Abstract:Introduction Recently, accurate machine learning and deep learning approaches have been dedicated to the investigation of breast cancer invasive disease events (IDEs), such as recurrence, contralateral and second cancers. However, such approaches are poorly interpretable. Methods Thus, we designed an Explainable Artificial Intelligence (XAI) framework to investigate IDEs within a cohort of 486 breast cancer patients enrolled at IRCCS Istituto Tumori “Giovanni Paolo II” in Bari, Italy. Using Shapley values, we determined the IDE driving features according to two periods, often adopted in clinical practice, of 5 and 10 years from the first tumor diagnosis. Results Age, tumor diameter, surgery type, and multiplicity are predominant within the 5-year frame, while therapy-related features, including hormone, chemotherapy schemes and lymphovascular invasion, dominate the 10-year IDE prediction. Estrogen Receptor (ER), proliferation marker Ki67 and metastatic lymph nodes affect both frames. Discussion Thus, our framework aims at shortening the distance between AI and clinical practice

Abstract:<p>More than 750 000 women in Italy are surviving a diagnosis of breast cancer. A large body of literature tells us which characteristics impact the most on their prognosis. However, the prediction of each disease course and then the establishment of a therapeutic plan and follow‐up tailored to the patient is still very complicated. In order to address this issue, a multidisciplinary approach has become widely accepted, while the Multigene Signature Panels and the Nottingham Prognostic Index are still discussed options. The current technological resources permit to gather many data for each patient. Machine Learning (ML) allows us to draw on these data, to discover their mutual relations and to esteem the prognosis for the new instances. This study provides a primary evaluation of the application of ML to predict breast cancer prognosis. We analyzed 1021 patients who underwent surgery for breast cancer in our Institute and we included 610 of them. Three outcomes were chosen: cancer recurrence (both loco‐regional and systemic) and death from the disease within 32 months. We developed two types of ML models for every outcome (Artificial Neural Network and Support Vector Machine). Each ML algorithm was tested in accuracy (=95.29%‐96.86%), sensitivity (=0.35‐0.64), specificity (=0.97‐0.99), and AUC (=0.804‐0.916). These models might become an additional resource to evaluate the prognosis of breast cancer patients in our daily clinical practice. Before that, we should increase their sensitivity, according to literature, by considering a wider population sample with a longer period of follow‐up. However, specificity, accuracy, minimal additional costs, and reproducibility are already encouraging.</p>

Evaluation of Machine Learning Algorithms for the Prognosis of Breast Cancer from the Surveillance, Epidemiology, and End Results Database

Abstract:Introduction Many researchers used machine learning (ML) to predict the prognosis of breast cancer (BC) patients and noticed that the ML model had good individualized prediction performance. Objective The cohort study was intended to establish a reliable data analysis model by comparing the performance of 10 common ML algorithms and the the traditional American Joint Committee on Cancer (AJCC) stage, and used this model in Web application development to provide a good individualized prediction for others. Methods This study included 63145 BC patients from the Surveillance, Epidemiology, and End Results database. Results Through the performance of the 10 ML algorithms and 7th AJCC stage in the optimal test set, we found that in terms of 5-year overall survival, multivariate adaptive regression splines (MARS) had the highest area under the curve (AUC) value (0.831) and F1-score (0.608), and both sensitivity (0.737) and specificity (0.772) were relatively high. Besides, MARS showed a highest AUC value (0.831, 95%confidence interval: 0.820–0.842) in comparison to the other ML algorithms and 7th AJCC stage (all P < 0.05). MARS, the best performing model, was selected for web application development (https://w12251393.shinyapps.io/app2/). Conclusions The comparative study of multiple forecasting models utilizing a large data noted that MARS based model achieved a much better performance compared to other ML algorithms and 7th AJCC stage in individualized estimation of survival of BC patients, which was very likely to be the next step towards precision medicine.
Multimodal Prediction of Five-Year Breast Cancer Recurrence in Women Who Receive Neoadjuvant Chemotherapy

Abstract:In current clinical practice, it is difficult to predict whether a patient receiving neoadjuvant chemotherapy (NAC) for breast cancer is likely to encounter recurrence after treatment and have the cancer recur locally in the breast or in other areas of the body. We explore the use of clinical history, immunohistochemical markers, and multiparametric magnetic resonance imaging (DCE, ADC, Dixon) to predict the risk of post-treatment recurrence within five years. We performed a retrospective study on a cohort of 1738 patients from Institut Curie and analyzed the data using classical machine learning, image processing, and deep learning. Our results demonstrate the ability to predict recurrence prior to NAC treatment initiation using each modality alone, and the possible improvement achieved by combining the modalities. When evaluated on holdout data, the multimodal model achieved an AUC of 0.75 (CI: 0.70, 0.80) and 0.57 specificity at 0.90 sensitivity. We then stratified the data based on known prognostic biomarkers. We found that our models can provide accurate recurrence predictions (AUC > 0.89) for specific groups of women under 50 years old with poor prognoses. A version of our method won second place at the BMMR2 Challenge, with a very small margin from being first, and was a standout from the other challenge entries.

Abstract:Machine learning (ML) has been recently introduced to develop prognostic classification models that can be used to predict outcomes in individual cancer patients. Here, we report the significance of an ML-based decision support system (DSS), combined with random optimization (RO), to extract prognostic information from routinely collected demographic, clinical and biochemical data of breast cancer (BC) patients. A DSS model was developed in a training set (n = 318), whose performance analysis in the testing set (n = 136) resulted in a C-index for progression-free survival of 0.84, with an accuracy of 86%. Furthermore, the model was capable of stratifying the testing set into two groups of patients with low- or high-risk of progression with a hazard ratio (HR) of 10.9 (p < 0.0001). Validation in multicenter prospective studies and appropriate management of privacy issues in relation to digital electronic health records (EHR) data are presently needed. Nonetheless, we may conclude that the implementation of ML algorithms and RO models into EHR data might help to achieve prognostic information, and has the potential to revolutionize the practice of personalized medicine.

Abstract:Background: To address the numerous unmeet clinical needs, in recent years several Machine Learning models applied to medical images and clinical data have been introduced and developed. Even when they achieve encouraging results, they lack evolutionary progression, thus perpetuating their status as autonomous entities. We postulated that different algorithms which have been proposed in the literature to address the same diagnostic task, can be aggregated to enhance classification performance. We suggested a proof of concept to define an ensemble approach useful for integrating different algorithms proposed to solve the same clinical task. Methods: The proposed approach was developed starting from a public database consisting of radiomic features extracted from CT images relating to 535 patients suffering from lung cancer. Seven algorithms were trained independently by participants in the AI4MP working group on Artificial Intelligence of the Italian Association of Physics in Medicine to discriminate metastatic from non-metastatic patients. The classification scores generated by these algorithms are used to train SVM classifier. The Explainable Artificial Intelligence approach is applied to the final model. The ensemble model was validated following an 80-20 hold-out and leave-one-out scheme on the training set. Results: Compared to individual algorithms, a more accurate result was achieved. On the independent test the ensemble model achieved an accuracy of 0.78, a F1-score of 0.57 and a log-loss of 0.49. Shapley values representing the contribution of each algorithm to the final classification result of the ensemble model were calculated. This information represents an added value for the end user useful for evaluating the appropriateness of the classification result on a particular case. It also allows us to evaluate on a global level which methodological approaches of the individual algorithms are likely to have the most impact. Conclusion: Our proposal represents an innovative approach useful for integrating different algorithms that populate the literature and which lays the foundations for future evaluations in broader application scenarios.

Abstract:Over the past decade, breast cancer has been the most common type of cancer in women. Different methods were proposed for breast cancer detection. These methods mainly classify and categorize malignant and Benign tumors. Machine learning is a practical approach for breast cancer classification. Data mining and classification are effective methods to predict and categorize breast cancer. The optimum classification for detecting Breast Cancer (BC) is ensemble-based. The ensemble approach involves using multiple ways to find the best possible solution. This study used the Wisconsin Breast Cancer Diagnostic (WBCD) dataset. We created a voting ensemble classifier that combines four different machine learning models: Extra Trees Classifier (ETC), Light Gradient Boosting Machine (LightGBM), Ridge Classifier (RC), and Linear Discriminant Analysis (LDA). The proposed ELRL-E approach achieved an accuracy of 97.6%, a precision of 96.4%, a recall of 100%, and an F1 score of 98.1%. Various output evaluations are used to evaluate the performance and efficiency of the proposed model and other classifiers. Overall, the recommended strategy performed better. Results are directly compared with the individual classifier and different recognized state-of-the-art classifiers. The primary objective of this study is to identify the most influential ensemble machine learning classifier for breast cancer detection and diagnosis in terms of accuracy and AUC score.

Abstract:Objective Current prognostic factors for endometrial cancer are not sufficient to predict recurrence in early stages. Treatment choices are based on the prognostic factors included in the risk classes defined by the ESMO-ESGO-ESTRO (European Society for Medical Oncology-European Society of Gynaecological Oncology-European Society for Radiotherapy and Oncology) consensus conference with the new biomolecular classification based on POLE, TP53, and microsatellite instability status. However, a minority of early stage cases relapse regardless of their low risk profiles. Integration of the immune context status to existing molecular based models has not been fully evaluated. This study aims to investigate whether the integration of the immune landscape in the tumor microenvironment could improve clinical risk prediction models and allow better profiling of early stages. Methods Leveraging the potential of in silico deconvolution tools, we estimated the relative abundances of immune populations in public data and then applied feature selection methods to generate a machine learning based model for disease free survival probability prediction. Results We included information on International Federation of Gynecology and Obstetrics (FIGO) stage, tumor mutational burden, microsatellite instability, POLEmut status, interferon γ signature, and relative abundances of monocytes, natural killer cells, and CD4+T cells to build a relapse prediction model and obtained a balanced accuracy of 69%. We further identified two novel early stage profiles that undergo different pathways of recurrence. Conclusion This study presents an extension of current prognostic factors for endometrial cancer by exploiting machine learning models and deconvolution techniques on available public biomolecular data. Prospective clinical trials are advisable to validate the early stage stratification.
Adaptive Machine Learning Approach for Importance Evaluation of Multimodal Breast Cancer Radiomic Features

Abstract:Breast cancer holds the highest diagnosis rate among female tumors and is the leading cause of death among women. Quantitative analysis of radiological images shows the potential to address several medical challenges, including the early detection and classification of breast tumors. In the P.I.N.K study, 66 women were enrolled. Their paired Automated Breast Volume Scanner (ABVS) and Digital Breast Tomosynthesis (DBT) images, annotated with cancerous lesions, populated the first ABVS+DBT dataset. This enabled not only a radiomic analysis for the malignant vs. benign breast cancer classification, but also the comparison of the two modalities. For this purpose, the models were trained using a leave-one-out nested cross-validation strategy combined with a proper threshold selection approach. This approach provides statistically significant results even with medium-sized data sets. Additionally it provides distributional variables of importance, thus identifying the most informative radiomic features. The analysis proved the predictive capacity of radiomic models even using a reduced number of features. Indeed, from tomography we achieved AUC-ROC using 19 features and using 7 of them; while from ABVS we attained an AUC-ROC of using 22 features and using only 3 features. Although the predictive power of DBT outperforms ABVS, when comparing the predictions at the patient level, only 8.7% of lesions are misclassified by both methods, suggesting a partial complementarity. Notably, promising results (AUC-ROC ABVS-DBT - ) were achieved using non-geometric features, thus opening the way to the integration of virtual biopsy in medical routine.

Abstract:This paper primarily addresses a dataset relating to cellular, chemical and physical conditions of patients gathered at the time they are operated upon to remove colorectal tumours. This data provides a unique insight into the biochemical and immunological status of patients at the point of tumour removal along with information about tumour classification and post-operative survival. The relationship between severity of tumour, based on TNM staging, and survival is still unclear for patients with TNM stage 2 and 3 tumours. We ask whether it is possible to predict survival rate more accurately using a selection of machine learning techniques applied to subsets of data to gain a deeper understanding of the relationships between a patient's biochemical markers and survival. We use a range of feature selection and single classification techniques to predict the 5 year survival rate of TNM stage 2 and 3 patients which initially produces less than ideal results. The performance of each model individually is then compared with subsets of the data where agreement is reached for multiple models. This novel method of selective ensembling demonstrates that significant improvements in model accuracy on an unseen test set can be achieved for patients where agreement between models is achieved. Finally we point at a possible method to identify whether a patients prognosis can be accurately predicted or not.

Abstract:Abstract Breast cancer is a deadly disease with a high mortality rate among PAN cancers. The advancements in biomedical information retrieval techniques have been beneficial in developing early prognosis and diagnosis systems for cancer patients. These systems provide the oncologist with plenty of information from several modalities to make the correct and feasible treatment plan for breast cancer patients and protect them from unnecessary therapies and their toxic side effects. The cancer patient’s related information can be collected using various modalities like clinical, copy number variation, DNA-methylation, microRNA sequencing, gene expression, and histopathological whole slide images. High dimensionality and heterogeneity in these modalities demand the development of some intelligent systems to understand related features to the prognosis and diagnosis of diseases and make correct predictions. In this work, we have studied some end-to-end systems having two main components : (a) dimensionality reduction techniques applied to original features from different modalities and (b) classification techniques applied to the fusion of reduced feature vectors from different modalities for automatic predictions of breast cancer patients into two categories: short-time and long-time survivors. Principal component analysis (PCA) and variational auto-encoders (VAEs) are used as the dimensionality reduction techniques, followed by support vector machines (SVM) or random forest as the machine learning classifiers. The study utilizes raw, PCA, and VAE extracted features of the TCGA-BRCA dataset from six different modalities as input to the machine learning classifiers. We conclude this study by suggesting that adding more modalities to the classifiers provides complementary information to the classifier and increases the stability and robustness of the classifiers. In this study, the multimodal classifiers have not been validated on primary data prospectively.

Prognostic power assessment of clinical parameters to predict neoadjuvant response therapy in HER2-positive breast cancer patients: A machine learning approach

Abstract:Background: About 15%-20% of breast cancer (BC) cases is classified as Human Epidermal growth factor Receptor type 2 (HER2) positive. The Neoadjuvant chemotherapy (NAC) was initially introduced for locally advanced and inflammatory BC patients to allow a less extensive surgical resection, whereas now it represents the current standard for early-stage and operable BC. However, only 20%-40% of patients achieve pathologic complete response (pCR). According to the results of practice-changing clinical trials, the addition of trastuzumab to NAC brings improvements to pCR, and recently, the use of pertuzumab plus trastuzumab has registered further statistically significant and clinically meaningful improvements in terms of pCR. The goal of our work is to propose a machine learning model to predict the pCR to NAC in HER2-positive patients based on a subset of clinical features. Method: First, we evaluated the significant association of clinical features with pCR on the retrospectively collected data referred to 67 patients afferent to Istituto Tumori "Giovanni Paolo II." Then, we performed a feature selection procedure to identify a subset of features to be used for training a machine learning-based classification algorithm. As a result, pCR to NAC was associated with ER status, Pgr status, and HER2 score. Results: The machine learning model trained on a subgroup of essential features reached an AUC of 73.27% (72.44%-73.66%) and an accuracy of 71.67% (71.64%-73.13%). According to our results, the clinical features alone are not enough to define a support system useful for clinical pathway. Conclusion: Our results seem worthy of further investigation in large validation studies and this work could be the basis of future study that will also involve radiomics analysis of biomedical images.
A Novel Approach for Predicting the Survival of Colorectal Cancer Patients Using Machine Learning Techniques and Advanced Parameter Optimization Methods

Abstract:Background: Colorectal cancer is one of the most prevalent forms of cancer and is associated with a high mortality rate. Additionally, an increasing number of adults under 50 are being diagnosed with the disease. This underscores the importance of leveraging modern technologies, such as artificial intelligence, for early diagnosis and treatment support. Methods: Eight classifiers were utilized in this research: Random Forest, XGBoost, CatBoost, LightGBM, Gradient Boosting, Extra Trees, the k-nearest neighbor algorithm (KNN), and decision trees. These algorithms were optimized using the frameworks Optuna, RayTune, and HyperOpt. This study was conducted on a public dataset from Brazil, containing information on tens of thousands of patients. Results: The models developed in this study demonstrated high classification accuracy in predicting one-, three-, and five-year survival, as well as overall mortality and cancer-specific mortality. The CatBoost, LightGBM, Gradient Boosting, and Random Forest classifiers delivered the best performance, achieving an accuracy of approximately 80% across all the evaluated tasks. Conclusions: This research enabled the development of effective classification models that can be applied in clinical practice.

Abstract:The choice of the most effective treatment may eventually be influenced by breast cancer survival prediction. To predict the chances of a patient surviving, a variety of techniques were employed, such as statistical, machine learning, and deep learning models. In the current study, 1904 patient records from the METABRIC dataset were utilized to predict a 5-year breast cancer survival using a machine learning approach. In this study, we compare the outcomes of seven classification models to evaluate how well they perform using the following metrics: recall, AUC, confusion matrix, accuracy, precision, false positive rate, and true positive rate. The findings demonstrate that the classifiers for Logistic Regression (LR), Support Vector Machines (SVM), Decision Tree (DT), Random Forest (RD), Extremely Randomized Trees (ET), K-Nearest Neighbor (KNN), and Adaptive Boosting (AdaBoost) can accurately predict the survival rate of the tested samples, which is 75,4\%, 74,7\%, 71,5\%, 75,5\%, 70,3\%, and 78 percent.

Abstract:Introduction: It has been estimated that 19,880 new cases of ovarian cancer had been diagnosed in 2022. Most epithelial ovarian cancer are sporadic, while in 15%-25% of cases, there is evidence of a familial or inherited component. Approximately 20%-25% of high-grade serous carcinoma cases are caused by germline mutations in the BRCA1 and BRCA2 genes. However, owing to a lack of effective early detection methods, women with BRCA mutations are recommended to undergo bilateral risk-reducing salpingo-oophorectomy (RRSO) after childbearing. Determining the right timing for this procedure is a difficult decision. It is crucial to find a clinical signature to identify high-risk BRCA-mutated patients and determine the appropriate timing for performing RRSO. Methods: In this work, clinical data referred to a cohort of 184 patients, of whom 7.6% were affected by adnexal tumors including invasive carcinomas and intraepithelial lesions after RSSO has been analyzed. Thus, we proposed an explainable machine learning (ML) ensemble approach using clinical data commonly collected in clinical practice to early identify BRCA-mutated patients at high risk of ovarian cancer and consequentially establish the correct timing for RRSO. Results: The ensemble model was able to handle imbalanced data achieving an accuracy value of 83.2%, a specificity value of 85.3%, a sensitivity value of 57.1%, a G-mean value of 69.8%, and an AUC value of 71.1%. Discussion: In agreement with the promising results achieved, the application of suitable ML techniques could play a key role in the definition of a BRCA-mutated patient-centric clinical signature for ovarian cancer risk and consequently personalize the management of these patients. As far as we know, this is the first work addressing this task from an ML perspective.

Abstract:Abstract Breast cancer (BC) is known as the most prevalent form of cancer among women. Recent research has demonstrated the potential of Machine Learning (ML) techniques in predicting the five-year BC risk using personal health data. Support Vector Machine (SVM), Random Forest, K-NN (K-Nearest Neighbour), Naive Bayes, Neural Network, Decision Tree (DT), Logistic Regression (LR), Discriminant Analysis, and their variants are commonly employed in ML for BC analysis. This study investigates the factors influencing the performance of ML techniques in the domain of BC prevention, with a focus on dataset size and feature selection. The study's goal is to examine the effect of dataset cardinality, feature selection, and model selection on analytical performance in terms of Accuracy and Area Under the Curve (AUC). To this aim, 3917 papers were automatically selected from Scopus and PubMed, considering all publications from the previous 5 years, and, after inclusion and exclusion criteria, 54 articles were selected for the analysis. Our findings highlight how a good cardinality of the dataset and effective feature selection have a higher impact on the model's performance than the selected model, as corroborated by one of the studies, which gets extremely good results with all of the models employed.

Breast Cancer Classification on Multiparametric MRI – Increased Performance of Boosting Ensemble Methods

Abstract:Introduction: This study aims to assess the utility of Boosting ensemble classification methods for increasing the diagnostic performance of multiparametric Magnetic Resonance Imaging (mpMRI) radiomic models, in differentiating benign and malignant breast lesions. Methods: The dataset includes mpMR images of 140 female patients with mass-like breast lesions (70 benign and 70 malignant), consisting of Dynamic Contrast Enhanced (DCE) and T2-weighted sequences, and the Apparent Diffusion Coefficient (ADC) calculated from the Diffusion Weighted Imaging (DWI) sequence. Tumor masks were manually defined in all consecutive slices of the respective MRI volumes and 3D radiomic features were extracted with the Pyradiomics package. Feature dimensionality reduction was based on statistical tests and the Boruta wrapper. Hierarchical Clustering on Spearman's rank correlation coefficients between features and Random Forest classification for obtaining feature importance, were implemented for selecting the final feature subset. Adaptive Boosting (AdaBoost), Gradient Boosting (GB), Extreme Gradient Boosting (XGBoost) and Light Gradient Boosting Machine (LightGBM) classifiers, were trained and tested with bootstrap validation in differentiating breast lesions. A Support Vector Machine (SVM) classifier was also exploited for comparison. The Receiver Operator Characteristic (ROC) curves and DeLong's test were utilized to evaluate the classification performances. Results: The final feature subset consisted of 5 features derived from the lesion shape and the first order histogram of DCE and ADC images volumes. XGboost and LGBM achieved statistically significantly higher average classification performances [AUC = 0.95 and 0.94 respectively], followed by Adaboost [AUC = 0.90], GB [AUC = 0.89] and SVM [AUC = 0.88]. Conclusion: Overall, the integration of Ensemble Learning methods within mpMRI radiomic analysis can improve the performance of computer-assisted diagnosis of breast cancer lesions.

Abstract:Abstract Background In clinical practice, a plethora of medical examinations are conducted to assess the state of a patient’s pathology producing a variety of clinical data. However, investigation of these data faces two major challenges. Firstly, we lack the knowledge of the mechanisms involved in regulating these data variables, and secondly, data collection is sparse in time since it relies on patient’s clinical presentation. The former limits the predictive accuracy of clinical outcomes for any mechanistic model. The latter restrains any machine learning algorithm to accurately infer the corresponding disease dynamics. Methods Here, we propose a novel method, based on the Bayesian coupling of mathematical modeling and machine learning, aiming at improving individualized predictions by addressing the aforementioned challenges. Results We evaluate the proposed method on a synthetic dataset for brain tumor growth and analyze its performance in predicting two relevant clinical outputs. The method results in improved predictions in almost all simulated patients, especially for those with a late clinical presentation (>95% patients show improvements compared to standard mathematical modeling). In addition, we test the methodology in two additional settings dealing with real patient cohorts. In both cases, namely cancer growth in chronic lymphocytic leukemia and ovarian cancer, predictions show excellent agreement with reported clinical outcomes (around 60% reduction of mean squared error). Conclusions We show that the combination of machine learning and mathematical modeling approaches can lead to accurate predictions of clinical outputs in the context of data sparsity and limited knowledge of disease mechanisms.

Abstract:Background: Breast cancer, characterized by its heterogeneity, is a leading cause of mortality among women. The study aims to develop a Machine Learning-Derived Liquid-Liquid Phase Separation (MDLS) model to enhance the prognostic accuracy and personalized treatment strategies for breast cancer patients. Methods: The study employed ten machine learning algorithms to construct 108 algorithm combinations for the MDLS model. The robustness of the model was evaluated using multi-omics and single-cell data across 14 breast cancer cohorts, involving 9,723 patients. Genetic mutation, copy number alterations, and single-cell RNA sequencing were analyzed to understand the molecular mechanisms and predictive capabilities of the MDLS model. Immunotherapy targets were predicted by evaluating immune cell infiltration and immune checkpoint expression. Chemotherapy targets were identified through correlation analysis and drug responsiveness prediction. Results: The MDLS model demonstrated superior prognostic power, with a mean C-index of 0.649, outperforming 69 published signatures across ten cohorts. High-MDLS patients exhibited higher tumor mutation burden and distinct genomic alterations, including significant gene amplifications and deletions. Single-cell analysis revealed higher MDLS activity in tumor-aneuploid cells and identified key regulatory factors involved in MDLS progression. Cell-cell communication analysis indicated stronger interactions in high-MDLS groups, and immunotherapy response evaluation showed better outcomes for low-MDLS patients. Conclusion: The MDLS model offers a robust and precise tool for predicting breast cancer prognosis and tailoring personalized treatment strategies. Its integration of multi-omics and machine learning highlights its potential clinical applications, particularly in improving the effectiveness of immunotherapy and identifying therapeutic targets for high-MDLS patients.

Abstract:Each year number of deaths is increasing extremely because of breast cancer. It is the most frequent type of all cancers and the major cause of death in women worldwide. Any development for prediction and diagnosis of cancer disease is capital important for a healthy life. Consequently, high accuracy in cancer prediction is important to update the treatment aspect and the survivability standard of patients. Machine learning techniques can bring a large contribute on the process of prediction and early diagnosis of breast cancer, became a research hotspot and has been proved as a strong technique. In this study, we applied five machine learning algorithms: Support Vector Machine (SVM), Random Forest, Logistic Regression, Decision tree (C4.5) and K-Nearest Neighbours (KNN) on the Breast Cancer Wisconsin Diagnostic dataset, after obtaining the results, a performance evaluation and comparison is carried out between these different classifiers. The main objective of this research paper is to predict and diagnosis breast cancer, using machine-learning algorithms, and find out the most effective whit respect to confusion matrix, accuracy and precision. It is observed that Support vector Machine outperformed all other classifiers and achieved the highest accuracy (97.2%).All the work is done in the Anaconda environment based on python programming language and Scikit-learn library.
