Abstract:The widespread utilization of silver nanoparticles (AgNPs) in industrial and commercial products inevitably raises the release into wastewater that might cause potential negative impacts on sewage treatment system. In this paper, long-term exposure experiments at four levels were conducted to determine whether AgNPs caused adverse impacts on nutrient removals in sequencing batch reactors (SBRs) and changes of microbial community structure. Compared with the control reactor (without AgNPs), carbon, nitrogen and phosphorus removal in presence of 0.1mg/L AgNPs was no difference. However, presence of 1.0 and 10mg/L AgNPs decreased the average removal efficiencies of COD from 95.4% to 85.2% and 68.3%, ammonia nitrogen from 98.8% to 71.2% and 49%, SOP from 97.6% to 75.5% and 54.1%, respectively. It was found that AgNPs could accumulate in sludge with the distribution coefficients of 39.2-114L/g, inhibit the protein and polysaccharide production in EPS, reduce the SOUR of sludge, and greatly increase LDH release from microbial cells. The illumina high-throughput sequencing results indicated that AgNPs concentration changed the structures of bacterial communities, associating with the effects of AgNPs on reactor performance. Sequence analyses showed that Proteobacteria, Bacteroidetes and Acidobacteria were the dominant phyla. It was notable that AgNPs addition reduced the contents of several nitrifying bacteria at genera level in sludge, leading to the lower removal of nitrogen.

Abstract:This work investigated the roles of sludge micropowder addition in microbial structure and partial nitrification and denitrification (PND) in an extended filamentous aerobic granular sludge-sequencing batch reactor (AGS-SBR) using high-ammonia wastewater. Type 1683 Acinetobacter with a high percentage became the dominant extended filaments, remarkably shifted and remained at a low level, acting as a framework for AGS recovery after micropowder addition. The sludge volume index (SVI5) decreased from 114 to 41.7 mL/g, mixed liquid suspended solids (MLSS) and extracellular polymers (EPS) both increased and balanced at 6836 mg/L and 113.4 mg/g•MLVSS, respectively. COD and NH4+-N were degraded to certain degrees in the end. However, the effluent NO2−-N accumulated to the peak value of 97.6 mg/L on day 100 (aeration stage), then decreased and remained at 45.3 mg/L with development of the stirring and micropowder supplemented in the SBR on day 160 (anoxic stage), while the influent NO2−-N always remained at zero. Interestingly, the influent/effluent NO3−-N both remained at zero throughout the whole experiment. These results demonstrated that PND was successfully obtained in this work. Sludge micropowder addition not only restrained the extended filaments’ overgrowth, but also contributed to PND realization with carbon released. Citrobacter and Thauera played an essential role in the PND process for high-ammonia wastewater treatment. Running condition, wastewater characteristic, and sludge structure played an important role in microbial composition.

Abstract:Patient population The study population consisted of 26 HIV-infected children who were patients of the Pediatric HIV Clinic at Strong Memorial Hospital (SMH) in Rochester, NY. The subjects were followed for a mean duration of 28.8 + 10.1 months during the course of the 36-month study period (April 1, 1993 to March 31, 1996). H1V infectivity was defined as a positive ELISA (HIV) confirmed by Western Blot for patients 15 months old or older and positive HIV culture or polymerase chain reaction (PCR) for patients younger than 15 months of age. The patient population comprised 12 males and 14 females ranging in age from 2 months to 13.5 years (mean: 4.8 + 3.6 years) at their time of entry into the study. All of the patients acquired HIV via maternal transmission and were taking antiretroviral drugs during the study period.

Abstract:The increasing application of engineered nanoparticles (NPs) has posed an emerging challenge to wastewater treatment processes. The short-term impacts of CuNPs, CuONPs, ZnONPs and AgNPs on anaerobic ammonium oxidation (anammox) process were investigated firstly in this study. CuONPs, ZnONPs and AgNPs up to 50 mg g(-1) suspended solid (SS) did not affect anammox activity, reactive oxygen species (ROS) production or cell membrane integrity. However, 1.25 mg g(-1) SS CuNPs significantly inhibited the anammox activity and the loads that caused 50% inhibition were 4.64 +/- 1.24 and 3.27 +/- 0.79 mg g(-1) SS for anammox granules and flocs, respectively. 5 mg g(-1) SS CuNPs caused serious accumulation of the toxic intermediate N2H4. Furthermore, CuNPs interacted with extracellular polymeric substances by specifically bonding to tyrosine or tryptophan-containing groups, C-O-C in polysaccharides and -OH in polymeric compounds. Therefore, this study calls for more attention to the risks of NPs to the anammox-based processes. (C) 2017 Elsevier Ltd. All rights reserved.
Engineered silver nanoparticle (Ag-NP) behaviour in domestic on-site wastewater treatment plants and in sewage sludge amended-soils

Abstract:Untreated sludge from small-scale on-site domestic wastewater treatment systems (septic tanks) was spiked with 20, 60 and 100 nm silver nanoparticles (Ag-NPs) to investigate Ag-NP behaviour in these systems that are widely distributed in rural areas. In addition, the release of Ag-NPs from a previously spiked clay-rich loam reference soil (LUFA 2.4) was evaluated, in the presence and absence of untreated sludge, to simulate the common practice of sludge disposal by spreading on agricultural land. Single particle ICP-MS was used to determine Ag-NP size distribution and the results were compared with total Ag (Ag-NP and ionic) measured in acid digested samples. As documented previously for large municipal scale wastewater treatment plants, Ag-NPs are found to be overwhelmingly (~98%) retained in the sludge in these small-scale systems. The Ag-NP retention efficiency on the LUFA reference soil amended with sludge is approximately 10 times greater than that of LUFA soil alone (in the absence of sludge). For soil spiked with 60 nm Ag-NPs, the calculated average diameter of Ag-NPs in the supernatant, after 24 h was 45 ± 3 nm (dissolution rate 7.2E-06 mol/m2·h for 60 nm Ag-NP), smaller than that of supernatant from the combined sludge/soil system (52 ± 2 nm), indicating lower Ag-NP dissolution rates in the sludge-amended soil. This study provides new information about the leachability of Ag-NPs from septic tank sludge and suggests that the effluent and sludge from septic tanks are potential sources of both nano- and dissolved ionic-Ag to environmental waters.
Removal of silver nanoparticles in simulated wastewater treatment processes and its impact on COD and NH(4) reduction.

Abstract:The increasing utilization of silver nanoparticles in industrial and consumer products has raised concern to wastewater treatment utilities due to its antimicrobial activity. In this work, the removal of citrate stabilized silver nanoparticles (Ag-NPs) during the wastewater treatment processes and its impact on treatment performance were examined. During simulated primary clarification, over 90% of the Ag-NPs remained in the wastewater, indicating that the majority of silver nanoparticles in sewage would enter the subsequent treatment units. During sequencing batch reactor processes, silver nanoparticles were effectively removed in each cycle throughout the 15-d experimental duration. Continuous input of silver nanoparticles into the wastewater did not significantly alter chemical oxygen demand (COD) removal. NH(4) removal was reduced at the beginning of the SBR experiment but quickly recovered at the later stage of the experiment. This study demonstrated that in the near future it is unlikely that citrate-stabilized Ag-NPs released into sewage will cause significant adversary effects on the COD and NH(4) removal of activated sludge processes in municipal wastewater treatment plants.

Abstract:In this study, the effects of silver (Ag NPs) and sliver sulfide nanoparticles (Ag2S NPs) on nitrogen removal and nitrogen functional microbes in constructed wetlands were investigated. The obtained results demonstrated that inhibition extent on nitrogen removal relied on NPs types and high concentrations NPs showed higher negative effects. 0.5 mg/L Ag NPs had no influence on NH4+-N removal, amoA and nxrA gene copies, whereas Ag2S NPs and Ag+ decreased NH4+-N removal by reducing abundances of nitrifying genes. The concentrations of NO3--N and TN in all 0.5 mg/L obviously increased compared with control, resulting from decreasing functional genes and denitrifying bacteria. And 0.5 mg/L Ag NPs exhibited largest inhibitory effects, with the highest NO3--N effluent concentrations. 2 mg/L Ag NPs decreased NH4+-N removal, but adverse effects gradually vanished with extension of time, whereas both Ag2S NPs and Ag+ at 2 mg/L influenced NH4+-N transformation and decreased the abundance of amoA and nxrA genes and the AOB Nitrosomonas in CWs. Moreover, 2 mg/L of Ag NPs reduced NO3--N removal by decreasing abundance of nirS and key denitrifying bacteria. To sum up, the inhibition mechanisms concluded from current results were possibly in that Ag NPs exhibited nanotoxicity rather than ionic toxicity.
Comparative analysis of upward and downward vertical flow constructed wetlands on the nitrogen removal and functional microbes treating wastewater containing Ag nanoparticles

Abstract:This study investigated impacts of silver nanoparticles (AgNPs) on nitrogen removal within constructed wetlands (CWs) with different flow directions. The obtained results showed that addition of AgNPs at 0.5 and 2 mg/L significantly inhibited NH4+-N removal, resulting from lower abundances of functional genes (amoA and nxrA) within CWs. And higher abundances of amoA and nxrA genes at 0.5 mg/L were observed in downward flow CW, leading to better NH4+-N removal, compared to upward flow CW. Besides, nitrifying genes amoA and nxrA in upward flow CW at 2.0 mg/L exhibited higher than downward flow CW, explaining better NH4+-N removal in upward flow CW. 0.5 mg/L AgNPs significantly declined NO3--N and TN removal, resulted from decreasing abundances of nirK, nirS and nosZ. In contrast, abundances of nirK, nirS and nosZ genes had slightly lower or higher than before adding AgNPs in upward flow CW, leading to lower NO3--N and TN effluent concentrations. High throughput sequencing also indicated the changes of functional bacterial community after exposing to AgNPs.

Abstract:Increased amount of nano-silver will be released into domestic and industrial waste streams due to its extensive application. However, great controversy still exists on the effects of silver nanoparticle (Ag-NP) on biological wastewater treatment processes and a toxicology model has not been built yet. Four sequencing batch reactors with activated sludge has been run for over three months with different silver species at a concentration of 1mg Ag/L in influent. Both freshly prepared Ag-NPs and aged Ag-NPs were tested with released silver ion as control. Results in this study showed that Ag-NPs, especially freshly prepared Ag-NPs, can help to maintain or even increase the diversity of microbial community in activated sludge and the biomass concentration even under long-term treatment. It indicates that the hormesis model need to be considered for the toxicology of Ag-NPs.
Speciation, controlling steps and pathways of silver release from the sludge generated from coagulation of wastewater spiked with silver nanoparticles

Abstract:Sludge generated in wastewater treatment facilities is an integral part for the introduction of silver nanoparticles (AgNPs) to the terrestrial environment, which would cause some adverse ecosystem responses. The understanding of silver release process from the sludge is important to evaluate their risks. In this study, the amount and speciation of the released silver were investigated by taking the sludge generated by wastewater coagulation with AgNPs added (denoted as sludgeC-AgNPs) an example, and kinetic analysis and density functional theory (DFT) calculations were first used to explore the controlling steps and pathways about the silver release. The results showed that sludgeC-AgNPs could release the dissolved silver and the colloidal silver. Beside Cl−, Ca2+ in the leaching solution could enhance the silver release of sludgeC-AgNPs, especially for the colloidal silver. The released colloidal silver restricted in size from 40 nm to 100 nm with irregular shape. Although the oxidative dissolution of Ag0 was the origin of the silver release pathways from the sludgeC-AgNPs, the silver diffusion was the controlling step due to the spontaneous binding between silver and the hydrolysates of polyaluminium chloride in sludgeC-AgNPs. However, Ca2+ in the leaching solution could occupy the binding site of silver on sludgeC-AgNPs, which would increase the diffusion rate of silver over the oxidative rate of Ag0. With this condition, the controlling step of silver release from sludgeC-AgNPs turned to the oxidative dissolution of Ag0. Our findings are important to assess the fate of AgNPs in wastewater treatment as well as sludge applications.

Abstract:The nitrogen pollution has caused serious environmental problems. The main aims of the present study were to remove the nitrogen from wastewater through ANAMMOX (ANaerobic AMMonium OXidation) process, and let the effluent meet the discharge standard which was significant and advance in the field of ANAMMOX application which was also very important to the environmental protection and water recycling. This study reports the performance of ANAMMOX-EGSB (Expanded Granular Sludge Bed) reactor after feeding with synthetic wastewater. The results showed that the effluent ammonium concentration was 11.90mgN/L and the nitrogen remove efficiency (NRE) reached up to 94.68% when the volumetric nitrogen loading rate (NLR) and volumetric nitrogen removal rate (NRR) were as high as 27.31 and 25.86kgN/(m3·d), respectively. The effluent ammonium concentration meets the Chinese integrated wastewater discharge standard (GB8978-1996) in which the ammonium concentration should be lower than 15mgN/L. The maximum ammonium, nitrite and nitrogen conversion rates (qmax) by the ANAMMOX granular sludge in the reactor were 907.13, 841.76, 1810.10mgN/(g VSS·d) and half saturation constants (KS) were 2.69, 0.44, 3.11mgN/L, respectively.
Strong suppression of silver nanoparticles on antibiotic resistome in anammox process

Abstract:This study comprehensively deciphered the effect of silver nanoparticles (AgNPs) on anammox flocculent sludge, including nitrogen removal performance, microbial community structure, functional enzyme abundance, antibiotic resistance gene (ARGs) dissemination, and horizontal gene transfer (HGT) mechanisms. After long-term exposure to 0–2.5 mg/L AgNPs for 200 cycles, anammox performance significantly decreased ( P < 0.05), while the relative abundances of dominant Ca. Kuenenia and anammox-related enzymes ( hzsA , nirK ) increased compared to the control ( P < 0.05). For antibiotic resistome, ARG abundance hardly changed with 0–0.5 mg/L AgNPs but decreased by approximately 90% with 1.5–2.5 mg/L AgNPs. More importantly, AgNPs effectively inhibited MGE-mediated HGT of ARGs. Additionally, structural equation model (SEM) disclosed the underlying relationship between AgNPs, the antibiotic resistome, and the microbial community. Overall, AgNPs suppressed the anammox-driven nitrogen cycle, regulated the microbial community, and prevented the spread of ARGs in anammox flocs. This study provides a theoretical baseline for an advanced understanding of the ecological roles of nanoparticles and resistance elements in engineered ecosystems.

Abstract:The ANAMMOX process with sequential biocatalyst addition (SBA-ANAMMOX process) was developed in sequencing batch reactor (SBR) to enhance nitrogen removal from ammonium-rich wastewater containing glucose content up to 800 mgCOD/L. The nitrogen removal performance of the SBR operated with 800 mgCOD/L was significantly enhanced after sequential adding 0.5 gVSS/d of high-rate ANAMMOX granules (HASGs). Kinetics analyses showed that the HASGs possessed high nitrite affinity and strong resistance to organic matter, both of which contributed to the enhancement of autotrophic nitrogen removal. Consequently. ANAMMOX became the dominant reaction to metabolize nitrite under organic conditions; denitrification was limited due to the lack of electron acceptors (nitrite and nitrate); while sulfate reduction emerged as an important process for consumption of organic matter in the SBA-ANAMMOX system. It is suggested that employing the SBA-ANAMMOX process for treatment of organic ammonium-rich wastewater, the simultaneous removal of nitrogenous compounds as well as organic matter was quite possible. Nitrogen Removal Performance of Anammox Process and Toxic Mechanism of Pharmaceutical Wastewater

Abstract:In order to develop effective process to remove nitrogen from pharmaceutical wastewater,the performance of anaerobic ammonia oxidation(anammox)for nitrogen removal from pharmaceutical wastewater was investigated using upflow anaerobic sludge bed (UASB) reactor.The acute toxicity of pharmaceutical wastewater,the influent and effluent of anammox UASB reactor as well as the cumulative toxicity of pharmaceutical wastewater to anammox sludge were also studied.The reactor operation results indicated that when the pharmaceutical wastewater was diluted more than 30 times,the toxicant concentration below the threshold and the operation performance of anammox reactor was very good,the average removal rate of ammonia and nitrite reached 87.8%and 95.6% respectively,the average volumetric total nitrogen loading rate was up to 10.38kg/(m~3·d).However,when the pharmaceutical wastewater was diluted less than 20 times,the toxicant concentration higher than the threshold and the operation performance of anammox reactor deteriorated,the average removal rate of ammonia and nitrite reduced to 24.6%and 26.0%,and then completely disappeared.The acute toxicity by photobacteria tests showed that the pharmaceutical wastewater,the influent and effluent of anammox process all had intense biotoxicity.When the relative luminosity was 50%,the dilution factors of the three samples were 70.5,5.19,7.77 respectively.After analnmox treatment,the toxicity of effluent enhanced.The cumulative toxicity by photobacteria tests suggested that the toxicants in pharmaceutical wastewater could cumulate in anamlnox sludge,leading to higher toxicity in the test Anammox sludge than that in the seed sludge.

Abstract:Silver nanoparticles (NPs) are entering soils with biosolids via wastewater treatment, and on-route, undergo changes (e.g., sulfidation) that alter silver bioavailability and toxicity. While additions of fresh Ag-NPs to soil have been show to influence bacterial diversity, it is unclear whether these effects are representative of realistic exposure pathways. To investigate the effects of wastewater treatment processing on the ecotoxicology of Ag-NPs, we ran sequencing batch reactors for 28 d to produce three batches of sludge: (1) a control, (2) Ag-NP sludge, and (3) AgNO3 sludge. The effects of processed silver on the diversity and composition of soil bacterial and fungal communities were compared to those of fresh Ag-NPs, Ag2S-NPs, and AgNO3, which were added with the control sludge, at two concentrations (1 and 10 mg Ag kg–1 dry soil) over time (3, 7, 30, and 90 d). The effects of processed Ag-NPs on the composition of soil bacterial communities were larger and more persistent than those of fresh Ag-NPs, Ag2S-NPs, and AgNO3. Treatment effects on fungi were relatively minor. However, the detailed information on the role of natural organic matters (NOMs) remains limited. Effects of humic acid (HA), alginate (AA) and bovine serum albumin (BSA) on the release amount, dynamics and speciation of silver from the sludgeC-AgNPs were investigated by a series of leaching experiments. The results showed that HA, AA and BSA in the leaching solution could enhance the silver release from the sludgeC-AgNPs. The concentrations of the dissolved and colloidal silver in the BSA solution were the highest at the initial stage of dynamic leaching. The controlling step of the silver release was internal diffusion in the HA and AA solution, while the release of dissolved silver was controlled by both chemical reaction and internal diffusion in the BSA solution. In addition, the released colloidal silver fractions in the BSA solution contained more particles with size >50 nm compared with the HA and AA solutions. The results suggested that the properties of NOMs may be the key factor affecting the transfer of AgNPs from the sludge to the terrestrial environment.

Abstract:The performance of high-loaded anaerobic ammonium oxidizing (ANAMMOX) upflow anaerobic sludge bed (UASB) reactors was investigated. Two ANAMMOX reactors (R1 with and R2 without effluent recycling, respectively) were fed with relatively low nitrite concentration of 240 mg-N L(-1) with subsequent progressive increase in the nitrogen loading rate (NLR) by shortening the hydraulic retention time (HRT) till the end of the experiment. A super high-rate performance with nitrogen removal rate (NRR) of 74.3-76.7 kg-N m(-3) day(-1) was accomplished in the lab-scale ANAMMOX UASB reactors, which was 3 times of the highest reported value. The biomass concentrations in the reactors were as high as 42.0-57.7 g-VSS L(-1) with the specific ANAMMOX activity (SAA) approaching to 5.6 kg-N kg-VSS(-1) day(-1). The high SAA and high biomass concentration were regarded as the key factors for the super high-rate performance. ANAMMOX granules were observed in the reactors with settling velocities of 73-88 m h(-1). The ANAMMOX granules were found to contain a plenty of extracellular polymers (ECPs) such as 71.8-112.1 mg g-VSS(-1) of polysaccharides (PS) and 164.4-298.2 mg g-VSS(-1) of proteins (PN). High content of hemachrome (6.8-10.3 μmol g-VSS(-1)) was detected in the ANAMMOX granules, which is supposed to be attributed to their unique carmine color.
Aggregation Behavior of Engineered Nanoparticles and Their Impact on Activated Sludge in Wastewater Treatment.

Abstract:The ever-increasing daily use of engineered nanoparticles will lead to heightened levels of these materials in the environment. These nanomaterials will eventually go into the wastewater treatment plant (WWTP), therefore, resulting into a pressing need for information on their aggregation behavior and kinetics in the wastewater aqueous matrix. In this work, we dispersed two different metal oxide nanoparticles (ZnO and TiO2) into the influent of two different WWTPs. Through the time-resolved dynamic light scattering analysis and transmission electron microscopy, the metal oxide nanoparticles (NPs) were quite stably existed in the wastewater matrix with aggregates of diameter 300-400 nm after 4.5 h or more suspension. We confirmed that the dissolved organic matters (DOMs) attributed to the stability of nanoparticles. No propensity of NPs to aggregate were observed in the presence of both monovalent and divalent electrolytes even at high concentrations up to 0.15 M in NaCl or 0.025 M in CaCl2, indicating that the destabilization of nanoparticles in the complicated wastewater matrix was not achieved by the compression of electrical double layer, therefore, their aggregation kinetics cannot be simply predicted by the classic Derjaguin-Landau-Verwey-Overbeek theory of colloidal stability. However, obvious aggregation of nanoparticles in the Al-2(SO4)(3) solution system was observed with the likely mechanism of bridging of the metal oxide nanoparticles and aggregates due to the formation of hydrous alumina (Al(OH)(3)center dot H2O) in the Al-2(SO4)(3) solution. In the wastewater matrix, we used the noninvasive measurement technology to detect the O-2 flux of activated sludge before and after treatment with 1, 10 and 100 mg L-1 NPs. The results confirmed that both ZnO and TiO2 NPs showed an adverse impact on the O-2 uptake of activated sludge when the exposure time extended to 4.5 h. (C) 2014 Elsevier Ltd. All rights reserved.

Abstract:Previous studies investigating the risk of engineered nanoparticles (NPs) to biological wastewater treatment have primarily tested NPs individually; however, limited data are available on the impact of NPs on the anaerobic ammonium oxidation (anammox) process. In this study, the toxicity of CuNPs on anammox granules was investigated individually and in combination with CuONPs or ZnONPs. Exposure to CuNPs at 5mgg-1 suspended solids (SS) decreased the anammox activity to 47.1±8.5%, increased the lactate dehydrogenase level to 110.5±3.4% and increased the extracellular N2H4 concentration by 16-fold but did not cause oxidative stress. The presence of CuONPs or ZnONPs at 5mgg-1 SS did not significantly aggravate or alleviate the toxicity of the CuNPs; however, the introduction of EDTA or S2- could attenuate the adverse effects of the CuNPs, CuONPs and ZnONPs on the anammox granules. EDTA captured Cu ions, whereas S2- shielded and deactivated Cu ions and passivated CuNPs. Therefore, our results indicated that the toxicity of NPs was dependent on the amount of active metal reaching the anammox cells. Overall, the results of this study have filled knowledge gaps and provided insights into the combined toxicity of NPs on anammox biomass.
