Abstract:A new biomimetic surface named nano-micro binary polymer brushes is fabricated by large-area bench-top dip-pen nanodisplacement lithography technique. It is composed of gelatin-modified poly(glycidyl methacrylate) nanolines which are spaced by microstripes of poly(N-isopropylacrylamide). Spatio-Temporal Control of Cell Adhesion: Toward Programmable Platforms to Manipulate Cell Functions and Fate

Abstract:Biophysical and biochemical signals of material surfaces potently regulate cell functions and fate. In particular, micro- and nano-scale patterns of adhesion signals can finely elicit and affect a plethora of signaling pathways ultimately affecting gene expression, in a process known as mechanotransduction. Our fundamental understanding of cell-material signals interaction and reaction is based on static culturing platforms, i.e., substrates exhibiting signals whose configuration is time-invariant. However, cells in-vivo are exposed to arrays of biophysical and biochemical signals that change in time and space and the way cells integrate these might eventually dictate their behavior. Advancements in fabrication technologies and materials engineering, have recently enabled the development of culturing platforms able to display patterns of biochemical and biophysical signals whose features change in time and space in response to external stimuli and according to selected programmes. These dynamic devices proved to be particularly helpful in shedding light on how cells adapt to a dynamic microenvironment or integrate spatio-temporal variations of signals. In this work, we present the most relevant findings in the context of dynamic platforms for controlling cell functions and fate in vitro. We place emphasis on the technological aspects concerning the fabrication of platforms displaying micro- and nano-scale dynamic signals and on the physical-chemical stimuli necessary to actuate the spatio-temporal changes of the signal patterns. In particular, we illustrate strategies to encode material surfaces with dynamic ligands and patterns thereof, topographic relieves and mechanical properties. Additionally, we present the most effective, yet cytocompatible methods to actuate the spatio-temporal changes of the signals. We focus on cell reaction and response to dynamic changes of signal presentation. Finally, potential applications of this new generation of culturing systems for in vitro and in vivo applications, including regenerative medicine and cell conditioning are presented.

Abstract:This paper considers high-quality conductivity data for plasticized ionomers in the context of polymer local and segmental processes. Dielectric spectroscopy was conducted on a neat PEO-based ionomer and six mixtures containing 6 wt % plasticizer with a wide range of dielectric constants. Conductivity increased dramatically but remained Vogel Fulcher Tamman (VFT)-like for all plasticized ionomers, indicating that the mechanism of ion transport was unchanged. Relaxation times of the polymer local beta and segmental alpha processes were analyzed for the plasticized ionomers, providing activation energies and relaxation strengths for the beta process and VFT fitting parameters for the alpha process. The glass transition temperature T(g) of the mixtures was found to be the critical characteristic governing conductivity, based on four criteria: VFT-like behavior of conductivity, decrease in the conductivity-segmental coupling index upon the addition of plasticizer, statistical insignificance of solvent quality (dielectric constant, donor number, and viscosity) on conductivity, and the creation of a conductivity master curve as a function of T(g)-normalized temperature.

Abstract:Microtransfer molding of poly(Disperse Red 1) methacrylate enables the creation of photomodifiable nanotopography based on a cm‐scale master. By controlling the exposure of the nanotopography to light, a broad range of new patterns can be created and then molded into a material that is not photosensitive. These new patterns have many potential applications in the study of cell behavior. Nanotopographic surfaces are a powerful tool for studying and controlling cell behavior. However, the fabrication of nanotopographic master patterns using conventional photolithography is expensive, which limits the range of designs that can be explored. In this study, a method is demonstrated for the photoreshaping of large‐area patterns of nanoridges. The original master pattern is created using conventional lithography, and an azopolymer replica is prepared using soft lithography. The manipulation of the nanoridges is achieved by projecting light with specific polarizations and exposure times, resulting in controllable widening, buckling, or removal of the ridges. The reprogrammed azopolymer master patterns can then be replicated, creating reproducible new nanotopographies that can be transferred into other materials using a molding procedure. Diffraction can be used for in situ monitoring of the reprogramming during exposure. Image‐analysis methods are used to characterize buckled ridges as a function of exposure time. The response of MCF10A epithelial cells are investigated to buckled nanoridges. A substantial impact of buckling on the dynamics and location of actin polymerization, as well as on the distribution and lengths of contiguous polymerized regions is also observed.

Micropatterning of proteins and mammalian cells on biomaterials

Abstract:Controlling the spatial organization of cells is vital in engineering tissues that require precisely defined cellular architectures. For example, functional nerves or blood vessels form only when groups of cells are organized and aligned in very specific geometries. Yet, scaffold designs incorporating spatially defined physical cues such as microscale surface topographies or spatial patterns of extracellular matrix to guide the spatial organization and behavior of cells cultured in vitro remain largely unexplored. Here we demonstrate a new approach for controlling the spatial organization, spreading, and orientation of cells on two micropatterned biomaterials: chitosan and gelatin. Biomaterials with grooves of defined width and depth were fabricated using a two-step soft lithography process. Selective attachment and spreading of cells within the grooves was ensured by covalently modifying the plateau regions with commercially available protein resistant triblock copolymers. Precise spatial control over cell spreading and orientation has been observed when human microvascular endothelial cells are cultured on these patterned biomaterials, suggesting the potential of this technique in creating tissue culture scaffolds with defined chemical and topographical features.

Abstract:Microfluidic device with patterned microstructures on the substrate surface was used to regulate cell adhesion,morphology,and functions in tissue engineering.We developed a microfluidic device which employing microscale patterned microstructures to achieve enhanced cell adhesion and migration,Biocompatible hydrogel substrates with micro-wavy and lattice-patterned microstructures were fabricated using standing surface acoustic waves and ultraviolet solidification.After seeding the L929 mouse fibroblast cells onto the patterned substrate of the microfluidic device,we determined that the viability and proliferation rate of cell migration can be greatly enhanced.Furthermore,L929 cells showed two types of gathering modes after 48 h of culturing.Cell growth was guided by the patterned substrate used in the microfluidic device and showed differences in the location distribution.Therefore,the developed microfluidic device with patterned microstructures can extend the application of in vitro cell culturing for future drug development and disease diagnosis.

Abstract:The possibility to reproduce in vitro the complex multiscale physical features present in the human tissues creates novel opportunities for biomedical advances and fundamental understanding of cell‐environment interfaces and interactions. While stiffness has been recognized as a key property in influencing cell behavior, so far systematic studies on the role of stiffness have been limited to values in the KPa‐MPa range, significantly below the stiffness of bone. Here, we report a platform enabling the tuning and control of the stiffness of a biocompatible polymeric interface up to values characteristic of the bone tissue, which are in the GPa range. The ability to fine tune the stiffness up to these large values is achieved by using extremely thin polymer films on glass and cross‐linking the films using UV light irradiation. We show that a higher stiffness is related to better adhesion, proliferation, and osteogenic differentiation, and that it is also possible to switch on/off cell attachment and growth by solely tuning the stiffness of the interface, without any surface chemistry or topography modification. Since the stiffness is tuned directly by UV irradiation, this platform is ideal for rapid and simple stiffness patterning, and stiffness gradients fabrication. This materials platform represents an innovative tool for combinatorial studies of the synergistic effect of tissue environmental cues on cell behavior, and creates new opportunities for next generation biosensors, single‐cell patterning, and lab‐on‐a‐chip devices. Programmed Topographic Substrates for Studying Roughness Gradient-Dependent Cell Migration Using Two-Photon Polymerization

Abstract:The mediation of the extracellular matrix is one of the major environmental cues to direct cell migration, such as stiffness-dependent durotaxis and adhesiveness-dependent haptotaxis. In this study, we explore another possible contact guidance: roughness dependent topotaxis. Different from previously reported studies on topotaxis that use standard photolithography to create micron or submicron structures that have identical height and different spatial densities, we develop a new method to programmatically fabricate substrates with different patterns of surface roughness using two-photon polymerization. Surface roughness ranging from 0.29 to 1.11 μm can be created by controlling the voxel distance between adjacently cured ellipsoid voxels. Patterned Ormocomp ® masters are transferred to polypropylene films using the nanoimprinting method for cell migration study. Our experimental results suggest that MG63 cells can sense the spatial distribution of their underlying extracellar roughness and modulate their migration velocity and direction. Three characteristic behaviors were identified. First, cells have a higher migration velocity on substrates with higher roughness. Second, cells preferred to migrate from regions of higher roughness to lower roughness, and their migration velocity also decreased with descending roughness. Third, the migration velocity remained unchanged on the lower roughness range on a graded substrate with a steeper roughness. The last cell migration characteristic suggests the steepness of the roughness gradient can be another environmental cue in addition to surface roughness. Finally, the combination of two-photon polymerization and nanoimprint methods could become a new fabrication methodology to create better 3D intricate structures for exploring topotactic cell migrations.

Abstract:We have developed a novel experimental set-up that simultaneously, (i) applies static and dynamic deformations to adherent cells in culture, (ii) allows the visualization of cells under fluorescence microscopy, and (iii) allows atomic force microscopy nanoindentation measurements of the mechanical properties of the cells. The cell stretcher device relies on a dielectric elastomer film that can be electro-actuated and acts as the cell culture substrate. The shape and position of the electrodes actuating the film can be controlled by design in order to obtain specific deformations across the cell culture chamber. By using optical markers we characterized the strain fields under different electrode configurations and applied potentials. The combined setup, which includes the cell stretcher device, an atomic force microscope, and an inverted optical microscope, can assess in situ and with sub-micron spatial resolution single cell topography and elasticity, as well as ion fluxes, during the application of static deformations. Proof of performance on fibroblasts shows a reproducible increase in the average cell elastic modulus as a response to applied uniaxial stretch of just 4%. Additionally, high resolution topography and elasticity maps on a single fibroblast can be acquired while the cell is deformed, providing evidence of long-term instrumental stability. This study provides a proof-of-concept of a novel platform that allows in situ and real time investigation of single cell mechano-transduction phenomena with sub-cellular spatial resolution.

Biointerfaces with ultrathin patterns for directional control of cell migration

Abstract:Abstract In the context of wound healing and tissue regeneration, precise control of cell migration direction is deemed crucial. To address this challenge, polydimethylsiloxane (PDMS) platforms with patterned 10 nm thick TiO x in arrowhead shape were designed and fabricated. Remarkably, without tall sidewall constraints, MC3T3-E1 cells seeded on these platforms were constrained to migrate along the tips of the arrowheads, as the cells were guided by the asymmetrical arrowhead tips which provided large contact areas. To the best of our knowledge, this is the first study demonstrating the use of thin TiO x arrowhead pattern in combination with a cell-repellent PDMS surface to provide guided cell migration unidirectionally without tall sidewall constraints. Additionally, high-resolution fluorescence imaging revealed that the asymmetrical distribution of focal adhesions, triggered by the patterned TiO x arrowheads with arm lengths of 10, 20, and 35 μm, promoted cell adhesion and protrusion along the arrowhead tip direction, resulting in unidirectional cell migration. These findings have important implications for the design of biointerfaces with ultrathin patterns to precisely control cell migration. Furthermore, microelectrodes were integrated with the patterned TiO x arrowheads to enable dynamic monitoring of cell migration using impedance measurement. This microfluidic device integrated with thin layer of guiding pattern and microelectrodes allows simultaneous control of directional cell migration and characterization of the cell movement of individual MC3T3-E1 cells, offering great potential for the development of biosensors for single-cell monitoring.

Abstract:Engineering 3D microstructures with predetermined properties is critical for stem cell niche studies. We have developed a multiphoton femtosecond laser-based 3D printing platform, which generates complex protein microstructures in minutes. Here, we used the platform to test a series of fabrication and reagent parameters in precisely controlling the mechanical properties of protein micropillars. Atomic force microscopy was utilized to measure the reduced elastic modulus of the micropillars and transmission electron microscopy was used to visualize the porosity of the structures. The reduced elastic modulus of the micropillars associated positively and linearly with the scanning power. On the other hand, the porosity and pore size of the micropillars associated inversely and linearly with the scanning power and reagent concentrations. While keeping the elastic modulus constant, the stiffness of the micropillars was controlled by varying their height. Subsequently, the single cell traction forces of rabbit chondrocytes, human dermal fibroblasts, human mesenchymal stem cells and bovine nucleus pulposus cells (bNPCs) were successfully measured by culturing the cells on micropillar arrays of different stiffness. Our results showed that the traction forces of all groups showed positive relationship with stiffness and that the chondrocytes and bNPCs generated the highest and lowest traction forces, respectively.

Abstract:Mechanical stress is a decisive factor for the differentiation, proliferation, and general behavior of cells. However, the specific signaling of mechanotransduction is not filly understood. One basic problem is the clear distinction between. the different extracellular matrix (ECM) constituents that participate in cellular adhesion and their corresponding signaling pathways. Here, a system is proposed that enables mechanical stimulation of human-skin-derived keratinocytes and human dermal fibroblasts that specifically interact with peptide sequences immobilized on a non-interacting but deformable substrate. The peptide sequences mimic fibronectin, laminin, and collagen type TV, three major components of the ECM. To achieve this, PDMS is activated using ammonia plasma and coated with, star-shaped isocyanate-terminated poly(ethylene glycol)-based prepolymers, which results in a functional coating that prevents unspecific cell adhesion. Specific cell adhesion is achieved by functionalization of the layers with the peptide sequences in different combinations. Moreover, a method that enables the decoration of deformable substrates with cell-adhesion-peptides in extremely defined nanostructures is presented. The distance and clustering of cell adhesion molecules below 100 nm has been demonstrated to be of utmost importance for cell adhesion. Thus we present a new toolbox that allows for the detailed analysis of the adhesion of human-skin-derived cells on structurally and biochemically decorated deformable substrates.
Femtosecond-laser-assisted spatial cell adhesion to microstructured surface geometry

Abstract:This article discusses the adhesion of C2C12 mouse myoblast cells to a microstructured polydimethylsiloxane (PDMS) surface patterned using femtosecond laser pulses. The wettability of the PDMS surface can be controlled by changing the writing-pulse energy; a hydrophilic surface is produced by low pulse energy, whereas high pulse energies lead to a superhydrophobic surface. The surface topography also varies with pulse energy. Images acquired with scanning electron microscopy show clear lines at low pulse energy, whereas at high energies, the lines are completely deformed by the presence of micro- and nano-structures. Thus, selective cell growth in the modified regions is affected by the energy of the laser pulses used for surface modification. In addition, the surface geometry (e.g., lines vs grids) of the modified regions affects the shape and alignment of C2C12 cells. Thus, we investigate the degree of cell alignment to modified lines fabricated with the same pulse energy and writing speed but with different inter-line spacings. The degree of alignment is quantified by the average value of a second-order Legendre polynomial. The results reveal that the degree of alignment of C2C12 cells to the surface lines decreases with the increase in spacing between lines.

A Scalable Method to Fabricate 2D Hydrogel Substrates for Mechanobiology Studies with Independent Tuning of Adhesiveness and Stiffness

Abstract:Mechanical signals from the extracellular matrix are crucial in guiding cellular behavior. Two-dimensional hydrogel substrates for cell cultures serve as exceptional tools for mechanobiology studies because they mimic the biomechanical and adhesive characteristics of natural environments. However, the interdisciplinary knowledge required to synthetize and manipulate these biomaterials typically restricts their widespread use in biological laboratories, which may not have the material science expertise or specialized instrumentation. To address this, we propose a scalable method that requires minimal setup to produce 2D hydrogel substrates with independent modulation of the rigidity and adhesiveness within the range typical of natural tissues. In this method, norbornene-terminated 8-arm polyethylene glycol is stoichiometrically functionalized with RGD peptides and crosslinked with a di-cysteine terminated peptide via a thiol-ene click reaction. Since the synthesis process significantly influences the final properties of the hydrogels, we provide a detailed description of the chemical procedure to ensure reproducibility and high throughput results. We demonstrate examples of cell mechanosignaling by monitoring the activation state of the mechanoeffector proteins YAP/TAZ. This method effectively dissects the influence of biophysical and adhesive cues on cell behavior. We believe that our procedure will be easily adopted by other cell biology laboratories, improving its accessibility and practical application.
Guided Cell Migration on Microtextured Substrates with Variable Local Density and Anisotropy

Abstract:This work reports the design of and experimentation with a topographically patterned cell culture substrate of variable local density and anisotropy as a facile and efficient platform to guide the organization and migration of cells in spatially desirable patterns. Using UV‐assisted capillary force lithography, an optically transparent microstructured layer of a UV curable poly(urethane acrylate) resin is fabricated and employed as a cell‐culture substrate after coating with fibronectin. With variable local pattern density and anisotropy present in a single cell‐culture substrate, the differential polarization of cell morphology and movement in a single experiment is quantitatively characterized. It is found that cell shape and velocity are exquisitely sensitive to variation in the local anisotropy of the two‐dimensional rectangular lattice arrays, with cell elongation and speed decreasing on symmetric lattice patterns. It is also found that cells could integrate orthogonal spatial cues when determining the direction of cell orientation and movement. Furthermore, cells preferentially migrate toward the topographically denser areas from sparser ones. Consistent with these results, it is demonstrated that systematic variation of local densities of rectangular lattice arrays enable a planar assembly of cells into a specified location. It is envisioned that lithographically defined substrates of variable local density and anisotropy not only provide a new route to tailoring the cell‐material interface but could serve as a template for advanced tissue engineering. Microtextured substrates with variable local density and anisotropy (see image) are designed to guide the organization and migration of fibroblasts in spatially desirable locations. Cell motility is sensitive to variation in the local density and anisotropy of rectangular lattice, with cell elongation and speed decreasing on symmetric lattice. Also, cells integrate orthogonal spatial cues when determining the direction of their orientation and movement.

Abstract:Control of stem cell behaviors at solid biointerfaces is critical for stem-cell-based regeneration and generally achieved by engineering chemical composition, topography, and stiffness. However, the influence of dynamic stimuli at the nanoscale from solid biointerfaces on stem cell fate remains unclear. Herein, we show that electrochemical switching of a polypyrrole (Ppy) array between nanotubes and nanotips can alter surface adhesion, which can strongly influence mechanotransduction activation and guide differentiation of mesenchymal stem cells (MSCs). The Ppy array, prepared via template-free electrochemical polymerization, can be reversibly switched between highly adhesive hydrophobic nanotubes and poorly adhesive hydrophilic nanotips through an electrochemical oxidation/reduction process, resulting in dynamic attachment and detachment to MSCs at the nanoscale. Multicyclic attachment/detachment of the Ppy array to MSCs can activate intracellular mechanotransduction and osteogenic differentiation independent of surface stiffness and chemical induction. This smart surface, permitting transduction of nanoscaled dynamic physical inputs into biological outputs, provides an alternative to classical cell culture substrates for regulating stem cell fate commitment. This study represents a general strategy to explore nanoscaled interactions between stem cells and stimuli-responsive surfaces.
Selective hydrophilic modification of Parylene C films: a new approach to cell micro-patterning for synthetic biology applications

Abstract:We demonstrate a simple, accurate and versatile method to manipulate Parylene C, a material widely known for its high biocompatibility, and transform it to a substrate that can effectively control the cellular microenvironment and consequently affect the morphology and function of the cells in vitro. The Parylene C scaffolds are fabricated by selectively increasing the material's surface water affinity through lithography and oxygen plasma treatment, providing free bonds for attachment of hydrophilic biomolecules. The micro-engineered constructs were tested as culture scaffolds for rat ventricular fibroblasts and neonatal myocytes (NRVM), toward modeling the unique anisotropic architecture of native cardiac tissue. The scaffolds induced the patterning of extracellular matrix compounds and therefore of the cells, which demonstrated substantial alignment compared to typical unstructured cultures. Ca(2+) cycling properties of the NRVM measured at rates of stimulation 0.5-2 Hz were significantly modified with a shorter time to peak and time to 90% decay, and a larger fluorescence amplitude (p < 0.001). The proposed technique is compatible with standard cell culturing protocols and exhibits long-term pattern durability. Moreover, it allows the integration of monitoring modalities into the micro-engineered substrates for a comprehensive interrogation of physiological parameters.

Abstract:A detailed understanding of the interface between living cells and substrate materials is of rising importance in many fields of medicine, biology and biotechnology. Cells at interfaces often form epithelia. The physical barrier that they form is one of their main functions. It is governed by the properties of the networks forming the cytoskeleton systems and by cell-to-cell contacts. Different substrates with varying surface properties modify the migration velocity of the cells. On the one hand one can change the materials composition. Organic and inorganic materials induce differing migration velocities in the same cell system. Within the same class of materials, a change of the surface stiffness or of the surface energy modifies the migration velocity, too. For our cell adhesion studies a variety of different, homogeneous substrates were used (polymers, bio-polymers, metals, oxides). In addition, an effective lithographic method, Polymer Blend Lithography (PBL), is reported, to produce patterned Self-Assembled Monolayers (SAM) on solid substrates featuring two or three different chemical functionalities. This we achieve without the use of conventional lithography like e-beam or UV lithography, only by using self-organization. These surfaces are decorated with a Teflon-like and with an amino-functionalized molecular layer. The resulting pattern is a copy of a previously created self-organized polymer pattern, featuring a scalable lateral domain size in the sub-micron range down below 100 nanometers. The resulting monolayer pattern features a high chemical and biofunctional contrast with feature sizes in the range of cell adhesion complexes like e.g. focal adhesion points.

Abstract:An efficient platform capable of cell adhesion needs to be developed to understand cell activities such as cell differentiation, diffusion, and migration. The basic sequence of cell adhesion involves cells communicating with their environment by generating mechanical and chemical signals. Thin polymeric films with micro- or nano-patterns are widely used to support cell growth with conformal contact at the biointerface. However, stable and biocompatible films with high reproducibility on a flexible substrate remain a challenge. As described here, we developed micro-pattern poly(tetrafluoroethyleneco-perfluoro-3,6-dioxa-4-methyl-7-octenesulfonic acid) (Nafion) films fabricated by a molding process. We present the fabrication and characterization of flexible, micro-patterned Nafion films and the evaluation of cell adhesion and alignment on these films. We found that cell adhesion and migration/direction could be modulated by controlling the surface architecture. This approach offers a new platform that constitutes a promising tool for use in flexible cell-based platforms and devices to observe cell-cell and cell-surface interactions.
