Abstract:An aza-BODIPY-CNOH probe attached aldoxime group demonstrated the specific detection for hypochlorous acid by the turn-on red emission signal. NMR and HRMS experiments confirmed that the fluorescence originated from the oxidation degradation of the non-fluorescence, aldoxime-based aza-BODIPY-CNOH probe into the red-fluorescence, nitrile oxide-based aza-BODIPY compound aza-BODIPY-CNO. The aza-BODIPY-CNOH probe showed good biocompatibility and was low toxic to living cells as shown from MTT experiments. Living RAW264.7 cells imaging indicated the aza-BODIPY-CNOH probe had good permeability and either exogenous or endogenous HClO caused the intracellular bright-red fluorescence, showing its potential hypochlorous acid-specific sensing ability in biological systems.

Abstract:A spectroscopic probe CMBT was synthesized and characterized. CMBT showed the specific recognition for HClO based on the turn-on blue fluorescence and naked-eye change from pink to colorless. NMR, IR, HRMS-ESI, and spectral analysis suggested that colorimetric and fluorescent change of CMBT to HClO originated from the conversion of CMBT to starting material coumarin-aldehyde 1 caused by the oxidization of HClO, which was responsible for the fluorescence recovery. The detection limit was calculated to be 1.61 mu M and 6.58 mu M for fluorescence and UV-vis analysis with a range up to 1 mM. HClO's fluorescence detection was successfully achieved in tap and river water samples. The prepared convenient paper test strips showed a distinct color change in varying concentrations of HClO. Abstract:A new phenanthroimidazole-based fluorescence probe for selective detection of HClO was synthesized and characterized using (HNMR)-H-1, (CNMR)-C-13, IR, and HRMS. With benzenesulfonohydrazide as the identification group, the probe demonstrated a fast fluorescence response from yellow-green to blue when the HC = N double bond was oxidized and broken into an aldehyde group by HClO. The probe showed high selectivity and sensitivity towards HClO with approximately 4.5-fold fluorescence enhancement and has been successfully applied in the molecular logic gate, determination of HClO in environmental water samples, and portable HClO detection.

Abstract:Hypochlorous acid (HOCl), one of the physiologically important reactive oxygen species (ROS), is engaged in a diverse variety of biological and pathophysiological functions. Excessive HOCl buildup can cause a variety of illnesses, therefore, the identification of events in real-time of abnormal levels of exogenous and endogenous HOCl in biological settings is extremely important. In the present paper, we reported a new near-infrared emissive fluorescent probe D-HBT-HOCl for the detection and bioimaging of HOCl in live cells. With regard to the construction of D-HBT-HOCl, we combined the dicyanoisophorone motif with a 2-aminobenzenethiol unit, resulting in the near-infrared emission wavelength of 700 nm and an ultra-large Stokes shift of 180 nm, which potentially endowed D-HBT-HOCl with superiority in biological imaging. In addition, the N, N-dimethylthiocarbamate served as an efficient recognition group for HOCl as well as a fluorescence quenching group in the probe. The results of the experiments showed that the newly designed probe exhibited excellent sensing capacity, including outstanding selectivity, a high signal-to-noise ratio, and a rapid response time for the detection of HOCl. What's more, this probe was further effectively leveraged for the imaging of both extrinsic and intrinsic HOCl in live cells, demonstrating its versatility, and possessed the potential to be used to further investigate the physiological and pathological activity of HOCl in biological systems.

Abstract:Herein, a red-emitting fluorescent probeDM-BDP-OClcontaining apara-DMTC benzyl pyridinium moiety at themesoposition of BODIPY as self-immolative portion for the detection of HOCl was designed and synthesized.DM-BDP-OClexhibited excellent specificity and a fast response for HOCl beyond other ROS/RNS. It was used for the accurately measurable detection of HOCl with a linear range from 0 mu M to 50 mu M, and the detection limit for HOCl reached 60 nM. Moreover, the probe could directly monitor fluctuations of exogenous and endogenous HOCl in living HeLa and RAW 264.7 cells. This work provided a powerful and convenient imaging tool for probing pathological and physiological actions of HOCl.

Abstract:As a significant reactive oxygen species (ROS), hypochlorous acid (HClO) plays a key role in the human immune system, ensuring the normal function of the organism. However, abnormal HClO levels have been linked to immune system disorders, neurodegenerative damage, and cancer. Therefore, rapid, and sensitive monitoring of HClO is important for the diagnosis of certain diseases. Here, we designed a probe XL for the detection of HClO in living cells, which is a mixture of phenothiazine and semi-cyanine. The probe XL is highly sensitive, selective and has an ultra-fast (<10 seconds) response to HClO in solution. The detection limit of probe XL for HClO was as low as 28 nM, and we also successfully used the test paper prepared with probe XL for the colorimetric fluorescence detection of HClO in real water samples. In addition, probe XL showed good response to both endogenous and exogenous HClO in cells. Finally, we successfully used probe XL for imaging HClO in zebrafish.

Abstract:Hypochlorite (ClO − ) is recognized as a bioactive substance that plays a crucial role in various physiological and pathological processes. The increase of ClO − content in cells is a key factor in the early atherosclerosis lesions, which are closely linked to cardiovascular and cerebrovascular diseases. Therefore, the development of an efficient and sensitive method for detecting hypochlorite in tap water, serum, and living cells, including animal model in vivo is of paramount importance. In this study, a novel fluorescent probe (Cy-F) based on the cyanine group was designed for the specific detection of ClO − , demonstrating exceptional selectivity, high sensitivity, and rapid response. The probe successfully detected ClO − in tap water and serum with a limit of detection (LOD) of 2.93 × 10 −7 M, showcasing excellent anti-interference capabilities. Notably, the probe exhibited good biocompatibility, low biological toxicity, and proved effective for detecting and analyzing ClO − in live cells and zebrafish. This newly developed probe offers a promising approach and valuable tool for detecting ClO − with biosafety considerations, paving the way for the design of functional probes tailored for future biomedical applications.

Abstract:Hypochlorous acid (HOCl) as a biologically important reactive oxygen species (ROS) plays crucial roles in the physiological and pathological processes. It is thus meaningful to explore an effective method for detecting basal HOCl in living systems. We herein report a large Stokes shift (about 115 nm) fluorescent probe LSS-HA for highly specific and ultrasensitive detection of HOCl with excellent water-solubility. Probe LSS-HA exhibited a remarkable fluorescence enhancement ((similar to) 227-fold) due to the adoption of high-quantum-yield fluorophore in aqueous solution. Besides, probe LSS-HA showed ultrasensitivity (detection limit is 3.30 nM) and fast response ( < 5 s) for HOCl. Furthermore, probe LSS-HA was successfully applied for imaging basal HOCl in living cells and zebrafish.

Abstract:A novel two-photon fluorescent probe () for hypochlorite (ClO(-)) has been developed based on specific ClO(-)-promoted oxidation of a C[double bond, length as m-dash]N bond. The probe shows linear fluorescence responses to ClO(-) with 23.5-fold enhancement. Two-photon fluorescence detection signals of to ClO(-) were observed with high selectivity and sensitivity. Abstract:Hypochlorite, as an important reactive oxygen species (ROS), plays a vital role in many physiological and pathological processes, but an excess concentration of hypochlorite (ClO-) may become toxic to humans and cause disease. Hence, the selective and rapid detection of hypochlorite (ClO-) is necessary for human safety. Here, we report a novel near-infrared (NIR) fluorescence "turn-on" and highly selective benzophenoxazinium chloride-based fluorescent probe, BPH (benzophenoxazinium dihydroxy benzaldehyde), for hypochlorite detection. Due to hypochlorite-induced vicinal diol oxidation to the corresponding ortho benzoquinone derivative, the photoinduced electron transfer (PET) process, which was operating from vicinal diol to the benzophenoxazinium chloride receptor moiety, was suddenly inhibited, as a result of which strong NIR fluorescence "turn-on" emission was observed. The detection limit of BPH was found to be 2.39 × 10-10 M, or 0.23 nM. BPH was successfully applied for exogenous and endogenous hypochlorite detection in live MDA-MB 231 cells.

Abstract:Due to the importance of hypochlorous acid (HClO) in biological and industrial, development of fluorescent probes for HClO has been an active research area. Here, a new red-emitting ratiometric fluorescent probe (P) was synthesized and well defined characterization via NMR, HR-MS, and fluorescence spectrum, which serves as a selective and sensitive probe for ClO− group. The probe showed a ratiometric fluorescent response to hypochlorite at the emission intensities ratio (I480/I612) increasing from 0.28 to 27.46. The emission intensities ratio (I480/I612) was linearly enhanced (I480/I612 = 0.064 X + 0.096) with the ClO− concentration range from 1 to 30 μM. The detection limitation for ClO− in aqueous solution is 0.47 μM. Moreover, this biocompatible red-emitting ratiometric fluorescent probe was utilized to the fluorescence imaging of ClO− in living cells and Zebrafish.

Abstract:Hypochlorite (ClO−) is one of the important reactive oxygen species (ROS), which playing an irreplaceable influence towards various physiological activities, it is significant to establish an analytical tool for identifying and monitoring of ClO−. Here, a ratiometric fluorescent probe MYQ with an extremely large emission shift containing 1,8-naphthalimide and benzopyran fluorophores was designed for the detection of ClO−. The probe MYQ presented red fluorescence emission at 655 nm, however, in the presence of ClO−, it exhibited strong blue fluorescence at 462 nm with fluorescent color change from red to blue. Meanwhile, MYQ showed excellent selectivity and selectivity toward ClO− over other common anions in the PBS (60% CH3OH, pH = 7.4) system. The limit of detection for ClO− was calculated to be 25.3 nΜ. The presumed mechanism for detecting ClO− was verified by mass spectrometry and liquid chromatography. Furthermore, the potential practicability of MYQ was demonstrated by strip test and cell imaging.

Abstract:The development of simple techniques for monitoring hypochlorous acid (HOCl) in living cells is urgently needed to disclose the important roles of HOCl in biological systems. Although a number of fluorescent probes have been exploited to detect HOCl, most of them are easily influenced by other competing reactive oxygen species (ROS) and reactive nitrogen species (RNS), and disturbed by many factors including variabilities in excitation, probe distribution and environmental conditions due to the recognition of HOCl based on the fluorescence changes in single wavelength. Herein, in this work, we present a simple fluorescent probe AETU-HOCl containing a new recognition group (2-amino-ethyl)thiourea (AETU) for the ratiometric detection of HOCl in living cells. Probe AETU-HOCl exhibits outstanding selectivity for HOCl over other ROS/RNS at higher concentrations (0.5 mM, 100 equiv.). More importantly, the rapid response (1 min) and high sensitivity (a detection limit of 3.9 nM) allow it to successfully visualize the intracellular HOCl levels by the ratiometric imaging. The present study would offer a simple, ratiometric, rapid, and ultrasensitive assay for the accurate determination of HOCl in the biological systems.

Abstract:The probe (XA‐CN) showed pH dependent deep‐red fluorescence and reached to the maximum photoluminescent intensity at pH 9. The 643 nm‐centered emissions disappeared at the presence of hypochlorite (ClO−). This is because the group ‐CN and the hydroxyl group was oxidized into aldehyde or carbonyl group by ClO−. As an oxidant, deodorant and bleaching agent, the hypochlorous acid (HClO) and hypochlorite (ClO−) are widely used in corrosion inhibitors, textile dyes, pharmaceutical intermediates and in our daily lives. However, excess usage or aberrant accumulation of ClO− leads to tissue damage or some diseases and even cancer. Therefore, it is necessary to develop a fluorescent probe that specifically identifies ClO−. In this article, we synthesized a deep‐red xanthene‐based fluorescent probe (XA‐CN). The strong electron deficient group dicyano endows the probe XA‐CN deep‐red fluorescent emission with high solubility, selectivity and sensitivity for ClO− detection. Studies showed that the probe demonstrated turn‐off fluorescence (643 nm) at the presence of ClO− in dimethylsulfoxide/phosphate‐buffered saline 1:1 (pH 9) solution with a limit of detection of 1.64 μM. Detection mechanism investigation revealed that the electron deficient group ‐CN and the hydroxyl group was oxidized into aldehyde or carbonyl groups at the presence of ClO−, resulting ultraviolet‐visible absorption of the probe blue shifted and turned‐off fluorescence. Furthermore, XA‐CN was successfully used for the detection of ClO− in tap water samples.

Abstract:As a vital reactive oxygen species (ROS), endogenous hypochlorous acid (HOCl) deeply engaged in masses of pathophysiological processes owing to its high reactivity, thus the detection of HOCl is awfully valuable for elucidating HOCl-related pathophysiology mechanism. Despite the extensive development of outstanding HOCl fluorescent probes so far, very few of them could be applied for monitoring HOCl in the pure water system. Herein, we present a novel rhodol-based fluorescent probe 6 for the unambiguous detection of HOCl in pure water, which was effectively designed and synthesized by using the spirocyclic hydrazide group as the HOCl sensitive site and introducing two hydroxyethyl groups for obtaining high hydrophilicity into the rhodol skeleton, a unique rhodamine structure. Compared to the control compound 5 without hydrophilic groups only capable to be used in the aqueous system containing organic solvent, 6 achieves the detection of HOCl not only in the above regular system but also in pure water, indicating that the introduction of hydrophilic hydroxyethyl group is an effective and reasonable approach to achieve the broader-spectrum detection of HOCl. More importantly, different with 5, highly water-soluble 6 regardless of dissolving in the regular system or pure water system can achieve the visual monitoring of HOCl with prominent sensitivity and high spatiotemporal resolution in living HepG2 cells and zebrafish, confirming the robust imaging ability of our probe 6 to HOCl in complex biosystem.

Abstract:Judiciously designed cyanine based fluorogenic probe (L) can exhibit interesting solvent polarity induced isomerization. The probe displayed a highly selective TURN-ON fluorescence response towards hypochlorite among various reactive oxygen species (ROS) and analytes in a mixed aqueous medium. The sensing process was attributed to the formation of a probe-OCl adduct which results in restricting the donor-acceptor extended conjugation. The detection limit was found to be as low as 3 \(\upmu \hbox {M}\). The proposed sensing mechanism is supported by mass spectrometric analysis and HPLC study.

Abstract:The development of selective fluorescent probes for rapid and sensitive detection of hypochlorite anion (OCl-) is of great importance for biomedical researchers to investigate the detailed functions and mechanisms of OCl- in living systems. Herein, based on the internal charge transfer (ICT) mechanism, employing 2-dicyanomethylene-3-cyano-4,5,5-trimethy1-2,5-dihydrofuran (TCF) fluorophore, we developed a novel visual and far-red fluorescent dual-channel probe TCF-OCl for the rapid and sensitive detection of OCl- in aqueous solution and living cells. TCF-OCl exhibits high OCl--selectivity even in the presence of other reactive oxygen species (ROS), which may be ascribed to the excellent reactivity of the arylboronate recognition moiety toward OCl-. ICT-based fluorescent probe TCF-OCl displays a considerable absorption spectrum (at 570 nm) enhancement and the color changes from faint yellow to red upon addition of OCl-. In addition, the results showed that TCF-OCl could quantitatively detect OCl- in the range of 0 to 50 mu M with the detection limit of 0.4 mu M by visual and fluorescent spectrometry methods. Importantly, TCF-OCl was successfully applied to the fluorescence imaging of OCl- levels in living cells, and it is expected to be a useful chemical tool for investigating the detailed functions and mechanisms of OCl- in living systems. Abstract:Hypochlorite (ClO − ), a weakly acidic reactive oxygen species, plays a crucial role in antibacterial and anti-inflammatory defense mechanisms. However, elevated levels of ClO − or disruptions in endogenous sites can lead to tissue damage and various diseases including cardiovascular disease, neuronal degeneration, and arthritis. To address this, the development of a specific fluorescent probe with a built-in self-calibration ratio mode for the analysis and biological imaging of ClO − is essential. In this study, a cyanine-based fluorescent probe (Cy-H) was designed for ratiometric fluorescent detection of ClO − , utilizing its aggregation behavior as a novel approach in this field. Upon exposure to ClO − , the phenolic hydroxyl group in probe Cy-H was oxidized into benzoquinone, leading to the formation of cyanine products that displayed a strong tendency to aggregate. As a result, the maximum emission peak of the probe shifted from 700 nm to 485 nm. Notably, a linear relationship was observed between the peak intensity ratio (I 485 /I 700 ) and the concentration of hypochlorite, with a limit of detection (LOD) of 0.49 μM. Furthermore, this probe was successfully employed for imaging analysis of hypochlorite in living cells and zebrafish.

Abstract:Hypochlorite (ClO − ) is a well-known redox regulator among reactive oxygen species and plays a crucial role in bacterial disinfection and body immunity. Therefore, there is a high demand for an analytical technique to monitor ClO − under physiological conditions. Herein, a novel fluorescence probe, RES-DPPT , was developed for detecting ClO − . This probe employs diphenylphosphinothioyl unit as the recognition group and resorufin as the fluorescent platform. RES-DPPT has high sensitivity and specificity, rapid response, and wide pH tolerance for monitoring ClO − . Abstract:A novel near-infrared-emitting aza-BODIPY-based fluorescent probe with two tellurium atoms at two upper benzyl rings has been prepared and explored for its fluorescent sensing properties towards hypochlorous acid/hypochorite (HClO/ClO−), which showed high selectivity and absolutely fluorescent "turn-on" phenomenon at 738 nm. The fluorescence of this probe was sufficiently quenched due to photoindued electron transfer by two tellurium atoms. Upon exposure to HClO/ClO−, a strong near-infrared emission at 738 nm appeared with fluorescence quantum yields changing from 0 to 0.11. This remarkable fluorescence change was ascribed to the oxidation of both electron-rich tellurium atoms. The detection limit of this probe towards HClO/ClO− was calculated to 0.09 μM in acetonitrile aqueous solution by the linear fluorescence change at 738 nm in the HClO/ClO−-concentration range of 0–30 μM. Interestingly, this probe was found to be applicable in a broad pH range (2–10). Meanwhile, the oxidized probe could be further responsive to biothiols with substantial fluorescence disappearance. The bioimaging experiments in RAW264.7 cells showed the appearance of intracellular near-infrared fluorescence after addition of HClO/ClO− and PMA, and the fluorescence could also be reversed to be silenced by further introduction of GSH, confirming its potential application for exogenous and endogenous detection of HClO/ClO− in living cells.
