Abstract:Tissue engineering offers an alternative method that can overcome some of the existing drawbacks of current articular defect repair methods because articular cartilage has a limited capacity to respond to injury. The solution may lie in the design of a three-dimensional load-bearing scaffold. Here we describe the tissue engineering of an osteochondral construct by coculturing osteogenic cells and chondrogenic cells on a three-dimenisonal load-bearing bioresorbable polymer scaffold. Porous polycaprolactone scaffolds were designed and fabricated via fused deposition modeling. Osteogenic cells were seeded and precultured in one-half of the partitioned scaffolds. Chondrogenic cells were later seeded into the other half. The cell-seeded scaffolds were cultured in a coculture medium. Both cell types proliferated, migrated, linked in their scaffold compartments, and integrated at the interface. Osteoblasts; and chondrocytes produced different extracellular matrices in each scaffold compartment. Mineralized nodules deposited in the osteogenic cell seeded compartment. High osteocalcin was detected in precultured osteogenic cell supernatant and high alkaline phosphatase was detected in the coculture supernatant of osteochondral constructs. This study suggests that a tissue-engineered osteochondral construct with a three-dimenisonal polycaprolactone scaffold has the potential for osteochondral defect repair.

Abstract:Extracellular matrix (ECM) derived from decellularized tissues and organs has been used as a biological scaffold in a variety of pre-clinical and clinical applications. However, the lack of mechanical properties and shape controllability is a drawback. In contrast, synthetic polymers can be easily engineered with good mechanical performance, but they have limited biological functionality. In this study, we explored a new method to electrospun cartilage-derived extracellular matrix (cECM) and polycaprolactone (PCL) composite nanofibrous membranes. The cartilage is a compact tissue for which electrospinning is difficult. To overcome this problem, the cartilage was sliced into pieces, milled into powders, and digested into a looser structure. The cECM/PCL (mass ratio 50:50) hybrid nanofibers appeared to be smooth, thinner, and uniform with enhanced mechanical properties and wettability compared to the electrospun PCL. Meanwhile, the presence of cECM in the cECM/PCL nanofibrous membranes significantly promoted chondrocyte proliferation in vitro and facilitated cartilage regeneration in vivo. All these results indicate that the cECM/PCL nanofibrous membranes fabricated with good mechanical properties and biocompatibility may represent a promising scaffold for cartilage regeneration. Moreover, this work may offer a convenient and cost-effective method to synthesize ECM-based hybrid nanofibrous scaffolds for other tissue applications.

Abstract:The main aim of this paper is to achieve the suitable SA-GEL (sodium alginate and gelatin) porous cartilage scaffold by 3D printing technology with optimal prediction parameters. Firstly, the characteristics of SA-GEL were analyzed, the influence of calcium chloride on the gel was explored, and the optimal cross-linking concentration and gelation temperature were determined. Secondly, a prediction model of the extrusion line width of SA-GEL was established, in which the printing pressure, the moving speed of the needle and the fiber interval were the important parameters affecting the printing performance of the SA-GEL composite material. Thirdly, the SA-GEL composite scaffolds were printed on the Bio-plotter platform, the C5.18 chondrocytes cells were cultured in the SA-GEL biomaterial scaffold, and the results show that the cells could survive well. These results show that, under the control of the printing parameters pressure 1.8 bar, moving speed 10.7 mm/s and the internal structure parameters of the scaffold is 0/45-1.2 (Printing interval: 1.2 mm, angle value: 45 degree), SA-GEL scaffold printing results can be obtained which have good mechanical properties and biocompatibility.
3D-Printed Nano-Structural Poly(Epsilon-Caprolactone) (pcl) Scaffolds for Corneal Stromal Regeneration

Abstract:Purpose : Regeneration of corneal stroma is believed to be among the greatest challenges in corneal tissue engineering. The aim of this study is to investigate the effect of a 3D-printed nano-structural poly(e-caprolactone) (PCL) scaffold on the material-cellular interaction, and to evaluate the efficiency for the corneal stroma regeneration in vitro. Methods : Two nano-structural scaffolds of different design and fiber distribution were fabricated by melt electrospinning 3D printing method using PCL. Human keratocytes were isolated from collagenase-digested limbal stromal tissue and seeded on the 3D-printed PCL scaffold. Cell morphology on scaffolds was studied by confocal microscopy, and cell proliferation was analyzed by PrestoBlue assay. Extracellular matrix(ECM) produced by keratocytes on the scaffolds was characterized by scanning electron microscope(SEM) and two-photon fluorescent microscopy. Results : The 3D-printed PCL scaffolds exhibited different micro-structure under SEM. As revealed in Figure.1, scaffold on the left presented offset fiber distribution, while the other one showed a fiber distribution at regular intervals. SEM and two-photon fluorescent microscopy demonstrated that keratocytes tended to grow along the fibrous network, and spread well on both scaffolds. Collagenous ECM produced by cells on the regular-fibrous scaffold was more abundant than on the offset-fibrous scaffold, forming a relatively thicker cross-section (Figure 2). Compared to a collagen fibrillar network secreted on regular-fibrous scaffolds, a lamellar ultrastructural ECM composed of aligned collagen was remodeled on offset-fibrous scaffolds, which was similar to native cornea stroma. Immunofluorescence staining showed that regular-fibrous scaffolds induced less secretion of type I collagen (COLI) and type V collagen (COLV) than offset-fibrous scaffolds. Conclusions : Our study showed that the fiber composition and topography of 3D-printed nanofibrous PCL scaffolds have a significant effect on the cell behavior through the material-cellular interaction. The refined design of nano-structure by melt electrospinning method stimulates self-organization and self-adaption of ECM, which helps mimicking native tissue microenvironments and offering the potential as cornea stroma regeneration substitutes.
Multi-material 3D bioprinting of porous constructs for cartilage regeneration

Abstract:The current gold standard for nasal reconstruction after rhinectomy or severe trauma includes transposition of autologous cartilage grafts in conjunction with coverage using an autologous skin flap. Harvesting autologous cartilage requires a major additional procedure that may create donor site morbidity. Major nasal reconstruction also requires sculpting autologous cartilages to form a cartilage framework, which is complex, highly skill-demanding and very time consuming. These limitations have prompted facial reconstructive surgeons to explore different techniques such as tissue engineered cartilage. This work explores the use of multi-material 3D bioprinting with chondrocyte-laden gelatin methacrylate (GelMA) and polycaprolactone (PCL) to fabricate constructs that can potentially be used for nasal reconstruction. In this study, we have investigated the effect of 3D manufacturing parameters including temperature, needle gauge, UV exposure time, and cell carrier formulation (GelMA) on the viability and functionality of chondrocytes in bioprinted constructs. Furthermore, we printed chondrocyte-laden GelMA and PCL into composite constructs to combine biological and mechanical properties. It was found that 20% w/v GelMA was the best concentration for the 3D bioprinting of the chondrocytes without comprising the scaffold's porous structure and cell functionality. In addition, the 3D bioprinted constructs showed neocartilage formation and similar mechanical properties to nasal alar cartilage after a 50-day culture period. Neocartilage formation was also observed in the composite constructs evidenced by the presence of glycosaminoglycans and collagen type II. This study shows the feasibility of manufacturing neocartilage using chondrocytes/GelMA/PCL 3D bioprinted porous constructs which could be applied as a method for fabricating implants for nose reconstruction.

Abstract:An epidemiological survey was conducted of the oral conditions in the region of lle de France of a representative sample of 2,062 children aged 6 to 15 years. The sample was selected among the population of Paris and the departments of Hauts de Seine, Seine St-Denis, Val-de-Marne, Val d'Oise and Seine et Marne; 1,962 children came from urban zones. The mean DMFT increased with age and was 0.98, 4.46 and 6.6 at the ages of 6, 9 and 12 years respectively. At 6 years of age, 65.9% of the children were free of caries in their permanent teeth decreasing to 17.4% and 11.9% at the ages of 12 and 15 years respectively. The pits and fissures were the most frequent sites of caries. The highest mean values of plaque and gingival indices were observed at the age of 11 years.
An additive manufacturing-based PCL-alginate-chondrocyte bioprinted scaffold for cartilage tissue engineering

Abstract:Regenerative medicine is targeted to improve, restore or replace damaged tissues or organs using a combination of cells, materials and growth factors. Both tissue engineering and developmental biology currently deal with the process of tissue self-assembly and extracellular matrix (ECM) deposition. In this investigation, additive manufacturing (AM) with a multihead deposition system (MHDS) was used to fabricate three-dimensional (3D) cell-printed scaffolds using layer-by-layer (LBL) deposition of polycaprolactone (PCL) and chondrocyte cell-encapsulated alginate hydrogel. Appropriate cell dispensing conditions and optimum alginate concentrations for maintaining cell viability were determined. In vitro cell-based biochemical assays were performed to determine glycosaminoglycans (GAGs), DNA and total collagen contents from different PCL-alginate gel constructs. PCL-alginate gels containing transforming growth factor-β (TGFβ) showed higher ECM formation. The 3D cell-printed scaffolds of PCL-alginate gel were implanted in the dorsal subcutaneous spaces of female nude mice. Histochemical [Alcian blue and haematoxylin and eosin (H&E) staining] and immunohistochemical (type II collagen) analyses of the retrieved implants after 4 weeks revealed enhanced cartilage tissue and type II collagen fibril formation in the PCL-alginate gel (+TGFβ) hybrid scaffold. In conclusion, we present an innovative cell-printed scaffold for cartilage regeneration fabricated by an advanced bioprinting technology.

Abstract:Nowadays, cartilage tissue engineering (CTE) is considered important due to lack of repair of cartilaginous lesions and the absence of appropriate methods for treatment. In this study, polycaprolactone (PCL) scaffolds were fabricated by three-dimensional (3D) printing and were then coated with fibrin (F) and acellular solubilized extracellular matrix (ECM). After extracting adipose-derived stem cells (ADSCs), 3D-printed scaffolds were characterized and compared to hydrogel groups. After inducing the chondrogenic differentiation in the presence of Piascledine and comparing it with TGF-β3 for 28 days, the expression of genes involved in chondrogenesis (AGG, COLII) and the expression of the hypertrophic gene (COLX) were examined by real-time PCR. The expression of proteins COLII and COLX was also determined by immunohistochemistry. Glycosaminoglycan was measured by toluidine blue staining. 3D-printed scaffolds clearly improved cell proliferation, viability, water absorption and compressive strength compared to the hydrogel groups. Moreover, the use of compounds such as ECM and Piascledine in the process of ADSCs chondrogenesis induction increased cartilage-specific markers and decreased the hypertrophic marker compared to TGF-β3. In Piascledine groups, the expression of COLL II protein, COLL II and Aggrecan genes, and the amount of glycosaminoglycan showed a significant increase in the PCL/F/ECM compared to the PCL and PCL/F groups.

Abstract:Abstract Cartilage has limited self-repair ability due to its avascular, alymphatic and aneural features. The combination of three-dimensional (3D) printing and tissue engineering provides an up-and-coming approach to address this issue. Here, we designed and fabricated a tri-layered (superficial layer (SL), middle layer (ML) and deep layer (DL)) stratified scaffold, inspired by the architecture of collagen fibers in native cartilage tissue. The scaffold was composed of 3D printed depth-dependent gradient poly(ε-caprolactone) (PCL) impregnated with methacrylated alginate (ALMA), and its morphological analysis and mechanical properties were tested. To prove the feasibility of the composite scaffolds for cartilage regeneration, the viability, proliferation, collagen deposition and chondrogenic differentiation of embedded rat bone marrow mesenchymal stem cells (BMSCs) in the scaffolds were assessed by Live/dead assay, CCK-8, DNA content, cell morphology, immunofluorescence and real-time reverse transcription polymerase chain reaction. BMSCs-loaded gradient PCL/ALMA scaffolds showed excellent cell survival, cell proliferation, cell morphology, collagen II deposition and hopeful chondrogenic differentiation compared with three individual-layer scaffolds. Hence, our study demonstrates the potential use of the gradient PCL/ALMA construct for enhanced cartilage tissue engineering.

Abstract:Introduction The use of scaffolds in tissue engineering is becoming increasingly important as solutions need to be found to preserve human tissues such as bone or cartilage. Various factors, including cells, biomaterials, cell and tissue culture conditions, play a crucial role in tissue engineering. The in vivo environment of the cells exerts complex stimuli on the cells, thereby directly influencing cell behavior, including proliferation and differentiation. Therefore, to create suitable replacement or regeneration procedures for human tissues, the conditions of the cells’ natural environment should be well mimicked. Therefore, current research is trying to develop 3-dimensional scaffolds (scaffolds) that can elicit appropriate cellular responses and thus help the body regenerate or replace tissues. In this work, scaffolds were printed from the biomaterial polycaprolactone (PCL) on a 3D bioplotter. Biocompatibility testing was used to determine whether the printed scaffolds were suitable for use in tissue engineering. Material and Methods An Envisiontec 3D bioplotter was used to fabricate the scaffolds. For better cell-scaffold interaction, the printed polycaprolactone scaffolds were coated with type-I collagen. Three different cell types were then cultured on the scaffolds and various tests were used to investigate the biocompatibility of the scaffolds. Results Reproducible scaffolds could be printed from polycaprolactone. In addition, a coating process with collagen was developed, which significantly improved the cell-scaffold interaction. Biocompatibility tests showed that the PCL-collagen scaffolds are suitable for use with cells. The cells adhered to the surface of the scaffolds and as a result extensive cell growth was observed on the scaffolds. The inner part of the scaffolds, however, remained largely uninhabited. In the cytotoxicity studies, it was found that toxicity below 20% was present in some experimental runs. The determination of the compressive strength by means of the universal testing machine Z005 by ZWICK according to DIN EN ISO 604 of the scaffolds resulted in a value of 68.49 ± 0.47 MPa.
3D printed PCL/GelMA biphasic scaffold boosts cartilage regeneration using co-culture of mesenchymal stem cells and chondrocytes: In vivo study

Abstract:Cartilage injury is difficult to self-repair, which poses a challenge to scaffold design and selection of seeded cells for cartilage tissue engineering. In this study, we presented a biphasic scaffold consisting of a cells-encapsulated biodegradable gelatin methacrylate (GelMA) hydrogel on the upper layer and a macroporous poly(ε-caprolactone) (PCL) scaffold filled with cells-encapsulated GelMA hydrogel on the lower layer to support cartilage regeneration using co-culture of bone marrow mesenchyml stem cells (BMSCs) and costal chondrocytes (CChon). We determined that the synergistic effect of BMSCs and costal chondrocytes was sufficient to induce chondrogenesis, but cartilage regeneration was better under TGF-β3 supplement. Memorably, when implanted in a rat osteochondral defect model for 12 weeks, the PCL/GelMA biphasic scaffolds with cells co-culture showed excellent cartilage regenerative capability, and enabled the Young's modulus of engineered cartilage comparable to that of native cartilage. Furthermore, the implantation of biphasic scaffold resulted in less mechanical pain than cartilage defects by gait analysis. Together, both in vitro and in vivo results demonstrated that the PCL/GelMA biphasic scaffold with co-culture of BMSCs and chondrocytes is a promising scaffold for cartilage regeneration, which pave a new way to solve the key issues of clinical cartilage repair.

Abstract:Many forms of heart disease are associated with the loss of cardiomyocytes both via apoptosis or necrosis, and despite the recent identification of resident cardiac stem cells, the native capacity for renewal and repair is inadequate. Cell transplantation strategies have emerged as a potential therapeutic approach for repairing injured myocardium. Many different cell types including embryonic stem cells have been transplanted in myocardial infarction (MI) models with resulting improvement in myocardial function. Here, we review the current state of knowledge with regard to the potential of embryonic stem (ES) cells to differentiate into cardiomyocytes in the embryonic stem cell derived-embryoid body (EB) in vitro system as well as for myocardial regeneration following myocardial infarction.
Extrusion-based 3D printing of osteoinductive scaffolds with a spongiosa-inspired structure

Abstract:Critical-sized bone defects resulting from trauma, inflammation, and tumor resections are individual in their size and shape. Implants for the treatment of such defects have to consider biomechanical and biomedical factors, as well as the individual conditions within the implantation site. In this context, 3D printing technologies offer new possibilities to design and produce patient-specific implants reflecting the outer shape and internal structure of the replaced bone tissue. The selection or modification of materials used in 3D printing enables the adaption of the implant, by enhancing the osteoinductive or biomechanical properties. In this study, scaffolds with bone spongiosa-inspired structure for extrusion-based 3D printing were generated. The computer aided design process resulted in an up scaled and simplified version of the bone spongiosa. To enhance the osteoinductive properties of the 3D printed construct, polycaprolactone (PCL) was combined with 20% (wt) calcium phosphate nano powder (CaP). The implants were designed in form of a ring structure and revealed an irregular and interconnected porous structure with a calculated porosity of 35.2% and a compression strength within the range of the natural cancellous bone. The implants were assessed in terms of biocompatibility and osteoinductivity using the osteosarcoma cell line MG63 and patient-derived mesenchymal stem cells in selected experiments. Cell growth and differentiation over 14 days were monitored using confocal laser scanning microscopy, scanning electron microscopy, deoxyribonucleic acid (DNA) quantification, gene expression analysis, and quantitative assessment of calcification. MG63 cells and human mesenchymal stem cells (hMSC) adhered to the printed implants and revealed a typical elongated morphology as indicated by microscopy. Using DNA quantification, no differences for PCL or PCL-CaP in the initial adhesion of MG63 cells were observed, while the PCL-based scaffolds favored cell proliferation in the early phases of culture up to 7 days. In contrast, on PCL-CaP, cell proliferation for MG63 cells was not evident, while data from PCR and the levels of calcification, or alkaline phosphatase activity, indicated osteogenic differentiation within the PCL-CaP constructs over time. For hMSC, the highest levels in the total calcium content were observed for the PCL-CaP constructs, thus underlining the osteoinductive properties.

Abstract:A novel sandwich-like composite 3D model integrated with deposited gelatin film and printed polycaprolactone (PCL)/polyvinyl pyrrolidone (PVP) scaffold is proposed for 3D cartilage cell culture. The 3D model includes three layers, top scaffold, middle gelatin film, and basal slide. The printed scaffold layer mimicking extracellular matrix is employed to provide 3D regeneration architecture; gelatin film layer is used for fixing printed scaffold and catching falling cells. Electrospray and electrohydrodynamic jet printing technologies are combined to construct sandwich-like composite 3D model. The blended constructing processes are investigated thoroughly both theoretically and experimentally. The retained charges on the gelatin film are influenced the electric field distribution and jet behaviors during printing. The characteristics of gelatin film and composite scaffold are studied. The thickness, surface roughness, and planeness of one layer of deposited gelatin film are 400 nm, 22 nm, and 2.27 mu m; the size of printed PCL/PVP composite fiber is 10 mu m. The fabricated sandwich-like composite 3D model is proved to facilitate adhesion and ingrowth of cartilage cells.
HBC-nanofiber hydrogel scaffolds with 3D printed internal microchannels for enhanced cartilage differentiation

Abstract:Articular cartilage injuries are a major orthopedic problem. Cartilage repair is a long-standing challenge due to the limited self-regenerative capability of cartilage. Tissue engineering offers a new and effective approach to cartilage repair. We report herein the fabrication of 3D scaffolds that mimic the native structure of cartilage, by first preparing poly(lactic-co-glycolic acid) (PLGA) electrospun nanofiber incorporated hydroxybutyl chitosan (HBC) hydrogels (HBC-NF hydrogels) and then injecting the hydrogels into a 3D printed poly(ε-caprolactone) (PCL) framework with internal microchannels for improved mechanical support and substance exchange. The thus-obtained HBC-NF hydrogels exhibited outstanding gelation properties with a gelling time of no more than 15 s at 37 °C. With the incorporation of the nanofibers, human mesenchymal stem cells (hMSCs) showed good proliferation in the HBC-NF hydrogels. The relative gene expression levels for mesenchymal condensation and matrix deposition significantly increased in the HBC-NF hydrogels due to the addition of the nanofibers, suggesting substantially enhanced cartilage differentiation. Furthermore, the injection of the HBC-NF hydrogels into the 3D printing PCL framework led to the formation of 3D scaffolds with significantly improved mechanical performance. More importantly, the construction of regulable internal microchannels for cell growth and the exchange of nutrients and waste products were achieved via co-printing of PCL and a sacrificial material, Pluronic F-127. The PCL reinforced HBC-NF hydrogel scaffolds with internal microchannels showed enhanced chondrogenesis and mechanical properties in vivo. In summary, the current work has demonstrated that PCL framework reinforced HBC-NF hydrogels with tunable internal microchannels provide an ideal biomimetic microenvironment for the growth and cartilage differentiation of hMSCs, therefore holding promise for potential applications in cartilage tissue engineering.

Abstract:Natural hydrogels are widely employed in tissue engineering and have excellent biodegradability and biocompatibility. Unfortunately, the utilization of such hydrogels in the field of three‐dimensional (3D) printing nasal cartilage is constrained by their subpar mechanical characteristics. In this study, we provide a multicrosslinked network hybrid ink made of photocurable gelatin, hyaluronic acid, and acrylamide (AM). The ink may be processed into intricate 3D hydrogel structures with good biocompatibility and high stiffness properties using 3D printing technology based on digital light processing (DLP), including intricate shapes resembling noses. By varying the AM content, the mechanical behavior and biocompatibility of the hydrogels can be adjusted. In comparison to the gelatin methacryloyl (GelMA)/hyaluronic acid methacryloyl (HAMA) hydrogel, adding AM considerably enhances the hydrogel's mechanical properties while also enhancing printing quality. Meanwhile, the biocompatibility of the multicrosslinked network hydrogels and the development of cartilage were assessed using neonatal Sprague–Dawley (SD) rat chondrocytes (CChons). Cells sown on the hydrogels considerably multiplied after 7 days of culture and kept up the expression of particular proteins. Together, our findings point to GelMA/HAMA/polyacrylamide (PAM) hydrogel as a potential material for nasal cartilage restoration. The photocuring multicrosslinked network ink composed of appropriate proportions of GelMA/HAMA/PAM is very suitable for DLP 3D printing and will play an important role in the construction of nasal cartilage, ear cartilage, articular cartilage, and other tissues and organs in the future. Notably, previous studies have not explored the application of 3D‐printed GelMA/HAMA/PAM hydrogels for nasal cartilage regeneration.

Abstract:Polycaprolactone (PCL) scaffolds have demonstrated an effectiveness in articular cartilage regeneration due to their biomechanical properties. On the other hand, alginate hydrogels generate a 3D environment with great chondrogenic potential. Our aim is to generate a mixed PCL/alginate scaffold that combines the chondrogenic properties of the two biomaterials. Porous PCL scaffolds were manufactured using a modified salt-leaching method and embedded in a culture medium or alginate in the presence or absence of chondrocytes. The chondrogenic capacity was studied in vitro. Type II collagen and aggrecan were measured by immunofluorescence, cell morphology by F-actin fluorescence staining and gene expression of COL1A1, COL2A1, ACAN, COL10A1, VEGF, RUNX1 and SOX6 by reverse transcription polymerase chain reaction (RT-PCR). The biocompatibility of the scaffolds was determined in vivo using athymic nude mice and assessed by histopathological and morphometric analysis. Alginate improved the chondrogenic potential of PCL in vitro by increasing the expression of type II collagen and aggrecan, as well as other markers related to chondrogenesis. All scaffolds showed good biocompatibility in the in vivo model. The presence of cells in the scaffolds induced an increase in vascularization of the PCL/alginate scaffolds. The results presented here reinforce the benefits of the combined use of PCL and alginate for the regeneration of articular cartilage.

Abstract:In this study, we aimed to improve the hydrophilicity, cellular activities and osteogenesis of polycaprolactone (PCL) scaffolds by adding gelatin (Gel) and nano-hydroxyapatite (n-HA). We fabricated biocomposite scaffolds using the extrusion deposition 3 D printing method. The physical and biological properties of the composite scaffold were evaluated. Scanning electron microscopy (SEM) of the fabricated composite scaffolds revealed that the Gel and n-HA particles were uniformly embedded in the internal PCL. The composite scaffold (PCL + Gel + n-HA) showed dramatically improved mechanical properties. The porosity, hydrophilicity and hydrophobicity and ability to promote the attachment and proliferation of osteoblasts were then compared between the pure PCL and composite scaffolds. The PCL + Gel + n-HA scaffold was implanted in the rabbits for 5 weeks, and the results of H&E and Masson staining analyses demonstrated that the scaffold had good biocompatibility and good osteogenesis. The strategy developed in this study has promising prospects for future clinical applications of enhancing bone defect repair.Graphical
