Abstract:The increasing demand for tea consumption calls for the development of more products with distinct characteristics. The sensory quality of tencha is significantly determined by innate differences among tea cultivars. However, the correlations between the chemical composition and sensory traits of tencha are still unclear. To enhance the understanding of the flavor formation mechanism in tencha and further to develop new cultivars resources, we investigated non-volatiles and volatile metabolites as well as sensory traits in tencha from different tea cultivars (Camellia sinensis cv. Yabukita, Longjing 43 and Baiye 1); the relationships between the flavor traits and non-volatiles/volatiles were further evaluated by partial least squares - discriminate analysis (PLS-DA), multiple factor analysis (MFA) and multidimensional alignment (MDA) analysis. A total of 64 non-volatiles and 116 volatiles were detected in all samples, among which 71 metabolites were identified as key flavor-chemical contributors involving amino acids, flavonol glycosides, flavones, catechins, ketones, alcohols, hydrocarbons, aldehydes, esters and acids. The levels of taste-related amino acids, flavonol glycosides and gallic acid varied significantly among the tencha samples made from different tea cultivars. All the samples exhibited typical quality characteristics of tencha. The tencha from Camellia sinensis cv. Longjing 43 and Camellia sinensis cv. Baiye 1 (cultivated in the open) exhibited higher levels of amino acids and gallic acid, which were associated with the umami taste and mellow taste of tea infusion. Abundant flavonol glycosides were related to the astringency, while partial tri-glycosides specifically quercetin-3-O-galactoside-rhamnoside-glucoside and total of flavonol galactoside-rhamnoside-glucoside were associated with mellow taste. The floral alcohols were identified as significant contributors to the refreshing aroma traits of tencha. The green, almond-like, acidic and fruity odorants were associated with a green and fresh aroma, while the green, cheesy and waxy odorants such as ketones, esters, acids and hydrocarbons were associated with seaweed-like aroma. This study provides insight into sensory-related chemical profiles of tencha from different tea cultivars, supplying valuable information on flavor and quality identification for tencha.
UHPLC Analysis of Major Functional Components in Six Types of Chinese Teas: Constituent Profile and Origin Consideration

Abstract:In this study, an effective ultra-high performance liquid chromatography (UHPLC) method for simultaneous determination of 15 constituents of catechins, caffeine, gallic acid and theaflavins was developed. Employing the developed UHPLC method, the contents of the 15 constituents were analyzed in 245 samples of 6 types of Chinese teas collected from different places. The catechin contents in these samples ranged from 16.50 to 94.35 mg/g. Theaflavins were not detected in green tea and white tea, and caffeine did not strongly relate to germplasm resource or fermentation degree. Principal component analysis (PCA) of UHPLC data revealed that white, oolong and green tea samples could be clustered closely, demonstrating notable difference of functional components between the three types of teas resulting from preparation process and varieties. The constituent profile could be primarily attributed to the structural transformation of catechins and pigments during tea processing. This study would advance our understanding of chemical profile of Chinese teas afforded by different processing technologies.
Taste-Characteristic Attributes and Related Contribution Compounds of White Tea

Abstract:This study focused on the taste-characteristic attributes of representative white teas and related taste-chemical compositions. In this study, the white tea made from 9 representive cultivars were taken as research objects, including Fuding Dabaicha, Fuan Dabaicha, Chun Yu2 etc. Sensory evaluation according to the national standard was used for taste-characteristic attributes analysis, high-performance liquid chromatography(HPLC) and other techniques were used for chemical detection, and chemometrics analysis was used to explore the relationship between taste-characteristic attributes and chemical compositions. In this study, 9 types of white teas of different cultivars reflected six main taste-characteristic attributes: Clear, fresh, sweet, mellow, thick, and strong. Glutamic acid was correlated with the umami taste attribute in white tea samples(R=0.525) positively. The fresh taste was positive correlated with ratio of glutamate to the sum of epigallocatechin gallate and epigallocatechin(Glu/(EGCG+EGC)) strongly(R=0.695). And the ratio of total kaempferide to total flavonol glycoside(Kae/TFOG) was positively correlated with the fresh taste(R=0.452). Meanwhile, the high ratio of total amino acids to total catechins(aa/Ca>0.30) and ratio of total amino acids to total alkaloids(aa/Alk>0.90) contributing to the mellow and thick taste in the samples greatly.
The Integration of Metabolomics, Electronic Tongue, and Chromatic Difference Reveals the Correlations between the Critical Compounds and Flavor Characteristics of Two Grades of High-Quality Dianhong Congou Black Tea

Abstract:Tea's biochemical compounds and flavor quality vary depending on its grade ranking. Dianhong Congou black tea (DCT) is a unique tea category produced using the large-leaf tea varieties from Yunnan, China. To date, the flavor characteristics and critical components of two grades of high-quality DCT, single-bud-grade DCT (BDCT), and special-grade DCT (SDCT) manufactured mainly with single buds and buds with one leaf, respectively, are far from clear. Herein, comparisons of two grades were performed by the integration of human sensory evaluation, an electronic tongue, chromatic differences, the quantification of major components, and metabolomics. The BDCT possessed a brisk, umami taste and a brighter infusion color, while the SDCT presented a comprehensive taste and redder liquor color. Quantification analysis showed that the levels of total polyphenols, catechins, and theaflavins (TFs) were significantly higher in the BDCT. Fifty-six different key compounds were screened by metabolomics, including catechins, flavone/flavonol glycosides, amino acids, phenolic acids, etc. Correlation analysis revealed that the sensory features of the BDCT and SDCT were attributed to their higher contents of catechins, TFs, theogallin, digalloylglucose, and accumulations of thearubigins (TRs), flavone/flavonol glycosides, and soluble sugars, respectively. This report is the first to focus on the comprehensive evaluation of the biochemical compositions and sensory characteristics of two grades of high-quality DCT, advancing the understanding of DCT from a multi-dimensional perspective.

A HPLC Method for Detection of 17 Characteristic Components in Tea Extract

Abstract:A HPLC method has been developed for the determination of tea for seven catechins ((+)-catechin, (−)-epicatechin, (−)-epigallocatechin, (−)-epigallocatechin gallate, (−)-epicatechin gallate, (−)-gallocatechins and (−)-gallocatechin gallate), two flavonols (quercetin, kaempferol), seven organic acids (Gallic acid, ferulic acid, protocatechuic acid, p-hydroxybenzoic acid,p-coumaric acid, erucic acid,vanillic acid) and caffeine. Using HPLC with gradient elution and photodiode array detection, seventeen compounds were separated and detected by optimizing the column temperature, mobile phase ratio and detection wavelength. The validity of this method was confirmed by the quantitative measurement of seventeen characteristic compounds. The correlation coefficient of all standards curve was 0.9967~1.0000 and the detection limits (s/N = 3) were 0.010~1.2 mg/L. The average recoveries of 17 characteristic components were between 81.50 and 124.80%. 59 green tea samples (origin: Hangzhou, China) were analyzed and 11 characteristic components were found in all samples and their content of different samples varied greatly. The catechins were the most abundant constituents in teas with up to 79.5~156.6 g/kg and the contents of (−)-epigallocatechin gallate were the highest. Quercetin and kaempferol were only found in some tea samples. Protocatechuic acid, p-hydroxybenzoic acid, ferulic acid and erucic acid were not detected in the tea samples. This method is ideally suited for routine analysis for the determination of bioactive components in green tea with good repeatability and accuracy of results. Furthermore, this method can be applied to all kinds of tea and tea products and provided a detection method for establishing the HPLC fingerprint of a certain tea.
Quantitative analysis and correlation evaluation on taste quality of green tea

Abstract:Summary There are many of green teas with different characteristics in China , and their qualities are often evaluated by sensory evaluation without quantifying chemical compounds . Though sensory characteristics are identified quickly , the contents of compounds are unknown . Determination of chemical compounds is usually used for tea quality evaluation and the results are relatively accurate and objective . But most of the studies focus on analyzing chemical compounds of a single flavor type . Reports about comparison of main compounds in different taste types are rare . So the chemical compounds of different taste types of green tea were quantified and their relationships with the taste quality were analyzed in this study to understand the material basis of formation of different taste types in green tea . Forty‐eight typical green teas in seven different taste types were selected from 287 tea samples by sensory evaluation . Thirteen chemical factors including tea polyphenols , amino acids , water extract , and caffeine were determined by standard methods . The relationships between compound contents and the taste quality were analyzed using SPSS ( statistic package for social science) 16 .0 software , and the tea quality predicting model based on the chemical compounds was established , which can provide certain methods for judgment of tea quality . Furthermore , the degree of correlation between every property and chemical components was carried out .The results indicated that the ester catechins , especially epigallocatechin gallate ( EGCG ) had a great contribution to the astringency . The green tea samples with astringency were of the high contents of water extract , tea polyphenols , ester catechins and EGCG . The green tea samples with strong taste were of high content of water extract . And the green teas with tastes of mellow , sweet and brisk were related to the low content of water extract and high content of amino acids . The factors of total water extracts , EGCG , gallic acid ( GA ) , caffeine and gallocatechin gallate ( GCG ) showed stronger correlation with tea quality . The equation based on stepwise regression analysis had good predictive ability for the taste quality and total quality , and the right ratios for taste score and total quality score were 79% and 83% respectively . Furthermore , the water extract , EGCG , caffeine , GCG , epicatechin gallate ( ECG) and tea polyphenols were well related to the divided taste properties .In conclusion , different kinds of taste characteristics are based on different contents of chemical components in green tea .
Screening the Cultivar and Processing Factors Based on the Flavonoid Profiles of Dry Teas Using Principal Component Analysis

Abstract:This study screened the effects of cultivar and processing method on the flavonoid profiles of different teas. Four tea cultivars 'Jiulongpao', 'Zhenong 902', 'Jinxuan' and 'Huangjingui' were respectively processed into green tea, oolong tea and black tea. The concentrations of 13 flavonol glycosides and 8 catechins in teas were simultaneously determined by UHPLC-DAD-MS/MS in combination with SRM mode. The intercultivar difference of flavonol glycosides in the same type of teas was greater than that of catechins. The contents of total flavonol glycosides and total catechins in black teas were respectively 14.2% similar to 28.8% and 82.9% similar to 84.6% lower than those of green teas made from the same cultivar. Myricetin glycosides, mono-glycosides, tri-glycosides and catechins are vulnerable to fermentation. Principal component analysis results indicated that the flavonol glycoside profiles of dry teas were well discriminated by tea plant cultivar rather than manufacturing procedure, whereas the catechin profiles of black teas were distinguished by fermentation process. Thus, tea plant cultivar is an important factor to be considered for consumption decision on dry teas, especially non-fermented and semi-fermented teas, due to remarkable intercultivar difference in flavonol glycosides.
Comparison of Chemical Constituents and Antioxidative Activities among Different Grades of Pu-erh Tea

Abstract:The antioxidative activity,aroma constituents and the major chemical components of three grades of Pu-erh tea,i.e.high,medium and low grade,were studied and compared.Results showed that the significant difference(P0.05)was shown in evaluation of the total antioxidative activity,scavenging hydroxyl free radical activity,scavenging DPPH free radical activity,total amount of alcohols compounds,total amount of heterocyclic oxygen compounds,total flavonoids and tea pigments between high and low grades.When compared with high or low grade,the medium grade of Pu-erh tea was found no significant difference(P0.05)in the others testing index except the total antioxidative activity,scavenging hydroxyl free radical activity,tea polyphenols and total flavonoids.Principal component analysis showed that the important indicator to distinguish the grade of Pu-erh tea seemed to be the scavenging hydroxyl free radical activity,the total antioxidative activity,tea polyphenols,total flavonoids and total amount of alcohols compounds.
A Study on Chemical Estimation of Pu-Erh Tea Quality

Abstract:Chemical compositions and infusion colour differences of seven pu-erh tea samples and their correlation to sensory quality were investigated. The results showed that the pu-erh tea contained 37.1 mg g(-1) caffeine, 15.7 mg g(-1) amino acids, 67.0 mg g(-1) polyphenols and 8.41 mg g(-1) total catechins, on average. Among the 17 tested volatile compounds, n-valeraldehyde was not detected. The most abundant volatile was beta-ionone and the next was linalool oxide II. Infusion colour analysis showed that the pu-erh tea had deep hue with DeltaE ranging from 66.8 to 79.2. Spearman's linear correlation analysis showed that total quality score (TQS) of the pu-erh tea was significantly correlated to concentration of amino acids, linalool oxide II and infusion colour indicator DeltaE. Five components were extracted from the 34 tested indicators by principal component analysis and were regressed on the TQS to produce six Pearson's linear regression equations for estimating sensory quality of pu-erh tea, among which two were statistically significant, ie TQS = 57.47 - 0.18geraniol + 0.33polyphenols - 1.14n-caproaldehyde - 1.38linalool oxide I + 0.21caffeine (p < 0.01) and TQS = 57.42-0.03Citral + 0.33polyphenols - 1.14n - caproaldehyde - 1.40linalool oxide I + 0.20caffeine (p < 0.01). Discovery and Flavor Characterization of High-Grade Markers in Baked Green Tea

Abstract:Green tea is a popular beverage around the world and possesses a unique flavor. The flavor qualities of green tea are closely related to its grade and this relationship has not yet been studied. Three baked green teas with similar flavor were studied, namely, Huangshan Maofeng, Taiping Houkui, and Shucheng Xiaolanhua. A total of 34 odor compounds were identified by solid phase microextraction (SPME) combined with two-dimensional comprehensive gas chromatography–olfactometry–mass spectrometry analysis (GC×GC-O-MS). The results of the clustering analysis showed that the content of D-limonene and linalool in the high-grade (Grade A) tea was much higher than the content in other grades, so they were identified as odor markers of Grade A baked green tea. The taste components of different grades of green tea infusion were analyzed by high-performance liquid chromatography–mass spectrometry (HPLC–MS) and HPLC. A combination of clustering analysis, principal component analysis (PCA), and orthogonal partial least squares discrimination analysis (OPLS-DA) indicated that galloylglucose, digalloylglucose, trigalloyglucose, strictinin, and gallic acid could be used as taste markers of Grade A baked green tea. Therefore, the results in this paper reveal the substances responsible for the odor and taste markers of high-grade baked green tea.

Abstract:Pan-fried green tea (PGT) is an easily acceptable tea drink for general consumers. In this study, volatile profiles and characteristic aroma of 22 representative Chinese PGT samples were extracted using stir bar sorptive extraction (SBSE) and analysed by gas chromatography-mass spectrometry (GC-MS), gas chromatography-olfactometry (GC-O) analysis, and odour activity value (OAV) calculations. In total, 88 volatile compounds were identified. Alcohols (45%), esters (19%), and ketones (16%) were the dominant volatiles, and geraniol (484.8 μg/kg) was the most abundant volatile component in PGT, followed by trans-β-ionone and linalool. In addition, the differences of aroma characteristics among PGT and other three types of green tea, namely baked green tea, steamed green tea, and sun-dried green tea, were also observed using partial least squares discriminant analysis (PLS-DA) and heatmap analysis, and it was found that β-myrcene, methyl salicylate, ( E )-nerolidol, geraniol, methyl jasmonate were generally present at higher content in PGT. This is the first comprehensive report describing the volatile profiles of Chinese PGT, and the findings from this study can advance our understanding of PGT aroma quality, and provide important theoretical basis for processing and quality control of green tea products.

Abstract:In this study, the chemical constituents of pu-erh tea, black tea, and green tea, as well as those of pu-erh tea products of different ages, were analyzed and compared using a chemical profiling approach. Differences in tea processing resulted in differences in the chemical constituents and the color of tea infusions. Human biological responses to pu-erh tea ingestion were also studied by using ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOFMS) in conjunction with multivariate statistical techniques. Metabolic alterations during and after pu-erh tea ingestion were characterized by increased urinary excretion of 5-hydroxytryptophan, inositol, and 4-methoxyphenylacetic acid, along with reduced excretion of 3-chlorotyrosine and creatinine. This study highlights the potential for metabonomic technology to assess nutritional interventions and is an important step toward a full understanding of pu-erh tea and its influence on human metabolism.

Abstract:Parameters of liquor color difference, taste related constituents and aroma related volatiles in 23 Chinese green tea (Camellia sinensis) samples were analyzed by color difference meter, high performance liquid and gas chromatographs respectively. Seven variables were extracted by principal component analysis from the data sets corresponding to the liquor color, taste and aroma and linear regression of total score of sensory preference (TSSP) upon the 7 extracted variables produced a significant relationship: TSSP = 105.82–0.56 (ΔE)−0.15 (total catechins) + 0.32 (geraniol) + 0.53 (GCG) + 0.44 (linalool oxide I) −0.54 (ascorbic acid) + 0.24 (n-valeraldehyde) (p = 0.003, standard error of the estimation = 2.97). The regressive relationship simplifies the process of chemical and instrumental assessment of sensory preference of green tea and could be used for green tea quality assessment.

Abstract:Crude tea polysaccharides (CTPS) were extracted from low-grade green tea, separately named CTPS-I by waterboiling extraction method and CTPS-II by compound enzyme method, and CTPS-III extracted from tea leaves by compound enzyme method. The content of saccharide and protein of them were determined. The homogeneity distribution and mass ratio of polysaccharides constituent were analyzed using HPGPC-ELSD, and there were 5, 4 and 7 kinds of homogeneous constituents accordingly in CTPS-I, II and III. There were six kinds of monosaccharide residues (rhamnose, arabinose, xylose, mannose, glucose and galatose) in CTPS confirmed by means of GC-MS. UV spectra discovered that CTPS-I had a distinctive peak absorption at 257 nm, while CTPS-II showed a shoulder-shaped peak absorption in 240-270 nm, and CTPS-III exhibited a weak double wave-shaped absorption in 250-360 nm. In addition, the mass ratio of free protein and nucleic acid in CTPS was determined. IR revealed the characteristics of tea polysaccharides complex including. related protein and nucleic acid. CD spectra showed that their conformations in water were different.

Abstract:Different aromatic components do indeed give different tea flavors. There is still little research on whether there is a certain regularity in the combination and content of aromatic components in different aroma types of Phoenix Dancong (PDC) tea. This potential regularity may be a key factor in unraveling the relationship between reproduction and evolution in PDC tea. Here, the 5 kinds of these 4 aroma types PDC tea (Zhuye, Tuofu, Jianghuaxiang, Juduo, Yashixiang) were used as research materials in this study, the headspace solid-phase microextraction combined with gas chromatography–mass spectrometry was used to analyze the aromatic components of these PDC teas. The results showed a total of 36 aromatic components identified in this study. When conducting cluster analysis, it was found that similarity degree arrangement sequence of 5 PDC teas was Juduo, Tuofu, Yashixiang, Zhuye and Jianghuaxiang. Among these aromatic components, the 7,9-Di-tert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione, the 2-Cyclopenten-1-one, 3-methyl-2-(2-pentenyl)-,(Z)-, the 2,4-Di-tert-butylphenol, the 3,7-dimethyl-1,5,7-Octatrien-3-ol, and the 2-Furanmethanol,5-ethenyltetrahydro-.alpha.,.alpha.,5-trimethyl-,cis- are common to 5 PDC teas. This study aims to elucidate the similarities in the aromatic components of 5 PDC teas, revealing the major aroma-endowed substances of various aroma, and providing theoretical reference for further exploring the relationship between aroma type discrimination, variety selection, and evolution of PDC teas.

Abstract:The potential of the electronic nose to monitor Longjing tea different grade based on dry tea leaf, tea beverages and tea remains volatiles was studied. The original feature vector was obtained from the response signals of the E-nose, and was analyzed by principal component analysis (PCA). To decrease the data dimension and optimize the feature vector, the front five principal component values of the PCA were extracted as the final feature vectors by PCA. The linear discrimination analysis (LDA) and the back-propagation neural network (BPNN) were proposed to identify Longjing tea grade. The results showed that the discrimination results and testing results for the tea grade were better based on the tea beverages than those based on the tea leaf and the tea remains based on the new five feature vectors; both of the LDA and BPNN methods achieved better discrimination for the tea grades based on the tea beverages and the analysis results of the two methods were accordance.
Identification of volatile components and analysis of aroma characteristics of Jiangxi Congou black tea

Abstract:Jiangxi Congou black tea is one of the earliest Congou black tea in China. In order to explore its sensory characteristics and molecular sensory basis, quantitative description analysis (QDA) and gas chromatography-mass spectrometry (GC-MS) were used to analyze the sensory characteristics and volatile components of 11 Congou black tea samples from Jiangxi Province in China. The results showed that the aroma characteristics of Jiangxi Congou black were composed of 11 aroma descriptors, namely, sweet-smell, floral, fruity, roasted, spicy, fresh, green, oil, off-odor, woody and smoky. There was also interaction effect between sensory attributes of aroma, which were significant negative correlation between floral, and fruity, and green. The presence of roasted incense could significantly enhance the olfactory perception of off-odor and smoky, and off-odor was an important attribute that affects the aroma quality of Jiangxi Congou black tea. Volatile components (i.e. 74 kinds) were identified, among which the highest content and species were alcohols, followed by esters, and the lowest were heterocycles and terpenes. The results of partial least square variable import project (PLS-VIP) showed that there were 29 volatile compounds that had an important contribution to the aroma characteristics of Congou black tea, and the volatile compounds that had an important influence on six aroma attributes were obtained.
Discrimination of LongJing Green-Tea Grade by Electronic Nose

Abstract:An investigation was made to evaluate the capacity of an electronic nose (E-nose, PEN2) to classify the tea quality grade. Four tea groups (A120, A280, A380 and A600) with a different quality grade were employed. In the experiment, the volume of vial and the headspace generated time were considered corresponding to the 5g tea samples, and the optimum experimental procedure was determined by using the variance analysis (ANOVA), multivariance analysis and principle components analysis (PCA). The results showed that the volume of vial affected the result of discrimination, and the headspace generated time had no significant effect on the E-nose response. The four tea groups were measured and response values at four different collection times were conducted by PCA, linear discrimination analysis (LDA) and artificial neural network (ANN). Only A120, A380 and A600 could be discriminated by PCA. However, the four tea groups were discriminated completely by LDA. The response value of the E-nose at 60s was optimum to be used for discrimination. The method of ANN (network topology 20-12-4) was performed and 90% of the total tea samples were classified correctly by using the back-propagation neural network.

Abstract:Maillard reaction products (MRPs) produced during thermal processing of tea are intimately related to its flavor. Our recent work revealed that both levels of L-theanine and D-galacturonic acid in tea leaves decreased dramatically during drying, whereas the specific MRPs from L-theanine and D-galacturonic acid remain elusive. Here, the MRPs formed from L-theanine and D-galacturonic acid were investigated and their taste characteristics and the involved mechanisms were explored. Two novel MRPs from L-theanine and D-galacturonic acid were identified as 1-(1-carboxy-4-(ethylamino)-4-oxobutyl)-3-hydroxypyridin-1-ium (MRP 1) and 2-(2-formyl-1Hpyrrole-1-yl) theanine (MRP 2). MRP 1 and MRP 2 accumulated in dark tea and black tea and were associated with sour (threshold, 0.25 mg/mL) and astringent tastes and an umami taste (threshold, 0.18 mg/mL), respectively. Molecular docking revealed that the taste characteristics of MRPs may be due to strong binding to umami taste receptor proteins (CASR, T1R1/T1R3) and the sour taste protein OTOP1 via hydrogen bonds and hydrophobic interactions.

Abstract:Aroma is an important factor in evaluating tea infusion quality. In this study, the aroma quality of 44 Dianhong Congou tea infusions was classified into high-quality group and non-high-quality group according to the sensory evaluation. The volatile components were investigated by using headspace solid phase microextraction combined with comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry. A total of 258 volatile components were identified, of which the three component types with the highest relative percentages were alcohols (17.05%), aldehydes (15.12%), and heterocycles (15.12%). Clear discrimination was achieved by orthogonal partial least squares discrimination analysis (R2Y = 0.94, Q2 = 0.82). In addition, 69 important differential compounds were identified based on variable importance in the projection value (VIP >1.0) and one-way ANOVA (p 1 were screened out, including (E, E)-2,4-heptadienal, linalool, 1-octene-3-one, geraniol, β-myrcene, (Z)-4-heptenal, and nonanal. Aroma character impact value indicated that linalool performed a positively supportive role in the aroma quality of Dianhong Congou tea infusions, while (E, E)-2,4-heptadienal negatively affected the aroma quality. These results contribute to providing a theoretical support for directional processing of high-quality black tea.
