Abstract:The field asymmetric ion mobility spectrometric (FAIMS) technique was introduced into the rapid analysis of illegally added ingredients in a traditional Chinese antidiabetic medicine for the first time. The metformin, phenformin and glibenclamide as well as the marketed medicine were detected and identified by a co-developed Thermal Desorption-FAIMS (TD-FAIMS) system. The temperature of the sampling tube for thermal desorption was set as 160 degrees C. The test pressure was 1.07x10(5) Pa. The range of the high field was set from 0% to 100% of the maximum field for the FAIMS spectrometer. The Compensation voltage (CV) scanned from. 6 V to 6 V. Eight groups of fingerprint points contained three variables of CV, E/N and Ion current (AI) were extracted from the spectra of the three reference substances and their combination mixtures. The results indicated that the medicine sample had the same spectrum as the mixture of three reference substances, which gave evidence of illegal addition into the traditional Chinese medicine. The result was confirmed by HPLC analysis by comparing spectrum of the medicine sample with those of metformin, phenformin and glibenclamide. The FAIMS instrument, which features its simplicity, high sensitivity and selectivity and fast response, demonstrates huge potential in the rapid analysis of traditional Chinese medicine.
Simultaneous identification of three emergent stimulant laxative adulterants in slimming foods using only one antibody

Abstract:Stimulant laxatives were recently found to be abused in slimming foods, resulting in harmful effects on consumers. To ensure the safety of relative products, sensitive yet multiplex immunoassays are crucial in rapid screening of stimulant laxatives. However, there are few immunoassays for these substances, and even less for broad-specific recognition. Thus, in this work, four theoretically promising haptens of emerging stimulant laxative bisacodyl were rationally designed using molecular modeling and synthesized to immune animals, whose feasibility was confirmed by the obtained broad-specific antibody. Based on this unique antibody, a highly sensitive multiplex competitive indirect enzyme-linked immunosorbent assay (ciELISA) was established with low limits of detection for bisacodyl, sodium picosulfate, and BHPM (0.23, 13.68, and 0.11 ng/mL). In spiked sample recovery test and real sample detection, this ciELISA exhibited acceptable consistency with the validation method, demonstrating high accuracy and applicability of our method. This reliable multiplex ciELISA proceeds the rapid screening of stimulant laxatives in slimming foods.

Abstract:In this work, novel ionic liquid (IL) functionalized polyacrylonitrile nanofibers mat (IL/PAN-NFsM) was firstly prepared through thiol-ene "click" reaction and evaluated for the establishment of a sensitive and high-throughput screening method. Because of its excellent pre-separation efficiency, IL/PAN-NFsM can adsorb trace-level targets from complex sample matrix within tens of seconds by performing a solid-phase extraction (SPE) process, and then served as sampling modules of direct analysis real time mass spectrometry (DART-MS) without any additional processing. This means the target analytes concentrated on IL/PAN-NFsM were directly desorbed, ionized, and detected by DART-MS. To verify the feasibility of the proposed method, three illegal added synthetic drugs (gliclazide, glimepiride, and gliquidone) were screened in six types of antidiabetic health-care tea samples. The results indicated that the sensitivity is in the level of ng g(-1), while total analysis time does not exceed 1.0 min per sample. Moreover, the stability expressed as relative standard deviation (RSD) varies from 1.7% to 17.3%. We proposed a new screening mode based on the direct combination of functionalized NFsM and DART-MS, which is expected to become a universal method in food safety analysis.
Simultaneous Identification, Confirmation and Quantitation of Illegal Adulterated Antidiabetics in Herbal Medicines and Dietary Supplements Using High-Resolution Benchtop Quadrupole-Orbitrap Mass Spectrometry.

Abstract:This paper presents an application of ultrahigh-performance liquid chromatography and quadrupole Orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap HR MS) for the screening, confirmation and quantification of 11 antidiabetics in herbal medicines and dietary supplements. The mass spectrometer was operated in Full MS/dd-MS(2) (data-dependent MS(2)) mode. The full MS scan acquired data for identification and quantification, and dd-MS(2) scan obtained product ion spectra for confirmation. UHPLC-Q-Orbitrap MS quantification was achieved using matrix-matched standard calibration curves with phenacetin as internal standard. The method validation that included selectivity, sensitivity, calibration curve, accuracy and precision, recovery, matrix effect and stability was evaluated. The response showed good linear relationship with the concentrations of analytes over wide ranges (e.g., 0.0004-1 μg/g for metformin) with all the coefficients of correlation (r(2)) >0.9991. The detection limits (LODs) were in the range of 0.05-0.5 ng/g for different analytes. The recoveries yielded results higher than 74.3% for all compounds. The accuracy was in the range of -6.75 to 3.85%, while the intra- and inter-day precision ranged from 0.048 to 11.5%. Among 63 batches of herbal medicines and 34 batches of dietary supplements samples, 7 batches of dietary supplements were positive, while all the herbal medicines were negative. Overall, the novel UHPLC-Q-Orbitrap has demonstrated great performance for identification, confirmation and quantification of antidiabetics in herbal medicines and dietary supplements, ensuring food safety and public health.
Rapid Screening of Emergent Febuxostat Adulteration in Functional Foods Based on a Simulation-Inspired High-Quality Antibody

Abstract:Due to the potential health risks of adulterated febuxostat in uric-acid-lowering foods, it is urgent to develop rapid detection methods. However, there are no fast analytical techniques for febuxostat yet. Herein, an efficient hapten simulation strategy was proposed to successfully produce a highly sensitive and selective monoclonal antibody toward febuxostat. Based on such a robust recognition element, easy colorimetric and ultrasensitive fluorescent lateral flow immunochromatographic immunoassays were first established, which can detect febuxostat as low as 60 μg/kg by the naked eye or 1.01 μg/kg by a commercial test strip reader with acceptable stability. Furthermore, in the recovery test and blind sample analysis, consistent results between our methods and the authorized liquid chromatography-tandem mass spectrometry method suggested the high accuracy and practicality of this work. The present work not only proposes a rational hapten design idea but also provides favorable tools for the rapid screening of febuxostat in functional foods.
Simultaneous Determination of Adulterants in Dietary Food Supplements Using Multivariate Data Analysis after Preconcentration with Novel Nanosorbents and Chromatographic Measurement

Abstract:Background: The increasing popularity of dietary supplements and, consequently, related adulteration emphasizes the rising need to examine the association of food supplements with fraud. Intentional or unintentional fraud in food supplements by hazardous chemicals compounds is a problem that many countries are struggling with. Much effort have been made to effectively and reliably control the quality of food supplements. Objective: Due to the importance of the subject, an analytical method for the simultaneous and reliable detection and quantitative determination of three key adulterants in dietary food supplements was developed. The proposed method benefits from analytical methods and multivariate calibration methods to progress the determination of adulterants in a complex matrix. Methods: HPLC assisted by multivariate curve resolution-alternating least square (MCR-ALS) analysis was used to detect adulterants in real samples after separation and preconcentration using novel mesoporous carbon nanoparticles. Solid-phase extraction (SPE) optimization was accomplished by central composite design (CCD). In order to obtain the best results, the MCR-ALS model was compared with the parallel factor analysis 2 (PARAFAC2) model and validated by estimation of linearity, detection limits, and recovery. Results: The detection limits and linear dynamics were calculated as 1.5, 4.27, and 4.77 µg/mL, and 1-50, 5-20, and 5-20 µg/mL for caffeine, ephedrine, and fluoxetine, respectively. Mean recovery for determination of caffeine, ephedrine, and fluoxetine using the developed method was reported as 101.75, 91.7, and 92.36, respectively. Conclusion: The results showed that to avoid negative health outcomes associated with the excessive consumption of adulterated food supplements releasing such products should be carefully regulated. The developed method was validated using statistical factors and showed acceptable and reliable results. Highlights: (1) The application of MCR-ALS coupled with HPLC-Diode-Array Detection data sets allowed the simultaneous identification and quantification of three key adulterants (caffeine, ephedrine, and fluoxetine) in dietary food supplements. (2) A small amount of the novel adsorbent was successfully used to preconcentrate the trace amounts of adulterants in samples. (3) This method benefits from the chemometrics tools and experimental design to significantly reduce the use of toxic solvents and complicated instruments to propose a less time-consuming method for quantification of multicomponents in the presence of uncalibrated interferents.

Abstract:Adulteration of herbal supplements with synthetic drugs is illegal. A rapid and reliable method which utilizes direct analysis in real time mass spectrometry (DART-MS) was developed for the identification of seven synthetic antidiabetic drugs used as adulterants in herbal dietary supplements. The supplement sample was simply extracted with methanol/water by manually shaking several times and directly analyzed using DART-MS. The presence of synthetic drug adulterants was confirmed through the accurate m/z values and MS/MS data obtained via quadruple time of flight mass spectrometry (QTOF MS). Parameters for the DART source were systematically optimized, and the limits of detection (LODs) in herbal supplement matrices were measured. This method was successfully applied to examine five commercial herbal dietary supplements, and two of them proved to be adulterated with metformin without labeling.

Abstract:A procedure was established and fully validated for the screening and quantification of fourteen synthetic antidiabetic adulterants in herbal pharmaceuticals and health foods, including metformin (MF), buformin (BF), phenformin (PHF), rosiglitazone (RGZ), pioglitazone (PGZ), chlorpropamide (CPM), glipizide (GPZ), tolbutamide (TBM), gliclazide (GCZ), glibenclamide (GBM), glimepiride (GMR), repaglinide (RGN), gliquidone (GQD) and nateglinide (NGN). The samples were extracted by methanol and separated by HPLC. Retention times and ultraviolet spectra were used for the preliminary screening, and the suspected adulterants were then confirmed by liquid chromatography-quadrupole-time of flight mass spectrometry (LC-Q-TOF-MS/MS) and quantified by HPLC. The developed procedure was successfully applied to assess twenty-four herbal samples, and PHF, GCZ, GBM, MF, GPZ and BF were found in many. To the best of our knowledge, this is the first report of simultaneous screening and quantification of these fourteen synthetic antidiabetic adulterants from any matrix.

Abstract:In this work, a rapid chemometrics-enhanced liquid chromatography–single stage mass spectrometry (LC–MS) was developed for the simultaneous green determination of six co-eluted sulfonylurea-type oral antidiabetic agents (SOADs) in health teas and human plasma samples. Shortening the chromatographic separation time and reducing the cost per analysis were achieved via using full scan mode of LC–MS under simple isocratic elution condition followed by an environment-friendly “mathematical separation” strategy. The problem of the complete separation of target analytes from each other and/or from the uncalibrated interferences in complex matrices was resolved by alternating trilinear decomposition (ATLD) method as a posteriori. Satisfactory qualitative and quantitative results were obtained even in the existence of unknown interferences and the “second-order advantage” was fully exploited. The average spiked recoveries for all target analytes were between 81.6% and 110.1% with standard deviations less than 7.7%. It was demonstrated that the proposed strategy could be promisingly used for green resolution and determination of co-eluted multi-analytes of interest in complex samples while avoiding elaborate sample pretreatment steps and complicated experimental conditions as well as more sophisticated high-cost instrumentations.

Abstract:The rapid and accurate detection of illegal adulteration of chemical drugs into dietary supplements is a big challenge in the food chemistry field. Detection of compounds without a standard reference is even more difficult; however, this is a common situation. Here in this study, a novel "standard‐free detection of adulteration" (SFDA) method was proposed and phosphodiesterase‐5 inhibitor derivatives were used as an example to figure out the possibility and reliability of this SFDA method. After analysis by quadrupole coupled time of flight–tandem mass spectrometry detection and multivariable statistics, six common fragment ions were chosen to indicate whether adulteration was present or not, while 20 characteristic fragment ions indicated whether adulteration was by nitrogen‐containing heterocycles or by anilines. Furthermore, the quantitative methods were conducted by high‐performance liquid chromatography–tandem mass spectrometry. In a word, this strategy allows for a quick determination of dietary supplement adulteration without any need for standard materials, improving the efficacy of food safety testing.

Abstract:The measurement of human insulin and its synthetic analogues in biological matrices has become increasingly important not only in clinical fields but also in doping control. The use of insulin and its analogues have been included in the list of prohibited substances published by the World Anti-Doping Agency (WADA). This study describes a qualitative method for detection of insulin analogues (lispro, aspart, glulisine, glargine, degludec, detemir) in human urine. The sample preparation consists of a preconcentration step using ultrafiltration followed by an immunoaffinity extraction with an antibody precoated ELISA plate. The obtained extracts are analyzed by conventional high-performance liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS). The limits of detection range between 10 pg/ml and 150 pg/ml. The applicability of the method was proven by the analysis of real urine samples obtained from diabetic patients treated with synthetic insulin analogues.

Abstract:Sodium saccharin is a common artificial sweetener. However, due to its possible carcinogenic effects and causing metabolic disorders, many countries have strictly regulated its use in food. In the study, we prepared a specific monoclonal antibody (mAb 2H11) using the new hapten (6-carboxylsaccharin) and developed a direct competitive enzyme-linked immunosorbent assay (dcELISA) for the screening of sodium saccharin residue in food. The half-maximum inhibition concentration (IC50 ) and working range (IC20 -IC80 , the concentrations causing 20% and 80% inhibition by sodium saccharin) were 32.5 and 6.47 to 164 ng/mL, which was 6.5 times more sensitive than the previously reported immunoassay. The average recoveries of sodium saccharin in spiked food samples detected by dcELISA ranged from 82.1% to 117%. Among 70 food samples bought in the physical stores and online, sodium saccharin residues were only detected in four samples purchased online (one canned pineapple, two winter jujube, and one kimchi). The content measured by dcELISA agreed well with those determined by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The developed dcELISA was proved to be a sensitive and accurate method for determining sodium saccharin in food. PRACTICAL APPLICATION: Quantitation of sodium saccharin residue in food is very necessary and important for consumers and regulatory agencies.

Abstract:<p>A high-efficiency approach for the synthesis of molecularly imprinted polymers has been developed and further for the solid-phase extraction of sulfonylurea herbicides in food samples. Molecular simulation approach combined chemometric selected metsulfuron-methyl (MSM) and 2-trifluoromethyl acrylic acid (TFMAA) as the template and the monomer to synthesize the molecularly imprinted polymers (MIPs). Experimental validation confirmed that the MSM-imprinted polymers showed a higher selectivity and affinity to sulfonylurea herbicides. The optimized molecularly imprinted solid-phase extraction (MISPE) conditions, including loading, washing, and eluting conditions, were established. The developed MISPE technology combined HPLC-MSMS was successfully used for the determination of sulfonylurea herbicides in foods. Compared with commercial SPE columns, MISPE showed high affinity, excellent selectivity and low matrix effect. The recoveries of sulfonylurea herbicides spiked in four matrices were between 86.4% and 100.2%, with the relative standard deviations (RSD) in the range of 0.9% -10.5%.</p>

Abstract:Adulterating herbal and dietary supplements with synthetic hypoglycemics is widely practiced, especially in developing and undeveloped countries. A simple, precise, and novel method was developed to simultaneously determine synthetic antidiabetics belonging to different classes, namely biguanide, sulfonylurea, and thiazolidinedione, in counterfeit herbal products as adulterants using HPTLC-MS. The method can simultaneously identify and quantify undeclared synthetic drugs metformin, pioglitazone, glipizide, and glimepiride in herbal as well as dietary supplements. The optimized mobile phase for the TLC system consists of cyclohexane:dichloromethane:1-propanol:saturated solution of ammonium acetate in acetic acid in the ratio of (7:5:2:2, v/v/v/v). The method is highly selective and sensitive for detecting chemicals in complex herbal molecules. Identification and quantification of drugs were performed using the densitometric method. The drugs were confirmed using triple quadrupole mass spectrometry through the MS interface. The screening of market preparations revealed the presence of metformin in two of eight products. This study shows that the method for simultaneous determination of these four drugs can be applied successfully to screen synthetic antidiabetics in the herbal complex matrix as adulterants.
Identification of fenfluramine adulteration in slimming foods using a highly sensitive immunoassay

Abstract:Fenfluramine, an appetite suppressant used for weight loss, can cause significant harm if overdosed. The unauthorized addition of fenfluramine to slimming foods has raised concerns. Currently, no rapid screening method is available for the quick detection of fenfluramine in the field. This study proposes six new haptens with varying spacer arm lengths and hydrophobicity, promising to elicit antibodies capable of being highly specific and strongly binding to fenfluramine. The study found that highly hydrophobic haptens with long spacer arms favored the generation of highly sensitive antibodies. Key interaction forces for antibody recognition of fenfluramine were revealed by intrinsic molecular mechanisms. Based on the above results, monoclonal antibody for fenfluramine was prepared and an ultrasensitive icELISA method with heterologous coating strategies was developed in slimming foods. The IC 50 of the method was 6.25 ng/mL, the linear detection range was 0.47–83.51 ng/mL and the detection limit was 0.10 ng/mL. Recovery rates in tea bags, tablets, capsules, coffee, and beverages ranged from 96.56 % to 108.90 %. The method was successfully applied to blind samples, showing strong correlation with HPLC-MS/MS results. Thus, the developed method is suitable for identifying fenfluramine adulteration in slimming foods.

Abstract:Traditional lateral flow immunoassays (TLFIAs) are valued for their simplicity, speed, and user-friendliness. However, the specificity of conventional test strips often necessitates large quantities of antigen-protein conjugates for target detection, which can be resource-intensive. Here, we present a strategy aimed at enhancing the universality of test strips while reducing the consumption of antigen-protein conjugates, without compromising sensitivity. By coating streptavidin on the test line and employing bifunctional antigen-protein conjugates (competitor to target and immunoprobe linker), the test strip was thus served as a universal module. We developed three universal lateral flow immunoassays (ULFIAs) for the detection of aflatoxin B1 (AFB1), carbendazim (CBZ), and enrofloxacin (ENR). Compared to traditional methods based on the same aggregation-induced emission fluorescent microspheres, the proposed ULFIAs demonstrated a significant increase in sensitivity, with enhancements of 11.0-fold for AFB1, 10.9-fold for CBZ, and 4.1-fold for ENR. Additionally, this approach substantially reduced the consumption of antigen-protein conjugates by 19.1-fold, 40.9-fold, and 23.8-fold, respectively, thereby promoting greener detection methods. This bifunctional antigen conjugate strategy offers a promising pathway for the sensitive detection of small molecule hazards.

Abstract:Pharmaceutical drugs like Sildenafil are illegally added to functional food such as nutritional supplements and herbal remedies to deliver drugs without a regular prescription to consumers. Rapid screening of illegal additives is desirable for the public security department. The seized samples are often large in number and unknown in composition; methods are needed for qualitative screening of unknown samples. Here, a new approach is presented based on atmospheric pressure solids analysis probe (ASAP) coupled with single-quadrupole mass spectrometer to rapidly screen 42 common illegal additives in six categories of functional food. The ASAP-MS method could be applied to solid or liquid sample analysis with a very simple pre-treatment and no LC chromatographic separation, using a home-built library; the identification of suspicious additives could be obtained rapidly. More importantly, the approach is sensitive enough for complex matrix samples like coffee samples. 21 batches of seized unknown samples were tested by the ASAP-MS, and the positive results were confirmed by LC-MS/MS(QQQ), indicating that the ASAP-MS method is effective and reliable. The ASAP-MS with home-built library is a promising method for rapid screening of illegal additives in functional food, which could be widely used in the grassroots police station that lack professional laboratory environment.

Abstract:In recent years, dietary supplements and herbal medicines are increasing in popularity all over the world. However, it is problematic that some manufacturers illegally included synthetic drugs in their products. Due to the extremely complex matrices of those products, most existing methods for screening illegal adulterations are time-consuming and liable to false positive. In this paper, a robust LC/MS/MS method for the high-throughput, sensitive and reliable determination of illegal adulterations from herbal medicines and dietary supplements was established. Minimal LC separation was employed and MRM was used to simultaneously monitor the three transitions under their respective optimal collision energy for each compound. Positive results were determined only if well-defined peaks appeared at all of the three transitions and the ratios among the peak areas were within given threshold. In this study, the method had been applied for the screening of nine most commonly adulterated therapeutic substances, such as sildenafil (Viagra) and famotidine, and the lower limits of detection of these compounds ranged from 0.05 to 1.5 ng/ml. Little sample preparation was needed for this method and the analysis time was less than 5 min/sample. The reliability has been demonstrated by the test with blank matrix. Over 200 products that were under suspicion by SDA of China had been assayed and till now no false negative or positive result was found. This method is rapid, simple, reliable and capable of screening multiple adulterants in one run.

Abstract:With the expansion of the functional food market, the qualification assessment of these products has become a major challenge, and efficient analytical tools are urgently needed. Here, a miniature mass spectrometer (MS) with self-aspiration capillary electrospray ionization (SACESI) source and ion trap analyzer was developed for rapid screening of various illegally added drugs in functional foods. No chromatographic separation was required, but a simplified two-step pretreatment method was developed to reduce the operational procedures and time consumption of the entire analysis. SACESI source uses capillary action to drive solution injection, which utilizes a simple structure and convenient operation to constitute a kind of disposable MS detection solution. To achieve accurate and automatic identification, an intelligent recognition algorithm with steps of spectrum preprocessing, characteristic peak matching, and support vector machine learning was constructed. The relative accuracy of rapid screening of 31 suspicious drugs in various samples is up to 99.78%. It achieves 100% correct identification for the 55 batches of actual samples captured by on-site inspection, which demonstrates the feasibility of the proposed analytical system and strategy in food safety applications.
