Assessment of Plant Growth Promoting and Abiotic Stress Tolerance Properties of Wheat Endophytic Fungi

Abstract:The aims of the present work were to isolate and characterize fungal endophytic communities associated with healthy wheat ( Triticum aestivum L. ) plants, collected from the North China. Segregated endophytes were screened for their PGP traits, abiotic stresses (heavy metals, salinity, drought, and temperature), and antibiotic sensitivity. A total of 16 endophytic fungi were isolated using the culture-dependent approach from different tissue parts of wheat plants. Based upon their internal transcribed spacer (ITS) rDNA gene sequencing, 15 out of 16 isolates were selected for further analysis. In the contemporary investigation, a number of the tested endophytes exhibited fairly good 1-aminocyclopropane-1-carboxylic acid deaminase (ACCD) (0.03±0.011 to 1.43±0.01 μ mol α -KB mg −1 protein hr −1 ), indole acetic acid (IAA) (1.125±0.04 to36.12±0.004 μ gml −1 ), and phosphate solubilizing index (PSI) (2.08±0.03to5.16±0.36) activities. More than 30% isolates gave positive result for siderophore and ammonia tests, whereas all exhibited catalase activity but only 2 (582PDA1 and 582PDA11) produced hydrogen cyanide. Trichoderma strains showed salt, heavy metals, and drought tolerance at high levels and also exhibited resistance to all the tested antibiotics. Strain 582PDA4 was found to be the most temperature (55°C) tolerant isolate. The findings of this study indicated that the microbial endophytes isolated from wheat plants possessing a crucial function to improve plant growth could be utilized as biofertilizers or bioagents to establish a sustainable crop production system.

Biocontrol Efficacy of Endophyte Pseudomonas Poae to Alleviate Fusarium Seedling Blight by Refining the Morpho-Physiological Attributes of Wheat.

Abstract:Some endophyte bacteria can improve plant growth and suppress plant diseases. However, little is known about the potential of endophytes bacteria to promote wheat growth and suppress the Fusarium seedling blight pathogen Fusarium graminearum. This study was conducted to isolate and identify endophytic bacteria and evaluate their efficacy for the plant growth promotion and disease suppression of Fusarium seedling blight (FSB) in wheat. The Pseudomonas poae strain CO showed strong antifungal activity in vitro and under greenhouse conditions against F. graminearum strain PH-1. The cell-free supernatants (CFSs) of P. poae strain CO were able to inhibit the mycelium growth, the number of colonies forming, spore germination, germ tube length, and the mycotoxin production of FSB with an inhibition rate of 87.00, 62.25, 51.33, 69.29, and 71.08%, respectively, with the highest concentration of CFSs. The results indicated that P. poae exhibited multifarious antifungal properties, such as the production of hydrolytic enzymes, siderophores, and lipopeptides. In addition, compared to untreated seeds, wheat plants treated with the strain showed significant growth rates, where root and shoot length increased by about 33% and the weight of fresh roots, fresh shoots, dry roots, and dry shoots by 50%. In addition, the strain produced high levels of indole-3-acetic acid, phosphate solubilization, and nitrogen fixation. Finally, the strain demonstrated strong antagonistic properties as well as a variety of plant growth-promoting properties. Thus, this result suggest that this strain could be used as an alternate to synthetic chemicals, which can serve as an effective method of protecting wheat from fungal infection.

Abstract:The fall armyworm (FAW) Spodoptera frugiperda , is a voracious pest of cereals native to the Americas and which invaded Africa in 2016. Chemical control is the main management option, which however remains ineffective and unsustainable. Fungal endophytes are increasingly used as alternative for the management of insect pests of economic importance. This study assessed the potential of eight endophytic fungal isolates to colonize maize plant and their ability to promote seedlings growth through seed and foliar inoculations, as well as their suppressive effects on FAW. Fungal colonization rates of different plant parts by the endophytes varied as per the inoculation methods. Beauveria bassiana ICIPE 279 colonized more than 60% of all the seedling parts while B . bassiana G1LU3 only colonized stem (25%) and leaf (5%) tissues through foliar inoculation. Trichoderma atroviride F2S21, T. asperellum M2RT4, T. harzianum F2R41, Trichoderma sp. F2L41, Hypocrea lixii F3ST1 and Fusarium proliferatum F2S51 successfully colonized all the plant parts and therefore were selected and further evaluated through seed inoculation for their endophytic persistence, effect on plant growth, and pathogenicity to Spodoptera frugiperda immature and adult stages. Weekly assessment showed varied effect of the endophytes on maize plant growth parameters compared to the control. During the first week, percentage colonization of the plant parts ranges between 90%–100%, 65%–100%, and 60%–100%, in the roots, stems, and leaves, respectively for all the five tested isolates. However, the colonization pattern/rates significantly decreased over time for H. lixii F3ST1 in the stems and leaves, and for T. harzianum F2R41 in the leaves and for T. asperellum M2RT4 in the roots. In addition, T. harzianum F2R41 outperformed all the other isolates in boosting the plant height, whereas H. lixii F3ST1 and T. asperellum M2RT4 outperformed all the other isolates in increasing the wet and dry shoots weight. Furthermore, the number of egg masses laid on endophytically-colonized maize plants varied among the treatments. Trichoderma asperellum M2RT4 and H. lixii F3ST1 endophytically-colonized maize plants significantly reduced the number of egg masses and the defoliation/feeding rates of the pest compared to the control. Additionally, T. harzianum F2R41 had the highest negative impact on the pupation and adult emergence of S. frugiperda with a female-biased sex ratio. Our findings indicate that T. asperellum M2RT4, T. harzianum F2R41, and H. lixii F3ST1 hold a potential to be developed as endophytic-fungal-based biopesticides for sustainable management of S. frugiperda and as plant growth promoters.

Abstract:Plant growth promoting fungi (PGPF) were employed in the present study to biocontrol Fusarium wilt disease in pepper plants. Two of the five fungal isolates were chosen based on biochemical characteristics such as their production of hydrocyanic acid, siderophores, and IAA, phosphate solubilization, and in vitro antifungal activities. The most potent fungal isolates were identified as Penicillium expansum (P. expansum) and Trichoderma harzianum (T. harzianum). Using GC-MS, it was found that PGPF extracts contain compounds with antifungal activity, antioxidants, and plant growth stimulators. The combined effect of T. harzianum and P. expansum increased the protection against fusarial wilt by (76.74%), followed by T. harzianum by (50%), then P. expansum by (17.64%). Significant improvement because of using the mixture (T. harzianum and P. expansum) showed an increase in shoot length (59.4%), root length (129%), and number of leaves (52.6%). Chlorophyll A and B levels in infected plants were consistently raised by 28.71% and 67.58%, respectively; as a result of application the mixture (T. harzianum and P. expansum). Also, there was an increase in soluble proteins and carbohydrates in infected plants treated with (T. harzianum) by 25.42% and 31.78% over untreated infected plants, respectively. It could be recommended that the use of targeted PGPF strains, especially a mixture of T. harzianum and P. expansum could be commercially used as therapeutic nutrients against Fusarium wilt of pepper plants.

Abstract:The biosynthesis of zinc oxide nanoparticles (ZnONPs) offers great potential for plant disease management due to their potent antimicrobial properties and environmental safety. However, the precise mechanisms underlying their antifungal mode of action and role in suppressing mycotoxins remain unclear. This study aims to elucidate the mechanisms by which ZnONPs suppress the pathogenic fungus Fusarium graminearium, known to cause Fusarium head blight in wheat. Additionally, it investigates how ZnONPs mitigate the production of mycotoxins, which pose risks to humans and ruminants. The study demonstrates that ZnONPs, bioproduced by Pseudomonas poae (P. poae), inhibit not only fungal growth, colony formation, and spore germination, but also significantly reduce mycotoxin production of F. graminearium by inhibiting the synthesis of deoxynivalenol (DON), downregulating the FgTRI gene, and causing morphological alterations of the toxisomes. The results also highlight that ZnONPs exert significant effects on fungi through multiple mechanisms, including cell wall damage and the generation of reactive oxygen species (ROS). Moreover, ZnONPs effectively inhibit F. graminearium in wheat leaves and coleoptiles. Fluorescence microscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), and optical microscopy all show that ZnONPs stop F. graminearium from getting into wheat plants and colonising them. Overall, the findings of this study provide evidence that ZnONPs are highly effective in reducing F. graminearium colonization in wheat plants and effectively decreasing mycotoxin production through multiple pathways.
Screening, Identification, and Optimization of Fermentation Conditions of an Antagonistic Endophyte to Wheat Head Blight

Abstract:Fusarium Head Blight (FHB, scab) is a destructive fungal disease that causes extensive yield and quality losses in wheat and other small cereals. Biological control of FHB is considered to be an alternative disease management strategy that is environmentally benign, durable, and compatible with other control measures. In this study, to screen antagonistic bacteria with the potential to manage FHB, 113 endophytes were isolated from the stems, leaves, panicles, and roots of wheat. Among them, six strains appeared to effectively inhibit Fusarium graminearum growth and one isolate, XS-2, showed a highly antagonistic effect against FHB. An in vitro antagonistic test of XS-2 on wheat heads confirmed that XS-2 could suppress the disease severity of FHB. The 16S rDNA sequence analysis revealed that XS-2 is a strain of Bacillus amyloliquefaciens. Antagonistic spectrum analyses showed that XS-2 had antagonistic effects against two and four types of cotton and fruit tree pathogens, respectively. The fermentation condition assays showed that glucose and peptone are the most suitable nutrient sources for XS-2, and that the optimal pH value and temperature for fermentation were 7.4 and 28 °C, respectively. Our study indicates that XS-2 has a good antagonistic effect on FHB and lays a theoretical foundation for the application of the strain as a biological agent in the field to control FHB.
Effect of Piriformospora indica-Induced Systemic Resistance and Basal Immunity Against Rhizoctonia cerealis and Fusarium graminearum in Wheat

Abstract:Wheat is among the top 10 and most widely grown crops in the world. However, wheat is often infected with many soil-borne diseases, including sharp eyespot, mainly caused by the necrotrophic fungus Rhizoctonia cerealis , and Fusarium head blight (FHB), caused by Fusarium graminearum , resulting in reduced production. Piriformospora indica is a root endophytic fungus with a wide range of host plants, which increases their growth and tolerance to biotic and abiotic stresses. In this study, the capability of P. indica to protect wheat seedlings against R. cerealis and F. graminearum was investigated at the physiological, biochemical, and molecular levels. Our results showed that P. indica significantly reduced the disease progress on wheat caused by F. graminearum and R. cerealis in vivo , but not showed any antagonistic effect on F. graminearum and R. cerealis in vitro . Additionally, P. indica can induce systemic resistance by elevating H 2 O 2 content, antioxidase activity, relative water content (RWC), and membrane stability index (MSI) compared to the plants only inoculated with F. graminearum or R. cerealis and control. RNA-seq suggested that transcriptome changes caused by F. graminearum were more severe than those caused by R. cerealis. The number of differentially expressed genes (DEGs) in the transcriptome can be reduced by the addition of P. indica: for F. graminearum reduced by 18% and for R. cerealis reduced 58%. The DEGs related to disease resistance, such as WRKY and MAPK, were upregulated by P. indica colonization. The data further revealed that the transcriptional resistance to F. graminearum and R. cerealis mediated by P. indica is quite different.

Abstract:The current study aimed to identify the survival of bio-control bacteria with antifungal activity against Fusarium oxysporum and assess their growth promoting activity in wheat crop field conditions. To evaluate the fungicidal activities of isolated bacteria using the dual culture method, both qualitative and quantitative bioassays were performed. Plant Growth Promoting activities such as Indole 3-Acetic Acid (IAA), phosphate solubilization, Hydrogen cyanide (HCN), and Siderophore production were assessed for three biocontrol bacterial isolates (BCB 07, BCB16, and BCB 83) out of 180 with 70% antagonistic activity against Fusarium oxysporum . Chitinase, protease, and cellulase interaction in isolates was also tested. BCB16 was selected as it had 70% antagonist activity against F. oxysporum but also had the highest PGPR (Plant Growth Promoting Rhizobacteria) traits when compared to the other two isolates. BCB16 was also tested for survival in talc powder and in wheat crop field conditions. Even after 4 months in talc powder, the survival rate remained stable. In a wheat crop field, BCB16 reduced the disease incidence of Fusarium oxysporum by 54.38%. When compared to fungus alone treatment, BCB16 increased average yield by 57% alone and 32% in challenged conditions. BCB16 was identified molecularly using the 16s rRNA gene. Bacillus amyloliquefaciens shared 97% of the deduced sequence. The sequence was submitted to genbank and assigned the accession number OM333889. Bacillus amyloliquefaciens has the potential to be used in the field as an alternative to synthetic fungicides against Fusarium oxysporum .

Wheat rhizosphere dynamics of Trichoderma gamsii A5MH and suppression of a Pythium root rot-Fusarium crown rot disease complex over two consecutive cropping seasons

Abstract:Abstract Aims Determine the wheat rhizosphere competence of Trichoderma gamsii strain A5MH and in planta suppression of the Pythium root and Fusarium crown rot pathogens Globisporangium irregulare and Fusarium pseudograminearum. Methods and results Wheat was continuously cropped (eight years) at a minimum tillage, low growing season rainfall (GSR ≤ 170 mm) site shown as highly conducive to Pythium root and Fusarium crown rots. Root isolation frequency (RIF) and qPCR were used to determine the rhizosphere dynamics of strain A5MH and the target pathogens at tillering, grain harvest, and in postharvest stubble over the final 2 years. Strain A5MH actively colonized the wheat rhizosphere throughout both growing seasons, had high root abundance at harvest [log 4.5 genome copies (GC) g−1] and persisted in standing stubble for at least 293-d postinoculation. Globisporangium irregulare was most abundant in roots at tillering, whereas F. pseudograminearum was only abundant at harvest and up to 9-fold greater in the drier, second year (GSR 105 mm). Strain A5MH decreased RIF of both pathogens by up to 40%, root abundance of G. irregulare by 100-fold, and F. pseudogaminearum by 700-fold, but was ineffective against crown rot in the second year when pathogen abundance was >log 6.0 GC g−1 root. Strain A5MH increased crop emergence and tillering biomass by up to 40%. Conclusions Further trials are required to determine if the A5MH-induced pathogen suppression translates to yield improvements in higher rainfall regions where non-cereal rotations reduce crown rot inoculum.

Minimal impacts on the wheat microbiome when Trichoderma gamsii T6085 is applied as a biocontrol agent to manage fusarium head blight disease

Abstract:Fusarium head blight (FHB) is a major fungal disease that causes severe yield and quality loss in wheat. Biological control can be integrated with other management strategies to control FHB. For this purpose, Trichoderma gamsii strain T6085 is a potential biocontrol agent to limit the infection of F. graminearum and F. culmorum in wheat. However, the possible impacts of T. gamsii T6085 on the broader microbiome associated with the wheat plant are not currently understood. Therefore, we identified bacteria and fungi associated with different wheat tissues, including assessment of their relative abundances and dynamics in response to the application of T6085 and over time, using amplicon sequencing. Residues of the prior year's wheat crop and the current year's wheat spikes were collected at multiple time points, and kernel samples were collected at harvest. DNA was extracted from the collected wheat tissues, and amplicon sequencing was performed to profile microbiomes using 16S v4 rRNA amplicons for bacteria and ITS2 amplicons for fungi. Quantitative PCR was performed to evaluate the absolute abundances of F. graminearum and T. gamsii in different wheat tissues. Disease progression was tracked visually during the growing season, revealing that FHB severity and incidence were significantly reduced when T6085 was applied to wheat spikes at anthesis. However, treatment with T6085 did not lessen the F. graminearum abundance in wheat spikes or kernels. There were substantial changes in F. graminearum abundance over time; in crop residue, pathogen abundance was highest at the initial time point and declined over time, while in wheat spikes, pathogen abundance increased significantly over time. The predominant bacterial taxa in wheat spikes and kernels were Pseudomonas , Enterobacter , and Pantoea , while Alternaria and Fusarium were the dominant fungal groups. Although the microbiome structure changed substantially over time, there were no community-scale rearrangements due to the T6085 treatment. The work suggests several other taxa that could be explored as potential biocontrol agents to integrate with T6085 treatment. However, the timing and the type of T6085 application need to be improved to give more advantages for T6085 to colonize and reduce the F. graminearum inoculum in the field.

Abstract:Fusarium head blight (FHB) is a disease that affects wheat crops worldwide and is caused by Fusarium graminearum. Effective and safe strategies for the prevention and treatment of the disease are very limited. Phomopsis liquidambaris, a universal endophyte, can colonize wheat. Two engineered strains, Phomopsis liquidambaris OE-Chi and IN-Chi, were constructed by transformation with a plasmid and integration of a chitinase into the genome, respectively. The OE-Chi and IN-Chi strains could inhibit the expansion of Fusarium sp. in plate confrontation assays in vitro. Colonization of the OE-Chi strain in wheat showed better effects than colonization of the IN-Chi strain and alleviated the inhibition of wheat growth caused by F. graminearum. The shoot length, root length and fresh weight of infected wheat increased by 164.9%, 115.4%, and 190.7%, respectively, when the plants were inoculated with the OE-Chi strain. The peroxidase (POD) activity in the wheat root increased by 38.0%, and it was maintained at a high level in the shoot, which suggested that the OE-Chi strain could enhance the resistance of wheat to F. graminearum. The root and shoot superoxide dismutase (SOD) activities were decreased by 11.8% and 19.0%, respectively, which may be helpful for colonization by the OE-Chi strain. These results suggested that the Phomopsis liquidambaris OE-Chi strain may be a potential endophyte in the biocontrol of FHB.

Colonization of Endophytic Bacillus amyloliquefaciens CC09 in Wheat Roots Observed under Transmission Electron Microscopy and Its Biocontrol Efficiency against Diseases

Abstract:Bacillus amyloliquefaciens can colonize many plant roots, promote plant growth and suppress pathogenic fungi. However, its colonization process and potential disease control efficacy on wheat roots and rhizosphere are rarely studied. By applying the rifampicin resistant mutant and plate diluting method, we have investigated the colonization capacity and population dynamics of endophytic B. amyloliquefaciens CC09 within wheat root tissues, surface and rhizosphere. Results showed that CC09 strain colonized well within root tissues, surfaces and rhizosphere with a density of 2.7×105, 2.0×105 and 5.5×105 cfu/g, respectively, at 5 d after inoculation, and maintained at these population levels thereafter. Moreover, the strain could colonize in the cortical cells, intercellular spaces, pericycle cells and canal tissues in wheat root tissues and did not show clear adverse impacts on the cell ultra-structures based on the transmission electron microscopy (TEM) observation. In addition, the cultural broth of the strain exhibited 90.7% control efficiency against wheat seedling blight caused by Fusarium graminearum using root-drenching pot assay under the greenhouse conditions. These results suggested that the endophytic B. amyloliquefaciens CC09 has the potential to be developed as biocontrol agent to control wheat disease caused by the pathogenic F. graminearum.

Abstract:Biotic and abiotic stress factors drastically limit plant growth and productivity through changing the physiological, biochemical, and cellular processes. In this study, 100 mM of lead (Pb) was used as an abiotic stress source, while Fusarium graminearum represented a biotic one on wheat plant. Compared to the control, Pb treatment and F. graminearum inoculation led to remarkable reductions in the wheat seedlings leaf area that reached 21 %, and 12.5 %, respectively. Moreover, the current results showed an enhanced activity of Phenylalanine ammonia lyase (PAL) that reached 173 % in the stressed wheat grains and seedlings, decreased mineral contents in N-3, P+3, K+, and Ca+2 in the shoot of wheat seedlings by 25, 42, 23 and 44 %; respectively, substantial increase (79 %) in the total soluble carbohydrates (TSC) and a highly significant reduction (28 %) in the total soluble proteins (TSP), compared with the non-treated control plants. On the other hand, wheat seed priming with weed extracts [i.e., Portulaca oleracea L. (purslane) and Beta vulgaris L. (chard)], or chemical solutions; mainly Calcium (Ca+2) and Salicylic acid (SA) applied exogenously, resulted in a noteworthy increase in the leaf area, compared with the stress treatments. Furthermore, seed priming ameliorated the toxic effects induced by Pb and F. graminearum treatments on the photosynthetic pigments; where it significantly increased the pigments content, while the chlorophyll (Chl) a/b ratio was reduced. Furthermore, priming treatments significantly increased the mineral contents (i.e., N, P, K, and Ca) and counteracted the imposed effects of stress treatments on TSC and TSP. The differential display polymerase chain reaction (DD-PCR) technique was performed to identify the variations in gene expression between the different treatments of wheat plants at three intervals of 7, 14, and 21 d old. This study aimed to investigate the use of plant extracts as potentially effective and environmentally safe green bio-control agents to control the infection of wheat plant by F. graminearum, ameliorate the biotic and abiotic stresses, and compete with the currently used deleterious chemical fungicides in the wheat farms.
Prevalence of Wheat Associated Bacillus spp. and Their Bio-Control Efficacy Against Fusarium Root Rot

Abstract:Bacillus spp. are the most prevalent group of bacteria in nature. Their prevalence depends upon multiple factors, namely, sporulation, antagonism, and production of secondary metabolites. The development of an eco-friendly approach to cope with edible crops diseases is very substantial for humans. In the present study, 658 isolates were obtained from wheat grown in the wheat rice cropping system and tested for their antagonistic activity against four wheat root rot pathogens, namely, Fusarium oxysporum, Fusarium moniliforme, Macrophomina phaseolina , and Rhizoctonia solani . Out of 658, 106 isolates were found antagonistic to either single or multiple fungi. Out of 106 antagonistic bacteria, 62 (23%) were rhizospheric, 28 (14%) were root endospheric, and 16 (9%) were leaf endospheric. Based on mean inhibition against all fungi, the bacterial strains SM-39 and SM-93 showed maximum antagonistic activity. The 16S rRNA gene analysis revealed that most of the antagonistic bacteria exhibiting ≥48% antagonism were Bacillus spp. (98%), except two were Klebsiella spp. (2%). The bacterial strains exhibited phylogenetic lineage with the type strains of the respective genus based on the 16S rRNA gene sequences. In the net house experiment, Bacillus velezensis (SM-39) and Bacillus cabrialesii (SM-93) significantly suppressed Fusarium root rot severity in wheat (42–62%). Plants treated with these strains had lower electrolytic leakage (29–36%), as compared to untreated (44%). Relative water content was much higher (46–58%) for plants inoculated with these strains. These antagonistic strains also considerably colonized the wheat rhizosphere with a cell population of 5.8–6.9.log CFU/g of soil. The rhizosphere of wheat grown in the wheat-rice cropping system could be the potential habitat of effective biocontrol agents.

Abstract:Piriformospora indica , a plant root-colonizing basidiomycete fungus, exhibits strong growth-promoting activity in symbiosis with a broad range of plants. Here, we report the potential of P. indica to improve growth, yield, and disease resistance in wheat in the field. In the present study, P. indica successfully colonized wheat through chlamydospores and formed dense mycelial networks that covered roots. Plants subjected to the seed soaking (SS) treatment with P. indica chlamydospore suspensions enhanced tillering 2.28-fold compared to the non-inoculated wheat in the tillering stage. In addition, P. indica colonization promoted vegetative growth significantly during the three-leaf, tillering, and jointing stages. Moreover, the P. indica -SS-treatment enhanced wheat yield by 16.37 ± 1.63%, by increasing grains per ear and panicle weight and decreased damage to wheat shoot and root architecture markedly, with high field control effects against Fusarium pseudograminearum (81.59 ± 1.32%), Bipolaris sorokiniana (82.19 ± 1.59%), and Rhizoctonia cerealis (75.98 ± 1.36%). Most of the primary metabolites, such as amino acids, nucleotides, and lipids, involved in vegetative reproduction were increased in P. indica -SS-treatment plants, whereas secondary metabolites, such as terpenoids, polyketides, and alkaloids, decreased following P. indica inoculation. The up-regulated processes of protein, carbohydrate, and lipid metabolism indicated that P. indica colonization increased growth, yield, and disease resistance via the acceleration of plant primary metabolism. In conclusion, P. indica improved morphological, physiological, and metabolic substance levels, and further promoted its growth, yield, and disease resistance in wheat.

Abstract:Background: Fusarium head blight (FHB), primarily caused by Fusarium graminearum, is a significant threat to global wheat production. It results in extensive yield losses and mycotoxin contamination. This study aims to evaluate various fungicides in vitro to determine their effectiveness in controlling FHB. Through a comprehensive analysis of the fungicides’ in vitro efficacy against F. graminearum, this research endeavours to establish a connection between laboratory observations and real-world field applications. Methods: The current investigation was conducted in the Plant Pathology laboratory of CSKHPKV, Palampur, during 2021-2022, wherein, nine fungicides viz., trifloxystrobin 25% + tebuconazole 50% (Nativo 75WG), carbendazim 12% + mancozeb 63% WP (SAAF), difenoconazole 25 EC (Score), carbendazim 50 WP (Bavistin), Propiconazole 25 EC (Tilt), tebuconazole 250 EC (Folicur), mancozeb 75WP (Indofil), copper oxychloride 50 WP (Blitox) and carboxin 37.5% +thiram 37.5% WS (Vitavax power) were evaluated at different concentrations against F. graminearum. In the in vitro assay, F. graminearum was subjected to varying concentrations of fungicides, ranging from low to high, to determine their inhibitory effects on the growth of mycelia, which are thread-like structures that form the vegetative part of fungi. The assay involved carefully monitoring the development of F. graminearum under different conditions to obtain accurate results. Result: The study’s results revealed that carbendazim 50 WP, trifloxystrobin 25% + tebuconazole 50% WG, and tebuconazole 250 EC at concentrations of 100 ppm or higher were the most effective in inhibiting the growth of mycelium. The varying degrees of effectiveness among the fungicides tested indicate that F. graminearum responds differently to various chemical treatments. The findings from this in vitro evaluation will provide valuable data for the ongoing efforts to develop effective and sustainable fungicide-based strategies to combat Fusarium head blight in wheat crops.

Abstract:Endophytic fungal-based biopesticides are sustainable and ecologically-friendly biocontrol agents of several pests and diseases. However, their potential in managing tomato fusarium wilt disease (FWD) remains unexploited. This study therefore evaluated effectiveness of nine fungal isolates against tomato fusarium wilt pathogen, Fusarium oxysporum f. sp. lycopersici (FOL) in vitro using dual culture and co-culture assays. The efficacy of three potent endophytes that inhibited the pathogen in vitro was assessed against FWD incidence, severity, and ability to enhance growth and yield of tomatoes in planta. The ability of endophytically-colonized tomato (Solanum lycopersicum L.) plants to systemically defend themselves upon exposure to FOL were also assessed through defence genes expression using qPCR. In vitro assays showed that endophytes inhibited and suppressed FOL mycelial growth better than entomopathogenic fungi (EPF). Endophytes Trichoderma asperellum M2RT4, Hypocrea lixii F3ST1, Trichoderma harzianum KF2R41, and Trichoderma atroviride ICIPE 710 had the highest (68.84-99.61%) suppression and FOL radial growth inhibition rates compared to EPF which exhibited lowest (27.05-40.63%) inhibition rates. Endophytes T. asperellum M2RT4, H. lixii F3ST1 and T. harzianum KF2R41 colonized all tomato plant parts. During the in planta experiment, endophytically-colonized and FOL-infected tomato plants showed significant reduction of FWD incidence and severity compared to non-inoculated plants. In addition, these endophytes contributed to improved growth promotion parameters and yield. Moreover, there was significantly higher expression of tomato defence genes in T. asperellum M2RT4 colonized than in un-inoculated tomato plants. These findings demonstrated that H. lixii F3ST1 and T. asperellum M2RT4 are effective biocontrol agents against FWD and could sustainably mitigate tomato yield losses associated with fusarium wilt.

Abstract:Fusarium graminearum can cause Giberella Ear Rot (GER) and seedling blight in maize, resulting in major yield losses. Besides GER, the infected grains are consequently contaminated with multiple mycotoxins of F. graminearum. Zearalenone and trichothecenes, such as deoxynivalenol and its acetylated forms, are among the major mycotoxins associated with F. graminearum infection in maize. In the current work, we explored the effect of the endophytic fungal genera of Epicoccum and Sordaria, to control F. graminearum infection in comparative trials with Piriformospora spp., an elusive endophytic genus. Furthermore, we investigated the effect of these endophytes on zearalenone, deoxynivalenol, and 15-acetyldeoxynivalenol levels using in vitro and in planta assays. As plants are endowed with several detoxification mechanisms comprising e.g., glucosylation of trichothecenes, the effect of the isolated fungal endophytes on the deoxynivalenol-3-glucoside level was also assessed. In general, results showed a considerable variability in the antifungal activity, both among species and among isolates within one species. Additionally, the effect on mycotoxin levels was variable, and not necessarily related to the antifungal activity except for zearalenone levels which were consistently reduced by the endophytes. These results highlight the great potential of certain endophytic fungal strains as new biocontrol agents in agricultural science.
Piriformospora indica Increases Resistance to Fusarium pseudograminearum in Wheat by Inducing Phenylpropanoid Pathway

Abstract:Fusarium crown rot (FCR), mainly caused by Fusarium pseudograminearum, not only seriously threatens the yield and quality of wheat, but also endangers the health and safety of humans and livestock. Piriformospora indica is a root endophytic fungus that colonizes plant roots extensively and can effectively promote plant growth and improve plant resistance to biotic and abiotic stresses. In this study, the mechanism of FCR resistance mediated by P. indica in wheat was revealed from the phenylpropanoid metabolic pathway. The results showed that the colonization of P. indica significantly reduced the progression of wheat disease, the amount of F. pseudograminearum colonization, and the content of deoxynivalenol (DON) in wheat roots. RNA-seq suggested that P. indica colonization could reduce the number of differentially expressed genes (DEGs) in the transcriptome caused by F. pseudograminearum infection. The DEGs induced by the colonization of P. indica were partially enriched in phenylpropanoid biosynthesis. Transcriptome sequencing and qPCR indicated that the colonization of P. indica up-regulated the expression of genes involved in the phenylpropanoid biosynthesis pathway. The metabolome analysis indicated that the colonization of P. indica increased the metabolites’ accumulation in the phenylpropanoid biosynthesis. Consistent with transcriptome and metabolomic analysis, microscopic observations showed enhanced lignin accumulation in the roots of the Piri and Piri+Fp lines, most likely contributing to the arrested infection by F. pseudograminearum. These results suggested that P. indica increased resistance to F. pseudograminearum in wheat by inducing the phenylpropanoid pathway.

Abstract:Endophytic fungi have recently garnered significant attention as next-generation bioinoculants due to their plausible role in ameliorating abiotic and biotic stresses. This adaptation is achieved via various signalling molecules and mechanisms established by these symbionts with their hosts. The present study screened 61 endophytic isolates of culturable mycobiome associated with wheat variety PBW725 during their crop cycle. Three endophytic isolates exhibited a minimum reduction in their growth and maximum biomass production during the drought stress developed using polyethylene glycol 6000. Further, these isolates also exhibited plant growth promoting properties by virtue of the production of indole acetic acid, gibberellic acid and ammonia. These isolates also exhibited the propensity to solubilise phosphate and zinc, produce siderophores and further exhibit extracellular enzymatic activities, contributing to plants' adaptability to abiotic stresses. The best isolate amongst the three was #5TAKL-3a, identified as Penicillium citrinum based on multilocus phylogenetic analysis. The isolate as a bioinoculant enhances various biochemical and physiological properties in planta . Hence our studies indicate that Penicillium citrinum #5TAKL-3a is a potential candidate bioinoculant for field trials to improve the adaptability of the wheat plant under drought stress.
