Abstract:Cadmium (Cd) contamination, which poses a serious threat to human health, has been recognized as a major threat to the agricultural system and crop production. Salicylic acid (SA) is a signaling molecule that plays an important role in against Cd toxicity. Previously, we found that spraying rice with SA could reduce the Cd accumulation in rice grains grown in Cd-contaminated soil. In this study, we studied the specific mechanism of SA spray on reducing Cd accumulation in rice grain. The results showed that treatment with SA could alleviate Cd toxicity in rice by increasing the activities of antioxidant enzymes that reduce hydrogen peroxide (H2O2) accumulation, but not by changing the pH, or total or available Cd of the soil. The key factor by which SA treatment reduced Cd accumulation in rice grains was by decreasing the Cd content in rice leaves at the flowering stage. This indicated that SA could modulate the Cd accumulation in shoots, reducing the Cd translocation to rice grains. Furthermore, SA could increase the H2O2 content, activating the SA-signaling pathway and modulating the expression levels of Cd transporters (OsLCT1 and OsLCD) in rice leaves to increase Cd tolerance and reduce Cd accumulation in the rice grain. Thus, spraying rice with SA may be effective measure to cope with Cd contamination in paddy soils.

Abstract:Corn and wheat cultivated in hydroponic culture solution with supplies of cadmium (c(Cd)0, 4.0 μmol/L) and phosphorus (c(P)0, 0.12, 0.6, 3.0 mmol/L) under controlled growth conditions in relation to the chemical forms of Cd, growth and physiological parameters, and enzyme activities of 21-day-old plants were investigated. The three predominant forms of Cd were NaCl-extractable Cd (related to Cd-binding proteins or polypeptides), acetic acid-extractable Cd (CdHPO4, Cd3(PO4)2, or Cd-polyphosphates), and water-extractable Cd (organic acid complexes). Increasing application of P tended to reduce the percentage of Cd combined with proteins and increase the amount of Cd-phosphates. Decreased cell plasma membrane permeability and K+ leakage of Cd-treated plants were observed with the P supply of 0.6 mmol/L. The moderate supply of P (0.6 mmol/L) also improved the contents of chlorophyll pigments, the activities of root dehydrogenase and leaf nitrate reductase in Cd-treated plants, whereas supply with an excess of P (3.0 mmol/L) showed not to be more effective in the alleviation of Cd toxic to plants. Subcellular accumulation of cadmium in corn and wheat plants at different levels of phosphorus

Abstract:Corn and wheat plants were grown in a nutrient culture solution at four levels of phosphorus (0, 0.12, 0.60 and 3.0 rnmol L"1) and two levels of cadmium (0 and 4.0 pmol L -1) in greenhouse for a 18-day period. The concentrations of phosphorus and cadmium in cell wall, cytoplasm and vacuoles of roots and leaves were examined by cell fractionation techniques. With increasing phosphorus in medium, the contents of P in cell wall, cytoplasm and vacuoles of corn and wheat roots and leaves increased. The highest content of P was observed in cell wall, next in vacuoles, and the lowest in cytoplasm. The wheat subcellular fractions in both roots and leaves had higher concentrations of phosphorus than those of corn. Increasing phosphorus in medium significantly inhibited the intracellular Cd accumulation in both species. However, at P concentration up to 3.0 mmol L"1, the Cd content in cell wall was increased. Increasing phosphorus resulted in reduction of the subcellular Cd content in corn and wheat leaves. Compared with corn, the wheat roots had a higher Cd content in the cell wall and vacuoles and a lower in cytoplasm, while in leaf subcellular fractions the wheat cell had a higher Cd content in its vacuoles and a lower one in its cytoplasm. The results indicate that phosphorus may be involved in sequestration of Cd ionic activity in both cell wall and vacuoles by forming insoluble Cd phosphate. © 1999 SCIENCE PRESS.

Abstract:Exogenous phosphorus (P) application can affect the degree of cadmium (Cd) accumulation in plants. In this study, a high-Cd accumulating rice cultivar (Fupin 36) was used to investigate the relationship between P and Cd. Pot experiments (hydroponics and soil culture) showed that Cd exposure inhibited plant biomass increased and interfered the uptake of nutrient elements. However, application of appropriate levels of P mitigated the deleterious effects of Cd and decreased Cd accumulation in rice, whereas application of a high level of P exacerbated Cd toxicity, indicating that P induced Cd accumulation in a dose-dependent manner. Kinetics analysis showed that Cd uptake in the roots was positively correlated with the P absorption pathway. Appropriate P application partially mitigated the lipid peroxidation impact and increased antioxidant enzymes activity (superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase) to ameliorate Cd-associated toxicity. Importantly, P application to upregulated the expression of Fe-responsive genes (e.g., OsNRAMP1 , OsIRT1 , OsYSL15 , OsIRO2 , and OsNAS2 ), and further positive feedback regulated the Cd accumulation by OsNRAMP1 and OsIRT1 as the Cd 2+ /Fe 2+ transporters. These findings indicated that P regulated Cd (im)mobilization by mediating the Fe homeostasis pathway in plants. In conclusion, P affected the Fe content by interfering Fe uptake-associated pathway (especially of OsNRAMP1 and OsIRT1 ) and feedback regulate the Cd accumulation in rice, as well as modulated the antioxidant defense to relieve the Cd toxicity. Additionally, appropriate levels of P application can enhance the Cd detoxification process and plant growth and ensure grain production and security.

Abstract:Rice is the main food crops with the higher capacity for cadmium (Cd) uptake, necessitating the urgent need for remediation measures to address Cd in paddy soil. Reasonable agronomic methods are convenient and favorable for fixing the issue. In this study, a pot experiment was employed to evaluate the effects of two foliar (NaH 2 PO 4 , SDP; KH 2 PO 4 , PDP) and two solid phosphate fertilizers (double-superphosphate, DSP; calcium-magnesium phosphate, CMP) on uptake and remobilization of Cd in rice plants under the low-P and rich-Cd soil. The results revealed that these four phosphorus fertilizer significantly down-regulated the relative expression of OsNRAMP5 involved in Cd absorption, while up-regulated OsPCS1 expression and increased distribution of Cd into the cell wall in roots. Furthermore, phosphorus fertilizer resulted in a significant decrease in the relative expression of OsLCT1 in stems and OsLCD in leaves, decreased the transfer factor of Cd from shoots to grains, and ulterior reduced the Cd accumulation in three protein components of globulin, albumin, and glutelin, making the average Cd concentration of brown rice decreased by 82.96%. These results comprehensively indicate that in situations with similar soil backgrounds, the recommended application of solid CMP and foliar PDP can alleviate the toxicity of Cd by reducing its absorption and remobilization.

Abstract:It is unknown how the Cd-mediated changes in the rhizospheric nitrifier community affect plant growth under different water management practices. This study examined the effect of Cd on microbial N transformation and its association with rice growth in two water regimes, continuous flooding (CF) and alternate wetting and drying (AWD), in a paddy soil. The 16S rRNA, amoA genes in the rhizosphere and endosphere, and OsNTRs were determined using qPCR. Nitrifier community analysis was performed by sequencing the amoA genes. Concentrations of N and Cd in shoots and roots, and IAA in the roots were determined by flow injection analyzer, ICP-MS and enzyme-linked immunosorbent (ELISA), respectively. The direct and indirect negative effects of Cd bioavailability in the rhizosphere on plant growth and ultra-structure of roots and leaves were greater under AWD than CF after 56-day growth. Nitrifier community was changed by water and Cd treatments. Crenarchaeota, Nitrosopumilus, Nitrosopira, and Nitrosovibrio tenuis related species were the dominant nitrifier players. Increasing Cd level decreased plant N uptake, root-IAA concentration, and the abundance of 16S rRNA gene in the rhizosphere and endosphere to a greater extent under AWD than under CF. This in turn decreased the colonization of endophytes and the surface area of the roots, leading to decreased N uptake from the soil. The results imply that appropriate water management and rhizospheric engineering in Cd-contaminated paddy fields may be important approaches to decreasing plant Cd uptake and rhizospheric nitrification.
More Effective Than Direct Contact: Nano Hydroxyapatite Pre-Treatment Regulates the Growth and Cd Uptake of Rice (oryza Sativa L.) Seedlings

Abstract:Cd contamination in rice urgently needs to be addressed. Nano hydroxyapatite (n-HAP) is an eco-friendly material with excellent Cd fixation ability. However, due to its own high reactivity, innovative application of n-HAP in the treatment of Cd contamination in rice is needed. In this study, we proposed a new application, namely n-HAP pre-treatment, which can effectively reduce Cd accumulation in rice and alleviate Cd stress. The results showed that 80mg/L n-HAP pre-treatment significantly reduced Cd content in rice shoot by 35.1%. Biochemical and combined transcriptomic-proteomic analysis revealed the possible molecular mechanisms by which n-HAP pre-treatment promoted rice growth and reduced Cd accumulation. (1) n-HAP pre-treatment regulated gibberellin and jasmonic acid synthesis-related pathways, increased gibberellin content and decreased jasmonic acid content in rice root, which promoted rice growth; (2) n-HAP pre-treatment up-regulated gene CATA1 expression and down-regulated gene OsGpx1 expression, which increased rice CAT activity and GSH content; (3) n-HAP pre-treatment up-regulated gene OsZIP1 expression and down-regulated gene OsNramp1 expression, which reduced Cd uptake, increased Cd efflux from rice root cells.
Effect of pH on growth and Cd accumulation in different rice varieties under hydroponics

Abstract:Currently, applying lime to cadmium (Cd)-contaminated paddy fields to increase pH and reduce Cd availability is an effective method to control excessive Cd levels in rice grain. However, under hydroponic conditions, the impact of increased pH on Cd accumulation in different rice varieties remains unclear. This study employed three rice varieties (Yuzhenxiang, Shaoxiang 100, Xiangwanxian 12) with different Cd accumulation characteristics under different pH and long-term treatment with 1 μM CdCl2, to study the effect of pH on growth and Cd accumulation in different rice varieties. The result showed that as pH shifted from 5 to 8, the SPAD values, shoot dry weight, and plant height of the three rice varieties significantly decreased. The main root length, root volume, and root dry weight of Yuzhenxiang, and Shaoxiang100 significantly decreased. Conversely, the root architecture indicators of Xiangwanxian 12 did not change significantly. As for element accumulation, increasing the pH significantly increased the content of Mn in both the shoots and roots of all three varieties. Yuzhenxiang significantly reduced Cd content in both the shoots and roots of rice, while Shaoxiang100 significantly increased Cd content in both parts. Xiangwanxian 12 showed a significant increase in Cd content in the shoots but a decrease in the roots. In terms of subcellular distribution, Yuzhenxiang significantly reduced Cd concentrations in the cell wall and organelles of root cells, resulting in lower Cd concentrations in the root tissue. Conversely, Shaoxiang100 significantly increased Cd concentrations in the cell wall, organelles, and soluble fractions of root cells, leading to higher Cd concentrations in the root tissue. Xiangwanxian 12 also exhibited a decrease in Cd concentrations in the cell wall, organelles, and soluble fraction of root cells, resulting in lower Cd concentrations in the root tissue. Additionally, the expression of the OsNRAMP5 and OsHMA3 gene was significantly increased in Shaoxiang 100, while no significantly change in Yuzhenxiang and Xiangwanxian 12. These results provide important guidance on the impact of pH on Cd accumulation during the vegetative growth stage of different rice varieties.
Zinc alleviates growth inhibition and oxidative stress caused by cadmium in rice

Abstract:A hydroponic experiment with two rice cultivars differing in cadmium (Cd) tolerance was conducted to investigate the alleviating effect of zinc (Zn) on growth inhibition and oxidative stress caused by Cd. Treatments consisted of all combinations of two Zn concentrations (0.2 and 1 mu M), three Cd concentrations (0, 1, and 5 mu M), and two rice cultivars (Bing 97252, Cd-tolerant; Xiushui 63, Cd-sensitive). Cd toxicity caused a dramatic reduction in plant height and biomass, chlorophyll concentration and photosynthetic rate, and an increase in Cd concentration in both roots and shoots, malondialdehyde (MDA) concentration, and superoxide dismutase (SOD) and peroxidase (POD) activities in shoots. The response of all these parameters was much larger for Xiushui 63 than for Bing 97252. Addition of Zn to the medium solution alleviated Cd toxicity, which was reflected in a significant increase in plant height, biomass, chlorophyll concentration, and photosynthetic rate, and a marked decrease in MDA concentration and activity of anti-oxidative enzymes. However, it was noted that Zn increased shoot Cd concentration at higher Cd supply, probably due to the enhancement of Cd translocation from roots to shoots. Therefore, further studies are necessary to determine the effect of Zn supply on Cd translocation from vegetative organs to grains or grain Cd accumulation before Zn fertilizer is applied to Cd-contaminated soils to alleviate Cd toxicity in rice.
Phosphate Addition Diminishes the Efficacy of Wollastonite in Decreasing Cd Uptake by Rice (oryza Sativa L.) in Paddy Soil.

Abstract:Cadmium (Cd) contamination in paddy soils poses food security risks and public health concerns. Exploring effective strategies to reduce rice grain Cd is an urgent need. In this study, field plot experiments were conducted to evaluate the effects of wollastonite application with or without phosphate (P) addition on Cd accumulation in rice (Oryza sativa L.). Co-application of P and wollastonite showed greater efficiency than wollastonite amendments alone in raising soil pH and CEC and decreasing soil Cd availability. Cd concentration in brown rice was decreased by 71% under the wollastonite treatment alone, but was decreased by only 29–39% when wollastonite was coupled with different P amendments. This seeming contradiction could be ascribed to the dramatic decline in the phytoavailability of manganese (Mn) and the increase in molar ratio of iron (Fe) to Mn (Fe/Mn) in Fe plaques on root surfaces in the presence of P additions. Significant negative correlations between Mn and Cd in rice plants and positive correlations between Fe/Mn in Fe plaque and Cd in rice plants indicated that P-induced soil Mn deficiency and reduced Mn in Fe plaque impeded the alleviation of Cd accumulation in rice. Application of wollastonite in Si-deficient paddy soils was effective in reducing rice Cd accumulation while boosting rice yield, but co-application of P and wollastonite was counterproductive and should be avoided. This work emphasized that a better understanding of the relationships between Cd and related mineral nutrient uptake would be helpful in developing more efficient measures to reduce rice grain Cd.
Inhibition effects of long-term calcium-magnesia phosphate fertilizer application on Cd uptake in rice: Regulation of the iron-nitrogen coupling cycle driven by the soil microbial community

Abstract:Cadmium (Cd) pollution in paddy soil seriously endangers food safety production. To investigate the effects and microbiological mechanisms of calcium-magnesium-phosphate (CMP) fertilizer application on Cd reduction in rice, field experiments were conducted in Cd-contaminated paddy soil. Compared with conventional compound fertilizer, CMP fertilizer treatments inhibited Cd uptake through plant roots, significantly decreasing Cd content in rice grains from 0.340 to 0.062 mg/kg. Soil pH and total Ca, Mg and P contents increased after CMP fertilizer application, resulting in a further decrease in soil available Cd content from 0.246 to 0.181 mg/kg. Specific extraction analysis recorded a decrease in both available Fe content and the ratio of nitrate to ammonium nitrogen, indicating that the soil Fe-N cycle was affected by the addition of CMP fertilizer. This finding was also recorded using soil bacterial community sequencing, with CMP fertilizer promoting the progress of nitrate-dependent Fe-oxidation driven by Thiobacillus (1.60-2.83%) and subsequent dissimilatory nitrate reduction to ammonium (DNRA) driven by Ignavibacteriae (1.01-1.92%); Fe-reduction driven by Anaeromyxobacter (3.09-2.23%) was also inhibited. Our results indicate that CMP fertilizer application regulates the Fe-N coupling cycle driven by the soil microbial community to benefit remediation of Cd contaminated paddy soil.
Hydrogen peroxide reduces root cadmium uptake but facilitates root-to-shoot cadmium translocation in rice through modulating cadmium transporters

Abstract:Cadmium (Cd) contamination in agricultural soils has become a serious worldwide environmental problem threatening crop production and human health. Hydrogen peroxide (H 2 O 2 ) is a critical second messenger in plant response to Cd exposure. However, its role in Cd accumulation in various organs of plants and the mechanistic basis of this regulation remains to be elucidated. In this study, we used electrophysiological and molecular approaches to understand how H 2 O 2 regulates Cd uptake and translocation in rice plants. Our results showed that the pretreatment of H 2 O 2 significantly reduced Cd uptake by rice roots, which was associated with the downregulation of OsNRAMP1 and OsNRAMP5 . On the other hand, H 2 O 2 promoted the root-to-shoot translocation of Cd, which might be attributed to the upregulation of OsHMA2 critical for Cd 2+ phloem loading and the downregulation of OsHMA3 involved in the vacuolar compartmentalization of Cd 2+ , leading to the increased Cd accumulation in rice shoots. Furthermore, such regulatory effects of H 2 O 2 on Cd uptake and translocation were notably amplified by the elevated level of exogenous calcium (Ca). Collectively, our results suggest that H 2 O 2 can inhibit Cd uptake but increase root to shoot translocation through modulating the transcriptional levels of genes encoding Cd transporters, furthermore, application of Ca can amplify this effect. These findings will broaden our understanding of the regulatory mechanisms of Cd transport in rice plants and provide theoretical foundation for breeding rice for low Cd accumulation.

Abstract:Rice consumption is a key Cd exposure pathway, which poses a health risk to humans. Reducing cadmium (Cd) concentrations in rice remains challenging. In this study, a pot experiment was conducted to examine the effects of foliar spray of Zn combined with organic matters (including Zn–lysine (Zn-Lys), Zn–fulvic acid (Zn-FA), Zn–amino acid (Zn-AA), and Zn combined with glutathione (Zn + GSH)) on Cd accumulation in rice grains. Compared with the control group, all treatment groups exhibited reduced Cd concentration in rice grains, while improving plant growth, and reducing Cd transport from other tissues to the grains. Zn–FA was found to be the most effective fertilizer, which considerably reduced Cd concentrations in grains from 0.77 ± 0.068 to 0.14 ± 0.021 mg/kg and yielded reductions of up to 81%, which is within the Chinese food maximum tolerable limit of 0.2 mg/kg. Furthermore, the analysis of the chemical forms of Cd of rice tissues indicated that the treatment groups had increased proportions of integrated with pectates and protein in the stems. Except for the group treated with Zn–Lys spray, the percentages of undissolved Cd phosphate in the leaves were increased in all treatment groups, which reduced Cd toxicity to rice plants. The foliar application of Zn combined with organic matters may be a promising strategy to decrease Cd concentration in rice grains cultivated in severely Cd-contaminated agricultural soil, particularly in the karst area in southwest China with limited available cultivable agricultural land.

Abstract:Cadmium (Cd) accumulation in rice is strongly controlled by liming, but information on the use of liming to control Cd accumulation in rice grown in slightly acidic soils is inconsistent. Here, pot experiments were carried out to investigate the mechanisms of liming on Cd accumulation in two rice varieties focusing on two aspects: available/exchangeable Cd content in soils that were highly responsive to liming, and Cd uptake and transport capacity in the roots of rice in terms of Cd accumulation-relative gene expression. The results showed that soil availability and exchangeable iron, manganese, zinc and Cd contents decreased with increased liming, and that genes related to Cd uptake (OsNramp5 and OsIRT1) were sharply up-regulated in the roots of the two rice varieties. Thus, iron, manganese, zinc and Cd contents in rice plants increased under low liming applications but decreased in response to high liming applications. However, yield and rice quantities were only slightly affected. These results indicated that Cd accumulation in rice grown in slightly acidic soils presents a contradictory dynamic equilibrium between Cd uptake capacity by roots and soil Cd immobilisation in response to liming. The enhanced Cd uptake capacity under low liming dosages increases risks to human health.
Foliar Spraying of Carbon Dots Reduces Cadmium Accumulation in Rice by Regulating Rhizosphere Immobilization, Root Development, and Subcellular Distribution

Abstract:Cadmium (Cd) is a toxic heavy metal widespread in rice paddies and threatens food safety and human health. Foliar exposure represents a cost-effective, simple, and time-independent approach to enhance rice resistance and minimize Cd accumulation. Herein, foliar spraying of carbon dots (CDs) was found to significantly reduce Cd accumulation in rice roots and shoots by 31.51 % and 17.93 %, respectively. Gene expression and mineral nutrient analyses indicated that CDs exposure inhibited Cd uptake by suppressing OsNramp1 and increasing competition of Fe and Mn with Cd for OsNramp5. Besides, CDs exposure down-regulated OsHMA2 for Cd transport from roots to aerial parts and up-regulated OsHMA3 for Cd vacuole sequestration. Additionally, CDs treatment promoted rice root development by increasing root biomass, cell walls, and mechanical resistance, which helped to anchor rice plants and impede Cd uptake. Furthermore, CDs spraying increased the organic carbon content and altered the microbial communities in the rice rhizosphere by increasing root exudation. This process facilitated the binding of dissolved Cd in pore water to organic matter and iron‑manganese oxides, ultimately reducing the bioavailability of Cd. This study underscores the effectiveness and mechanism of simple foliar spraying of CDs to mitigate Cd accumulation in rice.

Abstract:Rice ( Oryza sativa ) is one of the major cereal crops and takes up cadmium (Cd) more readily than other crops. Understanding the mechanism of Cd uptake and defense in rice can help us avoid Cd in the food chain. However, studies comparing Cd uptake, toxicity, and detoxification mechanisms of leaf and root Cd exposure at the morphological, physiological, and transcriptional levels are still lacking. Therefore, experiments were conducted in this study and found that root Cd exposure resulted in more severe oxidative and photosynthetic damage, lower plant biomass, higher Cd accumulation, and transcriptional changes in rice than leaf Cd exposure. The activation of phenylpropanoids biosynthesis in both root and leaf tissues under different Cd exposure routes suggests that increased lignin is the response mechanism of rice under Cd stress. Moreover, the roots of rice are more sensitive to Cd stress and their adaptation responses are more pronounced than those of leaves. Quantitative PCR revealed that OsPOX , OsCAD , OsPAL and OsCCR play important roles in the response to Cd stress, which further emphasize the importance of lignin. Therefore, this study provides theoretical evidence for future chemical and genetic regulation of lignin biosynthesis in crop plants to reduce Cd accumulation.

Abstract:Two contrasting rice (Oryza sativa L.) cultivars, i.e. Wuyujing 3 (WYJ3, Cd-tolerant) and Shanyou 63 (SY63, Cd-sensitive), were grown on a red soil (Ultisol) to study both individual and combined phytotoxicity of arsenic (As) and cadmium (Cd) in terms of Cd and As availability, their uptake and accumulation, antioxidant defense activity and oxidative damage. The antioxidant defense system examined in this study included enzymatic and non-enzymatic molecular antioxidants such as superoxide dismutase (SOD), peroxidase (POD), glutathione (GSH) and ascorbic acid (AsA). Results showed that As or Cd treatment decreased root and shoot biomass in both cultivars compared with their corresponding control (no Cd or As treatment), although less severe inhibition of plant growth was observed in WYJ3 than in SY63. Moreover, rice growth was inhibited more severely by Cd treatment than by As treatment, which could be explained by the higher amount of available Cd (60%) (0.1M HCl-extractable Cd) compared to the lower amount of available As (15%) (0.5M NaH2PO4-extractable As) in their postharvest soils. However, shoot biomass in cultivar SY63, and root and shoot biomass in cultivar WYJ3 were significantly higher in the As plus Cd treatment than in the Cd treatment alone, showing that the combined toxicity of these two heavy metals was not additive and on the contrary, As mitigated Cd-induced growth inhibition. The As plus Cd treatment also significantly decreased As or Cd concentrations both in roots and in shoots of the two rice cultivars compared with the As or Cd treatment alone, respectively. On the other hand, treatment with As or Cd alone significantly decreased the SOD and POD activities, and GSH and AsA concentrations, while the activities of these enzymes and the concentrations of GSH and AsA were significantly higher in the As plus Cd treatment than in the Cd treatment alone, resulting in less severe oxidative damage as indicated by the lower concentration of MDA in the As plus Cd treatment (P<0.05). However, no significant difference was observed in the antioxidant defense activity between the As plus Cd treatment and the As treatment alone. These results suggest that the combined toxicity of As and Cd in rice is lower than that of individual Cd or As, which might be attributed to the decreased uptake and accumulation of Cd and As, and the less oxidative stress caused by the interactive effects of As with Cd both in rhizosphere and in plants.

Abstract:Microbial remediation is a promising technique to reduce Cd accumulation in rice (Oryza sativa). In present study, a set of pot experiments were conducted to evaluate the effects of Cd-tolerate Pseudomonas TCd-1 inoculation on rice Cd uptake, soil enzyme activities and Cd bioavailability in the rhizosphere soils under Cd contaminated conditions. The results showed that at the ripening stage, with the inoculation of TCd-1, Cd contents in root, culm, leaf, hull and brown rice significantly reduced by 60.7%, 47.7%, 50.6%, 58.1% and 47.9%, respectively, and the cadmium bioconcentration factor (BCF) of rice lowered by 66.2% under 5 mg kg-1 Cd treatment. At the meantime, in the rhizosphere soils, pH increased by 0.05, the contents of exchangeable Cd (EX-Cd) and Fe-Mn oxides (OX-Cd) increased by 107.8% and 33.5%, whereas organic matter (OM-Cd) and residual (Res-Cd) decreased by 31.9% and 60.0%, respectively. The activity of acid phosphatase (ACP) increased by 28.3%, catalase (CAT), saccharase (SUC) activity decreased by 28.5% and 26.0%. Similarly, the Cd contents in root, culm, leaf, hull and brown rice reduced by 42.1%, 42.5%, 58.0%, 50.3%, and 68.8%, respectively, and the BCF lowered by 57.1%, under 10 mg kg-1 Cd treatment. Simultaneously, the soil pH increased by 0.06, the activities of CAT, SUC, urease (URE), ACP decreased by 26.4%, 34.6%, 63.8% and 15.3%, respectively. Furthermore, the correlation analysis showed that the inoculation of TCd-1 changed the correlation between rice Cd content and the biomass of roots, leaves, soil pH, CAT, PPO, URE activities, OM-Cd in rhizosphere soils. It suggested that Pseudomonas TCd-1 effectively reduced Cd uptake and Cd accumulation in rice was closely linked to the changes of soil pH, enzyme activities and Cd availability.

Abstract:Strategies to reduce cadmium (Cd) in rice grain, below concentrations that represent serious human health concerns, require that the mechanisms of Cd distribution and accumulation within rice plants be established. Here, a comprehensive hydroponic experiment was performed to investigate the differences in the Cd uptake, chelation and compartmentalization between high (D83B) and low (D62B) Cd-accumulation cultivars contrasting in Cd accumulation in order to establish the roles of these processes in limiting Cd translocation from root to shoot. D83B showed 3-fold higher Cd accumulation in the shoots than the cultivar D62B. However, a short-term Cd uptake experiment showed more Cd uptake by D62B than by D83B. The distribution of Cd in roots and shoots differed significantly. D83B translocated 38% of total Cd taken up to the shoots, whereas D62B retained most of the Cd in the roots. D62B had higher amounts of non-protein thiols (NPTs) and glutathione (GSH) than D83B. The NPT and Cd distribution ratio (CDR) in the anionic form in the roots of D62B increased gradually as Cd concentration increased. In D83B, in contrast, levels of CDR in the cationic form increased significantly from 22.10 to 43.37%, while NPT only increased slightly. Furthermore, the percentage of Cd ions retained in thiol-rich peptides, especially in the HMW complexes, was significantly higher in D62B compared with D83B. However, D83B possessed a greater proportion of potentially mobile (cationic) Cd in the roots and showed superior Cd translocation from root to shoot. Taken as a whole, the results presented in this study revealed that Cd chelation, compartmentalization and adsorption contribute to the Cd retention in roots.

Abstract:Cadmium (Cd) toxicity negatively impacts plant health and productivity. Nanosilica (SiO2NPs) and salicylic acid (SA) enhance plant performance and alleviate heavy metals stress. Yet, their combined effects against Cd-toxicity in rice remained less-explored. Thus, a hydroponic study investigated the individual and combined effects of SiO2NPs and SA on Cd-stress mitigation in rice at physio-biochemical, cellular, and molecular levels. Results indicated that Cd-alone treatment caused a significant reduction in rice growth and biomass and photosynthetic efficiency, which was associated with oxidative damage caused by enhanced Cd-accumulation in plant tissues. Cd-induction also potentiated its phytotoxicity by triggering enzymatic antioxidants against the extra production of reactive oxygen species (ROS). The addition of SiO2NPs and/or SA markedly minimized the Cd-induced toxicity by reducing Cd-bioaccumulation (42–56%), protecting photosynthetic efficiency, which were directly correlated with seedling biomass and restored cellular structures (leaf ultrastructure and surface morphology). The combined application of SiO2NPs and SA was more effective in activating antioxidant enzymes, phytohormones biosynthesis, and reducing oxidative damages caused by Cd than sole application. This was evident in the decreased production of ROS, malondialdehyde contents (29–37%), and recovered membrane stability. Moreover, SiO2NPs and/or SA relieved Cd-bioaccumulation (41–56%) by downregulating the Cd-related transporter genes (OsNramp1, OsNramp5, OsHMA2, and OsHMA3). Altogether, the cellular Cd-accumulation, photosynthesis, antioxidant defense, and phytohormones against oxidative stress can be ideal markers for cultivating rice in Cd-contaminated soils.
