Abstract:Secondary amines are oxidized by the methyltrioxorhenium/hydrogen peroxide system to the corresponding nitrones in excellent yield. The results provide a further example of the parallel between the chemistry of this metal system and that of the dioxiranes.

Abstract:Nitrocylcohexadienones have been applied as nitration reagents for mild, mono-nitrating reactions. The original synthesis of 2,3,5,6-tetrabromo-4-methyl-4-nitrocylcohexa-2,5-dien-1-one appeared to be difficult to pursue due to both the solvent system and reaction conditions. Therefore, we applied a modified solvent system and optimized the reaction conditions to prepare the dienone at 0°C, eventually overcome the difficulties.

Abstract:Owing to their distinctive 1,3-dipolar structure, the catalytic asymmetric hydrogenation of nitrones to hydroxylamines has been a formidable and longstanding challenge, characterized by intricate enantiocontrol and susceptibility to N-O bond cleavage. In this study, the asymmetric hydrogenation and transfer hydrogenation of nitrones were accomplished with a tethered TsDPEN-derived cyclopentadienyl rhodium(III) catalyst (TsDPEN: p-toluenesulfonyl-1,2-diphenylethylene-1,2-diamine), the reaction proceeds via a novel 7-membered cyclic transition state, producing chiral hydroxylamines with up to 99% yield and >99% ee. The practical viability of this methodology was underscored by gram-scale catalytic reactions and subsequent transformations. Furthermore, mechanistic investigations and DFT calculations were also conducted to elucidate the origin of enantioselectivity.

Abstract:Azo compounds (RN = NR') are an important class of organic molecules that find wide application in organic synthesis. Herein, we report an efficient, practical and metal-free oxidation of hydrazines (RNH-NHR') to azo compounds using 5 mol% HBr and hydrogen peroxide as terminal oxidant. This new method has been demonstrated by 40 examples with excellent yields. In addition, we showcased two examples of the one-pot sequential reactions involving our hydrazine oxidation/hydrolysis/Heck reaction or Cu-catalyzed N-arylation with aryl boronic acid. The role of the proton-responsive entity on the ligand scaffold is demonstrated by control experiments, including the use of a N-methylated pyrazole analogue. Abstract:An efficient and highly selective method for the oxidative conversion of primary amines to the corresponding nitriles using trichloroisocyanuric acid in the presence of catalytic TEMPO under mild reaction conditions is described. Other functional groups such as C,C-double bonds, benzyloxy etc. were found to be unaffected under the reaction conditions. This procedure provides a new entry to the synthesis of various aliphatic, aromatic and heterocyclic nitriles in excellent yield.

Abstract:A silver-assisted method for [3 + 2] annulation of nitrones with isocyanides has been developed. The developed protocol allows access to a variety of 2,3,4-trisubstituted 1,2,4-oxadiazolidin-5-one derivatives as single diastereomers in good to excellent yields using silver oxide as the catalyst and molecular oxygen as the terminal oxidant. A plausible mechanism involving a nucleophilic addition/cyclization/protodeargentation/oxidation pathway is proposed on the basis of experimental results.

Abstract:The oxidation of amines based on nitrogen-rich heterocycles is one of the appealing alternatives for promising nitro derivatives. Herein, we present a continuous-flow process to achieve oxidation of these amines to their nitro derivatives, taking advantage of hydrogen peroxide as an environmentally benign and inexpensive oxidant. Facile and sustainable access to valuable nitro compounds was ensured. The utility of the present continuous-flow oxidation process was demonstrated by synthesizing promising energetic compounds 5-amino-3-nitro-1,2,4-triazole (ANTA) and 1-methyl-3,4,5-trinitropyrazole (MTNP) on a multigram scale.

Abstract:We herein report a method for the direct a-amination of different carbonyl compounds by using aqueous ammonium as the N-source. Upon using ammonia in combination with hypochlorites as simple oxidants under phase-transfer conditions it is possible to carry out the direct oxidative a-amination of cyclic b-ketoesters, oxindoles as well as malonitriles and malonates with good to excellent yields. Furthermore, a first proof-of-concept for an asymmetric variant by employing chiral ammonium salts was obtained.

Abstract:Abstract Cleaner synthesis of amines remains a key challenge in organic chemistry because of their prevalence in pharmaceuticals, agrochemicals and synthetic building blocks. Here, we report a different paradigm for chemoselective hydrogenation of nitro compounds to amines, under mild, aqueous conditions. The hydrogenase enzyme releases electrons from H 2 to a carbon black support which facilitates nitro-group reduction. For 30 nitroarenes we demonstrate full conversion (isolated yields 78 – 96%), with products including pharmaceuticals benzocaine, procainamide and mesalazine, and 4-aminophenol – precursor to paracetamol (acetaminophen). We also showcase gram-scale synthesis of procainamide with 90% isolated yield. We demonstrate potential for extension to aliphatic substrates. The catalyst is highly selective for reduction of the nitro group over other unsaturated bonds, tolerant to a wide range of functional groups, and exhibits excellent stability in reactions lasting up to 72 hours and full reusability over 5 cycles with a total turnover number over 1 million, indicating scope for direct translation to fine chemical manufacturing.

Abstract:The efficient production of aliphatic primary amines is still a major challenge despite their production on a large scale. Particularly when considering the overall production route starting from alkenes, current strategies suffer in at least one reaction step from poor regio- or chemoselectivity. This work presents an efficient and selective synthesis protocol for primary aliphatic amines via their corresponding aldoximes. These are readily produced from the condensation of hydroxylamine and the respective aldehydes. This straightforward condensation can be carried out either with isolated aldehydes or with crude reaction solutions from the hydroformylation of alkenes. It allows a straightforward separation of the aldoximes via their precipitation, which then serve as intermediates for the final reduction. In a newly developed protocol for aldoxime reduction, yields of up to 90 % of the desired primary amines from several different aldoximes are achieved. The ruthenium/triphos catalyst system showed activities exceeding 7500 h-1, which is significantly faster than other amination protocols, and complete conversion is achieved within minutes. Our approach allows the synthesis of the polyamide-12 precursor (methyl 12 aminododecanoate) from the unsaturated, renewable oleochemical methyl 10 undecenoate via the hydroformylation, condensation, reduction sequence on up to 6 g scale with up to 68 % overall selectivity.

Abstract:Comprehensive Summary In this paper, we have developed a decarboxylative amination of carboxylic acids with nitroarenes for the synthesis of secondary amines. The protocol is performed at mild conditions without the use of noble metals as catalysts. A wide range structurally diverse secondary amines could be obtained in good yields (up to 94%) with good functional group tolerance. Abstract:A mild and highly efficient access to N-sulfonyl imines through the oxidation of corresponding N-tert-butylsulfinyl imines and N-p-tosyl-sulfinyl imines with UHP in the presence of catalytic amount MTO was developed. Under mild reaction conditions, this highly selective reaction, provides a general and efficient access to N-sulfonyl aldimines, ketimines and alpha-ketiminoesters in excellent yield. The unstable sulfonylimine intermediates can be afforded with high purity and ready for subsequent reaction without further purification.
