Abstract:The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, and lipid metabolism in juvenile hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lancolatus ♂). Five isonitrogenous and isolipidic diets with graded levels of phospholipids (14.10, 32.10, 52.50, 70.20, and 91.30 g/kg) were fed to triplicate groups of juvenile grouper (initial body weight 13.00 ± 0.02 g) for 61 days. Results showed that dietary phospholipids produced no statistical influence on growth performance of grouper. The fatty acids profiles of experimental fish are correlated well with dietary fatty acids composition. The increase of dietary phospholipids significantly elevated the crude lipid content in whole fish body, liver and muscle. Meanwhile, the increased dietary phospholipids significantly promoted hepatic triglyceride (TG) accumulation. The results of gene expression analysis revealed that dietary phospholipids did not significantly affect the expression of lipogenesis related genes, FAS, ACC and DGAT2. In addition, high dietary phospholipids inclusion significantly decreased the expression of fatty acids oxidation related genes, PPARɑ and CPT1. In summary, juvenile grouper don’t have a specific requirement for dietary phospholipids which inclusion aggravated lipid deposition through the regulation of fatty acids oxidation at the transcriptional level.
Effects of High-Lipid Dietary Protein Ratio on Growth, Antioxidant Parameters, Histological Structure, and Expression of Antioxidant- and Immune-Related Genes of Hybrid Grouper

Abstract:The hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus) is a new species of grouper crossed from giant grouper (E. lanceolatus) as the male parent and brown-marbled grouper (E. fuscoguttatus) as the female parent. We hypothesized that optimal levels of dietary protein may benefit liver function. High-lipid diets are energetic feeds that conserve protein and reduce costs, and are a hot topic in aquaculture today. Therefore, the objective of the research is to investigated the effects of dietary protein level in high-lipid diets on serum and liver biochemistry, liver histology, and liver immune and antioxidant indexes and gene mRNA expression of the juvenile hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus). Six iso-lipidic (161 g/kg) diets were formulated containing graded levels of protein (510 as control, 480,450, 420, 390 and 360 g/kg). Each treatment consisted of three replicates and 30 fish (6.70 ± 0.02 g) in one replicate. After an 8-week feeding experiment, the results indicated the following: (1) With the decreasing of dietary protein level, the specific growth rate (SGR) of groupers increased gradually and then decreased; SGRs of the 390 and 360 g/kg groups were significantly lower than other groups (p < 0.05). (2) In terms of serum and liver, the activity of antioxidant enzymes such as catalase (CAT) and superoxide dismutase (SOD), and the total antioxidant capacity (T-AOC) content, and the activity of immune enzymes such as lysozyme (LYS) and immunoglobulin (IgM) was significantly increased under the appropriate protein level. (3) Based on liver histology, we know that high or low dietary protein levels cause liver damage. (4) Dietary protein levels can significantly affect the mRNA expression levels of an anti-inflammatory factor gene (tgfβ), pro-inflammatory factor genes (il6, il8), heat shock proteins, and antioxidant and immune genes (hsp70 and hsp90, gpx, nrf2, keap1). It is concluded that the appropriate protein level can promote the growth performance of groupers, improve antioxidant activity and immune enzyme activity in serum and liver, and enhance the expression of immune genes.
Integrated Response of Growth Performance, Fatty Acid Composition, Antioxidant Responses and Lipid Metabolism to Dietary Phospholipids in Hybrid Grouper (epinephelus Fuscoguttatus ♀ × E. Lanceolatus ♂) Larvae

Abstract:Dietary phospholipids supplementation is essential for normal development of fish larvae due to their limited phospholipid biosynthetic ability. The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, antioxidant capacity and lipid metabolism of larval hybrid grouper. The larvae (40 day after hatching, 0.37 +/- 0.01 g) were fed the diet with graded level of phospholipids (1.87, 3.61, 5.53, 7.25 and 9.69%) four times daily for 23 days. Results showed that the specific growth rate of grouper larvae was significantly improved with the increase of dietary phospholipids, and optimal dietary phospholipids was estimated to be 9.17%. Fatty acids composition of whole fish body and liver was highly reflective of dietary fatty acid profile. In addition, the increase of dietary phospholipids resulted in a significantly elevated hepatic crude lipid and triglyceride content in a dose-dependent manner. The analysis of gene expression revealed that increased dietary phospholipids did not produce any statistical effect on the expression of lipogenesis genes including LXRalpha, FAS and ACC, but significantly facilitated the expression of DGAT2, a rate-limiting enzyme involved in triglyceride synthesis. The expression of genes, CPT1, involved in I3-oxidation was not significantly influenced as dietary phospholipids increased. However, phospholipids inclusion significantly reduced the expression of ATGL, an rate-limiting lipolytic enzyme. Additionally, the supplementation of dietary phospholipids significantly elevated the activity of antioxidant relate enzymes, CAT, SOD and T-AOC, but significantly decreased the MDA content, an end product of fatty acid peroxidation. In summary, dietary phospholipids supplementation was beneficial in improving growth performance and antioxidant capacity of grouper larvae, and meanwhile, promoted hepatic lipid deposition through regulating the synthesis and hydrolysis of triglyceride.

Influence of different oil sources on growth, disease resistance, immune response and immune-related gene expression on the hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatu), to Vibrio parahaemolyticus challenge

Abstract:The aim of this study was to investigate the effects of feeding alternative dietary oils to hybrid grouper fish (♀Epinephelus fuscoguttatus × ♂E. lanceolatu) on their growth, histological morphology of hepatocytes, disease resistance, immune response, and expression of immune-related genes. Seven experimental fish meal-based isonitrogenous and isolipidic diets were formulated containing 5% fish oil (FO; acting as controls) and various vegetable oils (VOs): corn oil (CO), sunflower oil (SO), tea oil (TO), olive oil (OO), rice oil (RO), and mixed oil (MO); comprising equal amounts of these oils). Each diet was fed to triplicate groups of 40 fish (initial mean body weight ± standard error = 15.09 ± 0.01 g) for eight weeks. The results show that 1) alternative dietary oils had no significant effects on weight gain rate, specific growth rate, protein efficiency ratio, and survival rate compared with controls (P > 0.05). The weight gain rate (WGR) and specific growth rate (SGR) of the SO group were lower than in the CO and OO groups. 2) These were no differences in morphological indexes among groups; except for the CO group, in which the condition factor and hepatosomatic index were lower than those in other groups. 3) Compared with controls, the whole-body moisture and crude protein contents in the VO groups were higher, while their crude lipid contents were lower. 4) The fatty acid contents in liver and muscle were affected by lipid type, and the contents of eicosapentaenoic acid and docosahexaenoic acid in liver and muscle in the VO groups were markedly lower than in controls. 5) Compared with control group, VO groups damaged the histological morphology of hepatocytes. 6) After a challenge with the Vibrio parahaemolyticus bacterium, there were no differences in mortality among groups. However, VO enhanced the activity of non-specific immune enzymes while down-regulating the expression of Nrf2 and inducing the expression of pro-inflammatory factors (IL1β, TNFα, TLR22, and MyD88) in the kidney. It can be concluded that dietary VO substitution does not affect the growth of fish but damaged the histological morphology of hepatocytes and induced the expression of pro-inflammatory factors in tissues. Finally, OO and CO were recommended as the appropriate lipid replacement for FO.

Abstract:The time honoured strategy of "no-touch" in the surgery of colon cancer does not stand an anatomical and patho-physiological criticism, also on the basis of some results of the literature. The personal experience of the problem leads to the proposal of an alternative realization of the procedure, that is an up-to-date of the classic method, and is really worth and easy to be overall adopted. The suggestion is to seal bipolarly the colon tract with the tumour by mean of the original grasping tie from the very beginning of the operation, and then to temporarily clamp the mesocolon close to the gut, before going on with the surgical dissection, lymphectomy and exeresis as usual. This procedure is able to completely isolate from the first steps of the operation the affected tract of the colon, so opposing the possible spreading of tumour cells in the portal and in the systemic venous stream during the following surgical manipulations. The rationale of the proposal is that of a more efficacy of the "no-touch" strategy based on strict anatomical and patho-physiological considerations.

Abstract:There is a trend towards using high-lipid diets in aquaculture, which can be optimized by using certain additives. This study investigated the effect of vitamin E (VE) supplementation of a high-lipid diet on the growth, serum and liver biochemical indexes, hepatic histology, and lipid metabolism of hybrid groupers (♀ Epinephelus fuscoguttatus × ♂ Epinephelus lanceolatus ). Six groups of iso-protein (50.23%) and iso-lipidic high-lipid (15.36%) experimental diets were prepared by adding VE at concentrations of 49.6 (basic diet, controls), 100, 110, 163, 206, and 245 mg/kg. Each treatment consisted of three replicates and 30 fish (10.20 ± 0.02 g) in an 8-week feeding experiment. The results show that 1) compared with controls, growth performance was not affected by VE addition. However, the weight gain rate (WGR) and specific growth rate (SGR) were significantly lower in the high-VE group (245 mg/kg) than in the 163 and 206 mg/kg groups. 2) Compared with controls, VE significantly increased the contents of alpha-tocopherol in the liver and muscle. 3) Adding VE can reduce the fatty livers caused by high-lipid diets and significantly reduce total serum cholesterol, reduce the activity of lipid synthesis enzymes, and significantly increase the activity of lipolytic enzymes in the liver. 4) Compared with controls, appropriate amounts of VE significantly increased the expressions of the liver lipid-metabolism-related genes PPAR-α , PPAR-β , PPAR-γ , H-FABP , A-FABP , and L-FABP . Based on observations of lipid metabolism enzyme activity, histological sections, and lipid-metabolism-related gene expression, we conclude that the addition of 163–206 mg/kg VE to a high-lipid diet can promote lipid metabolism in groupers.

Abstract:Background At present, fish meal (FM) resources are in short supply, and competition for food between humans and animals is becoming increasingly critical. Finding non-grain protein sources that can replace FM is the key to solving the rapid development of aquaculture. Methods Seven trial diets were prepared with 0 g/kg (EP0), 30 g/kg (EP3), 60 g/kg (EP6), 90 g/kg (EP9), 120 g/kg (EP12), 150 g/kg (EP15), and 180 g/kg (EP18) of enzyme-digested poultry by-product meal (EPBM) by replacing of FM. A total of 630 hybrid groupers ( Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) were equally portioned into 21 tanks. At 8:00 and 16:00 each day, groupers were fed until they were full for a cumulative period of 8 weeks. Results The results showed that 30 g/kg of EPBM significantly increased the rates of weight gain and special growth ( P < 0.05). Significantly higher activities of serum glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, catalase, and superoxide dismutase were observed in the EP3 group ( P < 0.05). The categories and numbers of the top 10 dominant bacteria in the phylum and genus levels were not significantly influenced by feed ( P > 0.05). In the proximal intestine and distal intestine, there were significantly higher expressions of SNAT3, LAAT1, CAT2 , and CAT1 in the EP3 group compared with the EP0 group ( P < 0.05). In the EP3 group, the expressions of PepT1, LAAT1, B 0, + AT , and CAT2 were significantly increased in MI than those in all other groups (except the EP0 group, P < 0.05). Conclusion When FM was replaced by 30 g/kg of EPBM, growth performance, antioxidant capacity, and the ability to transport amino acids and peptides of hybrid grouper were significantly improved.

Abstract:This study aimed to assess the impact of α-lipoic acid on the growth performance, antioxidant capacity and immunity in hybrid groupers (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus) fed with a high-lipid diet. Groupers (8.97 ± 0.01 g) were fed six different diets, with α-lipoic acid content in diets being 0, 400, 800, 1200, 1600, and 2000 mg/kg, named S1, S2, S3, S4, S5, and S6, respectively. The results show that the addition of 2000 mg/kg α-lipoic acid in the diet inhibited the growth, weight gain rate (WGR), and specific growth rate (SGR), which were significantly lower than other groups. In serum, catalase (CAT) and superoxide dismutase (SOD) were significantly higher in the S5 group than in the S1 group. In the liver, CAT, SOD and total antioxidative capacity (T-AOC) levels were significantly increased in α-lipoic acid supplemented groups. α-lipoic acid significantly upregulated liver antioxidant genes sod and cat, anti-inflammatory factor interleukin 10 (il10) and transforming growth factor β (tgfβ) mRNA levels. Conclusion: the addition of 2000 mg/kg of α-lipoic acid inhibits the growth of hybrid groupers. In addition, 400–800 mg/kg α-lipoic acid contents improve the antioxidant capacity of groupers and have a protective effect against high-lipid-diet-induced liver oxidative damage.

Abstract:This study investigated the effect of lipid levels on growth, body composition, lipid metabolism enzymes activity and related genes expression in hybrid grouper (♀ Epinephelus fuscointestinestatus × ♂ E. lanceolatu ). Three diets with different lipid levels of 6% (low-lipid feed, LF), 11% (medium-lipid feed, MF) and 16% (high-lipid feed, HF) were designed and each diet was fed to triplicate groups of 30 fish each (6.73 ± 0.00g) for eight weeks. The results revealed the following. 1) The maximum weight gain rate (WGR), specific growth rate (SGR), protein efficiency ratio (PER) and the lowest feed conversion ratio (FCR) values were found in the HF group. There was no significant difference in the survival rate (SR) among the groups ( P > 0.05). 2) Condition factor (CF), hepatosomatic index (HSI) and visceralsomatic index (VSI) increased with increasing dietary lipid. 3) Crude lipid content in the whole-body and muscle increased significantly as the dietary lipid level increased ( P < 0.05). 4) Principal component analysis of fatty acids showed that sum n-3 polyunsaturated fatty acids (∑n-3PUFA)/sum n-6 polyunsaturated fatty acids (∑n-6PUFA) was significantly higher in the HF group than in the other two groups ( P < 0.05). 5) Biochemical serum indexes showed higher triglycerides (TG), total cholesterol (TC), alkaline phosphatase (AKP) and low-density lipoprotein-cholesterol (LDL-C) levels but lower high-density lipoprotein-cholesterol (HDL-C) level in the HF group. 6) Liver lipid metabolism enzymes activity and biochemical indicators showed that fish fed the MF diet had higher lipoprotein lipase (LPL), hepatic lipase (HL) and triglyceride lipase (ATGL) activity but lower fatty acid synthase (FAS) activity and very-low density lipoprotein-cholesterol (VLDL-C) level. 7) The relative expression levels of lipoprotein lipase ( lpl ), adipocyte-type fatty acid-binding protein ( a-fabp ), liver-type fatty acid-binding protein ( l-fabp ), heart-type fatty acid-binding protein ( h-fabp ) and hormone-sensitive lipase ( hsl ) genes in the fish liver were higher in the MF group with increasing dietary lipid ( P < 0.05), while the fatty acid synthase ( fas ) gene was expressed at lower relative level ( P < 0.05). 8) Histological oil-red sections of the liver showed that fish in the HF group had larger and more hepatic lipid droplets than those fed the other diets. It was concluded that fish fed the HF diet had better growth performance but this diet was not conducive to the progress of lipid metabolism, resulting in lipid accumulation and affecting the quality of the fish. This study provides useful information for the formulation of environmentally friendly and cost-effective diets for hybrid grouper.

Screening of host gut-derived probiotics and effects of feeding probiotics on growth, immunity, and antioxidant enzyme activity of hybrid grouper (Epinephelus fuscoguttatus♀ × E. lanceolatus♂)

Abstract:In recent years, the widespread use of antibiotics in intensive grouper mariculture has resulted in the ineffectiveness of antibiotic treatment, leading to an increasing incidence of diseases caused by bacteria, viruses, and parasites, causing serious economic losses. Hence, it is crucial to develop alternative strategies to antibiotics for healthy and sustainable development of the mariculture industry. Here, we aimed to screen host gut-derived probiotics and evaluate its effects on growth and immunity of grouper. In this study, 43 bacterial strains were isolated from the intestine of the hybrid grouper (Epinephelus fuscoguttatus♀ × E. lanceolatus♂), and a potential probiotic strain G1-26, which can efficiently secrete amylase, protease, and lipase, was obtained using different screening mediums. Based on 16S rDNA sequencing, the potential probiotic strain G1-26 was identified as Vibrio fluvialis. The results of a biological characteristic evaluation showed that V. fluvialis G1-26 could grow at 25-45 °C, pH 5.5-7.5, salinity 10-40, and bile salt concentration 0-0.030%, and produce amylase, lipase, and protease under different culture conditions. Additionally, V. fluvialis G1-26 is sensitive to many antibiotics and does not exhibit aquatic biotoxicity. Subsequently, hybrid groupers were fed diets containing V. fluvialis G1-26 at different concentrations (0, 106, 108, and 1010 CFU/g) for 60 d. The results showed that V. fluvialis G1-26 at 108 CFU/g did not significantly affect the growth performance of the hybrid grouper (P > 0.05). V. fluvialis G1-26 supplementation at 108 and 1010 CFU/g significantly promoted the relative expression of immune-related genes in hybrid groupers (TLR3, TLR5, IL-1β, IL-8, IL-10, CTL, LysC, TNF-2, and MHC-2) and improved the activities of alkaline phosphatase, acid phosphatase, total superoxide dismutase, and total protein in the liver. In conclusion, V. fluvialis G1-26, a potential probiotic strain isolated from the intestine of the hybrid grouper, can be used as an effective immunopotentiator at an optimal dose of 108 CFU/g diet. Our results provide a scientific basis for the development and utilization of probiotics in the grouper mariculture industry.
Effects of Chinese herbal medicines on growth performance, intestinal flora, immunity and serum metabolites of hybrid grouper (Epinephelus fuscoguttatus♀×Epinephelus lanceolatu♂)

Abstract:The immunomodulatory roles of Chinese herbal Medicine (CHM) in aquatic animals have been well-recorded. However, how CHM impacts the intestinal microbiota and serum metabolism is not fully understood. In this study, the effects of different additive levels of CHM on the growth performance, immunity, intestinal flora and serum metabolism of hybrid grouper (♀Epinephelus fuscoguttatus × ♂Epinephelus lanceolatu) were investigated. The addition of 0.5%, 1.0%, 1.5% and 2.0% Chinese herbal medicine compound to feed could significantly improve the weight gain rate (WGR), specific growth rate (SGR) and survival rate (SR) of grouper, reduced feed coefficient, while had no significant difference on morphometric parameter. The most significant improvement for the parameters above was observed in 1.5% group. Different addition levels of CHM could also significantly enhance the activities of ACP, AKP, SOD, CAT and LZM in serum. Accordingly, the supplementation of CHM significantly induced up-regulation of immune genes such as IL-8, IL-1β, TNF-α, Nrf2, Lzm in the liver, spleen and head kidney of grouper, improved the resistance of grouper to V. harveyi as well. The intestinal flora analysis showed that at the phylum level, the main dominant species of intestinal microorganisms were Firmicutes, Proteobacteria, Bacteroidota, Actinobacteriota, Gemmatimonadota, Desulfobacterota, Fusobacteriota and Myxococcota. At the genus level, the high abundance was Lactobacillus, Streptococcus, Bacteroides, Escherichia, Romboutsia, Sphingomonas and Muribaculaceae. The abundance of probiotics (such as Lachnospiraceae, Lactobacillaceae, Streptococcaceae, etc) in CHM-supplement groups were higher (highest in 1.5% group) compared with control group. Moreover, a total of 11 common differential metabolic pathways were screened by LC-MS metabolism analysis of serum, they were Neuroactive ligand-receptor interaction, Purine metabolism, Linoleic acid, Glycerophospholipid metabolism, Taurine and hypotaurine metabolism, Arginine and proline metabolism, ABC transporters, Aminoacyl-tRNA biosynthesis, Arachidonic acid metabolism, Drug metabolism-cytochrome P450, alpha-Linolenic acid metabolism. Also, three common differential metabolites (PI(20:4(5Z,8Z,11Z,14Z)/18:1(11Z)), PC(20:3(8Z,11Z,14Z)/22:1(13Z)), PC(22:0/20:4(5Z,8Z,11Z,14Z)) associated with intestinal health, growth and disease resistance was found. These data will contributes to a comprehensive understand for the regulatory roles of CHM on fish, which is also beneficial for the disease control and sustainable development of aquaculture.

Abstract:Poly-β-hydroxybutyrate (PHB) is a bacterial metabolite produced by bacteria such as Halomonas sp. that serves as a carbon and energy storage compound for bacteria under nutrient-limited conditions. Two experiments were conducted to investigate the effects of dietary supplementation with Halomonas-PHB on hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lanceolatu ♂). In experiment I, juvenile groupers were fed basal diets supplemented with 3% Halomonas-PHB (3% HM-PHB) containing 1.4% PHB and 3% Halomonas (3% HM) without PHB, as well as a control diet, for seven weeks. The results showed no significant difference in survival rate, weight gain, and crude fat content between the 3% HM-PHB group and the control group; however, the crude protein of the 3% HM-PHB group was significantly lower than that of the control group. Furthermore, supplementation with 3% HM-PHB increased the fatty acids content in fish muscles, including long-chain unsaturated fatty acids C18:1n9, EPA, and DHA. In experiment II, groupers were fed a basal diet supplemented with 6.5% Halomonas-PHB (6.5% HM-PHB) containing 3% PHB and 6.5% Halomonas (6.5% HM) containing no PHB, as well as a basal diet (Control). After seven weeks of rearing, the fish were challenged with Vibrio anguillarum for 48 h. Although no significant difference in survival rate and growth was observed among different groups, the dietary supplement of 6.5% Halomonas-PHB improved the survival rate of V. anguillarum challenged grouper and significantly increased the gene expressions of catalase (CAT) and superoxide dismutase (SOD) in blood, interleukin 1 (IL1) and interleukin 10 (IL10) in the liver, spleen, head kidney, and blood (p < 0.05). In conclusion, dietary supplementation of Halomonas-PHB had no significantly positive effect on fish growth performance but increased the content of fatty acids, including long-chain unsaturated fatty acids C18:1n9, EPA, and DHA in fish muscle; it also improved the V. anguillarum resistance, possibly through increasing immune-related gene expression in different tissues and organs. Our findings offer compelling evidence that Halomonas-PHB can be utilized as a feed additive in intensive grouper farming to enhance the groupers' resistance to Vibrio.

Abstract:The supplementation of gossypol in excess is noted to cause detrimental effects such as the reduction of antioxidant enzymes and disruption of lipid metabolism in animals. Studies regarding the effects of different levels of gossypol are very rare; thus, this study was conducted to evaluate the effects of low and high dietary levels of gossypol and of supplementation with 0.13 % sodium butyrate (NaB) under high gossypol conditions on the growth performance and intestinal health of hybrid grouper ( Epinephelus fuscoguttatus ♀× Epinephelus lanceolatus ♂). Four treatments were used: Feed containing 40% fish meal was used as the control group [fishmeal (FM)], the FM diet plus 0.03% gossypol acetic acid (abbreviated as gossypol) as the low-level gossypol group (gL), FM + 0.15% gossypol was used as the high-level gossypol group (gH), and FM+0.15 % gossypol with 0.13 % NaB as the repair group (gHNaB). All diets were isonitrogenous and isolipidic. The results showed that the gL treatment significantly increased specific growth rate (SGR) and feed utilization; upregulated mRNA levels of distal intestinal transforming growth factor-β1 ( tgf β 1 ), jam, occludin, claudin3 , and zo1 ; and downregulated mRNA levels of il8, ifn γ, and akt . The gH treatment significantly reduced SGR and feed utilization; increased distal intestinal total nitric oxide synthase (NOS) activity and nitric oxide (NO) content; upregulated mRNA levels of distal intestinal tnf α, il1 β, il6, ifn γ, caspase2, caspase9 , and akt ; and downregulated mRNA levels of tgf β 1, jam , and zo1 . NaB supplementation significantly increased distal intestinal total NOS activity and NO content; downregulated distal intestinal tnf α, il1 β, ifn γ, pi3k p85 , and akt mRNA levels; and increased distal intestinal tgf β 1, jam, occludin , and zo1 mRNA levels. Above all, low- and high-level gossypol exhibited positive and negative effects on growth performance, distal intestinal anti-inflammatory capacity, and tight junctions, respectively, in hybrid groupers. NaB supplementation improved distal intestinal anti-inflammatory capacity and tight junctions in hybrid groupers to a certain extent.

Abstract:This experiment was conducted to determine the optimum amount of peanut cake (PNK) in the diet of hybrid grouper by studying the effect of PNK on the growth, antioxidant capacity, liver fat metabolism, and intestinal microbiota composition by replacing fish meal (FM) protein in the diet of the juvenile hybrid grouper. A total of 840 juvenile hybrid grouper with uniform body weight (13.20 ± 0.05 g) were selected from 7 treatments and 4 replicates (30 fish each) farmed for eight weeks. Results showed that the weight gain rate (WGR) of the low-substitution group was significantly higher than that of the high-substitution group (P < 0.05) and that the feed conversion and specific growth rates of the 44%, 55%, and 66% groups were significantly higher than those of the other groups (P < 0.05). The crude lipid content of the 44% group was significantly higher than those of other groups (P < 0.05), and the crude protein content of the 66% group was lowest and significantly lower than that of the control group (P < 0.05). The total protein content showed a decreasing trend and then an increasing trend. The total cholesterol content showed a gradually decreasing trend and was significantly higher in the control and 11% groups than those in the other groups (P < 0.05). The glutamic oxalacetic transaminase content in the 66% group was significantly higher than those in other groups (P < 0.05). The total antioxidant capacity and the malondialdehyde content of the 66% group were significantly higher (P < 0.05) and significantly lower (P < 0.05), respectively, than those of other groups. The oil red-stained sections of the liver showed that the deposited fat gradually increased with increasing levels of PNK replacement in the diet. A replacement level higher than 22% caused a decrease in WGR. When the replacement level was higher than 44%, a significant increase (P < 0.05) in harmful bacteria in their intestines was observed, and when the replacement level was higher than 55%, a significant increase (P < 0.05) in liver fat accumulation was observed. Based on the PNK replacement level corresponding to WGR, the appropriate PNK replacement level in the diet was 15%.

Abstract:The effect of hydrolyzed fish protein powder (HFP) on the growth, intestinal development, gene mRNA expression, and enzyme activity in the intestine and liver of juvenile hybrid grouper ( Epinephelus fuscoguttatus ♀ and Epinephelus lanceolatus ♂) was assessed after an 8-week feeding trial. Seven isonitrogenous (50%) and isolipidic (9%) diets were fed to hybrid grouper with 0% (CT), 1% (H1), 1.5% (H2), 2% (H3), 2.5% (H4), 3% (H5), and 4% (H6) HFP. No significant difference ( p > 0.05) in weight gain rate (WGR), specific growth rate (SGR), feed conversion ratio (FCR), and survival rate (SR) was observed in all the groups. The crude protein content in the H6 group was significantly higher than in the other groups ( p < 0.05). Intestinal lipase and trypsin activity were significantly higher in H3 and H5 groups ( p < 0.05). In the serum, superoxide dismutase (SOD) activity was significantly higher in H5 and H6 groups, while malondialdehyde (MDA) activity was lower ( p < 0.05) compared to other treatments. Insulin-like growth factor ( IGF-I ) and target of rapamycin ( TOR ) mRNA expression levels in the intestine and muscle were significantly higher in the H2 group and H1 group ( p < 0.05), respectively. The most abundant intestinal bacteria found at the genus level are Acinetobacter , Vibrio , and Flavobacteriaceae. The villus was significantly longer in hybrid grouper fed with different levels of HFP compared to the control, and fish in the H2 group had thicker intestinal muscle compared to the other groups ( p < 0.05). In conclusion, the addition of HFP to the low fishmeal (FM) diets of juvenile grouper improved the intestinal development and increased the levels of intestinal digestive enzymes.

Abstract:We investigated the effects of fish protein hydrolysate (FPH) on growth performance and humoral immune response of the large yellow croaker (Pseudosciaena crocea R.). One thousand and two hundred large yellow croakers [initial average weight: (162.75±23.85) g] were divided into four groups and reared in floating sea cages (3 m×3 m×3 m). The animals were fed with 4 diets: basal diet only (control) or diets supplemented with 5%, 10% and 15% (w/w) FPH. The results show that dietary FPH levels significantly influenced the growth and immunity of the large yellow croaker. Compared with the control group, total weight gain (TWG) in all treatment groups, relative weight gain (RWG) and specific growth rate (SGR) in fish fed with diets supplemented with 10% and 15% FPH were significantly increased (P<0.05). Similar results were observed in immune parameters [lysozyme activity, serum complements, immunoglobulin M (IgM)]. Lysozyme activity, complement C4 and IgM were also significantly increased (P<0.05) in fish fed with diets supplemented with 10% and 15% FPH, while complement C3 level was significantly increased (P<0.05) in all treatment groups. In general, with the supplementation of FPH, particularly at dose of 10%, the growth performance and immunity of the large yellow croaker can be improved effectively.
Effect of Fluosol‐DA/O2 on tumor‐cell and bone‐marrow cytotoxicity of nitrosoureas in mice bearing FSA‐II fibrosarcoma

Abstract:The perfluorochemical emulsion, Fluosol‐DA, combined with carbogen breathing, potentiates the effects of radiation and a number of chemotherapeutic agents in several rodent tumors. The interaction of Fluosol‐DA with drugs may be quite complex. In addition to increasing the oxygen supply in the tumor, Fluosol‐DA may alter the pharmacokinetics of the drug and function as a drug delivery system. A series of 4 nitrosoureas of varying lipophilicity were administered as single doses intravenously (i.v.) to C3H/Be/FeJ mice bearing subcutaneous FSa‐11C fibro sarcomas. Doses of 40 mg/kg of CCNU, 15 mg/kg of BCNU, 20 mg/kg of MeCCNU and 15 mg/kg of chlorozoticin followed by 2 hr of breathing 95% oxygen produced tumor growth delays of 7.5, 4.0, 3.8 and 2.7 days, respectively. When the drug injection was followed immediately by 0.3 ml of Fluosol‐DA and 2 hr of breathing 95% oxygen, the tumor growth delay produced by CCNU, BCNU, MeCCNU and chlorozoticin increased 2‐fold, 10‐fold, 4.5‐fold and 3.5‐fold, respectively. Administration of the drugs in Fluosol‐DA followed by 2 hr of 95% oxygen breathing resulted in a 3.5‐fold increase in tumor growth delay with CCNU, a 17‐fold increase with BCNU, a 12.5‐fold increase with MeCCNU and a 6‐fold increase with chlorozoticin compared to drug and 95% oxygen breathing. These results are quantified in terms of cell survival by the tumor excision assay. Effects on the bone marrow from each treatment were measured using the granulocyte‐monocyte colony‐forming units (CFU‐GM) assay. There was no correlation between the lipophilicity of the nitro sources tested and the tumor growth delay produced by each treatment.

Abstract:The effects of dietary tributyrin (TB) on growth, intestinal health, inflammatory response and antioxidant status in juvenile Pelteobagrus fulvidraco were investigated. The fish (initial weight 5.53 ± 0.02 g) were fed with diets containing 0 (control, TB0), 500 (TB1), 1000 (TB2) and 2000 (TB3) mg/kg tributyrin for 56 days. The results showed that significantly higher final weight, specific growth rate, protein efficiency ratio and protein retention were observed in fish fed with TB2 diet compared with those in TB0 ( P < 0.05). Significantly higher lipid retention, whole-body fat content, intraperitoneal fat weight, intraperitoneal fat index and intestine length index were found in TB2 than that in TB3 ( P < 0.05). Intestinal length, weight, villus length, muscular thickness, intestosomatic index and intestinal activities of trypsin, lipase, alkaline phosphatase, Na + /K + -ATPase in TB1 or TB2 were significantly higher than those in TB0 ( P < 0.05). The fish fed with TB2 or TB3 diet had significantly lower intestinal interleukin-1β, interleukin-6, tumou necrosis factor-α, malondialdehyde contents and significantly higher hepatic glutathione, superoxide dismutase activities than those in TB0 ( P < 0.05). Compared with TB0, the relative expressions of claudin-1, zomula occludens protein-1, occludin, lipoprotein lipase and fatty acid synthase were significantly increased in TB1, TB2 and TB3 ( P < 0.05). Together, 1000 mg/kg TB improved growth and lipid deposition, but decreased inflammatory response, probably through modulating tight junction proteins and lipid metabolism related genes expression and activating antioxidant system.
