Beam Energy Considerations for Gold Nano-Particle Enhanced Radiation Treatment

Abstract:Purpose: A novel approach using nano technology enhanced radiation modalities is investigated. The proposed methodology uses antibodies labeled with organically inert metals with a high atomic number. Irradiation using photons with energies in the kilo--electron volt (keV) range show an increase in dose due to a combination of an increase in photo-electric interactions and a pronounced generation of Auger and/or Coster-Kronig (A-CK) electrons. Methods: The dependency of the dose deposition on various factors is investigated using Monte Carlo simulation models. The factors investigated include: agent concentration, spectral dependency looking at mono--energetic sources as well as classical bremsstrahlung sources. The optimization of the energy spectrum is performed in terms of physical dose enhancement as well as the dose deposited by Auger and/or Coster-Kronig electrons and their biological effectiveness. Results: A quasi-linear dependency on concentration and an exponential decrease within the target medium is observed. The maximal dose enhancement is dependent on the position of the target in the beam. Apart from irradiation with low photon energies (10 - 20 keV) there is no added benefit from the increase in generation of Auger electrons. Interestingly, a regular 110kVp bremsstrahlung spectrum shows a comparable enhancement in comparison with the optimized mono--energetic sources. Conclusions: In conclusion we find that the use of nano-particle enhanced shows promise to be implemented quite easily in regular clinic on a physical level due to the advantageous properties in classical beams.

Study of the 199Au nanoparticles production parameters via irradiation of platinum target by using thermal neutrons

Abstract:In this study, the production parameters of 199Au nanoparticles (199AuNPs) have been investigated by a two-part study. The first part is about investigating the indirect method of producing non-carrier-added (NCA) 199Au radionuclide. MCNPX-2.6, TALYS-1.9, and ALICE/ASH-0.1 codes were applied as the theoretical approach to simulate the core of Tehran research reactor (TRR) for determining the activity of 199Au, specifying the production yield of 199Au, and calculating the excitation function of P198t(n,γ)P199t→A199u reaction. As the corresponding experimental approach, two 11 mg and 15.5 mg samples of enriched 198Pt metal powder were irradiated by thermal neutrons for 21 h and 10 min. The liquid-liquid extraction (LLX) technique has been used with a different solvent for each sample. LLX using ethyl acetate and LLX using Di-(2-Ethylhexyl) phosphoric acid (HDEHP) were applied for the 15.5 mg and the 11 mg samples respectively. The chemical yield of 199Au was calculated more than %99 for the 15.5 mg sample, and more than %80 for the 11 mg sample. The second part is about synthesizing 199AuNPs in an average size of 50 nm by using Turkevich method.
Photo-nuclear cross sections on $^{197}$Au, an update on the gold standard

Abstract:A method was developed for measuring photonuclear reactions concurrently at several discrete photon beam energies on a stack of different target materials via a single irradiation. Concentric ring targets of the materials (in order from front to back targets: Au, TiO$_2$, Zn, Os, and Au) were irradiated at the High Intensity Gamma-ray Source (HI$\gamma$S). As a proof of principle, we report the result of the cross section measurements from the front Au target. The excitation functions of the $^{197}$Au($\gamma$,n)$^{196}$Au and $^{197}$Au($\gamma$,3n)$^{194}$Au reactions were determined in the incident photon energy range of 13-31 MeV using quasi-monoenergetic photon beams provided at HI$\gamma$S. The cross sections of the combined ground state (2$^{-}$) and short-lived first isomeric state (m1, 5$^{+}$), and of the second isomeric state (m2, 12$^{-}$) in the $^{196}$Au production are obtained separately by subtracting the $\gamma$ rays from the internal conversion of the second isomeric state. The excitation function of the second isomeric state via the photon-induced reaction $^{197}$Au($\gamma$,n)$^{196m2}$Au was measured for the first time. By using the activation method rather than direct neutron counting, the exclusive cross sections for the ($\gamma$,n) and ($\gamma$,3n) reactions were determined. Comparing the yields from the front and back gold targets validates our ability to simulate the effect of photon scattering in the target stack and provides a method for assessing the systematic uncertainty of our technique.

Preparation of Orange-Red Fluorescent Gold Nanoclusters Using Denatured Casein As a Reductant and Stabilizing Agent, and Their Application to Imaging of HeLa Cells and for the Quantitation of Mercury(ii)

Abstract:We describe a method for the preparation of water-soluble gold nanoclusters (Au-NCs) from chloroauric acid using denatured-casein as both a reducing and stabilizing agent. The resulting Au-NCs were characterized by photoluminescence, UV–vis absorption, and X-ray photoelectron spectroscopies, and by transmission electron microscopy. The Au-NCs have an average diameter of 1.7 ± 0.2 nm and exhibit orange-red fluorescence emission peaking at 600 nm (with a Stokes’ shift as large as 237 nm), a quantum yield of 4.3 %, and good stability over the physiologically relevant range of pH values and ionic strength. Cytotoxicity studies showed the Au-NCs to display negligible effects in terms of altering cell proliferation or triggering apoptosis. Fluorescence imaging of HeLa cancer cells was accomplished by loading such cells with the Au-NCs. The fluorescence of the Au-NCs is found to be strongly quenched by Hg(II) ions, and thus the Au-NCs can be used for detecting and, possibly, imaging of Hg(II). An assay was worked out for the determination of Hg(II), and its limit of detection is 1.83 nM, which is 5.5 times lower than the maximum allowed concentration of Hg(II) in drinking water as defined by the US EPA.
Production of Medical Radioisotopes with High Specific Activity in Photonuclear Reactions with $γ$ Beams of High Intensity and Large Brilliance

Abstract:We study the production of radioisotopes for nuclear medicine in $(\gamma,x{\rm n}+y{\rm p})$ photonuclear reactions or ($\gamma,\gamma'$) photoexcitation reactions with high flux [($10^{13}-10^{15}$)$\gamma$/s], small diameter $\sim (100 \, \mu$m$)^2$ and small band width ($\Delta E/E \approx 10^{-3}-10^{-4}$) $\gamma$ beams produced by Compton back-scattering of laser light from relativistic brilliant electron beams. We compare them to (ion,$x$n$ + y$p) reactions with (ion=p,d,$\alpha$) from particle accelerators like cyclotrons and (n,$\gamma$) or (n,f) reactions from nuclear reactors. For photonuclear reactions with a narrow $\gamma$ beam the energy deposition in the target can be managed by using a stack of thin target foils or wires, hence avoiding direct stopping of the Compton and pair electrons (positrons). $(\gamma,\gamma')$ isomer production via specially selected $\gamma$ cascades allows to produce high specific activity in multiple excitations, where no back-pumping of the isomer to the ground state occurs. We discuss in detail many specific radioisotopes for diagnostics and therapy applications. Photonuclear reactions with $\gamma$ beams allow to produce certain radioisotopes, e.g. $^{47}$Sc, $^{44}$Ti, $^{67}$Cu, $^{103}$Pd, $^{117m}$Sn, $^{169}$Er, $^{195m}$Pt or $^{225}$Ac, with higher specific activity and/or more economically than with classical methods. This will open the way for completely new clinical applications of radioisotopes. For example $^{195m}$Pt could be used to verify the patient's response to chemotherapy with platinum compounds before a complete treatment is performed. Also innovative isotopes like $^{47}$Sc, $^{67}$Cu and $^{225}$Ac could be produced for the first time in sufficient quantities for large-scale application in targeted radionuclide therapy.

Abstract:The solvent extraction of gold from alkaline cyanide solution was studied by using 198Au as a radiotracer. The influence of several variables on the gold extraction, including the concentration of gold, the molar ratio of extractants to gold(I), the volume percentage of cosolvent and the pH value of the aqueous phase, was investigated. The results indicated that the radioactive tracer technique is a quick, accurate, and convenient tool to investigate the extraction behavior of an element existing in low concentrations. The experimental results indicated that the studied amines, tetradecyldimethylbenzylammonium chloride, N1923 and tri-n-octylamine, could be used as extractants for the recovery of gold from aqueous alkaline cyanide solutions.

Abstract:Radioisotopes of arsenic are of considerable interest to the field of nuclear medicine with unique nuclear and chemical properties making them well-suited for use in novel theranostic radiopharmaceuticals. However, progress must still be made in the production of isotopically pure radioarsenic and in its stable conjugation to biological targeting vectors. This work presents the production and irradiation of isotopically enriched (72)Ge(m) discs in an irrigation-cooled target system allowing for the production of isotopically pure (72)As with capability on the order of 10 GBq. A radiochemical separation procedure isolated the reactive trivalent radioarsenic in a small volume buffered aqueous solution, while reclaiming (72)Ge target material. The direct thiol-labeling of a monoclonal antibody resulted in a conjugate exhibiting exceptionally poor in vivo stability in a mouse model. This prompted further investigations to alternative radioarsenic labeling strategies, including the labeling of the dithiol-containing chelator dihydrolipoic acid, and thiol-modified mesoporous silica nanoparticles (MSN-SH). Radioarsenic-labeled MSN-SH showed exceptional in vivo stability toward dearsenylation.
Production assessment of non-carrier-added 199Au by (n,γ) reaction

Abstract:Gold-199 is a promising theranostic radionuclide for targeted radioimmunotherapy as well as for scintigraphy and dosimetry. 199Au can be produced in two methods in the direct and indirect routes of the reactor production via 197Au(n,γ)198Au(n,γ)199Au as the direct or 198Pt(n,γ)199Pt→199Au as the indirect method. This investigation described the development of a method for the reactor production of no-carrier-added (NCA) 199Au through neutron activation of natural Pt in Tehran Research Reactor (TRR) at a thermal neutron flux of 3.5 × 1013 n cm-2 s-1. Also, in this paper, the activity of 199Au has been estimated using the MCNPX code. In this case, first, the reactor core is simulated. Then the calculated results are compared with the corresponding experimental values. Moreover, two different chemical separation methods are investigated experimentally in details.

Abstract:Targeted alpha therapy (TAT) is a methodology that is being developed as a promising cancer treatment using the alpha-particle decay of radionuclides. This technique involves the use of heavy radioactive elements being placed near the cancer target area to create maximum damage to the cancer cells while minimizing the damage to healthy cells. Using gold nanoparticles (AuNPs) as carriers, a more efficient therapy methodology may be realized. AuNPs can be good candidates for transporting these radionuclides to the vicinity of the cancer cells since they are able to be labeled not just with the radionuclides, but also a host of other proteins and ligands to target these cells and serve as addi-onal treatment options. Research has shown that astatine and iodine are capable of adsorbing on the surface of gold, creating a covalent bond that is quite stable to be used in experiments.

Abstract:Introduction: Astatine-211 has attained significant interest in the recent times as a promising radioisotope for targeted alpha therapy (TAT) of cancer. In this study, we report the production of 211At via 209Bi (α, 2n) 211At reaction in a cyclotron and development of a facile radiochemical separation procedure to isolate 211At for formulation of nanoradiopharmaceuticals. Methods: Natural bismuth oxide target in pelletized form wrapped in Al foil was irradiated with 30 MeV α-beam in an AVF cyclotron. The irradiated target was dissolved in 2 M HNO3 followed by selective precipitation of Bi as Bi(OH)3 under alkaline condition. The radiochemically separated 211At was used for labeling cyclic RGD peptide conjugated gold nanoparticles (Au-RGD NPs) by surface adsorption. The radiochemical stability of 211At-Au-RGD NPs was evaluated in phosphate buffered saline (PBS) and human serum media. Results: The batch yield of 211At at the end of irradiation was ∼6 MBq.μA-1.h-1. After radiochemical separation, ∼80 % of 211At could be retrieved with >99.9 % radionuclidic purity. Au-RGD NPs (particle size 8.4±0.8 nm) could be labeled with 211At with >99 % radiolabeling yield. The radiolabeled nanoparticles retained their integrity in PBS and human serum media over a period of 21 h. Conclusions: The present strategy simplifies 211At production in terms of purification and would increase affordable access to this radioisotope for TAT of cancer.

Abstract:Boron neutron capture therapy (BNCT) is an anticancer modality realized through 10B accumulation in tumor cells, neutron irradiation of the tumor, and decay of boron atoms with the release of alpha-particles and lithium nuclei that damage tumor cell DNA. As high-LET particle release takes place inside tumor cells absorbed dose calculations are difficult, since no essential extracellular energy is emitted. We placed gold nanoparticles inside tumor cells saturated with boron to more accurately measure the absorbed dose. T98G cells accumulated ~50 nm gold nanoparticles (AuNPs, 50 µg gold/mL) and boron-phenylalanine (BPA, 10, 20, 40 µg boron-10/mL), and were irradiated with a neutron flux of 3 × 108 cm-2s-1. Gamma-rays (411 keV) emitted by AuNPs in the cells were measured by a spectrometer and the absorbed dose was calculated using the formula D = (k × N × n)/m, where D was the absorbed dose (GyE), k-depth-related irradiation coefficient, N-number of activated gold atoms, n-boron concentration (ppm), and m-the mass of gold (g). Cell survival curves were fit to the linear-quadratic (LQ) model. We found no influence from the presence of the AuNPs on BNCT efficiency. Our approach will lead to further development of combined boron and high-Z element-containing compounds, and to further adaptation of isotope scanning for BNCT dosimetry.

Abstract:Near‐infrared II (NIR‐II) imaging at 1100–1700 nm shows great promise for medical diagnosis related to blood vessels because it possesses deep penetration and high resolution in biological tissue. Unfortunately, currently available NIR‐II fluorophores exhibit slow excretion and low brightness, which prevents their potential medical applications. An atomic‐precision gold (Au) cluster with 25 gold atoms and 18 peptide ligands is presented. The Au25 clusters show emission at 1100–1350 nm and the fluorescence quantum yield is significantly increased by metal‐atom doping. Bright gold clusters can penetrate deep tissue and can be applied in in vivo brain vessel imaging and tumor metastasis. Time‐resolved brain blood‐flow imaging shows significant differences between healthy and injured mice with different brain diseases in vivo. High‐resolution imaging of cancer metastasis allows for the identification of the primary tumor, blood vessel, and lymphatic metastasis. In addition, gold clusters with NIR‐II fluorescence are used to monitor high‐resolution imaging of kidney at a depth of 0.61 cm, and the quantitative measurement shows 86% of the gold clusters are cleared from body without any acute or long‐term toxicity at a dose of 100 mg kg−1.

Abstract:Alpha-particle radiotherapy has gained considerable attention owing to its potent anti-cancer effect. 211At, with a relatively short half-life of 7.2 h, emits an alpha particle within a few cell diameters with high kinetic energy, which damages cancer cells with high biological effectiveness. In this study, we investigated the intravenous injection of 211At-labeled gold nanoparticles (AuNPs) for targeted alpha-particle therapy (TAT). Different kinds of surface-modified gold nanoparticles can be labeled with 211At in high radiochemical yield in 5 min, and no purification is necessary. The in vivo biodistribution results showed the accumulation of 5 nm 211At-AuNPs@mPEG at 2.25% injection dose per gram (% ID/g) in tumors within 3 h via the enhanced permeability and retention (EPR) effect. Additionally, we observed a long retention time in tumor tissues within 24 h. This is the first study to demonstrate the anti-tumor efficacy of 5 nm 211At-AuNPs@mPEG that can significantly suppress tumor growth in a pancreatic cancer model via intravenous administration. AuNPs are satisfactory carriers for 211At delivery, due to simple and efficient synthesis processes and high stability. The intravenous administration of 5 nm 211At-AuNPs@mPEG has a significant anti-tumor effect. This study provides a new framework for designing nanoparticles suitable for targeted alpha-particle therapy via intravenous injection.

Abstract:Auger-emitting radionuclides, exemplified by Pd-103, exhibit considerable therapeutic potential in cancer treatment due to their high cytotoxicity and localized biological impact. Despite these advantages, the separation of such radionuclides presents a complicated challenge, requiring intricate and time-intensive "wet chemistry" methods attributed to the exceptional chemical inertness of the associated metals. This study proposes an innovative solution to this separation challenge through the design and implementation of a piece of radionuclide separation equipment (RSE). The equipment employs a dry distillation approach, capitalizing on differences in partial vapor pressures between irradiated and resulting radioactive metals, with a diffusion-driven extraction method applied to separate Pd-103 radionuclides generated via the proton irradiation of Rh-103 at cyclotron. Our optimization endeavors focused on determining the optimal temperature for effective metal separation and adjusting the diffusion, evaporation, and deposition rates, as well as addressing chemical impurities. The calculations indicate 17% ± 2% separation efficiency with our RSE. Approximately 77 ± 2% and 49 ± 2% of the deposited Pd-103 were isolated on substrates of Nb foil and ZnO-covered W disc, respectively. The proposed innovative dry distillation method that has been experimentally tested offers a promising alternative to conventional separation techniques, enabling enhanced purity and cost-efficient cancer treatment strategies.

The approach is based on performing the nuclear reaction in inverse kinematics, namely sending a heavy-ion beam of appropriate energy on a light target (e.g. H, d, He) and collecting the isotope of interest. In this work, as a proof-of-concept, we studied the production of the theranostic radionuclide 67Cu (T 1/2 =62 h) via the reaction of a 70Zn beam at 15 MeV/nucleon with a hydrogen gas target. The 67Cu radionuclide, alongside other coproduced isotopes, was collected after the gas target on an Al catcher foil and their radioactivity was measured by off-line {\gamma}-ray analysis. After 36 h from the end of the irradiation, apart from the product of interest 67Cu, the main radioimpurity coming from the 70Zn+p reaction was 69mZn (T 1/2 =13.8 h) that can be reduced by further radio-cooling. Moreover, along with the radionuclide of interest produced in inverse kinematics, the production of additional radioisotopes is possible by making use of the forward-focused neutrons from the reaction and letting them interact with a secondary target. A preliminary successful test of this concept was realized in the present study. The main requirement to obtain activities appropriate for preclinical studies is the development of high-intensity heavy-ion primary beams.

Abstract:This work describes the production of two clinically relevant metal radioisotopes [Formula: see text] and [Formula: see text] with a medical cyclotron by the irradiation of liquid targets. New results are presented for the implementation of this methodology in a fully automated system, using commercially available equipment. Liquid target solutions containing enriched [Formula: see text] and [Formula: see text] were loaded, bombarded and transferred to synthesis modules where a purified solution containing the desired radiometal is obtained and can then be used to further radiolabeling within only one hour after End-Of-Bombardment (EOB). Typical production runs using enriched material lead to the production of 5 GBq and 6 GBq (0.14 MBq/([Formula: see text]Ah ⋅ mg) and 1.5 MBq/([Formula: see text]Ah ⋅ mg)) of [Formula: see text] and [Formula: see text]; although the technique can be used to obtain up to 25 GBq and 40 GBq, respectively, by simply scaling up the amount of the enriched material. Purified solutions containing [Formula: see text] and [Formula: see text] were obtained within 30 min after EOB and used to produce [Formula: see text]-ATSM and [Formula: see text]–DOTA–NOC, respectively, with quality parameters suitable for human use.

Abstract:Nuclear fission reactions can release massive amounts of energy accompanied by neutrons and γ photons, which create a mixed radiation field and enable a series of reactions in nuclear reactors. This study demonstrates a one-pot/one-step approach to synthesizing radioactive gold nanoparticles (RGNP) without using radioactive precursors and reducing agents. Trivalent gold ions are reduced into gold nanoparticles (8.6–146 nm), and a particular portion of 197Au atoms is simultaneously converted to 198Au atoms, rendering the nanoparticles radioactive. We suggest that harnessing nuclear energy to gold nanoparticles is feasible in the interests of advancing nanotechnology for cancer therapy. A combination of RGNP applied through convection-enhanced delivery (CED) and temozolomide (TMZ) through oral administration demonstrates the synergistic effect in treating glioblastoma-bearing mice. The mean survival for RGNP/TMZ treatment was 68.9 ± 9.7 days compared to that for standalone RGNP (38.4 ± 2.2 days) or TMZ (42.8 ± 2.5 days) therapies. Based on the verification of bioluminescence images, positron emission tomography, and immunohistochemistry inspection, the combination treatment can inhibit the proliferation of glioblastoma, highlighting the niche of concurrent chemoradiotherapy (CCRT) attributed to RGNP and TMZ.

Abstract:Conventional X-ray therapy (XRT) is commonly applied to suppress cancerous tumors; however, it often inflicts collateral damage to nearby healthy tissue. In order to provide a better conformity of the dose distribution in the irradiated tumor, proton therapy (PT) is increasingly being used to treat solid tumors. Furthermore, radiosensitization with gold nanoparticles (GNPs) has been extensively studied to increase the therapeutic ratio. The mechanism of radiosensitization is assumed to be connected to an enhancement of the absorbed dose due to huge photoelectric cross-sections with gold. Nevertheless, numerous theoretical studies, mostly based on Monte Carlo (MC) simulations, did not provide a consistent and thorough picture of dose enhancement and, therefore, the radiosensitization effect. Radiosensitization by nanoparticles in PT is even less studied than in XRT. Therefore, we investigate the physics picture of GNP-enhanced RT using an MC simulation with Geant4 equipped with the most recent physics models, taking into account a wide range of physics processes relevant for realistic PT and XRT. Namely, we measured dose enhancement factors in the vicinity of GNP, with diameters ranging from 10 nm to 80 nm. The dose enhancement in the vicinity of GNP reaches high values for XRT, while it is very modest for PT. The macroscopic dose enhancement factors for realistic therapeutic GNP concentrations are rather low for all RT scenarios; therefore, other physico-chemical and biological mechanisms should be additionally invoked for an explanation of the radiosensitization effect observed in many experiments.
