Abstract:5'-Guanosine monophosphate (5'-GMP) is one main source of freshness in broths. Herein, an electrochemical platform based on a novel ternary nanocomposite glassy carbon electrode modified with advantageously-united gold nanoparticles, 2,2'-bipyridine hydrated ruthenium (Ru(bpy)2Cl2) and sulfonated multi-walled carbon nanotubes (SMWCNTs)was prepared and used to detect 5'-GMP. After conditions optimization, the best performance of the electrochemical sensor was found in acidic media, including high specificity, sensitivity and selectivity. The electrochemical sensor exhibited a wide linear range under the optimal conditions. The enhanced sensitivity of this sensor was attributed to the Ru(bpy)2Cl2 and functionalized SMWCNTs that provided high electrical conductivity and electrocatalytic properties during electrochemical reaction. Precise analysis of 5'-GMP in actual broth samples showed satisfactory recovery. Thus, the sensor can be used in the market and food enterprises.
[Role of Ommaya reservoir in the management of neonates with post-hemorrhagic hydrocephalus].

Abstract:OBJECTIVE Intra-ventricular hemorrhage (IVH) is one of the most serious complications of preterm infants. Significant numbers of the surviving infants with severe IVH go on to develop post-hemorrhagic hydrocephalus (PHH). The management of PHH remains a very challenging problem for both neonatologists and pediatric neurosurgeons. This study aimed to evaluate the efficacy and safety of the use of Ommaya reservoirs and serial cerebrospinal fluid (CSF) drainage in the management of a series of neonates with PHH. METHOD Between January 1, 2003 and December 30, 2005, 15 consecutive newborn infants with IVH grades III to IV, complicated with progressive ventricular dilatation, underwent placement of an Ommaya reservoir. CSF was intermittently aspirated percutaneously from the reservoir. The amount and frequency of CSF aspiration were based on the clinical presentation and the follow-up results of serial cranial ultrasonograms or CT scans. The changes of CSF cell counts and chemistries were also followed. Patients whose progressive ventricular dilatation persisted despite serial CSF aspiration through Ommaya reservoir eventually had ventriculo-peritoneal shunts (V-P shunt) placed. All the patients were followed up in the outpatient clinic after discharge from the hospital and the neurodevelopmental outcomes were evaluated through 18-36 months of age. RESULT A total of 15 infants were included in this series. Of them, 11 were preterm infants who were at gestational ages of 29 to 34 weeks and 4 infants were full-term. All of the 4 full term infants presented with progressive ventricular dilatation after suffering from the intra-cranial hemorrhage (3 infants were due to vitamin K deficiency and 1 was due to birth trauma). Thirteen infants had grade III IVH, and 2 had grade IV IVH based on initial cranial ultrasonographic and CT scans. The mean age when IVH was diagnosed was (9 +/- 1) days in preterm infants and (22 +/- 7) days in full-term infants; the mean age when Ommaya reservoir was placed was (18 +/- 11) days in preterm infants and (31 +/- 7) days in full-term infants. All the infants tolerated the surgical procedure well. The Ommaya reservoir was tapped for an average of (21.5 +/- 4.6) times per patient. The mean CSF volume per tap was (10.2 +/- 1.3) ml/kg. The values of CSF protein, glucose and cell counts slowly reached normal levels at approximately 3 - 5 weeks after the placement of the reservoir. The velocity of head circumference increase per week was less than 1 cm in 13 patients in 1 - 4 weeks after the placement of the reservoir and the size of ventricles decreased gradually. By 12 - 18 months, 12 infants had normal size ventricles, and 1 patient still had mild ventricular dilation at 36 months. Two infants developed progressive hydrocephalus after serial CSF aspiration through Ommaya reservoir. One infant had a V-P shunt placed at 2 months of age and another infant died at 3 months of age at home after parents refused further therapy. Complications consisted of reservoir leaking and CSF infection at 16th day of placement in one patient after repeated tapping. By the end of 18 - 36 months of follow-up, 11 of 14 infants were considered normal, two patients had mild impairment in neurodevelopmental outcome (both had spastic bilateral lower limbs paresis, and one of whom also had amblyopia) and the other had seizure disorder. CONCLUSION The results from this series indicate that the placement of an Ommaya reservoir is relatively safe in newborn infants and is useful in the initial management of neonates with PHH and may be beneficial in improving their neurodevelopmental outcomes. A multicenter randomized trial may be needed to further validate the results of this report.

Abstract:Many species of copepods form dense aggregations, known as swarms. In the laboratory, we experimentally induced 5 different species of copepod to swarm in response to a point source of light. To map out the (X, y, z, t ) positions of swarm members. 2 right-angle views of the 3-dimensional swarm were videotaped. Since images of individual copepods appear indistinguishable on the paired 2-dixnensional projections, an algorithm was developed which matched the temporal changes of the vertical (z) positions of all images from the 2-dimensional projections of the 3-d~mensional copepod movement to produce (X, y, z, t ) positions of each individual. With the temporal/spatial positional data of swarm members, we tested the hypothesis that the fluid disturbance surrounding individual moving copepods. rather than the exoskeleton, maintains minimum separation distance. As the density of the swarm increased, the average nearest-ne~ghbor distance NND decreased, as did the mean mlnimum NND (MNND). For 3 of the 5 species, the MNND was significantly greater than that predicted from a random distribution, and was greater than twice the antennule or prosome length. While occasional physical contact may occur, resulting in escapes or attempted mating, it appears that most swarm members remain outside the f~e ld of self-generated fluid motion in the boundary layers surrounding their neighbors.

The survey indicated widely spread resistance to DDT; in a few cases ineffectivity of permethrin, tetramethrin and trichlorfon, respectively, was also documented.

Abstract:Most conducting polymer/graphene composites have excellent electrical conductivity. However, the background currents of these composites modified electrodes are much larger. In order to improve the sensitivities of these methods, it is necessary to decrease the background signal. In this paper, porous structure films of overoxidized polypyrrole/graphene (PPyox/GR) have been electrochemically coated onto glassy carbon electrode (GCE) and successfully utilized as an efficient electrode material for the quantitive detection of adenine and guanine, two of the most important components of DNA and RNA. The permselective polymer coatings with low background current could improve the selectivity and sensitivity of microelectrodes for the electropositive purine bases. The GRs into these polymers would further improve sensitivity by increasing the electroactive surface area. The electrochemical sensor can be applied to the quantification of adenine and guanine with a linear range covering 0.06–100µM and 0.04–100µM, and a low detection limit of 0.02μM and 0.01μM, respectively. More importantly, the proposed method was applied to quantify adenine and guanine in calf thymus DNA with satisfactory results.

Abstract:The common quantification methods of purines, including high-performance liquid chromatography, micellar electrokinetic chromatography, reverse phase ion pair chromatography, and capillary zone electrophoresis, have been used for the analysis of purines. These conventional techniques are limited by long operation times, large sample volumes, requirements for pretreatment and separation processes, or sophisticated, bulky and expensive equipment. Here, we have developed a simple method to prepare a probe for the quantitative analysis of purines using the surface-enhanced Raman scattering (SERS) method. The SERS probe is a rough Ag wire obtained with simple electrochemical treatment, which is optimized to reach the maximum SERS signal enhancement. The Raman enhancement factor (EF) of Ag wire after roughening can reach ∼106. Under optimized conditions, the rough Ag wire is capable of recognizing four purine bases (namely, guanine, adenine, hypoxanthine, and xanthine) in the range of 10−5 to 10−1 mg/mL with a linear relationship between SERS intensity of signature peak and purine concentration. The recovery rate of each purine base is higher than 80% in the mixture of four purines. The overall method was successfully applied to the monitoring of the level of purines in marketplace beer beverages. The novel wire-based SERS sensor used with portable Raman spectroscopy is prospective in diet safety and personalized point-of-care medical detection.

Abstract:Aiming at substantially improving the electrochemical sensing performance for guanine, we bring out a novel high-efficiency electrocatalyst composed of hierarchical sea urchin-like Prussian blue@palladium core-shell heterostructures supported on nitrogen-doped reduced graphene oxide (PB-U@Pd/N-RGO). The said PB-U@Pd/N-RGO was synthesized by a new and facile wet-chemistry method without using any surfactant and template. More impressively, we demonstrate a new strategy for morphology-controlled synthesis of Prussian blue via simply tuning the ratio of N,N'-dimethyl formamide and H2O in the mixed solvent. In-depth electrochemical characterizations demonstrate the superior electrocatalytic performance of PB-U@Pd/N-RGO toward the oxidation of guanine, which benefits from the synergistic effect of component materials and its unique core-shell heterostructure. Consequently, the constructed PBU@Pd/N-RGO-based electrochemical sensor displays superb comprehensive performance for the guanine assay. To be specifical, the developed sensor exhibits wide detection range (0.01-85.0 mu W), low detection limit (2.6 nM), good reproducibility, long-term stability, high sensitivity and selectivity together with acceptable recovery (97.0-104.0%) for the assay of guanine in biological samples. Such outstanding comprehensive performance is amongst the top of reports for the electrochemical sensing of guanine, promising practical applications in biosensing fields. (C) 2019 Elsevier Ltd. Abstract:Nanozyme based on the silver nanoparticles (AgNPs) decorated highly water-oxygen stable phosphorene (BP) nanohybrid with phosphorus-doped porous carbon microspheres (P-PCMs) is applied as an intelligent sensing platform for the electrochemical detection of 8-hydroxy-2′-deoxyguanosine (8-OHdG) as a biomarker of oxidative DNA damage in human urine sample. The density functional theory (DFT) is use for investigating the effect of silver on the conductivity level of BP and discussing a possible mechanism for the electrocatalytic oxidation of 8-OHdG. The Ag+ is in-situ reduced as AgNPs that grows onto the BP surface for the controllable preparation of BP-AgNPs with effective surface passivation among different metal ions decorated BP. P-PCMs with 4.9-fold enhancement in specific surface area (1636.73 m2 g−1) are prepared by hydrothermal carbonization of α-cyclodextrin as carbon sources, then calcinate in the presence of phosphoric acid as an activator and dopant. P-PCMs-BP-AgNPs are prepared by the ultrasound-assisted liquid-phase exfoliation with the addition of Ag+, and both P-PCMs and black phosphorus crystals are sufficiently grinded. The P-PCMs-BP-AgNPs nanohybrid displays good long-term water-oxygen stability, extraordinary specific surface area, superior electrocatalytic capacity with 303-fold enhancement, enzyme-like characteristics with <math>Imax</math> of 100 μA and <math>Km</math> of 29 μM. The machine learning (ML) model with artificial neural network (ANN) algorithm is employed for the intelligent output of 8-OHdG in real sample with acceptable recovery in work range from 0.2 to 125 μM.

Abstract:Taking advantage of strand-displacement DNA polymerization and parallel-motif DNA triplex system as dual amplifications, a new electrochemical label-free integrated aptasensor based on silver microspheres (SMSs) as a separation element and graphene-mesoporous silica-gold nanoparticle (NP) hybrids (GSGHs) as an enhanced element of the sensing platform was first reported. In this sensing design (schematic representation of the sensing procedure for adenosine triphosphate detection, Scheme 1 in manuscript text), which contains an enhanced three-step magnification process, SMSs with "clean" surface were first used to separate the undesirable aptamer and aptamer-adenosine triphosphate (ATP) complex attached on SMSs surface after aptamer-ATP interaction, which lead to the detachment of blocker DNA into the solution phase. Then, under the assistance of blocker DNA, an amplified method based on the inherent signal-transduction mechanism of the hairpin probe and strand-displacement property of DNA polymerase was introduced. The obtained duplex DNA was used to hybridize with an acceptor DNA assembled on electrode to form triplex DNA, which could bring a more obvious detection signal compared with the duplex DNA without the amplification. The electrochemical signal came from the GSGH-based enhanced sensing interface containing positively charged ferrocene-appended poly(ethyleneimine) (Fc-PEI). Using the above multiple effects, we could achieve the sensitive analysis of a model small molecule-ATP (an important "molecular currency" of intracellular energy transfer) in a wide detection range from 0.05 nM to 56.5 nM with the detection limit of 0.023 nM.
Highly selective detection of adenine and guanine by NH2-MIL-53(Fe)/CS/MXene nanocomposites with excellent electrochemical performance

Abstract:Adenine (A) and guanine (G) are mainly found in deoxyribonucleic acid (DNA) and ribonucleic acid (RNA) and play a crucial role in genetic information transfer and protein synthesis. In this study, NH2-MIL-53(Fe)/CS/MXene nanocomposites were prepared for detecting guanine and adenine. With high specific surface area, excellent water dispersion, and numerous active sites, MXene (transition metal carbides, nitrides, and carbonitrides) provides a good platform for loading primitive metal-organic frameworks (MOFs). At the same time, the problem of poor conductivity and dispersion of MOFs is solved. The electrochemical catalytic oxidation of adenine and guanine of NH2-MIL-53 (Fe)/CS/MXene nanocomposites was carried out by differential pulse voltammetry (DPV). Operating voltage of DPV: 0.7-0.9 V (vs. Ag/AgCl) for G, 1.0-1.2 V (vs. Ag/AgCl) for A, 0.8 V (vs. Ag/AgCl), and 1.1 V (vs. Ag/AgCl) for G and A. The concentration ranges for detecting A and G were 3-118 μM and 2-120 μM with detection limits of 0.57 μM and 0.17 μM (S/N = 3), respectively. The nanocomposite was used for detecting G and A in herring sperm DNA, and the content of G and A was found to be about 9 and 11 μM; the RSD values were 3.4 and 1.3%, respectively.

Abstract:A novel poly(bromocresol green) (PBG)/carbon quantum dots (CQDs) composite based sensor was successfully fabricated on glassy carbon electrode by electropolymerization. As-prepared PBG/CQDs modified electrode was successfully applied in the simultaneous determination of guanine and adenine. Electrochemical responses of as-prepared modified electrode were evaluated by cyclic voltammetry and differential pulse voltammetry. Owing to the synergistic effect of PBG and CQDs, the oxidation peaks of both guanine and adenine well separated from each other and the peak potential separation was large up to 310mV. The current of the oxidation peaks showed a linear dependence on the concentrations of guanine and adenine in the range of 0.50–142 and 0.30–130μM with detection limits of 0.016 and 0.026μM (S/N=3), respectively. Moreover, as-prepared PBG/CQDs modified electrode exhibited favorable selectivity, stability and reproducibility, which had promising application for the simultaneous determination of guanine and adenine.

Abstract:It is of great significance to propose simple methods to detect DNA bases sensitively for biological analysis and medical diagnosis. Herein, a highly crystalline polyimide covalent organic framework (TAPM-COF) has been successfully synthesized via a solvothermal route using pyromellitic dianhydride (PMDA) and tris(4-aminophenyl) amine (TAPA), which possessed large specific surface area (2286 m 2 g −1 ) and excellent thermal stability. Intriguingly, the crystallinity of the TAPM-COF improved significantly with the increase of water content in the reaction medium. To verify this phenomenon, we synthesized TPPM-COF with two pores by pyromellitic dianhydride (PMDA) and N,N,N′,N'-tetrakis(4-aminophenyl)-1,4-benzenediamine (TPDA), which bonding was similar to TAPM-COF. Furthermore, the prepared TAPM–COF–0.3 was used to construct a novel and independent electrochemical biosensor on glassy carbon electrode for simultaneously determination of adenine (A) and guanine (G) without other additives. However, to further improve signal of TAPM-COF in the field of electrochemical sensors, the crystalline TAPM–COF–0.3 can be readily integrated with amino-functionalized multiwalled carbon nanotubes (NH 2 -MWCNT) to form core-shell TAPM–COF–0.3@NH 2 -MWCNT driven by a π–π stacking interaction for more sensitive electrochemical sensing toward purine bases. In comparison to TAPM-COF/GCE, the TAPM-COF@NH 2 -MWCNT/GCE exhibited more favorable linear range and lower limit of detection. The work provided a new strategy for amplifying signal of COF in the field of electrochemical sensors.

Abstract:In this work, a novel sensor based on gold nanoparticle-modified zirconium-metal-organic frameworks (MOF-801) and carboxyl-functionalized carbon nanotube (MWCNT-COOH) composite material was built successfully for electrochemical detection of guanine and adenine in DNA sample, which exhibits diverse advantages such as fast, convenience and high efficiency. Guanine and adenine were detected by the cyclic voltammetry (CV) and differential pulse voltammetry (DPV). Under the optimal conditions, MOF-801/MWCNT-COOH/ AuNPs/GCE delivered linear DPV responses to guanine in the range of 0.8 ~ 60, 60 ~ 180 μM and the adenine in the range of 0.8 ~ 60, 60 ~ 180 μM with the low detection limit of 0.09 μM for guanine and 0.08 μM for adenine, respectively. The advantages of this method include simplicity, rapidity, high sensitivity, good selectivity as well as low cost. The modified GCE was successfully applied to the analysis of guanine and adenine in DNA samples.

Abstract:Herein, we report developing an electrochemical sensor based on a glassy carbon electrode modified by bismuth nanoparticles, graphene oxide, iron oxide, and poly-methyldopa namely Bi@Fe 3 O 4 -PMDA/GO/GCE for detecting guanine and adenine. Under optimized conditions (5 μL of Fe 3 O 4 -PMDA/GO, 0.5 mg mL −1 of Fe 3 O 4 -PMDA/GO solution, water as solvent of Fe 3 O 4 -PMDA/GO and acetate buffer (0.1 M, pH 6) as electrolytes), the electrochemical behaviors of guanine and adenine on the prepared modified electrode were investigated by cyclic voltammetry and differential pulse voltammetry. With a high specific surface area and numerous active sites, Bi@Fe 3 O 4 -PMDA/GO/GCE exhibited outstanding electrocatalytic properties enabling the determination of guanine and adenine over a wide concentration range with the low detection limit. The Bi@Fe 3 O 4 -PMDA/GO/GCE possessed the advantages of simplicity, speed, good sensitivity, and anti-interference performance. Using the DPV method, the resulting sensor exhibited an excellent response with a wide linear ranges from 0.5 to 300 μM for both analytes with LODs 0.027 and 0.032 μM for adenine and guanine, respectively. The designed electrode was satisfactorily employed for the analysis of the real sample. Therefore, Bi@Fe 3 O 4 -PMDA/GO/GCE demonstrating sufficient selectivity and sensitivity for the individual and simultaneous study could be applied in widespread fields, including biotechnology or microbiology.

Sensitive chemically amplified electrochemical detection of ruthenium tris-(2,2′-bipyridine) on tin-doped indium oxide electrode

Abstract:Optimized combination of chemical agents was selected for sensitive electrochemical detection of dissolved ruthenium tris-(2,2′-bipyridine) (Ru-bipy). The detection was based on the chemical amplification mechanism, in which the anodic current of a redox-active analyte was amplified by a sacrificial electron donor in solution. On indium-doped tin oxide (ITO) electrodes, electrochemical reaction of the analyte was reversible, but that of the electron donor was greatly suppressed. Several transition metal complexes, such as ferrocene and tris-(2,2′-bipyridine) complexes of osmium, iron and ruthenium, were evaluated as model analyte. A correlation between the amplified current and the standard potential of the complex was observed, and Ru-bipy generated the largest current. A variety of organic bases, acids and zwitterions were assessed as potential electron donor. Sodium oxalate was found to produce the largest amplification factor. With Ru-bipy as the model analyte and oxalate as the electron donor, the analyte concentration curve was linear up to 50μM, with a lower detection limit of approximately 50nM. Preliminary work was presented in which a Ru-bipy derivative was attached to bovine serum albumin and detected electrochemically. Although the combination of Ru-bipy, oxalate and ITO electrode has been used before for electrochemiluminescent detection of Ru-bipy and oxalate, as well as electrochemical detection of oxalate, its utility in amplified voltammetric detection of Ru-bipy as a potential electrochemical label has not been reported previously.

Abstract:An electrochemiluminescent (ECL) aptasensor for lysozyme (Lyz) based on a graphene quantum dot (GQD)-ruthenium bipyridine (Ru(bpy) 3 2+ )-silica (GQDs-Ru(bpy) 3 2+ @SiO 2 ) nanocomposite were prepared by encapsulating GQDs and the luminophore Ru(bpy) 3 2+ in silica nanospheres (SiO 2 ). The nanocomposite GQDs-Ru(bpy) 3 2+ @SiO 2 with uniform size exhibited a strong and reproducible ECL response. The nanoprobe (DNA-GQDs-Ru(bpy) 3 2+ @SiO 2 ) was obtained via amidation between GQDs-Ru(bpy) 3 2+ @SiO 2 and the complementary DNA sequence. The ECL platform was constructed by hybridization between the aptamer and the complementary strand DNA. The specific recognition between aptamer and the target lysozyme maintained the nanoprobe DNA-GQDs-Ru(bpy) 3 2+ @SiO 2 away from the electrode, which led to the decreasing ECL response. The aptasensor exhibited a linear relationship from 0.1 to 100 pg/mL with a detection limit of 1.7 × 10 −11 mg/mL. This study provided a simple and sensitive analytical approach for lysozyme.

Abstract:In the present paper, we report that CdSe quantum dots (QDs) can act as the coreactant of Ru(bpy) 3 2+ electrogenerated chemiluminescence (ECL) in neutral condition. Strong anodic ECL signal was observed at ~1.10 V at CdSe QDs modified glassy carbon electrode (CdSe/GCE), which might be mainly attributed to the apparent electrocatalytic effect of QDs on the oxidation of Ru(bpy) 3 2+ . Ru(bpy) 3 2+ can be intercalated into the loop of hairpin DNA through the electrostatic interaction to fabricate a probe. When the probe was bound to the CdSe QDs modified on the GCE, the intense ECL signal was obtained. The more Ru(bpy) 3 2+ can be intercalated when DNA loop has larger diameter and the stronger ECL signal can be observed. The loop of hairpin DNA can be opened in the presence of target DNA to release the immobilized Ru(bpy) 3 2+ , which can result in the decrease of ECL signal. The decreased ECL signal varied linearly with the concentration of target DNA, which showed the ECL biosensor can be used in the sensitive detection of DNA. The proposed ECL biosensor showed an excellent performance with high specificity, wide linear range and low detection limit.
