Abstract:Immunobiosensors were developed for detection of Escherichia coli O157:H7 based on the surface immobilization of monoclone antibodies onto indium tin oxide (ITO) electrodes. The immobilization of antibodies onto ITO chips was carried out by silanization. The effects of epoxysilane monolayer, the antibody layer on the electrochemical properties of the electrode, and the combined target bacteria were analyzed through cyclic voltammetry and electrochemical impedance spectroscopy. By using Randles model as the equivalent circuit, the concentration of the target bacteria can be quantitatively analyzed in terms of the change of electron transfer resistance. The biosensor could detect the target bacteria with a detection limit of 4times103 CFU/mL. A linear response was found between 4times1034times106 CFU/mL. This biosensor was characterized with high sensitivity, excellent selectivity, short detection time and easy operation. It has a promising application in clinical laboratory diagnoses, environmental detection and food safety

Abstract:A silicon microcantilever sensor was developed for the detection of Escherichia coli O157:H7. The microcantilever was modified by anti-E. coli O157:H7 antibodies on the silicon surface of the cantilever. When the aquaria E. coli O157:H7 positive sample is injected into the fluid cell where the microcantilever is held, the microcantilever bends upon the recognition of the E. coli O157:H7 antigen by the antibodies on the surface of the microcantilever. A negative control sample that does not contain E. coli O157:H7 antigen did not cause any bending of the microcantilever. The detection limit of the sensor was 1 × 106 cfu/mL when the assay time was < 2 h.

Abstract:Escherichia coli ( E. coli ) bacteria with varying solution concentrations have been successfully detected using silver nanoparticles (Ag NPs)-based localized surface plasmon resonance (LSPR) biosensors. The Ag NPs were effectively synthesized by a chemical method using trisodium citrate with L-Histidine (L-His) and deposited on the surface of Au thin film-coated half-cylinder BK-7 prisms. He-Ne laser with a wavelength of 632.8 nm was used to generate LSPR phenomena in Kretschmann configuration with prism/Au thin film/His-Ag NPs/ E. coli bacteria/air structure arrangements. The variation of E. coli bacteria concentration was carried out to determine the effect of E. coli bacteria concentration on the LSPR curve characteristics. The characterization results showed that the size of Ag NPs was 18.7 nm, and that of His-Ag NPs was 17.9 nm. Selected area electron diffraction results indicated the formation of diffraction rings with the presence of lattice planes (1 1 1), (2 0 0), (2 2 0), and (3 1 1), proving the face-centered cubic crystal structure of silver. The absorbance peak of Ag NPs shifted from a wavelength of 421–414 nm with an increase in band gap energy from 2.94 eV to 2.99 eV, along with a decreased average particle size. The functional groups observed in His-Ag NPs showed wavenumbers at 3320 to 3318 cm −1 , 2106 to 2129 cm −1 , and 1635 cm −1, showing the OH, CH, and C CO bonds, respectively. The SPR angle of the prism/Au thin film/air structure is 44.80°. Meanwhile, the LSPR angle for the prism/Au thin film/His-Ag NPs/air structure is 44.92°. There is an increase in the LSPR angle by 0.12°. Moreover, the minimum reflectance increases by 0.02. After detecting E. coli bacteria, the LSPR angle shifted by 0.26°, 0.38°, and 0.49° for concentrations of 6.0 × 10 8 CFU/mL, 6.0 × 10 7 CFU/mL and 6.0 × 10 6 CFU/mL respectively. However, the minimum reflectance rose from 0.09° to 0.14°, 0.20°, and 0.22°. Moreover, SPR testing with the structure of the prism/Au thin film/ E. coli bacteria/air was carried out to determine the contribution of His-Ag NPs for detecting E. coli bacteria. The results showed that no angular shift occurs. These results indicate that using Ag NPs encapsulated with L-His is essential in amplifying the SPR signal and detecting E. coli bacteria. There was a notable alteration in both the LSPR angle and minimum reflectance indicating that adding His-Ag NPs facilitated the interaction between the E. coli and the sensor surface, thereby enhancing the performance of LSPR-based sensors for E. coli detection for low limit of detection value at 0.47 CFU/mL.

Abstract:Biomolecule immobilization on nanomaterials is attractive for biosensors since it enables the capture of a higher concentration of bioreceptor units while also serving as a transduction element. The technique could enhance the accuracy, specificity, and sensitivity of the analytical measurements of biomolecules. However, it was found that the limitation in chemically binding biomolecules on nanoparticle surfaces could only cross-link between the C-terminal and N-terminal. Here, we report the facile one-step synthesis of amine-functionalized silica nanoparticles (AFSNPs). (3-Aminopropyl)triethoxysilane was used as a precursor to modify the functional surface of nanoparticles via the Stöber process. The biomolecules were immobilized to the AFSNPs through itaconic acid, a novel cross-linker that binds between the N-terminal and N-terminal and potentially improves proteins and nucleic acid immobilization onto the nanoparticle surface. The newly developed immobilization approach on AFSNPs for biomolecular detection enhanced the efficiency of ELISA, resulting in increased sensitivity. It might also be easily used to identify different pathogens for clinical diagnostics.

Abstract:Immunoassays based on enzyme-labeled antibodies have been widely used in the food safety field. However, the production process of enzyme-labeled antibodies is complicated and the low storage stability limits their application. Herein, antibody-horseradish peroxidase (HRP) co-assembled nanocomposites (AHC NCs) with outstanding advantages such as enhanced stability, lower cost, and substrate affinity were successfully prepared via a one-pot green method. Then the AHC NCs were employed as an alternative to traditional enzyme-labeled antibodies to develop a chemiluminescence enzyme immunoassay (CLEIA) toward Escherichia coli (E. coli) O157:H7. Under optimal conditions, E. coli O157:H7 can be detected in a linear range from 1 × 103 CFU mL-1 to 5 × 106 CFU mL-1, while the limit of detection (LOD) is as low as 2.2 × 102 CFU mL-1 (3σ). A series of repeatability studies showed reproducible results with a coefficient of variation of less than 7%. In addition, the proposed CLEIA was successfully applied to the analysis of spiked samples (tap water) and gave quantitative recoveries from 93.72% to 100.72%. This work demonstrates that the developed CLEIA can be applied as a universal platform for specific detection of diversified analytes.

Abstract:Immobilization of proteins on a biocompatible conductive interface is highly desirable for the fabrication of biosensors. In this study, a nanocomposite has been prepared by assembling well-distributed gold nanoparticles (AuNPs) on the surface of a polypyrrole-reduced graphene oxide (PPy-rGO) composite through electrostatic adsorption. This serves as a platform for immobilization of a capture antibody, which was conjugated onto the ferrocene doped polypyrrole-gold nanoparticles (PPy@Fc/AuNPs) composite. The design and performance of the biosensor was tested against detection of a whole-cell bacteria E. coli K12. This nanocomposite has a high surface area, good conductivity and biocompatibility, which is shown to be very suitable for enzyme-free detection of this bacteria. Results show excellent analytical performance with a linear range from 1.0 × 101 to 1.0 × 107 CFU mL-1 and a low detection limit of 10 CFU mL-1. The sensor has high selectivity, excellent reproducibility, and good stability.
Fabrication of a Label-Free Immunosensor Using Surface-Engineered AuPt@GQD Core-Shell Nanocomposite for the Selective Detection of Trace Levels of Escherichia coli from Contaminated Food Samples

Abstract:Fabrication of label-free immunosensors is highly necessitated due to their simplicity, cost-effectiveness, and robustness. Herein, we report the facile development of a label-free, direct, rapid, capacitive immunosensor for ultrasensitive and rapid recognition of trace levels of Escherichia coli from contaminated food samples. This was achieved using gold platinum core-shell nanoparticles loaded with graphene quantum dots (AuPt@GQDs) that were utilized as electrode modifiers. The incorporation of GQDs to the surface of AuPt core-shell nanoparticles was performed using the "greener" probe-sonication method. The electrochemical properties of AuPt@GQDs, determined using cyclic voltammetry and electrochemical impedance spectroscopy, suggested the optimized loading concentration of AuPt to be 0.05% in the core-shell nanocomposite to exhibit the highest current response. Furthermore, immobilization of anti-E. coli monoclonal antibodies (anti-E. coli mAb) onto the surface of modified electrodes was performed using amine coupling. The high specific binding of E. coli cells onto the surface of the immuno-electrode was measured as a direct function of change in transient capacitance with time that was measured at low and high frequencies. The resultant immunosensor (bovine serum albumin/anti-E. coli mAb/AuPt0.05@GQDs/FTO) demonstrated a detection range (5 to 4.5 × 103 cells/mL), with the detection limit as low as 1.5 × 102 cells/mL, and an excellent sensitivity ∼171,281.40 μF-1 mL cells-1 cm-2 without the use of any labels (R2-0.99). These findings were further verified using real sample analysis wherein the immuno-electrode demonstrated outstanding sensitivity, the highest noticed so far. More interestingly, the high resuability ∼48 weeks (RSD-5.92%) and excellent reproducibility in detection results (RSD ∼ 9.5%) testify its potential use in a clinical setting. The results reveal the usefulness of the surface-engineered AuPt@GQDs core-shell nanocomposite as an electrode modifier that can be used for the development of newer on-site monitoring devices to estimate trace levels of pathogens present as contaminants in food samples.

Abstract:The molybdenum disulfide (MoS2) nanosheets functionalized fiber optic surface plasmon resonance (SPR) immunosensor has been reported for the sensitive detection of Escherichia coli (E. coli). The MoS2 nanosheets were prepared by chemical exfoliation method. The synthesised nanostructures were characterized for their structural, morphological and optical properties. The E. coli monoclonal antibodies were successfully immobilized on the MoS2 functionalized sensing platform via hydrophobic interactions. An alternative method simplifying the antibodies immobilization process by functionalization of 2D nanomaterial (MoS2 nanosheets) for rapid (~15 mins) bacterial quantification is presented in this study. The immunosensor uses wavelength interrogation method and a strong linear relationship (R2 = 0.994) was observed between spectral response of immunosensor and different concentration of E. coli. The nonspecificity and cross-reactivity studies of the developed immunosensor were investigated with detection of Salmonella Typhimurium and Staphylococcus aureus. To demonstrate the practical application, spiked samples of water and orange juice were analysed with acceptable recovery results. The label-free immunosensor exhibits better performance, detection limit (94 CFU/mL), high sensitivity (2.9 nm/1000 CFU mL-1; 3135 nm/RIU) and profound specificity as compared to conventional fiber optic SPR sensor (detection limit: 391 CFU/mL, sensitivity: 0.6 nm/1000 CFU mL-1; 1646 nm/RIU). This sensing platform shows promising applications in regular water and food quality monitoring for various pathogenic microorganisms.
Surface Functionalization and Escherichia coli Detection Using Surface-Enhanced Raman Spectroscopy Driven by Functional Organic Polymer/Gold Nanofilm-Based Microfluidic Chip

Abstract:In this work, a microfluidic prototype based on polymeric materials was developed to monitor surface processes using surface-enhanced Raman spectroscopy (SERS), keeping the reagents free of environmental contamination. The prototype was fabricated on poly(methyl methacrylic acid) (PMMA). A micrometric membrane of a functional organic polymer (FOP) based on p-terphenyl and bromopyruvic acid monomers was formed on the PMMA surface to promote the formation of metal nanoclusters. Au nanosized film was deposited on the FOP membrane to give rise to the SERS effect. A microchannel was formed on another piece of PMMA using micromachining. A representative 3D model of the prototype layer arrangement was built and simulated in COMSOL Multiphysics® to approximate the electric field distribution and calculate the power enhancement factor as the Au film changes over time. The fabrication process was characterized using UV-visible and Raman spectroscopies and XPS. The prototype was tested using a Raman microscope and liquid solutions of cysteamine and Escherichia coli (E. coli). The simulation results demonstrated that the morphological characteristics of the Au layer give rise to the SERS effect, and the power enhancement factor reaches values as high as 8.8 × 105 on the FOP surface. The characterization results showed the formation of the FOP and the Au film on PMMA and the surface functionalization with amine groups. The Raman spectra of the prototype showed temporal evolution as different compounds were deposited on the upper wall of the microchannel. Characteristic peaks associated with these compounds were detected with continuous monitoring over time. This prototype offers many benefits for applications like monitoring biological processes. Some advantages include timely surface evaluation while avoiding environmental harm, decreased use of reagents and samples, minimal interference with the process by measuring, and detecting microorganisms in just 1 h, as demonstrated with the E. coli sample.

Abstract:Generally, immunoaffinity BLI biosensors detect target analytes via selective adsorption through antigen-antibody interactions. For increased sensitivity, various particles have been introduced into pre-bound analytes on BLI biosensors. With the growing interest in using proteins to assemble functional materials, Sup35NM amyloid fibrils with customized functions attract attention since they are ideal scaffolds for designable materials. In this work, we described a simple strategy through the genetic fusion of Z-domain with the protein Sup35NM for antibody binding and employed amyloid fibrils functionalized E. coli as signal enhancers. The target analytes attached to the BLI biosensor were loaded with antibody-coupled E. coli cells and the optical thickness of the layer greatly changed, thereby inducing a significant wavelength interference. Additionally, the IgG-trapping ability of the bacteria was characterized using ELISA and the BLI method. The signal was amplified in a sandwich assay format utilizing two model antigens, NS1 and PIC. The detection limit of NS1 decreased from 93.98 ng/mL to 0.82 ng/mL under optimized conditions and the signal of PIC is also improved by 8.4-fold. The whole detection takes 30 minutes. The findings demonstrate that functionalized E. coli may be utilized to amplify the signal of biosensors, in a rapid and sensitive manner.

Trifunctional Nanocomposites with Colorimetric Magnetic Catalytic Activities Labels in Sandwich Immunochromatographic Detection of Escherichia coli O157:H7

Abstract:The emergence of nanoenzymes has catalyzed the robust advancement of the lateral flow immunoassay (LFIA) in recent years. Among them, multifunctional nanocomposite enzymes with core-shell architectures are considered preferable for promoting the sensing ability due to their good biocompatibility, precise control over size, and surface properties etc. Herein, we developed a dual-channel ensured lateral flow immunoassay (DFLIA) platform utilizing a magnetic, colorimetric, and catalytic multifunctional nanocomposite enzyme (Fe3O4@TCPP@Pd) [TCPP, Tetrakis (4-carboxyphenyl) porphyrin] for the ultrasensitive and highly accurate rapid detection of Escherichia coli O157:H7 (E. coli O157:H7). Fe3O4@TCPP@Pd-mAb exhibits superior performance compared to traditional AuNPs, including enhanced sensitivity and an extended linear detection range, benefiting from its high brightness signal, strong magnetic separation ability, and high peroxidase activity (Vmax = 2.32 μM S1-). Moreover, the Fe3O4@TCPP@Pd-labeled mAb probe exhibited exceptional stability and high affinity toward E. coli O157:H7 (with an affinity constant of approximately 1.723 × 109 M-1), indicating its potential for the efficient capture of the pathogen. Impressively, the developed Fe3O4@TCPP@Pd-DFLIA achieved ultrasensitive detection for E. coli O157:H7 with pre- and postcatalytic naked-eye detection sensitivities of 255 cfu/mL and 77 cfu/mL, respectively, representing an approximately 41-fold improvement over the conventional AuNP-based LFIA and also possessed good specificity and reproducibility [relative standard deviation (RSD) < 10%]. Additionally, the established DFLIA exhibited satisfactory recoveries in detecting pork and milk samples, further validating the reliability of this platform for immunoassays and demonstrating its potential for utilization in bioassays and clinical diagnostics.
Pathogen-Imprinted Organosiloxane Polymers as Selective Biosensors for the Detection of Targeted E. coli

Abstract:Early detection of pathogens requires methods that are fast, selective, sensitive and affordable. We report the development of a biosensor with high sensitivity and selectivity based on the low-cost preparation of organosiloxane (OSX) polymers imprinted with E. coli-GFP (green fluorescent protein). OSX polymers with high optical transparency, no cracking, and no shrinkage were prepared by varying several parameters of the sol-gel reaction. The unique shape and chemical fingerprint of the targeted inactivated E. coli-GFP were imprinted into bulk polymers by replication imprinting where the polymer solution was dropcast onto a bacteria template that produced a replica of the bacterial shape and chemistry on the polymer surface upon removal of the template. Capture performances were studied under non-laminar flow conditions with samples containing inactivated E. coli-GFP and compared to inactivated S. typhimurium-GFP. Capture selectivity ratios are dependent on the type of alkoxysilanes used, the H2O:silane molar ratio, and the polymerization temperature. The bacteria concentration in suspension ranged from ~6 × 105 to 1.6 × 109 cells/mL. E. coli-imprinted OSX polymers with polyethylene glycol (PEG) differentiated between the targeted bacterium E. coli, and non-targeted bacteria S. typhimurium and native E. coli-GFP, achieving selectivity ratios up to 4.5 times higher than polydimethylsiloxane (PDMS) and OSX polymers without PEG.

Abstract:Graphical representation for chemical immobilization of E. coli‐specific bacteriophage with NH2‐MIL‐101(Fe) MOF a high photoluminescence rod‐shaped microcrystals for low‐level bacteria detection. As concern raised by the World Health Organization (WHO) of antibiotic‐resistant and bio‐defensive bacteria, a metal–organic framework (MOF) based optical biosensor came into consideration for precise, quick, and sensitive detection of Escherichia coli (E. coli) (ATCC 10799) using bacteriophage as a bio‐recognition element. In the present study, amine‐functionalized Fe‐based MOF, i.e., NH2‐MIL‐101(Fe), was synthesized by the solvothermal method (approx. 531–1106 nm in size and 20 mV zeta potentials by DLS) and further characterized by SEM, XRD, ATR‐FTIR, UV–VIS, and photoluminescent (PL) spectroscopy. The lytic bacteriophage was isolated from a sewage sample, purified, and concentrated using the ultra‐centrifugation method and achieved a high titer of 7.3 × 1012 PFU/ml. The concentration, stability, and accessible receptor binding domains (RBDs) of the biorecognition element for binding with their analytes play an important role in developing sensitive and specific biosensor systems. To fulfill the mentioned criteria, optimized glutaraldehyde concentration was estimated at 0.25%, at 30 °C for conjugating maximum bacteriophage titer of 8.6 × 105 PFU/ml for each 1 mg amine functionalized iron‐based MOF. The synthesized detection probe has shown excellent photoluminescence and antibacterial activity and achieved a detection limit of 652 CFU/ml over a bacterial detection concentration range from 5.78 × 101 to 5.78 × 106 CFU/ml for E. coli with 10–12 min of response time, high specificity, and long‐term stability even at room temperature. Therefore, it can be inferred that this MOF‐based strategy can be helpful in the specific and sensitive detection of various bacterial pathogens using bacteriophage as a bio‐recognition element.

Abstract:Noble metal nanoenzyme-based electrochemical biosensors has recently been used in the field of pathogenic bacteria detection. Nonetheless, the modification of bioligands on the surface of the nanoenzyme can shield its catalytic site and reduce its catalytic activity. Herein, we developed a bifunctional nanomaterial (Pt-Ni@erGO) composed of epoxy-rich graphene oxide (erGO) and platinum (Pt)-nickel (Ni) nanoparticles. erGO exhibits large specific surface area and sharp edges, which are beneficial to load high-density Pt-Ni nanoparticles and retain its catalytic activity, and insert/cut through the cell membranes. Pt-Ni@erGO displays high antibody-like activity and enhanced catalytic activity, which are conducive to the enhancement of the sensitivity. With the biosensor, we achieved the specific detection of Escherichia coli (E. coli) with a sensitivity of 38 CFU/mL and a linear range of 1.5 × 102–1.5 × 107 CFU/mL. In addition, the recovery for detecting E. coli in urine samples ranged 96.73 % to 98.85 %. Therefore, this work suggests that the designed bifunctional nanomaterial can serve as a promising electrochemical signal reporter and lead an innovative direction for the development of bifunctional probe with high antibody-like and catalytic activity.

Abstract:Escherichia coli O157:H7 is regarded as one of the most harmful pathogenic microorganisms related to foodborne diseases. This paper proposes a rapid-detection biosensor for the sensitive and quantitative analysis of E. coli O157:H7 in biological samples by surface-enhanced Raman scattering (SERS)-based lateral flow immunoassay (LFIA). A novel gold-shell silica-core (SiO2/Au) nanosphere (NP) with monodispersity, good stability, and excellent SERS activity was utilized to prepare high-performance tags for the SERS-based LFIA system. The SiO2/Au SERS tags, which were modified with two layers of Raman reporter molecules and monoclonal antibodies, effectively bind with E. coli O157:H7 and form sandwich immune complexes on the test lines. E. coli O157:H7 was quantitatively detected easily by detecting the Raman intensity of the test lines. Under optimal conditions, the limit of detection (LOD) of the SiO2/Au-based SERS-LIFA strips for the target bacteria was 50 cells/mL in PBS solution, indicating these strips are 2,000 times more sensitive than colloidal Au-based LFIA strips. Moreover, the proposed assay demonstrated high applicability in E. coli O157:H7 detection in biological samples, including tap water, milk, human urine, lettuce extract and beef, with a low LOD of 100 cells/mL. Results indicate that the proposed SERS-based LFIA strip is applicable for the sensitive and quantitative determination of E. coli O157:H7.

Abstract:Surface-enhanced Raman spectroscopy (SERS) has been successfully applied to detect various biomolecules, but it is still in challenge to assay living cells or bacteria sensitively, selectively and quantitatively in complex environments. In this paper, 4-ATP and DTNB are assembled on Ag nanoparticle (NP) -decorated poly (styrene-co-acrylic acid) (PSA) nanospheres and then sealed by silica shells to form sensitive SERS labels based on the localized surface plasmon resonance of Ag NPs and large light scattering cross-sections of PSA nanospheres. They are further developed as encoding tags for dual detection of S. aureus and E. coli after assembling corresponding aptamers, which demonstrate ultralow detection limits of 8 cell L-1 for S. aureus and 2 cell L-1 for E. coli. Such a bioassay indicates a point-of-care strategy of ultrasensitively biomedical detections by encoding specific SERS tags.

Abstract:Metal–organic frameworks (MOFs) are hybrid materials constructed by the linkage between an inorganic secondary building unit and an organic linker. A number of MOFs are luminescent in nature and can be structurally tuned for desirable geometry, surface functionality, and porosity. Luminescent MOFs have been endorsed for various biosensing applications. Lectins and carbohydrates have been used for the development of simple and convenient biosensing and bioimaging tools. Lectins are mostly present on the surface of microorganisms where they aid in pathogenesis. Due to this, they can be potential targets for a microbial biosensor. The present study, for the first time, explores the usage of a carbohydrate-conjugated FeMOF (Glyco-MOF) bioprobe for the selective determination of Pseudomonas aeruginosa and Escherichia coli . NH 2 -MIL-53(Fe) MOF was synthesized via a room temperature protocol and separately conjugated with galactose and mannose sugars via glutaraldehyde chemistry. The synthesized bioprobe is validated for structural integrity, luminescent nature, stability, and analyte assay. Electron microscopy studies validated the unhindered MOF's morphology and structural integrity, after bioconjugation. The synthesized bioprobes were able to detect P. aeruginosa and E. coli up to respective detection limits of 202 and 8 CFU/mL, respectively. The bioprobes are selective even in co-presence of possible interferants as well as being environmentally stable.
Highly Sensitive Label-free Biomolecular Detection Using Au-WS2 Nanohybrid Based SERS Substrates

Abstract:Recent advancements in nanotechnology have led to the development of surface-enhanced Raman spectroscopy (SERS) based rapid and low-cost technologies for ultra-sensitive label-free detection and identification of molecular analytes. Herein, we utilized the synergistic plasmonic and chemical enhancement effects of Au-WS2 nanohybrids to attain the high-intensity Raman signals of targeted analytes. To develop these nanohybrids, a series of monodispersed Au nanoparticles (NPs) of varying diameters from 20 to 80 nm was chemically synthesized and successively blended with liquid-phase exfoliated WS2 nano-flakes of average lateral size 90 nm. They provided a maximum enhancement factor (EF) of ~1.80×109 corresponding to the characteristic peaks at 1364 cm-1 and 1512 cm-1 for R6G analyte molecules. Theoretical studies based on the finite-difference time-domain simulations on Au-WS2 nanohybrid systems revealed a huge field-intensity enhancement with an EF of more than 1000 at the plasmonic hotspots, which was induced by the strong coupling of individual plasmon oscillations of the adjacent Au NPs upon light interactions. These electromagnetic effects along with the chemical enhancement effects of WS2 nanoflakes were found to be mainly responsible for such huge enhancement in Raman signals. Furthermore, these hybrids were successfully employed for achieving highly sensitive detection of the E. coli ATCC 35218 bacterial strain with a concentration of 104 CFU/mL in phosphate-buffered saline media, indicating their real capabilities for practical scenarios. The findings of the present study will indeed provide vital information in the development of innovative nanomaterial-based biosensors, that will offer new possibilities for addressing critical public health concerns.

Abstract:A novel label-free electrochemical DNA biosensor was constructed for the determination of Escherichia coli bacteria in environmental water samples. The aminated DNA probe was immobilized onto hollow silica microspheres (HSMs) functionalized with 3-aminopropyltriethoxysilane and deposited onto a screen-printed electrode (SPE) carbon paste with supported gold nanoparticles (AuNPs). The biosensor was optimized for higher specificity and sensitivity. The label-free E. coli DNA biosensor exhibited a dynamic linear response range of 1 × 10−10 μM to 1 × 10−5 μM (R2 = 0.982), with a limit of detection at 1.95 × 10−15 μM, without a redox mediator. The sensitivity of the developed DNA biosensor was comparable to the non-complementary and single-base mismatched DNA. The DNA biosensor demonstrated a stable response up to 21 days of storage at 4 °C and pH 7. The DNA biosensor response was regenerable over three successive regeneration and rehybridization cycles.

Abstract:A novel immunoassay protocol was demonstrated by the combination of fluorescent carbon nanodots (CNDs) and Ag@SiO2 surface-enhanced Raman scattering (SERS) tag nanoparticles into ensembles for a bifunctional nanoplatform. The CND-decorated Ag@SiO2 nanoparticles were constructed for sensitive fluorescence and SERS immunoassays. The silica shell thickness and amount of Ag@SiO2 nanoparticles were optimized for availability of strong fluorescence emission. The considerably large Raman scattering cross section of in situ-generated actual Raman reporter, 4,4'-dimercaptoazobenzene, from the apparent reporter p-aminothiophenol modified on the surfaces of Ag nanoparticles upon illumination of laser compensated for the reduction of SERS signals resulting from silica coating to a great degree. The antibody-modified bifunctional nanoparticles were captured by antibody-modified quartz slides in the presence of antigens in the sandwich structures for fluorescence and SERS immunoassays. The bifunctional nanoparticles could be used not only as bimodal probes for biodetection but also as bimodal tracers for bioimaging.
