Abstract:The interaction between vascular cell adhesion molecule‐1 (VCAM‐1) and very late antigen‐4 (VLA‐4) is known to play an important role in stabilizing the adhesion of lymphocytes to endothelial cells. Such cellular adhesion is crucial to many immunological processes including lymphocyte‐mediated cell lysis. In this study the expression of VCAM‐I on renal tubular epithelial cells is demonstrated on biopsy sections recovered during acute renal allograft rejection. Experiments performed using epithelial cells cultured from renal tubules show that VCAM‐1 is up‐regulated by addition of the inflammatory cytokines tumour necrosis factor‐alpha (TNF‐α) and interferon‐gamma (IFN‐γ). Combination of TNF‐α and IFN‐γ synergized to induce high levels of VCAM‐1 expression. Further experiments demonstrated that the cytokines produced by activated lymphocytes in mixed leucocyte culture also up‐regulate expression of VCAM‐1. Assays of the adhesion of lymphoid cells to cultured renal epithelial cells showed that cytokine pretreatment of the renal cells enhanced the binding of lymphoid cells. The proportion of bound lymphoid cells was significantly reduced by addition of an antibody capable of blocking the interaction of VCAM‐1 with VLA‐4. This result indicated that the VCAM‐1 induced on renal epithelial cells by inflammatory cytokines is functionally capable of binding VLA‐4, thereby enhancing the adhesion of potentially graft‐damaging lymphoid cells.

Finding a balance: Reproductive trade-offs in parasitic isopods of the family Cymothoidae

Abstract:In this study conducted along the coast of Odisha, India, reproductive trade-offs in isopods of the family Cymothoidae were investigated, focusing on the relationship between the number vs. volume of eggs and the percentage of brood to body volume. Ovigerous females from species with substantial sample sizes ( n > 30) were analyzed to understand their survival strategies. The findings highlight distinct strategies among different parasite types within Cymothoidae. Buccal parasites such as Cymothoa frontalis and Cymothoa indica , along with branchial parasites like Joryma sawayah and Agarna malayi , generally produced a higher number of smaller eggs, except for A . malayi , which produced fewer but larger eggs. In contrast, the external body surface-attaching parasite, Nerocila orbignyi , was observed to produce fewer eggs of larger size. Across all parasite types (intraspecific), there was a statistically significant negative correlation between egg number and egg volume ( p < 0.05). Buccal parasites were observed to have the highest percentage of marsupium volume relative to body volume, indicating a strategy to maximize egg storage. Conversely, external body surface-attaching parasites had the lowest percentage, possibly reflecting adaptation to a different reproductive environment. Additional observations revealed anatomical adaptations in all the studied isopod species which include the shift of the alimentary canal to the dorsal side to accommodate the development of the marsupial sac for egg incubation. In summary, these findings illustrate how parasitic isopods of the family Cymothoidae manage trade-offs between reproductive potential and success, crucial for their species' continuity and survival strategies in coastal ecosystems.

The parasites Vallisia indica, Allodiscocotyla chorinemi, Heterapta chorinemi, and Dionchus remorae exhibited site specificity on the gills of Scomberoides commersonianus, S. tol, S. lysan and S. tala, whereas the copepod Caligus sp. did not. Observations on site preference revealed microhabitat differences along the length of gill filaments, along the anterio-posterior axis of the gill, between external and internal gill filaments and on different gill arches. Site preference varied with parasite density on each gill and host. An interspecific association test between pairs of species revealed, in some cases, a positive association, and in other cases a negative association. There were apparently no association? between certain pairs of species. A comparison of intensity within pairs of parasite species infecting the same host revealed either an inverse or a direct correlation. The numerical dominance and prevalence of parasites differed on each host species. This study indicates that intra- and interspecific competition may occur among the gill parasites. The direction and speed of ventilation water-currents and certain intrinsic factors of the parasite themselves may determine their microhabitat restriction on the gills.

Abstract:The present study reports the parasitism of cymothoid isopod Renocila bijui Aneesh & Bruce, 2020 from two host fish species, Chrysiptera unimaculata (Cuvier, 1830) and Acanthurus xanthopterus Valenciennes, 1835; for the first time. It was also observed and collected from its originally described host species, A. triostegus (Linnaeus, 1758) during the study period, confirming its host-specificity to the acanthurid fish group. The observation of R. bijui from C. unimaculata , a pomacentrid fish, indicates its preference for another coral-reef associated fish species as host. Further studies may give more insight into the distribution and behaviour of this isopod species in Indian waters.

Abstract:In the present study, the fish parasitic Cymothoid, Nerocila phaiopleura Bleeker, 1857 attached with a Baitfish, Spratelloides delicatulus (Bennett, 1832) is reported for the first time from Lakshadweep Islands. Collected parasites were confirmed as N. phaiopleura based on morphological diagnosis. In total, eleven numbers of N. phaiopleura were collected from the anterior part of the individuals of S. delicatulus . Smaller size lesions in the body of host fish were also noticed. The report of this parasitic isopod from Lakshadweep Islands provides evidence on its geographic range extension and occurrence of the parasite, N. phaiopleura with a baitfish species, S. delicatulus , which is a new host.

Abstract:Cymothoidae are parasitic isopods that infest fishes inhabiting marine, brackish, and fresh-water environments. The systematics, distribution, and host specificity of cymothoids is often unknown. In this study, morphological observations and molecular analysis using the COI and 16S rRNA genes were conducted on a juvenile individual of Cymothoa indica infesting the wide-banded hardyhead silverside, Atherinomorus lacunosus, and an ovigerous female of Mothocya collettei infesting the hound needlefish, Tylosurus crocodilus, both collected from Okinawa Island, Japan. This is the first time that these two species of cymothoid have been collected in the Ryukyu Islands, and the juvenile C. indica in particular represents a new northern range limit for its species. That specimen was identified using molecular analysis and showed almost identical morphology to previously reported juveniles, but it differed in the numbers of robust setae on pereopods 6 and 7. The ovigerous female of M. collettei showed a high morphological similarity to M. collettei but had some morphological traits consistent with Mothocya affinis. DNA analysis indicated that these two nominal species of Mothocya are very closely related, and morphological and molecular studies using larger populations of both are needed.

Abstract:A morphological description and molecular characterisation of the parasitic isopod Cymothoa eremita Brünnich, 1783, collected off the fish host Nemipterus furcosus (Valenciennes, 1830) from the South China Sea, is presented here. Since most cymothoid descriptions are primarily based on females, this paper highlights the morphological characteristics from 20 ovigerous female and 20 male C. eremita cymothoids, isolated from 300 fish individuals. Furthermore, our findings indicated significant variations in the morphological features of C. eremita , including: 1) average length and width dimensions, 2) the presence or absence of eyes, 3) the length of pleotelson, 4) the configuration of its posterior and lateral margins, and 5) the structure of the cephalon. These characteristics were compared among diverse fish host species reported in different geographical regions. Twenty one of 300 N. furcosus were infested with C. eremita, with a prevalence of 7%, and mean intensity of 1.9 (two cymothoids per individual fish) respectively. A total of 687 base pairs (bp) from the mitochondrial cytochrome oxidase subunit I (COI) gene were sequenced for C. eremita as a complementary measure to the morphological analysis undertaken in this study. These sequences were utilized to reconstruct a phylogenetic tree, incorporating our sequenced specimens and relevant sequences sourced from the GenBank database. The Maximum Likelihood (ML) phylogenetic tree comprised 45 cymothoid species, with partial COI gene sequences showing monophyletic clustering of most species within the same genus, which is in congruence with their morphological divergence. Continued efforts to conduct studies on the biology, taxonomy, ecology and genetics of both cymothoid and fish host are crucial to better understand the patterns of host-parasite specificity and co-evolutionary changes, which may indirectly contribute towards cymothoid speciation and diversification.

Prevalence, site and tissue preference of myxozoan parasites infecting gills of cultured fish in Punjab (India)

Abstract:Native carp species cultured in Indian farms in Punjab (catla Catla catla, rohu Labeo rohita, mrigal Cirrhinus mrigala, exotic carps such as silver carp Hypophthalmichthys molitrix, grass carp Ctenopharyngodon idella, common carp Cyprinus carpio and a catfish Sperata seenghala) were examined for the presence of myxozoan parasites infecting gills. Firstly, the gills were examined under a zoom-stereomicroscope for the presence of plasmodia. The number of plasmodia per gill was counted to determine the index for the intensity of infection. Infected tissues were processed for histology, and 3-4 µm sections of infected gills were stained with haematoxylin & eosin and Luna's method. A total of 19 species of myxosporean were found infecting various cell types in the gills. Of these, 14 species belonged to the genus Myxobolus, 3 species to the genus Thelohanellus and 2 species to the genus Henneguya. Species belonging to the genus Myxobolus formed the interlamellar and intralamellar vascular (LV) type plasmodia, and species belonging to the genus Thelohanellus and Henneguya formed intrafilamental vascular (FV) type plasmodia. Mixed infections comprising 2, 3 or 4 different myxozoan species were noted in individual fish. The most common type of parasitism was polyparasitism due to 4 myxobolids co-occuring in fish with an infection rate of 23.16%. All species caused mild to severe haemorrhagic gill disease with little clinical symptomatology.

Parasitic copepods affect morphospace and diet of larvae of a temperate reef fish

Abstract:The effects of ectoparasites on larvae of the clingfish Gobiesox marmoratus were evaluated at the dietary and morphometric levels. The larvae and ectoparasites were collected by nearshore plankton samplings during October, November and December 2013 off El Quisco Bay, central Chile. The standardized abundance of total larvae and those ectoparasitized larvae (PL) was positively related and high parasite prevalence was found throughout the sampling period (up to 38%). Geometric morphometrics analyses indicate main changes in the shape through early ontogeny and subtle but significant variations between PL and non-parasitized larvae (NPL). Prey composition varied between PL and NPL; small size (<6 mm standard length, LS ) parasitized larval G. marmoratus ate mostly gastropod larvae, whereas small non-parasitized specimens ate mainly cirripede nauplii. All larger (>8 mm LS ), pre-settlement stages parasitized by Trifur and, or Caligus copepods had content in their gut, suggesting that ectoparasites did not diminish prey capture in host with larger size. Morphometric and dietary changes occurring during larval development were decoupled, both for PL and NPL. The maintenance of a slender, more hydrodynamic body through pelagic development and the ingestion of less-mobile prey in PL suggests non-lethal effects of ectoparasitism on rocky-reef fish larvae.
Microhabitat distributions and species interactions of ectoparasites on the gills of cichlid fish in Lake Victoria, Tanzania

Abstract:Heterogeneous exposure to parasites may contribute to host species differentiation. Hosts often harbour multiple parasite species which may interact and thus modify each other's effects on host fitness. Antagonistic or synergistic interactions between parasites may be detectable as niche segregation within hosts. Consequently, the within-host distribution of different parasite taxa may constitute an important axis of infection variation among host populations and species. We investigated the microhabitat distributions and species interactions of gill parasites (four genera) infecting 14 sympatric cichlid species in Lake Victoria, Tanzania. We found that the two most abundant ectoparasite genera (the monogenean Cichlidogyrus spp. and the copepod Lamproglena monodi) were non-randomly distributed across the host gills and their spatial distribution differed between host species. This may indicate microhabitat selection by the parasites and cryptic differences in the host-parasite interaction among host species. Relationships among ectoparasite genera were synergistic: the abundances of Cichlidogyrus spp. and the copepods L. monodi and Ergasilus lamellifer tended to be positively correlated. In contrast, relationships among morphospecies of Cichlidogyrus were antagonistic: the abundances of morphospecies were negatively correlated. Together with niche overlap, this suggests competition among morphospecies of Cichlidogyrus. We also assessed the reproductive activity of the copepod species (the proportion of individuals carrying egg clutches), as it may be affected by the presence of other parasites and provide another indicator of the species specificity of the host-parasite relationship. Copepod reproductive activity did not differ between host species and was not associated with the presence or abundance of other parasites, suggesting that these are generalist parasites, thriving in all cichlid species examined from Lake Victoria.
Habitat shift of adult Caligus undulatus (Copepoda: Siphonostomatoida: Caligidae) from host fish to plankton in response to host behavior

Abstract:Before 2019, adults of the sea louse Caligus undulatus were reported exclusively in plankton from ocean samples worldwide and were not known to parasitize fish hosts. In 2019, the first instance of this caligid parasitizing a fish host, Japanese sardinella Sardinella zunasi, was reported in the Seto Inland Sea, Japan. The presently reported study aimed to investigate the biology and ecology of adult C. undulatus in plankton communities in the Seto Inland Sea and surrounding waters from March 2020 to November 2021. The occurrence of sea lice in plankton communities was restricted to the period of August-January, mainly between October and December with maximum plankton abundance (10.5 ind. per 1000 m3) recorded on 30 November 2020. All post-naupliar stages of C. undulatus were found on the host fish, and they represented a typical life cycle pattern known for Caligus species. The sex ratios in both planktonic and parasitic adults were not significantly different. The frequency of occurrence of planktonic and parasitic adult females with egg strings was 68 and 46%, respectively. The number of eggs per string was significantly higher in parasitic adult females (mean ± SD: 16.9 ± 8.6) than in planktonic females (10.4 ± 10.8). These data suggest that adult females were detached from their hosts and continued to produce eggs without feeding. Seasonal migration of S. zunasi to brackish water for spawning may result in the detachment of mature caligids from the host and may be effective in protecting the offspring, which are less tolerant of less brackish water.
Suitability of invasive gobies as paratenic hosts for acanthocephalans of the genus Pomphorhynchus sp.

Abstract:Ponto-Caspian gobies became highly abundant in many regions outside their native distribution range (e.g. in the Rhine River system). In the newly invaded habitats, the parasite communities of the invasive gobies are characterized by a lower species richness compared to their native range. Interestingly, acanthocephalans of the genus Pomphorhynchus are highly abundant, although they do not become mature and mostly remain encapsulated in the abdominal cavity as preadults. Thus, gobiids could either represent a dead-end host for Pomphorhynchus sp. declining its population (dilution effect) or act as a paratenic host that could increase the infection pressure if the infected gobies are preyed upon by appropriate definitive hosts (spill back). To determine which of the 2 scenarios the gobiids contribute to, we conducted 2 infection experiments using smaller and larger individuals of the definitive host chub (Squalius cephalus), infected with preadults of Pomphorhynchus sp. collected from the abdominal cavity of Neogobius melanostomus. The results showed that preadults were able to complete their development and mature in the definitive host with mean recovery rates of 17.9% in smaller and 27.0% in larger chubs. Successful infections were observed in 62.0% and 80.0% of the smaller and larger chubs, respectively. Our study demonstrated that gobies can theoretically serve as a paratenic host for acanthocephalans of the genus Pomphorhynchus, and that infection might spill back into the local fish community if infected gobies are preyed upon by suitable definitive hosts of Pomphorhynchus sp. such as large barbel or chub.

Functional diversity among coral reef fishes as consumers of ectoparasites

Abstract:Nearly all organisms must cope with parasitic infestation, and most research on parasite ecology in marine systems has focused on the variety of both direct and indirect impacts that parasites can have on hosts. In coral reef ecosystems, gnathiid isopods are one of the most common ectoparasites of fishes. For individuals infested with gnathiids, there is a heavy reliance on intraspecific mutualisms where specialist species groom or "clean" parasites off host fishes. However, gnathiids spend most of their time free-living and not associated with a host and are thus susceptible to consumption by non-cleaner fish species. Here, we investigated if non-cleaner and facultative (less specialized) cleaner fish consume ectoparasitic gnathiid isopods as often or in similar quantities as dedicated (highly specialized) cleaners. To do so, we sampled the gut contents of 797 fishes, representing 61 species and including both cleaner and non-cleaner fishes, to compare their consumption of gnathiid isopods. We found that although there is some site level variation, cleaner fishes eat significantly more gnathiids, and eat them more frequently, compared to non-cleaner fishes. Our results highlight the importance of both dedicated and facultative cleaners as consumers of ectoparasites and show that their role is unlikely to be supplanted by non-cleaner consumers. Furthermore, we suggest that different cleaner species act as complementary rather than redundant specialists.

Abstract:The copepod family Shiinoidae Cressey, 1975 currently comprises nine species of teleost parasites with unusual morphology and a unique attachment mechanism. Female shiinoids possess greatly enlarged antennae that oppose a rostrum, an elongate outgrowth of cuticle that originates between the antennules. The antennae form a moveable clasp against the rostrum which they use to attach to their host. In this study, we use micro-computed tomography (microCT) to examine specimens of Shiinoa inauris Cressey, 1975 in situ attached to host tissue in order to characterize the functional morphology and specific muscles involved in this novel mode of attachment and to resolve uncertainty regarding the segmental composition of the regions of the body. We review the host and locality data for all reports of shiinoids, revise the generic diagnoses for both constituent genera Shiinoa Kabata, 1968 and Parashiinoa West, 1986, transfer Shiinoa rostrata Balaraman, Prabha & Pillai, 1984 to Parashiinoa as Parashiinoa rostrata (Balaraman, Prabha & Pillai, 1984) n. comb., and present keys to the females and males of both genera.

Abstract:In recent years there have been several accounts of the occurrence of diclidophoroidean trematodes parasitic on the gills of fishes, e.g. Price (1943), Sproston (1946), Dawes (1947), Brinkmann (1952), and Chauhan (1953), and these accounts have included descriptions of the morphology of the parasites. The distribution records have revealed a generally high degree of host specificity and, in some species, a preference even for certain gill arches of the particular hosts, while the morphological descriptions have shown that there is a considerable variation in the form of the parasites, extending to various degrees of deviation from bilateral symmetry (PI. I, figs. 1–11). These morpho-logical variations are present in spite of the fact that the different parasites occupy such broadly similar habitats in the gill chambers of their respective hosts. But, as far as I am aware, this is the first attempt to investigate the distribution and morphology of the parasites in relation to their micro-habitats.

Abstract:Parasites are widely known to afflict nearly all animal groups, including cephalopods, which host a diverse range of endo– and ectoparasites. These parasites, from protozoans to metazoans, can exert either detrimental or symbiotic effects on their cephalopod hosts. Within cephalopods lies a hidden biodiversity of parasites, including viruses, bacteria, fungi, and other taxa such as Euglenozoa, Myzozoa, Microsporidia, Ciliophora, Dicyemids, Myxozoa, Trematoda, Cestoda, Nematoda, Acanthocephala, Annelida, and Crustacea. Among these, cestodes are the most prevalent group, often infesting a wide range of cephalopods, followed by dicyemids, which target renal organs. This distribution pattern observed globally shows Mediterranean Sea exhibiting the highest number of cases in terms of host–parasite interactions. Although global attention is given to cephalopod parasites, research on their presence and distribution in Indian waters remains limited, despite the rich marine ecosystems they inhabit. Most studies in this regard were conducted decades ago, with no recent updates on parasite progress. Abstract:Myxozoans are highly specialized metazoan parasites with a very diverse host range that live on aquatic hosts. In both wild and farm-raised fish, myxozoans are the most frequent parasites. The aim of the current study was to evaluate any myxozoan parasite found in different organs of Schizothorax richardsonii . Live fish were transported with artificial aeration to the University of Kashmir. Thirty-five fish specimens were examined during the study period, and only gills of 6 fish were determined to be infected. Taxonomic study was carried out with the help of different keys, and histopathologcal study was assayed in order to study the diseased organs of the fish. Besides, characterization of partial 18S rDNA gene was also done in the present study as per the standard procedures. The findings showed that the plasmodia were big, oval shaped, and white in color, measuring 1.5 × 2.0 mm. They were referred to as intralamellar vascular-type plasmodia (LV 3 ). Histological changes in the gills included inflammation, gill epithelium lifting, hypertrophy of lamellar cells, and fusion of nearby gill lamellae. The myxospore body had a tiny ICP at the anterior end and was spherical to ovoidal in form, measuring 11.19 × 7.66 μm. Polar tubules were pyriform in shape and measuring 6.12 × 1.95 μm, having coils up to 7 in number. The gill plasmodium index showed a moderate level of infection (GPI 2). According to their phylogenetic relationships, M. richardsonii n. sp. was sister species to Myxobolus sp. KLT and Myxobolus dermiscalis –infecting scales of Labeo rohita from Myanmar and India, respectively. The novel sequence KU885918 and 30 additional sequences from closely related sequences showed 97% homogeneity or above in NCBI GenBank database. Moreover, the tree showed grouping based on the tissue tropism, host, and geographical location of the infected host. Morphological, histological, and molecular characterizations were used to identify a novel myxosporean parasite, Myxobolus richardsonii n. sp. in cold water fish, Schizothorax richardsonii (Heckel), obtained from Wullar Lake in Kashmir Himalayas, India.

Abstract:Different groups parasitize the gills of fishes, and the organisms can interact in positive, negative, or neutral ways, depending on parasitic abundance, environmental conditions, and the availability of resources. Here, we studied the distribution of ectoparasites on the gills of the mullet Mugil curema in the Cabedelo port region (Brazil), asking if (1) the gill parasites of M. curema fix on specific host gill arches and/or (2) co-occur in the same gill arches within a host individual; (3) if the abundance of one parasite taxon affects the abundance of another on the same gill arches; and (4) considering distinct major groups, if parasites present patterns of co-occurrence on hosts and gill arches different from the expected at random. We used generalized linear models, generalized linear mixed models, and models with zero inflation to define the microhabitats (arches) with the highest abundances of parasites and null models to analyze the co-occurrences of gill parasite groups. Three taxa of Monogenea (Ligophorus brasiliensis Abdallah, Azevedo & Luque, 2009, Ligophorus sp. 1 and Ligophorus sp. 2) and six species of Copepoda were recorded (Bomolochus nitidus Wilson, 1911; Caligus praetextus Bere, 1936; Ergasilus atafonensis Amado & Rocha, 1997; Ergasilus bahiensis Amado & Rocha, 1997; Ergasilus caraguatatubensis Amado & Rocha, 1997 and Ergasilus lizae Krøyer, 1863). We found that, in general, the gill parasites were more abundant/prevalent on specific arches (I and II) of the hosts, shared such preferences and co-occurred on the gill arches. The presence of one taxon influenced the occurrence of another only in the arches I and II; as a result, the infracommunities were characterized as interactive in the gills of this host species.
MM 3 Potential energy surfaces of a-3-linked L-fucobiose and fucotriose and their sulfated counterparts q

Abstract:The adiabatic potential energy surfaces (PES) of a-L-Fuc-(1 ! 3)-a-L-Fuc and their counterparts disulfated at 2,2 0 and 4,4 0, and tetrasulfated at 2,2 0,4,4 0, which are representative of fucoidan structures, were obtained using the MM3 force field, and plotted as contour maps and as 2D graphs representing the energy versus the w angle. The surfaces of the corresponding trisaccharides were also obtained and represented by a single 3D contour map for which the energy is plotted against the two w glycosidic angles. For the nonsulfated disaccharide, similar populations of two minima occur. A substantial sulfate effect is observed. Whereas sulfation on both of the 2-positions shift the global minimum to positive w angles, sulfation on both of the 4-positions deepen the well at negative w values. A similar effect occurred in their galactose counterparts. Sulfation on the 2and 4-positions carry the additive effect of both groups. The same trend was observed for both linkages present in the trisaccharides, with minor differences. For instance, the 4,4 0,400 trisulfated compound exhibits a trend by which the glycosidic linkage closer to the nonreducing end appears to be highly flexible, with similar energies in both conformers. Raising the dielectric constant on nonsulfated oligosaccharides was found to give a better agreement with experimental determinations. 2004 Elsevier Ltd. All rights reserved.
