Abstract:Five diets that contained fresh squid meat as the basic constituent and were supplemented with different amounts of highly unsaturated fatty acids (HUFA) and astaxanthin were fed to pond-reared Penaeus monodon broodstock. Diet A was sole squid meat. Diets B and C were supplemented with astaxanthin 50 and 100 mg kg(-1) respectively. Diets D and E were supplemented with HUFA 5 and 10 g kg(-1) and astaxanthin 50 mg kg(-1) respectively. The result showed that the group fed diet E had the best reproductive performance in all experimental groups. It had a higher proportion of spawns (71.5%), spawning rate (0.047), a shorter latency period (7.7 +/- 0.3 d), higher absolute fecundity (x 10(3)) (361.6 +/- 5.5) and egg production/female (x 10(3)) (597.0 +/- 18.0) than all the other experimental groups. The fatty acid composition in broodstock diets strongly affected the tissue and fecundity of broodstock. Good correlations between the content of 20:4n-6 in eggs and the fecundity (r(2)=0.6109) and egg production (r(2)=0.9876) of broodstock were found. On the other hand, 22:6n-3 and DHA/EPA ratio was negatively correlated with the fecundity of broodstock (r(2)=0.5362, 0.8702 respectively). The result also showed that the balance between n-3 and n-6 fatty acid families, total polyunsaturated fatty acids and total saturated fatty acid and 20:5n-3 (EPA) and 22:6n-3 (DHA) may play vital roles in maturation and reproductive performance of P. monodon broodstock.
Identification of Gonadal Associated Genes in Black Tiger Shrimp ( Penaeus Monodon ) Using Transcriptome Analysis and High‐throughput Sequencing

Abstract:Black tiger shrimp (Penaeus monodon) is a commercially important species. It is reported to have low reproductive maturation rates in captivity. To improve the production of this species via sustainable aquaculture, genetic techniques are required to improve its maturation rates. However, to date, reports on P. monodon reproduction at a transcriptional level are limited. To address this, we conducted transcriptome sequencing of the ovaries and testes of P. monodon using high-throughput sequencing to identify sex-related pathways and genes. A total of 75.74 Gb of clean data were generated from nine gonadal tissue libraries. We obtained 62,276 unigenes with a mean length of 1452 bp and N50 of 2423 bp. A total of 11,411 significantly differentially expressed genes (DEGs) were identified between the testis and developed ovaries. Enrichment analysis of the DEGs identified 415 significantly enriched GO terms and 106 enriched KEGG pathways. Genes related to the P. monodon ovaries were enriched in the steroid biosynthesis, mRNA surveillance, nucleocytoplasmic transport, oxidative phosphorylation and DNA replication pathways. Five upregulated and five downregulated ovarian genes were confirmed using RT-PCR. This investigation has thus identified important sex-related functional genes, GO terms and pathways in P. monodon. In particular, we identified several important genes and pathways related to gonadal development, such as GnRH/GnRHR-like, Hsp90, PCNA, cathepsin L, Wnt signalling pathway and ovarian steroidogenesis pathway, which provide important insights into the mechanism analysis and sex regulation of shrimp gonadal development. The results will provide reference data for future research into the reproductive biology of shrimp, which will aid in improved black tiger shrimp breeding in captivity.
A Comprehensive Study on Nutritional Quality, Physiological Enzyme Activity and Genetic Diversity in Six Populations of Penaeus Monodon

Abstract:Penaeus monodon is a crucial economic crustacean globally and a focal point of aquatic research. Germplasm resources provide a diverse food source for humans, meeting various nutritional needs; they also maintain plant and animal gene banks, enhancing resilience, adaptability, and promoting ecological balance in terms of genetic diversity. To accurately assess the germplasm resources of P. monodon, we conducted a comprehensive investigation. We analyzed muscle characteristics, nutrient content, amino acids, fatty acids, and genetic diversity using whole-genome sequencing in six P. monodon groups. The results showed that the crude protein content of the six groups of P. monodon was 21.17–24.60 g/100 g, crude fat content was 0.60–0.80 g/100 g, essential amino acid content was 5.44–6.41 g/100 g, total amino acid content was 15.68–18.63 g/100 g, and total fatty acid content was 354.78–587.43 mg/100 g. Genetic differentiation index (Fst) values ranged from 0.029 to 0.604 for six groups. PmSZ has a good genetic condition. There was no significant difference in crude fat in conventional nutrients, and the difference in biochemical indexes T-AOC, CAT, and SOD was not significant (P > 0.05). The noteworthy point is that the analysis of genetic diversity reveals a moderate level of genetic diversity. PmDF and PmZH exhibit the highest degree of differentiation, while PmRP and PmZH show the lowest degree of differentiation. Our survey results establish a critical foundation for comprehending and harnessing the germplasm resources essential for selectively breeding robust P. monodon populations.

Abstract:To explore the effect of concentrated dephenolic cottonseed protein (CDCP) partly instead of fish meal on the non-specific immune of Penaeus monodon, we designed a single factor test to study the four types of non-specific immunity of P. monodon hepatopancreas after acute salinity stress. A total number of 5,400 P. monodon were selected with an initial body weight of 1.76±0.15g and randomly divide into 6 groups (3 replicates per group, 900 tails per replicate). Shrimp were feed with two protein level diets (Diet A and Diet B). After 8 weeks of feeding, P. monodon were randomly selected with an average weight of 14.71±1.06g in each feed group, and subjected to acute salinity stress of 10ppt and 20ppt after temporary rearing, and the heat shock protein 70 (HSP70), Na+-K+-ATPase, cortisol and the content of alkaline phosphatase (ALP) were measured. The results of the study showed that when the CDCP partially replaced fish meal of P. monodon diet, the hepatopancreas HSP70, and the contents of Na+-K+-ATPase and cortisol were significantly higher than those of the control group. In the acute salinity change, there was no significant difference in the survival rate of P. monodon at 48h under the two salinity stresses, and the four immune indexes showed different trends. The expression level of Cortisol was more prominent than that of HSP70, which could better reflect the stress degree of P. monodon. Most of the indicators showed a trend of rising and then falling and then rising compared with the original level, and the changes of the four indicators of hepatopancreas in the Diet B group were significantly higher than those in the Diet A group at most time point of the two salinities.

Abstract:Sea cucumber peptides (SCPs) have been proven to have many active functions; however, their impact on testosterone synthesis and the corresponding mechanism are not yet clear. This study attempts to explore the effects of SCPs on sex hormone regulation in acute exhaustive swimming (AES) male mice and the possible mechanisms. In the present study, SCP intervention significantly prolonged exhaustive swimming time and reduced exercise metabolite accumulation. The reproductive ability-related parameters including penile index, mating ability, testicular morphology, and sperm storage were dramatically improved by SCP intervention. Notably, SCPs markedly reversed the AES-induced decrease in serum testosterone (T), estradiol (E2), and follicle-stimulating hormone (FSH) levels. Moreover, treatment with a high dose of SCP (0.6 mg per g bw) significantly enhanced the expression of testosterone synthesis-related proteins in testis, meanwhile markedly increasing the gene expression of StAR, Hsd17b3, Hsd17b2, Ldlr, and Cyp19a1. Serum metabolomics results indicated that SCP intervention notably upregulated the expression of 1-stearoyl-2-arachidonoyl-sn-glycerol but downregulated the concentrations of succinate and DL-lactate. Furthermore, serum metabolomics combined with testicular transcriptome, western blot, and correlation analyses demonstrated that SCPs may regulate testosterone synthesis via the Ca2+/PKA signaling pathway. This study indicated that the SCP could be a potential dietary supplement to improve the symptoms of decreased sex hormones related to exercise fatigue.

Abstract:The decrease of seawater pH can affect the metabolism, acid-base balance, immune response and immunoprotease activity of aquatic animals, leading to aquatic animal stress, impairing the immune system of aquatic animals and weakening disease resistance, etc. In this study, we performed high-throughput sequencing analysis of the hepatopancreas transcriptome library of low pH stress penaeus monodon, and after sequencing quality control, a total of 43488612-56271828 Clean Reads were obtained, and GO annotation and KEGG pathway enrichment analysis were performed on the obtained Clean Reads, and a total of 395 DEGs were identified. we mined 10 differentially expressed and found that they were significantly enriched in the Metabolic pathways (ko01100), Biosynthesis of secondary metabolites (ko01110), Nitrogen metabolism (ko00910) pathways, such as PIGA, DGAT1, DGAT2, UBE2E on Metabolic pathways; UGT, GLT1, TIM genes on Biosynthesis of secondary metabolites; CA, CA2, CA4 genes on Nitrogen metabolism, are involved in lipid metabolism, induction of oxidative stress and inflammation in the muscular body of spot prawns. These genes play an important role in lipid metabolism, induction of oxidative stress and inflammatory response in the muscle of the shrimp. In summary, these genes provide valuable reference information for future breeding of low pH-tolerant shrimp.
Dynamics of fatty acid regulatory genes during ovarian development in Penaeus monodon

Abstract:The delay in ovarian maturation in farmed black tiger shrimp Penaeus monodon has resulted in the widespread practice of feeding broodstock with the polychaetes Perinereis nuntia and their unilateral eyestalk ablation. Although this practice alters fatty acid content in shrimp ovaries and hepatopancreas, its effects on fatty acid regulatory genes have yet to be systematically examined. Here, microarray analysis was performed on hepatopancreas and ovary cDNA collected from P. monodon at different ovarian maturation stages, revealing that 72 and 58 genes in fatty acid regulatory pathways were differentially expressed in hepatopancreas and ovaries respectively. Quantitative real-time PCR analysis revealed that ovarian maturation was associated with higher expression levels of acetyl-CoA acetyltransferase, acyl-CoA dehydrogenase, acyl-CoA oxidase 3 and long-chain fatty acid transport protein 4 in hepatopancreas, whereas the expression levels of 15 fatty acid regulatory genes were increased in shrimp ovaries. To distinguish the effects of different treatments, transcriptional changes were examined in P. monodon with stage 1 ovaries before polychaete feeding, after one-month of polychaete feeding and after eyestalk ablation. Polychaete feeding resulted in lower expression levels of enoyl-CoA hydratase and acyl-CoA synthetase medium-chain family member 4 , while the expression level of phosphatidylinositide phosphatase SAC1 was higher in shrimp hepatopancreas and ovaries. Additionally, eyestalk ablation resulted in a higher expression level of long-chain fatty acid-CoA ligase 4 in both tissues. Together, our findings describe the dynamics of fatty acid regulatory pathways during crustacean ovarian development and provide potential target genes for alternatives to eyestalk ablation in the future.

Abstract:We analysed fatty acids (FAs) composition and the potential functions of FAs in hepatopancreas and testis of freshwater caridean shrimp Macrobrachium nipponense during testis development. The hepatopancreas presented a high unsaturated FA content of about 40%, whereas the testis showed a high level of saturated fatty acids (SFAs) with a decreasing content of 97-86% of total FAs during maturation. Several specific FAs in testis showed significant changes throughout maturation. Among SFAs, C14:0 contributed with a relatively constant high proportion of about 22%, whereas C16:0 and C18:0 contents were also at high levels but showed significant increases during maturation. Representatives of highly unsaturated fatty acids (HUFAs), such as C18:1n-9, C18:2n-6, C20:5n-3 (EPA), C20:4n-6 (AA) and C22:6n-3 (DHA) showed significant increases despite their low proportion in the gonad. These results suggest that a high content of HUFAs in hepatopancreas may represent a reservoir for sperm production. The FA C14:0 may provide a particular function. The variations of the FA profile in testis at different developmental stages reveals that C18:1n-9, C18:2n-6, C20:5n-3, C20:4n-6 and C22:6n-3 may play important roles in spermiogenesis.

Abstract:Testis maturation, germ cell development and function of sperm, are related to lipid composition. Phosphatidylcholines (PCs) play a key role in the structure and function of testes. As well, increases of polyunsaturated fatty acids (PUFA) and highly unsaturated fatty acids (HUFA), especially arachidonic acid (ARA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA) are essential for male fertility. This study is the first report to show the composition and distribution of PCs and total fatty acids (FAs) in three groups of seminiferous tubules (STs) classified by cellular associations [i.e., A (STs with mostly early germ cells), B (STs with mostly spermatids), and C (STs with spermatozoa)], in three morphotypes of Macrobrachium rosenbergii, [i.e., small male (SM), orange claw male (OC), and blue claw male (BC)]. Thin layer chromatography exhibited levels of PCs reaching maxima in STs of group B. Imaging mass spectrometry showed remarkably high signals corresponding to PC (16:0/18:1), PC (18:0/18:2), PC (18:2/20:5), and PC (16:0/22:6) in STs of groups A and B. Moreover, most signals were detected in the early developing cells and the intertubular area, but not at the area containing spermatozoa. Finally, gas chromatography-mass spectrometry indicated that the major FAs present in the testes were composed of 14:0, 16:0, 17:0, 18:0, 16:1, 18:1, 18:2, 20:1, 20:2, 20:4, 20:5, and 22:6. The testes of OC contained the greatest amounts of these FAs while the testes of BC contained the least amounts of these FAs, and there was more EPA (20:5) in the testes of SM and OC than those in the BC. The increasing amounts of FAs in the SM and OC indicate that they are important for spermatogenesis and spermiogenesis. This knowledge will be useful in formulating diets containing PUFA and HUFA for prawn broodstocks in order to improve testis development, and lead to increased male fecundity.

Abstract:Gonadal development is a prerequisite for the reproductive success of an organism, and might be affected by environmental factors such as emergent pollutants. Although marine crustaceans are threatened by ubiquitous emergent pollutants such as microplastics (MPs) and bisphenol A (BPA) under realistic scenarios, studies about the impacts of these pollutants on the gonadal development of crustacean species are rare. In this study, the effects of MPs and BPA, alone or in combination, on gonadal development were investigated in whiteleg shrimp (Litopenaeus vannamei). The results obtained demonstrated that whiteleg shrimp exposed to MPs and BPA had significantly smaller gonad-somatic index (GSI) and an obvious delay in the gonad developmental stage. In addition, exposure of whiteleg shrimp to pollutants tested resulted in a reduction in the oxygen consumption rate, elevation in the ammonia excretion rate, decline in the O: N ratio, and downregulation in the expression of metabolism-related genes, indicating significant disruptions of shrimp metabolism by the pollutants. Furthermore, in addition to a few exceptions, both the in vivo contents of gonadal development-related hormones (GIH and MIH) and the expression of genes encoding regulatory hormones (GIH, MIH, and CHH) were upregulated by the exposure of whiteleg shrimp to the pollutants investigated, suggesting a significant obstruction of endocrine regulation. Moreover, MP-BPA coexposure was shown to be more toxic to whiteleg shrimp than the corresponding single exposures and significantly greater amount of BPA accumulated in the gonads (both testis and ovaries) of shrimp with the coexistence of MPs, which may be caused by the Trojan horse effect and summation of the toxic impacts on common targets. In general, the data obtained in this study demonstrated that MPs and BPA at environmentally realistic concentrations significantly inhibited the gonadal development of whiteleg shrimp probably by interfering with metabolism and disrupting endocrine regulation.
Effects of arachidonic acid supplementation in maturation diet on female reproductive performance and larval quality of giant river prawn (Macrobrachium rosenbergii)

Abstract:The giant river prawn (Macrobrachium rosenbergii) is one of the most farmed freshwater crustaceans in the world. Its global production has been stalling in the past decade due to the inconsistent quality of broodstock and hatchery-produced seeds. A better understanding of the role of nutrition in maturation diets will help overcome some of the production challenges. Arachidonic acid (20:4 n-6, ARA) is a fatty acid precursor of signaling molecules important for crustacean reproduction, prostaglandins E and F of the series II (PGE2 and PGF2α), and is often lacking in maturation diets of shrimp and prawns. We examined the effects of ARA in a combination of different fish oil (FO) and soybean oil (SO) blends on females’ reproductive performance and larval quality. Adult females (15.22 ± 0.13 g and 11.12 ± 0.09 cm) were fed six isonitrogenous and isolipidic diets containing one of two different base compositions (A or B), supplemented with one of three levels of Mortierella alpine-derived ARA (containing 40% active ARA): 0, 1 or 2% by ingredient weight. The two base diets differed in the percentages of (FO and SO with diet A containing 2% SO and 2% FO and diet B containing 2.5% SO and 1.5% FO, resulting in differences in proportional contents of dietary linoleic acid (18:2n-6, LOA) and docosahexaenoic acid (22:6n-3, DHA)). After the eight-week experiment, prawns fed diet B with 1 and 2% ARA supplement (B1 and B2) exhibited the highest gonadosomatic index (GSI), hepatosomatic index (HSI), egg clutch weight, fecundity, hatching rate, number of larvae, and reproductive effort compared to those fed other diets (p ≤ 0.05). Larvae from these two dietary treatments also had higher tolerance to low salinity (2 ppt). The maturation period was not significantly different among most treatments (p ≥ 0.05). ARA supplementation, regardless of the base diet, significantly improved GSI, HSI, egg clutch weight and fecundity. However, the diets with an enhanced ARA and LOA (B1 and B2) resulted in the best reproductive performance, egg hatchability and larval tolerance to low salinity. These dietary treatments also allow for effective accumulation of ARA and an n-3 lcPUFA, DHA in eggs and larvae.

Abstract:The present study was undertaken to determine the effects of polyunsaturated fatty acids (PUFAs) on histomorphological structure of liver, developmental stages of ovary, serum calcium (Ca2+) level, sperm quality and spontaneous spawning performance of striped gourami, Colisa fasciatus. Fishes were collected from the Brahmaputra River, Mymensingh, Bangladesh. Treated group was fed with supplemental diet enriched with 1 % squid extracted phospholipids for 3 months while the control group was given the same feed except phospholipids. For histomorphology of liver and ovaries, samples were collected on monthly basis. Treated group exhibited higher gonadal maturation resulted in spontaneous spawning compared to control. During spawning time, normal morphological structure of hepatocytes with lipid granules was observed in the liver of treated group, whereas less lipid granules with altered histological appearance were noticed in control group. Moreover, serum Ca2+ concentration was measured significantly higher (P < 0.01) in PUFA-treated female in contrast with control one. Again, sperm quality was determined by staining the sperm with trypan blue dye. Live sperm depicted transparent appearance in contrast to the dead one that retained black spots. The amount of live sperm was significantly higher (98.86 ± 0.87 %) in PUFA-treated males than that of control (90.58 ± 0.67 %). Therefore, the present study clearly indicated these enhanced spawning performances of C. fasciatus owing to PUFAs supplementation in diet.

Abstract:The Pacific whiteleg shrimp (Penaeus vannamei) is a highly significant species in shrimp aquaculture. In the production of shrimp larvae, noticeable variations in the reproductive capacity among female individuals have been observed. Some females experience slow gonadal development, resulting in the inability to spawn, while others undergo multiple maturations and contribute to the majority of larval supply. Despite numerous studies that have been conducted on the regulatory mechanisms of ovarian development in shrimp, the factors contributing to the differences in reproductive capacity among females remain unclear. To elucidate the underlying mechanisms, this study examined the differences in the ovarian characteristics between high and low reproductive bulks at different maturity stages, focusing on the cellular and molecular levels. Transmission electron microscopy analysis revealed that the abundance of the endoplasmic reticulum, ribosomes, mitochondria, and mitochondrial cristae in oocytes of high reproductive bulk was significantly higher than that of the low reproductive bulk in the early stages of ovarian maturation (stages I and II). As the ovaries progressed to late-stage maturation (stages III and IV), differences in the internal structures of oocytes between females with different reproductive capacities gradually diminished. Transcriptome analysis identified differentially expressed genes (DEGs) related to the mitochondria between two groups, suggesting that energy production processes might play a crucial role in the observed variations in ovary development. The expression levels of the ETS homology factor (EHF) and PRDI-BF1 and RIZ homology domain containing 9 (PRDM9), which were significantly different between the two groups, were compared using qRT-PCR in individuals at different stages of ovarian maturation. The results showed a significantly higher expression of the EHF gene in the ovaries of high reproductive bulk at the II and IV maturity stages compared to the low reproductive bulk, while almost no expression was detected in the eyestalk tissue of the high reproductive bulk. The PRDM9 gene was exclusively expressed in ovarian tissue, with significantly higher expression in the ovaries of the high reproductive bulk at the four maturity stages compared to the low reproductive bulk. Fluorescence in situ hybridization further compared the expression patterns of EHF and PRDM9 in the ovaries of individuals with different fertility levels, with both genes showing stronger positive signals in the high reproductive bulk at the four ovarian stages. These findings not only contribute to our understanding of the regulatory mechanisms involved in shrimp ovarian development, but also provide valuable insights for the cultivation of new varieties aimed at improving shrimp fecundity.

Abstract:Globally, penaeid shrimp are the most farmed and traded aquatic organisms, although they are easily susceptible to microbial pathogens. Moreover, there is a desire to increase the nutritional value of shrimp , especially the levels of n-3 polyunsaturated fatty acids (PUFAs), such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), which also possess immunomodulatory and anti-inflammatory properties. Some aquatic animals can synthesize EPA and DHA from dietary plant-sourced alpha-linolenic acid (ALA), but penaeid shrimps' ability to synthesize these n-3 PUFAs is unknown. Here, molecular biology techniques, including gas chromatography–mass spectrometry, qPCR, ELISA, etc., were used to demonstrate that exogenous ALA or Vibrio parahaemolyticus could modulate EPA and DHA levels and immune genes in Penaeus vannamei by inducing key enzymes involved in n-3 PUFAs biosynthesis, such as delta desaturases and elongation of very long-chain fatty acid (ELOVLs). Most importantly, knockdown or inhibition of ∆6 desaturase significantly decreased EPA and DHA levels and immune gene expression even with exogenous ALA treatment, consequently affecting shrimp antibacterial immunity and survival. This study provides new insight into the potential of P. vannamei to synthesize n-3 PUFAs from exogenous ALA or upon bacteria challenge, which could be leveraged to increase their nutritional content and antimicrobial immunity.

Effects of Dietary Folic Acid Supplementation on Sex Differences in Oriental River Prawn, Macrobrachium nipponense

Abstract:To better understand the sex differences in the utilization of folic acid for male and female prawns, Macrobrachium nipponense, three experimental diets with graded folic acid contents (A: 1.22 mg/kg, B: 5.44 mg/kg, and C: 10.09 mg/kg) were provided to prawns for 8 weeks. The experimental study demonstrated that prawns fed diets containing 1.22 mg/kg showed the best values on body weight gain (BWG), specific growth rate (SGR), and individual weight for both males and females. Male prawns fed above 1.22 mg/kg of dietary folic acid significantly decreased the crude protein contents in their bodies (p < 0.05), while no significant differences were observed in females among all treatments (p > 0.05). The protease activity of the hepatopancreas in females was significantly (p < 0.05) improved by a dietary level of 10.09 mg/kg of folic acid. However, the opposite trend was observed in males, with the highest protease activity observed at a dietary level of 1.22 mg/kg folic acid. Increasing dietary folic acid levels did not suppress early maturation but led to an increase in the fecundity of females. Furthermore, prawns fed with 10.09 mg/kg of folic acid exhibited improved tolerance against thermal stress in both males and females. Transcriptome analysis revealed that during thermal stress, the "Oxidative phosphorylation" and "Pantothenate and CoA biosynthesis" signaling pathways were significantly enriched in females, and the "Fatty acid biosynthesis" signaling pathway was significantly enriched in males. The results of this study preliminarily evaluate the differences between male and female M. nipponense in response to different dietary folic acid levels and are helpful in promoting the health and growth of aquaculture production of this species.

Abstract:An 8-week feeding experiment was conducted to investigate the effects of dietary n-3 highly unsaturated fatty acids (n-3 HUFAs) on growth performance, fatty acid profiles, hematological characteristics and hepatopancreatic histology in juvenile Pacific white shrimp (Litopenaeus vannamei) (initial average weight, 0.14 ± 0.01 g). Six isoproteic and isolipidic diets were formulated with graded levels of n-3 HUFA (0.24, 0.53, 0.86, 1.25, 1.58 and 1.97 % of dry matter), and the 0.24 % group was used as control group. The results revealed that shrimp fed the diet containing 0.86 % n-3 HUFA had highest weight gain (WG) and specific growth rate (SGR), and the feed conversion ratio (FCR) showed the minimum at the same level. Shrimp muscle fatty acid profiles reflected that of diet. The contents of triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) in serum decreased along with n-3 HUFA levels increasing. The activity of superoxide dismutase (SOD) in hepatopancreas was highest significantly at the level of 0.86 % n-3 HUFA group than that in other groups. The content of malondialdehyde (MDA) in hepatopancreas increased with the increasing of n-3 HUFA levels, which reached the maximum value in 1.97 % group and was significantly higher than that in control group. The levels of n-3 HUFA could obviously affect on the cell morphology and lipid deposition in hepatopancreas of L. vannamei. Results of this study suggested that shrimp fed moderate dietary n-3 HUFA could enhance growth, feed utilization, antioxidant capacity and reduces excess lipid deposition.

Abstract:Sexually dimorphic traits such as growth and body size are often found in various crustaceans. Methyl farnesoate (MF), the main active form of sesquiterpenoid hormone in crustaceans, plays vital roles in the regulation of their molting and reproduction. However, understanding on the sex differences in their hormonal regulation is limited. Here, we carried out a comprehensive investigation on sexual dimorphic responses to MF in the hepatopancreas of the most dominant aquacultural crustacean-the white-leg shrimp (Litopenaeus vannamei). Through comparative transcriptomic analysis of the main MF target tissue (hepatopancreas) from both female and male L. vannamei, two sets of sex-specific and four sets of sex-dose-specific differentially expressed transcripts (DETs) were identified after different doses of MF injection. Functional analysis of DETs showed that the male-specific DETs were mainly related to sugar and lipid metabolism, of which multiple chitinases were significantly up-regulated. In contrast, the female-specific DETs were mainly related to miRNA processing and immune responses. Further co-expression network analysis revealed 8 sex-specific response modules and 55 key regulatory transcripts, of which several key transcripts of genes related to energy metabolism and immune responses were identified, such as arginine kinase, tropomyosin, elongation of very long chain fatty acids protein 6, thioredoxin reductase, cysteine dioxygenase, lysosomal acid lipase, estradiol 17-beta-dehydrogenase 8, and sodium/potassium-transporting ATPase subunit alpha. Altogether, our study demonstrates the sex differences in the hormonal regulatory networks of L. vannamei, providing new insights into the molecular basis of MF regulatory mechanisms and sex dimorphism in prawn aquaculture.
Dietary polyunsaturated fatty acids affect volume and metabolism of Drosophila melanogaster sperm

Abstract:Dietary fatty acids can accumulate in sperm and affect their function in vertebrates. As Drosophila melanogaster shares several pathways of lipid metabolism and shows similar lipid-dependent phenotypes but lacks some hormones that in vertebrates regulate lipid metabolism, there is currently no clear prediction as to how dietary fatty acids affect the sperm of D. melanogaster. We manipulated the amount and identity of dietary polyunsaturated fatty acids (PUFA) in the food of D. melanogaster males (a treatment known to affect membrane fluidity) and measured changes in sperm parameters. We found that (a) males reared on food containing PUFA-rich, plant-derived lipids showed a slower increase in sperm volume over male age compared to males reared on yeast-derived lipid food which is richer in saturated fatty acids. (b) The resistance of sperm membrane integrity to osmotic stress was not altered by dietary lipid treatment, but (c) food containing yeast-derived lipids induced a 46% higher in situ rate of production of reactive oxygen species in sperm cells. These findings show that dietary lipids have similar effects on sperm parameters in Drosophila as in vertebrates, affect some, but not all, sperm parameters and modulate male reproductive ageing. In concert with recent findings of sex-specific seasonal variation of diet choice in the wild, our results suggest a substantial dietary impact on the dynamics of male reproduction in the wild.
