Abstract:Springtime bud-break and shoot development induces substantial carbon (C) costs in trees. Drought stress during shoot development can impede C uptake and translocation. This is therefore a channel through which water shortage can lead to restricted shoot expansion and physiological capacity, which in turn may impact annual canopy C uptake. We studied effects of drought and re-hydration on early season shoot development, C uptake and partitioning in five individual 10-year old Picea mariana [black spruce] trees to identify and quantify dynamics of key morphological/physiological processes. Trees were subjected to one of two treatments: (i) well-watered control or (ii) drought and rehydration. We monitored changes in morphological [shoot volume, leaf mass area (LMA)], biochemical [osmolality, non-structural carbohydrates (NSC)] and physiological [rates of respiration (R d ) and light-saturated photosynthesis ( A sat )] processes during shoot development. Further, to study functional compartmentalization and use of new assimilates, we 13 C-pulse labeled shoots at multiple development stages, and measured isotopic signatures of leaf respiration, NSC pools and structural biomass. Shoot water potential dropped to a minimum of −2.5 MPa in shoots on the droughted trees. Development of the photosynthetic apparatus was delayed, as shoots on well-watered trees broke-even 14 days prior to shoots from trees exposed to water deficit. R d decreased with shoot maturation as growth respiration declined, and was lower in shoots exposed to drought. We found that shoot development was delayed by drought, and while rehydration resulted in recovery of A sat to similar levels as shoots on the well-watered trees, shoot volume remained lower. Water deficit during shoot expansion resulted in longer, yet more compact (i.e., with greater LMA) shoots with greater needle osmolality. The 12 C: 13 C isotopic patterns indicated that internal C partitioning and use was dependent on foliar developmental and hydration status. Shoots on drought-stressed trees prioritized allocating newly fixed C to respiration over structural components. In conclusion, temporary water deficit delayed new shoot development and resulted in greater LMA in black spruce. Since evergreen species such as black spruce retain active foliage for multiple years, impacts of early season drought on net primary productivity could be carried forward into subsequent years.

Abstract:The occurrence of extreme drought poses a severe threat to forest ecosystems and reduces their capability to sequester carbon dioxide. This study analysed the impacts of a central European summer drought in 2015 on gross primary productivity (GPP) at two Norway spruce forest sites representing two contrasting climatic conditions—cold and humid climate at Bílý Kříž (CZ-BK1) vs. moderately warm and dry climate at Rájec (CZ-RAJ). The comparative analyses of GPP was based on a three-year eddy covariance dataset, where 2014 and 2016 represented years with normal conditions, while 2015 was characterized by dry conditions. A significant decline in the forest GPP was found during the dry year of 2015, reaching 14% and 6% at CZ-BK1 and CZ-RAJ, respectively. The reduction in GPP coincided with high ecosystem respiration (Reco) during the dry year period, especially during July and August, when several heat waves hit the region. Additional analyses of GPP decline during the dry year period suggested that a vapour pressure deficit played a more important role than the soil volumetric water content at both investigated sites, highlighting the often neglected importance of considering the species hydraulic strategy (isohydric vs. anisohydric) in drought impact assessments. The study indicates the high vulnerability of the Norway spruce forest to drought stress, especially at sites with precipitation equal or smaller than the atmospheric evaporative demand. Since central Europe is currently experiencing large-scale dieback of Norway spruce forests in lowlands and uplands (such as for CZ-RAJ conditions), the findings of this study may help to quantitatively assess the fate of these widespread cultures under future climate projections, and may help to delimitate the areas of their sustainable production.

Abstract:In the future, periods of strongly increased temperature in concert with drought (heat waves) will have potentially detrimental effects on trees and forests in Central Europe. Norway spruce might be at risk in the future climate of Central Europe. However, Douglas-fir is often discussed as an alternative for the drought and heat sensitive Norway spruce, because some provenances are considered to be well adapted to drier and warmer conditions. In this study, we identified the physiological and growth responses of seedlings from two different Douglas-fir provenances to increased temperature and atmospheric drought during a period of 92 days. We analysed (i) plant biomass, (ii) carbon stable isotope composition as an indicator for time integrated intrinsic water use efficiency, (iii) apparent respiratory carbon isotope fractionation as well as (iv) the profile of polar low molecular metabolites. Plant biomass was only slightly affected by increased temperatures and atmospheric drought but the more negative apparent respiratory fractionation indicated a temperature-dependent decrease in the commitment of substrate to the tricarboxylic acid cycle. The metabolite profile revealed that the simulated heat wave induced a switch in stress protecting compounds from proline to polyols. We conclude that metabolic acclimation successfully contributes to maintain functioning and physiological activity in seedlings of both Douglas-fir provenances under conditions that are expected during heat waves (i.e. elevated temperatures and atmospheric drought). Douglas-fir might be a potentially important tree species for forestry in Central Europe under changing climatic conditions.

Comparison of morphological and physiological response to drought stress among temperate forest understory forbs and graminoids

Abstract:Drought stress can profoundly affect plant growth and physiological vitality, yet there is a notable scarcity of controlled drought experiments focused on herbaceous species of the forest understory. In this study, we collected seeds from five forb and four graminoid species growing in the temperate forest understory of the Czech Republic. These seeds were germinated under controlled glasshouse conditions and subjected to moderate drought stress for five weeks. We assessed biomass partitioning, stomatal and leaf morphology, leaf gas exchange, minimum leaf conductance (gmin), and chlorophyll fluorescence parameters. The comparison of two ecological guilds revealed that graminoids exhibited a higher root-to-shoot ratio, improved water-use efficiency, greater carboxylation efficiency, and enhanced non-photochemical quenching under drought conditions compared to forbs. In contrast, forbs had significantly lower gmin, along with higher total biomass and total leaf area. Despite these differences in morpho-physiological functional traits, both groups experienced a similar relative reduction in biomass during drought stress. Key predictors of biomass accumulation under drought included photochemical quenching, stomatal traits, total leaf area and gmin. A negative correlation between biomass and gmin suggests that plants with lower residual water losses after stomatal closure can accumulate more biomass under drought stress. Additionally, gmin was positively correlated with guard cell length, suggesting that larger stomata contribute to higher residual water loss. Graminoids exhibited morpho-physiological modifications that enhanced drought resistance, indicating a greater emphasis on stress tolerance as a survival strategy. In contrast, forbs maintained higher biomass and total leaf area, reflecting a competitive strategy for maximizing resource acquisition.

Abstract:Abstract The ability of tree species to acclimate and tolerate projected increases in drought frequency and intensity has fundamental implications for future forest dynamics with climate change. Inquiries to date on the drought tolerance capacities of tree species, however, have focused almost exclusively on mature trees with scant in situ work on seedlings, despite the central role that regeneration dynamics play in forest responses to changing conditions. We subjected naturally established seedlings of co-dominant subalpine conifer species (Abies lasiocarpa and Picea engelmannii) in the southern Rocky Mountains to 2 years of in situ summer precipitation exclusion, simulating summer drought conditions similar to a failure of the North American monsoon. We compared the morphological and physiological responses of seedlings growing in drought vs. ambient conditions to assess the relative changes in drought tolerance traits as a function of seedling size. Drought treatments had a marked impact on soil moisture: volumetric water content averaged ≈5–8 % in drought treatments and ≈8–12 % in ambient controls. We detected no significant shifts in morphology (e.g. root biomass, leaf:stem area ratio) in response to drought for either species, but net photosynthesis in drought treatments was 78 % lower for spruce and 37 % lower for fir. Greater stomatal control associated with increasing stem diameter conferred greater water use efficiencies in larger seedlings in both species but was not significantly different between drought and ambient conditions, suggesting an overall lack of responsivity to water stress and a prioritization of carbon gain over investment in drought mitigation traits. These results indicate a canonization of traits that, while useful for early seedling establishment, may portend substantial vulnerability of subalpine seedling populations to prolonged or recurrent droughts, especially for spruce.

Sap flow and growth response of Norway spruce under long-term partial rainfall exclusion at low altitude

Abstract:Introduction: Under ongoing climate change, more frequent and severe drought periods accompanied by heat waves are expected in the future. Under these conditions, the tree's survival is conditioned by fast recovery of functions after drought release. Therefore, in the presented study, we evaluated the effect of long-term water reduction in soil on tree water use and growth dynamics of Norway spruce. Methods: The experiment was conducted in two young Norway spruce plots located on suboptimal sites at a low altitude of 440 m a.s.l. In the first plot (PE), 25% of precipitation throughfall was excluded since 2007, and the second one represented the control treatment with ambient conditions (PC). Tree sap flow, stem radial increment, and tree water deficit were monitored in two consecutive growing seasons: 2015-2016, with contrasting hydro-climatic conditions. Results: Trees in both treatments showed relatively isohydric behavior reflected in a strong reduction of sap flow under the exceptional drought of 2015. Nevertheless, trees from PE treatment reduced sap flow faster than PC under decreasing soil water potential, exhibiting faster stomatal response. This led to a significantly lower sap flow of PE, compared to PC in 2015. The maximal sap flow rates were also lower for PE treatment, compared to PC. Both treatments experienced minimal radial growth during the 2015 drought and subsequent recovery of radial growth under the more the humid year of 2016. However, treatments did not differ significantly in stem radial increments within respective years. Discussion: Precipitation exclusion treatment, therefore, led to water loss adjustment, but did not affect growth response to intense drought and growth recovery in the year after drought.
Seasonality of PSII thermostability and water use efficiency of in situ mountainous Norway spruce (Picea abies)

Abstract:The stability of monocultural, even-aged spruce forests at lower altitudes in Central Europe is seriously threatened by the prospects of global climate change. The thermostability and water use efficiency of their photosynthetic apparatus might play a vital role in their successful acclimation. In this study, photosystem II (PSII) performance (OJIP transient, rapid light curves) and thermostability were analyzed in Norway spruce (Picea abies (L.) Karst.) throughout the growing season of the exceptionally warm year 2018 (May–September) in the Western Carpathians, Slovakia. These measurements were accompanied by analysis of pigment concentrations in the needles. In addition, gas-exchange temperature curves were produced weekly from June until September to obtain intrinsic water use efficiencies. At the beginning of the growing season, needles exposed to heat stress showed significantly higher basal fluorescence and lower quantum yield, performance index, critical temperature thresholds of PSII inactivation and non-photochemical yield in comparison to other months. The overall thermostability (heat-resistance) of PSII peaked in July and August, reflected in the lowest basal fluorescence and the highest quantum yield of PSII, critical temperature thresholds and yield of non-photochemical quenching under heat stress. Additionally, the ratio between chlorophyll and carotenoids was the highest in August and had a positive impact on PSII thermostability. Moreover, the high-temperature intrinsic water use efficiency was significantly higher during July and August than in June. Results show that 15-year-old trees of Picea abies at 840 m a.s.l. exhibited acclimative seasonal responses of PSII thermostability and intrinsic water use efficiency during an exceptionally warm year. Our results suggest that mountainous P. abies at lower altitudes can acclimate their photosynthetic apparatus to higher temperatures during summer.

Abstract:Dry spells strongly influence biomass production in forest ecosystems. Their effects may last several years following a drought event, prolonging growth reduction and therefore restricting carbon sequestration. Yet, our understanding of the impact of dry spells on the vitality of trees' above-ground biomass components (e.g., stems and leaves) at a landscape level remains limited. We analyzed the responses of Pinus sylvestris and Picea abies to the four most severe drought years in topographically complex sites. To represent stem growth and canopy greenness, we used chronologies of tree-ring width and time series of the Normalized Difference Vegetation Index (NDVI). We analyzed the responses of radial tree growth and NDVI to dry spells using superposed epoch analysis and further explored this relationship using mixed-effect models. Our results show a stronger and more persistent response of radial growth to dry spells and faster recovery of canopy greenness. Canopy greenness started to recover the year after the dry spell, whereas radial tree growth remained reduced for the two subsequent years and did not recover the pre-drought level until the fourth year after the event. Stem growth and canopy greenness were influenced by climatic conditions during and after drought events, while the effect of topography was marginal. The opposite responses of stem growth and canopy greenness following drought events suggest a different impact of dry spells on trees ́ sink and source compartments. These results underscore the crucial importance of understanding the complexities of tree growth as a major sink of atmospheric carbon.

Abstract:Drought is an important driver of plant survival, growth, and distribution. Water deficit affects different pathways of metabolism, depending on plant organ. While previous studies have mainly focused on the metabolic drought response of a single organ, analysis of metabolic differences between organs is essential to achieve an integrated understanding of the whole plant response. In this work, untargeted metabolic profiling was used to examine the response of roots, stems, adult and juvenile needles from Pinus pinaster Ait. full-sib individuals, subjected to a moderate and long lasting drought period. Cyclitols content showed a significant alteration, in response to drought in all organs examined, but other metabolites increased or decreased differentially depending on the analyzed organ. While a high number of flavonoids were only detected in aerial organs, an induction of the glutathione pathway was mainly detected in roots. This result may reflect different antioxidant mechanisms activated in aerial organs and roots. Metabolic changes were more remarkable in roots than in the other organs, highlighting its prominent role in the response to water stress. Significant changes in flavonoids and ascorbate metabolism were also observed between adult and juvenile needles, consistent with previously proven differential functional responses between the two developmental stages. Genetic polymorphisms in candidate genes coding for a Myb1 transcription factor and a malate dehydrogenase (EC 1.1.1.37) were associated with different concentration of phenylalanine, phenylpropanoids and malate, respectively. The results obtained will support further research on metabolites and genes potentially involved in functional mechanisms related to drought tolerance in trees.

Abstract:Climate change-related drought stress is expected to shift carbon partitioning toward volatile organic compound (VOC) biosynthesis. The effect of drought stress on VOC synthesis remains unknown in several tree species. Therefore, we exposed eastern white pine ( Pinus strobus ) plants to severe drought for 32 days and performed physiological analysis (chlorophyll content, leaf water content, and root/shoot index), biochemical analysis (non-structural carbohydrates, proline, lipid peroxidation, and antioxidant assay), and total root VOC analysis. Drought stress decreased the relative water and soil moisture contents. Root proline accumulation and antioxidant activity increased significantly, whereas leaf chlorophyll synthesis and fresh weight decreased significantly in drought-treated plants. A non-significant increase in sugar accumulation (leaves and roots), proline accumulation (leaves), antioxidant activity (leaves), and lipid peroxidation (leaves and roots) was observed in drought-treated plants. Drought stress caused a non-significant decline in root/shoot ratio and starch accumulation (leaves and roots) and caused a significant increase in root abscisic acid content. Drought-treated plants showed an increase in overall monoterpene synthesis (16%) and decline in total sesquiterpene synthesis (3%). Our findings provide an overall assessment of the different responses of VOC synthesis to severe water deficit that may help unravel the molecular mechanisms underlying drought tolerance in P. strobus .

Abstract:Understanding how trees prioritize carbon gain at the cost of drought vulnerability under severe drought conditions is crucial for predicting which genetic groups and individuals will be resilient to future climate conditions. In this study, we investigated variations in growth, tree-ring anatomy as well as carbon and oxygen isotope ratios to assess the sensitivity and the xylem formation process in response to an episode of severe drought in 29 mature white spruce ( Picea glauca [Moench] Voss) families grown in a common garden trial. During the drought episode, the majority of families displayed decreased growth and exhibited either sustained or increased intrinsic water-use efficiency (iWUE), which was largely influenced by reduced stomatal conductance as revealed by the dual carbon‐oxygen isotope approach. Different water-use strategies were detected within white spruce populations in response to drought conditions. Our results revealed intraspecific variation in the prevailing physiological mechanisms underlying drought response within and among populations of Picea glauca . The presence of different genetic groups reflecting diverse water-use strategies within this largely-distributed conifer is likely to lessen the negative effects of drought and decrease the overall forest ecosystems' sensitivity to it.

Abstract:Summary Drought affects the complex interactions between Norway spruce, the bark beetle Ips typographus and associated microorganisms. We investigated the interplay of tree water status, defense and carbohydrate reserves with the incidence of bark beetle attack and infection of associated fungi in mature spruce trees. We installed roofs to induce a 2‐yr moderate drought in a managed spruce stand to examine a maximum of 10 roof and 10 control trees for resin flow (RF), predawn twig water potentials, terpene, phenolic and carbohydrate bark concentrations, and bark beetle borings in field bioassays before and after inoculation with Endoconidiophora polonica and Grosmannia penicillata. Drought‐stressed trees showed more attacks and significantly longer fungal lesions than controls, but maintained terpene resin defenses at predrought levels. Reduced RF and lower mono‐ and diterpene, but not phenolic concentrations were linked with increased host selection. Bark beetle attack and fungi stimulated chemical defenses, yet G. penicillata reduced phenolic and carbohydrate contents. Chemical defenses did not decrease under mild, prolonged drought in our simulated small‐scale biotic infestations. However, during natural mass attacks, reductions in carbon fixation under drought, in combination with fungal consumption of carbohydrates, may deplete tree defenses and facilitate colonization by I. typographus.

Abstract:An increase in frequency, intensity and duration of drought events affects forested ecosystems. Trees react to these changes by adjusting stomatal conductance to maximize the trade-off between carbon gains and water losses. A better understanding of the consequences of these drought-induced physiological adjustments for tree growth could help inferring future productivity potentials of boreal forests. Here, we used samples from a forest inventory network in Canada where a decline in growth rates of black spruce (Picea mariana (Mill.) B.S.P.) and jack pine (Pinus banksiana Lamb.) occurred in 1988-1992, an exceptionally dry period, to verify if this growth decline resulted from physiological adjustments of trees to drought. We measured carbon and oxygen isotope ratios in growth rings of 95 spruces and 49 pines spanning 1985-1993. We used 13C discrimination (Δ13C) and 18O enrichment (Δ18O) as proxies for intrinsic water use efficiency and stomatal conductance, respectively. We studied how inter-annual variability in isotopic signals was linked to climate moisture index, vapor pressure deficit and annual snowfall amount. We found significantly lower Δ13C values over 1988-1990, and significantly higher Δ18O values in 1988-1989 and 1991 compared to the 1985-1993 averages. We also observed that a low climatic water balance and a high vapor pressure deficit were linked with low Δ13C and high Δ18O in the two study species, in parallel with low growth rates. The latter effect persisted into the year following drought for black spruce, but not for jack pine. These findings highlight that small differences in physiological parameters between species could translate into large differences in post-drought recovery. The stronger and longer lasting impact on black spruce compared to jack pine suggests a less efficient carbon use and a lower acclimation potential to future warmer and drier climate conditions.

Abstract:Aleppo pine (Pinus halepensis Mill.) is a pioneer species, highly competitive due to exceptional resistance to drought. To investigate the stress resistance in the first and second year of development, a steady-state drought experiment was implemented. Photosynthesis (A(net)), stomatal conductance and transpiration (E) were measured on three different sampling dates together with phloem soluble sugars, amino acids and non-structural proteins. Needle ascorbic acid (AsA) and reactive oxygen species were measured to evaluate the seedlings' drought stress condition in the final sampling. Drought impaired A(net) and E by 35 and 31%, respectively, and increased AsA levels up to 10-fold, without significant impact on the phloem metabolites. Phloem sugars related to temperature fluctuations rather than soil moisture and did not relate closely to A(net) levels. Sugars and proteins decreased between the second and third sampling date by 56 and 61%, respectively, and the ratio of sugars to amino acids decreased between the first and third sampling by 81%, while A(net) and water-use efficiency (A(net)/E) decreased only in the older seedlings. Although gas exchange was higher in the older seedlings, ascorbic acid and phloem metabolites were higher in the younger seedlings. It was concluded that the drought stress responses depended significantly on developmental stage, and research on the physiology of Aleppo pine regeneration should focus more on temperature conditions and the duration of drought than its severity.
Seasonal Responses of Hydraulic Function and Carbon Dynamics in Spruce Seedlings to Continuous Drought

Abstract:Drought is expected to increase in the frequency and duration associated with climate change. Although hydraulic function and carbon (C) storage have been widely recognized as key components to plant survival under a single drought, the physiological responses to continuous drought remain largely unknown, particularly for high northern temperate and boreal forests which are sensitive to water stress. In this study, we quantified the survival, growth, gas exchange, water relations, and nonstructural carbohydrates (NSCs) in 3-year-old Jezo spruce ( Picea jezoensis ) seedlings responding to continuous drought stress. Seedlings were maintained in drought conditions for 392 days, covering two growing and one dormant winter season. Seedlings subjected to drought showed a significant decrease in net photosynthesis rate ( A net ) and stomatal conductance ( g s ) in both growing seasons, and biomass in the second growing season. The seedling mortality continuously increased to 35.6% at the experimental end. Notably, responses of C storage and leaf water potential to drought varied greatly depending on seasons. Living seedlings exposed to drought and control treatments had similar NSC concentrations in both growing seasons. However, seedlings with concentrations of both the soluble sugars and starch less than 1% in root died in the winter dormant season. In the second growing season, compared with the control treatment, droughted seedlings had significantly lower leaf water potential and stem wood-specific hydraulic conductivity ( K w ). Meanwhile, the leaf predawn water potential did not recover overnight. These suggest that C starvation might be an important reason for seedlings that died in the winter dormant season, while in the growing season drought may limit seedling survival and growth through inducing hydraulic failure. Such seasonal dependence in hydraulic dysfunction and C depletion may lead to higher mortality in spruce forests facing extended drought duration expected in the future.

Abstract:Under drought stress, temperate forest understorey forbs prioritize biomass and leaf area retention, while graminoids enhance water‐use efficiency and photoprotection for survival. Drought stress can profoundly affect plant growth and physiological vitality, yet there is a notable scarcity of controlled drought experiments focused on herbaceous species of the forest understorey. In this study, we collected seeds from five forb and four graminoid species common in European temperate forests. Seeds were germinated under controlled glasshouse conditions and subjected to moderate drought stress for 5 weeks. We assessed biomass partitioning, stomatal and leaf morphology, leaf gas exchange, minimum leaf conductance (gmin), and chlorophyll fluorescence parameters. Comparison of the two ecological guilds revealed that graminoids had a higher R/S, improved WUE, greater carboxylation efficiency, and enhanced non‐photochemical quenching under drought conditions compared to forbs. In contrast, forbs had significantly lower gmin, with higher total biomass and total leaf area. Despite these differences in morpho‐physiological functional traits, both groups experienced a similar relative reduction in biomass after drought stress. Key predictors of biomass accumulation under drought included photochemical quenching, stomatal traits, total leaf area and gmin. A negative correlation between biomass and gmin suggests that plants with lower residual water loss after stomatal closure can accumulate more biomass under drought stress. Additionally, gmin was positively correlated with guard cell length, suggesting that larger stomata contribute to higher residual water loss. Contrasting strategies in morpho‐physiological responses to drought define the differences between the two groups. In graminoids, drought resistance suggests greater emphasis on stress tolerance as a survival strategy. In contrast, forbs were able to maintain higher biomass and total leaf area, indicating a competitive strategy for maximizing resource acquisition.

Abstract:In recent years, global forests have been facing an increase in tree mortality owing to increasing droughts. However, the capacity for plants to adjust their physiology and biochemistry during extreme drought and subsequent recovery is still unclear. Here, we used 1.5-year-old Pinus massoniana Lamb. seedlings and simulated drought conditions to achieve three target stress levels (50%, 85%, and 100% loss of stem hydraulic conductivity (PLC)), followed by rehydration. Needle water status, gas exchange, and biochemical parameters were assessed during drought and recovery. The results showed that drought had significantly negative impacts on needle water status and gas exchange parameters, with gas exchange declining to 0 after PLC85 was achieved. Soluble protein concentration (SPC), soluble sugar concentration (SSC), malondialdehyde (MDA) content, superoxide dismutase (SOD) activity, and needle water-use efficiency showed fluctuations. The activity of antioxidant enzymes and the values of osmotic regulators were then gradually decreased as the physiological and biochemical functions of seedlings were disturbed. Seedlings showed a stronger ability to recover from PLC50 than PLC85 and PLC100. We conclude that the physiological and biochemical recovery of P. massoniana seedlings is more likely to be inhibited when plants experience increasing drought stress that induces 85% and greater loss of hydraulic conductance.
