Spatio-temporal plasticity of gill microbiota in estuarine fish

Abstract:Coastal marine and estuarine systems are subject to enormous endogenous and exogenous pressures, particularly climate change, while at the same time being highly productive sources and nurseries for fish populations. Interactions between host and microbiome are increasingly recognized for their importance for fish health, with growing evidence indicating that increasing environmental pressures impact host resilience and favor the raise of opportunistic bacterial taxa. The microbial composition of the gill mucus reflects environmental conditions and represents an entry route for pathogens into the fish body. High-throughput sequencing of prokaryotic populations from 250 samples of two fish species with highly different habitat preferences, as well as seasonal and spatial distributions in the Elbe estuary system, allowed us to describe the variation of the microbiota along a salinity gradient and under fluctuating environmental conditions. The analysis of estuarine fish core microbiota in relation to variable bacterial components indicated dysbiotic states under sustained hypoxia and high nutrient loads largely driven by increased prevalence of facultatively aerobic ( Acinetobacter ) and anaerobic heterotrophs ( Shewanella , Aeromonas ). By correlating bacterial abundances with environmental and physiological parameters in a co-occurrence network approach, we describe plasticity in microbiota composition, identify potential biomarkers for fish health monitoring and reconstruct movement patterns of the fish. Our results can help to shape future minimal-invasive and cost-effective monitoring programs, and identify factors that need to be controlled in the estuary to promote fish and stock health.

Impacts of temperature and turbidity on the gill physiology of darter species

Abstract:Fish gills are complex organs that have direct contact with the environment performing numerous functions, including gas exchange and ion regulation. Determining if gill morphometry can change under different environmental conditions to maintain and/or improve gas exchange and ion regulation is important for understanding if gill plasticity can improve survival with increasing environmental change. We assessed gill morphology (gas exchange and ion regulation metrics), hematocrit and gill Na + /K + ATPase activity of wild-captured blackside darter ( Percina maculata ), greenside darter ( Etheostoma blennioides ), and johnny darter ( Etheostoma nigrum ) at two temperatures (10 and 25 °C) and turbidity levels (8 and 94 NTU). Samples were collected August and October 2020 in the Grand River to assess temperature differences, and August 2020 in the Thames River to assess turbidity differences. Significant effects of temperature and/or turbidity only impacted ionocyte number, lamellae width, and hematocrit. An increase in temperature decreased ionocyte number while an increase in turbidity increased lamellae width. Hematocrit had a species-specific response for both temperature and turbidity. Findings suggest that the three darter species have limited plasticity in gill morphology, with no observed compensatory changes in hematocrit or Na + /K + ATPase activity to maintain homeostasis under the different environmental conditions.

Abstract:In thermally variable ecosystems, temperatures can change extensively on hourly and seasonal timescales requiring ectotherms to possess a broad thermal tolerance (critical thermal minima [CTmin] and maxima [CTmax]). However, whether fish acclimate in the laboratory similarly as they acclimatize in the field under comparable thermal variation is unclear. We used temperature data from a tidal salt marsh to design 21-day lab-acclimation treatments (static: 12, 17, 22, 27 °C; daily variation with mean 22 °C: i) range 17-27 °C, ii) range 17-27 °C with irregular extremes within 12-32 °C). We compared thermal limits in lab-acclimated and field-acclimatized eurythermal arrow goby ( Clevelandia ios ). Variable temperature-acclimated and acclimatized fish had similar CTmin and CTmax. Notably, arrow gobies showed rapid plasticity in their absolute thermal tolerance within one tidal cycle. The daily mean and max temperatures experienced were positively related to CTmax and CTmin, respectively. This study demonstrates that ecologically informed lab acclimation treatments can yield tolerance results that are applicable to wild fish.

Abstract:The fourfinger threadfin fish (Eleutheronema tetradactylum) is an economically significant species renowned for its ability to adapt to varying salinity environments, with gills serving as their primary organs for osmoregulation and immune defense. Previous studies focused on tissue and morphological levels, whereas ignored the cellular heterogeneity and the crucial gene information related to core cell subsets within E. tetradactylum gills. In this study, we utilized high-throughput single-cell RNA sequencing (scRNA-seq) to analyze the gills of E. tetradactylum, characterizing 16 distinct cell types and identifying unique gene markers and enriched functions associated within each cell type. Additionally, we subdivided ionocyte cells into four distinct subpopulations for the first time in E. tetradactylum gills. By employing weighted gene co-expression network analysis (WGCNA), we further investigated the cellular heterogeneity and specific response mechanisms to salinity fluctuant. Our findings revealed the intricate osmoregulation and immune functions of gill cells, highlighting their crucial roles in maintaining homeostasis and adapting to fluctuating salinity levels. This comprehensive cell-type atlas provides valuable insights into the species adaptive strategies, contributing to the conservation and management of this commercially significant fish as well as other euryhaline species.
Air Pocket for a Non-Air-Breathing Fish: Developing Respirometry Methodology for the critically endangered Delta Smelt

Abstract:Respirometry quantifies O2 consumption to estimate metabolic activity across stressor(s), and the resulting data is central for devising conservation strategies for fishes. Due to its rapid adoption and widespread utility, protocols have been established to foster quality control and increase consistency. However, the application of established respirometry protocols on the Delta Smelt (Hypomesus transpacificus) have been largely unsuccessful, with reports over the past 12 years documenting high mortality rates that can only be circumvented with very short measurement duration (~4 h). This prevents the measurement of standard metabolic rate (SMR) and critical O2 tension (Pcrit), and ultimately obstructed physiological research, management actions and conservation tools development. In this study, we detailed the anecdotal, behavioral and empirical evidence that culminated in successful measurements of Delta Smelt SMR and Pcrit at 10, 12, 15, 17 and 19 degrees C. We discovered the Delta Smelt is physotomous and requires frequent refilling of its air bladder to maintain buoyancy. Inspired by air-breathing fish respirometry, the inclusion of an air pocket within their chamber led to considerably more robust respirometry measurements compared to past attempts. The influence of fasting and time of day on O2 consumption were also examined. Altogether, this methodology paves the way for acquiring critical knowledge needed to inform Delta Smelt research and conservation (e.g. the development of metabolic index). The lessons learned may also have implications for other aspects of Delta Smelt conservation such as decreasing conservation aquaculture operating costs and improving survival during fish transport and supplementation.

The Effect of Hypoxia on Gill Morphology and Ionoregulatory Status in the Lake Qinghai Scaleless Carp, Gymnocypris Przewalskii

Abstract:Goldfish and crucian carp at low temperature exhibit plasticity in gill morphology during exposure to hypoxia to enhance gas exchange. Hypoxia-induced changes in gill morphology and cellular ultrastructure of the high altitude scaleless carp from Lake Qinghai, China, were investigated to determine whether this is a general characteristic of cold water carp species. Fish were exposed to acute hypoxia (0.3 mg O-2 l(-1)) for 24 h followed by 12 h recovery in normoxic water (6 mg O-2 l(-1) at 3200 m altitude), with no mortality. Dramatic alterations in gill structure were initiated within 8 h of hypoxia and almost complete by 24 h, and included a gradual reduction of filament epithelial thickness (> 50%), elongation of respiratory lamellae, expansion of lamellar respiratory surface area (> 60%) and reduction in epithelial water-blood diffusion distance (< 50%). An increase in caspase 3 activity in gills occurred following 24 h exposure to hypoxia, indicating possible involvement of apoptosis in gill remodeling. Extensive gill mucous production during hypoxia may have been part of a general stress response or may have played a role in ion exchange and water balance. The large increase in lamellar surface area and reduction in diffusion distance presumably enhances gas transfer during hypoxia (especially in the presence of increased mucous production) but comes with an ionoregulatory cost, as indicated by a 10 and 15% reduction in plasma [Na+] and [Cl-], respectively, within 12-24 h of hypoxia. Within 12 h of hypoxia exposure, 'wavy-convex '-mitochondria rich cells (MRCs) with large apical crypts and numerous branched microvilli were transformed into small 'shallow-basin' cells with a flattened surface. As the apical membrane of MRCs is the site for active ion uptake from the water, a reduction in apical crypt surface area may have contributed to the progressive reduction in plasma [Na+] and [Cl-] observed during hypoxia. The changes in the macro-and ultra-structure of fish gills, and plasma [Na+] and [Cl-] during hypoxia were reversible, showing partial recovery by 12 h following return to normoxia. Although the large morphological changes in the gill observed in the scaleless carp support the hypothesis that gill remodeling during hypoxia is a general characteristic of cold water carp species, the reduced magnitude of the response in scaleless carp relative to goldfish and crucian carp may be a reflection of their more active lifestyle or because they reside in a moderately hypoxic environment at altitude.

Abstract:1. The general structure of the gills of different fishes is compared and it is concluded that, though essentially the same, there are certain differences by which they can be recognized. Possible ways in which they may have evolved from one another are considered. 2. A detailed account is given of the structure of the secondary lamellae, where gaseous exchange takes place, and it is shown that two epithelial sheets are separated by a vascular axis mainly composed of pillar cells overlain by a basement membrane on each side. Blood pathways through the gills are discussed in relation to their respiratory function. 3. The embryonic development of gills is described and evidence regarding homo‐logies of different structures, particularly the pillar cells, is reviewed. 4. The gills of fish having different modes of life show variations in (a) the number of arches, (b) the number and length of the gill filaments, and (c) the size and frequency of the secondary lamellae. Ways in which measurements of gill area may be carried out and some of the complications involved are reviewed and a summary given of measurements made for a wide variety of species. Measurements of the thickness of the water‐blood barrier are also discussed; the more active fish generally have thinner water‐blood barriers and larger gill areas. 5. The different mechanisms of gill ventilation are summarized and characteristics of gill resistance in elasmobranchs and teleosts are compared. Gas exchange is discussed in relation to available techniques and the current terminology and symbols, and to indicate the value of analogies between gill exchangers and systems studied by engineers. 6. It is outlined how studies of the functioning of gills during coughing, parasitic infection, and in polluted waters add to knowledge of their role in respiration.

Effects of Thermal Stress on the Gut Microbiome of Juvenile Milkfish (Chanos chanos)

Abstract:Milkfish, an important aquaculture species in Asian countries, are traditionally cultured in outdoor-based systems. There, they experience potentially stressful fluctuations in environmental conditions, such as temperature, eliciting changes in fish physiology. While the importance of the gut microbiome for the welfare and performance of fish has been recognized, little is known about the effects of thermal stress on the gut microbiome of milkfish and its interactions with the host’s metabolism. We investigated the gut microbiome of juvenile milkfish in a thermal stress experiment, comparing control (26 °C) and elevated temperature (33 °C) treatments over three weeks, analyzing physiological biomarkers, gut microbiome composition, and tank water microbial communities using 16S amplicon sequencing. The gut microbiome was distinct from the tank water and dominated by Cetobacterium, Enterovibrio, and Vibrio. We observed a parallel succession in both temperature treatments, with microbial communities at 33 °C differing more strongly from the control after the initial temperature increase and becoming more similar towards the end of the experiment. As proxy for the fish’s energy status, HSI (hepatosomatic index) was correlated with gut microbiome composition. Our study showed that thermal stress induced changes in the milkfish gut microbiome, which may contribute to the host’s habituation to elevated temperatures over time.

Effect of combined stress (salinity and temperature) in European sea bass Dicentrarchus labrax osmoregulatory processes

Abstract:European sea bass Dicentrarchus labrax undertake seasonal migrations to estuaries and lagoons that are characterized by fluctuations in environmental conditions. Their ability to cope with these unstable habitats is undeniable, but it is still not clear how and to what extent salinity acclimation mechanisms are affected at temperatures higher than in the sea. In this study, juvenile sea bass were pre-acclimated to seawater (SW) at 18°C (temperate) or 24°C (warm) for 2weeks and then transferred to fresh water (FW) or SW at the respective temperature. Transfer to FW for two weeks resulted in decreased blood osmolalities and plasma Cl- at both temperatures. In FW warm conditions, plasma Na+ was ~15% lower and Cl- was ~32% higher than in the temperate-water group. Branchial Na+/K+-ATPase (NKA) activity measured at the acclimation temperature (Vapparent) did not change according to the conditions. Branchial Na+/K+-ATPase activity measured at 37°C (Vmax) was lower in warm conditions and increased in FW compared to SW conditions whatever the considered temperature. Mitochondrion-rich cell (MRC) density increased in FW, notably due to the appearance of lamellar MRCs, but this increase was less pronounced in warm conditions where MRC's size was lower. In SW warm conditions, pavement cell apical microridges are less developed than in other conditions. Overall gill morphometrical parameters (filament thickness, lamellar length and width) differ between fish that have been pre-acclimated to different temperatures. This study shows that a thermal change affects gill plasticity affecting whole-organism ion balance two weeks after salinity transfer.
Seasonal changes in mitochondrial bioenergetics and physiological performance of the bluegill sunfish, Lepomis macrochirus, from a shallow, Midwest river

Abstract:As global temperature shifts due to anthropogenic impacts, seasonal temperatures in shallow aquatic ecosystems are expected to increase. Previous studies on freshwater fishes that experience significant temperature changes during the annual seasons found pronounced physiological restructuring not observed in animals inhabiting more thermally stable environments. Studies evaluating mitochondrial bioenergetics in fish are often performed on animals acclimated to constant temperatures in the laboratory. However, natural habitats are much more complex. Fishes may experience substantial daily and seasonal variation in temperature, energy requirements and resource availability, which are impossible to emulate on acclimation studies. Our study explores the effects of these more complex natural environments on whole-organism thermal tolerance and mitochondrial bioenergetics in bluegill sunfish (Lepomis macrochirus), a native fish to the temperate zone of North America. Compensatory mechanisms and variations in physiological thresholds were observed in specimens acclimatized to the fall season compared to specimens acclimatized to spring and summer seasons. Somatic indices, such as relative weights and hepatosomatic indices, showed significant differences across seasons and critical thermal maxima significantly decreased in the cold acclimatized specimens. Liver mitochondria from L. macrochirus also showed significantly higher uncoupled proton conductance, cytochrome c oxidase (COX) activity, and reduced respiratory control ratios in individuals sampled in the colder season. These findings suggest that mechanisms regulating proton conductance and COX activity modulate mitochondrial function across seasons to sustain physiological fitness in ectotherms inhabiting shallow, inland aquatic habitats.
Evidence of a temperature–oxygen squeeze within floodplain thermal refuge habitats

Abstract:Vertical heterogeneity in lakes and estuaries can present cold‐adapted fishes with a temperature–oxygen squeeze, such that the epilimnion is stressfully warm and the cooler hypolimnion is hypoxic, thereby restricting fishes to the metalimnion. In temperate floodplain rivers, patches of lentic habitat (e.g., alcoves) have the potential to provide thermal refuge for cold‐water fishes during summer, but little is known about whether these smaller habitat features present fish with temperature–oxygen constraints. In this study, we measured temperature and oxygen profiles in six cold‐water alcoves on the Willamette River floodplain (Oregon, U.S.A.) to characterise potential trade‐offs in temperature and oxygen for a cold‐water fish, coastal cutthroat trout (Oncorhynchus clarkii clarkii). To evaluate how fish responded to trade‐offs, we used synergistic methods (needle probe thermocouple, surgically implanted iButtons, and external temperature transmitting radio tags) to monitor fish body temperatures at different temporal scales and compare them to temperature–depth profiles. The cold‐water alcoves displayed a linear relationship between dissolved oxygen and temperature, where cooler temperatures came at the expense of reduced oxygen. Fish body temperatures were intermediate to temperatures recorded at the surface and bottom of the alcove. The span of depths selected by these fish represents less than 20% of the available vertical habitat in these alcoves. These results demonstrate that refuges formed by cool hyporheic upwelling can generate a temperature–oxygen squeeze where fish use only a subset of the refuge habitat feature. Oxygen constraints on thermal refuge use may be a blind spot for climate adaptation planning for cold‐water fishes, because dissolved oxygen data are limited compared to water temperature data, and many cool refuges are fed by subsurface flows which are potentially hypoxic.

Optimal morphometric factors responsible for enhanced gas exchange in fish gills

Abstract:Fish gills are one of the most primitive gas/solute exchange organs, having the highest ventilation volume, present in nature. Such performance is attributed to a functional unit of gill - secondary lamella - that can extract oxygen from an ambience even at a very low partial pressure. For centuries, gills have stood as one of the simplest but an elegant gas/solute exchange organs. Although the role of various morphometric factors of fish gills on gas/solute exchange capabilities have been reported, there has been limited understanding on what makes fish gills as an excellent gas/solute exchange system. Therefore, in the current study, we have theoretically studied the variation of few structural and parametric ratios, which were known to have role in gas/solute exchange, with respect to the weight of fishes. Thereafter, modelling and simulation of convection-diffusion transport through a two dimensional model of secondary lamella were carried out to study different factors affecting the performance of gills. The results obtained from both the studies (theoretical and computational) were in good agreement with each other. Thus, our study suggested that fish gills have optimized parametric ratios, at multiple length scales, throughout an evolution to arrive at an organ with enhanced mass transport capabilities. Further, our study also highlighted the role of length of primary and secondary lamella, surface area of secondary lamellae and inter-lamellar distance on gas/solute exchange capabilities of fish gills. Thus, these defined morphological parameters and parametric ratios could be exploited in future to design and develop efficient gas/solute exchange microdevices.
Regulation of PGC-1α of the Mitochondrial Energy Metabolism Pathway in the Gills of Indian Medaka (Oryzias dancena) under Hypothermal Stress

Abstract:Ectothermic fish exposure to hypothermal stress requires adjusting their metabolic molecular machinery, which was investigated using Indian medaka (Oryzias dancena; 10 weeks old, 2.5 ± 0.5 cm) cultured in fresh water (FW) and seawater (SW; 35‰) at room temperature (28 ± 1 °C). The fish were fed twice a day, once in the morning and once in the evening, and the photoperiod was 12 h:12 h light: dark. In this study, we applied two hypothermal treatments to reveal the mechanisms of energy metabolism via pgc-1α regulation in the gills of Indian medaka; cold-stress (18 °C) and cold-tolerance (extreme cold; 15 °C). The branchial ATP content was significantly higher in the cold-stress group, but not in the cold-tolerance group. In FW- and SW-acclimated medaka, the expression of genes related to mitochondrial energy metabolism, including pgc-1α, prc, Nrf2, tfam, and nd5, was analyzed to illustrate differential responses of mitochondrial energy metabolism to cold-stress and cold-tolerance environments. When exposed to cold-stress, the relative mRNA expression of pgc-1α, prc, and Nrf2 increased from 2 h, whereas that of tfam and nd5 increased significantly from 168 h. When exposed to a cold-tolerant environment, prc was significantly upregulated at 2 h post-cooling in the FW and SW groups, and pgc-1α was significantly upregulated at 2 and 12 h post-cooling in the FW group, while tfam and nd5 were downregulated in both FW and SW fish. Hierarchical clustering revealed gene interactions in the cold-stress group, which promoted diverse mitochondrial energy adaptations, causing an increase in ATP production. However, the cold-tolerant group demonstrated limitations in enhancing ATP levels through mitochondrial regulation via the PGC-1α energy metabolism pathway. These findings suggest that ectothermic fish may develop varying degrees of thermal tolerance over time in response to climate change. This study provides insights into the complex ways in which fish adjust their metabolism when exposed to cold stress, contributing to our knowledge of how they adapt.

Abstract:Gills and gut are the two primary osmoregulatory organs in fish. Recently, studies have expanded beyond the osmoregulatory mechanisms of these organs to explore the microbiota communities inhabiting them. It is now known that microbial communities in both organs shift in response to osmotic stress. However, there are limited studies identifying the major contributors and co-occurrence among these microbiota in both organs under seawater and freshwater transfer conditions. The current data mining report performed a bioinformatics analysis on two previous published datasets from our group, aiming to provide insights into host-bacteria relationships under osmotic stress. We divided the samples into four groups: control seawater gills (LSW); control seawater gut (TSW); freshwater transfer gills (LFW); and freshwater transfer gut (TFW). Our results showed that LSW had higher diversities, richness, and evenness compared to TSW. However, both the LFW and LSW did not show any significant differences after the freshwater transfer experiment. We further applied co-occurrence network analysis and, for the first time, reported on the interactions of taxa shaping the community structure in these two organs. Moreover, we identified enriched ectoine biosynthesis in seawater samples, suggesting its potential role in seawater environments. Increased mRNA expression levels of Na+/K+-atpase, and cftr, were observed in gills after 6 h of ectoine treatment. These findings provide a foundation for future studies on host-bacteria interactions under osmotic stress.
Effects of different temperatures on Leiocassis longirostris gill structure and intestinal microbial composition

Abstract:Fish are poikilothermic vertebrates and their physiological activities are affected by water temperature. In recent years, extreme weather has occurred frequently, and temperature changes have adversely affected the growth of farmed fish. To explore the changes in gill tissue structure caused by changing the water temperature and the relationship between the intestinal microbiota and the Leiocassis longirostris host adaptation mechanism, gill tissue sections and intestinal microbial 16S rRNA amplicon sequencing were conducted under different temperature stress (low temperature 4 °C, normal temperature 26 °C and high temperature 32 °C). The results showed that heat stress and cold stress caused injury and swelling, terminal congestion, cell vacuolation, and necrosis of the gill tissue of L. longirostris . For intestinal microbiota, the abundances of Pseudomonadota and Bacillota increased at the cold stress, while the abundances of Fusobacteriota and Bacteroidota increased at the heat stress. The number of opportunistic bacteria, mainly Aeromonas and Acinetobacter , was the highest under cold stress. In addition, the richness of the intestinal microbiota decreased significantly at heat and cold stresses, while evenness increased. Prediction of intestinal microbiota function showed that most common functions, such as metabolism of cofactors and vitamins, energy metabolism and replication and repair, were decreased significantly at heat stress and cold stress, and phylogenetic relationship analysis revealed significant differences among the groups. In conclusion, the change of temperature altered the gill tissue structure, and affected the structure and homeostasis of the intestinal microbiota, thus affecting the survival time of L. longirostris , and cold stress had a greater effect than heat stress.

Abstract:Accurately quantifying air–sea heat and gas exchange is crucial for comprehending thermoregulation processes and modeling ocean dynamics; these models incorporate bulk formulae for air–sea exchange derived in unstable atmospheric conditions. Therefore, their applicability in stable atmospheric conditions, such as desert-enclosed basins in the Gulf of Eilat/Aqaba (coral refugium), Red Sea, and Persian Gulf, is unclear. We present 2-year eddy covariance results from the Gulf of Eilat, a natural laboratory for studying air–sea interactions in stable atmospheric conditions, which are directly related to ocean dynamics.

Abstract:One of the main expected responses of marine fishes to ocean warming is decrease in body size, as supported by evidence from empirical data and theoretical modeling. The theoretical underpinning for fish shrinking is that the oxygen supply to large fish size cannot be met by their gills, whose surface area cannot keep up with the oxygen demand by their three-dimensional bodies. However, Lefevre et al. (Global Change Biology, 2017, 23, 3449-3459) argue against such theory. Here, we re-assert, with the Gill-Oxygen Limitation Theory (GOLT), that gills, which must retain the properties of open surfaces because their growth, even while hyperallometric, cannot keep up with the demand of growing three-dimensional bodies. Also, we show that a wide range of biological features of fish and other water-breathing organisms can be understood when gill area limitation is used as an explanation. We also note that an alternative to GOLT, offering a more parsimonious explanation for these features of water-breathers has not been proposed. Available empirical evidence corroborates predictions of decrease in body sizes under ocean warming based on GOLT, with the magnitude of the predicted change increases when using more species-specific parameter values of metabolic scaling.

Abstract:BackgroundChanges in ambient temperature seriously affect physiological regulation and biochemical reactions in ectotherms. However, transient elevation in oceanic temperature occurs naturally during the day. Short-term elevation in the ambient temperature affects different physiological responses in marine fish, including cellular protein stability and osmotic balance of the internal environment. Since fish gills are vital osmoregulatory organ which directly contacts external environment, activation of cytoprotective responses to maintain gill cell viability and biological function is essential for fish survival under challenging environmental conditions. The purpose of this study was to investigate the short-term effects of elevated temperature on physiological regulation in the gills of a marine teleost, blue green damselfish (Chromis viridis).ResultsAs part of the stress response, plasma glucose levels were induced by short-term hyperthermic exposure (12 h). Furthermore, upregulation of the levels of gill heat shock proteins (HSPs) and ubiquitinated proteins was essential for preventing the accumulation of protein aggregations in branchial cells of C. viridis under hyperthermic stress. The specific activity of branchial Na+/K+-ATPase (NKA), however, significantly reduced while the amount of protein was similar between normal and high-temperature groups.ConclusionsThe present study provided the evidence to illustrate that activation of the branchial protein quality control mechanism to carry out cytoprotective response was involved in coping with thermal stress. However, plasma osmolality and muscle water content, respectively, that slightly but evidently increased and decreased might result from impaired osmoregulatory ability due to hyperthermia-decreased gill NKA activity.

Abstract:The intracellular build-up of thermally damaged proteins following exposure to heat stress results in the synthesis of a family of evolutionarily conserved proteins called heat shock proteins (Hsps) that act as molecular chaperones, protecting the cell against the aggregation of denatured proteins. The transcriptional regulation of heat shock genes by heat shock factor 1 (HSF1) has been extensively studied in model systems, but little research has focused on the role HSF1 plays in Hsp gene expression in eurythermal organisms from broadly fluctuating thermal environments. The threshold temperature for Hsp induction in these organisms shifts with the recent thermal history of the individual but the mechanism by which this plasticity in Hsp induction temperature is achieved is unknown. We examined the effect of thermal acclimation on the heat-activation of HSF1 in the eurythermal teleost Gillichthys mirabilis. After a 5-week acclimation period (at 13, 21 or 28 degrees C) the temperature of HSF1 activation was positively correlated with acclimation temperature. HSF1 activation peaked at 27 degrees C in fish acclimated to 13 degrees C, at 33 degrees C in the 21 degrees C group, and at 36 degrees C in the 28 degrees C group. Concentrations of both HSF1 and Hsp70 in the 28 degrees C group were significantly higher than in the colder acclimated fish. Plasticity in HSF1 activation may be important to the adjustable nature of the heat shock response in eurythermal organisms and the environmental control of Hsp gene expression.
