Abstract:Highlights • C. odorata aerial part lipophilic extract (COLE) was prepared via a facile green chemistry approach. • COLE demonstrated potent and selective anticancer activity against human breast and colon cancer cells. • Compared to the hydrophilic extract, COLE displayed superior antibacterial properties. • COLE also exhibited potent antioxidant and metal chelation activities. • UPLC-ESI-QTOF-MS and GC-MS analysis revealed that COLE was predominantly endowed with phenolic and terpene metabolites. There has been growing interest in the biological and pharmacological potential of C. odorata in recent times. While a number of reports had highlighted the beneficial attributes of various extracts prepared from C. odorata , the biological properties of the lipophilic extract remained unexplored. This work aimed at examining the anticancer, antimicrobial and antioxidant properties of C. odorata aerial part lipophilic extract (COLE). The results revealed that COLE possessed potent and selective anticancer properties against human breast and colon cancer cells. The extract presented strong cytotoxic activity against MCF-7 cells (IC 50 of 9.18 μg/mL), MDA-MB-231 (IC 50 of 12.19 μg/mL), HT-29 (IC 50 of 19.48 μg/mL), and HCT-116 cells (IC 50 of 14.12 μg/mL). Meanwhile, COLE was found to be non-toxic at the concentrations tested against normal (HEK-293 and CCD-841 CoN) cells. In addition, COLE demonstrated significant antibacterial activity against B. cereus (MIC value of 0.157 mg/mL), and moderate activity against L. monocytogenes (MIC value of 0.63 mg/mL) and E. coli (MIC value of 1.06 mg/mL). Furthermore, COLE effectively scavenged free radical species, including ABTṠ + (IC 50 of 46.80 μg/mL), DPPḢ (IC 50 of 27.78 μg/mL), and superoxide anion (IC 50 of 64.07 μg/mL), likely due to the high phenolic (262.48 mg gallic acid equivalent/g) and flavonoid content (70.20 mg quercetin equivalent/g). UPLC-MS analysis revealed the putative identities of 85 secondary metabolites whereas the GC-MS analysis detected 36 volatile constituents in COLE. Also, it was revealed that terpenes and phenolic compounds were the predominant specialized metabolites present in COLE, and putatively responsible for the remarkable biological properties. Taken together, these results revealed the richness of C. odorata lipophilic extract in terms of its metabolite profile and biological activities. In conclusion, COLE is worthy of further investigation to take advantage of its good anticancer, antioxidative and antimicrobial properties in pharmacological or nutraceutical application.

Abstract:Aim: Hepatocellular carcinoma (HCC) is the world's second leading cause of cancerrelated mortality and the fifth most prevalent cancer overall. Several synthetic and plant-based remedies are in practice to treat diverse liver disorders. Because of their minimal side effects and protective characteristics, plant phenolics have the potential to become alternative therapeutics, replacing currently existing HCC medications. The present study identifies the plant phenolics as having the capacity to inhibit HCC with low side effects and cost efficiency. Background: Hepatocellular carcinoma (HCC) is the leading cause of cancer-related mortality, despite the proven effectiveness of screening programs for at-risk individuals, the majority of patients have disease progression or tumor characteristics that preclude curative therapies at the time of diagnosis. Acteoside (Verbascoside) is a naturally occurring phenylethanoid glycoside found throughout the plant kingdom. Acteoside is a physiologically active chemical with the number of pharmacological and protective effects against various liver illnesses. Objectives: Currently used HCC medications have a variety of side effects. Plant-based chemicals offer the possibility of treating HCC with minimal side effects. The work is targeted to find the best phytochemical (plant phenolic) lead molecule for future drug development research against Hepatocellular carcinoma. Methods: The targets were selected based on an analysis of relevant literature, and the 3D structures of the selected receptors were obtained in. pdb format from the RCSB-Protein data bank (PDB, http://www.rscb.org/pdb). Based on a review of the literature, sixty plant secondary metabolites, or plant phenolics, were selected. The ligand structures were obtained and downloaded in.sdf format from the NCBI PubChem chemicals database (https://pubchem.ncbi.nlm.nih.gov/). Molecular docking between the receptor and ligands was accomplished using the Molegro Virtual Docker 6.0 (MVD) software. Results: The target RAF1, BRAF chain 1, TIE2 chain 2 FGFR1, FGFR2, AXL, and FGFR4 showed the best binding effectiveness with acteoside compared to their respective positive control. RET chain 1 and BRAF chain 2 acteoside showed prominent binding efficacy after Curcumin, and Epigallocatechingallate, respectively, against positive control. Present findings clearly point towards the potentiality of acteoside in inhibiting various HCC targets. Conclusion: Acteoside may be used as a prominent lead molecule in the future treatment of hepatic cancer with its multifaceted binding efficiencies against various target proteins.
Untargeted LC/HRMS Metabolomics Analysis and Anticancer Activity Assay on MCF-7 and A549 Cells from Coleus amboinicus Lour Leaf Extract

Abstract:Background: Cancer remains the leading cause of death globally, with breast cancer being the foremost cause among women and lung cancer ranking second for both women and men. Objectives: This study aimed to identify the metabolomic content of Coleus amboinicus leaves and evaluate their anticancer activities against breast and lung cancer cells, thereby providing insights into potential alternative treatments for these cancers and initiating research on active isolates from C. amboinicus leaves. Methods: The research methodology involved maceration using ethanol, followed by multistage partitioning with solvents n-hexane, chloroform, and ethyl acetate. Phytochemical screening was performed using standard reagents to detect the presence of alkaloids, phenolics, polyphenols, flavonoids, steroids/triterpenoids, and saponins. Metabolomic profiling was conducted using LC/HRMS, and the anticancer activities against lung cancer cells (A549) and breast cancer cells (MCF-7) were assessed using the MTT assay. Results: The results showed that the C. amboinicus extract contains various secondary metabolite groups such as alkaloids, phenolics and polyphenols, flavonoids, steroids, triterpenoids, and saponins. Conclusions: The diverse metabolomic profile of the C. amboinicus leaf extract demonstrated potential activity against cancer, as evidenced by in vitro tests on lung (A549) and breast (MCF-7) cancer cells. C. amboinicus leaf extract shows promise as an active ingredient in the prevention and alternative natural treatment of lung and breast cancer. Further research and testing, both in vivo and clinically, are warranted.

Abstract:Background: Edible fruits and resins provide various benefits to mankind including potential medicinal applications. This study aimed to determine the cytotoxicity, selectivity, and clonogenicity of fruits and exudates of certain Saudi medicinal plants (Anethum graveolens (BEP-09), Opuntia ficus-indica (L.) Miller (BEP-10), Boswellia serrata Roxb. ex Colebr. (BEP-11), and Commiphora myrrha (BEP-12)) against human liver adenocarcinoma (HepG2). Methods: Initial cytotoxicity and cell line selectivity against different cell lines were screened using MTT assay. The most promising extract was subjected to gas chromatography-mass spectrometry (GC-MS) analysis to determine the main phytoconstituents. Clonogenicity was checked for the most active extract. Results: The selected plants' fruits and resins possess a significant cytotoxic activity estimated as IC50. The fruit of BEP-10 was found to be the most active extract against liver cancer cells (IC50 = 2.82) comparable to both doxorubicin (IC50 = 1.40) and camptothecin (IC50 = 1.11). It showed a selectivity index of 4.47 compared to the normal human foetal lung fibroblast (MRC5) cells. BEP-10 showed a dose-dependent clonogenic effect against HepG2 cells comparable to the effect of doxorubicin. The GC-MS chromatogram of BEP-10 extract revealed the presence of eight small polar molecules, representing 73% of the total identified compounds and the rest three molecules (27%) were non-polar constituents. The furan derivatives represent the chief components in BEP-10 (16.3%), while the aldehyde 5-(hydroxymethyl)-2-furancarboxaldehyde was found to be the main molecule (13.2%). Conclusion: The fruits of BEP-10 have a potential cytotoxic effect particularly against HepG2. The identified phytoconstituents in the tested plant extract might contribute to the investigated cytotoxic activity.

Abstract:Ethnopharmacological relevance: Ageratina adenophora (Sprengel) R.M.King & H.Rob. has been used as traditional indigenous medicine all across the globe for its diverse therapeutic applications such as anticancer, analgesic, antipyretic, thermogenic, antiseptic, antimicrobial as well as astringent. The various ethnic groups of India use plant parts to treat cuts and wounds, venomous insect bites, skin lesions, blisters, scabies and other skin irritations, gastritis and indigestion problems, cough, stomach ache and dysentery. The Portuguese traditionally extract the juice from the plant and use it for cancer, diabetes, liver disorder, gallbladder and stomach ailments. Nigerian healers use different parts of the plant to treat diabetes, fever and inflammation. Aim of the study: The aim of this study is to investigate the cytotoxic potential of A. adenophora hydroalcoholic leaves extract (AHL) on Colorectal cancer (CRC) cell lines (HCT-116, HCT-15 and HT-29), synergistic potential with chemotherapeutic drugs 5FU and Cisplatin as well as reactive oxygen species (ROS) generation, based on the sample collected from Mao district of Manipur, India. Identification of bioactive phytocompounds in AHL was also performed by HRLCMS. Methods: The AHL was evaluated for its cytotoxic as well as antiproliferative activities by 3-(4, 5-dimethylthiazol-2-yl)-2, 5 diphenyltetrazolium bromide (MTT) assay, clonogenic and cell migration assays. The total phenolic content (TPC) and total flavonoid content (TFC) were quantified by Folin-ciocalteu and Aluminium chloride assays respectively. Caspase 3 activation was evaluated using Caspase-3 Assay Kit. Apoptosis detection by flow cytometry was carried out using annexin V-FITC/PI apoptosis detection kit. The apoptotic cells were also visualized by Giemsa and 4',6-Diamidino-2-phenylindole (DAPI) staining. The intracellular Reactive oxygen species (ROS) generation was also evaluated using fluorescent probe 2',7'-dichlorodihydrofluorescein di-acetate (H2DCFDA) in flow cytometry. The combination effects of AHL with chemotherapeutic drugs 5FU and Cisplatin were also evaluated. The identification of phytochemical constituents of AHL were analysed by HR-LCMS. Results: The AHL induced cytotoxic activity significantly in HCT-116 with IC50 of 65.65 ± 2.10 μg/mL, but non-cancerous cell HeK-293 was least cytotoxic. Colony formation and cell migration were inhibited in a dose and time dependent manner. The cell morphology upon AHL treatment was significantly altered with apoptotic features. The extract was rich in total phenolic (82.09 ± 0.35mgGAE/g) and total flavonoid (58.31 ± 0.55 mgQAE/g) contents. AHL induced apoptosis as detected by AnnexinV/PI, via activation of caspase 3 and elevated production of Reactive oxygen species (ROS). AHL in combination with 5FU and Cisplatin acts synergistically and potentiates the therapeutic properties of the extract. Sesquiterpenes, phenolic as well as flavonoid derivatives with anticancer properties were detected in AHL by HRLCMS, and these phytoconstituents may be attributed for anticancer property of AHL. Conclusion: The present study evaluates the effectiveness of AHL against Colorectal cancer cell lines. AHL is cytotoxic and induces apoptosis in HCT-116 cells by caspase 3 activation and increased ROS production that can be attributed to sesquiterpenoids. Thus, the plant A. adenophora has therapeutic potential for Colorectal cancer and can be further exploited for developing anticancer drug.

Abstract:India is considered a hub for a wide range of phytochemicals due to its rich biodiversity, which indeed flourished with many folk medicines and treatments. Phytochemicals are secondary metabolites synthesized in plants to protect themselves from deteriorating environmental stress and defense against pathogens. In the present study, we aimed to explore the strong anticancer potential of the bioactive compound present in the leaf extract of Coldenia procumbens (L.). The bioactive compound oleanolic acid was first time reported in the plant and was successfully isolated, characterized with various spectroscopic analyses. The in vitro study results explored that oleanolic acid has shown a potent cytotoxic effect against human hepatocellular carcinoma cell lines with a low IC 50 value (40.66μg/mL) and induced apoptotic cell death. Overall, the present findings clearly demonstrated that the natural triterpenoid compound oleanolic acid isolated from C. procumbens showed a potent cytotoxic effect against HepG2 cells and explored the strong anticancer potential against liver cancer.

Abstract:On a global basis, colorectal cancer is one of the foremost causes of cancer-related death rate. The present research encompasses the fabrication of CuO nanoparticles employing leaf extract of Coleus amboinicus (CA–CuO NPs) and evaluating the anti-cancerous capability against HCT 116 cells (Colon cancer). The fabricated CA–CuO NPs were assessed by UV–Vis. spectrophotometer, FT-IR, SEM, EDAX and DLS. The cytotoxic action upon the HCT 116 cells was evaluated using MTT, trypan blue exclusion assay, phase contrast microscopy, DAPI, and MMP Staining. The oxidative stress markers such as nitric oxide (NO), lipid peroxidation (LPO), and reactive oxygen species (ROS) levels were analyzed. The antioxidant markers including catalase (CAT) and superoxide dismutase (SOD) were further analyzed. Using RT-PCR, the expression levels of genes including Bax , p53 , and Bcl-2 were analyzed. The peak at 225 nm in the UV spectrum disclosed the formation of CA–CuO NPs. The possible existence of phytoconstituents responsible for reducing CuO ions was confirmed by FT-IR results. The SEM images showed the formation of irregular rice-shaped and elongated tetragonal-shaped CuO nanoparticles. The EDAX results confirm the existence of C, Cu, O and S peaks. The DLS findings revealed average particle size of CA–CuO NPs was 118.6 nm. The MTT assay demonstrated a dose-dependent, substantial suppression of HCT 116 cell growth, with an IC 50 of 12.5 μg/mL indicating 49.71% inhibition. The proportion of viable cells in the trypan blue exclusion assay steadily dropped as treated cells suffered morphological deformity as observed by phase contrast microscopy. The DAPI and MMP staining revealed an increase in dead cells in a dose-wise manner. Oxidative stress indicators including NO, LPO, and ROS levels were considerably higher in the treated groups than in the control. The antioxidant markers such as CAT and SOD declined significantly in treated groups. The endpoints of the gene expression study revealed the overexpression of tumor suppression genes Bax and p53 and anti-apoptotic gene, Bcl-2 was unexpressed. The biogenically synthesized CA–CuO NPs exhibited exceptional anti-cancerous activity towards HCT 116 cell line.

Antiproliferative effects of extracts from Salvia officinalis L. and Saliva miltiorrhiza Bunge on hepatocellular carcinoma cells

Abstract:Salvia species have been used as valuable medicinal and herbal plant in many countries. Salvia officinalis L. and Salvia miltiorrhiza Bunge are widely used in traditional medicine for a long time. In the present study, cytotoxicity of ethanol and acetone extracts prepared from leaves and roots of two Salvia species was investigated using hepatocellular carcinoma cells (HepG2) and normal human liver cells (WRL-68). The cytotoxicity and anti-proliferative abilities of the extracts were evaluated by measuring cell viability (MTS assay), lactate dehydrogenase (LDH) leakage, the cellular ATP level, morphological changes using an inverted microscope, and apoptosis using flow cytometry. The results indicated that ethanol and acetone extracts of leaves and roots of S. officinalis (SO-L-E, SO-L-A, SO-R-E and SO-R-A, respectively) and ethanol and acetone extracts of roots of S. miltiorrhiza (SM-R-E and SM-R-A, respectively) significantly inhibited the proliferation of HepG2 cells in a time- and dose-dependent manner when the concentration was less than 150μg/mL. The cytotoxity of SO-L-E, SO-R-E and SO-R-A were significantly less in WRL-68 when compared to HepG2 cells in vitro. The increase of LDH leakage, decrease of ATP and the changes in morphology of HepG2 cells further confirmed the cytotoxic effect of these extracts to HepG2 cells. Furthermore, SO-L-E, SO-L-A, SO-R-E ethanol extract of leaves of S. miltiorrhiza (SM-L-E) and SM-R-E were able to induce apoptosis in HepG2 cells. This study shows the potential of the extracts to be used in the prevention and/or treatment of liver cancer or as ingredients in functional foods and provides scientific support for development and utilization of S. officinalis and S. miltiorrhiza, especially the roots of S. officinalis.

Abstract:Caralluma europaea Guss. ( C. europaea ) is a medicinal plant used for cancer treatment. However, these treatments may be associated with complications that need to be investigated. This work aims to evaluate not only the chemical composition but also the hepatoprotective and anticancer properties of C. europaea extracts. The chemical constitution of the hydroethanolic extract was explored using gas chromatography-mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC). The hydroethanolic extract, flavonoids, and polyphenols-rich extract at 100, 15, and 50 mg/kg, respectively, were administered to acetaminophen-treated rats for seven days. We used Western blotting and Real-Time quantitative Polymerase Chain Reaction (RT-qPCR) to determine the protein and the mRNA levels of cancer stemness markers in pancreatic cancer cell lines MIA PaCa-2 and BxPC-3 exposed to increasing doses of C. europaea extracts. In silico analysis was used to evaluate the effects of phenolic compounds revealed in C. europaea on caspase-3 and HSP90, and on liver damage on CYP2E1. The primary phenolics detected by GC-MS and HPLC were ferulic acid and benzofurazan. The positive control group showed an increase in AST, ALT, ALP, triglycerides, and VLDL levels. C. europaea extracts demonstrated hepatoprotective effects by ameliorating acetaminophen-induced alterations of biochemical and hispathological parameters. Immunoblotting and RT-qPCR profiling of cancer stemness markers indicated a reduction in the expression levels of Oct-4 and Nanog proteins, as well as a reduction in the mRNA levels of CD133 by 50–60% and Sox2 by 80–90% in pancreatic cancer cells. Molecular docking showed that naringenin presented the highest docking Gscore on CYP2E1 (−8.199) and HSP90 (−7.742). In conclusion, C. europaea extracts could be considered as a safe and promising therapeutic strategy to sensitize pancreatic cancer cells to chemotherapy.

Abstract:HCC is one of the fastest-rising causes of cancer-related death. Novel therapeutic approaches for treatment are warranted. The goal of this study is to find effective components from Chinese herbal medicines, which is an important alternative source of anticancer medicine. To this end, six different herbs were selected from various traditional literatures. Soxhlet extractor was used to distill the strong polar and weak polar components of each herb. The inhibitive effect of each component was determined using liver cancer cells BEL7404. From total of 12 extractions, it was found that the combined crude lysate of Amana edulis from water and ethanol system had the best efficacy. At the concentration of 0.1 mg/mL, this component has the highest inhibition rate up to 70%. To investigate the underlying molecular reasons, we observed that the component can significantly induce the liver cancer cells apoptosis and retard the cell reproduction at G2/M stage. Verification experiments showed that this component also has apparent inhibitive effects on other liver cancer cells, such as HepG2 and Huh7. On the other hand, it has less effectiveness on another cell line HepaRG, which retains many characteristics of primary human hepatocytes. The results suggested that there might be highly efficient antihepatoma ingredient in the water and ethanol extraction of Amana edulis. The pure substances remain to be isolated and further research on their targets is required.

Abstract:Cloves (Syzygium aromaticum) have been used as a traditional Chinese medicinal herb for thousands of years. Cloves possess antiseptic, antibacterial, antifungal, and antiviral properties, but their potential anticancer activity remains unknown. In this study, we investigated the in vitro and in vivo antitumor effects and biological mechanisms of ethyl acetate extract of cloves (EAEC) and the potential bioactive components responsible for its antitumor activity. The effects of EAEC on cell growth, cell cycle distribution, and apoptosis were investigated using human cancer cell lines. The molecular changes associated with the effects of EAEC were analyzed by Western blot and (qRT)-PCR analysis. The in vivo effect of EAEC and its bioactive component was investigated using the HT-29 tumor xenograft model. We identified oleanolic acid (OA) as one of the components of EAEC responsible for its antitumor activity. Both EAEC and OA display cytotoxicity against several human cancer cell lines. Interestingly, EAEC was superior to OA and the chemotherapeutic agent 5-fluorouracil at suppressing growth of colon tumor xenografts. EAEC promoted G0/G1 cell cycle arrest and induced apoptosis in a dose-dependent manner. Treatment with EAEC and OA selectively increased protein expression of p21(WAF1/Cip1) and γ-H2AX and downregulated expression of cell cycle-regulated proteins. Moreover, many of these changes were at the mRNA level, suggesting transcriptional regulation by EAEC treatment. Our results demonstrate that clove extract may represent a novel therapeutic herb for the treatment of colorectal cancer, and OA appears to be one of the bioactive components.
Anti-inflammatory and Apoptotic Effects of Levisticum Officinale Koch Extracts on HT 29 and Caco-2 Human Colorectal Carcinoma Cell Lines

Abstract:Background: Colorectal cancer is among the deadliest cancers in the world. Due to the occurrence of side effects related to current standard therapy, researchers are seeking better alternative treatments. For many years, herbs have been a promising source for discovering therapeutic compounds. Therefore, the primary objective of this research was to examine the distinctive apoptotic and anti-inflammatory properties exhibited by Levisticum officinale Koch (lovage) on HT-29 and Caco-2 cell lines. Materials and methods: The maceration method was used to prepare different extracts (ethanol, dichloromethane, petroleum, and residues) from the plant. These extracts were then tested on two colon cancer cell lines - HT-29 and Caco-2 - using the MTT assay to determine the half-maximal inhibitory concentration (IC50) values. In addition, we evaluated the expression levels of several inflammatory genes (IKKb, IKKa, and REIB) using real-time PCR. We also assessed Cox-2 protein expression using western blot analysis. The western blot was also used to analyze apoptosis-related proteins, including Caspase-3, BAX, and Bcl-2. Results: The dichloromethane extract of Levisticum officin (DELO) exhibited a high cytotoxic effect on Caco-2 and HT-29 cell lines, with IC50 values of 106.0±2 μg/mL in HT-29 cells and 175.3±4 μg/mL in Caco-2 cells after 72 hours. None of the lovage extracts showed a significant cytotoxic effect on non-cancerous cells (3T3 cell line). Furthermore, the group treated with DELO showed a lower expression level of inflammatory genes and COX-2 protein compared to the control group. Notably, treatment with DELO resulted in an increase in Caspase-3 protein and BAX/Bcl-2 ratio in both HT-29 and Caco-2 cells. Conclusion: According to this study, DELO has the potential to act as an anti-inflammatory and anti-cancer agent. Further research on the compounds present in DELO and their effect on various signaling pathways could help in the development of new drugs for diseases where inflammation or cells escape from apoptosis play a crucial role.

Abstract:Background: Celastrus orbiculatus Thunb. is a medicinal plant that has been widely used for thousands of years in China, and the ethyl acetate extract (Celastrus orbiculatus Thunb. Extract, COE) from its stem was reported to exert antitumor and anti-inflammatory effects in various preclinical studies. However, the anti-non-small-cell lung cancer activity of COE and its potential mechanism are not yet fully understood. Purpose: To investigate the antitumor effects of COE on non-small-cell lung cancer (NSCLC) cells and explore its molecular mechanism from the perspective of Hippo signaling, YAP nuclear translocation, and reactive oxygen species (ROS) generation. Methods: The effects of COE on proliferation, cell cycle arrest, apoptosis, stemness, and senescence in NSCLC cell lines were determined by CCK-8, clone formation, flow cytometry, and beta-galactosidase staining assays. The ef-fects of COE on Hippo signaling were investigated by Western blotting. The intracellular expression and dis-tribution of YAP were analyzed by immunofluorescence assay. DCFH-DA probe combined with flow cytometry was used to detect intracellular total ROS levels in NSCLC cells after COE treatment. Xenograft tumor model was established, and the animal living image system was employed to analyze the effects of COE on the Hippo-YAP signaling in vivo. Result: COE significantly inhibited NSCLC activity in vitro and in vivo, mainly by proliferation inhibition, cycle arrest, apoptosis promotion, senescence promotion, and stemness downregulation. COE strongly activated Hippo signaling and inhibited YAP expression and nuclear retention. Activation of Hippo signaling induced by COE was associated with ROS-mediated phosphorylation of MOB1. Conclusion: This study demonstrated that COE inhibited NSCLC through activating Hippo signaling and sup-pressing YAP nuclear translocation, in which ROS may play a role in the phosphorylation of the MOB1 protein.

Abstract:Cancer treatment frequently carries side effects, therefore, the search for new selective and effective molecules is indispensable. Hymenaea courbaril L. has been used in traditional medicine in South America to treat several diseases, including prostate cancer. Leaves’ extracts from different polarities were evaluated using the 3-(4,5-methyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) cell viability assay to determine the cytotoxicity in prostate p53-null cells, followed by bio-guided fractionations to obtain the most cytotoxic fraction considering the selectivity index. The most cytotoxic fraction was analyzed by GC/MS to identify the active compounds. The majority compound, caryophyllene oxide, induced early and late apoptosis, depolarized the mitochondrial membrane, leading to several morphological changes and shifts in apoptotic proteins, and caspases were evidenced. Depolarization of the mitochondrial membrane releases the pro-apoptotic protein Bax from Bcl-xL. The apoptosis process is caspase-7 activation-dependent. Caryophyllene oxide is a safe anti-proliferative agent against PC-3 cells, inducing apoptosis with low toxicity towards normal cells.

Abstract:Flux-weakening control for induction motor (IM) in voltage extension region (outside the inscribed circle but in the hexagon) is meaningful to yield a further maximum torque. However, as the voltage-limit trajectory migrates out of the inscribed circle, torque ripple becomes more severe. Meanwhile, the insufficient voltage margin results in the degradation of current dynamic performance in transition period (from base speed region to flux-weakening region), especially in harsh conditions, e.g., a step speed command. To address the problems above, this paper gives a quantitative analysis of the torque ripple and an explicit discussion on the current dynamic performance. A novel “Voltage Reference Adjustable” flux-weakening controller with “Self-Locking Limit Block” (SLLB) is proposed. There are two advantages. The first is the capability to operate in any voltage extension regions, offering a tradeoff between obtaining the maximum torque and suppressing the torque ripple. The second is an optimized voltage distribution, achieving a better track characteristic of d- and q-axis currents with the help of triggered SLLB in transition period. Experimental results on a commercial IM control system verify the validity of the proposed scheme.
A New Natural Source against A549 Lung Cancer Cells Anthemis cotula and its Biological Activities and Phenolic Contents

Abstract:The objective of this study was to assess the biological activities of Anthemis cotula L. and explore its potential therapeutic effects on A549 lung cancer cells. Within this particular framework, the aerial components of the plant were collected and subjected to extraction using a Soxhlet apparatus to obtain extracts in the form of water, ethanol, and methanol. The determination of the phenolic content of the samples was conducted using the Folin-Ciocalteu reagent, while the quantification of flavonoid content was conducted via an aluminum chloride assay, and the measurement of the plant's total antioxidant, total oxidant, and oxidative stress index was conducted using Rel Assay kits. The antimicrobial properties of the substances were examined via the agar dilution technique. The MTT assay was employed to determine the antiproliferative activity against A549 lung cancer cells, and the LC-MS/MS instrument was utilised to screen for the presence of standard compounds. The following compounds were identified in the study: acetohydroxamic acid, catechin hydrate, vanillic acid, resveratrol, kaempferol, fumaric acid, gallic acid, protocatechuic acid, 4-hydroxybenzoic acid, caffeic acid, salicylic acid, 2-hydroxycinnamic acid, naringenin, quercetin, luteolin, and alizarin. Plant extracts were found to be effective against microorganisms in the range of 25 to 200 μg/mL; furthermore, the water extract exhibited the highest total antioxidant status value of 8.201 ± 0.113 μmol/L. Similarly, the ethanol extract demonstrated the highest total oxidant status value of 11.247 ± 0.178 μmol/L, while, lastly, the methanol extract displayed the highest oxidative stress index value of 0.151 ± 0.001. Strong antiproliferative activity was observed in all the plant extracts, and the ethanol extract with the highest concentration exhibited a total phenolic content of 61.78 ± 4.38 mg/g. The ethanol extract exhibited the highest recorded total flavonoid content, measuring 66.84 ± 3.13 mg/g. In the present context, it has been observed that the plant demonstrates significant biological activities; consequently, A. cotula is believed to possess significant potential as a valuable resource in the development of pharmacological designs.

Abstract:Background: The use of plant essential oils as pharmaceuticals is a fast-growing market especially in China. Throughout the 20th century, a rapid increase took place in the use of many essential oil-derived products in the medicinal industry as nutraceuticals, medicinal supplements, and pharmaceuticals. Purpose: The objective of this study was to explore the chemical composition of Croton crassifolius essential oil as well as its potential anti-tumour properties and related anti-proliferative, autophagic, and apoptosis-inducing effects. Methods: Supercritical CO2 fluid extraction technology was used to extract CCEO and the chemical constituents of the essential oil were identified by comparing the retention indices and mass spectra data taken from the NIST library with those calculated based on the C7-C40 n-alkanes standard. The cytotoxic activity and anti-proliferative effects of CCEO were evaluated against five cancer cell lines and one normal human cell line via CCK-8 assays. In addition, flow cytometry was used to detect cell cycle arrest. The efficacy of CCEO treatments in controlling cancer cell proliferation was assessed by cell cycle analysis, clonal formation assays, RT-qPCR, and western blot analysis. Autophagic and apoptosis-inducing effects of oils and the associated molecular mechanisms were assessed by flow cytometry, cell staining, reactive oxygen species assays, RT-qPCR, and western blot analysis. Conclusion: Forty compounds representing 92.90% of the total oil were identified in CCEO. The results showed that CCEO exerted a measurable selectivity for cancer cell lines, especially for A549 with the lowest IC50 value of 25.00 ± 1.62 μg/mL. Assessment of the anti-proliferative effects of CCEO on A549 cells showed that the oil inhibited cell proliferation and colony formation in a dose- and time-dependent manner. Investigation of the molecular mechanisms of cell cycle regulation confirmed that the oil arrested A549 cells in G2/M phase by decreasing the expression of cyclin B1-CDK1 and cyclin A-CDK1 and increasing the expression of cyclin-dependent kinase inhibitor (CKI) P21 at both the transcriptional and translational levels. Autophagy staining assays and western blot analysis revealed that CCEO promoted the formation of autophagic vacuoles in A549 cells and increased the expression of autophagy-related proteins beclin-1 and LC3-II in a dose-dependent manner. A series of apoptosis analyses indicated that CCEO induces apoptosis through a mitochondria-mediated intrinsic pathway. This study revealed that CCEO is a promising candidate for development into an anti-tumour drug of the future.

Abstract:Houttuynia cordata (Thunb.), an important medicinal plant of Northeast India, Korea, and China, is used to treat various ailments and for anticancer research. Knowing its traditional practices, we are interested in the mode-of-action of HCT on HepG2 to co-relate the traditional practice with modern drug therapeutics. UPLC-Q-ToF-Ms analysis of HCT reveals identification of 14 metabolites. Network pharmacology analysis of the 14 compounds showed interaction with 232 different targets with their potential involvement in hepatocellular carcinoma. Whole extracts impart cytotoxicity on variety of cell lines including HepG2. There was a significant morphological alteration in treated HepG2 cells due to impairment of cytoskeletal components like β and γ- tubulin. Arrest at G1-S checkpoint was clearly indicated downregulation of Cyclin D1. The root extracts actuated apoptosis in HepG2 as evident from altered mitochondrial membrane potential, Annexin V- FITC, BrdU-PI, AO/EtBr assays, and modulations of apoptotic protein expression but without ROS generation. Whole extracts caused abrogation of epithelial to mesenchymal transition with repression of Snail, N-Cadherin, Vimentin, MMP-9, and upregulation of Pan-Cadherin. Pathway analysis found GSK-3β in Wnt/β-Catenin signaling cascade to be involved through Hepatocellular carcinoma (hsa05225) pathway. The GSK-3β/β-Catenin/PDL-1 signaling was found to be inhibited with the downregulation of pathway components. This was further confirmed by application of EGF, an inducer of the GSK-3β/β-Catenin pathway that neutralized the effect of Houttuynia cordata (Thunb.) root extract on the said pathway. Network pharmacology analysis also confirms the synergy network with botanical-bioactive-target-disease which showed Kaempferol to have the highest degree of association with the said pathway.
Molecular Docking and Simulation-Binding Analysis of Plant Phytochemicals with the Hepatocellular Carcinoma Targets Epidermal Growth Factor Receptor and Caspase-9

Abstract:Among primary liver cancers, hepatocellular carcinoma (HCC) is one of the most common forms and it has been categorized as the joint-fourth largest reason of cancer-related deaths globally. Different factors such as alcohol abuse, hepatitis B and C, viral infections, and fatty liver diseases are mainly related to the pathogenesis of HCC. In the current study, 1000 total various plant phytochemicals were docked to proteins involved in HCC. The compounds were docked to the active site amino acids of epidermal growth factor receptor and caspase-9 as receptor proteins in order to explore their inhibiting potential. The top five compounds against each receptor protein were explored as potential drug candidates on the basis of their binding affinity and root-mean square deviation values. The top two compounds against each protein were found to be liquoric acid (S-score −9.8 kcal/mol) and madecassic acid (S-score −9.3 kcal/mol) against EGFR, and limonin (S-score −10.5 kcal/mol) and obamegine (S-score −9.3 kcal/mol) against the caspase-9 protein. The selected phytochemicals were further assessed through drug scanning using Lipinski's rule of five to explore their molecular properties and druggability. According to the ADMET analysis, the selected phytochemicals were found to be non-toxic and non-carcinogenic. Finally, the molecular dynamics simulation study revealed that liquoric acid and limonin were stabilized within the binding pockets of EGFR and capase-9, respectively, and stayed firmly bound throughout the simulation. In light of the current findings, the phytochemicals reported in this study, especially liquoric acid and limonin, could be used as potential drugs for the treatment of HCC in the future.

Abstract:We investigated the biological activities of 14 medicinal plants from Saudi Arabia as anti-inflammatory, antioxidant and anti-proliferative properties using hepatocellular carcinoma (HCC) cell lines. Hexane extracts exhibited the maximum inhibitory activities against albumin denaturation compared with those of other solvents. Baccharoides schimperi hexane extract showed the highest anti-inflammatory while Ocimum basilicum methanol extract showed significantly high hydroxyl radical scavenging activity (p < 0.001). B. schimperi methanol extract showed highest cytotoxicity and anti-proliferation activity (IC50; 25 µg/ml) in hepatocellular carcinoma cell line. Annexin V and caspase-9 activities were induced significantly (p < 0.001) by B. schimperi and the response increased in a dose dependent compared with untreated cells (p < 0.001). The docking scores showed a low docking energy binding of 2-Cyclohexylpiperidine of B. schimperi with COX-2, (-16.62 kcal/mol), high affinity for apoptotic and anti-inflammatory potency It can be concluded that, B. schimperi active ingredients as 2-Cyclohexylpiperidine and phytol contributed to its biological activity.
