioxide extraction. The effects of various values of temperatures (50-70 degrees C), pressures (200-400 bar), and percentages of methanol modifier (80%-100%) on the extraction yields of lotaustralin were evaluated. Extract identification was performed using high-performance liquid chromatography. The experimental data obtained were fitted to second-order polynomial equations and assessed using analysis of variance. The highest yields predicted from the experiments were 2.05 g kg(1) lotaustralin at the optimal values, i.e. temperature 65 degrees C, pressure 316 bar, 88% methanol modifier at a flow rate of 0.4 mL min(1), and dynamic extraction time 90 min.

Abstract:Curcuminoids consisted curcumin, demethoxycurcumin and bisdemethoxycurcumin, were extracted from turmeric using subcritical solvent by varying conditions of temperature (110-150 °C), time (1-10 min), pressure (5-100 atm), solid-to-solvent ratio, and mixing ratio of solvent. Preliminary lab-scale experiments were conducted to determine the optimum extraction temperature and mixing ratio of water and ethanol for the pilot-scale extraction. The maximum yield of curcuminoids in the pilot-scale system was 13.58% (curcumin 4.94%, demethoxycurcumin 4.73%, and bisdemethoxycurcumin 3.91% in dried extracts) at 135 °C/5 min with water/ethanol mixture (50:50, v/v) as a solvent. On the other hand, the extraction yields of curcuminoids were obtained as 10.49%, 13.71% and 13.96% using the 50%, 95% and 100% ethanol, respectively, at the atmospheric condition (60 °C/120 min). Overall results showed that the subcritical solvent extraction is much faster and efficient extraction method considering extracted curcuminoids contents and has a potential to develop a commercial process for the extraction of curcuminoids.

Abstract:Background: Curcuminoids, the bioactive compounds found in turmeric, exhibit potent antioxidant, anti-inflammatory, and neuroprotective properties. This study aims to enhance the extraction of curcuminoids from turmeric using environmentally friendly solvents supercritical CO2 (scCO2) combined with natural deep eutectic solvents (NADESs) in one process, and to evaluate the resulting biological activity. Methods: A Box–Behnken statistical design was applied to optimize scCO2 extraction conditions—pressure, CO2 volume, and temperature—to maximize curcuminoid yield. Next, the menthol and lactic acid NADESs were selected, and these two solvents were combined into a single turmeric extraction process. The biological activity of the resulting extract was evaluated using antioxidant assays (ferric reducing antioxidant power and 2,2-diphenyl-1-picrylhydrazyl) and enzyme inhibition assays (acetylcholinesterase, butyrylcholinesterase, and tyrosinase). Toxicity assessments were conducted on the aquatic invertebrates Daphnia pulex, Artemia sp., and Chironomus aprilinus. Results: The most effective extraction was achieved using a menthol–lactic acid NADES as a cosolvent, integrated at a 1:20 ratio of plant material to NADESs while in combination with scCO2. The optimized scCO2–NADES extraction resulted in a high curcuminoid yield (33.35 mg/g), outperforming scCO2 extraction (234.3 μg/g), NADESs ultrasound-assisted extraction (30.50 mg/g), and alcohol-based solvents (22.95–26.42 mg/g). In biological assays, the extract demonstrated significant antioxidant activity and effective inhibition of enzymes (acetylcholinesterase, butyrylcholinesterase, and tyrosinase). Toxicity studies showed a concentration-dependent response, with EC50 for Chironomus aprilinus at the level of 0.098 μL/mL and Daphnia pulex exhibiting high sensitivity to the extract. Conclusions: This study highlights the potential of combining NADESs and scCO2 extraction in one process, demonstrating the effectiveness of scCO2–NADES extraction in maximizing curcuminoid yield and enhancing bioactivity.

Abstract:A bio-active phytochemical, curcumin, was isolated from dried rhizomes of Curcuma longa using Soxhlet, microwave, ultra-sonic and supercritical carbon dioxide assisted extraction techniques. The quantification of curcumin in resultant extracts was performed using pre-validated HPLC methodology. The critical parameters viz. effect of pre-irradiation and soaking solvent on the curcumin yield were studied. The extraction efficiency of all the above described techniques was established in terms of percent curcumin yields and extraction rate constants. Prior to extraction, microwave and ultra-sonic irradiation of dry curcuma powder resulted in 68.57 and 40.00% curcumin yield, respectively, whereas water soaked irradiated curcuma powder yielded 90.47 and 71.42% curcumin recovery respectively, during a total extraction period of five minutes. The maximum extraction rate constant of 47.49×10−2min−1 was observed when using microwave assisted acetone extract of water soaked curcuma rhizomes. The comparison of Soxhlet, microwave, ultra-sonic and supercritical carbon dioxide assisted extraction in terms of percent yield and required extraction period showed that microwave assisted extraction technique was more efficient for the curcumin extraction from powdered C. longa rhizomes.

Abstract:The supercritical fluid extraction (SFE) of free anthraquinones (including chrysophanol, emodin, aloe-emodin, rhein, and physcion) from Rheum Palmatum L. was studied, in which the effects of operation conditions, such as extraction temperature and pressure, flow rate of CO2, static and dynamic extraction time, and the kind of entrainers, on the extraction rate were discussed. An orthogonal experimental design was employed to optimize the extraction conditions with respect to pressure, temperature and the amount of favorite entrainer. The extracted anthraquinones were analysed by high-performance liquid chromatography (HPLC). Results show that the temperature and pressure in the separation tank, and the flow rate of CO2 have a little effect on the extraction yield. The optimum conditions for maximun yield are found as following: the static extraction time 60 min, dynamic extraction time 30 min, ethanol used as entrainer 300 mL?(100 g raw material)?1, extraction temperature 45 °C and extraction pressure 45 MPa. The response anthraquinones yield under these optimum conditions is 1.15%.

Abstract:Curcuma longa [Linn] (turmeric), of the Zingiberaceae family, has a great importance in the food, textile, and pharmaceutical industries. The aim of this work was to identify the best processing conditions to maximize the yields of essential oil and pigments, as well as their content of ar-turmerone, (alpha and beta)-turmerone, and the curcuminoids, respectively. Autoclave pressure and distillation time were the variables studied for the steam distillation process. The highest yields of essential oil (0.46 wt %) and pigment (0.16 wt %)-expressed as curcumin, demethoxycurcumin, and bisdemethoxycurcumin-were obtained at a pressure of 1.0 x 10(5) Pa and a time of 2 h. On the other hand, with extraction by volatile solvents, the best yield of essential oil (5.49 wt %) was obtained when using 0.175, 0.124, 0.088 mm particles (Foust, A. S.; Wenzel, L. A.; Clump, C. W.; Maus, L.; Andersen, L. B. Princípios das Operações Unitárias; Editora Guanabara Dois S.A.: Rio de Janeiro, Brazil, 1982), at 40 degrees C, and 6 h of extraction. However, the best yield of pigment (7.98 wt %) was obtained under the same conditions, except for the temperature (30 degrees C).

Abstract:Curcumin is the major constituent of turmeric (Curcuma longa L.). It has attracted widespread attention for its anticancer and anti-inflammatory activities. The separation of curcumin and its two close analogs, demethoxycurcumin and bisdemethoxycurcumin, has been challenging by conventional techniques. In this study, an environmentally friendly method based on supercritical fluid chromatography was established for the rapid and facile separation of the three curcuminoids directly from the methanol extract of turmeric. The method was first developed and optimized by ultra performance convergence chromatography, and was then scaled up to preparative supercritical fluid chromatography. Eluted with supercritical fluid CO2 containing 8-15% methanol (containing 10mM oxalic acid) at a flow rate of 80 mL/min, curcumin, demethoxycurcumin and bisdemethoxycurcumin could be well separated on a Viridis BEH OBD column (Waters, 250 mm x 19 mm, 5 mu m) within 6.5 min. As a result, 20.8 mg of curcumin (97.9% purity), 7.0 mg of demethoxycurcumin (91.1%), and 4.6 mg of bisdemethoxycurcumin (94.8%) were obtained after a single step of supercritical fluid chromatography separation with a mean recovery of 76.6%. Showing obvious advantages in low solvent consumption, large sample loading, and easy solvent removal, supercritical fluid chromatography was proved to be a superior technique for the efficient separation of natural products.

Abstract:Natural aromas like cinnamaldehyde are suitable solvents to extract curcuminoids, the active ingredients found in the rhizomes of Curcuma longa L. In a pursuit to find other nature-based solvents, capable of solving curcumin, forty fragrances and flavours were investigated in terms of their solubilisation power. Aroma compounds were selected according to their molecular structure and functional groups. Their capabilities of solving curcumin were examined by UV–Vis spectroscopy and COSMO-RS calculations. The trends of these calculations were in accordance with the experimental solubilisation trend of the solubility screening and a list with the respective curcumin concentrations is given; σ-profiles and Gibbs free energy were considered to further investigate the solubilisation process of curcumin, which was found to be based on hydrogen bonding. High curcumin solubility was achieved in the presence of solvent (mixtures) with high hydrogen-bond-acceptor and low hydrogen-bond-donor abilities, like γ- and δ-lactones. The special case of DMSO was also examined, as the highest curcumin solubility was observed with it. Possible specific interactions of selected aroma compounds (citral and δ-hexalactone) with curcumin were investigated via 1H NMR and NOESY experiments. The tested flavours and fragrances were evaluated regarding their potential as green alternative solvents.

Abstract:Humans have utilized plants for various purposes, including sustenance and medical treatment for millennia. Researchers have extensively investigated medicinal plants' potential in drug development, spurred by their rich array of chemical compounds. Curcumin, a valuable bioactive compound, is extracted from Turmeric, known by the scientific name Curcuma Longa L. Notably, curcumin boasts potent antioxidant and anti-inflammatory properties, making it a promising candidate for treating cancer and other microbial diseases. Therefore, the simulation study of the extraction of this important medicinal compound by water, which is a green solvent, was carried out. This study employed molecular dynamics simulation for the first time to explore the extraction of Curcuma Longa L. extract using subcritical water. The simulations were carried out at constant pressure and different temperatures, using the Compass force field in the Lammps simulation package. The findings revealed an increase in the amount of Curcuma longa extract with rising temperature, indicating a weakening of hydrogen bonds in water molecules. Water lost its polar state with increasing temperature and became a suitable non-polar solvent for extracting non-polar compounds. The average absolute relative deviation (AARD) for calculated and simulated density data was 6.45%.

Abstract:The aim of this work was to study the effect of dynamic maceration factors upon the curcumin content of Curcuma longa L., Zingiberaceae, extracts and to determine the optimum set of parameters for the extraction of curcumin using a 25 full factorial design and the response surface methodology. Under the established conditions, the content of soluble solids and curcumin in the extracts ranged from 0.8 to 3.4%, and from 0.1 to 1.8%, respectively. The most influential variable observed for the extraction was the ethanolic strength of the solvent. The optimized condition involves an extraction time of 12 h, agitation speed of 30 rpm, drug to solvent ratio of 1/6, extraction temperature of 80 ºC and the solvent with ethanolic strength of 70%. The data reported herein are useful for further developments of curcuma phytopharmaceutical intermediate products with optimized characteristics.
High-efficient column chromatographic extraction of curcumin from Curcuma longa

Abstract:Curcumin is an important food additive and a potential therapeutic agent for various diseases from turmeric, the rhizome of Curcuma longa L. High-efficient column chromatographic extraction (CCE) procedures were developed for the extraction of curcumin from turmeric. Turmeric powder was loaded into a column with 2-fold 80% ethanol. The column was eluted with 80% ethanol at room temperature. For quantitative analysis with a non-cyclic CCE, 8-fold eluent was collected as extraction solution. For large preparation with a cyclic CCE, only the first 2-fold of eluent was collected as extraction and other eluent was sequentially circulated to the next columns. More than 99% extraction rates were obtained through both CCE procedures, compared to a 59% extraction rate by the ultrasonic-assisted maceration extraction with 10-fold 80% ethanol. The CCE procedures are high-efficient for the extraction of curcumin from turmeric with minimum use of solvent and high concentration of extraction solution.

Abstract:Solubility, extraction and stabilization of curcumin extracted from Curcuma longa are still a challenge. Either the solubility power of the extracting solvent mixture is not at its optimum or non-sustainable solvents are used or the curcumin instability against oxidation is a problem. In order to tackle these problems, we consider here a promising alternative solvent medium, which is a ternary system composed of water/sodium salicylate (an anionic, preservative hydrotrope)/ethyl acetate. The effects of the structuring of the system are also studied. The position of the new critical point and the orientation of tie-lines in the liquid/liquid area were determined and further examined via interfacial tension and diffusion ordered spectroscopy (DOSY NMR) measurements. The critical point between the mono-phasic region and the liquid/liquid area is a classical one, around which no special nano-structuring is found, in contrast to some other ternary mixtures with hydrotropes. The system was investigated in terms of its curcumin solubility and its extraction power thereof. Good extraction yields (~90% of the Soxhlet reference) could be achieved in the area of best solubility. Surprisingly, the hydrophobic curcumin shows a significant solubility in systems that contain a significant amount of water in presence of sodium salicylate. These mixtures are far from the critical point but show significant structuring. The solubility is even higher than in pure ethyl acetate. At the point of highest solubility (water/sodium salicylate/ethyl acetate 7/13/80 w/w/w) 15.48 mg of curcuminoids per g Curcuma longa are successfully extracted.The partition coefficient in the lower liquid/liquid two-phasic region was determined for a subsequent purification of curcumin by precipitation. Surprisingly, curcumin accumulates at the "oil-water" interface in the more polar phase. Sodium salicylate could be removed from the precipitated curcumin by washing it with water (no trace of sodium salicylate has been detected with NMR after the washing process).The preservative effect of salicylate against the oxidative effect of light on curcumin was found to be significant. A better stabilization of curcumin was observed far away from the critical point and was most efficient in the region, where aggregates could be detected. So, extraction, solubilization, and chemical stabilization of a light sensitive substance could be performed using an anti-oxidative hydrotrope.

Abstract:The essential oils of three medicinally important Curcuma species (Curcuma alismatifolia, Curcuma aromatica and Curcuma xanthorrhiza) were extracted using conventional hydro-distillation (HD) and solvent free microwave extraction (SFME) methods. The volatile compounds from the rhizome essential oils were subsequently analysed by GC–MS. The isolation of essential oils of each species was carried out following the six principles of green extraction and comparison was made between their chemical composition, antioxidant, anti-tyrosinase and anticancer activities. SFME was found to be more efficient than HD in terms of energy savings, extraction time, oil yield, water consumption and waste production. Though the major compounds of essential oils of both the species were qualitatively similar, there was a significant difference in terms of quantity. The essential oils extracted through HD and SFME methods were dominated by hydrocarbon and oxygenated compounds, respectively. The essential oils of all Curcuma species exhibited strong antioxidant activity, where SFME was significantly better than HD with lower IC50 values. The anti-tyrosinase and anticancer properties of SFME-extracted oils were relatively better than that of HD. Further, among the three Curcuma species, C. alismatifolia essential oil showed the highest rates of inhibition in DPPH and ABTS assay, significantly reduced the tyrosinase activity and exhibited significant selective cytotoxicity against MCF7 and PC3 cells. The current results suggested that the SFME method, being advanced, green and fast, could be a better alternative for production of essential oils with better antioxidant, anti-tyrosinase and anticancer activities for application in food, health and cosmetic industries.

Abstract:The extraction condition of curcumin from Curcuma longa L was optimized through four factors and three levels orthogonal experiment based on the results of single factor tests. Under the optimal conditions:the concentration of ethanol 80 %, extraction temperature 70°C, the ratio of liquid to material 20, and extraction time 3 h, a crude extract with the yield of curcumin 56.8 mg/g could be obtained. The isolation and purification of curcuminoids from the crude extract was performed on high performance counter current chromatography employing an optimized solvent system n‐hexane‐ethyl acetate‐methanol‐water (2/3/3/1, v/v). From 97 mg crude sample (in which the purity of curmumin was 68.56 %), 67 mg curmumin, 18 mg demethoxycurcumin and 9.7 mg bisdemethoxycurcumin with HPLC purity of 98.26 %, 97.39 % and 98.67 % were respectively obtained within 70 min. The antioxidant activities and cytotoxicity of purified curcumin was comparable to that of the commercial product, indicating that the biological activity of curcumin could be maintained by this method.This article is protected by copyright. Abstract:The essential oil composition and total phenolic content (TPC) of curcuminoids were studied in rhizomes of nine Curcuma longa L. accessions. Curcuminoids, present in commercially available turmeric rhizomes, play vital roles in various pharmacological activities. A simple, rapid, and sensitive high performance liquid chromatography photodiode array (HPLC-PDA) method was optimized for simultaneous determination of curcuminoids, namely, a mixture of curcumin, demethoxy curcumin (DMC), and bisdemethoxy curcumin (BDMC) in rhizomes of C. longa. Chromatographic separation was performed on an RP C18 column within 13 minutes (11.4 to 12.95 minutes). Elution was accomplished by the application of acetonitrile and 1.5% acetic acid in water in a gradient system with flow rate of 2.0 mL min−1. PDA was employed for qualitative and quantitative analysis. The calibration curves were found linear (0.99) for all cucuminoids; the limit of detection and quantification ranged between 1.01 µ g mL−1 to 1.16 µ g mL−1 and 2.30 µ g mL−1 to 3.05 µ g mL−1, respectively, while recovery values ranged between 97.97% to 98.32%. The amount of curcumin varied from 0.46% to 2.17%, DMC from 0.13% to 0.92% and BDMC from 0.06% to 0.52%. The validated method was successively used to determine the above compounds in C. longa rhizomes. The TPC in rhizomes ranged from 14.12 mg g−1 to 27.72 mg g−1. The chemical composition of rhizome essential oil, analyzed by gas chromatography mass spectrometry (GCMS,) showed large variations in major compounds like ar-tumerone (7.31–38.66%), β-curcumene (1.58–24.53%), and curlone (1.55–15.97%).

Abstract:In this work a method is developed for the extraction and purification of the three curcuminoids, curcumin (CUR), demethoxycurcumin (DMC), and bisdemethoxycurcumin (BDMC), from commercially available crude curcumin. In a previous publication the extraction of pure curcumin by repeated crystallization has been described. The focus of this paper is on the following chromatographic treatment of the mother liquor from the crystallization to obtain pure DMC and BDMC and to increase the yield of pure CUR. In the chromatographic process, a mixture of chloroform and methanol is used as the mobile phase, and silica gel is used as the stationary phase. Each fraction isolated in the chromatographic process was characterized by high-performance liquid chromatography (HPLC) and mass spectrometry (LC-MS) techniques, and the pure CUR, DMC, and BDMC solid phases were fully characterized by powder Xray diffraction (PXRD), differential scanning calorimetry (DSC), and thermal gravimetric analysis (TGA). Stability studies were performed on the purified curcuminoids where the degradation products were observed and analyzed by HPLC/LC-MS. Overall, the combined purification method recovered from the crude: 88.5%, 79.7%, and 68.8% of CUR, DMC, and BDMC, respectively, in highly pure form CUR (100%), DMC (98.6%), and BDMC (98.3%).

Abstract:A reversed-phase high performance liquid chromatographic method was established for simultaneous determination of curcumin, demethoxycurcumin and bidemethoxycnrcumin in the rhizome of Curcuma longa L., a typical herbaceous plant. A Kromasil(TM) C-18 column was used with a mixture of acetonitrile and phosphoric acid aqueous solution ( pH = 2. 5) as the mobile phase. The flow rate was 0. 9 mL/min and the detection wavelength was set at 425 nm. The three constituents, were separated within 10 min. The method is rapid, simple and accurate, and it can be used for the evaluation of Curcuma longa L.

Abstract:A coupled ultrasonic extraction process of curcumin from Curcuma longa L enhanced by the graphene-oxide-assisted solvent was developed. The yield was investigated in relation to extraction temperature, extraction time, ethanol volume fraction, and graphene oxide concentration. Response surface methodology was used to develop a quadratic model of the yield and extraction process. The optimum extraction parameters with the highest yield were obtained and the economic value was then assessed. Results showed that the coupling process could reduce the extraction time, extraction temperature, and ethanol volume fraction while improving the extraction yield. The optimal process parameters were 50.0°C extraction temperature, 1.5 h extraction time, 60.0% ethanol volume fraction, and 1.0 mg/g graphene oxide concentration. The highest extraction yield was 64.9 mg/g, which was better than that have been reported. It can save an estimated $1507 in production expenses for every ton of turmeric under the optimal extraction process. Moreover, the reduction in extraction temperature and ethanol volume fraction also means a safer and cleaner production environment, which highlights the greater potential application prospect of the extraction process developed in this paper.
