Abstract:Ischemic stroke is caused by an interruption in the flow of blood to the brain and a risk factor for death and disability. Recent genome-wide association studies have identified more than 40 common sequence variants associated with ischemic stroke. However, the results are not always the same in populations with different genetic backgrounds. In the present study, we evaluated a hypothesis that a North Asian population living in a geographic area with unusually harsh environmental conditions would develop unique genetic risks. We investigated the candidate genes for ischemic stroke and risk factors in a Chinese Mongolian population which has not been explored previously. A total of 167 stroke cases and 176 controls were included in the study. Genotyping was performed by amplicon sequencing. The association was detected with single nucleotide polymorphisms (SNPs) located within genes NINJ2 (rs12425791) and ALDH2 (rs2238151) as well as intergenic rs9536591 were significantly associated with ischemic stroke, of which SNP rs12425791 of the NINJ2 gene was the strongest association. ALDH2 gene encodes mitochondrial aldehyde dehydrogenase, involved in the oxidative pathway of alcohol metabolism. Sex, age, body mass index and high blood pressure might be the risk factors. The current work also demonstrated genetic heterogeneity exists between Chinese and other populations. Our study provided the new insights into the genetic basis and environmental factors of ischemic stroke in Mongolian population.
The relationship between mannan-binding lectin gene polymorphisms and serum protein level in Mongls and Huis in China

Abstract:Objective:To analyze the gene polymorphisms at code 54 of exon 1 in mannose-binding lectin(MBL) gene and the palsma MBL concentrations in Mongols and Huis in China.Methods:MBL plasma concentrations were measured by ELISA method with human MBL ELISA kit,Single nucleotide polymorphisms of MBL gene were determined with sequence analysis method.Geneticanalysis was deployed with SHEsis software,and the statistics package for social science,SPSS11.0 was used in the study.Results:The median values of MBL plasma in 79 Mongols and 69 Huis were 1 686 ng/ml and 1 909 ng/ml respectively and no significant difference was found between them(Z=0.63,P=0.4).In 79 Mongols,the variant allele frequencies of the codon 54 of MBL gene was 0.203,62.0% of the 79 cases were A/A exon type,while 35.4% were A/B type and 2.50% were B/B type instead.In all 69 Huis,the variant allele frequencies was 0.268,58.0% of 40 cases were A/A exon type,while 30.4% were A/B type and 11.6% were B/B type instead.The frequency of point mutation at +230 site was 0.203.No significant difference of variant allele frequencies was found between the Mongols and Huis nationalities(χ2=1.772,P=0.183).Neither C nor D exon type were found in the study.The A/A wild type was associated with the highest plasma concentration with median values,the A/B type was associated with lower MBL levels and the B/B type with the lowest levels(χ2=86.526,P<0.01).Conclusion:There are only three genotypes,A/A,A/B and B/B types are identified in the Mongols and Huis in the study,and A/A type has predominance higher plasma protein level than in A/B type,which is also higher than in B/B type.
Analysis of APOE and SLCO1B1 Gene Polymorphism and Correlation with Dyslipidemia in China

Abstract:Background: The aim of the study was to analyze the polymorphism of APOE and SLCO1B1 genotype and correlation with blood lipid levels in patients with dyslipidemia in Hunan Province, China. Methods: Concentration of total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were retrospectively analyzed from 396 patients with dyslipidemia from 2016 to 2020. The APOE and SLCO1B1 genotypes were detected by PCR-fluorescent probe. SPSS21.0 software was used to analyze the gene frequency and their correlation. Results: The frequency of E3/E3 genotype was the highest in the APOE gene, reaching 64.14%. The concentrations of LDL-C and TC in E4 phenotype group were the highest and were statistically different from those in E2 and E3 phenotype groups (p < 0.05). There were significant differences in E4 phenotype between the dyslipidemia group and the normolipidemia group (p < 0.05). The frequency of *1b/*1b genotype was the highest in the SLCO1B1 gene, reaching 41.41%. The concentrations of TC and LDL-C in the intermediate OA1P1B1 function group were higher than those in the normal OA1P1B1 function group and the differences were statistically significant (p < 0.05). Conclusions: The genotypes of APOE and SLCO1B1 gene are correlated with concentration of TC and LDL-C, and their genotypes frequency distribution have regional differences.

Abstract:This Account outlines our efforts in ligand development for asymmetric hydrogenation. The successful development of three classes of ligands is presented, including (1) ligands with phosphocyclic motifs, (2) ligands with atropisomeric backbones, and (3) bisphosphine ligands inspired by the structure of 2,3- O-isopropylidene-2,3-dihydroxyl-1,4-bis(diphenylphosphino)butane (DIOP). With this large ligand toolbox, we have prepared many pharmaceutically valuable chiral products efficiently.
Differences in Genotype and Allele Frequency Distributions of Polymorphic Drug Metabolizing Enzymes CYP2C19 and CYP2D6 in Mainland Chinese Mongolian, Hui and Han Populations

Abstract:What is known and Objective: Cytochrome P450 2C19 (CYP2C19) and CYP2D6 are important xenobiotic metabolic enzymes and both show considerable genetic variability between Orientals and Caucasians. There are known marked heterogeneity in susceptibility to various cancers and hypertension among Chinese Mongolian, Hui and Han ethnic groups, but the molecular mechanisms are unknown. Our objective was to investigate the patterns of distribution of CYP2C19 and CYP2D6 polymorphisms among healthy Chinese subjects to determine whether any observed inter-ethnic variability might be worth further investigation as possible contributors to the known differences in disease prevalence. Methods: Blood samples were collected from 454 unrelated Chinese healthy subjects (214 Han, 111 Hui, 129 Mongolian) for genotyping analysis. The single nucleotide polymorphisms (SNPs) CYP2C19*2 (681G>A in exon 5), CYP2C19*3 (636G>A in exon 4) and CYP2D6*10 (188C>T in exon 1) were determined by the polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) method. Results and Discussion: Significantly higher frequencies of the CYP2C19 poor metabolic genotypes were observed in Chinese Han (18.7%), Chinese Hui (25.0%) and Chinese Mongolian (10.9%) subjects than has been reported for Caucasians (1.73.0%, P < 0.01). The prevalent defective allele CYP2C19*2 occurred more frequently in both Chinese Hui (32.4%) and Han (29.7%) than in Chinese Mongolian (18.2%, P < 0.01) subjects. The CYP2C19*2 and CYP2C19*3 defective alleles were significantly more frequent in Chinese Han and Chinese Hui ethnic groups than have been reported for Caucasians (11.116.3% and 00.2%, P < 0.01). CYP2D6*1/*10 heterozygotes and CYP2D6*10/*10 homozygotes were observed more frequently in Chinese Han (43.1% and 27.2%), Hui (40.6% and 30.7%) and Mongolian subjects (31.3% and 9.6%, both P < 0.01) than have been reported for Caucasians (5.5% and 0.3%, P < 0.01). In Chinese Mongolians, the CYP2D6*10 allele occurred at a frequency (25.2%, P < 0.01) intermediate between those reported for Caucasians and the other two Chinese ethnic populations. What is new and Conclusions: This is first report of interethnic differences in frequencies of functional CYP2C19 and CYP2D6 genes among Chinese Mongolian, Hui and Han populations. These differences may be important in explaining reported inter-ethnic differences in disease prevalence and response to drugs.
Analysis of Dyslipidemia Correlation and Polymorphisms of ApoE and SLCO1B1 Gene in Dyslipidemia People

Abstract:Objective To investigate the frequency distribution of ApoE and SLCO1B1 genotypes in abnormal blood lipid lev-els in Xiangya Hospital,explore the correlation of ApoE and SLCO1B1 genotypes with various indicators of dyslipidemia,and provide evidence for prevention and treatment of atherosclerotic diseases.Methods Blood lipid data including TG,TC,LDL-C and HDL-C in 87 cases of dyslipidemia people in Xiangya Hospital from June in 2016 to April in 2017 were collected.The ApoE and SLCO1B1 genotypes were detected by PCR fluoroscopy in 87 cases of dyslipidemia.The distribution of gene fre-quency was analyzed and the differences of blood lipid indexes among the genotypes were compared.Results The frequen-cies of each ApoE genotype in 87 cases of dyslipidemia were E2/E2 1.15%,E2/E3 13.79%,E2/E4 1.15%,E3/E3 56.32%,E3/E4 26.44% and E4/E4 1.15%,respectively.The highest proportion of allele frequency was E3 with the per-cent of 76.44%,E2 and E4 occupied 8.62% and 14.94%,respectively.The concentration of LDL-C in E4 phenotype group was higher than that in E2 group and E3 group,and there was no significant difference in the levels of TG,TC,LDL-C and HDL-C.The frequencies of each SLCO1B1 genotype were *1a/*1a 6.90%,*1a/*1b 36.70%,*1a/*15 13.79%,*1b/*1b 26.44% and *1b/*15 16.09%.The highest frequency of each haplotype was *1b with the percent of 52.87%,*1a and *15 occupied 32.18% and 14.94%,respectively,and no * 5 type was detected.There were no significant differ-ences in TG,TC,LDL-C and HDL-C concentrations between SLCO1B1 groups.Conclusion ApoE gene frequency distribu-tion was uneven.Its polymorphism be related with the lipid levels.The frequency distribution of SLCO1B1 gene had racial differences,and its polymorphism was not related with lipid level.According to the the genotypes of ApoE and SLCO1B1, clinician can choose the right dose of drug to prevent coronary atherosclerotic disease.

Abstract:Cytochrome P450 2D6 (CYP2D6) and endothelial nitric oxide synthase (eNOS) are important in the cardiovascular disease susceptibility and drug response. The methylation of cytosine in the promoter of genes could influence expression. The interindividual differences of drug response could only be explained partly by the polymorphisms of drug disposition genes. This study was aimed to investigate the genetic and epigenetic polymorphisms of CYP2D6 and eNOS in Chinese Tibetan, Mongolian, Uygur, and Han volunteers. Blood samples were collected from 842 unrelated Chinese healthy subjects (323 Tibetan, 134 Mongolian, 162 Uygur, and 223 Han) for genotyping and part of the DNA samples were subjected to cytosine methylation analysis. Significant genetic and epigenetic interethnic polymorphisms of eNOS and CYP2D6 were found in the four Chinese groups. The 4b-G-T wild-type haplotype of eNOS was the most common in Chinese Tibetan (87.1%), Mongolian (79.9%), Uygur (76.4%), and Han (79.5%), respectively. eNOS T-786C creates a new CpG site and a significantly higher methylation level was found in −786CC than that in −786TC in Chinese Han (P = 0.0485) and total Chinese (P = 0.0104). CYP2D6 A-678 (rs28633410) makes the symmetrical CpG site changed to be an asymmetrical CpA site, and the methylation level of cytosine in the CpG context of G-678A was significantly higher than that in the CpA. The results of the present study could be useful for understanding the interindividual disparity of cardiovascular disease susceptibility and drug response among different Chinese ethnic groups and would provide more evidences for the precise medication of Chinese.
A case-control study of the association between the polymorphisms of APOE and SLCO1B1 and the severity of coronary artery disease

Abstract:Objective The single nucleotide polymorphisms (SNPs) of APOE and SLCO1B1 were examined to explore their association with the risk and severity of coronary heart disease(CAD). Methods A total of 1267 cases of consecutive coronary heart disease (CAD)-suspected inpatients visiting department of Cardiology in Peking University Peoples' Hospital from March 2017 to november were recruited into this case-control study, and then 391 CAD cases and 223 non-CAD controls were enrolled for final analysis after screening by coronary angiography and exclusion criteria. The severity of the CAD cases were evaluated according to Gensini scores. The SNPs of APOE(388T>C, 526C>T) and SLCO1B1(388A>G, 521T>C) were detected using Real-time PCR and further verified using Sanger sequencing. Environmental risk factors were collected, and the correlations between SNPs of APOE and SLCO1B1 and the risk and severity of CAD were performed by SPSS version 16.0. Results The SNPs of all the subjects included in CAD group and non-CAD group were successfully detected, with an accordance of 100％ to Sanger sequencing. The distribution of APOE and SLCO1B1 gene were subjected to Hardy-Weinberg. The distributions of APOE gene ε3/ε3 genotypes and ε3 allele were most commonly found in both CAD group and non-CAD group (ε3/ε3: 70.8％,73.1％;ε3: 83.5％,85.2％;respectively). APOE genotypes and alleles were comparable between the CAD cases and non-CAD controls (P>0.05). The frequencies of APOE gene ε4+genotype were more likely to be found in the subgroup of CAD with Gensini score≥72 (P<0.05). The distributions of SLCO1B1 gene *1b/*1b genotypes and *1b allele were most commonly found in both CAD group and non-CAD group (*1b/*1b: 37.3％, 36.8％; *1b: 60.1％, 61.7％; respectively). There was no significant difference in genotype and allele frequencies of SLCO1B1 between the two groups and among subgroups with different severity of CAD (P>0.05). Conclusion This study observed no association between SNPs of APOE, SLCO1B1 and the risk of CAD in this population. However, APOE gene ε4 +genotype may increase the severity of CAD.
The prognostic difference study on the individualized clopidogrel administration in Hmong and Dong patients based on the CYP2C19 gene polymorphism after percutaneous coronary intervention

Abstract:This study explored the distribution characteristics of CYP2C19 gene polymorphism among Hmong and Dong patients in the Qiandongnan region of Guizhou province after percutaneous coronary intervention (PCI). The aim was to assess the clinical impact of individualized clopidogrel administration based on CYP2C19 genotypes. A total of 208 patients were classified into ultra-fast, fast, intermediate, and slow metabolic groups. They were randomly assigned to clopidogrel individualized administration (IA) or conventional treatment (CA) groups. Patients were followed for 6 months to evaluate major adverse cardiovascular events (MACE) and adverse reactions. The CYP2C19 genotype distribution was in Hardy-Weinberg equilibrium, showing consistency in the population. While no significant ethnic differences were found in genotype and metabolic distribution, allele distribution varied, with Hmong patients exhibiting a higher proportion of CYP2C19*1 alleles than Dong patients. Following individualized administration, the IA group demonstrated lower incidences of non-fatal myocardial infarction and emergency revascularization compared to the CA group. Bleeding events were higher in the IA group, but the total MACE incidence was lower. No statistical difference in MACE and adverse drug reactions (ADR) was observed in the CA group across metabolic types, but MACE incidence was higher in intermediate and slow metabolic groups. In the IA group, no significant difference in MACE was noted among metabolic types, but ADR incidence varied significantly, particularly in dyspnea. The study highlighted significant CYP2C19 allele distribution differences between Hmong and Dong patients post-PCI in Qiandongnan. Patients with slow metabolic profiles demonstrated higher MACE incidence with conventional clopidogrel dosage, whereas CYP2C19-guided therapy reduced MACE without increasing bleeding risk. These findings supported clinical individualized clopidogrel administration in post-PCI patients in the Qiandongnan region, contributing to rational clopidogrel use.
Functional Allele and Genotype Frequencies of CYP1A2 ,CYP2B6 and Inos among Mainland Chinese Tibetan, Mongolian, Uygur and Han Populations

Abstract:What is known and objectiveCytochrome P450 1A2 (CYP1A2), CYP2B6 and inducible nitric oxide synthase (iNOS) are involved in the metabolism and action of many important therapeutic drugs, and genetic variants have been associated with interethnic differences in response to treatment, including chemotherapy.MethodsEight hundred and forty-two unrelated Chinese healthy subjects (323 Tibetan, 134 Mongolian, 162 Uygur and 223 Han) were recruited for genotyping. Frequencies of CYP1A2 -163C>A, CYP2B6 516G>T and iNOS -954G>C were determined by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods.Results and discussionThe frequency of CYP1A2-163A was higher in Chinese Mongolian (0698) than in Chinese Tibetan (0633), Uygur (0633) and Han populations (0608, P < 005), respectively. The frequency of CYP1A2-163A in the Chinese population (total, 0636) was intermediate between those reported in Caucasians (0682, P < 005) and Africans (0549, P < 001). The frequency of CYP2B6 516T in Chinese Uygur (0287) was significantly higher than those in Chinese Tibetan (0147, P < 001) and Mongolian (0179, P < 001), respectively, but was similar to the frequency in Chinese Han (0226). The frequencies of CYP2B6 516T were in the order of Africans (0500) > Caucasians (0286) > Chinese (0200). The variant iNOS-954C was rare in Chinese Tibetan (0005), Mongolian (0004), Uygur (0000) and Han (0007), respectively, but showed higher frequencies in African ethnic groups. The frequencies of iNOS-954C were in the order of Africans (0098) > Chinese (0004) > Caucasians (0000).What is new and conclusionThis is the first report of the distribution frequencies of functional CYP1A2, CYP2B6 and iNOS genes among mainland Chinese Tibetan, Mongolian, Uygur and Han populations. These results should help inform studies of interethnic differences in disease susceptibility or drug responses.
Relationship between gene polymorphism of LOX-1 and coronary artery disease in elderly Chinese Han population

Abstract:Objective To study the relationship between polymorphism of lectin-like oxidized LDL receptor HLOX-1) gene and coronary artery disease in elderly Chinese Han population.Methods 165 subjects were divided into 2 groups according to results of coronary angiography:38 cases with normal coronary artery served as controls and 127 cases with coronary artery disease constituted coronary heart disease group.PCR single strand conformational polymorphism(PCR-SSCP) method was used to analyze LOX-1 501G/C and 3'UTR loci C/T gene polymorphism.The relationships of C genotype and T genotype with high sensitivity C-reactive protein and fibrinogen were analyzed.Results No polymorphism of Exon 4 was detected in all subjects.There was no significant difference in the 3' UTR polymorphism between patients with coronary artery disease and controls.C/T genotype could be seen at LOX-1 3'UTR.Compared with control group.The number of diseased coronary arteries and degree of coronary stenosis had no statistically significant relation with difference in frequency of 3'UTR locus C/T gene.hs-CRP and fibrinogen levels had no statistically significant difference between C genotype and T genotype.Conclusions The results show that the LOX-1 polymorphism may be not associated with the risk of coronary artery disease in the Chinese Han population.

Abstract:Abstract Background Apolipoprotein E (ApoE) and solute carrier organic anion transporter family member 1B1 (SLCO1B1) regulate lipid metabolism. However, the relationship between genetic polymorphisms of APOE and SLCO1B1 and cerebral infarction (CI) remains unclear. Methods A total of 938 CI patients and 1028 control participants were included in the study. The rs429358 and rs7412 single nucleotide polymorphisms (SNPs) in the APOE gene and rs2306283 and rs4149056 SNPs in the SLCO1B1 gene were analyzed by fluorescence polymerase chain reaction (PCR). Results The genotype ɛ3/ɛ3 was the most common APOE genotype, with ɛ3 being the allele with the highest frequency, followed by ɛ4 and ɛ2. Statistically significant differences of genotype ɛ2/ɛ2 (χ 2 = 3.866, P = 0.049), ɛ2/ɛ3 (χ 2 = 20.030, P < 0.001), ɛ3/ɛ4 (χ 2 = 16.960, P < 0.001), and ɛ4/ɛ4 (χ 2 = 4.786, P = 0.029) between CI patients and controls were detected. The SLCO1B1 genotype *1b/*1b and haplotype *1b showed the highest frequency in the study sample. There was no statistically significant difference in the frequencies of SLCO1B1 genotypes and haplotypes among CI patients comparing with controls. Moreover, ε4 carriers had significantly higher low-density lipoprotein-cholesterol (LDL-C) and apolipoprotein B (Apo-B) and lower apolipoprotein A1 (Apo-A1)/Apo-B levels than ε2 and ε3 carriers, but ε2 carriers showed lower LDL-C and Apo-B and higher Apo-A1/Apo-B than ε3 and ε4 carriers. Further, logistic regression analysis revealed that high LDL-C, high ApoB, smoking, hypertension and the ε4 allele were risks for the presence of CI. Conclusions This study indicated that the APOE SNPs rs429358 and rs7412 may be associated with susceptibility to cerebral infarction in southern Chinese Hakka population.

Abstract:Objective The aim of this study was to characterize the population frequency of SLCO1B1 polymorphic variants in three distinct healthy Asian populations, namely Chinese (n=100), Malay (n=100) and Indian (n=100), and to explore the association between haplotype-tagged single nucleotide polymorphisms (htSNPs) on hepatic SLCO1B1 mRNA expression.Methods The distribution of polymorphic variants in the SLCO1B1 gene at eight loci that spanned approximately 48 kb was investigated in the three different Asian ethnic groups and in 32 non-cancerous liver tissues from Chinese patients.Results Of the 26 polymorphisms screened, we found eight polymorphic variants that differed in genotypic and allelic frequencies between the Chinese, Malay and Indian populations. Significant interethnic differences were observed in the genotype frequency distributions across the promoter SNP [g.-11187G > A (P=0.030)] as well as three coding region SNPs [c.388G > A (P < 0.001); c.571T > C (P < 0.001); c.597C > T (P < 0.001)] in the healthy subjects. Haplotype analysis revealed 12 different haplotypes in both the Chinese and Malay populations and 18 haplotypes in the Indian population. In both the Malay and Indian populations, the htSNPs were c.388A > G, c.571T > C and c.597C > T, whereas in the Chinese population they were g.-11187G > A, c.388A > G and c.597C > T. The c.388A > G and c.597C > T htSNPs accounted for more than 70% of the variations between the three major haplotypes in each Asian ethnic group. In terms of the c.388A > G htSNPs, genotypic-phenotypic association analyses revealed that there was no effect on SLCO1B1 expression in hepatic tissues; in addition, no genotypic-phenotypic associations were evident with regards to the c.597C > T htSNP.Conclusion Future studies should investigate the phenotypic effects of the c.388A > G htSNP on the disposition of OATP1B1 substrates in Asian populations.

Abstract:Objective To discover the relationship between ALDH2 Glu487Lys gene polymorphism and coronary artery disease in Han population.Methods 490 patients with coronary artery disease(CHD group) which were identified by coronary angioplasty(coronary narrow ≥50%) and 433 patients without CAD were recruited from Zhongshan Hospital(control group).Taqman 5'nuclease polymerase chain reaction assay was performed to screen the ALDH2 Glu487Lys gene polymorphism.The difference of the distribution of ALDH2 gene was compared between CAD group and control group;We also carried out the subgroup analysis according to the risk factors for coronary heart disease.Results The difference of genotype distribution(P=0.22) between CAD group and control group showed no statistical signification.Subgroup analysis showed obvious statistical difference of ALDH2 gene type distribution(P<0.05) between CAD group and control group in the following four subgroups:no smoking,no drinking,no hypertention history and BMI<25(kg/m2) population.Conclusion There is no association between ALDH2 genetic polymorphism and coronary artery disease in Chinese Han population,but the ALDH2-AA genotype may affect coronary artery disease in Chinese Han population without drinking history,somking history,hypertention or BMI<25(kg/m2).

Abstract:AIMS:To explore the association between six single-nucleotide polymorphisms in OLR1, PON1, MTHFR gene and the angiographical characteristics of coronary atherosclerosis to determine if any of the conventional factors correlate with genetic polymorphisms in the advent of the disease.METHODS:We examined rs1801131, rs1801133, rs3736232, rs3736234, rs854563 and rs662 by TaqMan® SNP Genotyping Assays in 1075 subjects that underwent angiography. The angiographical characteristics of coronary atherosclerosis were defined by the Gensini Score system.RESULTS:The T allele of rs1801133 was associated with coronary atherosclerosis severity with the OR = 1.49, 95% CI = 1.04-2.14. In MTHFR gene, haplotype T-A was a susceptibility haplotype to coronary atherosclerosis (OR = 1.27, 95% CI = 1.06-1.51) whereas haplotype C-A had a protective effect (OR = 0.83, 95% CI = 0.70-0.99). In addition, several synergistic effects between rs1801133 and conventional risk factors such as diabetes and smoking were found.CONCLUSIONS:Collectively, the results demonstrate that the T allele of rs1801133 conferred an increased risk for coronary atherosclerosis. The MTHFR C-A haplotype was a protective haplotype, while T-A haplotype was a susceptibility haplotype. The presence of the T allele of rs1801133 increases the odds of coronary atherosclerosis severity when associated with conventional risk factors.

Abstract:BackgroundThe present study is aimed to evaluate difference of lipid metabolism related gene single nucleotide polymorphisms (SNPs) with ischemic stroke (IS) in Han and Uighur population of Xinjiang, China.MethodsFour hundred eight patients with ischemic stroke and 347 unrelated healthy individuals of age and sex matched were genotyped for Apolipoprotein A5 (ApoA5), lipoprotein lipase (LPL), Cholesteryl ester transfer protein (CETP) and low-density lipoprotein receptor (LDL-R) genes. Their mutation difference was analyzed by SNaP shot techniques. GeneMapper4.1 SPSS20.0 software was used for data management and analysis. Using a single locus analysis, the distribution difference of genotype loci in ischemic stroke cases and controls were detected to assess the genetic risk factors of ischemic stroke.ResultsSignificance differences of genotype distribution in ischemic stroke cases and controls were observed in LDLR rs688 in Han and Uighur population in recessive model from analysis of single gene locus. It also was found that dramatic difference of triglyceride (TG) of LPL rs328 and systolic blood pressure in CETP rs708277 of total population. In binary logistic regression analysis of total studied population, ischemic stroke was observed significantly associated with LDLR rs688 both addictive model (TT/CC, adjusted OR = 1.47, 95% CI = 1.04–2.07) and recessive model (TT/CT + CC, adjusted Odds ratio (OR) = 2.66, 95% Confidence Interval (CI) = 1.37–5.14). In Han population, ischemic stroke was observed significantly associated with rs688 both in addictive model (TT/CC, adjusted OR = 3.27, 95% CI = 1.06–10.05). In Uighur population, no significant association was found between gene polymorphisms and the risk of ischemic stroke. Combined analysis of multiple gene and loci, interaction effects of LDLR rs688 C/T, ApoA5 rs662799 A/G and CETP rs708272 C/T denoted a significant influence on IS susceptibility.ConclusionSingle nucleotide polymorphisms of lipid metabolism relative gene were significantly associated with the morbidity of ischemic stroke in Han population. The interaction effects of rs688 C/T with ApoA5 rs662799 A/G and CETP rs708272 C/T promoted the occurrence of IS.

Abstract:Objective To analyze risk factors for metabolic syndrome in Mongolia versus Han nationalities in Xilinhaote city. Methods Using the epidemiology investigation data of health examination,we calculated the prevalence of metabolic syndrome of Mongolia and Han nationalities, then used logistic regression model to explore risk factors of two nationalities. Results The crude prevalence of MS in Mongolia and Han nationality was 34.3% and 24.6% respectively. The multivariate logistic regression showed that male, the meat-rich diets and aging(OR:2.18, 1.92, 1.04 respectively)were the risk factors for Mongolia nationality, and smoking, family history of hypertension, drinking, the meatrich diets, aging (OR: 1.89, 1.84, 1.72, 1.61 and 1.04 respectively) were the risk factors for Han nationality. Conclusions Xinlinhaote population has higher MS prevalence, and different nationalities have different risk factors. We should take preventive actions to control it.

Abstract:BACKGROUND Dyslipidemia is a common complication in patients with diabetes mellitus (DM) that increases the risk of cardiovascular disease. Genetic polymorphisms have been implicated in the development of dyslipidemia. AIM To investigate the association between polymorphisms of candidate genes involved in lipid metabolism and dyslipidemia in Chinese patients with DM. METHODS A cross-sectional study was conducted on 1098 Chinese patients with DM recruited from multiple healthcare centers. Demographic and clinical data were collected, and dyslipidemia was defined according to the National Cholesterol Education Program Adult Treatment Panel III guidelines. Genomic DNA was extracted from blood samples and genotyping for selected polymorphisms of candidate genes (APOE , LPL , CETP , and others) was performed using PCR and DNA sequencing techniques. Statistical analyses were performed using logistic regression models adjusted for potential confounding factors. RESULTS The study population consisted of 578 males (52.6%) and 520 females (47.4%), with a mean age of 58.4 ± 12.2 years. The prevalence of dyslipidemia was 64.8%. Significant associations were found between dyslipidemia and the APOE rs7412 T/T, APOE rs429358 C/C, LPL rs328 G/G, and CETP rs708272 G/G genotypes after adjusting for covariates. Subgroup analyses showed generally consistent associations across subgroups, although some variations in effect sizes were observed. CONCLUSION This study identified significant associations between genetic polymorphisms of APOE , LPL , and CETP genes and dyslipidemia in Chinese patients with DM.

Abstract:Objectives To investigate the epidemiologic features and risk factors in ischemic stroke among the adults both in Mongolian and Han nationalities in the rural area of Fuxin district, Liaoning province and to provide the bases for prevention and treatment. Methods The data of health conditis including sex, age(≥35), nationality,history of hypertension,cardio-cerebrovascular diseases, smoking and alcohol consumption in 30 214 rural residents we collected and studies. Results The prevalence of ischemic stroke in the rural residents of Mongolian nationality was 2.36%, and the standardized rate was 2.48%, while in the Han nationality was 1.73%, and the standardized rate was 1.78%. With increasing age and levels of hypertension, the prevalence of stroke showed a trend of significant increase. Multivariate logistic stepwise regression analysis showed that the age, sex, hypertension, and family history of hypertension were the same risk factors in both nationalities, the risk of having stroke in the hypertensive populations was 5 to 8 times higher than that in the non-hypertensive populations.and at the same time, the education also had some effects on the prevalence of stroke in the rural residents of Han nationality. Conclusions The prevalence of ischemic stroke in the rural residents of Mongolian nationality in fuxin county, Liaoning province is higher than that of Han nationality. This may be associated with the lifestyle and genetic factor. It is a great task to improve the awareness of risk factors of hypertension and relevant knowledge of the rural residents in order to reduce the prevalence of stroke.
