Abstract:Corydalis turtschaninovii (CT) is a traditional Chinese medicine which is known to have analgesic effects, and is under investigation for the management of chronic pain. Our study aims to establish a UPLC–MS/MS method for pharmacokinetic determination of nine bioactive alkaloids of raw and processed CT in rat plasma. Nitidine chloride was selected as internal standard. After protein precipitation with methanol, the plasma samples were separated on a reversed phased column with a mobile phase of acetonitrile and water (including 0.1% formic acid). The MRM parameters were optimized as follows: m/z 354.0 → 188.1 for protopine, m/z 321.0 → 293.1 for coptisine, m/z 371.1 → 189.1 for allocryptopine, m/z 357.1 → 193.3 for tetrahydropalmatine, m/z 324.1 → 176.1 for tetrahydrocoptisine, m/z 340.1 → 176.2 for tetrahydroberberine, m/z 368.1 → 289.1 for corynoline, m/z 370.5 → 192.1 for corydaline, m/z 367.2 → 351.2 for dehydrocorydaline, and m/z 406.0 → 300.3 for the IS. The linearity, accuracy, precision, stability, recovery and matrix effect of the method were well validated. This method was successfully employed for a pharmacokinetic study of raw and vinegar-processed CT in rats. The absorption of the nine alkaloids was accomplished in an hour. The double peak phenomenon of the nine alkaloids may be ascribed to enterohepatic recirculation. Compared with the raw group, AUC0→t and Cmax of the nine alkaloids were significantly elevated in the vinegar-processed group. Our findings suggest that vinegar-processing could increase the bioavailability of the nine alkaloids of CT in rats. The pharmacokinetic information obtained will provide basis for application of processed CT in future clinical therapy.
The comparative pharmacokinetic study of Yuanhu Zhitong prescription based on five quality-markers

Abstract:Background: According to the compatibility theory, therapeutic effects of Chinese medicine prescription are generally attributed to the synergism of multi-herbs. Quality-markers, as the crucial effective components, play a key role in the interaction of compatibility. Pharmacokinetic studies could illustrate the interaction between multiple components in dynamics perspective. Purpose: This study aims to establish a rapid, reliable and sensitive ultra performance liquid chromatography coupled with tandem mass spectrometry method for simultaneous determination of corydaline, tetrahydropalmatine, protopine, imperatorin and isoimperatorin (quality-markers of Yuanhu Zhitong prescription) in rat plasma, and then applied to the comparative pharmacokinetic study for clearing the interaction of compatibility in Yuanhu Zhitong prescription. Methods: Five quality-markers were separated on a Waters Acquity UPLC BEH C18 column (2.1 mm × 50 mm, 1.7 µm) by gradient elution with 0.1% formic acid-water and 0.1% formic acid-acetonitrile. Detection was performed in the positive ionization and multiple reaction monitoring mode. The analytical method was validated and successfully applied to the comparative pharmacokinetic study after oral administration of Yuanhu Zhitong prescription and single-herb extracts. Results: Calibration curves showed good linearity over the concentration ranges of 0.25-500 ng/ml for corydaline, tetrahydropalmatine and isoimperatorin, 0.1-200 ng/ml for protopine, and 0.5-500 ng/ml for imperatorin, respectively. Compared with Rhizoma corydalis group, AUC0-t and AUC0-∞ significantly increased (p < 0.01 for corydaline and tetrahydropalmatine, and p < 0.05 for protopine) after oral administration of Yuanhu Zhitong prescription extract. Meanwhile, Cmax of corydaline and tetrahydropalmatine increased remarkably, from 93.00 µg/l to 196.35 µg/l for corydaline (p < 0.05) and 181.62 µg/l to 311.22 µg/l for tetrahydropalmatine (p < 0.01). In addition, MRT0-t and MRT0-∞ of corydaline, as well as Tmax of protopine in Yuanhu Zhitong prescription group were obviously delayed compared to Rhizoma corydalis group (p < 0.01). Conclusion: These achieved results indicated that the compatibility of Rhizoma corydalis and Radix Angelicae dahuricae lead to greater absorption of corydaline, tetrahydropalmatine and protopine, which would be help to better understand the compatibility mechanism of Yuanhu Zhitong prescription.
Development and Validation of a HPLC-ESI-MS/MS Method for Simultaneous Quantification of Fourteen Alkaloids in Mouse Plasma after Oral Administration of the Extract of Corydalis Yanhusuo Tuber: Application to Pharmacokinetic Study

Abstract:The tuber of Corydalis yanhusuo is a famous traditional Chinese medicine and found to have potent pharmacological effects, such as antinociceptive, antitumor, antibacterial, anti-inflammatory, and anti-depressive activities. Although there are several methods to be developed for the analysis and detection of the bioactive ingredients' alkaloids, so far, only few prominent alkaloids could be quantified, and in vitro and in vivo changes of comprehensive alkaloids after oral administration are still little known. In this study, we first developed a simple and sensitive high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (HPLC-ESI-MS/MS) method to quantify the comprehensive alkaloids of extracts of C. yanhusuo in mouse plasma, using nitidine chloride as an internal standard. As results, at least fourteen alkaloids, including an aporphine (oxoglaucine), a protopine (protopine), five tertiary alkaloids (corydaline, tetrahydroberberine, tetrahydropalmatine, tetrahydrocolumbamine, and tetrahydrocoptisine) and seven quaternary alkaloids (columbamine, palmatine, berberine, epiberberine, coptisine, jatrorrhizine, and dehydrocorydaline) could be well quantified simultaneously in mouse plasma. The lower limits of quantification were greater than, or equal to, 0.67 ng/ mL, and the average matrix effects ranged from 96.4% to 114.3%. The mean extraction recoveries of quality control samples were over 71.40%, and the precision and accuracy were within the acceptable limits. All the analytes were shown to be stable under different storage conditions. Then the established method was successfully applied to investigate the pharmacokinetics of these alkaloids after oral administration of the extract of Corydalis yanhusuo in mice. To the best of our knowledge, this is the first document to report the comprehensive and simultaneous analyses of alkaloids of C. yanhusuo in mouse plasma. It was efficient and useful for comprehensive pharmacokinetic and metabolomic analyses of these complex alkaloids after drug administration.

Abstract:BackgroundYanhusuo (Corydalis yanhusuo W.T. Wang; YHS), is a well-known traditional Chinese herbal medicine, has been used in China for treating pain including chest pain, epigastric pain, and dysmenorrhea. Its alkaloid ingredients including tetrahydropalmatine are reported to inhibit cytochromes P450 (CYPs) activity in vitro. The present study is aimed to assess the potential of total alkaloid extract (TAE) from YHS to effect the activity and mRNA levels of five cytochromes P450 (CYPs) in rat.MethodsRats were administered TAE from YHS (0, 6, 30, and 150 mg/kg, daily) for 14 days, alanine aminotransferase (ALT) levels in serum were assayed, and hematoxylin and eosin-stained sections of the liver were prepared for light microscopy. The effects of TAE on five CYPs activity and mRNA levels were quantitated by cocktail probe drugs using a rapid chromatography/tandem mass spectrometry (LC-MS/MS) method and reverse transcription-polymerase chain reaction (RT-PCR), respectively.ResultsIn general, serum ALT levels showed no significant changes, and the histopathology appeared largely normal compared with that in the control rats. At 30 and 150 mg/kg TAE dosages, an increase in liver CYP2E1 and CYP3A1 enzyme activity were observed. Moreover, the mRNA levels of CYP2E1 and CYP3A1 in the rat liver, lung, and intestine were significantly up-regulated with TAE from 6 and 30 mg/kg, respectively. Furthermore, treatment with TAE (150 mg/kg) enhanced the activities and the mRNA levels of CYP1A2 and CYP2C11 in rats. However, the activity or mRNA level of CYP2D1 remained unchanged.ConclusionsThese results suggest that TAE-induced CYPs activity in the rat liver results from the elevated mRNA levels of CYPs. Co-administration of prescriptions containing YHS should consider a potential herb (drug)–drug interaction mediated by the induction of CYP2E1 and CYP3A1 enzymes.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Corydalis yanhusuo is a well-known Chinese herbal medicine that is commonly applied as an analgesic agent in clinic using for thousands of years. Resent research showed that the quaternary ammonium alkaloids from Corydalis yanhusuo have a significant effect on myocardial ischemia. However, the corresponding anti-myocardial ischemia components that represent the efficacy of the quaternary ammonium alkaloids have not been elucidated.AIM OF THE STUDY: Explore the anti-myocardial ischemia components of Corydalis yanhusuo and develop a method of quality control for Corydalis yanhusuo. Chemical fingerprints of quaternary ammonium alkaloids extracted from Corydalis yanhusuo samples from 37 different sources were identified using UPLC-Q-TOF MS. The protective effects of the 37 samples with respect to H9C2 cell hypoxia-reoxygenation were detected by MTT assays. The fingerprint-efficacy relationship between the chemical fingerprints and cardioprotection afforded by Corydalis yanhusuo was investigated using three chemometric methods.RESULTS: Because of their inherent differences in chemical compositions, the protective effects to H9C2 cell hypoxia-reoxygenation were different. The results of three chemometric methods showed that the source of the Corydalis yanhusuo has an important influence on both the chemical fingerprint and efficacy. In particular, dehydrocorybulbine, 13-methyldehydrocorydalmine, dehydrocorydaline, columbamine, and palmatine appear to be the main effective components for quality control of this TCM.CONCLUSION: This work provides a general model of combination of UPLC and cardioprotection efficiency to study the fingerprint-efficacy relationship of Corydalis yanhusuo which can offer some references for detecting principal components of Corydalis yanhusuo on cardioprotection efficiency. Fingerprint-efficacy studies also provide a powerful method of quality control in Corydalis yanhusuo and other TCMs.

Abstract:Corydalis yanhusuo W. T. Wang, also known as yanhusuo , yuanhu , yanhu and xuanhu , is one of the herb components of many Chinese Traditional Medicine prescriptions such as Jin Ling Zi San and Yuanhu-Zhitong priscription. C. yanhusuo was traditionally used to relieve pain and motivate blood and Qi circulation. Now there has been growing interest in pharmacological effects of alkaloids, the main bioactive components of C. yanhusuo . Eighty-four alkaloids isolated from C. yanhusuo are its important bioactive components and can be characterized into protoberberine alkaloids, aporphine alkaloids, opiate alkaloids and others and proper extraction or co-administration methods modulate their contents and efficacy. Alkaloids from C. yanhusuo have various pharmacological effects on the nervous system, cardiovascular system, cancer and others through multiple molecular mechanisms such as modulating neurotransmitters, ion channels, gut microbiota, HPA axis and signaling pathways and are potential treatments for many diseases. Plenty of novel drug delivery methods such as autologous red blood cells, self-microemulsifying drug delivery systems, nanoparticles and others have also been investigated to better exert the effects of alkaloids from C. yanhusuo . This review summarized the alkaloid components of C. yanhusuo , their pharmacological effects and mechanisms, and methods of drug delivery to lay a foundation for future investigations.

Abstract:Corydalis saxicola Bunting, also named Yanhuanglian in China, is widely used in folk prescriptions to treat hepatitis, hepato-cirrhosis and hepatic cancer. Its active components consists mainly of alkaloids, which have poor solubility in water. The previous study showed that the absolute bioavailabilities (BA) of the alkaloids were only about 10% after oral administration to rats, compared to intravenous administration. In this paper, the solid dispersion (SD) method was used to improve the BAs of dehydro-cavidine (YHL-1), coptisine (YHL-2), and dehydroapocavidine (YHL-3), the main alkaloids of the extract of Corydalis saxicola Bunting, by improving their solubility. The results showed that using PEG 4000 as a carrier at an amount equivalent to 7 folds of the extract could significantly improve the solubility of the compounds in water, pH 1.2 HCl and pH 7.2 PBS. DSC and X-ray detection indicated that the drug might be in a dissolved or amorphous state in SD, which was different from that in the physical mixture. The formulation was further studied in terms of BA enhancement by cumulative urinary excretion after oral administration to rats. A new method using high performance liquid chromatography (HPLC) coupled to a diode-array detector (DAD) was established. The results showed that the relative BAs (RE BAs) of YHL-1, YHL-2 and YHL-3 were all significantly improved by about 1 fold after oral administration of SD solution, compared to the extract suspension (p < 0.05).

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE:Tianshu capsule (TSC) is a traditional Chinese medicine (TCM) preparation and has significant clinical effect on migraine. The composition characteristics of TSC formula are worth exploring for the treatment of migraine. AIM OF THE STUDY:Identify the compounds in vivo after oral administration of TSC, Gastrodin (GAS), ferulic acid, senkyunolide G and senkyunolide I in the blood of healthy and migraine rats were used as representative compounds for time-dependent processes investigation. And we focus on explaining the characteristics of TSC treatment on migraine from a pharmacokinetic perspective. MATERIALS AND METHODS:In this study, Ultra-high performance liquid chromatography with Orbitrap tandem mass spectrometry (UPLC-Orbitrap-MS/MS) system were used to detect the compounds in rat plasma and brain after oral administration of TSC. A sensitive, selective and reliable ultra-performance liquid chromatography coupled with triple quadrupole mass spectrometry (UPLC-TQ-MS/MS) system method was established for the simultaneous quantitative analysis of multi-components and evaluate the pharmacokinetic behavior of four main compounds. RESULTS:A total of 46 compounds were significantly identified in vivo of rat, including 35 compounds in plasma, 7 compounds only in brain and 4 compounds both in plasma and brain. The quantitative method has been confirmed to be feasible for multi-components content determination, pharmacokinetic parameters indicated that the four compounds absorbed into blood rapidly, and senkyunolide I also cross the blood-brain barrier (BBB) quickly into brain. GAS has relatively high concentrations in plasma, and the parameters AUC0-t, CLz/F displayed significant differences between the normal and migraine rats. And the AUC0-t and Cmax of GAS and ferulic acid exhibited dose-dependent way. CONCLUSIONS:we added compounds for the pharmacokinetic study of TSC, providing powerful help for clinical medication in the treatment of migraine.
An ultra-high-performance liquid chromatography-tandem mass spectrometry method for the simultaneous quantitation of 10 alkaloids of Corydalis Decumbentis Rhizoma preparation in dog plasma and its application to a pharmacokinetic study

Abstract:Objective: A sensitive and rapid ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was successfully applied to the determination of 10 alkaloids in beagle dog plasma following a single oral dose of Xiatianwu capsules and enteric-coated capsules, with theophylline serving as the internal standard (IS). Methods: Plasma samples were preprocessed using liquid-liquid extraction (LLE) with ethyl acetate ahead to separation using a gradient elution procedure on a Waters ACQUITY UPLC HSS T3 column (1.8 μm, 100 × 2.1 mm). The mobile phase was composed of 0.1% formic acid solution and acetonitrile at the flow rate of 0.3 ml/min. Multiple reaction monitoring (MRM) was used to determine the analytes in the positive ion mode. Results: The calibration curves for 10 analytes demonstrated a high degree of linearity (r ≥ 0.9920). The lower limit of quantification (LLOQ) values for 10 alkaloids were all more than 1.074 ng/ml, and matrix effects varied from 94.25% to 106.15%. The mean extraction recovery of quality control samples at low (LQC), medium (MQC) and high (HQC) concentrations, as well as IS, was all more than 76.60%. The intra-day and inter-day precision (RSD) also satisfied the requirement. Simultaneously, the variation of assay accuracies (RE) was between 13.05% and 9.38%. Conclusion: The test was validated in accordance with regulatory bioanalytical guidelines and was found to be suitable for use in a pharmacokinetic investigation of these compounds in beagle dogs after oral administration of Xiatianwu general capsules and enteric-coated capsules. The Cmax of 10 alkaloids ranged from 52.61 to 192.46 ng/ml after oral administration of Xiatianwu capsules, and from 67.50 to 247.36 ng/ml. The Tmax was between 0.59 and 1.33 h of Xiatianwu capsules, and between 1.08 and 2.00 h of enteric-coated capsules. The t1/2 ranged from 3.18 to 7.47 h of general capsules, and from 6.01 to 11.36 h. AUC0-t ranged from 181.06 to 722.74 ng·h/ml of Xiatianwu capsules, and from 275.03 to 884.17 ng·h/ml of enteric-coated capsules. The Cmax of enteric-coated capsules were significantly increased except for tetrahydropalmatine and berberine. Tmax of general capsules were less than 1 h, and of enteric-coated capsules were less than 2 h. The t1/2 of dehydrocorydaline, palmatine, tetrahydrojatrorrhizine, jatrorrhizine and coptisine in enteric-coated capsules was longer than that in ordinary capsule. The AUC0-t and AUC0-∞ of bicuculline, dehydrocorydaline, protopine, magnoflorine, tetrahydrojatrorrhizine, jatrorrhizine, berberine and coptisine were all significantly higher in enteric-coated capsules.
Fu'cupping Physical Permeation-Enhancing Technique Enhances the Therapeutic Efficacy of Corydalis yanhusuo Gel Plaster

Abstract:Corydalis yanhusuo , a traditional Chinese medicine, is widely used to treat various pains, and its active ingredients are alkaloids. This study aimed to develop a new type of transdermal gel plaster containing the extract of C. yanhusuo . Studies have shown that Fu'cupping physical permeation-enhancing technique can promote the penetration of alkaloids and improve the efficacy of drugs. A transdermal gel plaster containing the extract of C. yanhusuo was prepared and optimized using an orthogonal experimental design. The skin permeation ability of the gel plaster was studied in vitro , while the anti-inflammatory and analgesic effects of the prepared patch alone or with Fu'cupping physical permeation-enhancing technique were evaluated in a rat model. The formulation of a gel plaster containing C. yanhusuo extract was successfully prepared with an optimized composition consisting of glycerin (15 g), sodium polyacrylate (2 g), silicon dioxide (0.3 g), ethanol (2 g), aluminum oxide (0.1 g), citric acid (0.05 g), the C. yanhusuo extract (3 g), and water (15 g). The cumulative transdermal permeation of dehydrocorydaline, corypalmine, tetrahydropalmatine, and corydaline in 24 h was estimated to be 569.7 ± 63.2, 74.5 ± 13.7, 82.4 ± 17.2, and 38.9 ± 8.1 μg/cm 2 , respectively. The in vitro diffusion of dehydrocorydaline and corydaline followed the zero-order kinetics profile, while that of corypalmine and tetrahydropalmatine followed a Higuchi equation. The prepared gel plaster significantly reduced paw swelling, downregulated inflammatory cytokines, and mitigated pain induced by mechanical or chemical stimuli. The Fu'cupping physical permeation-enhancing technique further improved the anti-inflammatory and analgesic effects of the patch. The combined application of the Fu'cupping physical permeation-enhancing technique and the alkaloid gel plaster may be effective against inflammation and pain. Abstract:Abstract Salvia miltiorrhiza Bunge is a Traditional Chinese Medicine used for the treatment of diseases related to the heart and liver. Owing to the abundance of salvianolic acids and tanshinones, S. miltiorrhiza exhibits various pharmacological activities, such as stimulating blood circulation, eliminating blood stasis, relieving menorrhea and pain, and improving polydipsia. Neocryptotanshinone (NCTS) is one of the tanshinones isolated from S. miltiorrhiza and exhibits anti‐inflammatory, hypoglycemic, and preventive effects against cardiovascular diseases; however, its pharmacokinetic properties have not been reported to date. Therefore, a simple and reliable LC–MS/MS method was developed to determine absorption, distribution, and excretion of NCTS in rat physiological samples. We demonstrated that NCTS had high oral absolute bioavailability of 85.96% ± 25.32%, and it was rapidly distributed in both the heart and brain, and the fecal excretion of NCTS was the highest. This study provides a theoretical basis for further development of NCTS as a potential clinical candidate.

Abstract:Second trimester maternal serum screening (MSS) is performed by measuring concentrations of alpha fetoprotein (AFP), unconjugated estriol (uE3), beta human gonadotropin (bhCG) and inhibin-A. This technology is commonly used to identify pregnancies at increased risk for trisomy 21, trisomy 18, open neural tube defects and most recently, steroid sulfatase deficiency ichthyosis, and the Smith–Lemli–Opitz Syndrome (Table I). We report here on, two children with the 22q11.2 deletion syndrome whose mothers both had MSS results consistent with an increased risk for trisomy 18. The first patient was a female infant transferred to our hospital for evaluation and management of multiple congenital anomalies. The child was born to a 24-year-old G II P II who had MSS with a 1 in 23 risk that the infant had trisomy 18. Following a normal level II ultrasound exam amniocentesis was declined. The child was born at 36 weeks gestation at less than the 10th centile for height, weight, and head circumference, with the following anomalies: overriding sutures, sclerocorneas, microphthalmia with colobomas, posteriorly rotated ears, bulbous nasal tip, imperforate anus, long fingers and postaxial polydactyly of the right hand and right foot. FISH probes documented the 22q11.2 deletion. The second childwasborn at 29weeks gestation to a gravida V para IV SAB I, 36-year-old woman who had MSS with a 1 in 40 risk of trisomy 18. An ultrasound at 20 weeks gestation was normal. A follow-up ultrasound 4 weeks later identified a congenital heart defect and an amniocentesis was done with normal results (46,XY). At birth he presented with an interrupted aortic arch type B, VSD, hypospadias, posteriorly rotated ears, and asymmetric crying facies. 22q11.2 FISH probe established the diagnosis. Approximately 1 in 200 women who have MSS will be positive for trisomy 18 [Yankowitz et al., 1998]. The incidence of 22q11.2 deletion has been reported to be between 1/4,000 [Wilson et al., 1994] and 1/ 6,395 [Devriendt et al., 1998]. If MSS for trisomy 18 was not also identifying fetuses at risk for 22q11.2 deletion syndrome, then the expected incidence of these two independent events occurring together would be between 1/800,000 and 1/1,279,000 screening tests. Given the small probability that these events would occur by chance, we suggest that MSS positive for trisomy 18 may also be identifying fetuses at risk for the 22q11.2 deletion. Abstract:HPLC-UV and HPLC-MS techniques were used in fingerprint analysis of the medicinal extract of Corydalis yanhusuo. An HPLC profile of medicinal extract of Corydalis yanhusuo was established as a characteristic fingerprint and employed to assess the consistency and differences between extracts. To determine the major constituents of medicinal extract of Corydalis yanhusuo for quality control, 12 major chromatographic peaks were characterized. To explore the in vivo metabolism of the major active constituents' rat plasma was analyzed by HPLC-MS following oral administration of medicinal extract of Corydalis yanhusuo at different time intervals. These results indicate that the quaternary alkaloids are significant for the biological activity of medicinal extract of Corydalis yanhusuo. It is concluded that the chemical fingerprint combined with the metabolic fingerprint is a useful means of controlling the quality of medicinal extract of Corydalis yanhusuo and clarifying the possible mechanism of action.

Abstract:Corynoxeine, a natural active alkaloid found in Genus Uncaria, has been reported to have anti-depressant effects. In this study, a sensitive and efficient ultra-high performance liquid chromatography tandem mass spectrometry method for quantifying corynoxeine in rat plasma and tissues was established, validated and applied to investigate the pharmacokinetics and tissue distribution differences between normal rats and chronic unpredictable mild stress (CUMS)-induced depression model rats following oral administration. All bio-samples were prepared by methanol protein precipitation method with theophylline as internal standard (IS). Chromatographic separation was conducted on an Agilent ZORBAX Eclipse Plus C18 column using mobile phase A (acetonitrile) and B (0.1% formic acid in water) in gradient elution mode with a flow rate of 0.3 mL/min. Mass spectrometric detection was performed in multiple-reaction monitoring mode with positive electrospray ionization source. The transitions of m/z 383.0→160.2 for corynoxeine and m/z 181.1→124.0 for IS were chosen for quantification. The method showed good linearity, stability, accuracy, precision, recovery, and non-significant matrix effect, which were within the acceptable ranges. The pharmacokinetic results revealed that the absorption and bioavailability of corynoxeine in depression rats decreased compared to normal rats. The tissue distribution of corynoxeine trended to be mostly in the intestine and stomach and the distribution of this compound in intestine tissue of depression rats was significantly increased compared to the normal rats. The pharmacokinetics and tissue distribution profiles of corynoxeine were altered in CUMS-induced depression rats compared to normal rats and these experimental findings could provide beneficial information to the mechanism research and clinical applications of corynoxeine.

Abstract:Shuanghua Baihe tablet is a traditional Chinese patent medicine which showed special advantages in the treatment of recurrent aphthous stomatitis. Scientists have improved and implemented the LC-MS/MS method, which is specific and sensitive, for comparative pharmacokinetics study of five alkaloids, including palmatine, berberine, epiberberine, jatrorrhizine and coptisine in rat plasma after oral administration of Rhizoma Coptidis extract and Shuanghua Baihe tablets. The results showed that Shuanghua Baihe tablets could promote the absorption of these five alkaloids and improved their bioavailability compared with R. Coptidis extract. To further investigate the related mechanism, everted intestinal sac model in vitro was used to indicate that alteration of in vivo pharmacokinetics of five alkaloids could be attributed to, at least in part, the absorption changes by coadministration of other herbs. These discoveries served as a theoretical basis for clinical use of Shuanghua Baihe tables.

Abstract:Xianglian pill, a TCM prescription, consists of Rhizome coptidis and Radix Aucklandiae, and has been used to treat gastrointestinal disease for many years. Berberine, jatrorrhizin, coptisine, and palmatine are four representative alkaloids in Xianglian pill. Knowing that the drug disposal process in vivo is closely related to the toxicity and efficacy of a drug, it is important to classify the disposal properties of these alkaloids. In this study, the pharmacokinetics and tissue distribution of the four alkaloids were investigated. The analytical samples were analyzed using validated HPLC‐MS/MS method. The results showed that the four alkaloids could be slowly absorbed. The Cmax values of berberine, jatrorrhizin, coptisine, and palmatine were 11.420, 2.287, 2.584, 3.102 ng/mL, respectively. Subsequently, the tissue distribution studies showed that they were quickly distributed to various tissues with rich blood flow in the body.

Abstract:Introduction Corydalis DC., the largest genus of Papaveraceae, comprises numerous species known for their abundant alkaloid content and historical use in clinical medicine. Recently, a new species of genus Corydalis named Corydalis huangshanensis Lu Q. Huang & H. S. Peng was discovered in the Huangshan Mountains of Anhui Province, China. Objective To compare the chemical characteristics of C. huangshanensis and other 13 Corydalis species, aiming to elucidate the potential medicinal value of this new species. Materials and Methods The chemical constituents of C. huangshanensis and other 13 medicinal plants of genus Corydalis were analyzed using ultra‐high‐performance liquid chromatography Q‐Exactive Plus hybrid quadrupole‐Orbitrap mass spectrometer (Q‐Orbitrap) mass technology. The differences in the alkaloids in the 14 species were distinguished by chemometrics. Results The mass spectrometry fragmentation information and relative content of 72 alkaloids were obtained. Orthogonal partial least squares discriminant analysis (OPLS‐DA) and cluster heat mapping analysis showed that these 14 species were divided into two groups. The clustering relationship between C. huangshanensis and C. decumbens (Thunb.) Pers. was similar, exhibiting similar chemical compositions and characteristics. These results indicate the potential pharmacological effects of C. huangshanensis. Conclusion This study enhances our understanding of the chemical classification of Corydalis and provides a basis for speculations on the medicinal value of C. huangshanensis.

Abstract:Liver fibrosis is a pathological result of liver injury that usually leads to a pathophysiological wound healing response. The total alkaloids of Corydalis saxicola Bunting (TACS) have been used for hepatoprotective effects on the liver. However, its exact therapeutic mechanisms of liver fibrosis are not yet well understood. To explore the potential anti-fibrosis mechanism of TACS, metabolomics coupled with network pharmacology were applied to reveal the underlying mechanisms. Ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS) combined with multivariate statistical analyses were performed to estimate changes in metabolic profiles. As a result, a total of 23 metabolites in rats with liver fibrosis were altered; of these, 11 had been downregulated and 12 had been upregulated compared with the control group. After TACS treatment, the levels of 13 metabolites were significantly restored compared with the CCl4-treated group, of which 4 metabolites were up-regulated and 9 metabolites were down-regulated. Many of these metabolites are involved in the bile acid metabolism, glutathione metabolism, tryptophan metabolism and purine metabolism. Then, three key targets, including cytochrome P450 family1 subfamily A member 1 (CYP1A1), ornithine decarboxylase 1 (OCD1) and monoamine oxidase Type B (MAOB) were predicted as potential therapeutic targets of TACS against liver fibrosis through network pharmacology analysis. Finally, palmatine, tetrahydropalmatine and dehydrocavidine were screened as potential active compounds responsible for the anti-fibrosis effect of TACS by molecular docking analysis. This study reveals that TACS exerted anti-fibrosis effects by regulating the liver metabolic pathway with multiple components and multiple targets, which is helpful to further clarify the hepatoprotective mechanisms of natural plant extracts.
