Abstract:Several chitosan sodium tripolyphosphate (TPP) nanoparticles embedded with Torreya grandis aril essential oils (TEOs) were synthesized using an emulsion-ionic gelation technique. Mannosylerythritol lipid A (MEL-A), a type of biosurfactant, was selected as the emulsifier. In order to replace acetic acid, an ionic liquid (IL) was employed to dissolve chitosan. The physical properties, diameters, morphology, embedding rate, and antibacterial effects of those essential oil loaded chitosan (CS) nanoparticles were characterized. The results demonstrated that chitosan nanoparticles can be successfully prepared in an ionic liquid containing system and the diameters for nanoparticles in acetic acid and ionic liquid solutions are 144.1 ± 1.457 and 219.0 ± 4.045 nm. After loading with essential oils, the size increased to 349.6 ± 10.55 and 542.9 ± 16.74 nm, respectively. Antibacterial properties were investigated by the observation of the inhibition zone against S. aureus. The results revealed that TEO loaded nanoparticles synthesized in acid and IL aqueous systems have stronger antibacterial activities than CS nanoparticles.

Abstract:The present experiments were undertaken to investigate the mechanism of spasmolytic action of 4-methylumbelliferone (4-MU). It has been reported that the spasmolytic actions of trihydroxybenzene derivatives and trihydroxypropiophenone (THPP) in the biliary system are partially due to the inhibition of catechol-O-methyltrasferase (COMT) activity. On the other hand, resorcinol, a dihydroxybenzene isomer is used as a synthetic material for 4-MU. Utilizing epinephrine as a substrate and S-adenosylmethionine as a methyl donor, the effects of pyrogallol and 4-MU on rat liver COMT activity were determined. Pyrogallol showed inhibition of enzyme activity depending on the concentration added, however, 4-MU was ineffective at any concentration. Contractile responses of the isolated rabbit aorta to transmural electrical stimulation and exogenously applied epinephrine were potentiated by pretreatment with pyrogallol, but were rather inhibited at high concentrations of 4-MU. The phasic and tonic contractions in the isolated rabbit sphincter of Oddi induced by KCl in normal nutrient solution were little affected by either THPP or 4-MU in low concentrations, but were inhibited, in high concentrations. On the other hand, these contractions were inhibited by papaverine in any concentration, but not so by atropine. The contractile responses of the isolated rabbit sphincter of Oddi to CaCl2 and BaCl2 in Ca++ free medium were significantly inhibited by THPP, papaverine and 4-MU, but not so by atropine. The transmural electrical stimulation applied to the sphincter of Oddid caused contractions of the two components, the initial one during stimulation and the other following stimulation. These two components were inhibited by papaverine and 4-MU, depending on the concentration added, but were only slightly inhibited by THPP in high concentrations. It was observed, however, that atropine usually converted the initial component to a relaxation. The late component was unaffected by atropine. The mechanism of spasmolytic action of 4-MU has been discussed herein.
Physicochemical properties and antibacterial activity of polylactic acid/starch acetate films incorporated with chitosan and tea polyphenols

Abstract:Polylactic acid (PLA)/starch acetate (SA) base film was prepared by scraping film method with PLA as the base film material, modified SA as the filling phase and polyethylene glycol (PEG) as plasticizer. Chitosan (CS)/tea polyphenol (TP) antibacterial agent was coated on the surface of PLA/SA base film by coating method. The morphology and structure of the composite film were characterized by FTIR, XRD, SEM and thermogravimetric analysis, and the mechanical, optical and antibacterial properties were tested and analyzed. The results showed that CS/TP compound antibacterial agent was well combined with PLA/SA base film. Compared with the PLA/SA base film, when the content of TP (based on the mass of PLA/SA base film, the same below) was 0.4%, the tensile strength of the composite film increased by 29.33%, had good mechanical strength (24.96 ± 0.65 MPa), and the elongation at break increased by 29.34%; There was no obvious change in transparency, the light transmittance decreased by 0.86%; At the same time, the antibacterial composite film had good antibacterial effect on Escherichia coli , Staphylococcus aureus and Bacillus subtilis . PLA/SA/CS/TP antibacterial composite film could be used in food packaging, especially those designed to protect food from light and microbial decay.

Abstract:The overuse of antibiotics has resulted in a surge of drug-resistant bacteria, making the pursuit of natural antimicrobials an urgent and significant trend. Encapsulation and nanoparticulation are effective ways to enhance the antibacterial properties of natural drugs. In this study, we encapsulated tannic acid (TA) with chitosan (CS) and poly (lactide-co-glycolide) (PLGA) using the emulsion-solvent evaporation method to enhance the antimicrobial effect of TA. We prepared a bilayer membrane spherical nanoemulsion of TA-PLGA-CS (TPC) with uniform size of 559.87 ± 1.16 nm, and zeta potential of 59.53 ± 1.07 mV. TPC could be stably stored for 90 days at 4°C without affecting the properties of the emulsion, and the minimum bactericidal concentration against four strains of Escherichia coli (E. coli) remained unchanged for 60 d. The results indicated that TPC enhanced the inhibitory effect of TA against E. coli. Scanning electron microscope images revealed that TPC treatment caused damage to the bacterial cell membrane. In addition, in vivo experiments indicated that TPC exhibited a superior therapeutic effect on artificial colibacillosis in chickens infested with Avian pathogenic Escherichia coli, as evidenced by the changes in body weight and a reduction bacterial load in heart. Furthermore, TPC reversed the down-regulation of catalase, glutathione peroxidase1 (GPX1), and GPX7 gene expression levels in intestinal tissues. Compared to the model group, TPC treatment elevated serum glutathione peroxidase activities and lowered myeloperoxidase and lactate dehydrogenase levels, offering antioxidant protection that was slightly better than that of doxycycline hydrochlorid group. In summary, we prepared a novel TA antimicrobial preparation with significant antioxidant potential and inhibitory effect against E. coli both in vitro and in vivo.

Abstract:Abstract Tea tree oil (TTO) is widely known essential oil extracted from Melaleuca alternifolia leaves naturally having antimicrobial and antibacterial activities. Due to its highly volatile nature it rapidly evaporates causing loss of efficiency and shorten the effects. Microencapsulation technique was incorporated to ensure the core material is being protected from the immediate contact with the environment and offers controlled release. In this study, microencapsulation of Tea Tree Oil was done by employing complex coacervation technique using Chitosan - Gum acacia system as the coating material and utilized tannic acid as the crosslinking agent. All the materials used in this process are from natural sources which are safe for the human and the environment. In designing the operating process condition for TTO encapsulation, we found that wall ratio of 2:5 and 3.6 pH gave the best yield along with better efficiency. The proposed method studied the surface morphology of the microcapsules with an efficiency and yield of 84.50% and 69.9 % respectively.

Abstract:In order to overcome the problem of fast release of antifouling agent capsaicin, enzymatic and pH double response poly(lactic-co-glycolic acid) @ chitosan @ capsaicin (@PLGA) dual-core shell microcapsules were prepared by composite emulsion method to achieve controlled release and the long-lasting antibacterial. In the first stage, due to the excellent biodegradability PLGA shell quickly decomposes and ruptures, releasing the internal CAP and to achieve the intelligent controlled release of enzymatic response. Rapid response within 12 h and achieves over 90% bacterial inhibition. The antibacterial rate at pH 4 is about five times more than that at pH 8. With the decrease of pH value caused by bacterial reproduction, the protonation effect of CS is triggered to release internal CAP, realizing controlled release sterilization of pH response in the second stage, the bacterial inhibition rate remains stable at over 90%. On the 7th day of the long-lasting inhibition experiment, a small increase in the number of colonies could be observed, but it was soon inhibited again to low-level state. This is the feature of the dual enzymatic and pH response. The bacterial inhibition rate was stable at about 90% for 24 days, which proves that compared with the single response nanocapsules , the prepared microcapsules have more stable and durable phased release antibacterial property.

Abstract:Tea tree oil (TTO) is a broad-spectrum natural antimicrobial agent. Its application is limited due to its volatile property. To reduce its volatility and improve its sustained-release performance, a synergistic sustained-release system was prepared. Montmorillonite (Mt) was ion-exchanged with sodium ions (Na+) and zinc ions (Zn2+), then organically modified with cetyl trimethyl ammonium bromide (CTAB) and chitosan (CS) to improve its sustained release performance. The results indicated that Mt was exfoliated, but still maintained its ordered layer structure. The surface of CS-Zn-OMt was rough, because some pores and surfaces were filled. The addition of Zn2+ increased the loading of CS (37.8 − 40.8%) and induced more uniform distribution of CS on Mt. Also, CS-Zn-OMt achieved a high loading capacity for TTO and the loading value was 222.0 mg*g−1. Furthermore, the release studies showed that CS-Zn-OMt exhibited sustained-release behavior due to the release from the outer layer of CS and the Mt channel simultaneously. The results of antibacterial test showed that the addition of CS inhibited the drift of Mt and showed good antibacterial performance because of strong effective drug-carrier interaction. The developed system can be potentially applied for the delivery of volatile essential oils for antibacterial infection.Graphical

Abstract:Antimicrobial food packaging with a sustained release of antimicrobial agents plays a key role in maintaining food quality and food safety. In this study, polyelectrolyte gellan gum (GG)-chitosan (CS) multilayer film was fabricated by layer-by-layer assembly technology with the incorporation of thyme essential oil (TEO) coarse emulsion (TEOC) or nanoemulsion (TEON). The microstructure of GG-CS three-layered film was observed by scanning electron microscopy (SEM). The red fluorescent signals in confocal laser scanning microscopy (CLSM) images indicated smaller droplets and more uniform distribution of TEON prepared by ultrasonic treatments, which was consistent with the particle size results and porous structure in SEM images. The film incorporated with 6% TEON exhibited higher antimicrobial activity than that with 6% TEOC, and it significantly reduced E. coli populations in liquid model system. Moreover, films incorporated with TEON showed improved mechanical flexibility (elongation at break) and UV blocking property in comparison to TEOC. This developed GGCS three-layered film loading with TEON was expected to be a potential antimicrobial material with sustained release property for food packaging applications.
Impact of tea tree essential oil and citric acid/choline chloride on physical, structural and antibacterial properties of chitosan-based films

Abstract:The purpose of this study was to develop a novel antimicrobial packaging film by using chitosan (CS), deep eutectic solvent (DES, a plasticizer) and tea tree essential oil (TTO, a natural antibacterial agent). The physical properties of the films (i.e., water content, solubility, water vapor permeability, color, thickness, water contact angle, and mechanical properties) demonstrated that TTO reduced the hydrophilicity of the films. At the same time, DES significantly improved the flexibility of the films. Micro-structural characterization showed that DES and TTO interacted with the CS matrix and were uniformly dispersed in films. The interactions among these main components in the films were verified by Fourier transform infrared spectroscopy and X-ray diffraction analysis. Thermogravimetric analysis further confirmed that these films had good thermal stability. In addition, the antibacterial assays verified that the CS-DES-TTO films could inhibit the activity of planktonic Sh. flexneri . The films prepared in this study showed the characteristics required of active packaging materials.

Abstract:The use of nanotechnology has contributed to conquer the disadvantages of oil-loaded food packaging by altering their properties. In this study, chitosan (CS) composite films were fabricated using a novel nano-encapsulation system: ethanol extract of propolis (EEP)-phosphatidylcholine (PC)-tea seed oil (TSO) nano-microcapsules. X-ray diffraction results indicated that the nano-microcapsules were biocompatible with CS. Uniformly dispersed EEP-PC-TSO nano-microcapsules in CS improved the UV-blocking ability, thermal stability, structural densification, yellowing, and water vapor permeability of the composite films, the release of TSO occurs gradually and under controlled conditions. Compared with a simple mixture film of CS/PC/TSO/EEP, the CS film based on nano-microcapsules containing EEP-PC-TSO was more effective at reducing total bacterial count, thiobarbituric acid reactants (TBARS), and total volatile basic nitrogen (TVB-N) values, extending the shelf life of pork tenderloin. In summary, this CS composite film based on EEP-PC-TSO nano-microcapsules could be applied as a potential preservative film for food packaging.

Abstract:Many active ingredients in food products are destroyed by gastric acids and enzymes as they pass through the stomach, thereby reducing the desired effect of the target ingredient. In addition, traditional microcapsule preparation methods usually break up the dispersion by mechanical stirring or oscillation, which generates droplets of uneven size and inconsistent inclusion content, and mechanical external forces may cause denaturation and inactivation of active substances. This study aimed to use microfluidics to generate Oil-in-water-in-oil (O/W/O) droplet structures with good monodispersity and highly tunable properties, and to further prepare microcapsules with high encapsulation rate and targeted intestinal release by taking advantage of the cross-linking properties of alginate that reacts with Ca 2+ under acidic conditions. Finally, the particle size distribution, release effect, encapsulation effect and oxidative stability of the core-shell microcapsules were evaluated. In this study, two types of microcapsules, homogeneous and core-shell microcapsules, were prepared by selecting tea polyphenols and vitamin E as models for hydrophilic and lipophilic bioactive compounds, respectively. Thin shell microcapsules with a shell thickness of about 25 μm were prepared using PDMS chip conditioning. Large microcapsules with a particle size of 230 μm were generated by using capillary glass tube chips, which had an excellent antioxidant property with a retention rate of 88.66 ± 1.25% of VE after 20 days of storage at 4 °C, while all of the VE was oxidized in the bare environment. In addition, in simulated gastric fluid, the release of VE was only about 20%, while in artificial intestinal fluid, the release was close to 100%, so the microcapsules also had intestinal targeting properties.

Abstract:The product of ozonolysis, glycero-(9,10-trioxolane)-trioleate (ozonide of oleic acid triglyceride, [OTOA]), was incorporated into polylactic acid/polycaprolactone (PLA/PCL) blend films in the amount of 1, 5, 10, 20, 30 and 40% w/w. The morphological, mechanical, thermal and antibacterial properties of the biodegradable PLA/PCL films after the OTOA addition were studied. According to DSC and XRD data, the degree of crystallinity of the PLA/PCL + OTOA films showed a general decreasing trend with an increase in OTOA content. Thus, a significant decrease from 34.0% for the reference PLA/PCL film to 15.7% for the PLA/PCL + 40% OTOA film was established using DSC. Observed results could be explained by the plasticizing effect of OTOA. On the other hand, the PLA/PCL film with 20% OTOA does not follow this trend, showing an increase in crystallinity both via DSC (20.3%) and XRD (34.6%). OTOA molecules, acting as a plasticizer, reduce the entropic barrier for nuclei formation, leading to large number of PLA spherulites in the plasticized PLA/PCL matrix. In addition, OTOA molecules could decrease the local melt viscosity at the vicinity of the growing lamellae, leading to faster crystal growth. Morphological analysis showed that the structure of the films with an OTOA concentration above 20% drastically changed. Specifically, an interface between the PLA/PCL matrix and OTOA was formed, thereby forming a capsule with the embedded antibacterial agent. The moisture permeability of the resulting PLA/PCL + OTOA films decreased due to the formation of uniformly distributed hydrophobic amorphous zones that prevented water penetration. This architecture affects the tensile characteristics of the films: strength decreases to 5.6 MPa, elastic modulus E by 40%. The behavior of film elasticity is associated with the redistribution of amorphous regions in the matrix. Additionally, PLA/PCL + OTOA films with 20, 30 and 40% of OTOA showed good antibacterial properties on Pseudomonas aeruginosa, Raoultella terrigena (Klebsiella terrigena) and Agrobacterium tumefaciens, making the developed films potentially promising materials for wound-dressing applications.
Characteristics, Controlled-release and Antimicrobial Properties of Tea Tree Oil Liposomes-incorporated Chitosan-based Electrospun Nanofiber Mats

Abstract:In this paper, a notable chitosan/poly(ethylene oxide) nanofiber mats containing tea tree oil liposomes (TOL-CENs) were successfully fabricated using electrospinning process. The microstructures and morphology were characterized by scanning electron microscopy. The porosity, fluid absorbability, water vapor permeability and mechanical properties of nanofiber mats were also estimated by ethanol density method, gravimetric method, dish method and tensile test, respectively. Compared to the chitosan/poly(ethylene oxide) composite freeze-dried sponges containing tea tree oil liposomes, TOL-CENs had greater porosity, water absorption, breathability and better mechanical properties. In addition, the controlled-release properties and long-term bactericidal capability of the material were also assessed. From the analysis of the release kinetics and mechanism, it was found that the significant decreased terpinen-4-ol concentration gradient from liposomal surface to the outside of material was the key to the sustained terpinen-4-ol release in virtue of liposomal encapsulation. TOL-CENs exhibited long-term and more excellent microbicidal effects against Staphylococcus aureus, Escherichia coli and Candida albicans than chitosan/poly(ethylene oxide) nanofiber mats. The combination of tea tree oil liposomes and chitosan in nanofiber mats synergistically destroyed cell membrane, prevented cell adhesion and caused the irregular aggregation of cytoplasm, resulting in cell disintegration observed by transmission electron microscope. In summary, TOL-CENs had potential application value as a long-term antimicrobial nonwoven materials.

Abstract:The exploration of safe antibiotic substitutes is one of the research hotspots in animal husbandry. Adding suitable plant essential oils into feed could improve the growth performance and immune capacity of animals. In order to make plant essential oil play a better role in feed application, sodium alginate and chitosan were used as the wall materials, and blended plant essential oils (BEO) as the core material to prepare BEO microcapsules by the sharp-hole condensation method. On the basis of single-factor experiments, the optimal preparation conditions for BEO microcapsules were obtained by response surface experiments. The physicochemical properties were characterized and analyzed by Fourier-transform infrared spectroscopy (FTIR) and field scanning electron microscope (FSEM). Meanwhile, the release mechanism was studied by simulating a gastrointestinal sustained-release experiment. The results showed that under the optimal preparation conditions, the encapsulation efficiency of BEO microcapsules could reach 80.33 ± 2.35%. FTIR and SEM analysis displayed that the microcapsules obtained had uniform color and size and a complete and compact structure. In vitro study indicated that the release amount of BEO microcapsules in the simulated intestinal fluid is higher than that in the simulated intestinal fluid, which was consistent with animal digestive and absorptive characteristics.

Abstract:A functional textile immobilized by microcapsules of the lime peel essential oils of C. aurantifolia (LPEO) was prepared and characterized. A varied amount of Chitosan/Alginate (CH/AG) ratios, followed by a mass of LPEO and concentration of sodium tripolyphosphate (STPP) crosslinker, was optimized sequentially to coacervate LPEO using a Tween 80 emulsifier. An antibacterial assay against both Gram-positive and Gram-negative bacteria was further evaluated for the embedded microcapsules. The LPEO (0.2 g) was effectively coacervated by CH/AG (5:3) crosslinked by 2% of STTP to give a yield, oil content (OC), and encapsulation efficiency (EE) of 53.45 ± 2.16%, 65.08 ± 2.60% and 85.04 ± 0.70% respectively. A rough spherical shape of LPEO microcapsules was homogeneously observed with an average particle size of 0.757 mm. An Avrami's kinetic model revealed the release mechanism of the core following zero-order kinetics ( k = 1.11 ± 0.13 × 10 −9 s −1 , Ea = 70.21 kJ/mol). The LPEO microcapsules demonstrated good thermal stability up to 122 °C and maintained 38% OC at ambient temperature for four weeks. A 70.34 ± 4.16% of the LPEO microcapsules were successfully overlaid onto the gauze with citric acid binder and sodium phosphate catalyst. Overall, the immobilized microcapsules exhibited strong inhibition against S. aureus and moderate against S. epidermidis, E. coli, and K. pneumonia .

Abstract:Aquatic products have an important role in global agriculture, but the challenges associated with preservation have limited their marketability. Essential oil (EO), such as sweet orange oil (SOEO), has been widely used for preservation due to its excellent antibacterial ability. However, the volatilization of EO limits its application in food preservation. In this study, SOEO was extracted from sweet orange peel by steam distillation and then stored in microcapsules. The components of the microcapsules were as follows: the porous starch was chosen as an adsorbed substrate to store SOEO (PS/SOEO), and sodium alginate (SA) and chitosan (CMCS) were used as shell material to delay the volatilization of SOEO using the sharp pore coagulation method. Our results showed that the main antibacterial ingredients in SOEO were aldehydes (33.93%) and d-limonene (15.38%). The microcapsules were of an irregular shape (oval), and the size of the microcapsules was 1.2 ± 0.1 cm as measured by a digital micrometer. Scanning electron microscopy (SEM) results showed that there were a lot of pores on the surface of the starch after modification, but sodium alginate and chitosan could well encapsulate these pores. The results of Fourier transform infrared (FTIR) spectroscopy and X-ray diffraction (XRD) analysis also showed that SOEO was successful encapsulated into the porous starch. The results of compression test and releasing kinetics studies suggested that CMCS and SA improved the mechanical and slow-releasing ability of SOEO microcapsules. The best antibacterial performance was obtained when 0.8 g of SOEO microcapsules was added. Finally, the shelf life of crawfish could be extended to 6 days by SOEO microcapsule (1/10 g, SOEO microcapsule/crawfish) under room temperature. These results provide a systematic understanding of the antibacterial capabilities of sweet orange essential oil microcapsules, which can contribute to the development of preservation methods for aquatic products.

Abstract:Journal of Bioactive and Compatible Polymers, Ahead of Print. As known, the chitosan is a biodegradable biopolymer with antibacterial properties, therefore it is used in the fields of pharmacy, medical, wastewater treatment, biotechnology, cosmetics, textiles, and agriculture. Apart from these, the chitosan has an important place in the food industry with its widespread use. In this research article, the chitosan were encapsulated with the taurine and garlic extracts by the spray dryer. The CSA and CSB compounds synthesized as final products were analyzed by Fourier transformed infrared spectroscopy (FTIR) and High Performance Liquid Chromatography (HPLC). The effect of the encapsulation process on the molecular weight of the polymer structure was investigated using the cryoscopy method. The compound CSA represents 1/2 encapsulation of chitosan with taurine and increased garlic extracts, respectively, while CSB represents 1/1 encapsulation of chitosan with increased taurine and fixed garlic extracts. The % antioxidant activity of the final products was determined by DDPH method. The inhibition zone and surface activity proporties of the CSA and CSB were carried out against Listeria monocytogenes, Staphylococcus aureus, E. coli, and Salmonella bacteria. The results obtained as a result of the analyzes were evaluated, and optimum values were determined for use in food packaging.

Abstract:To obtain long‐lasting preservation materials, the tea polyphenol liposomes (TP‐Lips)/lysozyme (LZM)–chitosan (CS) composite coating with the gradual sustained property was prepared by tape casting method. These coatings were characterised, and their physicochemical properties were measured. Meanwhile, the antibacterial mechanisms of coatings were studied using the spoilage bacteria of aquatic products (Shewanella putrefaciens and Pseudomonas fluorescens) as target strains. Compared with CS coating, the incorporation of TP‐Lips makes the cracks and granular matter of the coatings increase. Except for the oxygen permeabilities (OP) and carbon dioxide permeabilities (CDP), the rest of the physicochemical properties are decreased, including tensile strength (Ts), elongation at break (EB) and light transmittance (T). Antibacterial mechanisms indicate that TP and LZM have a synergistic antibacterial effect. The slow‐release system composed of liposomes and coating prolongs the action time of TP and LZM. Hence, the TP‐Lips/LZM‐CS coating could be a hopeful material in food preservation field with excellent antibacterial properties. The coexisting system of tea polyphenol liposomes (TP‐Lips) and lysozyme (LZM) is constructed in the chitosan coating, and the gradual release of TP from liposomes to the external environment. And it is clarified that TP and LZM have synergistic antibacterial effect.
