Abstract:PurposeThis study was aimed to identify the expression pattern of vascular endothelial growth factor (VEGF) in non-small cell lung cancer (NSCLC) and to explore its potential correlation with the progression of NSCLC.MethodsGene expression profile GSE39345 was downloaded from the Gene Expression Omnibus database. Twenty healthy controls and 32 NSCLC samples before chemotherapy were analyzed to identify the differentially expressed genes (DEGs). Then pathway enrichment analysis of the DEGs was performed and protein-protein interaction networks were constructed. Particularly, VEGF genes and the VEGF signaling pathway were analyzed. The sub-network was constructed followed by functional enrichment analysis.ResultsTotal 1666 up-regulated and 1542 down-regulated DEGs were identified. The down-regulated DEGs were mainly enriched in the pathways associated with cancer. VEGFA and VEGFB were found to be the initiating factor of VEGF signaling pathway. In addition, in the epidermal growth factor receptor (EGFR), VEGFA and VEGFB associated sub-network, kinase insert domain receptor (KDR), fibronectin 1 (FN1), transforming growth factor beta induced (TGFBI) and proliferating cell nuclear antigen (PCNA) were found to interact with at least two of the three hub genes. The DEGs in this sub-network were mainly enriched in Gene Ontology terms related to cell proliferation.ConclusionEGFR, KDR, FN1, TGFBI and PCNA may interact with VEGFA to play important roles in NSCLC tumorigenesis. These genes and corresponding proteins may have the potential to be used as the targets for either diagnosis or treatment of patients with NSCLC.

Abstract:Acute myeloid leukemia (AML) is a disease of the hematopoietic progenitor cells associated with heterogeneous clonal proliferation. Vascular endothelial growth factor (VEGF) and its receptors play important roles in the regulation of angiogenesis during physiological and pathological processes. It is thought that AML cells have an autocrine VEGF pathway that contributes to the development and progression of AML. In addition, growing evidence has suggested that numerous microRNAs are involved in AML. The present study aimed to investigate the relationship between VEGF dysregulation and microRNA profiles in AML cells and patients. VEGF-overexpressing and VEGF-knockdown leukemia cells were constructed and changes in the patterns of microRNA expression were analyzed using a microRNA array. Subsequently, mononuclear cells from the blood of patients with AML showing high or low expression levels of VEGF were obtained and were used to assess the patterns of microRNA expression by reverse transcription-quantitative polymerase chain reaction. The results of the present study suggested that downregulation of VEGF markedly altered the profile of microRNAs in AML cells, while upregulation of VEGF did not. Examination of clinical samples from patients with AML showed that several microRNAs were closely associated with the expression level of VEGF, including miR-20a, miR-93, miR-16-5p, miR-17-5p, miR-124-5p and miR-17-3p. These results suggested that VEGF may be a pivotal protein that can both receive and initiate signals in leukemia cells.
Effects of NANOG Expression on Proliferation and Cell Cycle of Acute Lymphoblastic Leukemia Cells

Abstract:Objective To explore the expression of NANOG gene in acute lymphoblastic leukemia (ALL) cell lines and the effect of down-regulation of NANOG on the proliferation and cell cycle of ALL cells.Methods Three kinds of ALL cell line MOLT-4,CCRF-HSB2 and Jurkat were chosen as study object.The expression of NANOG was detected by reverse transcription-polymerase chain reaction (RTPCR) and Western blot in MOLT-4,CCRF-HSB2 and Jurkat cells.The lentiviral vector (pLB-shNANOG-1 and pLB-shNANOG-2) carrying NANOG specific shRNA were constructed.After infection of MOLT-4 cells with the lentivirus constructs,green fluorescent protein positive cells were harvested by flow cytometry as interference group.With empty plasmid (pLB-sh control) infected MOLT-4 cells and normal cells were used as wild-type group and control group.The efficiency of RNA interference was detected by real-time quantitative PCR and Western blot.Cell proliferation was evaluated using CCK-8 cell counting kit.Cell cycle was analyzed by flow cytometry.The expression of p53-related gene was detected by real-time quantitative PCR.Results The virus titers were (1.83-3.12)×108 IU/mL.Each interfering sequence cloud stably down-regulate the expression of NANOG.Analysis of cell proliferation indicated that the MOLT-4 cells expressing NANOG shRNA grew significantly slower than wild-type cells and control cells,and this difference became more obvious after 72 hours (P＜0.001).Cell cycle analysis indicated that the percentage of S-phase cells in wild-type or control group cells was obviously increased when compared to MOLT-4 cells expressing NANOG shRNAs (P＜0.01).The percentage of MOLT-4 cells expressing NANOG shRNAs in the G0/G1-phase was higher than in wild-type cells and control cells (P＜0.001),while the percentage of cells in the G2/M-phase was not statistical different among these cells (P＞0.05).The expression of gene TP53 and CDKN1A were increased in groups of shNANOG-1 and shNANOG-2 than that of groups of control and wild-type (P＜0.05).But the expression of gene MDM2 and CCND1 was decreased (P＜0.05).Conclusions NANOG is expressed in some human ALL cell lines.Down-regulation of NANOG could inhibit cell proliferation and arrest in the cell cycle of G0/G1 phase through a MDM2-TP53 CDKN1A pathway in leukemic cells.
Expression and clinical study of vascular endothelial growth factor and its receptors FLT-1, FLK-1/KDR in non-small cell lung cancer

Abstract:Objective To investigate the relationship between the expression of vascular endothelial growth factor (VEGF), its receptors FLT-1, FLK-1/KDR and tumor angiogenesis, clinical staging, lymph node metastasis in non-small cell lung cancer (NSCLC). Methods Immunohistochemical staining and enzyme-linked immunosorbent assay (ELISA) were used to detect the levels of VEGF and its receptors FLT-1,FLK-1/KDR in the tissue and serum of 112 patients with NSCLC, and analysed their clinical significances. Results The expression of VEGF and its receptors FLT-1, FLK/KDR in lung tissue of NSCLC were significantly higher than that in normal alveolar tissue and paracancerous tissue (P<0.05) and they increased significantly in proportion with clinical stages Ⅰ, Ⅱ, Ⅲ-Ⅳ (P<0.05). The positive rates of VEGF and its receptors FLT-1, FLK-1/KDR in lymph node metastasis group were significantly higher than those without lymph node metastasis group (P<0.05). Se- rum VEGF in NSCLC was significantly higher than that in the control group and also increased according to clinical staging (Ⅰ, Ⅱ, Ⅲ-Ⅳ; P<0.05). Serum VEGF was significantly higher in the lymph node metastasis group than that of the absence group (P<0.05) . Conclusions The expression of VEGF and its receptors FLT-1, FLK/KDR in NSCLC increased significantly, and positively correlated with tumor development, progression and lymph node metastasis. Serum VEGF is an important marker of hyperplasia and metastasis in NSCLC.

Abstract:Introduction U2AF1 gene is associated with various types of hematological malignancies in adults. However, the expression level of U2AF1 gene and its prognostic significance are unclear in pediatric ALL patients. The study aimed to study the mRNA level of U2AF1 in pediatric ALL patients and its clinical relevance with long-term survival. Methods We quantitatively determined U2AF1 gene expression at diagnosis in 132 children with ALL by real-time PCR. According to the patients' median U2AF1 value, the patients' samples were classified into low U2AF1 and high U2AF1 expression groups. Twenty-two bone marrow samples from 22 patients with ITP were recruited as control. The correlation between the expression level of U2AF1 and clinical treatment outcome was analyzed. Results Pediatric patients with ALL showed higher U2AF1 mRNA levels than controls (P = .034). The relapse rates of patients in low U2AF1 levels group were obviously higher than those of U2AF1 high expression group (28.8% vs 12.1%, P = .030). Patients of low U2AF1 expression presented worse 5-year EFS than those of high U2AF1 expression (60% vs 81%, P = .035). For T-ALL, patients with low U2AF1 mRNA level showed lower BM blast percentages (P = .031), worse EFS (37.8% vs 92.3%, P = .003), and CIR (62.2% vs 7.7%, P = .003) than those in high U2AF1 expression group. Multivariate analysis confirmed low U2AF1 mRNA level could be used as an independent risk indicator of poor EFS and CIR of children with T-ALL. Conclusion Low U2AF1 mRNA level is related to inferior prognosis and can be served as a prognostic indicator for risk stratification in children with T-ALL.
Overexpression of Lymphoid Enhancer‐binding Factor‐1 (LEF1) is a Novel Favorable Prognostic Factor in Childhood Acute Lymphoblastic Leukemia

Abstract:INTRODUCTION:Lymphoid enhancer-binding factor-1 (LEF1) is a target gene and central mediator of the Wnt signaling pathway. High LEF1 expression has been reported as a prognostic marker in several types of hematologic malignancies of adult patients.METHODS:In this study, LEF1 expression was analyzed by real-time polymerase chain reaction (PCR) in 122 children with newly diagnosed ALL treated on the China NPCAC97 protocols. Patients' samples were dichotomized at the median value of control group and divided into LEF1(low) and LEF1(high) groups.RESULTS:The LEF1 mRNA levels in patients with ALL were significantly higher than those of normal controls, and the LEF1 levels were dramatically decreased following induction therapy. In addition, LEF1(high) patients had lower white blood cell (WBC) count at diagnosis and lower minimal residual disease (MRD) levels at the time of complete remission as compared to LEF1(low) patients. Finally, our studies showed that high LEF1 expression is associated with favorable CR rate and overall survival (OS) in childhood ALL (5-year OS: LEF1(high) 92% vs. LEF1(low) 73%, P = 0.009). High LEF1 level was associated with a favorable relapse-free survival in standard-risk patients and also related to a better OS within the subgroup of patients with BCR-ABL-negative ALL.CONCLUSION:Overexpression of LEF1 is a favorable prognostic factor in childhood ALL. The prognostic impact of LEF1 may assist treatment stratification and suggest the need of alternative regimens.

[Effects of NANOG Gene Down-Regulation on the Apoptosis of T-cell Acute Lymphoblastic Leukemia Cells].

Abstract:OBJECTIVE:To explore gene expression of NANOG in T-cell acute lymphoblastic leukemia (T-ALL) cell lines and the effects of NANOG gene down-regulation on apoptosis of leukemia cells. METHODS:Real-time PCR (RT-PCR) and Western blot were used to detect the expression level of NANOG gene and protein in MOLT-4, CCRF-HSB2 and Jurkat cells. To test the efficiency of RNA interference, MOLT-4 cells were firstly infected by lentiviral vectors, which were successfully constructed with NANOG specific shRNA. NANOG expression levels were subsequently re-evaluated by RT-PCR and Western blot. The percentages of early apoptotic cells (Annexin V⁺/7-AAD⁻) and late apoptotic cells (Annexin V⁺/7-AAD⁺) were analyzed by flow cytometry. The expression of apoptosis-related genes was also detected. RESULTS:Both NANOG gene and protein expression was positive in MOLT-4 and CCRF-HSB2 cells. The lentiviral vectors pLB-shNANOG-1, pLB-shNANOG-2, and pLB-sh control were successfully constructed, as evidenced by the viral titers (1.83-3.12)× 10⁸ IU/ml. The experimental data on infection of MOLT-4 cells with such lentiviral vectors revealed that both shRNA interfering sequences (shNANOG-1 and shNANOG-2) could stably down-regulate NANOG gene and protein expressions. The percentages of early apoptotic cells in groups of shNANOG-1[(8.06 ± 1.61)%]and shNANOG-2[(5.67 ± 1.59)%]were significantly increased as compared to that of MOLT-4 group[(1.13 ± 0.40)%]or sh-control [(1.15±0.49)%](P<0.05). However, no statistical difference among them was observed for late apoptotic cells (P>0.05). The gene expression of TP53, PMAIP1, and CASP9 of either shNANOG-1 or shNANOG-2 group was augmented as compared to that of MOLT-4 group or sh-control (P<0.05). Reversely, a significant down-regulation of Bcl-2 gene expression was observed (P<0.05). CONCLUSION:NANOG can be expressed in various human T-ALL cell lines. Down-regulation of NANOG can trigger leukemia cellular apoptosis through mitochondria-dependent apoptosis pathway.
[Activation of Signal Transducers and Activators of Transcription Induced by Vascular Endothelial Growth Factor in CD34+ Hematopoietic Progenitor Cells in Vitro].

Abstract:OBJECTIVE:To investigate the activation pattern of signal transducers and activators of transcription (STAT) induced by vascular endothelial growth factor (VEGF) in CD34+ hematopoietic progenitor cells, and gain an insight into the molecular mechanism and signal transduction pathway of VEGF that has an effect on CD34+ hematopoietic progenitor cells.METHODS:After isolated from umbilical cord blood by using a high-gradient magnetically activated cell sorting system (MACS), CD34+ cells were stimulated by VEGF (50 ng/ml) for different time (0, 15, 30, 45, 60, 90 min) to detect the tyrosine phosphorylation and nuclear translocation of STAT-3 and STAT-5 with Western blot and immunocytochemistry methods. The expression of VEGF receptor-2 (VEGFR2) on the membrane of CD34+ progenitor cells was examined by immunocytochemistry. ATWLPPR, an effective peptide screened from phage epitope library by affinity for membrane-expressed VEGFR2 and blocking the binding of VEGF to VEGFR2, was used to determine whether the activation of STAT pathway induced by VEGF was blocked.RESULTS:Tyrosine phosphorylation of STAT-3 and STAT-5 was undetectable in unstimulated CD34+ cells, but was evident at 15 min in response to VEGF stimulation. VEGF resulted in a rapid and transient tyrosine phosphorylation of STAT-3 and STAT-5. The maximal tyrosine phosphorylation was catched at 30 and 45 min, respectively (P = 0.0001), and returned to basal levels at 90 min. Immunocytochemistry confirmed that increased phosphorylated STAT-3 was translocated into the nuclei at 30 min (P = 0.0001), and mainly in cytoplasms again at 90 min after stimulation with VEGF. However, compared with unstimulated CD34+ cells, there was only increased phosphorylation of STAT-5 appeared mainly in cytoplasms, but no significant nuclear translocation was found after stimulation with VEGF (P > 0.05). The presence of VEGFR2 was confirmed using anti-VEGFR2 antibody staining by immunocytochemistry, moreover, the phosphorylation of STAT-3 and STAT-5 failed to be activated by the co-culture with ATWLPPR and VEGF, suggesting that activation of the STAT pathway be specifically mediated by VEGFR2 in CD34+ progenitor cells.CONCLUSIONS:STAT signaling pathway participates in the signal transduction of VEGF via VEGFR2 in CD34+ hemopoietic progenitor cells.
Correlation Between the Expression of VEGF and N-ras/pERK1/2 Signal Pathway and Its Clinical Significance in Acute Myeloid Leukemia

Abstract:OBJECTIVE: To investigate the expression of VEGF, N-Ras and pERK1/2,in order to clarify the relationship between the expression of VEGF and N-Ras/ERK signal pathway in the development of human acute myeloid leukemia(AML). METHODS: The expression level of VEGF, N-Ras and pERK1/2 was checked in the samples of bone marrow mononuclear cells from 46 diagnosed AML patients using immunohistochemistry method, and mutation analysis at N-ras codon 12 was finished using the method of PCR/RFLP, PCR/SSCP and DNA sequencing analysis. RESULTS: The expression of VEGF, N-Ras and pERK1/2 were significantly higher in 28 cases of newly diagnosed AML patients [(52.1±0.31)%,(59.8±0.32)%,(41.9±0.38)%,respectively] than that of 20 cases of normal control [(20.5±0.16)%,(20.9±0.12)%,(14.1±0.14)%;P=0.034,P=0.008,P=0.005, respectively],and that of 18 cases of complete remission AML(CR AML)[(18.6±0.18)%,(28.2±0.27)%,(15.1±0.19)%;P=0.029, P=0.018 ,P=0.008,respectively]; meanwhile, there was no significant different for the expression level of VEGF, N-Ras and pERK1/2 between the group of CR AML and normal control (P0.05, P=0.411, P=0.343). There was no mutation at N-ras codon 12 in all of patients suffering from AML. Close relationship was founded between the expression of VEGF, N-Ras and pERK1/2 (VEGF N-Ras:r=0.510,P=0.003;VEGF pERK1/2:r=0.513,P=0.003;VEGF pERK1/2:r=0.464,P=0.009). CONCLUSIONS: VEGF, cooperating with N-Ras/ERK signal pathway play some roles, and N-Ras/ERK signal pathway was seldom affected, if not all, by N-ras mutation, in the development of human acute myeloid leukemia.
Targeting lactate dehydrogenase A ( LDHA ) exerts antileukemic effects on T‐cell acute lymphoblastic leukemia

Abstract:BACKGROUND: T-cell acute lymphoblastic leukemia (T-ALL) is an uncommon and aggressive subtype of acute lymphoblastic leukemia (ALL). In the serum of T-ALL patients, the activity of lactate dehydrogenase A (LDHA) is increased. We proposed that targeting LDHA may be a potential strategy to improve T-ALL outcomes. The current study was conducted to investigate the antileukemic effect of LDHA gene-targeting treatment on T-ALL and the underlying molecular mechanism.METHODS: Primary T-ALL cell lines Jurkat and DU528 were treated with the LDH inhibitor oxamate. MTT, colony formation, apoptosis, and cell cycle assays were performed to investigate the effects of oxamate on T-ALL cells. Quantitative real-time PCR (qPCR) and Western blotting analyses were applied to determine the related signaling pathways. A mitochondrial reactive oxygen species (ROS) assay was performed to evaluate ROS production after T-ALL cells were treated with oxamate. A T-ALL transgenic zebrafish model with LDHA gene knockdown was established using CRISPR/Cas9 gene-editing technology, and then TUNEL, Western blotting, and T-ALL tumor progression analyses were conducted to investigate the effects of LDHA gene knockdown on T-ALL transgenic zebrafish.RESULTS: Oxamate significantly inhibited proliferation and induced apoptosis of Jurkat and DU528 cells. It also arrested Jurkat and DU528 cells in G0/G1 phase and stimulated ROS production (all P < 0.001). Blocking LDHA significantly decreased the gene and protein expression of c-Myc, as well as the levels of phosphorylated serine/threonine kinase (AKT) and glycogen synthase kinase 3 beta (GSK-3β) in the phosphatidylinositol 3'-kinase (PI3K) signaling pathway. LDHA gene knockdown delayed disease progression and down-regulated c-Myc mRNA and protein expression in T-ALL transgenic zebrafish.CONCLUSION: Targeting LDHA exerted an antileukemic effect on T-ALL, representing a potential strategy for T-ALL treatment.

Overexpression of PTPN21 Promotes Proliferation of EGF-stimulated Acute Lymphoblastic Leukemia Cells Via the MAPK Signaling Pathways

Abstract:OBJECTIVES:This study aims to investigate the role of excessive Protein Tyrosine Phosphatase Non-Receptor Type 21 (PTPN21) in the proliferation of Acute Lymphoblastic Leukemia (ALL) cells with EGF stimulation.METHODS:PTPN21 was overexpressed in ALL cell lines by lentiviral transfection. Apoptosis was assayed by Annexin V/7-AAD staining. The proliferation and cell cycle of EGF-treated ALL cells were assessed by MTT and Ki-67/7-AAD staining respectively. The phosphorylation of Src tyrosine kinase and mediators of distinct MAPK pathways were assessed by Western blot.RESULTS:Overexpression of PTPN21 had minimal effect on the apoptosis of ALL cells, but significantly promoted the proliferation and cell cycle progression of ALL cells stimulated with EGF. The activity of Src tyrosine kinase and the MAPK pathways was elevated. Inhibition of MAPK pathways by specific inhibitors mitigated this pro-proliferative effect of excessive PTPN21 on EGF-stimulated ALL cells.CONCLUSION:PTPN21 may facilitate ALL progression by promoting cell proliferation via the Src/MAPK signaling pathways.
Amino acid transporter LAT1 in tumor-associated vascular endothelium promotes angiogenesis by regulating cell proliferation and VEGF-A-dependent mTORC1 activation

Abstract:Abstract Background Tumor angiogenesis is regarded as a rational anti-cancer target. The efficacy and indications of anti-angiogenic therapies in clinical practice, however, are relatively limited. Therefore, there still exists a demand for revealing the distinct characteristics of tumor endothelium that is crucial for the pathological angiogenesis. L-type amino acid transporter 1 (LAT1) is well known to be highly and broadly upregulated in tumor cells to support their growth and proliferation. In this study, we aimed to establish the upregulation of LAT1 as a novel general characteristic of tumor-associated endothelial cells as well, and to explore the functional relevance in tumor angiogenesis. Methods Expression of LAT1 in tumor-associated endothelial cells was immunohistologically investigated in human pancreatic ductal adenocarcinoma (PDA) and xenograft- and syngeneic mouse tumor models. The effects of pharmacological and genetic ablation of endothelial LAT1 were examined in aortic ring assay, Matrigel plug assay, and mouse tumor models. The effects of LAT1 inhibitors and gene knockdown on cell proliferation, regulation of translation, as well as on the VEGF-A-dependent angiogenic processes and intracellular signaling were investigated in in vitro by using human umbilical vein endothelial cells. Results LAT1 was highly expressed in vascular endothelial cells of human PDA but not in normal pancreas. Similarly, high endothelial LAT1 expression was observed in mouse tumor models. The angiogenesis in ex/in vivo assays was suppressed by abrogating the function or expression of LAT1. Tumor growth in mice was significantly impaired through the inhibition of angiogenesis by targeting endothelial LAT1. LAT1-mediated amino acid transport was fundamental to support endothelial cell proliferation and translation initiation in vitro. Furthermore, LAT1 was required for the VEGF-A-dependent migration, invasion, tube formation, and activation of mTORC1, suggesting a novel cross-talk between pro-angiogenic signaling and nutrient-sensing in endothelial cells. Conclusions These results demonstrate that the endothelial LAT1 is a novel key player in tumor angiogenesis, which regulates proliferation, translation, and pro-angiogenic VEGF-A signaling. This study furthermore indicates a new insight into the dual functioning of LAT1 in tumor progression both in tumor cells and stromal endothelium. Therapeutic inhibition of LAT1 may offer an ideal option to potentiate anti-angiogenic therapies.

[Effect of Autocrine VEGF on Chronic Myeloid Leukemia Cell Line K562]

Abstract:Vascular endothelial growth factor (VEGF) is a specific mitogen for vascular endothelial cells. It has been associated with angiogenesis, growth, metastasis and poor prognosis in solid tumors. Lately, it has been known that VEGF expression is higher in bone marrow from chronic myeloid leukemia (CML) patients than that in normal subjects. However, it is not clarified that the effect of VEGF on the abnormal proliferation of CML cells. In order to explore the effect of autocrine VEGF on CML cells, K562 cells were transfected with the VEGF(121) cDNA sense vector (K562/S) or with the VEGF(121) cDNA antisense vector (K562/As). K562 cells were transfected with the pcDNA(3) vector (K562/V) as the control. Cell proliferation was determined by MTT and colony forming assay in vitro. Flow cytometric Annexin-V-FITC/PI dual labeling technique was performed to observe the effect of VEGF(121) cDNA transfection on apoptosis of K562 cells. Results indicated that K562/S transfectants exhibited a 3-fold increase in VEGF secretion, and K562/As transfectants exhibited a 49% reduction in VEGF secretion. K562/As showed a reduced growth rate and colony forming efficiency as compared to K562/V. K562/S showed an increasing growth rate and colony forming efficiency as compared to K562/V. K562/As had more apoptotic cells than K562/V and K562/S in the same culture condition. These data suggest that VEGF plays an important role in the abnormal proliferation and apoptosis in CML cells through an autocrine mechanism.
[Expression of Angiogenic Factors and Their Clinical Significances in Acute Myeloid Leukemia].

Abstract:BACKGROUND & OBJECTIVETo investigate the expression of vascular endothelial growth factor (VEGF) & basic fibroblast growth factor (bFGF) in primary acute myeloid leukemia (AML) cells, and study plasma VEGF& bFGF concentration in patients with AML and the two factors' clinical significance.METHODSVEGF&bFGF and their receptors mRNA expression were detected by RT-PCR; VEGF & bFGF plasma level was analyzed by ELISA.RESULTSIn 107 AML patients, the positive mRNA expression rate of VEGF, KDR and Flt1 was 56.1%, 49.5%, and 2.8%, respectively; for bFGF and FGFR1-4, the positive rate was 21.5%, 35.5%, 9.4%, 35.5% and 23.4%, respectively. VEGF concentration in AML (154.75+/-109.98) pg/ml was significantly higher than that in AML-CR (72.05+/-23.39) pg/ml and the normal control (99.91+/-41.87) pg/ml (P< 0.05). Nevertheless, the level of bFGF had no significant difference among AML, AML-CR and the normal control. VEGF level (124.05+/-76.57) pg/ml in patients, who got complete remission (CR) after 2 cycles of chemotherapy, was remarkably lower than that (211.243+/-169.88) pg/ml in patients without CR (P< 0.05). Furthermore, the higher the concentration of VEGF & bFGF, the shorter the survival was.CONCLUSIONVEGF& bFGF and their receptors mRNA in leukemic patients could be expressed to some degree,and our investigation suggested VEGF &bFGF and their receptors probably be produced by autocrine or paracrin. Abnormally high level of VEGF may be a poor factor for chemotherapy and survival in AML. The prognosis of patients with AML may be improved by treating VEGF and their receptors as therapy targets.

Abstract:OBJECTIVE:To evaluate the correlation between MicroRNA-191 (miR-191) and T lymphoblastic leukemia/lymphoma (T-ALL/LBL) to probe its underlying molecular mechanism.METHODS:The expression of miR-191 was examined by real-time PCR (RT-PCR) in 20 T-ALL/LBL tissue samples and 20 lymphoid reactive hyperplasia (LRH) tissue samples. The correlation between miR-191 and the clinicopathological feature of T-ALL/LBL was analyzed. Antisense miR-191 lentiviral vectors was constructed and transfected into T-ALL/LBL Jukat cells. After transfection, the expression of miR-191 was examined by RT-PCR. The cell activity was evaluated by CCK-8 asssy. The cell cycle and apoptosis were determined by flow cytometry.RESULTS:Compared with LRH samples, the results of RT-PCR showed significant upregulation of miR-191 in 20 T-ALL/LBL tissue samples (1.875±0.079 vs 1.000, P<0.05). The expression level of miR-191 was negatively associated with prognosis. Compared with LV-NC-GFP and control groups, the expression of miR-191 significantly decreased after transfection of antisense miR-191 lentiviral vectors (0.578±0.012 vs 1.011±0.053 and 1.000, P<0.05), the percentages of apoptotic cells and the cell in G0/G1 phase significantly increased (P<0.05).CONCLUSIONS:miR-191 might play a significant role in the development of T-ALL/LBL, implicating a new target for therapy.

Abstract:Objective YAP1 plays a dual role as an oncogene and tumor suppressor gene in several tumors; differentiating between these roles may depend on the YAP1 phosphorylation pattern. The specific function of YAP1 in B cell acute lymphoblastic leukemia (B-ALL), however, is currently unclear. Thus, in the present study, the role of YAP1 in B-ALL was investigated using relevant cell lines and patient datasets. Methods The effects of shRNA-mediated knockdown on YAP1 and LATS1 levels in the NALM6 and MOLT-4 cell lines were examined using Western blotting, quantitative real-time polymerase chain reaction, flow cytometry, immunostaining, and nude mouse subcutaneous tumorigenesis experiments. Gene expression levels of Hippo pathway-related molecules before and after verteporfin (VP) treatment were compared using RNA-Seq to identify significant Hippo pathway-related genes in NALM6 cells. Results Patients with ALL showing high YAP1 expression and low YAP1-Ser127 phosphorylation levels had worse prognoses than those with low YAP1 protein expression and high YAP1-Ser127 phosphorylation levels. YAP1-Ser127 phosphorylation levels were lower in NALM6 cells than in MOLT-4 and control cells; YAP1 was distributed in the nuclei in NALM6 cells. Knockdown of YAP1 inhibited MOLT-4 and NALM6 cell proliferation and arrested the NALM6 cell cycle in the G0/G1 phase. Before and after VP treatment, the expression of the upstream gene LATS1 was upregulated; its overexpression promoted YAP1-Ser127 phosphorylation. Further, YAP1 was distributed in the plasma. Conclusion LATS1 may downregulate YAP1-Ser127 phosphorylation and maintain B-ALL cell function; thus, VP, which targets this axis, may serve as a new therapeutic method for improving the outcomes for B-ALL patients.
Tumor Necrosis Factor- α Promotes the Tumorigenesis, Lymphangiogenesis, and Lymphatic Metastasis in Cervical Cancer via Activating VEGFC-Mediated AKT and ERK Pathways

Abstract:Background: Lymphatic metastasis is a common phenomenon of cervical cancer. Tumor necrosis factor-α (TNF-α) was found to be closely associated with lymphatic cancer metastasis. However, the mechanism through which TNF-α regulates lymphatic metastasis in cervical cancer remains unclear. Methods: In this study, cervical cancer cells were cultured in Dulbecco's modified Eagle's medium (DMEM) with or without TNF-α for 48 h, and then the corresponding conditional medium (CM-TNF-α or CM) was collected. The level of vascular endothelial growth factor (VEGFC) in the corresponding CM was then detected using an enzyme-linked immunosorbent assay (ELISA). Next, human lymphatic endothelial cells (HLECs) were cultured in CM-TNF-α or CM for 48 h. Cell viability was measured using the cell counting kit-8 (CCK-8) assay, and angiogenesis was detected using a tube formation assay. Subsequently, the expressions of AKT, p-AKT, ERK, and p-ERK in HLECs were detected using western blotting. In addition, to further investigate the effect of TNF-α on the progression of cervical cancer, a C33A subcutaneous xenograft model was established in vivo. Results: We found that TNF-α significantly stimulated cervical cancer cells to secrete VEGFC. Additionally, the CM collected from the TNF-α-treated cervical cancer cells notably promoted the proliferation, migration, and angiogenesis of HLECs; however, these changes were reversed by MAZ51, a VEGFR3 inhibitor. Moreover, TNF-α obviously elevated D2-40 and VEGFC protein expressions in tumor tissues, promoting lymphangiogenesis and lymphatic metastasis in vivo. Meanwhile, TNF-α markedly upregulated p-AKT and p-ERK expressions in tumor tissues, whereas these changes were reversed by MAZ51. Conclusion: Collectively, TNF-α could promote tumorigenesis, lymphangiogenesis, and lymphatic metastasis in vitro and in vivo in cervical cancer via activating VEGFC-mediated AKT and ERK pathways. These results may provide new directions for the treatment of cervical cancer.
Significance of TEL-AML1 Fusion Gene Expression in Children with Acute Lymphoblastic Leukemia

Abstract:Objective To study the frequency and clinical significance of transloation,ets,leukemia-acute myeloblasticleukemia1 (TEL-AML1) fusion gene in children with acute lymphoblastic leukemia (ALL).Methods Base on MIC(morphology,immunology and cytogenetics),nested reverse transiption-polymerase chain reaction(RT-PCR) was performed to detect TEL-AML1 fusion gene.Results TEL-AML1 fusion gene was found in 22.2% (20/90) of patients with newly diagnosed ALL and 23.7% (19/80) in B lineage ALL cases.The fusion gene was found in 1 T lineage ALL case.Fifteenth days′ complete remission rate and early mortality were no difference between TEL-AML1 fusion gene positive group and negative group.Conclusion Therapeutic results are not different between TEL-AML1 fusion gene positive group and negative group lately.

Abstract:ALL (Acute lymphoblastic leukemia) is the most common pediatric malignancy and T-ALL (T-cell acute lymphoblastic leukemia) comprises about 15% cases. Compared with B-ALL (B-cell acute lymphoblastic leukemia), the prognosis of T-ALL is poorer, the chemotherapy is easier to fail and the relapse rate is higher. Previous studies mainly focused in Notch1-related long non-coding RNAs (lncRNAs) in T-ALL. Here, we intend to investigate lncRNAs involved in T-ALL covering different subtypes. The lncRNA PPM1A-AS was screened out for its significant up-regulation in 10 T-ALL samples of different subtypes than healthy human thymus extracts. Besides, the PPM1A-AS expression levels in 3 T-ALL cell lines are markedly higher than that in CD45 + T cells of healthy human. We further demonstrate that PPM1A-AS can promote cell proliferation and inhibit cell apoptosis in vitro and can influence T-ALL growth in vivo . Finally, we verified that PPM1A-AS can regulate core proteins, Notch4, STAT3 and Akt, of 3 important signaling pathways related to T-ALL. These results confirm that lncRNA PPM1A-AS can act as an oncogene in T-ALL and maybe a potential clinical target of patients resistant to current chemotherapy or relapsed cases.
